Ce travail de thèse est une étude in silico des structures tridimensionnelles des protéines, qui a fait l’objet de 5 publications scientifiques.D’une manière plus précise, les travaux s’articulent autour de trois thématiques originales et complémentaires dans le domaine de la bioinformatique structurale : la caractérisation d’un nouvel échelon de description de la structure des protéines (les unités protéiques), intermédiaire entre les structures secondaires et les domaines.Le deuxième axe de cette thèse porte sur le développement d’une nouvelle méthode de prédiction des structures protéiques, appelée ORION.Cette méthode permet une détection accrue d’homologues lointains grâce à la prise en compte de l’information structurale sous forme d’un alphabet structural (les blocs protéiques).Une seconde version améliorée a été rendue accessible à la communauté scientifique par le biais d’une interface web : http://www.dsimb.inserm.fr/ORION/.Le dernier axe de cette thèse, s’oriente autour du développement d’outils, pour la prédiction de l’orientation et l’évaluation de la membrane dans les structures de protéines membranaires effectué dans le cadre de plusieurs collaborations.Les outils développés (ANVIL et MAIDEN) ont été mise à la disposition de la communauté scientifique par le biais d’une interface web appelée OREMPRO et accessible à l’adresse suivante : http://www.dsimb.inserm.fr/OREMPRO/. / This thesis deals with three complementary themes in the field of structural bioinformatics : the characterization of a new level of description of the protein structure (Protein Units) which is an intermediate level between the secondary structures and protein domains. Development of a Comprehensive Linear Response History Analysis Procedure for Seismic Load Analysis

This thesis reviews the parameters required to perform linear response history analysis according to Chapter 16 of the American Standard ASCE 7-10. A careful analysis is presented about the selection of ground motions using real records and using artificial records generated such that their response spectrum matches with a defined target spectrum; three different techniques are studied for the generation of these artificial records. Also, this document revises the scaling of ground motion techniques in the American Standard ASCE-7 as well as in other seismic codes. It presents a detailed analysis of the variables influencing the scaling of ground motions, and it suggests a new scaling technique for linear response history analysis. The assumptions made establishing the flexibility of the diaphragms are also analyzed as well as dynamic methods to include accidental torsion when doing a linear response history analysis. Other modeling issues such as the orientation of the ground motion axis, scaling of element forces and displacements, orthogonal loading, solution techniques, P-Delta effects, modeling of the basement, and calculation of drifts are also studied in the context of linear response history analysis. The thesis concludes with suggested code language for linear response history analysis intended to be considered in future editions of the American Standard ASCE 7. / Master of Science

The development and validation of a brand-building framework for platform-centric startups

Platform-centric startups that generate value by facilitating interactions between two or more interdependent groups digitally transform and disrupt the business environment, leading to significant changes in how people purchase, sell, communicate, and even live. Some of these, such as Uber and Airbnb, have become among the most beloved and successful brands, though they had serious challenges in building up their brands at the early stages due to lack of resources. The purpose of this dissertation is to develop a framework of branding in platform-centric startups by identifying the pillars of brand‐building and linking the relevant factors. This study employs qualitative method to propose a four-level framework including passion, innovation, co-creation, and moralization for brand-building in startups. The novelty of this framework reveals that the locus of brand-building is gradually expanding from the founders to the company, then to the customers, and finally to a larger societal constituency. The framework can assist founders and stakeholders in monitoring the growth of a new venture in the real business world while guiding branding scholars to investigate the discipline of startup branding.

yes / Cybersecurity is a serious issue that many organizations face these days. Therefore, cybersecurity management is very important for any organization. Organizations should learn to deal with these cyber threats through effective management across all business functions. The main purpose of this study is to identify the factors that affect cybersecurity within an organization and analyze relationships among these factors. The modified total interpretive structural modeling (M-TISM) technique is used to build a hierarchical model and define the common interactions between the factors. This study presents the impact of collaboration, training, resources and capabilities, information flow, technology awareness, and technological infrastructure on effective cybersecurity management. In addition, the study also explains the interrelationships among the identified factors in the M-TISM model.

Predicting Compression Failure of Fiber-reinforced Polymer Laminates during Fire

A thermo-structural model was developed to predict the failure of compressively loaded fiber-reinforced polymer (FRP) laminates during fire. The thermal model was developed as a one-dimensional heat and mass transfer model to predict the thermal response of a decomposing material. The thermal properties were defined as functions of temperature and material decomposition state. The thermal response was used to calculate mechanical properties. The structural model was developed with thermally induced bending caused by one-sided heating. The structural model predicts out-of-plane deflections and compressive failure of laminates in fire conditions. Laminate failure was determined using a local failure criterion comparing the maximum combined compressive stress with the compressive strength. Intermediate-scale one-sided heating tests were performed on compressively loaded FRP laminates. The tests were designed to investigate the effect of varying the applied stress, applied heat, and laminate dimensions on the structural response. Three failure modes were observed in testing: kinking, localized kinking, and forced-response deflection, and were dependent on the applied stress level and independent of applied heating. The times-to-failure of the laminates followed an inverse relationship with the applied stress and heating levels. The test results were used to develop a relationship which relates a non-dimensionalized applied stress with a non-dimensionalized slenderness ratio. This relationship relates the applied stress, slenderness ratio, and temperature of the laminate at failure and can be used to determine failure in design of FRP laminate structures. The intermediate-scale tests were also used to validate the thermo-structural model with good agreement. / Master of Science

O núcleo resistente, considerado um dos principais elementos componentes dos sistemas estruturais de edifícios de andares múltiplos, quando associado ao sistema de contraventamento, consegue conferir à estrutura um razoável acréscimo de rigidez. O núcleo é ainda o único elemento estrutural capaz de resistir isoladamente a todos os esforços atuantes na estrutura de um edifício, contribuindo na determinação mais precisa dos seus deslocamentos. Baseado nestes fatores, acredita-se que as diferentes modelagens, adotadas para o núcleo, proporcionarão diferentes resultados para a mesma estrutura de contraventamento. Este trabalho tem como principal objetivo analisar a influência que estas modelagens exercem na estrutura de contraventamento. Inicialmente, são apresentadas algumas das modelagens mais utilizadas para o núcleo. Com estas modelagens foram processadas algumas estruturas, e os diversos resultados obtidos utilizados no processo comparativo. Para o processamento foram consideradas análises em teoria de primeira e segunda ordem e, para as fundações, vinculação rígida. Não serão consideradas as deformações por esforço cortante nos elementos. / Resistant cores, are considered one of the principal elements of the structural systems of multistory buildings. When associated to the bracing structure, it gives to the structure a reasonable stiffness increment. The core is still the only structural element capable to resist separately all the efforts in the structure of a building, contributing to a more accurate determination of its displacements. Based on these factors, it is believed that different modeling adopted for the cores will provide different results for the same bracing structure. The main aim of this work is to analyze the influence that these modeling exerts on the bracing structure. First, some of the most used core modeling were presented. Then some structures were processed, using these modeling. The several obtained results were compared. For all the analysis it was considered the first and second order theory and rigid connections for the foundations. Shear deformation were not considered in the elements.

Modeling the structure, dynamics, and interactions of biological molecules

Biological molecules are essential parts of organisms and participate in a variety of biological processes within cells. Understanding the relationship between sequence, structure, and function of biological molecules are of fundamental importance in life science and the health care industry. In this dissertation, a multi-scale approach was utilized to develop coarse-grained molecular models for protein and RNA simulations. By simplifying the atomistic representation of a biomolecular system, the coarse-grained approach enables the molecular dynamics simulations to reveal the biological processes, which occur on the time and length scales that are inaccessible to the all-atom models. For RNA, an "intermediate" coarse-grained model was proposed to provide both accuracy and efficiency for RNA 3D structure modeling and prediction. The overall potential parameters were derived based on structural statistics sampled from experimental structures. For protein, a general, transferable coarse-grain framework based on the Gay-Berne potential and electrostatic point multipole expansion was developed for polypeptide simulations. Next, an advanced atomistic model was developed to model electrostatic interaction with high resolution and incorporates electronic polarization effect that is ignored in conventional atomistic models. The last part of my thesis work involves applying all-atom molecular simulations to address important questions and problems in biophysics and structural biology. For example, the interaction between protein and miRNA, the recognition mechanism of antigen and antibody, and the structure dynamics of protein in mixed denaturants. / text
