Implementação do biobanco e do laboratório central e validação do protocolo de laboratório do ELSA-Brasil / Implementation of the biobank and the central laboratory and validation of laboratory protocol ELSA-Brazil

Introdução: O Estudo Longitudinal de Saúde do Adulto é um estudo de coorte multicêntrico com o objetivo de identificar os fatores de risco associados ao diabetes tipo 2 e à doença cardiovascular na população brasileira. Nosso principal objetivo é explicar a concepção e implementação das rotinas do laboratório central e biobanco do Estudo Longitudinal de Saúde do Adulto (ELSA-Brasil); destacando as forças e limitações do protocolo. O segundo objetivo é descrever os pré-testes usados para validar o protocolo de antes do início do estudo. Métodos: O estudo optou pela centralização dos exames em um laboratório central no Hospital Universitário da Universidade de São Paulo (USP), Com base em dados recentes que confirma a estabilidade da glicose em amostras congeladas, até os testes de glicose no sangue foram centralizadas. Porém o processamento das amostras foi realizado nos laboratórios locais o que reduziu os custos do transporte das amostras para o laboratório central. A estratégia implicou na necessidade de implementação de equipes nos laboratórios em cada Centro de Investigação para coletar e processar as amostras antes do transporte. O estudo incluiu exames para a avaliação do metabolismo da glicose, resistência à insulina, perfil lipídico, eletrólitos, ácido úrico, hormônios da tireóide, função hepática e contagem total de células do sangue. Além disso, as amostras de DNA, urina plasma, e de soro foram colhidas e Além desses exames, o estudo também extraiu DNA de leucócitos, colheu e estocou amostras de urina, plasma e soro. Para garantir a homogeneidade do protocolo todos os membros das equipes locais passaram por treinamento e certificação centralizado, com visitas cruzadas entre os centros durante o campo para controle de qualidade. Resultados: A opção por um laboratório central garantiu a uniformidade da metodologia utilizada para a realização dos exames, evitando as variações inevitáveis entre laboratórios. Durante 26 meses, cerca de 375.000 testes foram realizados no laboratório central. Não houve perda de amostras biológicas durante o estudo. A implementação do biobanco usando palhetas armazenados em repositórios de nitrogênio foi realizada sem problemas importantes desde 2008 até agora. Conclusão: O ELSA-Brasil mostrou a exequibilidade de exames multicêntricos no Brasil com todos os exames realizados em um laboratório central, de uma maneira custo-efetiva. A logística de armazenamento de amostras biológicas foi feito com custos aceitáveis e de qualidade, sendo um modelo para estudos futuros / Background: The Longitudinal Study of Adult Health is a multicenter cohort study aimed to identify risk factors associated with type 2 diabetes and cardiovascular disease in Brazilian population. Our main objective is to explain the conception and implementation of the routines of the central laboratory and biobank of the Longitudinal Study of Adult Health (ELSA-Brasil) highlighting the strength and the limitations of the protocol The second objective is to describe the pre-tests used to validate the protocol before the start of the study. Methods: The study made an option to centralize the exams in one central laboratory at the University Hospital, São Paulo University (\"USP\"). Based on recent data that confirms the stability of glucose in freeze samples, even blood glucose tests were centralized. However, biological samples were processed in the local laboratories, reducing the weight of the material to be transported, and diminishing costs of transportation to the central lab. Especially trained local teams collected and processed biological samples before transportation. The study included tests for glucose metabolism, insulin resistance, lipid profile, electrolytes, uric acid, thyroid hormones, hepatic function, and total blood cell count. In addition, DNA, urine, plasma and serum samples were collected and stored. In order to guarantee protocol homogeneity, all team members underwent centralized training and certification, and cross-visits in each research center were done. Results: The choice of a central laboratory assured uniformity of the methodology used for the exams, avoiding the variations between laboratories During 26 months, approximately 375,000 tests were done in the central- laboratory. There was no loss of biological samples during the study. The implementation of the biobank using straws stored in nitrogen repositories was performed without important problems since 2008 until now. Conclusion: The ELSA-Brazil showed the feasibility of a multicenter study in Brazil with all the analyses performed in a central laboratory, in a cost-effective way. The logistic of storage of biological samples was done with acceptable costs and quality being a model for future studies

Foi realizado estudo sobre a influência da variação de alguns parâmetros do tratamento térmico de têmpera e revenimento, bem como aplicação de criogenia, na tenacidade a fratura do aço aeronáutico 300M, com a utilização da técnica de corpo cilíndrico com entalhe. Primeiramente, realizou-se a construção da curva de transição dúctil frágil, através de ensaio Charpy, para amostras submetidas a três tratamentos térmicos, 1 - Têmpera seguida de Revenimento, 2 - Têmpera seguida de criogenia e revenimento. Não foi observada influência significativa da aplicação da criogenia (tratamento de numero 2) na curva de transição dúctil-frágil em comparação com o tratamento térmico número 1. Em um segundo momento, com o auxílio da técnica de ensaio de corpo cilíndrico com entalhe, realizou-se a medição da tenacidade à fratura das amostras do aço 300M submetido a quatro diferentes tratamento térmicos, Q, V, C e D. Os tratamentos variavam entre si principalmente pela temperatura de austenitização. Os valores de dureza para as amostras submetidas a criogenia (V e D) foram maiores que as dos tratamentos sem criogenia (Q e C), e também apresentaram tenacidade à fratura menor. Com relação à temperatura de austenitização, observou-se que os tratamentos C e D, austenitizados a 980°C, propiciaram a completa dissolução de precipitados primários da matriz, elevando a tenacidade à fratura com relação aos tratamentos realizados na faixa de 870°C, Q e V, superando também os efeitos negativos para a tenacidade à fratura devido ao aumento no tamanho de grão. / This work studies the influence of the variation for some parameters on quenching and tempering heat treatment, and also deep cryogenic application, on aeronautic steel 300M fracture toughness. First, the ductile-brittle curve was constructed, by Charpy test, for samples submitted for three heat treatments, 1 Quenching and Tempering, 2 Quenching, deep cryogenic and tempering. No significant influence of deep cryogenic treatment addition (heat treatment 2) on ductile-brittle curve was observed in comparison with heat treatment 1. After that, the fracture toughness for 300M steel, submitted for heat treatments Q, V, C e D, was measured using circumferentially notched specimens technique. The main difference among these heat treatments is about austenitizing temperature. For heat treatments with deep cryogenic treatment (V and D), the hardness values were bigger than the ones without this and, analyzing fracture toughness, it was lower than the treatments without cryogenic treatment. Regarding austenitizing temperature, all heat treatments with higher austenitizing temperature at 980°C (C and D), allows complete dissolution of carbides and prior precipitates particles. Its effect was observed that this characteristic increase the fracture toughness when it is compared with others heat treatments with lower austenitizing temperature at 870°C (Q and V). This improvement is enough to overcome the grain size grown negative effects for fracture toughness.

Les échantillons archivés dans les banques d'échantillons de l’environnement (Environmental Specimen Banks en anglais, ESB) sont, parmi les échantillons disponibles pour effectuer des recherches sur l'environnement, les mieux caractérisés géographiquement et issues de longues séries chronologiques. Le mercure (Hg) est un métal lourd hautement toxique et très répandu qui reste l'un des contaminants majeur de l'environnement; les ESB ont joué un rôle de premier plan dans les recherches effectuées sur ce dernier. Ce travail de thèse résume et discute dans un premier temps les modes d’emplois classiques des ESB pour l'étude du mercure dans les écosystèmes aquatiques. Une étude, issue du projet “Seabird Tissue Archival and Monitoring Project” (STAMP), est ainsi présentée pour illustrer comment les œufs d'oiseaux marins peuvent être utilisés pour surveiller les concentrations en Hg environnemental dans les écosystèmes marins subarctique et Arctique. Ce travail présente également un panel d’échantillons archivés dans les ESB du monde entier ; ces échantillons étant estimés comme les plus robustes, fournissent une ressource précieuse pour mener des recherches dans le domaine émergent de la chimie des isotopes stables de mercure. Les signatures isotopiques de fractionnement du mercure sont utilisées pour la répartition des sources et le traçage du cycle biogéochimique du Hg. Ceci est illustré par les résultats de deux études effectuées sur des œufs d'oiseaux marins d’Alaska archivés. Etant donné que les recherches sur les isotopes stables du mercure continuent d'évoluer, les ESB peuvent fournir une source d’échantillons robuste et économique pour élargir et diversifier l'inventaire des mesures des isotopes de Hg, et être utilisées pour développer et tester des hypothèses. Les échantillons archivés dans les ESB sont disponibles à des collaborateurs externes afin d'effectuer des recherches à fort impact, et devraient être pleinement exploités pour répondre aux nouvelles préoccupations environnementales. / The specimens archived in Environmental Specimen Banks (ESBs) are among the longest time-series, most geographically robust, and highest integrity samples available for performing environmental research. Mercury (Hg) is a highly toxic and ubiquitous heavy metal that remains one of the world’s most prominent environmental contaminants, and ESBs have played a prominent role in Hg research. We review and discuss some of the ways in which ESBs have traditionally been used to study Hg in aquatic ecosystems, and show an example of how seabird eggs from the Seabird Tissue Archival and Monitoring Project (STAMP) are used for monitoring environmental Hg concentrations in subarctic and Arctic marine ecosystem. We also highlight some of the most robust sample sets archived in ESBs worldwide, which provide a valuable resource for conducting research in the emerging field of Hg stable isotope chemistry. Signatures of Hg isotope fractionation are used for source apportionment and for tracing biogeochemical cycling of Hg. We present results from two additional studies in Alaska that show how banked seabird eggs can be used for each of these purposes. As the research surrounding Hg stable isotopes continues to mature, ESBs can provide a robust and economical sample archive to expand and diversify the inventory of Hg isotope measurements, and be used to develop and test hypotheses. Samples archived in ESBs are available for request by external collaborators in order to perform high impact research, and should be fully utilized to address emerging environmental concerns.

Excessive usage of wildlife is the second most common cause of species decline on our planet, and thus one of the direct threats to biodiversity. Since biodiversity has a virtually existential significance for the well-being of people, it needs to be cared for in order to slow down its decline. The aim of this thesis is to analyze and evaluate legal regulations at the international, EU, and Czech level, to present the instruments and institutes used to regulate the trade in CITES specimens, and to propose solutions for potential problems. Where deemed necessary or appropriate, a comparison of international legal regulations with EU and Czech legislation is made. This thesis is divided into 6 chapters that are further internally subdivided into subchapters. The first chapter deals with the importance of trade in endangered species of fauna and flora in relation to the loss of biodiversity. The second chapter provides an overview of the legal regulation governing trade in endangered species at different levels (international, EU, and Czech). The third chapter introduces the individual bodies responsible for the implementation of the Convention CITES (or Regulation No. 338/97) at international, EU, and Czech level. Stability of End Notched Flexure Specimen

This paper deals with two-dimensional Finite Element Analysis of the End Notched Flexure (ENF) specimen. The specimen is known to be unstable if the crack length is shorter than some critical crack length acr. A geometric linear two-dimensional Finite Element (FE) analysis of the ENF specimen is performed to evaluate acr for isotropic and orthotropic elastic materials, respectively. Moreover, the Mode II Energy Release Rate (ERR) JII and the compliance of the specimen are calculated. The influence of anisotropy is studied. Comparisons are made with the results from beam theory. This work is an extension of previous work.

Ihre Anwendbarkeit wird durch die Auswertung von Versuchen an verstärkten Stahlbetonplatten mit einer Reihe veränderlicher Ausgangswerte überprüft. Als Ergebnis dieser Arbeit stehen dem Anwender einfach nutzbare, praxisgerechte Bemessungstabellen zur Verfügung. In dieser Version weisen die Grafiken eine bessere Qualität auf. / This article introduces a calculation method and a design concept for reinforced concrete elements strengthened with textile concrete. Its applicability is verified by an analysis of tests with strengthened steel reinforced concrete slabs with a certain variety of design parameters. Results of this work are easy manageable, praxis-orientated design tables that are available for users. This version contains figures with a better quality.

The Development of Leibniz’s Substance Ontology From 1666-1688

Leibniz’s early conception of individual substance ontology is one of the most puzzling, and fascinating, within the history of philosophy. It is the purpose of this paper to show that: 1) Leibniz did develop a coherent scheme that embodied his substance ontology, 2) the exposition of his early substance ontology is in A Specimen of the Discoveries of the Admirable Secrets of Nature in General, written in 1688 and, 3) the scheme is not sufficiently represented in the Discourse on Metaphysics. Leibniz slowly developed a multifaceted view of substance within the twenty years previous to the writing of the Discourse. This view is comprised of the matter/form complex, the predicate-in-subject thesis and, the phenomenal characteristics of material interaction. These three facets can also be viewed as ontological, teleological/ epistemological, and phenomenological, respectively. These facets were developed concurrently and are interdependent. The understanding of any facet requires the understanding of all of them. From the exploration of Leibniz’s development of substance ontology, one can understand his presentation of rational theology in the Discourse. Leibniz develops the ontology to account for the infinite nature of material division. The unification of material bodies requires explanation. Leibniz has the desire to create a method of deriving a priori knowledge of God, the universe, and humanity; he believes his substance ontology creates the firm basis needed to accomplish this task. The Discourse on Metaphysics does not itself represent the complete scheme Leibniz developed. It shall be shown that A Specimen of the Discoveries of the Admirable Secrets of Nature in General, composed in 1688, is a definitive exposition of Leibniz's early substance ontology. The Discourse on Metaphysics can be viewed as an exposition of rational theology based upon the ontology Leibniz had developed.

Investigation Of The Effect Of Orientation And Heat Treatment On The Stress Corrosion Cracking Susceptibility Of 7050 Aluminum Alloy

In the present work, the effect of variation in specimen orientation and heat treatment on the Stress Corrosion Cracking (SCC) susceptibility of 7050 aluminum alloy was investigated in 3,5% NaCl solution and under freely corroding conditions. For this purpose, Constant Extension Rate Tests (CERT) was performed on precracked Compact Tension (CT) specimens and the Direct Current Potential Drop technique was applied to measure the crack lengths. In addition to crack length versus time curves, the relationship between the crack growth rate and the stress intensity factor was determined. Fractographic analysis was utilized extensively to support the findings related with basic mechanisms of cracking. The alloy was found to be in the most susceptible state in the SL orientation, in which the crack propagation direction is parallel to the rolling direction. The resistance to SCC is higher in the TS but at maximum in LT orientation where the loading direction is parallel to the rolling direction. In the peak aging treatment, T651, alloy is susceptible to SCC in SL orientation. When the over aging treatment, T7651, is applied the resistance is increased and the two step over aging treatment, T73651, has resulted in an additional improvement in this orientation. On the other hand, the alloy showed higher resistance to SCC in TS and LT orientations in T651 condition compared to the T7651 and T73651 treatments. In these orientations, the alloy is less susceptible in T73651 condition than in T7651 treatment.

Evaluation Of Cement Mortars By Ultrasound

Ultrasonic testing of concrete is often used for the assessment of its uniformity, strength, modulus of elasticity, durability and etc. therefore, the related parameters of testing such as the transducer frequency, the specimen geometry and etc. are well-known. On the other hand, most of the concrete properties are affected by the cement and the mechanical as well as some durability properties of cements are determined through cement mortars. Applications of ultrasound on determining the properties of cement mortars are quite limited. Therefore, the required specimen dimensions, transducer frequencies have not yet been established for cement mortars. In this study, ultrasonic pulse velocity (UPV) of mortars was determined with different transducers of different frequencies for different size ans shape of specimens. Within the scope of the experimental program, three different ultrasonic frequencies (54 kHz, 82 kHz, and 150 kHz) were utilized and the relation between ultrasonic testing frequency and specimen shape was experimentally investigated. It was concluded that the mechanical properties of mortar was adversely affected by the water-to-cement ratio. It was also observed that, when the length/wavelength ratio increases, the measured UPV with different transducer frequencies tends to converge to a single value. Finally, it was also concluded that an increase in moisture content of the mortar mixtures causes an increase in UPV and a decrease in compressive strength.
