The thesis discusses the transformation of southern European countries, Italy and Spain, from traditionally countries of emigration into countries of immigration. It starts from historical migration flows which result in so-called Mediterranean model of immigration characteristic for the region in the last two decades. The paper analyses the composition of immigrants in both countries, focusing on the ethnic composition, the motives of residence and employment status compared with overall employment in the EU context. Since the south European countries concerned are significantly affected by inflows of migrants by sea in recent years, the thesis also focuses on problematic areas of the Mediterranean, detention centers and cooperation with source countries. The main aim of this work is to analyze how these two countries handle the phenomenon of immigration and what recourse did they choose in creating their own immigration policies towards both legal and illegal immigration. The second objective is then to bring some comparison of the two countries in the approaches to integration of immigrants.

Population genetics of the Mediterranean fruit fly Ceratitis capitata in the Western Cape Province, South Africa : invasion potential and dispersal ability

Thesis (MSc)--Stellenbosch University, 2011. / ENGLISH ABSTRACT: The Mediterranean fruit fly (medfly), Ceratitis capitata, is a highly invasive species throughout the world and considered as one of the most successful agricultural and economical pests. The increase of global trade in fruit and human travel combined with the biology of the medfly has allowed the species to spread from its proposed Afrotropical origin, to a number of locations throughout the world. In the Western Cape various control strategies have been implemented to control medfly populations, including insecticides and more environmentally-friendly techniques such as the Sterile Insect Technique (SIT). In order to be effective, however, an SIT program requires some knowledge of the population structure and the movement of individuals between pest-occupied sites. The identification of sites from which re-invasion is most likely to occur and knowledge regarding the primary routes through which pests are likely to re-establish are critical to ensure successful SIT programmes. To provide this important information to SIT and area-wide pest control programs in South Africa, sampling at two different spatial scales (regional- and fine-scale) in South Africa was undertaken. Regional scale sampling was done at 13 locations in the Western Cape and fine scale sampling was done at 13 locations within the Ceres-valley. All individuals were genotyped at 11 polymorphic microsatellite markers and selected individuals from the regional scale were sequenced for the mitochondrial gene COI. Our results show that populations at regional- and fine-scale in the Western Cape are characterized by high levels of genetic diversity (HEregional = 0.805; HEfine = 0.803). Little or weak population differentiation was detected at the regional- and fine-scales, suggesting overall high levels of gene flow among sampling locations. These findings were supported by coalescent based methods indicating sufficient levels of gene flow to prevent population differentiation between neighbouring (200m) and distant (350km) populations. However, natural dispersal in C. capitata has been shown to rarely exceed 10 km. As such, high levels of gene flow between distant populations are more likely the result of humanmediated dispersal, linked to the movement of fresh produce within South Africa. This high level of gene flow has important implications for pest management practices, as my results suggests that area-wide pest management should be undertaken at a regional scale, rather than on a farm or valley scale. My results are placed within a management framework, and I argue for more stringent control when fruit are transported within South Africa. Of particular interest for future studies is the investigation of gene flow at broader spatial scales (i.e. the whole of South Africa) and a comparison of the genetic diversity, population differentiation and gene flow patterns of C. capitata with that of Ceratitis rosa will be important to establish a successful pest management strategy in South Africa. / AFRIKAANSE OPSOMMING: Die Mediterreense vrugtevlieg (medvlieg), Ceratitis capitata, is ‘n indringerspesie wêreldwyd en word beskou as een van die mees suksesvolle ekonomiese en landbou peste. My resultate toon dat bevolkings op beide skale gekarakteriseer word deur hoë vlakke van genetiese diversiteit (HEstreeks = 0.805; HEplaaslik = 0.803) en geen of swak bevolkings differensiasie. Hierdie resultate dui daarop dat daar hoë vlakke van geenvloei tussen bevolkings is. Hierdie bevindinge word verder ondersteun deur metodes gebaseer op die statistiese eienskappe van die genealogiese verhouding tussen allele onder sekere mutasie en demografiese modelle, wat voldoende vlakke van geenvloei aandui tussen nabye (200m) sowel as verafgeleë (350km) bevolkings om bevolkings differensiasie te verhoed. Natuurlike beweging in C. capitata is egter selde meer as 10 km, sodanig kan die hoë vlakke van geenvloei toegeskryf word aan die verspreiding van individue met menslike hulp, spesifiek in die vervoer van varsprodukte in Suid-Afrika. Hierdie hoë vlak van geenvloei het verreikende implikasies vir pes beheer praktyke, omdat my resultate voorstel dat area-wye pes beheer onderneem moet word op ‘n streeks-skaal eerder as op ‘n plaas-tot-plaas of vallei wye area. Ek plaas my resultate in ‘n bestuursraamwerk, waarin ek streng beheer van vrugtevervoer in Suid-Afrika beklemtoon. Verdere navorsing moet fokus op die ondersoek van geenvloei op ‘n landswye skaal (hele Suid-Afrika) in C. capitata, sowel as die vergelyking van die genetiese diversiteit, bevolkings differensiasie en geenvloei patrone van C. capitata met die van Ceratitis rosa om ‘n suksesvolle pes beheer strategie vir Suid-Afrika te formuleer.

Modélisation de la variabilité climatique de la circulation et des masses d'eau en Méditerranée : impacts des échanges océan-atmosphère.

Cette thèse a pour but de progresser sur des points essentiels concernant le réalisme de la représentation de la formation et du trajet des masses d'eau en Mer Méditerranée, ainsi que de leur variabilité. A cet effet, plusieurs modèles océaniques régionaux de la Méditerranée, de résolutions horizontales différentes, sont développés et utilisés. Une configuration réaliste permettant de représenter la variabilité interannuelle des conditions aux limites de ces modèles (atmosphère, océan Atlantique, fleuves, mer Noire) est utilisée pour réaliser des simulations à long terme des 50 dernières années en Méditerranée. Deux événements rares, caractérisant la variabilité décennale en Méditerranée, sont étudiés : l'Eastern Mediterranean Transient (EMT) et la Western Mediterranean Transition (WMT). L'EMT est la période, au début des années 1990, pendant laquelle le site principal de formation d'eau dense dans le bassin oriental méditerranéen est passé du sous-bassin Adriatique au sous-bassin Egée. La capacité des simulations à long terme à reproduire la séquence d'événements composant l'EMT est tout d'abord démontrée. Parmi les éléments de préconditionnement et de déclenchement de l'EMT suggérés dans la littérature, nous montrons que les facteurs essentiels sont les flux hivernaux intenses au-dessus du sous-bassin Egée pendant les hivers 1992 et 1993. Le réalisme de la formation et de la propagation de l'eau crétoise profonde (Cretan Deep Water, CDW) pendant l'EMT est ensuite analysé dans les simulations de référence et de sensibilité. La propagation de la CDW au fond de la Méditerranée orientale n'est reproduite qu'avec des conditions atmosphériques modifiées. La WMT a commencé à l'hiver 2005 dans le Golfe du Lion, pendant lequel a été formé un volume très important d'eau profonde ouest-méditerranéenne (Western Mediterranean Deep Water, WMDW), caractérisée par une température et une salinité inhabituellement élevées. Les simulations reproduisent l'intensité de la convection profonde dans le Golfe du Lion pendant l'hiver 2005, qui est est due à la forte perte de flottabilité en surface. Elles indiquent également que des tourbillons cycloniques profonds, d'une centaine de kilomètres de diamètre, sont responsables du transport rapide de la nouvelle WMDW vers le Sud. Puis, les simulations à long terme permettent de replacer la WMT dans la variabilité décennale de la Méditerranée nord-occidentale. Elles montrent que l'EMT a potentiellement doublé le volume de nouvelle WMDW formé en 2005 dans le Golfe du Lion, mais qu'il n'est pas responsable de la température et de la salinité élevées de la nouvelle WMDW. Ces caractéristiques inhabituelles sont dues à l'absence de convection intense dans le Golfe du Lion pendant les années 1990, ce qui a favorisé l'accumulation de sel et de chaleur dans la Méditerranée nord-occidentale.

Small-island interactions : pottery from Roman Malta

This thesis is an investigation of Roman pottery from the Maltese islands from the 1st century BC to the mid-4th century AD, and how pottery can help assess Malta's economic role in the wider central Mediterranean region. The archipelago's locally produced vessels, its range of ceramic exports, and the quantification of the types of amphorae, fine, and cooking wares the islands imported, were studied and the data were used to compare with the pottery available from the small islands of Pantelleria, Lampedusa and the Kerkennah isles. The aim is to revisit the theme of the economic role of the Maltese islands and other similar-sized islands in the region by moving away from the tradition of unilateral and monographic narratives, which more often than not, omit the wealth of information that can be garnered from pottery. In the first instance, a detailed study of three complete and new ceramic assemblages, including amphorae, fine, cooking and coarse wares, was undertaken. The opportunity to quantify identifiable imports and compare them with local products - the first of its kind for fine, cooking and coarse wares - provided valuable proxy data for comparing Malta with neighbouring islands and centres, and demonstrated what proportion of ceramic vessels were locally supplied, and how these changed over time. These data were also fed into a series of network analyses, which plotted the common pottery links shared between small-island and mainland sites in the region. The analyses were interpreted in conjunction with a critique of existing pottery quantification methods, and the potential acceptance for utilising all known pottery data irrespective of the quality and quantity of the published data available. Most importantly, the import trends obtained from this study were incorporated into the existing narrative of how small islands and their local industries featured in the central Mediterranean's regional economy, highlighting the types of archaeologically visible industries that existed; how these developed symbiotically alongside other larger supply networks; and what effect this might have had on the integration of small islands in the Roman Mediterranean.

Fraction primaire et secondaire de l'aérosol organique : méthodologies et application à un environnement urbain méditerranéen, Marseille

La mise en place de politiques efficaces visant à la réduction des niveaux de concentrations en PM exige la connaissance préalable des sources primaires et secondaires de l’aérosol organique, une fraction majoritaire des PM demeurant encore mal appréhendée. Les travaux réalisés au cours de cette thèse s’inscrivent dans le cadre du projet FORMES qui avait pour principal objectif d’évaluer et de contraindre les principales méthodes de quantification de l’influence des différentes sources de la fraction organique de l’aérosol en milieu récepteur et d’en optimiser les procédures. La caractérisation physico-chimique de l’aérosol s’est articulée autour de deux campagnes de mesures intensives de 15 jours chacune dans deux environnements urbains très contrastés : Marseille en été, et Grenoble en hiver. Ce travail de thèse s’est concentré sur le cas de Marseille, un environnement très complexe, combinant une activité photo oxydante très intense à un ensemble d’émissions primaires qui incluent les sources industrielles et les émissions par les bateaux.L’analyse CMB appliquée sur le cas de Marseille a montré que les sources primaires sont dominées par les émissions véhiculaires contribuant à 17 % du carbone organique (OC). Bien que les sources industrielles contribuent faiblement à la masse de l’aérosol (2.5 % de l’OC), ces émissions contrôlent les concentrations des HAP et de certains métaux lourds. Ces sources contribuent également, en moyenne sur la période, à environ 30% du nombre des particules ultrafines (Dp<50 nm), ce qui augmente probablement leurs effets sanitaires. Contrairement à Grenoble, où la combustion de bois est une source prépondérante (environ 70 % de l’OC), à Marseille ces émissions ne constituent qu’une source minoritaire, contribuant à 0.8 % de l’OC. Toutefois, la principale information révélée par la déconvolution de sources par CMB est que les sources primaires considérées ne permettent d’expliquer, que 22 % de l’OC mesuré ; 78 % du carbone restant non expliqué. Cette fraction est associée majoritairement à l’aérosol organique secondaire (SOA). En combinant les résultats CMB aux mesures 14C, il a été montré que plus que 70% de cet aérosol est très vraisemblablement d’origine biogénique. En conséquence, les contributions du SOA formé à partir de l’isoprène, l’&#945;-pinène et le &#946;-caryophyllène ont été examinées moyennant une approche basée sur les marqueurs de cet aérosol. Le SOA issu de ces précurseurs a été estimé comme contribuant uniquement à 4.3 % de l’OC, laissant une grande fraction de ce carbone non-attribuée. Cette sous-estimation est la conséquence de trois causes : (i) les incertitudes associées à l’approche utilisée, (ii) des précurseurs biogéniques non-considérés et (iii) le vieillissement de l’aérosol secondaire au cours de son transport dans l’atmosphère, comme suggéré par les mesures d’organosulfates et de la fraction polycarboxylique de type HULIS. Cette dernière fraction peut contribuer à près de 40% de l’OC non-attribué. / Tougher particulate matter regulations around the world and especially in Europe point out the need of source apportionment studies in order to better understand the different primary and secondary sources of organic aerosol, a major fraction of particulate matter that remains not well constrained. The work carried out in this thesis takes part of the FORMES project whose main objective is the source apportionment of the organic aerosol using different approaches, including mainly CMB modelling, AMS/PMF and radiocarbon (14C) measurements. The aerosol characterisation was performed within two intensive field campaigns conducted in two contrasted urban environments: Grenoble during winter and Marseille during summer. The present work focuses on the Marseille case study that presents a particularly complex environment, combining an intense photochemistry to a mixture of primary emissions including shipping and industrial emissions. Primary organic carbon (POC) apportioned using CMB modelling contributed on average for only 22% and was dominated by vehicular emissions accounting on average for 17% of OC. Even though, industrial emissions contribute for only 2.3% of the total OC, they are associated with ultrafine particles and high concentrations of Polycyclic Aromatic Hydrocarbons and heavy metals such as Pb, Ni and V, which most likely relate them with acute health outcomes. Whereas in Grenoble the organic aerosol was dominated by wood burning smoke (70% of OC), this source was negligible in Marseille contributing for less than 1% of OC. The main result from this source apportionment exercise is that 78% of OC mass cannot be attributed to the major primary sources and remains un-apportioned; this fraction is mostly associated with secondary organic aerosol. Radiocarbon measurements suggest that more than 70% of this fraction is of modern origins, assigned predominantly to biogenic secondary organic carbon (BSOC). Therefore, contributions from three traditional BSOC precursors, isoprene, &#945;-pinene and &#946;-caryophellene, were considered using a marker based approach. The aggregate contribution from BSOC derived from these precursors was estimated at only 4.2% of total OC. As a result, these estimates underpredict the high loading of OC. This underestimation can be associated with (i) uncertainties underlying the marker-based approach, (ii) presence of other SOC precursors and (iii) further processing of fresh SOC, as indicated by organosulfates (RSO4) and HUmic LIke Substances (HULIS) measurements. This HULIS can contribute up to 40% of the unattributed OC.

Conidia of the pathogen, which is dispersed by wind, water droplets and by insects, can penetrate the intact grape berry cuticle, but disease expression occurs only under predisposing conditions. Since relatively high infection rates often occur in vineyards, predisposing factors must play a fundamental role in primary infection and subsequent disease occurrence. Insects can play a very important role in this regard by depositing inocula at wound sites during feeding and by providing fresh wounds during their oviposition and feeding activities. The aim of this study was (i) to determine the potential of the Mediterranean fruit fly to transfer B. cinerea and other bunch and fruit rot fungi in natura, (ii) to investigate the transport, deposition and subsequent disease expression on grape berries in vitro, and (iii) to investigate fruit fly activities and the nature of deposited conidia and mycelia of B. cinerea by aid of digital photography and epifluorescence microscopy, respectively. Two Sensus fruit fly traps containing the para-pheromone, Capilure, were installed in orchards and five neighboring vineyards on four farms in the Stellenbosch region. Ceratitis fruit flies were collected weekly, identified and counted to determine the fluctuations in fruit fly population. Following field collection, the fruit flies were plated on Kerssies' B. cinerea selective medium and the number of flies yielding the pathogen was recorded. Two fruit fly species, C. capitata and C. rosa, were captured during the study period. Ceratitis rosa numbers comprised only 1% of the total number of fruit flies captured. Ceratitis capitata numbers, and the percentage B. cinerea contaminated flies generally increased after harvest in the different orchards and vineyards. Following harvest, the percentage flies yielding B. cinerea was higher in vineyards compared to orchards. Furthermore, in each vineyard an increase in percentage B. cinerea contaminated fruit flies was preceded by a corresponding increase in its neighboring orchard. The levels of both Penicillium and Alternaria contaminated fruit flies stayed high throughout the investigation period, especially after harvest of the orchard cultivars. Low incidence of Aspergillus, Mucor and Rhizopus spp. were recorded on C. capitata. These findings suggest that the Mediterranean fruit fly may play an important role in the dispersal of inocula of fungi associated with postharvest decay from early-maturing stone fruit orchards to mid- and late-maturing wine grape vineyards, and in disease induction under conditions unfavourable for natural infection. Three experiments were conducted to determine the potential of fruit flies in provoking B. cinerea decay. In the first experiment, transport of conidia and disease expression were investigated on rachis segments bearing unwounded berries only. In the second experiment, the effect of wounding on disease expression was investigated. In the third experiment, the effect of inoculum type (mycelia and conidia) on transportation and disease expression was investigated on rachis segments bearing unwounded berries, and on segments with wounded berries. The table grape cultivar, Dauphine, and the wine grape cultivar, Shiraz, were used at véraison, two weeks before harvest and harvest, and the transport studies were conducted in ethanol-disinfected perspex cages. Disease expression was studied in dry (~56% RH), ethanol-disinfected perspex chambers incubated at 22°C. The isolations from berries revealed that the flies deposited, without preference, high amounts of B. cinerea at various positions on the grape berry's surface. The freezing studies showed that the deposited conidia germinated and penetrated the berry skin at various positions. However, B. cinerea developed more often at the pedicel end than on the cheek or style end, which indicated a peculiar interaction between B. cinerea, the fruit fly and host tissue at this part of the berry. This phenomenon was substantiated by the finding that B. cinerea also developed more often at the pedicel end of berries that were not frozen. Further evidence for this interaction was found on intact berries exposed to flies that carried mycelia after feeding on berries without sporulating colonies of the pathogen, but showing symptoms of slippery skin. Significantly more decay developed on wounded berries compared to the unwounded berries and more so at the wound site. In addition, female fruit flies were responsible for significantly more decay development than male fruit flies. The study thus proved that the Mediterranean fruit fly can promote B. cinerea disease development under conditions unfavorable to natural infection. The activities of the Mediterranean fruit fly, Ceratitis capitata, on grape berries were monitored by aid of digital photography. In addition, the deposition of conidia and mycelia of Botrytis cinerea at three sites (pedicel end, cheek and style end) on the grape berry, germination of the fungal structures after dry (±56% RH) and moist (±93% RH) incubation and wounds inflicted during ovipositioning were examined with an epifluorescence microscope. The observations revealed that the fruit fly's activities were generally restricted to the grape berry. They visited the grape berry cheek more often, but visitations to the pedicel end of berries increased substantially from véraison to harvest, indicating the possibility of nutrient leakages at this site. Microscopy revealed that the flies deposited conidia singular, in feeding packages and in faecal excrements on the berry surface. The conidia in feeding packages were ensheathed by salivical fluids and occurred in clusters of 10 to 50 conidia. An average of 60% of the conidia in feeding packages germinated under dry conditions (±56% RH). Conidia that passed through the intestinal tract of the fruit fly and that were deposited in faecal excrements were deformed and low in viability. These conidia did not occur in cluster format, but were proportionally spread with the faeces on the surface of the grape berry. Conidia that were deposited singular and in faecal excrements did not germinate unless incubated under moist conditions (± 93% RH). Wounds inflicted by female fruit flies during ovipositioning were most frequently observed on the cheek. This predisposition to B. cinerea infection of grape berries by the activities of fruit flies, suggested an important role for the flies in the initiation of Botrytis bunch rot epidemics in vineyards. / AFRIKAANSE OPSOMMING: DIE ROL VAN DIE MEDITERREENSE VRUGTEVLIEG, CERATITIS CAPITATA, IN BOTRYTIS CINEREA TROSVERROTTING VAN DRUIWE Botrytis-trosverrotting van druiwe word deur Botrytis cinerea Pers. :Fr. veroorsaak. Ceratitis-vrugtevlieë is weekliks versamel, geïdentifiseer en getel om fluktuasies in die vrugtevliegpopulasie te bepaal. Na die veldversameling is die vrugtevlieë op Kerssies se B. cinerea-selektiewe medium uitgeplaat. Gedurende die studie is twee spesies vrugtevlieë, C. capitata en C. rosa, gevang. Na oesstyd het die aantal Ceratitis-vrugtevlieë en die persentasie vrugtevlieë, besmet met B. cinerea, in die verskillende boorde en wingerde toegeneem. Na oestyd was die persentasie vrugtevlieë wat B. cinerea gedra het, hoër in die wingerde as in die boorde. Elke toename in die persentasie B. cinerea-besmette vrugtevlieë in 'n wingerd is voorafgegaan deur 'n ooreenkomstige toename in die aangrensende vrugteboord. Die aantal vrugtevlieë besmet met Penicillium en Alternaria spp. het tydens die navorsingstydperk deurgaans hoog gebly, veral nadat die vrugteboord-kultivars geoes is. Die voorkoms van Aspergillus-, Mucor- en Rhizopus spp. op Ceratitis-vrugtevlieë was deurgaans laag. Hierdie bevinding wys daarop dat vrugtevlieë 'n belangrike rol speel in die verspreiding van swarninokula, wat met na-oes verrotting geassosieer word, van vroegrypwordende steenvrugteboorde na mid- en laatrypwordende wyndruifwingerde. Drie eksperimente is in vitro onderneem om vrugtevlieë se potensiaal om B. cinereaverrotting te veroorsaak te bepaal. In die eerste eksperiment is ragi met slegs ongewonde korrels gebruik om die oordrag van konidia en siekte-ontwikkeling te ondersoek. In die tweede eksperiment is die effek van verwonding op siekte-ontwikkeling ondersoek. In die derde eksperiment is die effek van inokulumtipe (miselia en konidia) op verspreiding en siekte-ontwikkeling ondersoek deur ragis-segmente met gewonde korrels sowel as ragissegmente met ongeskonde korrels te gebruik. Die tafeldruif-kultivar Dauphine en die wyndruif-kultivar Shiraz, by kleurbreuk, twee weke voor oes en by oestyd, is in die eksperimente gebruik. Die oordragstudies is in etanol-ontsmette perspex-hokke uitgevoer. Siekte-ontwikkeling is bestudeer in droeë (±56% RH), etanol-ontsmette perspex-kamers en geinkubeer by 22°C. By ondersoek is gevind dat vlieë, sonder voorkeur, groot hoeveelhede B. cinerea op verskeie dele op die druiwekorrel-oppervlak deponeer. Bevriesingstudies het aangetoon dat die gedeponeerde konidia op verskeie dele van die korrelontkiem en die skil binnedring. Botrytis cinerea het egter meer dikwels by die korrelsteelkant as by die stempelkant, of op die wang, ontwikkel. Hierdie bevinding het 'n eiesoortige interaksie tussen B. cinerea, die vrugtevlieg en gasheerweefsel by die korrelsteelkant van die korrel aangetoon. Die verskynsel is gestaaf deur die bevinding dat B. cinerea ook meer dikwels by die korrelsteelkant van die korrels wat nie gevries is nie, ontwikkel het. Verdere bewys van hierdie interaksie is gevind by ongeskonde korrels wat aan die vlieë wat miselia gedra het blootgestel is. Die siekte het beduidend meer dikwels op gewonde as ongewonde korrels en verder aansienlik meer dikwels op die wondoppervlakte ontwikkel. Dit was ook duidelik dat vroulike vrugtevlieë baie meer vir verrotting verantwoordelik was as manlike vrugtevlieë. Die studie bewys dus dat Mediterreense vrugtevlieë die ontwikkeling van B. cinerea kan bevorder in omstandighede wat ongunstig is vir natuurlike infeksie. Die aktiwiteite van die Mediterreense vrugtevlieg C. capitata op die druiwekorrels is met behulp van digitale fotografie waargeneem. Verder is die deponering van konidia en miselia van B. cinerea op die verskillende dele (korrelsteelkant, wang en stempelkant) van die korrel, ontkieming van die swamstrukture na droeë (±56% RH) en nat (±93% RH) inkubasie en wonde wat tydens oviposisionering veroorsaak is, met epifluoressensie-mikroskopie ondersoek. Die waarnemings het onthul dat die vrugtevlieg se aktiwiteite gewoonlik tot die druiwekorrel beperk is. Hulle het korrelwange meer dikwels besoek. Besoek aan die korrelsteelkant het aansienlik toegeneem van kleurbreuk tot oestyd, wat op die moontlikheid van voedingstof-lekkasie by die deel aandui. Mikroskoopstudies het aangedui dat vlieë konidia enkel, in voedingspakkies en in fekale uitskeidings op die korreloppervlakte deponeer. Die konidia in die voedingspakkies is deur speekselvloeistof omhul en het in groepe van 10 tot 50 konidia voorgekom. Gemiddeld 60% van die konidia in voedingspakkies het in droeë omstandighede (±56% RH) ontkiem. Konidia wat deur die spysverteringskanaal van die vrugtevlieg gegaan het en in die fekale ekskresie gedeponeer is, was misvorm en het lae lewensvatbaarheid gehad. Laasgenoemde konidia was nie in groepe gedeponeer nie, maar is proporsioneel met die feces op die oppervlak van die druiwekorrel versprei. Konidia wat enkel en in feces gedeponeer is, het nie ontkiem nie, tensy toestande vogtig (±56% RH) was. Wonde wat deur die vroulike vrugtevlieë tydens oviposisionering veroorsaak is, is meer dikwels op die wang van die korrelopgemerk. Hierdie predisposisie van druiwekorrels tot B. cinerea-infeksie, meegebring deur die aktiwiteit van die vrugtevlieg, dui daarop dat die rol wat die vrugtevlieg in die inisiëring van Botrytis trosverrottingepidemies in wingerde speel, van beduidende belang is.

The effects of phosphorus (P) deficiency on growth and nitrogen fixation of Virgilia trees from the Cape Floristic Region (CFR)

Thesis (MSc)--Stellenbosch University, 2013. / ENGLISH ABSTRACT: The aim of this study was to determine how P nutrition affects biological nitrogen fixation (BNF) via effects on the N2-fixing bacteria in the nodules of Virgilia species native to the Cape Floristic Region (CFR), South Africa. This was evaluated in 3 separate studies: The first study aimed to determine how phosphorus deficiency affects N nutrition of two legume tree species from the Mediterranean Fynbos ecosystem. This study showed that during prolonged P deficiency, V. divaricata maintained a constant biomass, while V. oroboides showed a decreased biomass. V. oroboides showed a decrease in nutritional concentrations, which resulted in the decrease of symbiotic nitrogen fixation (SNF). Both plants utilized atmospheric N more efficiently per nodule under P deficiency. Maximum photosynthesis decreased in V. oroboides, while V. divaricata maintained its photosynthesis. Both species also had greater carbon construction costs during P deficiency. V. divaricata showed clear adaptive features during P-deficiency, as it maintained its growth respiration. The two legume species appear to have different adaptations to P deficiency, which may influence their performance and distribution in their naturally low P environment. The second study aimed to evaluate if soil environmental conditions and mineral nutrient concentration play a major role in microbial communities in plant rhizosphere and nodulation during N2 fixation in legumes. Therefore this study firstly aimed to determine the composition of the N2 fixing bacterial population in the rhizosphere and nodules of V. divaricata. Secondly, it aimed to determine the contribution of these bacteria to N2 fixation during conditions of P deficiency in the Fynbos environment. In the study, the effects of phosphate (P) nutrition on N2 fixing bacterial community structures in Virgilia divaricata rhizosphere and nodules were examined in a pot experiment. V. divaricata were germinated in Fynbos soil as natural inoculum, transferred to clean sand cultures and supplied with 500 μM P and 5 μM P. The N2 fixing bacterial communities in the rhizosphere and nodules were examined based on the PCR-DGGE banding patterns of 16S rDNA and sequencing methods. The GenBank blast results revealed that V divaricata was efficiently nodulated by a wide range of root-nodule bacterial strains, including Burkholderia phytofirmans, Burkholderia sp. and Bradyrhizobium sp. during low P supply. The 15N natural abundance data also confirmed that 40-50% of the N nutrition was acquired through symbiotic N2 fixation. This is not only evidence of nodulation, but also an indication of the adaptation of a range of N2 fixing bacterial strains / species to the nutrient poor, sandy, acidic soil of the Mediterranean-type ecosystems of the Fynbos. The third study examined the physiological effects, such as N2 fixation parameters, plant dependence on N2 fixation, N preference, legume plant growth, carbon costs and amino acid biosynthesis during P deficiency and mineral N supply as NH4NO3 in a slow-growing, Fynbos legume tree, Virgilia divaricata. Continued application of NH4NO3 to the legume plant showed a greater increase in plant dry matter compared to plants with two nitrogen sources (mineral N and atmospheric N2) or plants that relied on atmospheric N2 fixation. Carbon construction costs were more pronounced in plants supplied with two N sources and during P deficiency. Maximum photosynthetic rates per leaf area increased during prolonged P deficiency, irrespective of the N sources. Though plants nodulated, plant dependence on N2 fixation decreased with the addition of NH4NO3. Roots and nodules of the P deficient plants showed an increase in asparagine content, most strikingly so in plants treated with a single source of N. These studies reveal that different legume species of the same genus, may employ contrasting adaptations in order to maintain N nutrition under P deficiency. / AFRIKAANSE OPSOMMING: Die doel van hierdie studie was die bepaling van die wyse waarop fosfaat (P) voeding die biologiese stikstof binding (BNF) deur middel van die effek op N2-bindingsbakterië in die wortelknoppies van Virgilia spesies wat inheems tot die Kaap floraryke area (CFR), Suid Afrika is, affekteer. Drie aparte eksperimente is uitgevoer om die doel te evalueer: Die eerste studie het gepoog om te bepaal hoe 'n fosfaat tekort N voeding van twee peulplant spesies van die Mediterreense Fynbos ekosisteem affekteer. Hierdie studie het getoon dat V. divaricata 'n konstante biomassa tydens verlengde P tekort behou, terwyl V. oroboides ‟n verlaagde biomassa getoon het. Die N2-bindende bakteriële gemeenskappe in die risofeer en wortelknoppies is op grond van die PCR-DGGE band patrone van die 16S rDNA en volgorde bepalingsmetodes ondersoek. Die GenBank Blast resultate het getoon dat V. divaricata doeltreffend deur 'n wye reeks wortel-wortelknoppie bakteriële stamme genoduleer is, insluitende insluitende Burkholderia phytofirmans, Burkholderia sp. en Bradyrhizobium sp. tydens lae P toediening. Die natuurlike 15N voorkoms data het ook bevestig dat 40-50% van die N voeding deur simbiotiese N2 binding bekom is. Dit dien nie net as bewys vir wortelknoppie vorming nie, maar ook 'n aanduiding van die aanpassing van 'n reeks N2 bindende bakteriële stamme/ spesies tot die voedingsarme, sanderige, suur grond van die Mediterreanse ekosisteem van die Fynbos. Die derde studie het die fisiologiese effekte soos bv. N2 fikserings faktore, die afhanklikheid van plante op N2 fiksering, N voorkeur, peulgewas groei, koolstof kostes en aminosuur biosintese tydens P tekort en minerale N toediening soos NH4NO3 in 'n stadig-groeiende, Fynbos peulgewasboom, Virgilia divaricata ondersoek. Volgehoue toediening van NH4NO3 aan die peulplant toon 'n groter toename in plant droë weefsel. Tydens P tekort is die koolstof bou koste meer verhoog in plante wat met twee N bronne voorsien is. Tydens verlengde P tekort het die maksimum fotosintese tempo per blaaroppervlakte toegeneem, ongeag die N bron. Alhoewel die plante wortelknoppies gevorm het, het die plant se afhanklikheid van N2 binding tydens die toediening van NH4NO3 afgeneem.
