Performance practice is a term familiar to serious musicians. For the performer, this means assimilating and applying all the education and training that has been pursued in a course of study. Performance practice entails many aspects such as development of the craft of performing on the instrument, comprehensive knowledge of pertinent literature, score study and listening to recordings, study of instruments of the period, notation and articulation practices of the time, and issues of tempo and dynamics. The orchestral literature of Eastern Europe, especially Germany and Russia, from the mid-nineteenth century through the mid-twentieth century provides some of the most significant and musically challenging parts for the tuba. The works of Igor Stravinsky, Sergei Prokofiev, and Dmitri Shostakovich, along with their orchestral contemporaries, represents a significant portion of this literature. This study examines a seminal work in the orchestral genre from each of these three Russian composers. The role of the tuba in each work is discussed. Excerpts of the tuba part are examined in terms of performance issues such as range, rhythm, phrasing, and scoring. Comparisons and contrasts are drawn as to how each composer used the tuba and the effectiveness of the utilization.

Diploma thesis „Náměstí Míru – extension and rehabilitation of public space in Brno“ deals with the analysis of the current state of defined part of Masaryk district, the evaluation of the problems of the imaginary city center and the design of buildings along with the working transport solution. The aim of the thesis is to define „Náměstí Míru“ as a functional public space, that people can identify with. To modify traffic solutions to avoid confused situations and collisions between pedestrians, trams and cars. To design new buildings to replace brownfields, create new housing units, and offer opportunities for using an active parterre. Emphasis is placed on modern urbanistic approaches and views of the 21st century, compact building, multifunctionality of buildings, and prioritization of city planning and public spaces with priority on human, his interests, needs, and everyday life.

The diploma thesis is the proposal of the Christian community and spiritual center near Church of St. Anthony of Padua in Ostrava Kunčičky. The building program corresponds to the catchment area of Kunčičky and includes an existing church building (conversion for the contemporary liturgy) with the addition of a new parish with a living space, a parish office and a meeting room. The existing building of the parish (adaptation for commercial use, bistro and offices) and the community center includes: a social hall with the necessary background, 2 flats 1 + kk and apartment for collective accommodation, clubroom and classroom for leisure activities, music, art workshop and warehouse. Part of the area is new square as the new centre of Kunčičky, the park and green area "open garden" with a children's and sports playground. The proposal is based on the creation of a new public space in the vicinity of the existing church. The pre-church is widened and creates a new square, the center of the city with the potential of meeting, landmark, festivities, Christmas markets, etc. On the contrary, the community center is behind the church and creates a new public space "open garden". Around it all community events are concentrated. Among these spaces is a "filter" set, as a soothing element of two poles. The complex offers people from this locality also a wide range of the city, a new offer of cultural, social and spiritual activities.

This dissertation examines the medieval libraries of the order of Regular Canons of St. Augustine in Bohemia. The research is concentrated on manuscripts that were written before the end of the 15th century, together with handwritten parts bound together with old prints and fragments removed from manuscripts. The aim of the study is to fill gaps in existing research and provide a systematic overview of the libraries of canonries in Bohemia, namely Roudnice, Jaroměř, Karlov in Prague, Sadská, Rokycany, Třeboň, and Borovany. The research of these libraries is based mainly on preserved manuscripts. Their affiliation to the libraries can be identified with certainty thanks to ownership remarks, in some cases also by typical shelf marks or notes about the content of manuscripts, possibly by other circumstances connected with the history of the books. In addition, there are contemporary sources of the history of some libraries (library catalogues, inventories, records of gifts and testaments, notes about payments for manuscripts, records of the purchase of the books, or reports of the sale of manuscripts in exile). The aim of this thesis "Theology of Labour of St. John Paul II." is to summarize John Paul II.'s theological view of human labour, its impact on Christian spirituality and evangelization and pastoral efforts of the church. There is also attention paid to influence of his theology on further social magisterial texts. At the end, there is a reflection of impacts of this teaching to everyday economic and political life. The thesis is based on magisterial documents of John Paul II., especially on the encyclical Laborem exercens and his two further social encyclicals, Sollicitudo rei socialis and Centisimus annus. Other inspiring ideas come especially from reflections on St. Joseph and from the theology of body. There is a discussion on relation of human labour to the Mystery of The Holy Trinity, on perichoretical and sharing character of human labour. As a part of discussion on spirituality, there is the work state of life defined as analogy to Christian state of life. Human labour is a key to the social question, it is a tool for our salvation and therefore the mission of the church. St Vitus' Cathedral in Prague: Liturgy, Symbolism and Architectural Imitation in Medieval Bohemia In this thesis, the focus is put on the Wenceslas Chapel in St Vitus' Cathedral, the unusual design of which seems to have two levels of meanings: On the one hand, it was probably designed as a recreation of an earlier rotunda that had been founded by St Wenceslas. On the other hand, it seems that the intention was made the chapel and the whole cathedral would resemble the Church of the Holy Sepulchre in Jerusalem, where the Calvary Chapel occupied the same position as the Wenceslas Chapel. This was most likely inspired by presence of the Imperial insignia containing the valuable Passion relics that Charles had acquired in 1350 and placed in the basilica, probably near the Wenceslas Chapel. Perhaps from that reason Charles had the new Gothic chapel decorated with semi-precious stones, because its red spots were associated with the blood of Christ and the Passion. The Wenceslas Chapel together with two Passion chapels at Karlstein Castle, containing Charles IV's private passion treasure, gave rise to the "Passion" architecture of the Luxembourg period. The aim of the thesis is to describe and analyze the function of the Jesuit order in the royal city of Jihlava in the years 1625-1682 with special focus on the question how the Jesuit college managed to anchor itself in an environment that was mostly Lutheran for almost a century. The thesis is based on sources of both municipal and Jesuit provenance - i.e. mostly of Latin and German provenance, stored dominantly in the Moravian Land Archive in Brno and in the funds of the State District Archive in Jihlava. Thus, the thesis expands the topic of an already defended bachelor thesis, which focused on the establishment and institutional security of the Jesuit college in the context of the political and religious development of Moravia. The thesis thoroughly describes the relationship between the royal city and its burghers and the Jesuit college. From the viewpoint of the burghers, I follow the relationship on a study of pious gifts given to the Jesuit college by the burghers on ground of their testaments. This allows determining the level of attention paid to the Jesuit college by the burghers and whether they preferred it to other church institutions in the city. Detecting structural variants in the DNA of the inbred Scandinavian wolf

Only 40 years ago, just three individuals made the journey from Finland/Russia to found the current wolf population in the southwest of Sweden. This population, that to this date descends from less than 10 founders, has a substantial increased extinction risk due to inbreeding. Several previous studies have used SNPs to monitor the level of inbreeding and homozygosity in the population, as well as measure immigration and the inflow of new genetic material. This study uses both short- and long-read data to discover structural variants (SVs) and small indels in the population, so that they may be used to extend the analyses and provide more insight into the current state of the Scandinavian wolf population. After the calling of the SVs, strict filtering and manual curation were applied to the data, thereby removing many false positive calls and increasing confidence in the remaining SVs. Short-read and long-read SV-callers found 31,800 and 57,821 SVs respectively, with relatively little overlap between the two sets. By far, the most common SV-types were deletions and insertions, at about 30,000 each with varying length ranging from a 50 base pairs to several tens of Mbp. Analyses on the data, such as PCAs and parent-offspring trio analyses, reveal high-confidence calls and consistent results between SV-types and SV-callers, as well as a low estimated genotyping error rate. PCAs performed on the SVs resembled those performed on SNPs, which strengthens the credibility of the identified variants. Finally, this study suggests several alternative steps for possible improvement to the dataset, along with some proposals for subsequent research topics that may use the variants discovered in this study.
