Des agents chimiosensibilisants pour lutter contre la résistance aux antibiotiques chez les bactéries Gram-négatif : criblage et caractérisation / Chemosensitizers against Gram negative bacteria : screening & characterization

Les bactéries Gram-négatif possèdent une structure d'enveloppe, ainsi que des pompes d'efflux (i.e., systèmes de transport actif permettant de détoxifier la cellule bactérienne) qui les rendent naturellement résistantes aux antibiotiques. Ces deux caractéristiques constituent de véritables barrières qui s'opposent à l'accumulation d'une grande variété d'antibiotiques près de leur cible, à l'intérieur de la bactérie. La perturbation des mécanismes s’opposant à l’accumulation d’antibiotiques par des agents chimiosensibilisants représente une stratégie prometteuse.L’objectif de cette thèse était de mieux comprendre les mécanismes d'inhibition de la résistance qui s’oppose à l’accumulation d’antibiotiques chez les bactéries Gram- négatif.Dans un premier temps l'activité de divers agents chimiosensibilisants synthétiques a été caractérisée. Trois dérivés ont été identifiés pour augmenter significativement l'activité synergique avec les antibiotiques, préalablement observée avec le géraniol. Ces dérivés ont montré une activité inhibitrice des pompes d'efflux ou perméabilisatrice de la membrane externe, pouvant être à l'origine des synergies observées.Dans un second temps, une méthode de criblage a été mise au point, en permettant la détection spécifique des agents chimiosensibilisants tout en décrivant leur mécanisme d'action.Ces travaux de thèse ont participé à proposer une solution thérapeutique brevetée au stade pré-clinique. Ils ont en outre permis de mettre en place des outils originaux pour identifier de nouveaux chimiosensibilisants, mais aussi pour mieux comprendre comment perturber les barrières s'opposant à l'accumulation d'antibiotiques. / Gram-negative bacteria are naturally resistant to many classes of antibiotics thanks to their ability to control the accumulation of drugs. Decreasing membrane barrier permeability and producing efflux pumps that expel drugs outside bacteria, represent the prevalent mechanisms of this resistance. One of the most promising solutions consists in restoring antibiotic activity by targeting such barriers to accumulation, with chemosensitizers.The purpose of my PhD was to better understand the inhibition of resistance that opposes the accumulation of antibiotics in Gram-negative bacteria.In the first stage of the study, the activity of various synthetic chemosensitizers has been characterized. Three compounds were identified to significantly increase the synergistic activity with antibiotics, that was previously observed with geraniol. These derivatives showed an efflux pump inhibition or an outer membrane permeabilization effect, that could be related to the observed synergy.In the second stage of the study, a screening method has been developed for the specific detection of chemosensitizers, while describing their mechanism of action.This work participated in proposing a patented therapeutic solution in the preclinical stage. This study has led to new tools to identify novel chemosensitizers, but also to better understand how to impair the barriers opposing the accumulation of antibiotics.

A Theoretical Study of the Synergy and Lazy Annotation Algorithms

Given a program with assertions, the assertion checking problem is to tell whether there is an execution of the program that violates one of the assertions. One approach to this problem is to explore different paths towards assertion violations, and to learn “blocking” conditions whenever a path is blocked from reaching the violations. The Synergy algorithm of Gulavani et al. [FSE 2006] and the Lazy Annotation algorithm of McMillan [CAV2010] are two recent algorithms that follow this approach to assertion checking. Each technique has its own advantages. Synergy uses concrete tests which are very cheap as compared to theorem prover calls. The tests also help by giving us the place to perform the refinement (called the frontier) for an abstraction which is too coarse. Synergy uses partition refinement while maintaining its abstraction. The Lazy Annotation algorithm basically partitions each location in to regions that are safe and unsafe. The safe regions are those from which we cannot reach the error states, and the unsafe regions are the remaining ones. The annotations that this algorithm maintains correspond to the safe regions. The advantage that annotations have over partition refinement is that annotations can recover from irrelevant predicates used for annotating, where as once a partition is refined with an irrelevant predicate, it cannot recover from it. In this work, we make a theoretical study of the algorithms mentioned above. The aim of the study is to answer questions like: Is one algorithm provably better than the other, in terms of the best-case execution (counting the number of refinement steps) on input programs? Is the termination behavior of one always better than the other? We show that the Synergy and Lazy Annotation algorithms are incomparable, i.e., neither of them is provably better than the other, in terms of their best-case execution times. We also show how we can view the two algorithms on a common ground, in the sense that we show how to translate a snapshot of one algorithm into a snapshot of the other. This allows us to import the heuristics of one algorithm into the other, and there by propose new and potentially improved versions of these algorithms. By viewing them o n a common ground, we are also able to view the final proofs generated by the algorithms in either representation. We go on to study the proposed new versions of the Synergy and Lazy Annotation, comparing their best-case running times and their termination behaviour. We show that the following pairs of algorithms are incomparable: Mod-Syn (Lazy Annotation-style refinement imported into Synergy) and Synergy, Mod-Syn and Lazy Annotation, Synergy and SEAL(Synergy heuristics imported into Lazy Annotation). We show that the SEAL algorithm always performs better than the Lazy Annotation algorithm.

This diploma thesis analysis the evolution of Helika company after entering the foreign partner. The evolution has been involved by the depression and its impacts on the industry. Through financial analysis is compared with the development of the business sector and its competitors across the market. The conclusion of the thesis is evaluation whether the decision of entering the foreign partner was suitable act and whether the company achieved to required effects.

Exploring the Utilization of Startle as a Therapy Tool in Individuals with Stroke

abstract: Stroke is a debilitating disorder and 75% of individuals with stroke (iwS) have upper extremity deficits. IwS are prescribed therapies to enhance upper-extremity mobility, but current most effective therapies have minimum requirements that the individuals with severe impairment do not meet. Thus, there is a need to enhance the therapies. Recent studies have shown that StartReact -the involuntary release of a planned movement, triggered by a startling stimulus (e.g., loud sound)- elicits faster and larger muscle activation in iwS compared to voluntary-initiated movement. However, StartReact has been only cursorily studied to date and there are some gaps in the StartReact knowledge. Previous studies have only evaluated StartReact on single-jointed movements in iwS, ignoring more functional tasks. IwS usually have abnormal flexor activity during extension tasks and abnormal muscle synergy especially during multi-jointed tasks; therefore, it is unknown 1) if more complex multi-jointed reach movements are susceptible to StartReact, and 2) if StartReact multi-jointed movements will be enhanced in the same way as single-jointed movements in iwS. In addition, previous studies showed that individuals with severe stroke, especially those with higher spasticity, experienced higher abnormal flexor muscle activation during StartReact trials. However, there is no study evaluating the impact of this elevated abnormal flexor activity on movement, muscle activation and muscle synergy alterations during voluntary-initiated movements after exposure to StartReact. This dissertation evaluates StartReact and the voluntary trials before and after exposure to StartReact during a point-to-point multi-jointed reach task to three different targets covering a large workspace. The results show that multi-jointed reach tasks are susceptible to StartReact in iwS and the distance, muscle and movement onset speed, and muscle activations percentages and amplitude increase during StartReact trials. In addition, the distance, accuracy, muscle and movement onsets speeds, and muscle synergy similarity indices to the norm synergies increase during the voluntary-initiated trials after exposure to StartReact. Overall, this dissertation shows that exposure to StartReact did not impair voluntary-initiated movement and muscle synergy, but even improved them. The aim of this thesis is to evaluate the previous and current situation of the K. s.r.o. company market position, including a comparison of the strategic administration of the former and new management. A complete strategic analysis was performed in the analysed company - i.e. external and internal analysis of the company including the evaluation of risks and results of the financial analysis. Thanks to the strategic analysis of the K. company, important information was gained, necessary for the subsequent evaluation of the previously and currently implemented company strategy. The result of the thesis is an evaluation of conclusions and determination of recommendations which will lead to a general improvement of the current position of the company and to maintaining its competitive strength.

Para este fin, se analizan las variables de estrategia gerencial, creación de valor, función comercial, capital humano, aspectos operativos y tecnológicos y gobierno corporativo. Finalmente, a la luz de la información bibliográfica revisada, y con la finalidad de que pueda servir de apoyo a las empresas para la toma de decisiones en relación a los procesos de F&A, se recomienda llevar a cabo un estudio empírico que permita cuantificar los efectos sobre las variables empresariales e identificar cuáles son las condiciones que favorecen el éxito de las F&A. / This paper presents relevant research that discusses the effects that Mergers and Acquisitions (M&A) have on business variables, with particular emphasis on the construction and destruction of value. Textile Hybrids explores knitted textiles by challenging properties of elasticity and flexibility through yarn composition, technical construction and combinations with wood. This study is placed in the field of textile spatial design and suggests experimental ways to explore three-dimensionality in a knitted textile by changing its properties through material synergies. The outcome is a three piece series of modular three-dimensional, standalone textile objects. The construction, assembly and flexibility of each piece make it possible to separate all components for reassembly, recycling or reusing, suggesting further research possibilities into more tangible contexts within textile spatial design, architecture, furniture design and product design. This study is derived from an interest to explore different ways a textile can exist on its own in a spatial context such as the home, without solely being the material covering a load-bearing framework i.e a couch or a chair.

An Exploratory Study on the Post-Acquisition Process of Technological Acquisition – a case study of HMS

Although M&amp;A’s have the greatest probability of failure in organizations, the major reasons for this outcome is the integration process. M&amp;As have been a popular strategy for accessing new markets, introducing new products, expanding their knowledge base, and enhancing competitive advantage. In this study, we explore a case of technology acquisition, and we propose a theoretical framework based on literature that identifies phases in post-acquisition process. The process involves three stages: knowledge absorption, operational phase and commercial phase that is explored and evaluated with the empirical data of the case study. Then, thematic analysis was utilized in this study to identify common themes related to the role of acquisition. The related functions and benefits were grouped under acquisition roles. Since the case study was a successful acquisition, it was easy to figure out the roles and dimensions of integration from it. Both the methods contribute in addressing, the necessary phases that need to be organized with integration and deriving different functionalities to achieve common goals. Finally, we present a discussion and bring out the relationships that emerged from this study from different themes and have been mapped to stages in the post-acquisition process, resulting in outcomes from each role. Thus, this study puts an emphasis on the range of factors that create value from successful technological acquisition and conclude as post-acquisition process with integration elements is the initial pivotal position for the consequences.
