Performance, dry matter intake, seminal parameters and proteomics of seminal plasma and sperm membrane of Morada Nova sheep fed the diet cashew nut base / Desempenho, consumo de matÃria seca, parÃmetros seminais e proteÃmica do plasma seminal e da membrana espermÃtica de carneiros Morada Nova alimentados com dieta à base de castanha de caju

Lucas dos Santos Fonseca 27 December 2013 (has links)
The cashew nut meal is alternative food for ruminant nutrition which is rich in lipids, proteins and minerals. However, its nutrition influences on male development and reproduction are still unknown. In this context, seminal and sperm protein analysis can show metabolic consequences from cashew nut meal on sheep reproductive efficiency. Therefore, the current dissertation aims evaluate the effects of 13% of cashew nut meal in the ration on the weight gain, food intake, carcass yield, weight of sexual organs, sperm quality and, seminal and sperm membrane proteins of Morada Nova Rams. Twenty rams were divided in two groups: cashew nut and control, which received 13% or 0% of cashew nut meal in the diet, respectively. During ninety days, the animals were kept in individual boxes and evaluated for ration consumption and weight gain. The sperm collections by eletroejaculation were made by weekly and, the volume, sperm concentration and motility were analyzed. After ninety days, the seminal and sperm membrane proteins were analyzed by two-dimensional polyacrylamide gel electrophoresis. The gels were digitalized, and their images were evaluated by computer software. At the end of the experiment, the rams were slaughtered and, the weight gain, carcass yield and weight of sexual organs were measured. There was not effect of cashew nut meal addition in the diet on the weight gain, carcass yield and weight of sexual organs. But, after sixty days, there was a reduction on food intake in the cashew nut group (P < 0,05). The sperm quality was not influenced by the diet. We observed an effect of cashew nut diet on the expression of seminal and sperm proteins. Seven spots from seminal plasma were expressed differently between cashew nut and control groups (P < 0,05). The spot 2206 (21,82 kDa; pI: 5,04) was negatively correlated with the individual motility score (r = -0,49). Additionally, seven spots from sperm membrane protein also differ between rams from cashew nut and control groups (P < 0,05). In conclusion, the addition of 13% of cashew nut meal in the diet alters the expression of sheep seminal plasma and sperm membrane proteins.

Plasma Seminal: Efeito na motilidade, funcionalidade de membrana e dispersão da cromatina espermática do sêmen equino refrigerado a 5°C e tratado com N-Acetil-L-Cisteína / Seminal Plasma: Effect on motility, membrane functionality, and sperm chromatin dispersion of equine semen treated with N-Acetyl-L-Cysteine at 5°C

Rodrigues, Murilo Farias January 2013 (has links)
In this study, three concentrations of N-acetyl- l-cysteine (NAC) were evaluated by sperm motility, membrane functionality (hypo-osmotic test), and sperm chromatin integrity. the hypo-osmotic test showed only 32.4% (P <0.05) of the cells having a functional membrane 12h into the cooling process, while the fresh semen (0h) was 53.5%. The percentage of motile cells decreased within 12h of cooling from 38.9 to 19.7%, with no difference between 12 and 24h of cooling. The tests of sperm chromatin dispersion and hypo-osmotic shock revealed similarities between the periods of cooling at 24 to 48h. In Experiment 2, the nine stallion ejaculates were divided into 4 equal parts, diluted in half with Kenney milk powder supplemented with 5.0, 2.5 and 0.5mM NAC and a group not supplemented (0.0mM). The evaluations were made fresh (0h), 24 and 48h after cooling. The analysis of motility was assessed in all experiments up to 24 hours. In experiment 2, the addition of 0.5 to 5.0mM NAC resulted in areas of similar sperm chromatin dispersion (59.7 and 55.5μM2, respectively). However, the area of the chromatin dispersion between non-supplemented group=0.0mM (65.3μM2) and 2.5mM NAC (52.8μM2) differ (P <0.05). The percentage of cells with plasma membrane function were similar between the supplemented with 0.5mM NAC and non-supplemented (0.0mM) groups with (39.7 and 39.8%, respectively), but higher than 2.5mM (34.5%) and 5.0mM (34.2%). Progressive motility of ejaculates supplemented with NAC were similar, but only the group with 0.5mM NAC which resulted in 35.2% of mobile cells showed similarities with the non-supplemented group (36.2%). The greatest damage to sperm chromatin in equine semen diluted with 50% seminal plasma kept cooled to 5°C was due to 24h cooling. The addition of 2.5 and 5.0mM NAC inhibits the mobility and functionality of the equine sperm membrane, but does not cause a deleterious effect on the integrity of the DNA.

Os caracteres analisados não forneceram indícios de um agrupamento apoiando as seções propostas por Radlkofer, mais são caracteres de utilidade potencial na taxonomía e identificação das espécies do gênero / Abstract: In this work were studied the ontogeny, morphology and anatomy of the seeds of some species of the genus Paullinia L. (Sapindaceae), collected in severa I areas in the states of São Paulo, Rio de Janeiro and Paraná. The ontogeny of the fleshy structure of the seed of Paullinia trigonia Vel!. was study for conceptual and morphological purposes. In order to evaluate the importance of the characters of the seed for taxonomic value, the morphology and the anatomy of the seed coat, scar, and fleshy structure were studied in mature seeds of nine species of the genus Paullinia: P. elegans Cambess., P. spicata Benth., P. cupana Kunth, P. carpopodea Cambess., P. meliaefolia A. L. Juss., P. coriacea Casar., P. crista ta Radlk., P. racemosa Wawra and P. trigonia Vell., species belonging to five out of thirteen sections established by Radlkofer. The results indicated that in the ontogeny of the fleshy structure, the outer integument participates in the formation of the fleshy structure in two regions in the seed: in the basal portion of the raphe and in the area near the micropyle. The funicle does not contribute in the formation of the fleshy structure. The fleshy structure was definited as sarcotesta. The term aril referring to this white structure is not used in our work. Aspects relating to the development of the fleshy structure and of the staminate and pistilate flowers, are compared and discussed with other representatives of the genus and the Sapindaceae family. The morphologic characteristics of the seeds present remarkable differences between the species. Three characters altogether allow distinguishing the species easily: 1. Structural size and shape of the seed. 2. Morphology of the white fleshy structure, characterized here as sarcotesta for nine species examined, proportion that covers the seed, and fissures mainly in the anti-raphe region. 3. Shape and size of the scar, lobules in the raphe and anti-rafe regions, dimension and type of fusion of the projections, as well as the degree of the constriction in the intermediate area between the lobes and the projections of the scar. The most useful anatomical characters to differentiate the species are the ones presented in the seed coat: dimension of the exotestal stratum and number of layers and dimensions of the mesotesta. The characters analyzed here do not offer indications of a clustering supporting the sections proposed by Radlkofer, but they are characters of potential utility in the taxonomy and identification of the species of the genus / Mestrado / Mestre em Biologia Vegetal

Influência da disponibilidade de sombra a pasto sobre as características seminais e tolerância ao calor de touros da raça Brahman (Bos taurus indicus) / Influence of shadow availability on semen characteristics and heat tolerance on Brahman bulls (Bos taurus indicus)

Paulo Fantinato Neto 17 December 2010 (has links)
In Brazil, similarly to others tropical countries, bovines are bred under environmental conditions of extensive system. This breed system type may lead to heat stress even on zebu animals. Under heat stress conditions, testicular degeneration can occur and, therefore, there is a search for animals that are heat tolerant. Therefore, the aim of this work was to evaluate the heat tolerance of Brahman bulls using the heat tolerance test and to verify the occurrence of any possible effect of shadow availability on pasture on semen quality. Ten Brahman bulls aging between 24 and 30 months were used in this work. For biological semen evaluation, three semen samples were collected from each animal two weeks before beginning. The bulls were then separated into two groups: 5 animals were allocated on pasture with shadow availability and 5 bulls were allocated on pasture without any kind of shadow. The heat tolerance test were performed in three consecutive typical summer days and the semen samples were collected each 14 days during 2 months, in a total of 4 semen samples per animal. Testicular consistence and scrotal circumference were measured just before every semen collection. The semen\'s characteristics evaluated were volume, aspect, mass movement, motility, straight movement, sperm concentration and morphological exam. Data obtained from experimental proceedings were analyzed by Statistical Analysis System program (SAS Institute Inc., 2004). No difference was observed (P>0.05) in any of the characteristics analyzed when Brahman bulls were maintained on pastures with or without shadow availability. The performance of tested animals on the heat tolerance test shows that these animals are heat tolerant and present good thermolitic ability.

Lucchese, Letícia 23 November 2007 (has links)
The diagnosis of human papillomavirus infection (HPV) in men has gained importance due to the fact they are frequently asymptomatic, who may be a virus reservoir, with an important role in the transmission and perpetuation of the disease. The goal of this study was to identify the presence of HPV in the semen in order to evaluate its influence in semen quality as well as in seminal oxidative stress. Sixty-eight men were analyzed, from which 30 were positive for DNA/HPV and 38 negative. Of the positive cases, 8/30 had HPV infection with the subtypes 16 or 18, which are considered with high prevalence for oncogenic risks. The viral infection and the classification of the virus subtypes were determined with the nested-PCR technique, and the enzymatic activities of superoxide dismutase (SOD) and catalase (CAT) were determined by spectrophotometry technique. There was no significant correlation among the studied epidemiological variables and the presence of the virus. The semen parameters sperm concentration and motility were evaluated manually according to the World Health Organization (WHO) criteria and the sperm morphology evaluated according to the WHO and Tygerberg´s strict criteria. The results show a higher incidence of sperm with abnormal morphology in patients with HPV infection compared to patients without the infection. Sperm concentration and motility did not show a difference between patients with or without HPV infection. In patients with the HPV, CAT levels were higher than patients without the HPV infection. Seminal oxidative stress may be related to the pathophysiology of infertility in patients with seminal HPV infection. This infection signalize for diminishing semen quality based on the damage of the sperm morphology seen in our study.

Runelöv, Martin January 2015 (has links)
We investigate the applicability of network analysis to the problem of finding seminal publications in scientific publishing. In particular, we focus on the network measures betweenness centrality, the so-called backbone graph, and the burstiness of citations. The metrics are evaluated using precision-related scores with respect to gold standards based on fellow programmes and manual annotation. Citation counts, PageRank, and random selection are used as baselines. We find that the backbone graph provides us with a way to possibly discover seminal publications with low citation count, and combining betweenness and burstiness gives results on par with citation count.

Seminal Plasma Metabolome in Relation to Semen Quality and Urinary Phthalate Metabolites Among Chinese Adult Men

Wang, Yi Xin, Wu, Yan, Chen, Heng Gui, Duan, Peng, Wang, Liang, Shen, He Qing, Lu, Wen Qing, Sun, Bin, Wang, Qi, Zhang, Bo, Chavarro, Jorge E., Zhang, Jie, Pan, An 01 August 2019 (has links)
Background: A growing body of evidence has found links between endocrine disruptor phthalates and male reproductive disorders, but the mechanisms underlying these relationships are poorly known. Seminal plasma metabolomes may mediate associations of phthalate exposure with impaired semen quality. Objective: To identify seminal plasma metabolomes associated with poor semen quality and evaluate their associations with urinary phthalate metabolites among 660 Chinese adult men. Method: The seminal plasma metabolic profiles were acquired using an untargeted approach based on liquid chromatography-high resolution mass spectrometry. We explored the differences in seminal plasma metabolites between participants with poor and good semen quality and evaluated cross-sectional associations between discriminatory metabolic biomarkers and urinary phthalate metabolites. Results: Differences between poor and good semen quality groups were observed in relation to 25 seminal plasma metabolites, mostly related to the metabolism of polyunsaturated fatty acids (PUFA) and acylcarnitine (all p < 0.05). After adjusting for various confounders and multiple tests, metabolites were all significantly associated with one or more individual sperm quality parameters (motility, concentration, total count, and morphology) (all p < 0.05). Among identified metabolic biomarkers, seminal plasma L-palmitoylcarnitine, linoelaidyl carnitine, and oleic acid were inversely associated with urinary mono-(2-ethylhexyl) phthalate (MEHP), and seminal plasma L-acetylcarnitine was inversely associated with the proportion of di-(2-ethylhexyl)-phthalate metabolites (DEHP) excreted as MEHP in urine (%MEHP) (all p < 0.05). Mediation analysis revealed that oleic acid and L-acetylcarnitine mediated significant proportions (6.7% and 17%, respectively) of the positive associations between urinary DEHP metabolites and the percentage of spermatozoa with an abnormal head. Conclusions: Elevated urinary phthalate metabolites may impact semen quality by causing metabolic disorders of seminal plasma PUFAs and acylcarnitine. These pathways warrant further investigation.

Biochemical components of seminal plasma of llamas (Lama glama) at three ages

Delgado Callisaya, Pedro Angel 01 January 2002 (has links) (PDF)
This study was conducted at the installation of the Rural Academic Unit-Tiahuanaco of the BCU, located in the community of Achaca, third municipal section of Ingavi province, department of La Paz. It is 57 km from the La Paz-Desaguadero international highway, at 68 degrees 42 minutes 28 seconds latitude South by 16 degrees 35 minutes 41 seconds longitude West, at an altitude of 3856 meters above sea level. The study went from October 2000 to September 2001. The study consisted of determining the concentrations of the biochemical components in llama seminal plasma at three ages. Components studied were glucose, inorganic phosphate, creatinine, total protein, albumin, globulins, cholesterol, calcium, potassium, sodium, and magnesium. Twelve male llamas of 3, 4, and 5 years were selected and acquired from the Choquecota area of the Carangas province, department of Oruro. Four animals were chosen at each age and were subjected to a training period of semen collection during 2 months, using the artificial hindquarters designed for this effect. The 6 that best responded to the training were used for the investigation. Eight collections were obtained from each animal over the course of the study, and they were used for laboratory analysis. The results were analyzed using a hierarchical factorial design that involved a mixed analysis (nested and crossed) of the factors of age and collections. (The averages of two collections corresponding to each week were analyzed.) Each weekly collection average per age was an experimental unit. Four experimental units were obtained for each age, and the analysis of the data was done with the SAS statistics package version 6.12. From the analyses done the following results were obtained: the concentrations of glucose (6.246 [plus or minus] 0.716 mg/dl), creatinine (3.459 [plus or minus] 1.27 mg/dl), cholesterol (67.28 [plus or minus] 18.21 mg/dl), potassium (8.249 [plus or minus] 1.78 mEq/L), and sodium (123.187 [plus or minus] 18.39 mEq/L) did not show significant differences between ages or collections (p>0.05). The concentrations of calcium (12.138 [plus or minus] 3.64 mg/dl) and magnesium (1.943 [plus or minus] 0.52 mEq/L) showed significant (p<0.05) differences in age only and not in collections. Globulins (1.574 [plus or minus] 0.51 g/dl) showed differences between collections (p<0.05) but not between ages. Total protein (3.732 [plus or minus] 0.45 g/dl), albumin (2.158 [plus or minus] 0.46 g/dl), and inorganic phosphate (9.42 [plus or minus] 2.42 mg/dl) showed differences both between ages and between collections (p<0.05).

Paternal programming: the role of seminal plasma in pregnancy hemodynamics and offspring growth

Swanson, Rebecca Michele 08 December 2023 (has links) (PDF)
Seminal plasma is commonly known to serve as a transport medium for sperm as it moves through the female reproductive tract for fertilization, however, more recent evidence demonstrates seminal plasma induces an expansive inflammatory response in the uterus. Murine models have found this inflammatory response is important for clearing pathogens and poor-quality sperm, eliciting the secretion of cytokines, chemokines, and embryokines that aid in embryo attachment and growth, placental angiogenesis, and blunting maternal immunity to the embryo. However, there is minimal research on the impacts of seminal plasma uterine priming in bovine, and more specifically embryo growth, uterine blood flow, offspring growth and metabolism, and production efficiency. There is significant evidence that malnutrition and environmental stress during gestation alters uterine blood flow resulting in poor placental efficiency and poor fetal growth and development which persists postnatally. Animal production is vital in providing high-quality protein for human consumption but recent challenges of public misconception, consumer preferences, high input costs, and environmental impacts threaten the security of these production systems. Growth efficiency is imperative for improving economic and environmental sustainability, and in turn ensuring the longevity of animal production systems. Knowing the impact of seminal plasma on the uterus, and its potential role in placental efficiency and subsequent offspring growth and metabolic function, and the negative impacts these can have on economic and environmental sustainability drive the need to better understand seminal plasma uterine priming in bovine.
