Morfologia e morfometria das células do endotélio da córnea de equinos utilizando a microscopia eletrônica de varredura

O conhecimento da morfologia endotelial nas diferentes regiões da córnea é de suma importância para avaliação de endotélios corneanos saudáveis e doentes e de suas respostas ao uso de medicações. O objetivo do presente estudo foi descrever a morfologia do endotélio nas regiões central e periférica superior da córnea saudável de equinos, medir a área média das células pentagonais, hexagonais e heptagonais presentes nas regiões avaliadas, calcular o polimegatismo de cada tipo celular e correlacionar estes parâmetros entre os diferentes formatos celulares. Foram estudados dez equinos, machos ou fêmeas, de diferentes idades, provenientes de um abatedouro licenciado. Imagens da superfície posterior do endotélio da córnea foram obtidas com uso de microscopia eletrônica de varredura. A morfologia do endotélio de diferentes regiões da córnea foi avaliada. Além disso, foi correlacionada a variabilidade do tamanho celular médio com a morfologia endotelial. A análise estatística foi conduzida usando o teste de análise de variância (ANOVA) seguido do teste de Tukey (Post-Hoc), com nível de significância de 5%. As amostras avaliadas foram compostas em sua maioria por células hexagonais (60,5%), pentagonais (21,4%) e heptagonais (16,9%), células com quatro, oito ou nove lados compuseram 1,3%. A área celular média das células pentagonais foi 203,58 μm², das hexagonais foi 223,84 μm² e nas heptagonais foi 270,54 μm². O polimegatismo encontrado foi de 16% nas células pentagonais e hexagonais e de 20% nas heptagonais. A morfologia das células endoteliais de equinos saudáveis não diferiu entre as regiões central e periférica superior da córnea, sugerindo que a região central é representativa da região periférica. As células com sete lados apresentaram polimegatismo maior em relação às células de seis e de cinco lados. / The knowledge of the endothelial morphology in the different regions of the cornea is important for the evaluation of healthy and sick corneal endothelium and its response to drugs. In order to describe the endothelial morphology in the central and superior peripheral regions of the equines’ cornea, evaluate area from pentagonal, hexagonal and heptagonal cells present in the evaluated regions, calculate the polimegathism and correlate these parameters, two healthy corneas were collected of ten equine, male or female, of different ages. Images of the posterior surface of the corneal endothelium were taken with scanning electron microscope. The endothelial morphology was studied in the different regions of the equines’ cornea. In addition, the polimegathism and morphology was also correlated. A statistical analysis was conducted using the Analysis of Variance (ANOVA) followed by Tukey's Test (Post-Hoc), with a 5% level of significance. In the central region, the endothelium consisted of 58.8% hexagonal cells, 22.6% pentagon, 17.1% heptagonal and 1.4% cells with either four, eight or nine sides. In the superior peripheral region, 62.1% of the cells were hexagonal, 20.2% pentagon, 16.6% heptagonal and 1.1% cells with four or eight sides. The average cell area of the pentagonal cells was 210.77 ± 31.53 μm² in the central region and 196.39 ± 34.35 μm² in the superior peripheral region; in hexagonal cells, the average cell area found in the central region was 216.12 ± 37.09 μm² and 231.56 ± 34.95 μm² in the superior peripheral region; and in cells with seven sides, the average cell area was 261.76 ± 55.29 μm² in the central region and in the superior peripheral region was found 279.32 ± 52.37μm². When not considering the corneal regions, the polymegathism found was 16% in the pentagonal and hexagonal cells and 20% in the heptagonal cells. The morphology results obtained did not differ between the central and peripheral regions of the cornea, suggesting that the central region is representative of the peripheral region. The highest coefficient of variation was seen in cells with seven sides.

Diversas substâncias são utilizadas para biomodificação radicular com objetivo de remover tecido mineralizado e expor as fibras colágenas da matriz dentinária, favorecendo a quimiotaxia e migração de células do ligamento periodontal. Este estudo avaliou diferentes concentrações, modos e tempos de aplicação de ácido cítrico na biomodificação radicular. Dentes humanos tiveram duas áreas de 3x2 mm delimitadas apicalmente a junção cemento-esmalte com a utilização de fresa cilíndrica, instrumentadas com 50 movimentos de raspagem utilizando curetas de Gracey 5/6 e em seguida, espécimes foram obtidos e divididos em 6 grupos (45 amostras/grupo): soro fisiológico (controle), ácido cítrico (0.5%, 1%, 2%, 15% e 25%), com tempos de 1, 2 ou 3 minutos para cada grupo, nos modos de aplicação: a) aplicação passiva (bolinha de algodão); b) fricção suave (pincel); c) fricção vigorosa (bolinha de algodão), com renovação da solução a cada 30 segundos. As amostras foram submetidas à desidratação em concentrações crescentes de álcool etílico e HMDS, sendo em seguidas metalizadas e levadas para observação em microscopia eletrônica de varredura. Um examinador treinado, calibrado e cego avaliou as fotomicrografias obtidas utilizando o Índice de Remoção de Smear Layer proposto por Sampaio em 1999, atribuindo escores de 1 a 8. A análise estatística foi realizada comparando-se as proporções dos escores obtidos, e mostrou que a concentração de 25 % aplicada por fricção suave no tempo de 3 minutos é recomendada para utilização do ácido cítrico na biomodificação radicular. / Abstract: The root scaling produces a smear layer which contains microorganisms and toxins that obliterates the dentinal tubules and covers the root surface, affecting the periodontal healing process. Different substances have been used to promote root biomodification with the purpose of creating the demineralization of the root surface and the collagen fibrils' exposure. The aim of this study was to evaluate different concentrations, modes and application times of citric acid in the root biomodification. Human teeth had an area of 3x2mm delimited from the apical to the cement-enamel junction. This area was scaled, with 50 strokes by hand instruments, to produce smear layer. The specimens were divided in 6 groups. (45 samples/group): physiologic solution (control) and citric acid (0.5%, 1%, 2%, 15% and 25%). The application time was 1', 2'and 3' to each group. The application modes were: a) topic application, with cotton pellets; b) brushing, with a soft brush; c) burnishing, with cotton pellets. In all modes, the solution was renewed every 30". The samples were submitted to ethylic alcohol and HMDS dehydration and then they were evaluated using the scanning electron microscopy. A blind and trained examiner evaluated photomicrographs based on Sampaio's index (1999). Data were statistically analyzed by means of a binomial test (p≤0.05). The statistical analysis showed that the best results were achieved with the 25% concentration applicated by brushing for 3 minutes. Crack growth behavior in an aluminum alloy under very low stress amplitudes

The near-threshold behavior of long cracks is studied in this paper using precracked flat dogbone specimens of a commercial aluminum alloy in peak-aged and overaged conditions. After introducing the initial crack in compression precracking, the crack was propagated approximately with the constant range of the stress intensity factor at values just above or below the corresponding threshold values. It was found that there were two major mechanisms which kept the crack from continuous extension. First, the crack front was pinned by primary precipitates. This effect was rather pronounced and lead to significant kinking in the crack front and ductile ridges on the fracture surface. The second mechanism was shear-controlled crack extension of very long cracks with plastic zones ahead of the crack tip, very similar to stage-I small cracks. Interaction with primary precipitates deflected the shear-controlled cracks but did not change the crack extension mode.

Celulózová nanovlákna a materiály v aerosolu / Cellulose Nanofibers and Aerogel Materials

Tato diplomová práce se zabývá přípravou nových kompozitů na bázi mikrofibrilované celulozy a multi-walls carbon nanotubes (MFC/MWCNTs). Tři různé metody byly použity pro modifikaci MWCNTs. Byl zkoumán vliv použitých modifikací na kvalitu MWCNTs suspense. Dále byly připraveny kompozity obsahující MFC a MWCNTs a vliv MWCNTs na mechanické a elektrické vlastnosti a morfologii byl zkoumán. MWCNTs suspense byly charakterizovány za použití UV-VIS spektrofotometrie, Rastrovacího elektronového mikroskopu (SEM) a Termogravimetrické analýzy (TGA). Elektrické vlastnosti byly měřeny pomocí Keithly Electrometer/High Resistivity Meter. Mechanické vlastnosti v tahu byly měřeny za použití Miniature Materials Tester a pro studium morfologie byl použit SEM. Žádný významný vliv MWCNTs na mechanické vlastnosti nebyl pozorován. Hodnoty získané z měření rezistivity vykazují typický perkolační trend a výrazného snížení rezistivity bylo dosaženo přidáním 1 – 2 wt.% MWCNTs. Nicméně hodnoty se velice liší pro horní a spodní stranu téhož vzorku. Ze získaných výsledků vyplývá, že klíčovým krokem ke zdokonalení vlastností materiálu jsou dobrá suspense MWCNTs a také dosažení vzájemné kompatibility obou složek.

Nanoscale Electronic Properties in GaN Based Structures for Power Electronics Using Electron Microscopy

abstract: The availability of bulk gallium nitride (GaN) substrates has generated great interest in the development of vertical GaN-on-GaN power devices. The vertical devices made of GaN have not been able to reach their true potential due to material growth related issues. Power devices typically have patterned p-n, and p-i junctions in lateral, and vertical direction relative to the substrate. Identifying the variations from the intended layer design is crucial for failure analysis of the devices. A most commonly used dopant profiling technique, secondary ion mass spectroscopy (SIMS), does not have the spatial resolution to identify the dopant distribution in patterned devices. The possibility of quantitative dopant profiling at a sub-micron scale for GaN in a scanning electron microscope (SEM) is discussed. The total electron yield in an SEM is shown to be a function of dopant concentration which can potentially be used for quantitative dopant profiling. Etch-and-regrowth is a commonly employed strategy to generate the desired patterned p-n and p-i junctions. The devices involving etch-and-regrowth have poor performance characteristics like high leakage currents, and lower breakdown voltages. This is due to damage induced by the dry etching process, and the nature of the regrowth interface, which is important to understand in order to address the key issue of leakage currents in etched and regrown devices. Electron holography is used for electrostatic potential profiling across the regrowth interfaces to identify the charges introduced by the etching process. Comparing the pitting corrosion behavior of prominent Zr-based bulk metallic glasses

Five well-known Zr-based alloys of the systems Zr–Cu–Al–(Ni–Nb, Ni–Ti, Ag) (Cu 5 15.4–36 at.%) with the highest glass-forming ability were comparatively analyzed regarding their pitting corrosion resistance and repassivation ability in a chloride-containing solution. Potentiodynamic polarization measurements were conducted in the neutral 0.01 M Na₂SO₄ 1 0.1 M NaCl electrolyte and local corrosion damages were subsequently investigated with high resolution scanning electron microscopy (HR-SEM) coupled with energy dispersive x-ray spectroscopy (EDX). Both pitting and repassivation potential correlate with the Cu concentration, i.e., those potentials decrease with increasing Cu content. Pit morphology is not composition dependent: while initially hemispherical pits then develop an irregular shape and a porous rim. Corrosion products are rich in Cu, O, and often Cl species. A combination of low Cu and high Nb or Ti contents is most beneficial for a high pitting resistance of Zr-based bulk metallic glasses. The bulk glassy Zr₅₇Cu₁₅.₄Al₁₀Ni₁₂.₆Nb₅ (Vit 106) and Zr₅₂.₅Cu₁₇.₉Al₁₀Ni₁₄.₆Ti₅ (Vit 105) alloys exhibit the highest pitting resistance.

IN SITU MORPHOLOGICAL AND STRUCTURAL STUDY OF HIGH CAPACITY ANODE MATERIALS FOR LITHIUM-ION BATTERIES

Lithium-ion batteries(LIBs) have dominated the energy storage market in the past two decades. The high specific energy, low self-discharge, relatively high power and low maintenance of LIBs enabled the revolution of electronic devices and electric vehicle industry, changed the communication and transportation styles of the modern world. Although the specific energy of LIBs has increased significantly since first commercialized in 1991, it has reached a bottleneck with current electrode materials. To meet the increasing market demand, it is necessary to develop high capacity electrode materials.<div><br></div><div>Current commercial anode material for LIB is graphite which has a specific capacity of 372 mAh g-1. Other group IV elements (silicon (Si), germanium (Ge), tin (Sn)) have much higher capacities. However, group IV elements have large volume change during lithiation/delithiation, leading to pulverization of active materials and disconnection between electrode particles and current collector, resulting in fast capacity fading. To address this issue, it is essential to understand the microstructural evolution of Si, Ge and Sn during cycling.<br></div><div><br></div><div>This dissertation is mainly focused on the morphological and structural evolution of Sn and Ge based materials. In this dissertation, anin situ focused ion beam-scanning electron microscopy (FIB-SEM) method is developed to investigate the microstructuralevolution of a single electrode particle and correlate with its electrochemical performance. This method is applied toall projects. The first project is to investigate the microstructural evolution of a Sn particle during cycling. Surface structures of Sn particles are monitored and correlated with different states of charge. The second project is to investigate the morphological evolution of Ge particles at different conditions. Different structures (nanopores, cracks, intact surface) appear at different cycling rates. The third project is to study selenium doped Ge (GeSe) anodes. GeSe and Ge particles are tested at the same condition. Se doping forms Li-Ge-Se network, provides fast Li transport and buffers volume change. The fourth project is to study the reaction front of Ge particle during lithiation. Micron-sized Ge particles have two reaction fronts and a wedge shape reaction interface, which is different from the well-known core-shell mode. The fifth project is to investigate antimony (Sb)-coated porous Ge particles. The Sb coating suppresses electrolyte decomposition and porous structure alleviates volume change. The results in this dissertation reveal fundamental information about the reaction mechanism of Sn and Ge anode. The results also show the effects of doping, porous structuring and surface coating of anode materials.</div>

Vascular Corrosion Casting: Review of Advantages and Limitations in the Application of Some Simple Quantitative Methods

Vascular corrosion casting has been used for about 40 years to produce replicas of normal and abnormal vasculature and microvasculature of various tissues and organs that could be viewed at the ultrastructural level. In combination with scanning electron microscopy (SEM), the primary application of corrosion casting has been to describe the morphology and anatomical distribution of blood vessels in these tissues. However, such replicas should also contain quantitative information about that vasculature. This report summarizes some simple quantitative applications of vascular corrosion casting. Casts were prepared by infusing Mercox resin or diluted Mercox resin into the vasculature. Surrounding tissues were removed with KOH, hot water, and formic acid, and the resulting dried casts were observed with routine SEM. The orientation, size, and frequency of vascular endothelial cells were determined from endothelial nuclear imprints on various cast surfaces. Vascular volumes of heart, lung, and avian salt gland were calculated using tissue and resin densities, and weights. Changes in vascular volume and functional capillary density in an experimentally induced emphysema model were estimated from confocal images of casts. Clearly, corrosion casts lend themselves to quantitative analysis. However, because blood vessels differ in their compliances, in their responses to the toxicity of casting resins, and in their response to varying conditions of corrosion casting procedures, it is prudent to use care in interpreting this quantitative data. Some of the applications and limitations of quantitative methodology with corrosion casts are reviewed here.

Studium beta fáze v Al-Mg-Si slitinách pomocí nekonvenčních metod elektronové mikroskopie / Study of beta phase in Al-Mg-Si alloys by means of unconventional methods of electron microscopy

Aluminium Al-Mg-Si alloys are the most commonly used in automotive and construction industry. Hexagonal ’-phase is one of the metastable phases occured in this type of alloys. Unlike classic square -phase, this ’-phase is characterized by different crystalographic orientation to the matrix and shape. Standard method used for identification of aluminium alloys is scanning electron microscopy (SEM), because of its quickness and efficiency, but in case of very thin or damaged structures (as a result of metallographic process) it’s insufficient. Scanning low energy electron microscopy (SLEEM) can be appropriate for identification of mentioned precipitates due to its physical principles resulting in many advantages compared to SEM. So the most important benefits are interaction volume reduction (which leads to improvement of surface sensitivity), increase of material contrast (ability to change matrix / precipitates contrast) as well as crystalographic contrast.

The effect of netarsudil on pore densities of Schlemm's canal inner wall endothelium in human eyes

BACKGROUND: Netarsudil, a Rho kinase and norepinephrine transport (NET) inhibitor, is a new FDA approved drug used for decreasing raised intraocular pressure (IOP) in ocular hypertensive and primary open-angle glaucoma (POAG) patients. Previous studies reported that netarsudil increased outflow facility and lowered IOP by increasing active outflow areas around the circumference of the eye and dilating the episcleral veins (ESV; Kiel and Kopczynski, 2015; Ren et al., 2016). However, the mechanisms by which netarsudil increases outflow facility have not yet been fully elucidated. Moreover, the effects of netarsudil on the inner wall (IW) endothelium I-pores and B-pores of the Schlemm’s canal (SC) have also not been investigated yet. AIM: The goal was to determine if netarsudil-treatment increased the effective filtration areas (EFA) by increasing pore density in both high- and non-flow type areas, compared to untreated control eyes. METHODS: In this study, the effects of netarsudil on the pore densities on IW of SC were investigated by serial block-face scanning electron microscopy (SBF-SEM). Two pairs of eyes were perfused with green fluorescent tracers in order to determine the outflow pattern prior to treatment. Then, one eye of each pair was perfused with netarsudil, while the fellow eye of each pair was perfused with vehicle solution. All eyes were then perfused with red fluorescent tracers in order to determine the outflow pattern once they were treated with netarsudil. Both pairs of eyes were perfused and fixed at 15 mmHg. Global imaging was performed for all eyes to visualize high- and non- flow areas in the trabecular meshwork (TM) and ESV’s. A SBF-SEM was used to image eight wedges of tissue including the IW of SC and TM (high- and non-flow areas from four eyes) for a total of 16,378 images. The study analyzed the percentage of pore-types (GV-associated I-pores, Non-GV associated I-pores, B-pores), the median pore spans, the GV-associated I-pore locations, and the pore densities (per IW nuclei and IW area) between the equivalent control and netarsudil-treated flow areas. RESULTS: In global images, an increase in high-flow areas were observed in netarsudil-treated eyes due to recruitment from low-flow and non-flow areas. A greater percentage of GV-associated I-pores, B-pores, and total pores were found in high-flow in contrast to non-flow areas in both control and netarsudil-treated eyes (all P ≤ 0.05). However, the percentage of GV-associated I-pores in non-flow areas were significantly greater in treated compared to control eyes (P ≤ 0.05). Qualitative observations from two pairs of eyes showed a trend of greater I-pore, B-pore, and total pore density/per IW nucleus and density/per IW surface area in high-flow in contrast to non-flow areas for both treated and control eyes. No difference in I-pore, B-pore, and total pore density/per IW nucleus and density /per IW surface area were observed in equivalent flow-type areas when comparing control and netarsudil-treated eyes. In addition, there was a significant greater percentage of I-pores located on the side of GVs than the top of GVs in all cases (P ≤ 0.05). CONCLUSIONS: Netarsudil increased high-flow areas. A greater pore density was found in high-flow in contrast to non-flow areas. Netarsudil also significantly increased the proportion of GV-associated I-pores in non-flow areas when compared to control eyes. Our results suggests that one mechanism of netarsudil increasing outflow facility is acting through recruiting the high-flow areas around the circumference of the eye, which is associated with higher pore density and increasing the proportion of GV-associated I-pores in non-flow areas.
