Wertigkeit von Entzündungsparametern in der postoperativen Phase nach Implantation von Hüft- und Kniegelenk-Totalendoprothesen / relevance of inflammatory parameters after hip and knee arthroplasty

Ziel dieser Arbeit ist, die Wertigkeit von Entzündungsparametern (CRP, BSG, Leukozyten und Temperatur) in der postoperativen Phase anhand einer retrospektiven Studie mit 531 Patienten nach Implantation von Knie- und Hüft-Totalendoprothesen zu beurteilen. Hierbei sollten zum einen die Auswirkungen von Vorerkrankungen (Asthma bronchiale, Rheumatoider Arthritis, Nikotinabusus, Gicht/Hyperurikämie, Z. n. Thrombosen) auf den postoperativen Verlauf der Entzündungsparameter und zum anderen postoperative Komplikationen (Thrombosen, bronchopulmonale Infekte, Harnwegsinfekte, Wundinfekte, Protheseninfekte) anhand der Entzündungsparameter erfasst werden. Die Auswertung erfolgte mittels SPSS für Windows durch den Mann-Whitney Test als nicht parametrischen Test für unabhängige Variablen. Im Durchschnitt stieg der CRP-Wert von 0,6 mg/dl präoperativ auf etwa 8,5 mg/dl um den 3. postoperativen Tag an und fiel dann durchschnittlich auf 1,4 mg/dl um den 14. postoperativen Tag ab. Die BSG-Werte stiegen von etwa 18 mm/h präoperativ auf 52 mm/h um den 3. postoperativen Tag an. Der höchste BSG-Wert (56 mm/h) wurde erst um den 7. postoperativen Tag erreicht. Danach fiel der BSG-Wert leicht ab auf durchschnittlich etwa 43 mm/h um den 14. postoperativen Tag. Die Temperaturkurve verlief ähnlich: Auch hier stieg die Temperatur rasch auf einen Höchstwert von 37,1°C um den 2. postoperativen Tag. Es folgte ein langsamer Abfall der Temperatur, bis hin zu Normwerten ab dem 10. postoperativen Tag. Die Leukozyten zeigten im Durchschnitt einen geradlinigen Verlauf mit durchschnittlichen Werten zwischen 7 und 8 G/l, allerdings bei einer großen Standartabweichung. Patienten mit Asthma bronchiale zeigten präoperativ signifikant erhöhte BSG und Leukozytenwerte. Patienten mit Gicht/Hyperurikämie fielen durch signifikant niedrigere Temperaturwerte am 6. und 12. postoperativen Tag sowie durch Trends zu niedrigeren Werten am 10. und 16. postoperativen Tag auf. Signifikant höhere Werte fanden sich bei der CRP-AUC-Kurve vom 7.-14. postoperativen Tag. Des Weiteren fanden sich Trends zu höheren Werten am 14. postoperativen Tag bei der BSG und am 3. postoperativen Tag bei den Leukozyten. Bei Patienten mit Z.n. Thrombose zeigte sich der postoperative BSG-Verlauf vom 3.-14. Tag signifikant erniedrigt, ebenso waren die postoperativen Temperaturwerte vom 15. und 16. Tag signifikant niedriger. Der einzig signifikante Unterschied zwischen Rauchern und Nichtrauchern fand sich in der präoperativen BSG, wo Raucher signifikant niedrigere Werte hatten. Des Weiteren zeigten sich Trends zu höheren Leukozytenwerten am 3. und 7. postoperativen Tag sowie Trends zu niedrigeren Temperaturen am 1., 2., 7., 8. und 14. postoperativen Tag. Patienten mit Rheumatoider Arthritis fielen durch signifikant erhöhte präoperative CRP- und BSG-Werte auf. Zusätzlich traten Trends zu höheren Werten am 14. postoperativen Tag für CRP, BSG und Leukozyten auf. Dem Trend zur Temperaturerhöhung am 2. postoperativen Tag folgten erneute Trends am 9. und 11. postoperativen Tag sowie signifikant höhere Temperaturen am 10. und 12. postoperativen Tag. Patienten, die einen Harnwegsinfekt erlitten, hatten signifikant erhöhte Werte am 14. postoperativen Tag für CRP und BSG. Auch die CRP-AUC-Kurve zeigte diesen Verlauf signifikant. Bei Patienten, die an einem bronchopulmonalem Infekt erkrankten, war der CRP-Wert des 3. postoperativen Tages signifikant erhöht. Dies ließ sich auch in der CRP-AUC-Kurve vom 0.-3. postoperativen Tag signifikant nachweisen. Darüber hinaus lag ein Trend am 3.-7. postoperativen Tag zu höheren Werten vor. Bei Patienten mit tiefer Beinvenenthrombose fiel eine signifikante Temperaturerhöhung am 7., 8. und 11. postoperativen Tag auf, sowie ein Trend am 9., 10. und 12. postoperativen Tag. Die CRP-Werte stiegen um den 14. postoperativen Tag signifikant an. Gleiches Verhalten zeigte die CRP-AUC-Kurve vom 7.-14. postoperativen Tag. Patienten mit Wundinfektionen fielen durch Trends zu niedrigeren Temperaturwerten am 4. und 5. postoperativen Tag auf. Außerdem stieg um den 7. postoperativen Tag die BSG trendwertig an. Die Leukozyten waren präoperativ signifikant erhöht. Dagegen war bei Patienten mit TEP-Infektion der CRP-Wert um den 14. postoperativen Tag signifikant erhöht. Die Temperaturwerte des 8. postoperativen Tages zeigten einen Trend zu niedrigeren Zahlen. Die Ergebnisse entsprechen in etwa denen anderer Studien. Insgesamt lässt sich eine Tendenz ablesen: BSG und Temperatur sind vor allem bei den erwähnten Vorerkrankungen erhöht, Leukozyten spielen kaum eine Rolle und die CRP-Bestimmung findet ihren Platz hauptsächlich in der Diagnostik von Komplikationen. Außerdem wird klar, dass kein Entzündungsparameter spezifisch für eine bestimmte Vorerkrankung oder Komplikation ist. Darüber hinaus sind nicht Einzelwerte, sondern viel mehr der Verlauf der Entzündungsparameter entscheidend. / Blood parameter like CRP (C-reactive protein), ESR (erythrocyte sedimentation rate), white blood cells (leukocytes) and body temperature were messured after total hip and knee arthroplasty in 531 patients. The effects of bronchial asthma, rheumatiod arthritis, smoking, gout and a deep vein thrombosis in the past on inflammatory parameters were investigated, on the oher side we tried to check out, whether deep vene thrombosis, bronchial or pulmonary infections, urinary tract infections, wound infections or infections of the endoprothesis influence the developing of inflammatory parameters.

LISST Instruments as a Tool in Phytoplankton Ecology

Laser in situ scattering and transmissometry (LISST) instruments are used to measure the particle size distributions (PSDs) and volume concentration of individual and groups of phytoplankton in water. The objective of this research was to test the LISST’s ability in detecting monospecific blooms in-situ and the ability to detect aggregation after diatoms were subjected to different temperatures and bacteria concentrations. The PSDs of ten harmful algal bloom (HAB) species were measured with the LISST characterizing the peak location, peak height, peak width, and peak range resulting in a scattering signature for each species. Each species had specific characteristics that would allow for their detection with the LISST, though microscope observations would be needed for complete accuracy. The LISST was able to detect HABs placed in natural seawater collected off the Texas coast. Blooms of four HAB species before they reached full” bloom concentrations were detected making the LISST a possible low cost, effective tool in the early detection and monitoring of HABs. The diatom, Odontella aurita, was used to test how well the LISST could monitor aggregation, an important process in the termination of many phytoplankton blooms. Increasing temperature causes an increase in transparent exopolymer particle (TEP) production in diatoms, which is a critical sticky particle that increases the probability of aggregation. An increase in temperature can also cause an increase in bacteria concentration that can positively effect TEP production and thus aggregation. O. aurita was grown at 20 °C and 28 °C and showed a significant increase in TEP abundance with temperature (p = 0.002), though no relationship between TEP production and bacteria concentration existed. Coomassie stained particles (CSP) are proteinaceous gel-like particles, which are currently understudied. CSP was consistently produced though it did not appear to be dependent upon any single factor. The increase in ocean temperatures has implications for an increase in phytoplankton blooms making the monitoring and understanding of these blooms even more important as they can affect the carbon cycle and potentially the microbial loop.

Laser Scattering as a Tool to Determine the Effect of Temperature on Diatom Aggregation

Diatoms are estimated to contribute 25 percent of the primary production on Earth and therefore they play a significant role in the global carbon cycle. Diatom blooms often terminate with the formation of aggregates that sink rapidly from surface waters, affecting the flux of organic carbon from the surface to deep waters and the sea floor. The role of carbon-rich transparent exopolymeric particles (TEP) in aggregate formation as ocean temperature increases has yet to investigated in continuous cultures. I hypothesize that temperature increase can influence the production of TEP, a fraction of total suspended exopolymers. To test the hypothesis, a laser in situ scattering and transmissometry instrument (LISST-100X, Sequoia Instruments) successfully counted and sized six individual diatom species in batch culture: Chaetoceros muelleri, Coscinodiscus wailesii, Thalassiosira weissflogii, Phaeodactylum tricornutum, Skeletonema costatum, and Skeletonema marinoi and successfully demonstrated its efficacy in detecting diatom aggregates using S. costatum. Four replicate continuous cultures were sampled for particle size distribution (PSD), nutrients, chlorophyll a, total carbohydrates, prokaryote concentration, and TEP at temperatures of 22.5, 27 and then 20 degrees C. While TEP particles were scarce, acid polysaccharide (APS)-coated C. muelleri cells were observed, forming dense webs on the filters. Both carbohydrate per cell and APS area per cell were found to significantly correlate with temperature (p<0.05) while significant difference between APS concentration at each temperature was only found between 27 and 22.5 or 20 degrees C (p<0.05). Net changes in PSDs with increasing temperature showed that distributions of relative volume concentration decreased in the smallest size bins and increased in the largest size bins. Our results show that increasing the temperatures of nitrogen-limited C. muelleri cultures did not cause increased TEP formation but instead resulted in increased cell-surface coating. Increasing concentration of cell coatings and TEP particles will cause diatoms to aggregate more readily, enhancing their sinking rate away from the ocean surface. Increased ocean temperature has great implications for diatom blooms and other microorganisms, causing greater export of carbon out of the surface waters and potentially altering the microbial loop.

The sinking rate and transparent exopolymer particle (TEP) production of Hemiaulus hauckii

Diatom-diazotroph associations contribute extensively to vertical flux in the modern ocean, and are linked to extensive sedimentary layers in the paleo-record formed either by rapidly sinking aggregates or more slowly sinking cells and chains. Although diatom sinking rates have been extensively studied, the sinking rates of these diatom symbioses are poorly documented. The effect of silicate and phosphate limitation on the sinking rate of Hemiaulus hauckii and its symbiont Richelia intracellularis were determined in semi-continuous cultures using a SETCOL. TEP (transparent exopolymer particle) was measured concurrently for linkages to aggregate formation. Although cells occurred in chains of 1-20 cells long, aggregate formation did not occur in semi-continuous cultures. There was no significant effect of P or Si limitation on sinking rates compared to controls. Although H. hauckii possess a symbiont that lacks gas vesicles, sinking rates (0.04-0.72 m/day) were within the range predicted by size-rate relationships. A portion of each H. hauckii experimental measurement ascended as well with significant differences found between the lowered P 80% exchange and the lowered Si with 80% exchange (p=0.05) and between the lowered Si with 80% exchange and the Si control batch culture (p=0.039). TEP production did not change significantly between treatments. However, a fraction of TEP in each population was positively buoyant as well, with ascent rates between 0.32-0.49 m/day. This study confirms an earlier report of positive buoyancy in TEP as well as noting that a significant fraction of the Hemiaulus symbiosis can be ascending within the population. / text

Réalisation d'un détecteur de radioactivité pour un système microfluidique

Pour établir le comportement pharmacocinétique de nouveaux radiotraceurs en imagerie moléculaire, il est nécessaire d’approfondir l’analyse réalisée à partir d ’une image par l’ajout d’une mesure dynamique de la radioactivité dans le sang. L’Université de Sherbrooke développe présentement une plateforme microfluidique d’échantillonnage et d’analyse permettant la mesure de la radioactivité du plasma en temps réel. L’objectif du présent projet de maîtrise était de réaliser le composant optoélectronique responsable de la détection des positrons et de l’intégrer à la puce microfluidique. L’option retenue a été l’utilisation de photodiodes PIN en silicium. Un procédé de fabrication, ainsi qu’une série de photomasques ont été développés afin de produire une première itération de prototypes. Les détecteurs ont été conçus de manière à optimiser leur sensibilité en fonction du type de rayonnement à détecter. En effet, la région de détection doit être suffisamment épaisse et sensible pour absorber le maximum de particules énergétiques. Également, il est essentiel de minimiser le courant de fuite en noirceur afin d’obtenir un photocourant directement proportionnel à l’énergie des radiations incidentes. Les caractéristiques électriques obtenues avec les premiers détecteurs ont été démontrées proches des performances de détecteurs commerciaux similaires. De plus, il a été possible d’intégrer un canal microfluidique au substrat contenant les photodiodes et d’en réaliser l’encapsulation sans altérer les performances électriques initiales des détecteurs. Une courbe de l’activité radioactive du l8F a été mesurée, celle-ci se comparant à l’activité théorique associée à ce radioisotope communément utilisé en TEP. Enfin, un spectre en énergie des émissions radiatives du 18F a été mesuré et comparé aux performances de systèmes utilisant des photodiodes commerciales. D a été démontré que le prototype offrait un rapport signal sur bruit similaire aux systèmes basés sur des photodiodes commerciales. Des problèmes durant la fabrication ont cependant compromis les performances de la deuxième itération de détecteurs. Une série de recommandations finales afin d’éviter ces problèmes a été émise suite à la deuxième itération réalisée lors de ce projet de maîtrise.

Recalage multi-modal automatique : technique de multi-résolution parallèle appliquée à la TEP et l'IRM / Multi-modal automatic registration : A parallel multi-resolution approach applied to PET-MRI

Résumé : Le recalage automatique des images issues de la tomographie par émission de positrons (TEP) et de l’imagerie par résonance magnétique (IRM) du petit animal pose un problème difficile à résoudre, tant sur l’aspect de la précision, du taux de réussite et de convergence que sur la rapidité d’exécution. En fait, la plupart des techniques de recalage actuelles sont développées et appliquées aux cerveaux humains, mais ne sont pas aussi efficaces lorsqu’appliquées sur des données animales. L’anisotropie impor¬tante des voxels (résolution fine dans le plan de l’acquisition, mais grande épaisseur de coupe) et la dégradation des images associée à ce type d’acquisition s’additionne au manque d’information d’intensité et de complexité anatomique de ce type de jeu de données. Ce mémoire met l’accent sur les techniques multimodales de recalage automatique et de leurs limites, appliquées particulièrement à la TEP et l’IRM du petit animal. Dans l’article principal présenté dans ce mémoire, nous proposons une mesure qui utilise un recalage multirésolution en parallèle (imbriqué dans la fonction d’énergie) au lieu d’une approche classique de multirésolution séquentielle, influen¬çant directement la procédure du recalage. En combinant les niveaux de basse et haute résolution des images, nous nous assurons une plus grande insensibilité par rapport au bruit, d’une ouverture accrue permettant une meilleure convergence et rapidité d’exécution. L’article démontre que notre nouvelle approche automatique est un algorithme de recalage robuste et efficace avec un taux de réussite élevé. Nous présentons également dans ce mémoire certains détails d’implantation de l’outil, qui a été créé par l’auteur de ce document, reposant sur le recalage classique et la nouvelle méthode décrite dans ce mémoire.||Abstract: Automatic registration of small animal Positron Emission Tomography (PET) and Magnetic Resonance Imaging (MRI) data represents a difficult problem in terms of convergence speed, accuracy and success rate. In fact, most existing registration methods are developed and applied to human brain volumes but these are not as effective for small animal data because of the lack of intensity information in the images and often the large anisotropy in voxel dimensions (very small in-plane resolution and large slice thickness). This master thesis focuses on multi-modal automatic registration techniques and their limitations, especially applied to PET-MRI registration. In the main article of this master thesis, we propose a new registration measure that combines multi-resolution in parallel (in the same energy function) instead of a classic sequential multi-resolution, which influence the procedure of the registration as well. By combining low and high resolution levels of images, we can gain from the low noise sensitivity and aperture at coarse levels and higher contrast and details at higher levels, which helps convergence accuracy and speed. The paper shows that our new approach is therefore an automatic, robust and efficient registration algorithm with a high success rate. We also present in this document some implementation details on the tool which was created by the author of this thesis based on the classic registration and the new approach described in this thesis.

Synthèse et étude de nouveaux précurseurs pour une imagerie bimodale optique/TEP ou TEMP

Résumé Ce travail de thèse en cotutelle a été réalisé à l’Institut de Chimie Moléculaire de l’Université de Bourgogne (France) ainsi qu’au département de Chimie de l’Université de Sherbrooke (Québec). Ces travaux sont consacrés à la modification de la structure des BODIPY en vue d'une application in vivo pour l'imagerie bimodale optique/TEP ou TEMP. Ces modifications concernent l'hydrosolubilisation des fluorophores, mais aussi le décalage des domaines d’absorption et d’émission vers la fenêtre thérapeutique, zone dans laquelle la lumière est moins atténuée par les tissus biologiques. Des solutions permettant la bioconjugaison des fluorophores sur un dérivé de la bombésine, peptide permettant de cibler des récepteurs membranaires surexprimés dans certains types de cellules cancéreuses, sont également présentées. Enfin, une stratégie d’introduction d’un radioélément sur le BODIPY est décrite. Elle implique l’introduction sur l’atome de bore d’un groupe partant permettant sa radiofluoration. Différents dérivés de BODIPY ont également été utilisés pour la construction d’édifices multichromophoriques à transfert d’énergie. Ces travaux sont décrits dans la seconde partie de cette thèse. Ils comprennent notamment des études photophysiques permettant de mettre en évidence le transfert d’énergie photoinduit donneur-accepteur entre différents chromophores Dans les différents système élaborés, de type BODIPY-pophyrine et BODIPY-corolle, le BODIPY est alternativement donneur, ou accepteur d’énergie.

Continuum description of deformable organs based on tetrahedral meshes : application to dosimetry and imaging for hadron therapy / Représentation continue des organes déformables basée sur des maillages tétraédriques : application à la dosimétrie et l'imagerie pour l'hadronthérapie

Dans le cadre du projet européen ENVISION (2010-2014) et en collaboration avec l'équipe CAS-PHABIO de l'IPNL, cette thèse constitue une contribution méthodologique et technique dans le domaine de la dosimétrie et de l'imagerie de contrôle par émission des positons (TEP) pour les organes en mouvement. Les méthodes actuelles utilisent le recalage déformable d'images CT pour estimer le mouvement des organes internes. Le recalage déformable permet d'estimer le déplacement de chaque voxel d'une image à une autre. La dose radio-thérapeutique ainsi que l'activité TEP sont accumulées sur des voxels. Ces approches ont des difficultés quand il s'agit de prendre en compte la variation de densité à l'intérieur des organes et l'aspect non-répétitif du mouvement respiratoire. Les travaux antérieurs de l'équipe ont permis de développer un premier modèle biomécanique complet du système respiratoire qui, corrélé avec des signaux externes, pourrait prendre en compte la variabilité du mouvement respiratoire. Cette thèse présente une approche qui permet d'intégrer un tel modèle biomécanique dans un système de planification de traitement pour l'hadronthérapie. Dans cette thèse, nous avons choisi d'investiguer de près l'utilisation des maillages tétraédriques déformables dans la dosimétrie et la reconstruction d'images TEP afin d'estimer les avantages et inconvénients de ce type de géométrie. En conclusion, notre approche peut être utilisée avec n'importe quel modèle de déformation basé sur une géométrie tétraédrique et dont le mouvement est décrit par le déplacement des nœuds des maillages et donc contrairement aux méthodes basés images, notre approche n'est pas nécessairement dépendante de l'existence des images internes à tout moment. Dans le futur, les méthodes développées dans cette thèse pourraient être utilisées avec un modèle biomécanique complet du système respiratoire afin de quantifier, par exemple, les effets de la variabilité de la respiration sur le dépôt de dose / Respiratory-induced organ motion is a technical challenge to nuclear imaging and to charged particle therapy dose calculations for lung cancer treatment in particular. Internal organ tissue displacements and deformations induced by breathing need to be taken into account when calculating Monte Carlo dose distributions as well as when performing tomographic reconstructions for PET imaging. Current techniques based on Deformable Image Registration (DIR) cannot fully take into account the density variations of the tissues nor the fact that respiratory motion is not reproducible. As part of the ENVISION (2010-2014) European project, in collaboration with the CAS-PHABIO team from IPNL (the Nuclear Physics Institute from Lyon), this PhD project presents a methodological contribution to physical dose calculations and PET-based treatment verification for hadron therapy in the case of moving tumours. Contrary to DIR-based methods where motion is described by relative voxel displacement, each organ is represented as a deformable grid of tetrahedra where internal motion is described by mesh vertex transformations calculated using continuum mechanics. First, this PhD project proposes a new method to calculate four dimensional dose distribution over tetrahedral meshes, which are deformed using biomechanical modeling based on Finite Element Analysis (FEA). The second part of the PhD is focused on motion compensation for PET image reconstruction using deformable tetrahedral meshes

Ceramic Membrane combined with Powdered Activated Carbon (PAC) or Coagulation for Treatment of Impaired Quality Waters

Ceramic membranes (CM) are robust membranes attributed with high production, long life span and stability against critical conditions. While capital costs are high, these are partially offset by lower operation and maintenance costs compared to polymeric membranes. Like any other low-pressure membrane (LPM), CM faces problems of fouling, low removal of organic matter and poor removal of trace organic compounds (TOrCs). Current pretreatment approaches that are mainly based on coagulation and adsorption can remove some organic matter but with a low removal of the biopolymers component which is responsible for fouling. Powdered activated carbon (PAC) accompanied with a LPM maintains good removal of TOrCs. However, enhanced removal of TOrCs to higher level is required. Submicron powdered activated carbon (SPAC), obtained after crushing commercial activated carbon into very fine particle, and novel activated carbon (KCU 6) which is characterized with larger pores and high surface area were employed. A pre-coating approach, which provides intimated contact between PAC and contaminants, was adopted for wastewater and (high DOC) surface water treatment. For seawater, in-line coagulation with iron III chloride was adopted. Both SPAC and KCU 6 showed good removal of biopolymers at a dose of 30 mg/L with > 85 % and 90 %, respectively. A dose of 40 mg/L of SPAC and 30 mg/L KCU 6 pre-coats were successful used in controlling membrane fouling. SPAC is suggested to remove biopolymers by physical means and adsorption while KCU 6 removed biopolymers through adsorption. Both KCU 6 and SPAC attained high removal of TOrCs whereas KCU 6 showed outstanding performance. Out of 29 TOrCs investigated, KCU 6 showed > 87 % TOrCs rejection for 28 compounds. In seawater pretreatment, transparent exopolymer particles (TEP) were found to be an important foulant. TEP promoted both reversible and irreversible fouling. TEP are highly electronegative while alumina CM is positively charged which support strong TEP–alumina binding. The influence of TEP fouling was minimized with a low dose of 0.5 – 1 mg/l Fe coagulant. Bacteria were almost completely removed; Silt Density Index (SDI) value was maintained to 2 % per minute and a constant permeate turbidity of 0.05 NTU was achieved.

Effectiveness of seawater reverse osmosis (SWRO) pretreatment systems in removing transparent exopolymer particles (TEP) substances

Transparent exopolymer particles (TEP) have been reported as one of the main factors of membrane fouling in seawater reverse osmosis (SWRO) process. Research has been focused on algal TEP so far, overlooking bacterial TEP. This thesis investigated the effects of coagulation on removal of bacterial TEP/TEP precursors in seawater and subsequent reduction on TEP fouling in ultrafiltration (UF), as a pretreatment of SWRO. Furthermore, the performance of pretreatment (coagulation + UF) has been investigated on a bench-scale SWRO system. TEP/TEP precursors were harvested from a strain of marine bacteria, Pseudoalteromonas atlantica, isolated from the Red Sea. Isolated bacterial organic matter (BOM), containing 1.5 mg xanthan gum eq./L TEP/TEP precursors, were dosed in Red Sea water to mimic a high TEP concentration event. Bacterial TEP/TEP precursors added to seawater were coagulated with ferric chloride and aluminum sulfate at different dosages and pH. Results showed that ferric chloride had a better removal efficiency on TEP/TEP precursors. Afterwards, the non-coagulated/coagulated seawater were tested on a UF system at a constant flux of 130 L/m2h, using two types of commercially available membranes, with pore sizes of 50 kDa and 100 kDa, respectively. The fouling potential of coagulated water was determined by the Modified Fouling Index (MFI-UF). Transmembrane pressure (TMP) was also continuously monitored to investigate the fouling development on UF membranes. TEP concentrations in samples were determined by the alcian blue staining assay. Liquid chromatography-organic carbon detection (LC-OCD) was used to determine the removal of TEP precursors with particular emphasis on biopolymers. Finally, SWRO tests showed that TEP/TEP precursors had a high fouling potential as indicated by MFI-UF, corresponding to the TMP measurements. Coagulation could substantially reduce TEP/TEP precursors fouling in UF when its dosage was equal or higher than 0.2 mg Fe/L. The flux decline experiments showed that coagulation + UF pretreated water had a smaller fouling potential than MF pretreated water. This thesis also provides useful and practical information on controlling bacterial TEP/TEP precursors fouling in UF and RO systems.
