Vertical Axis Wind Turbines : Electrical System and Experimental Results

The wind power research at the division of Electricity at Uppsala University is aimed towards increased understanding of vertical axis wind turbines. The considered type of wind turbine is an H-rotor with a directly driven synchronous generator operating at variable speed. The experimental work presented in this thesis comprises investigation of three vertical axis wind turbines of different design and size. The electrical, control and measurement systems for the first 12 kW wind turbine have been designed and implemented. The second was a 10 kW wind turbine adapted to a telecom application. Both the 12 kW and the 10 kW were operated against dump loads. The third turbine was a 200 kW grid-connected wind turbine, where control and measurement systems have been implemented. Experimental results have shown that an all-electric control, substituting mechanical systems such as blade-pitch, is possible for this type of turbine. By controlling the rectified generator voltage, the rotational speed of the turbine is also controlled. An electrical start-up system has been built and verified. The power coefficient has been measured and the stall behaviour of this type of turbine has been examined. An optimum tip speed ratio control has been implemented and tested, with promising results. Use of the turbine to estimate the wind speed has been demonstrated. This has been used to get a faster regulation of the turbine compared to if an anemometer had been used.

F-actin rearrangements and analysis of physical environment of invasive hyphal growth.

Invasive growth through a substrate requires a massive amount of penetrative force, and this is generated in the space of a few microns in a growing tip. This process is known to be critical in the root hair, pollen tube, rhizoids, and the topic of this thesis, hyphal growth. However defining the mechanisms underlying the tip growth remains a contentious issue. Shortcomings in control of direction and regulation of growth began to undermine early turgor-based theories, and the cytoskeletal protein actin, ubiquitous in nature and with crucial roles in structure and motility became a target for investigation. A major breakthrough came with the discovery that a characteristic actin depleted zone (ADZ) occurs at the growing tip of hyphae during invasive but not non-invasive hyphal growth. The ADZ is likely to have an important role in generating the greater protrusive force required for invasive growth. However, since its discovery, little has been determined about the characteristics of the ADZ. Uncertainty in the description of the physical environment the hyphae face adds a layer of complexity to interpretation of results. This thesis aims to address this issue, studying the impact of increasing agarose substrate concentration on the presence and dimensions of the ADZ in the oomycete A. bisexualis. Furthermore, agarose is examined by compression and imaging to compare the physical characteristics of the agar samples over the range of concentrations, and determine whether increasing agarose concentration influences agarose gel structure. Results suggest a difference in the number of ADZ observed in non-invasive compared with invasive samples, however no significant differences in the number or dimensions of ADZ were found amongst the 1-4% w/v agarose concentrations. The 0% sample showed 20.7 percent of hyphae exhibited depleted zones, while 1, 2, 3 and 4% samples showed 56.9%, 48.8%, 40.9% and 54.2% respectively. ADZ dimensions did not correlate with agarose concentration. The average ADZ area:hyphal diameter ratio was 0.634, 0.526, 0.430, 1.09, and 0.65 for 0-4% agarose concentrations respectively. Additionally, investigation of gel compression forces revealed gel strength increases with agarose concentration. The force required to compress the agarose increased from 1.85 Psi in 1% agarose to 4.85, 7.09 and 12.22 Psi in 2, 3 and 4% agarose concentrations respectively. SEM imaging, however, suggests heterogeneity of the fibrous interconnected network of agarose gels at a microscopic scale with variable porous structure at all agarose concentrations. This scale is relevant to hyphal tip growth. In combination, these results suggest F-actin depletion may be a response mechanism to provide greater force for invasive growth. Additionally, this response is not dependent on the concentration of the agarose media, possibly due to the variability encountered within the media. These results contribute another important step forward in unraveling the elusive mechanism of tip growth.

Aerodynamic and acoustic analysis of the tip-leakage flow past a single ailfoil / Analyse aérodynamique et acoustique de l’écoulement de jeu d’un profil isolé

L'écoulement de jeu est un phénomène très important dans les turbomachines. Il provient du mouvement relatif entre la pale et la paroi d'extrémité, et la différence de pression à travers la pale. L'écoulement de jeu est extrêmement complexe pour sa nature tridimensionnelle et instable, et son existence conduit à de nombreux effets défavorables, par exemple, les pertes de performance aérodynamique et les émissions de bruit. C'est pourquoi l'écoulement de jeu a motivé de nombreuses recherches expérimentales et numériques. Afin d'améliorer la compréhension du écoulement de jeu et le bruit de large bande associé, une campagne de recherche a été menée au LMFA. En ce qui concerne l'écoulement de jeu, cette campagne de recherche comprend une expérience avec des technologies de mesure avancées, un calcul zonal LES et une série de calculs RANS / URANS. L'expérience et les simulations considèrent une configuration simple de l'écoulement de jeu à un faible nombre de Mach. Les résultats expérimentaux et numériques sont analysés de façon systématique et approfondie dans la présente étude. Enfin, des efforts sont déployés pour la modélisation et la prédiction du bruit à large bande avec des résultats expérimentaux et numériques. On observe dans l'expérience un système à multiple-tourbillon, avec une tourbillon de jeu intense. Les différentes analyses sur les caractéristiques d'écoulement montrent un bon accord entre l'expérience et le ZLES dans la région du écoulement de jeu. L'approche zonale (RANS-LES) s'avère être un outil puissant pour fournir une description détaillée du écoulement de jeu, avec un coût de calcul limité. Cependant, les calculs RANS et URANS surestiment globalement la diffusion de la tourbillon. En outre, l'oscillation du tourbillon de jeu est étudiée en utilisant des champs instantanés de PIV et l'amplitude d'oscillation est évaluée. La réponse dynamique de la tourbillon de jeu est également étudiée avec URANS aux fréquences choisies. Deux modèles de prédiction du bruit en champ lointain, correspondant à deux sources acoustiques différentes, sont reformulés et mis en oeuvre avec les données de champ proche des simulations numériques. Ces prédictions sont comparées aux mesures à champ lointain. En utilisant les données ZLES, le modèle de l’écoulement de jeu sur-estime le bruit généré dans la région de jeu. Le modèle de bruit de bord de fuite est implémenté avec les données ZLES et les données RANS et fournit une très bonne prédiction dans une large bande de fréquence. / The tip-leakage flow is a common flow feature in turbomachines. It originates from the relative motion between the blade tip and the end-wall, and the pressure difference across the blade. The tip-leakage flow is extremely complex for its three-dimensional unsteady nature, and its existence leads to many unfavourable effects, such as aerodynamic performance losses and noise emissions. These issues have motivated extensive experimental and numerical researches from both aerodynamic and aeroacoustic points of view. In order to improve the understanding of the tip-leakage flow and its associated broadband noise, a research campaign has been carried out at LMFA. Regarding the tip-leakage flow, this research campaign includes an experiment with advanced measurement technologies, a zonal LES computation and a series of RANS/URANS computations. Both the experiment and the simulations consider a single-airfoil configuration at low Mach number. Experimental and numerical results are analysed systematically and thoroughly in the current study. Finally, efforts are put on the broadband noise modelling and prediction based on the experimental and numerical results. A multi-vortex system with an intense tip-leakage vortex is observed in the experiment. The various analyses of the flow characteristics show a good agreement between the experiment and the ZLES in the blade tip region. The zonal (RANS-LES) approach proves itself to be a powerful tool to provide a detailed description of the tip-leakage flow, with a limited computational cost. However, the RANS and URANS computations globally over-estimate the diffusion of the tip-leakage vortex. Furthermore, the random oscillation of the tip-leakage vortex is investigated using PIV instantaneous flow fields and the wandering amplitude is evaluated. The dynamic response of the tip-leakage vortex is also studied with URANS at selected frequencies. Two far-field noise prediction models, corresponding to two different acoustic sources, are reformulated and implemented with the near-field data from the numerical simulations. These predictions are compared to the far-field measurements. Using the ZLES data as input, the blade-tip self-noise model is found to over-estimate the noise generated in the blade-tip region. The trailing-edge noise model is implemented with the time-averaged ZLES and the RANS near-field data, and yields a very good prediction within a broad range of frequency.

A radiação laser tem sido amplamente utilizada em diversos dispositivos para aplicações tecnológicas. Uma área particularmente interessante que tem sido explorada para o desenvolvimento destes dispositivos é a medicina. Um campo da medicina que tem se beneficiado desse esforço é a termoterapia a laser. No entanto, algumas desvantagens do uso em larga escala desta tecnologia são devidas à falta de uniformidade da absorção da radiação pelo tecido humano. Uma maneira proposta de resolver esta problemática tem sido a utilização de um método fototérmico indireto para converter a energia da luz do laser em calor; por meio de uso de pontas de fibras ópticas revestidas. Contudo, o uso dessas pontas apresenta a desvantagem da limitada estabilidade química do revestimento. Neste trabalho, apresentamos os resultados do desenvolvimento de pontas cristalinas de Nd2O3 altamente estáveis quimicamente em fibras monocristalinas de safira (Al2O3) obtidas diretamente a partir da fase líquida (fundida), fazendo-se uso da técnica Laser-Heated Pedestal Growth (LHPG). A metodologia desenvolvida permite a produção de pontas de diâmetros variados, o que possibilita definir a dimensão da região de aquecimento dentro dos limites de interação ponta cristalina/meio. Para o crescimento dos cristais no formato de fibras, chamados de fibras monocristalinas, usando-se a técnica LHPG, preparamos pedestais de Al2O3 puros e de composição Nd2O3, utilizando-se a técnica de extrusão a frio. Fibras monocristalinas de safira de 0,67 mm de diâmetro e 200 mm de comprimento, livres de poros ou trincas, com pontas cristalinas aproximadamente cônicas foram fabricadas em atmosfera aberta. Para avaliarmos a eficiência da conversão de luz em calor, as fibras foram bombeadas com luz de um laser de comprimento de onda 810 nm. Uma correspondência linear entre a potência do laser e a temperatura da ponta cristalina foi determinada em nossos sistemas e valores de até 250 oC podem ser alcançados; o que torna a ponta cristalina da fibra promissora para aplicações em termoterapia a laser e para a área de soldagem em micro(nano) eletrônica. Neste trabalho apresentamos também um estudo sistemático sobre o processo de puxamento de fibras do compósito eutético Nd2O3-NdAl11O18, através da técnica LHPG, com velocidades de puxamento variando entre 0,08 - 0,92 mm/min. As fibras eutéticas foram fabricadas com sucesso, livres de poros ou trincas. A microestrutura analisada ao microscópio eletrônico de varredura (MEV) foi denominada como do tipo regular, presente em todas as amostras. A despeito da sua complexidade, a microestrutura desse compósito foi controlada e a relação entre o espaçamento médio entre as fases e a velocidade de puxamento &#9552v = constante = (8,2 ± 0,3) µm3/s foi mantida em todo o intervalo das velocidade de puxamento utilizadas. Devido às propriedades mecânicas e ópticas das fases eutéticas individuais, este compósito apresenta-se como candidato com enorme potencial para aplicações dentro da ciência e engenharia de materiais. Ao nosso conhecimento, este trabalho parece ser o primeiro relato sobre a microestrutura eutética do sistema binário Al2O3-Nd2O3. / The laser radiation has been extensively used as devices for technological application, for which the medicine is a hot topic. In this field, the laser-induced thermotherapy is the primary beneficiary of the main researches and applications. However, the heterogeneity associated to the light aborsorption by the human tissue is major impediment preventing the large using of the thermotherapy. To solve this situation, an indirect photothermal method has been employed in order to convert laser radiation into heat by using coated optical fiber tips. Nevertheless, the limited chemical stability of the coating is a crucial problem associated to the large scale application of these coated tips. In the present work, we prepared crystal tips of Nd2O3, chemically stable, coupled in the Al2O3 (sapphire) crystal fiber, being both obtained by the laser-heated pedestal growth technique (LHPG), providing the production of tips with different diameters. Also, the cold extrusion technique was employed to produce the Al2O3 e Nd2O3 pedestals. Besides, the Al2O3 crystal fibers with 0.67 mm in diameter and 200 mm in length, free of cracks and pores, were successfully produced in an air atmosphere. In order to account the efficiency of the light-to-heat conversion, the fibers were pumped with laser light centered at 810 nm. Finally, linear correspondence between laser power and crystal tip temperature was determined in our systems, making them appropriated to the laser-induced thermotheraphy applications and welding in micro- and nano-electronics. Furthermore, we presented a profound investigation about the pulling process of Nd2O3-NdAl11O18 eutectic fibers obtained by LHPG technique. These eutectics were successfully pulled free of cracks and pores. Using scanning electron microscopy (SEM), we were capable of identificating the regular microstructure of our eutectics. Changes in the microstructure were observed in the eutectic fibers pulled using fiber pulling rates from 0.08 to 0.92 mm/min. Despite the complexity of their microstructures, the relation &#9552v = (8,2 ± 0,3) µm3/s = constant, defined by Jackson and Hunt, applied very well for this system. Owing to the mechanical and optical properties of the eutectic phases, these compounds present huge potential for applications in the materials science and engineering related fields. To our knowledge, this work seems to be the first report on the eutectic microstructure of the Al2O3-Nd2O3 binary system.

Les travaux de thèse se sont attachés à développer des outils permettant :(i) d'estimer par nanoindentation le niveau de contraintes résiduelles locales de type biaxial introduit dans un alliage d'aluminium 2050-T8. Nous proposons une méthode d'estimation qui couple des essais expérimentaux de nanoindentation, l'observation des empreintes résiduelles et une analyse numérique de l'effet de contraintes résiduelles élastiques biaxiales sur la géométrie des empreintes résiduelles d'indentation. et (ii) de déterminer par nanoindentation le comportement mécanique d'un film d'oxyde ultra-mince, d'épaisseur variant entre 15 et 20nm, formé sur un acier inoxydable biphasé de type Duplex. Nous avons développé une méthode de caractérisation de films ultra-minces qui couple la réalisation et l'analyse d'essais expérimentaux à l'aide d'indenteurs dont le défaut de pointe est déterminé, et la reproduction de ces essais en simulation numérique 3D à l'aide des géométries réelles des indenteurs. / Works performed during this thesis were dedicated to development of tools allowing:(i) To estimate using nanoindentation tests the level of biaxial residual stresses introduced within a 2050-T8 aluminium alloy. We propose an estimation method that couples experimental nanoindentation tests, residual imprints observation and numerical evaluation of the impact of elastic biaxial residual stresses on the geometries of residual indentation imprints. And (ii) the determination by nanoindentation of the mechanical behavior of an ultra-thin oxide film, between 15 to 20nm thickness, formed on a dual phased Duplex stainless steel. We developed a characterization method that couples analysis of experimental nanoindentation tests using several indenters with known tip defects, and reproduction of these tests in 3D finite element simulations using the true indenters' geometries.

The auditory mechano-electrical transduction machinery : components and interactions / La machinerie de la transduction mécano-électrique auditive : composants et interactions

La (Pcdh15) est localisée dans les touffes ciliaires des cellules ciliées internes et externes de la cochlée. Elle forme des liens interstéréociliaires entre les différents stéréocils, des protrusions riches en actine qui forment ensemble la touffe ciliaire. L’absence de Pcdh15 entraîne la désorganisation des touffes ciliaires et la perte de la mécanotransduction auditive. Pcdh15 forme en effet la partie basse du lien de bout de cil (en anglais tip-link), le lien contrôlant l’ouverture du canal de transduction, qui se situe au point d’insertion inferieur de ce lien. Il existe trois isoformes de Pcdh15 (CD1, CD2 et CD3). J’ai étudié la distribution de ces isoformes dans la touffe ciliaire à différents stades de sa maturation. Différents modèles murins « KO conditionnel » ont été générés, ils m’ont permis d’analyser les conséquences de l’absence de chacune des isoformes individuellement, ainsi que celles de l’absence de Pcdh15-CD2 et Pcdh15-CD3, et celles de l’absence simultanée des trois isoformes. J’ai ainsi pu conclure que Pcdh15-CD2 est la seule isoforme essentielle pour la formation des tip-links dans les cellules ciliées matures. Dans les touffes ciliaires matures, Pcdh15 joue également un rôle dans l’interaction entre les touffes ciliaires et la membrane tectoriale, dans le contrôle de la taille des stéréocils, et dans la formation des liens interstéréociliaires apicaux. Pour ces fonctions, mais pas pour la formation du tip-link, Pcdh15-CD1 et Pcdh15-CD2 (les isoformes présentes dans la touffe ciliaire mature) sont redondantes. Cependant, Pcdh15-CD1 ne peut compenser que partiellement l’absence de Pcdh15-CD2 et Pcdh15-CD3. Pour étudier comment Pcdh15 interagit avec les autres protéines impliquées dans le syndrome de Usher, les interactions avec l’harmonine et la whirline (deux protéines d’échafaudage qui sont colocalisées avec Pcdh15 à l’apex des stéréocils) ont été analysées in vitro. / Protocadherin-15 (Pcdh15 is located in the stereociliary hair bundles of inner and outer hair cells (IHCs and OHCs) of the cochlea, where it forms fibrous links between different stereocilia. Absence of Pcdh15 leads to deafness due to the disorganization of hair bundles and absence of mechano-electrical transduction. The latter is explained as Pcdh15 forms the lower component of the tip-link, that gate hair cell mechano-electrical transduction channels. There are three different splice isoforms of Pcdh15 (CD1, CD2 and CD3), I studied their distribution in the developing and mature auditory hair cells. Different conditional Pcdh15 knockout mouse models were generated, permitting analysis of the absence of each of the different Pcdh15 isoforms individually, of the combined absence of Pcdh15-CD2 and Pcdh15-CD3, and of the absence of all isoforms. I was able to conclude that Pcdh15-CD2 is essential for the formation of tip-links in mature hair cells. In mature hair bundles Pcdh15 also plays a role in the coupling of the hair bundles to the tectorial membrane, in the control of the size of the stereocilia, and in the formation of apical links between stereocilia. The different Pcdh15 isoforms present in mature hair bundles (Pcdh15-CD1 and Pcdh15-CD2) are functionally redundant for these functions, but not for tip-link formation. In immature hair bundles, the different Pcdh15 isoforms are functionally redundant, although Pcdh15-CD1 can only partially compensate the absence of Pcdh15-CD2 and Pcdh15-CD3. To discover how Pcdh15 interacts with other proteins implicated in Usher syndrome, interactions with harmonin and whirlin were analyzed by biophysical techniques.

Trocantheric Femoral Fractures - a retrospective cohort study comparing reoperation rates before and after implementation of new treatment protocol

Introduction: Hip fractures are very common in Sweden. Keeping reoperation rates as low as possible is critical. One factor correlated to mechanical complications and thereby reoperation rates is the tip-apex distance. In April of 2017 the Orthopedic clinic at Karlstad’s Central Hospital in Sweden implemented new routines for the choice of treatment method of pertrocantheric and subtrocantheric femoral fractures. Aim: The primary aim of this study was to compare reoperation rates of pertrocantheric and subtrocantheric femoral fractures before and after implementation of new routines. Secondly, we wanted to study the association between the tip-apex distance and reoperation rates for pertrocantheric femoral fractures treated at Karlstad’s Central Hospital. Method: This was an internal quality control designed as a retrospective cohort study. Data on patients treated for pertrocantheric or subtrocantheric femoral fractures were retrieved from the Swedish Fracture Registry. Tip-apex distances were measured on patients’ radiographic images. P-values for comparison of the two study periods were created using the chi2 test. Association between the tip-apex distance and reoperation rate was presented as odds ratio. Result: The reoperation rates were the same for both time periods. The reoperation rates of unstable fractures were lower for the study period after implementation of new routines. Neither of these results were statistically significant. There was also no statistically significant association between the tip-apex distance and reoperation rates. Conclusion: No statistically significant decrease in reoperation rates was seen after implementation of new routines. There was no association between the tip-apex distance and reoperation rates.

Development and Application of Theta Tips as a Novel nESI-MS Ion Source and Protein Identification Using Limited Trypsin Digestion and Mass Spectrometry

<div><div><div><div>Mass spectrometry is a widely used tool for efficient chemical characterization and identification. The development of electrospray ionization as a soft ionization method enables mass spectrometry for large biomolecule investigation. Protein as one of the most important classes of biomolecules, its structural changes including folding, unfolding, aggregation, degradation and post-translational modification all influence protein bioactivity. Protein characterization and identification are important for protein behavior mechanism understanding, which may further contribute to disease treatment development. Protein conformation changes are normally very fast, and the initial stages, which significantly influence the conformation changing pathway, normally occur in milliseconds or shorter time scale. Such a fast structural change is hard to be monitored using traditional bulk solution manipulations, and fast sample preparation methods are required. </div><div><br></div><div>In this thesis project, theta tips are applied as a microreactor and nESI-MS emitter to perform fast protein manipulation immediately before MS analysis. Theta tips can be operated in two modes. The first mode is for submillisecond time scale reactions. Proteins and reagents are loaded into different channels and directly sprayed out simultaneously. Proteins and reagents mix and react in the Taylor cone and subsequent droplets for submillisecond time scale. Through this method, pH induced protein folding is investigated and protein folding intermediates were captured. The second mode is for milliseconds or longer reactions. Differential voltages can be applied to each channel before ionization and spray. The electric field between the two channels induces in-tip electroosmosis, which lead to an in-tip mixing and reaction. In this mode, the reaction time is not limited by the droplet lifetime as in the first mode, but is controlled by electroosmosis time. By changing the electroosmosis square wave frequency and cycles, the mixing time can be elongated to milliseconds or longer, which is suitable for slower reaction study. </div><div>Joule heating is discovered during theta tip electroosmosis when samples are dissolved in buffer. The Joule heating effect is high enough to heat up the aqueous solution to at least 75 oC based on Raman thermometry measurement, while the actual peak temperature could be higher. The Joule heating effect in theta tip electroosmosis can be easily controlled by electroosmosis voltage, time, buffer concentration etc.. Proteins are thermally denatured by the Joule heating effect, and the denaturation extent correlates with Joule heating parameters. </div><div>With this results in hand, we are developing a protein melting temperature measurement method using theta tip Joule heating effect and mass spectrometry. This new melting temperature measurement method measures changes in protein mass and charge state distributions. Therefore, it could sensitively detect ligand loss and protein tertiary structural changes, which is an important compensation to current protein melting temperature measurement techniques like CD or DSC. Since the heating time is short and protein concentration for MS is low, protein aggregation and thermal fragmentation are highly avoided so a complete protein thermal unfolding process is monitored. Theta tip electroosmosis combining MS characterized protein thermal denaturation behavior from a new aspect.</div><div><br></div><div>Besides single protein folding and unfolding, protein identification and post-translational modification are important for proteomics study. The traditional bottom-up, top-down and middle-down methods are not able to both preserve intact protein mass and efficiently generate enough fragment peaks easily without performing gas phase dissociation. In this thesis, we also developed a new way to identify proteins combining limited trypsin digestion and mass spectrometry. Intact protein mass was preserved for protein size and PTM identification. Enough tryptic peptides were also generated for protein identification through database search.</div></div></div></div><div><br></div>

his Diploma thesis concentrates on problems related to cutting tools, their wear, durability, and cutting conditions. The work provides basic analysis of technology applied to two selected parts. Further, there is also analysis of cutting tools suppliers. The work gives brief characteristics of the basic types of wear and their origins. In the experimental section is a statistical evaluation of selected tools, calculation of optimal cutting conditions and durability, and finally selected tools wear evaluated. The technical and economical evaluation compares cutting conditions provided in theory with the real cutting conditions.
