Vers des assemblages de complexes métalliques oligonucléaires, servant d’antenne solaire au niveau moléculaire

Idéalement, ces systèmes seraient capables de fournir l’énergie et la séparation de charge nécessaire pour catalyser des réactions à transfert multiélectroniques, tel que l’hydrolyse de l’eau ou la réduction du gaz carbonique. La réalisation d’un tel système catalytique créerait une source d’énergie renouvelable, sous forme d’énergie chimique, crée directement à partir de l’énergie solaire. Le système envisagé, schématisé sous la forme d’une antenne, possède trois parties distinctes. Tout d’abord, des chromophores forment un état excité en captant l’énergie de la lumière visible du soleil. Vient ensuite un centre de liaison qui lie tous les chromophores et qui collecte l’énergie de cet état excité à travers un transfert d’électron. Cet électron est de nouveau transféré vers la dernière partie, un centre réactionnel catalytique. Cet assemblage permet de créer une séparation de charge entre le chromophore et le centre réactionnel qui sont séparés par le centre de liaison, évitant ainsi la recombinaison de charge. Le projet se focalise sur la synthèse, la caractérisation et l’application en photocatalyse d’assemblages chromophore–centre de liaison–catalyseur. Tout d’abord, une étude de chromophores à base de fluorène et de rhénium a été effectuée dans le but d’évaluer le transfert électronique entre ces deux composants. Ensuite, des centres de liaisons à base de dimère de rhodium tétraamidinate ont été créés et étudiés afin d’établir leurs caractéristiques photophysiques et électrochimiques. Puis un d’entre eux a été assemblé avec des chromophores de rhénium, créant ainsi des espèces moléculaires discrètes contenant d’un à quatre chromophores. Et pour finir, ces assemblages ont été combinés avec un catalyseur à base de cobalt, puis ont été testés dans des expériences de photoproduction d’hydrogène. Cette dernière partie a requis l’élaboration d’un photoréacteur qui est aussi décrite en détail dans cet ouvrage. / This research project involves synthetic metallosupramolecular systems developed to mimic the natural process of photosynthesis. Ideally, these systems would be able to provide the energy and the charge separation needed to catalyze multielectron-transfer reactions, such as water-splitting or carbon dioxide reduction. The realization of such a catalytic system would create a renewable energy source, in the form of chemical energy, created directly from solar energy. The system envisioned has three distinct parts in the form of an antenna. First of all, chromophores go into an excited state, while capturing the visible light energy of the Sun. Then comes a hub which binds all the chromophores and collects this excited state energy through an electron transfer. This electron is then transferred again to the last part, a catalytic reaction center. This assembly creates a charge separation between the chromophore and the reaction center which are separated by the hub, thus avoiding the recombination of charge. The project focuses on the synthesis, characterization and application in photocatalysis of chromophore-hub-catalyst assemblies. First of all, a study of fluorene and rhenium based chromophores was made to assess the electronic transfer between these two components. Then, tetraamidinate rhodium dimer based hubs have been created and studied in order to establish their photophysical and electrochemical characteristics. Then one of these assemblies was formed with chromophores of rhenium, thus creating discrete molecular species containing one to four chromophores. And finally, these assemblies were combined with a cobalt-based catalyst and were tested for hydrogen photoproduction. The latter required the development of a photoreactor which is also described in detail in this thesis.

O tratamento baseia-se na irradiação e destruição do tumor, preservando os órgãos normais quanto ao excesso de radiação. Radionuclídeos emissores β- como 90Y, 131I, 177Lu e 188Re, são úteis para o desenvolvimento de radiofármacos terapêuticos e, quando associados a AcM como o Anti-CD20 são importantes principalmente para o tratamento de Linfomas Não Hodgkins (LNH). 188Re (Eβ- = 2,12 MeV; Eγ= 155 keV; t1/2 = 16,9 h) é um radionuclídeo atrativo para RIT. O Centro de Radiofarmácia do IPEN possui um projeto que visa a produção do radiofármaco 188Re-Anti-CD20. Com isso,este estudo foi proposto para avaliar a eficácia desta técnica de marcação para tratamento em termos compartimentais e dosimétricos. O objetivo deste trabalho consistiu na compararação da marcação do AcM anti-CD20 com 188Re com a marcação do anticorpo com 90Y, 131I, 177Lu e 99mTc (pelas suas características químicas similares) e 211At, 213Bi, 223Ra e 225Ac. Através do estudo de técnicas de marcação relatadas em literatura, foi proposto um modelo compartimental para avaliação de sua farmacocinética e estudos dosimétricos, de alto interesse para a terapia. A revisão de dados publicados na literatura, possibilitou demonstrar diferentes procedimentos de marcação, rendimentos de marcação, tempo de reação, impurezas e estudos de biodistribuição. O resultado do estudo mostra uma cinética favorável para o 188Re, pelas suas características físicas e químicas frente aos demais radionuclídeos avaliados. O estudo compartimental proposto descreve o metabolismo do 188Re-anti-CD20 através de um modelo compartimental mamilar, que pela sua análise farmacocinética, realizada em comparação aos produtos marcados com emissores β-: 131I-antiCD20, 177Lu-anti-CD20, o emissor γ 99mTc-anti-CD20 e o emissor α 211At-Anti-CD20, apresentou uma constante de eliminação de aproximadamente 0,05 horas-1 no sangue do animal. A avaliação dosimétrica do 188Re-Anti-CD20 foi realizada através de duas metodologias: pelo método de Monte Carlo e pelo uso de uma fonte pontual β- através da Fórmula de Loevinger via programa Excel. Através da Fórmula de Loevinger fez-se a validação do método de Monte Carlo para a dosimetria do 188Re-Anti-CD20 e dos demais produtos. As doses e as taxas de doses obtidas pelos dois métodos foram avaliadas em comparação à dosimetria do 90Y-Anti-CD20, 131I-Anti-CD20 e do 177Lu-Anti-CD20, obtidas pela mesma metodologia. O estudo de dose foi realizado utilizando modelos matemáticos considerando um camundongo nude de 25g, simulando diferentes tamanhos de tumor e diferentes formas de distribuição do produto dentro do animal. De acordo com os resultados obtidos, pela energia de emissão β-, 188Re-Anti-CD20 apresenta maior deposição de energia para tumores volumosos em relação aos demais produtos avaliados. Em uma simulação com 100% do produto captado pelo tumor, 89% da dose total manteve-se absorvida pelo tumor, preservando a integridade de ógãos críticos como coração (2%), pulmões (5%), coluna (4%), fígado (0,014%) e rins (0,0007%). Em uma simulação onde há uma biodistribuição do produto no organismo do animal, 38% da dose total é absorvida pelo tumor e >3% é absorvida pela coluna. Nessa situação mais próxima da realidade, a extrapolação dos dados para um humano de 70kg, mostrou que a dose absorvida no tumor corresponde a cerca de 33%; na coluna 7% e o coração receberia uma dose de 35% do total. - 8 - ABSTRACT (EN) The thesis is submitted as a commented set of reviewed publications documenting and depicting the possibilities of mass spectrometry in the field of chemical, biological and pharmaceutical research; namely for the purposes of structure elucidation of selected organometallic complexes, analyses of drugs and their metabolites, monitoring of important biological markers. In course of experimental work, the following objectives were studied and solved:  Proposal and realization of micro-scale preparation of selected rhenium complexes with aromatic ligands, utilizing tetrabutyammonium tetrachlorooxorhenate as a starting material; preparation and structure characterization of oxorhenium(V) complexes with 1,2-dihydroxybenzene, 1,2,3-trihydroxybenzene, and 2,3- dihydroxynaphtalene as ligands by means of ESI/MS, APPI/MS and LDI-MS; ESI/MS and UV/Vis study of kinetic behavior of complexes arising from the reaction of tetrabutylamonnium tetrachlooxorhenate with pyrogallol and catechol as ligands. A Re-Os Study of Sulfides from the Bagdad Porphyry Cu-Mo Deposit, Northern Arizona, USA

Use of Re-Os systematics in sulfides from the Bagdad porphyry Cu-Mo deposit provide information on the timing of mineralization and the source of the ore -forming elements. Analyzed samples of pyrite, chalcopyrite and molybdenite mainly from the quartz monzonite and porphyritic quartz monzonite units are characterized by a moderate to strong potassic alteration (secondary biotite and K- feldspar). Rhenium concentrations in molybdenite are between 330 and 730 ppm. Two molybdenite samples from the quartz monzonite and porphyritic quartz monzonite provide a Re-Os isotope age of 71.7 ± 0.3 Ma. A third sample from a molybdenite vein in Precambrian rocks yields an age of 75.8 ± 0.4 Ma. These molybdenite ages support previous suggestions of two mineralization episodes in the Bagdad deposit. An early event at 76 Ma and a later episode at 72 Ma. Pyrite Os and Re concentrations range between 0.008-0.016 and 3.9-6.8 ppb, respectively. Chalcopyrite contains a wide range of Os (6 to 91 ppt) and Re (1.7 to 69 ppb) concentrations and variable ¹⁸⁷Os/¹⁸⁸Os ratios that range between 0.13 to 22.27. This variability in the chalcopyrite data may be attributed to different copper sources, one of them the Proterozoic volcanic massive sulfides in the district, or to alteration and remobilization of Re and Os. Analyses from two pyrite samples yield an eight point isochron with an age of 77 ± 15 Ma and an initial ¹⁸⁷Os/¹⁸⁸Os ratio of 2.12. This pyrite Re-Os isochron age is in good agreement with the molybdenite ages. We interpret the highly radiogenic initial 1870s/188Os as an indication that the source of Os and, by inference, the ore-forming elements for the Bagdad deposit, was mainly the crust. This conclusion agrees with previous Pb and Nd isotope studies and supports the notion that a significant part of the metals and magmas have a crustal source.

Développement et caractérisation de dérivés dipyrrométhène pour des applications dans le domaine du photovoltaïque

Ce projet de recherche mené en collaboration industrielle avec St-Jean Photochimie Inc. / PCAS Canada vise le développement et la caractérisation de dérivés dipyrrométhène pour des applications dans le domaine du photovoltaïque. La quête du récoltage des photons se situant dans le proche-infrarouge a été au centre des modifications structurales explorées afin d’augmenter l’efficacité de conversion des cellules solaires de type organique et à pigments photosensibles. Trois familles de composés intégrant le motif dipyrrométhène ont été synthétisées et caractérisées du point de vue spectroscopique, électrochimique, structural ainsi que par modélisation moléculaire afin d’établir des relations structures-propriétés. La première famille comporte six azadipyrrométhènes au potentiel de coordination tétradentate sur des centres métalliques. Le développement d’une nouvelle voie synthétique asymétrique combinée à l’utilisation d’une voie symétrique classique ont permis d’obtenir l’ensemble des combinaisons de substituants possibles sur les aryles proximaux incluant les noyaux 2-hydroxyphényle, 2-méthoxyphényle et 2- pyridyle. La modulation du maximum d’absorption dans le rouge a pu être faite entre 598 et 619 nm. De même, la présence de groupements méthoxyle ou hydroxyle augmente l’absorption dans le violet (~410 nm) tel que démontré par modélisation. La caractérisation électrochimique a montré que les dérivés tétradentates étaient en général moins stables aux processus redox que leur contre-parti bidentate. La deuxième famille comporte dix dérivés BODIPY fusionnés de façon asymétrique en position [b]. L’aryle proximal a été modifié de façon systématique afin de mieux comprendre l’impact des substituents riches en électron et de la fusion de cycles aromatiques. De plus, ces dérivés ont été mis en relation avec une vaste série de composés analogues. Les résultats empiriques ont montré que les propriétés optoélectroniques de la plateforme sont régies par le degré de communication électronique entre l’aryle proximal, le pyrrole sur lequel il est attaché et le noyau indolique adjacent à ce dernier. Les maximums d’absorption dans le rouge sont modulables entre 547 et 628 nm et la fluorescence des composés se situe dans le proche- infrarouge. L’un des composé s’est révélé souhaitable pour une utilisation en photovoltaïque ainsi qu’à titre de sonde à pH. La troisième famille comporte cinq complexes neutres de RuII basés sur des polypyridines et portant un ligand azadipyrrométhène cyclométalé. Les composés ont montré une forte absorption de photons dans la région de 600 à 800 nm (rouge à proche- infrarouge) et qui a pu être étendue au-delà de 1100 nm dans le cas des dérivés portant un ligand terpyridine. L’analyse des propriétés optoélectroniques de façon empirique et théorique a montré un impact significatif de la cyclométalation et ouvert la voie pour leur étude en tant que photosensibilisateurs en OPV et en DSSC. La capacité d’un des complexes à photo-injecter un électron dans la bande de conduction du semi-conducteur TiO2 a été démontré en collaboration avec le groupe du Pr Gerald J. Meyer à University of North Carolina at Chapel Hill, premier pas vers une utilisation dans les cellules solaires à pigments photosensibles. La stabilité des complexes en solution s’est toutefois avérée problématique et des pistes de solutions sont suggérées basées sur les connaissances acquises dans le cadre de cette thèse. / This research project carried out in industrial collaboration with Saint-Jean Photochemicals Inc. / PCAS Canada aims at the development and characterization of dipyrromethene derivatives for photovoltaic applications. The quest for harvesting near- infrared photons was the central focus and various structural modifications were explored to improve the power conversion efficiency of organic and dye-sensitized solar cells (OPV and DSSC, respectively). Three families of chromophores which embedded a dipyrromethene motif were synthesized and characterized through spectroscopy, electrochemistry, X-ray diffraction and computationnal modelization in order to establish their structure-properties relationship. The first family includes six azadipyrromethenes with potential for tetradentate coordination on metallic centers. The development of a new asymmetric synthetic route together with the classical symmetric one allowed access to all possible combinations of derivatives including 2-hydroxyphenyl, 2-methoxyphenyl and 2-pyridyl substituents in the proximal position of the dipyrromethene. Modulation of the absorption maxima in the red ranged between 598 and 619 nm. Also, having methoxy or hydroxy substituents provided an increase of the violet absorption (~410 nm) as established by modelization. Electrochemical characterization showed that the tetradentate azadipyrromethenes were generally less stable towards redox processes as compared to their bidentate counter- parts. The second family includes ten asymmetric benzo[b]-fused BODIPYs where the proximal aryl was systematically modified in order to assess the impact of electron-rich substituents and fused aromatic cycles. The derivatives were further compared to a wide series of related BODIPYs. Empirical results showed the optoelectronic properties are dictated by the extend of electronic communication between the proximal aryl, the pyrrol to which it is attached and the adjacent indolic moiety. Absorption maxima in the red were modulated between 547 nm and 628 nm and the fluorescence was in the near-infrared. One compound proved to be a potential candidate for photovoltaic and pH probe applications. The third family includes five neutral RuII polypyridine complexes bearing a cyclometalated azadipyrromethene ligand. The compounds exhibit strong light absorption in the 600 – 800 nm range (red to near-infrared) that tails beyond 1100 nm in the terpyridine-based adducts. Analysis of the optoelectronic properties showed a significant impact of this novel cyclometalation strategy for dipyrromethene derivatives and paved the way for further incorporation of the resulting complexes as photosensitizers in OPV and DSSC. In collaboration with the group of Pr Gerald J. Meyer at the University of North Carolina at Chapel Hill, the capacity of one compound to photo-inject its electron into the conduction band of the TiO2 semiconductor was established, a first step towards their use in dye-sensitized solar cells. The structural instability in solution of the complexes hindered their full potential for photovoltaic applications and suggestions to improve them are proposed based on the knowledge acquired in the course of this thesis.
