The complexes of oxidative phosphorylation (OXPHOS) are situated in the inner mitochondrial membrane in higher structural and functional complexes, so-called supercomplexes, which facilitates substrate channeling. ATP synthase is also able to organize in higher structures. In mammalian mitochondria, ATP synthase is usually present in a dimeric form. There is evidence of its trimerization and even tetramerization. Furthermore, it seems that ATP synthase catalyzing the phosphorylation of ADP to ATP, adenine nucleotide translocator (ANT) ensuring the exchange of ADP for newly synthesized ATP across the inner mitochondrial membrane and phosphate carrier (PiC) allowing the import of inorganic phosphate (Pi) into the matrix of mitochondria are assembled in a supercomplex called ATP synthasome. This association among the components of phosphorylative apparatus seems to increase the efficiency of processes leading to the ATP synthesis. First, we studied amounts of the components of phosphorylative apparatus in connection with various ATP synthase contents among mitochondria isolated from nine rat tissues. Estudo e caracterização geológica e geotécnica de rejeitos de mineração: Adrianópolis (PR) / Geological and geotechnical study and characterization in tailing: Adrianópolis (PR)

Os rejeitos de mineração provenientes do beneficiamento do minério, quando dispostos de maneira inadequada, podem causar a contaminação do solo e das águas superficiais e subterrâneas. Dependendo do tipo de minério tratado, tais rejeitos podem conter elevadas concentrações de metais potencialmente tóxicos e passar a atuar como fontes persistentes de liberação de tais metais através dos ecossistemas. Assim sendo, a caracterização desses rejeitos é extremamente importante. Neste contexto, foram realizadas caracterizações geológicas e geotécnicas nos rejeitos de mineração provenientes da região do Vale do Ribeira (município de Adrianópolis-PR), visando avaliar o grau de mobilidade e toxicidade dos metais contidos na porção superficial deste depósito. Para tanto, foram realizadas caracterizações mineralógicas (DRX e MEV/EDS), granulométricas, determinação dos parâmetros físico-químicos (pH, Eh e CE), além de ensaios que verificaram a mobilidade, especiação química e toxicidade das amostras: ensaios de solubilização, lixiviação, ensaio de variação de pH, extração sequencial e ecotoxicidade aquática envolvendo o organismo teste Daphnia similis. Além dos rejeitos foram realizadas análises mineralógicas e determinação dos parâmetros físico-químicos em amostras de estéril. A partir destas caracterizações, foi possível verificar que a composição mineralógica predominantemente no rejeito e no estéril é o carbonato, reflexo da litologia local. Tal composição refletiu também nos elevados valores de pH, que variaram de 7,6 a 8,0. Esses rejeitos apresentaram granulometria fina (predominância de areia muito fina, fina e silte), além de altas concentrações de metais, principalmente Pb (concentração média de 5.236,67 mg.kg-1 ) e Zn (7.726,53 mg.kg-1 ). Dos metais analisados, o Pb excedeu o valor preconizado na norma ABNT NBR 10.004:2004, tanto nos extratos lixiviados como nos solubilizados, para todas as amostras analisadas, o que permitiu classificar tais resíduos como Classe I - Perigoso. No ensaio de variação de pH, de forma geral, o Zn apresentou maior liberação na condição inicial mais ácida (pH 3), com média de liberação de 11,54 mg.L-1 após sessenta minutos de ensaio; embora nas outras condições também tenha ocorrido liberação considerável. A partir da extração sequencial, foi possível verificar que todos os metais (com exceção do Cu) estavam associados à fase de óxidos/hidróxidos, porém as fases consideradas de maior mobilidade (trocável e carbonática) estavam presentes em concentrações significativas para todos os metais analisados. As cinco amostras analisadas mostraram-se heterogêneas sendo que a RP5 e a RP7, apresentaram maior mobilidade dos metais Pb e Zn, além de terem apresentado maior toxicidade nos ensaios de ecotoxicidade. Para o ensaio de ecotoxicidade aquática, tais amostras apresentaram imobilidade dos neonatos até mesmo nas diluições menos concentradas (diluições contendo somente 6,2 e 3,1% de extrato solubilizado). Assim, concluiu-se que o rejeito pode ser considerado perigoso do ponto de vista ambiental, já que as altas concentrações de metais potencialmente tóxicos presentes estão passíveis de liberação e migração no ecossistema, podendo interagir com a biota aquática e terrestre. / Tailings from the ore processing when improperly disposed, can cause contamination of soil, surface water and groundwater. Depending on the treated ore, the mining waste may contain high concentrations of toxic metals and start acting as persistent metals sources to surrounding ecosystems. Therefore, the characterization of this material is extremely important. In this context, geological and geotechnical characterization were performed on tailings from Ribeira Valley (Adrianópolis-PR), in order to evaluate the mobility and toxicity of metals present in the superficial portion of the deposit. For the current paper the following tests were performed: mineralogical characterization (XRD and SEM/EDS), particle size distribution, determination of physical and chemical properties (pH, Eh and EC), solubilization and leaching tests, pH variation, sequential extraction and aquatic ecotoxicity were carried out. Besides the tailings analyzes, mineralogical characterization and pH measurements were also performed in samples from Perau (waste rock). The results showed mineralogical compositions predominantly carbonate in both tailing and Perau samples, as a reflection of the local lithology. This composition also reflected in the higher pH values, which ranged from 7.6 to 8.0. The tailing had fine particle size (predominance of silt, very fine and fine sand), and high concentrations of metals, especially Pb (average concentration of 5236.67 mg.kg-1) and Zn (7726.53 mg.kg-1). The metal Pb was above of the recommended value in ABNT NBR 10.004:2004 in leaching tests, solubilization tests and for all samples. This fact allowed the classification of the waste as Class I-Dangerous. In testing of pH variation, the Zn showed higher release in initial acidic condition (pH 3), releasing average 11.54 mg.L-1 after sixty minutes of test; although in others pH conditions have also been considerable release. In sequential extraction, the results found that all metals (except Cu) were associated with phase oxides/hydroxides, but the phases considered more mobile (exchangeable and carbonate) were also present in significant concentrations for all metals. The five samples were considered heterogeneous and two of them (RP5 and RP7) had greater mobility of Pb and Zn, besides a higher toxicity in ecotoxicity tests. In aquatic ecotoxicity test, the samples showed immobility of neonates organisms even in less concentrated dilutions (dilutions containing only 6.2 and 3.1% solubilized extract). In conclusion, the waste may be considered problematic for environment, because the high concentrations of toxic metals present in tailing can be released from it and migrate to the ecosystems, causing exposure to terrestrial and aquatic organisms.

Etude des compartiments d’hydrolyse et de méthanogenèse d’un procédé de méthanisation en voie épaisse multi-étapes phasé en température : Le procédé Arkométha / Study of the hydrolysis and methanogenic compartments of a high solid temperature phased anaerobic digester : The Arkométha ® process

Le procédé Arkométha® est un système de digestion anaérobie de rejets organiques fonctionnant en voie épaisse et comprenant plusieurs compartiments phasés en température. Ce travail s’est intéressé aux performances des différentes phases du procédé pour le traitement de résidus agricoles. Dans un premier temps, le compartiment d’hydrolyse a été étudié. Les paramètres les plus influents ont été identifiés (température et durée de l’hydrolyse) à l’aide d’un plan d’expérience. Ils ont ensuite été testés sur un pilote afin d’évaluer leur influence sur la solubilisation et la production de méthane ultérieure. Des tests ont alors été conduits à l’échelle laboratoire en mode continu au moyen de réacteurs en séries. Le compartiment méthanogène, constitué de deux à trois secteurs, a été étudié en mettant en évidence l’efficacité de chaque secteur. L’effet de la température a ici également été étudié, avec un impact important. Les deux compartiments ont été mis en série permettant l’étude du procédé complet. Ceci a permis de mettre en lumière certains risques liés à des combinaisons de température peu favorables. Par ailleurs, une comparaison avec un procédé mono-étape a été effectuée. Outre l’étude des performances de chaque compartiment et l’analyse de leur articulation, un résultat intéressant de ce travail réside dans l’affinement de la notion d’hydrolyse dans le cadre de la méthanisation en voie épaisse, communément confondue avec la solubilisation. En effet, nous avons montré que le gain apporté par l’utilisation d’un réacteur séquencé avec un compartiment dédié à l’hydrolyse ne réside pas dans l’amélioration de la solubilisation, mais dans l’augmentation conséquente de la cinétique de méthanisation dans les étapes suivantes. De façon intéressante, alors que la température joue un rôle primordial dans la solubilisation de la matière organique, elle est relativement peu influente pour l’amélioration de la cinétique de dégradation, où le temps de séjour devient prépondérant. / The Arkométha ® process is a system treating organic wastes in several sectors working at high solid concentration and being temperature phased. The present work focuses on the performances of the different phases of the process in treating agricultural wastes. In a first time, the hydrolysis sector has been studied. The most influent parameters have been identified (temperature and retention time), using design of experience. They have been tested afterwards at a pilot scale in order to assess the influence on solubilization and later methane production. Then, tests have been conducted at the lab scale in continuous mode using serial reactors. The methanogenic compartment, composed by two to three sectors has been studied highlighting the efficiency of each sector. The effect of temperature has also been studied, with a strong impact. The two compartments being used in series enabled the study of the complete process. This allowed highlighting some risks linked with certain temperatures combinations. On the other hand, a comparison with a single step reactor has been made. Beside the analysis of the relative performances of each compartment, a major result of this work lies in the distinction between hydrolysis and solubilization. Eventually, we showed that the gain brought by the hydrolysis compartment is not only related to solubilization of organic matter, but also in the increase in kinetics in the downwards sectors. Interestingly, while temperature has a major role in solubilization, it is of less importance in improving the degradation kinetics, where the retention time becomes of higher importance.

The oil sludge is a complex viscous mixture which is considered as a hazardous waste. It is then necessary to develop methods and technologies that optimize the cleaning process, oil extraction and applications in industry. Therefore, this study aimed to determine the composition of the oil sludge, to obtain and characterize microemulsion systems (MES), and to study their applications in the treatment of sludge. In this context, the Soxhlet extraction of crude oil sludge and aged sludge was carried out, and allowing to quantify the oil (43.9 % and 84.7 % - 13 ?API), water (38.7 % and 9.15 %) and solid (17.3 % and 6.15 %) contents, respectively. The residues were characterized using the techniques of X-ray fluorescence (XRF), Xray diffraction (XRD) and transmission Infrared (FT-IR). The XRF technique determined the presence of iron and sulfur in higher proportions, confirming by XRD the presence of the following minerals: Pyrite (FeS2), Pyrrhotite (FeS) and Magnetite (Fe3O4). The FT-IR showed the presence of heavy oil fractions. In parallel, twelve MES were prepared, combining the following constituents: two nonionic surfactants (Unitol L90 and Renex 110 - S), three cosurfactants (butanol, sec-butanol and isoamyl alcohol - C), three aqueous phase (tap water - ADT, acidic solution 6 % HCl, and saline solution - 3.5 % NaCl - AP) and an oil phase (kerosene - OP). From the obtained systems, a common point was chosen belonging to the microemulsion region (25 % [C+S] 5 % OP and AP 70 %), which was characterized at room temperature (25?C) by viscosity (Haake Rheometer Mars), particle diameter (Zeta Plus) and thermal stability. Mixtures with this composition were applied to oil sludge solubilization under agitation at a ratio of 1:4, by varying time and temperature. The efficiencies of solubilization were obtained excluding the solids, which ranged between 73.5 % and 95 %. Thus, two particular systems were selected for use in storage tanks, with efficiencies of oil sludge solubilization over 90 %, which proved the effectiveness of the MES. The factorial design delimited within the domain showed how the MES constituents affect the solubilization of aged oil sludge, as predictive models. Portanto, neste trabalho objetivou-se determinar a composi??o da borra de petr?leo, obter e caracterizar sistemas microemulsionados (SME), e estudar as suas aplica??es no tratamento da borra. Neste contexto, utilizou-se a extra??o Soxhlet da borra de petr?leo bruta e da borra envelhecida, permitindo quantificar o teor de ?leo (43,9 % e 84,7 % - 13 ?API), de ?gua (38,7 % e 9,15 %) e de s?lidos (17,3 % e 6,15 %), respectivamente. Os res?duos foram caracterizados atrav?s das t?cnicas de fluoresc?ncia de raios X (FRX), de difra??o de raios X (DRX) e Infravermelho de transmiss?o (IFT). O FRX determinou a presen?a de ferro e enxofre em maiores propor??es, comprovando-se pelo DRX a presen?a dos seguintes minerais: Pirita (FeS2), Pirrotita (FeS) e Magnetita (Fe3O4). O IFT mostrou a presen?a de fra??es pesadas de petr?leo. Em paralelo foram obtidos doze SME, combinando os seguintes constituintes: dois tensoativos n?o i?nicos (Unitol L90 e Renex NP110 - T), tr?s cotensoativos (butanol, sec-butanol e ?lcool isoam?lico - C), tr?s fases aquosas (?gua de torneira - ADT, solu??o ?cida HCl 6 % e solu??o salina NaCl 3,5 % - Fa) e uma fase ?leo (querosene - Fo). A partir da obten??o destes sistemas, foi escolhido um ponto em comum na regi?o de microemuls?o (25 % [C+T], 5 % Fo e 70 % Fa), os quais foram caracterizados, a temperatura ambiente (25?C), atrav?s da viscosidade (Re?metro de Haake Mars), di?metro de part?culas (Zeta Plus) e estabilidade t?rmica. Misturas com essa composi??o foram aplicadas na solubiliza??o da borra do petr?leo, sob agita??o, na propor??o de 1:4, variando tempo e temperatura. As efici?ncias de solubiliza??o foram obtidas desconsiderando o teor de s?lidos, situado em uma faixa de 73,5 % a 95 %. Assim, escolheram-se dois sistemas como destaque para aplica??o em tanques de armazenamento, com efici?ncias de solubiliza??o da borra de petr?leo acima de 90 %, comprovando-se assim a efic?cia do SME. Os planejamentos experimentais fatoriais delimitaram dentro do dom?nio a influ?ncia de constituintes dos SME na solubiliza??o da borra de petr?leo envelhecida, com modelos preditivos. Isolatings were performed from rhizosphere of vegetables and soil after harvest of cultivated species. The vast majority of the strains showed biochemical profiles identical to Pseudomonas (fluorescent group). While profiles identical to Burkholderia, Stenotrophomonas and Acinetobacter constituted other groups of representatives. Characterization of isolates took into account the capacity for synthesis of compounds possibly involved in mechanisms to stimulate plant growth, namely: biofilms, siderophores, acyl homoserine lactones (AHLs), indole-3-acetic acid (IAA) and indole related compounds, as well as solubilization of calcium phosphate. A total of 94 isolates were evaluated in the greenhouse for their ability to promote the growth of cabbage (Brassica oleracea var. acephala) in pots containing unsterilized loam soil. In addition, consortia were evaluated between components of the collection and rhizobia for the same purpose. In addition, it compared the effects of application of certain producing IAA isolates and the application of synthetic IAA on the growth of cabbage. Laboratory experiments showed that in vitro, 100% of the isolates formed biofilm, 71% synthesized siderophores, 31% produced AHLs, 95% produced IAA and 74% solubilized P. However, bioassays in the greenhouse showed that only 12% of the isolates induced significant responses to promote the growth of cabbage. Of these, a subgroup composed of 10 strains of rhizobacteria given increments in growth parameters of both the biomass and the root system of vegetables. Biofilms, siderophores, IAA and solubilization of P were common characteristics of the subgroup. No synergistic effect was observed among isolates in consortia, although it was found that the two strains of rhizobia consortium promoted the growth of inoculated plants. The isolates R1132 and S311 were higher than the application of synthetic IAA. Os experimentos de laborat?rio indicaram que, in vitro, 100% dos isolados formaram biofilme, 71% sintetizaram sider?foros, 31% produziram AHLs, 95% produziram AIA e 74% solubilizaram P. No entanto, os bioensaios na casa de vegeta??o evidenciaram que apenas 11% dos isolados bacterianos induziam respostas significativas de promo??o do crescimento da couve. Desses, um subgrupo formado por 10 estirpes de rizobact?rias conferiu incrementos nos par?metros de crescimento, tanto da biomassa a?rea quanto do sistema radicular da hortali?a. Biofilmes, sider?foros, AIA e solubiliza??o do P foram caracter?sticas comuns do subgrupo. N?o foi observado efeito sin?rgico entre os isolados em cons?rcios, embora tenha sido verificado que as duas estirpes de riz?bios consorciadas promoveram o crescimento das plantas inoculadas. Identification of candidate plant growth promoting endophytes from Echium plantagineum roots

Magister Scientiae - MSc (Biotechnology) / The yearly increase of global population will result in a greater demand for crop production, but with the climates changes and a lack of available agricultural land it will become increasingly more difficult to provide sufficient crops to feed everyone adequately. Application of the PGPE has proven over the past researches to be able enhance growth of plants via various growth promoting mechanisms. To identify suitable growth promoting bacteria candidate, E. plantagineum plant was used to isolate endophytes from the root after surface sterilization. The isolates bacteria were used to inoculate Brassica napus L seeds. The effects of isolate's ability to promote growth were evaluated based on the certain growth parameters after 42 days in the green house. Isolate CP5 produced highest results in all growth parameter. Isolates CP5 was selected as potential candidate as significant improvement was shown by this isolate. This isolate was tested for the ability to produce ACC deaminase, solubilize phosphate, synthesize IAA and siderophore production. Furthermore isolate CP5 growth promotion abilities was tested on Brassica napus L under antimony stress. / 2021-08-31

O objetivo deste trabalho foi avaliar in vitro as diferentes bactérias isoladas destas espécies florestais que possam ter efeito benéfico no crescimento e desenvolvimento inicial das plantas e caracterizálas pelo sequenciamento do gene 16S rRNa e perfil eletroforético do gene BOX-A 1R. As bactérias foram isoladas da rizosfera e da raiz de mudas de cada espécie, em meio de cultura NFb. No total, 29 isolados foram cultivados em meio de cultura DYGS enriquecido com triptofano. A estimativa colorimétrica do Ácido Indolacético (AIA) foi feita no espectrofotômetro, utilizando o sobrenadante da cultura de células e solução de Salkowski, com resultado positivo para o isolado Pn 6 Sphingobium chlorophenolicum com 61,69 μg.mL-1. Os isolados também foram testados para a eficiência de solubilização de fosfato de cálcio que foi realizado e contabilizado até o 15° dia após a inoculação em placas de Petri com meio de cultura NBRIP sólido, onde 27 isolados apresentaram resultados positivos. A caracterização genética permitiu a diferenciação em gêneros e também em espécies de um mesmo gênero. Os isolados que deram resultados positivos para AIA e solubilização de fosfato... / The biodiversity conservation implies cultivating native forest species that can be used in reforestation projects, recovery of degraded areas as well as getting others like wood and medicinal products. The plant growth promoting rhizobacteria (PGPR) use is being studied in cultures of economic importance. In this work four native forest species, no nodulating leguminous had been chosen: Pterogyne nitens, Albizia hasslerii, Copaifera langsdorffii, and Stryphnodendron adstringens. The aim of this work was the in vitro evaluation of the different isolates from those forest species with beneficial effects in growth and initial development of the plants and characterizes them by 16S rRNA gene sequencing and BOX-A 1R. The bacteria had been isolated from the rhizosphere and from the roots (endophytic) of each species seedlings on NFb growth media. A total of 29 isolates had been cultivated in growth media DYGS enriched with tryptophan. The indolacetic acid (IAA) production was estimated by colorimetric test in the spectrophotometer, using the supernatant of the cells culture and Salkowski´s reagent, with positive result for the isolated Pn 6 Sphingobium chlorophenolicum with 61,69 μg.mL-1. Isolates had been also tested for the efficiency of P- solubilizing that was carried through and measured until the 15 º day after the inoculation in Petri plates with solid growth media NBRIP, where 27 isolates had presented positive results. The genetic characterization allows us to differentiate by the genus and species from the same genus. The isolates that had positive results for IAA and P-solubilizing had not matched between itself, what suggests nursery tests with combinations between them to evaluate the isolate efficiency as a possible PGPRs.

Nevertheless, the chemical characteristics of rock phosphates (RP), particularly the level of toxic elements, can affect the efficiency of microbial solubilization. Fluoride (F-) released from fluorapatites has been shown to significantly inhibit P solubilization by Aspergillus niger, stimulating the search for alternatives to overcome F- toxicity. Thus, the aim of this study was to select A. niger mutants efficient at P solubilization in the presence of F-. Three selected mutants were also characterized as to their P solubilization mechanisms and the solubilization potential of different P sources. Twenty-nine mutants were obtained that presented changes in their phosphate solubilization activity in comparison to the wild type (WT). The mutant FS1-331 showed higher solubilization of Araxá RP, while FS1-555 promoted higher soluble P when grown in media with calcium phosphate supplemented with F- and in those with pure P sources. These mutants also showed higher tolerance to F- than the WT and displayed changes in the production of organic acids. The higher production of oxalic acid by FS1-331 and FS1-555 correlated with their improved capacity of Araxá RP solubilization. A mutant with decreased P solubilization capacity showed lower production of organic acids, corroborating the importance of these compounds for RP solubilization. FS1-331 was more efficient at solubilizing Araxá, Catalão, and Patos de Minas RPs than the WT and FS1-555. The variation in P solubilization capacity of the mutants obtained in this work may help clarify the RP solubilization mechanisms by A. niger. Moreover, the mutants with better performance selected in this work show potential for use in microbial RP solubilization systems with P sources rich in fluoride. / O uso dos micro-organismos solubilizadores de fosfato (P) é uma alternativa para o uso sustentável do P tendo em vista a diminuição das reservas fosfáticas de alta qualidade. As características químicas dos fosfatos de rocha (FR), em especial o nível de elementos tóxicos, podem influenciar a eficiência de solubilização microbiana. O fluoreto (F-) liberado de fluorapatitas inibi significativamente o processo de solubilização por Aspergillus niger FS1. Assim, o objetivo deste estudo foi selecionar mutantes de A. niger eficientes na solubilização de fosfato na presença de F-. Três mutantes selecionados foram também caracterizados quanto aos mecanismos de solubilização de fosfato e ao potencial de solubilização de diferentes fontes de P. Vinte e nove mutantes com alterações na solubilização de fosfato foram obtidos. O mutante FS1-331 demonstrou maior potencial de solubilização de fosfato de Araxá, enquanto que o mutante FS1-555 aumentou o P solubilizado em meio de cultura com fosfato de cálcio suplementado com F- e naqueles com fontes sintéticas de P. Os mutantes avaliados mostraram maior tolerância ao F- do que o tipo selavagem e tiveram a produção de ácidos orgânicos alterada. A maior produção de ácido oxálico pelos mutantes FS1-331 e FS1-555 relacionou-se com o melhor desempenho dessas estiprpes na solubilização de fosfato de Araxá. Um mutante com solubilização de P diminuída (FS1-375) apresentou menor produção de ácidos orgânicos quando comparado ao tipo selvagem, corroborando a importância desses compostos para o processo de solubilização de FRs. O mutante FS1-331 foi o mais eficiente na solubilização dos FRs de Araxá, Catalão e Patos de Minas. A variação na solubilização de P das estirpes fúngicas estudadas pode contribuir para a melhor compreensão do processo de solubilização de fosfatos por A. niger. Além disso, os mutantes mais eficientes obtidos apresentam alto potencial de aplicação em sistemas microbianos de solubilização de fosfatos com fontes de P ricas em fluoreto.
