Previous issue date: 2013-03-08 / Ethylene is the inductor and acceleratorhormone of the maturation and senescence of climacteric fruits, as is the case of Cantaloupe melons (Cucumismelo var. Cantalupensis, Naud cv. Vedrantais) and tomatoes (LicopersicumesculentumL. cv.
Micro-Tom). It is known that the reduction of production and/or the action of this hormone prolongs the shelf life of these fruits. Thus, in order to reduce the production
of such hydrocarbon, were genetically transformed melon trees with clones of the ACC oxidase 'antisense', of melon (pMEL1AS, Ayubet al., 1996) and of apple (pAP4AS, Silva et al., 2004). As expected, in both cases, the ethylene production was reduced, prolonging shelf life of fruits in 7 days. However, the production of volatile compounds was also affected. The transgenic melons produced, on average,
70% less esters than the WT, independently of having been transformed with pMEL1AS or pAP4AS. As the intervention made acted in the reduction of the production of ethylene. It was emitted the hypothesis that, with the supplementation
of the hormone, the synthesis of flavors would be restored. Such fact was proven in pMEL1AS melons, but not in pAP4AS. The exact causes of this difference were not yet made clear. It is believed that because of the greater reduction of the ethylene production in the fruit pAP4AS, besides affecting genes directly related to the ripening classical metabolic pathways, genes related to other pathways of synthesis
of hormones have also been affected, as is the case of cytokininsor polyamines. By observation of the phenotype of pAP4AS plants we realized that these had more vegetative growth and lateral sproutings. Considering such observations, it was launched the hypothesis that with the increase of the levels of cytokinins one could interfere in the responses to ethylene, delaying the normal evolution of the fruits. In
pAP4AS melons it was proved that there are higher levels of cytokinins, both in the roots and fruits. However, to confirm that these changes are effectively consequence of higher levels of cytokinins, exogenous application of that growth regulators was made. Nevertheless, were not observed any expected responses. So we decided to test the application of cytokinin in another plant model, the tomato tree cv. Micro-
Tom, who is also a producer of climacteric fruits and that provides greater ease of cultivation and obtainment of greater quantity of fruit in protected environments and in areas with limited space. As molecular variables for assessment of the treatments in melons were determined the accumulation of transcript of genes of HPL, LOX and AAT, besides the ACCO and ACCS. In tomatoes, were performed physicochemical
analyzes. For melons, when quantifying the accumulation of transcripts of genes of ACC synthase (ACCS), it was found that genes ACCS1 and ACCS3 showed higher expressions in WT fruits, suggesting that they have a strong relation with the evolution of the climacteric crisis. Moreover, the gene ACCS5 was more expressed in fruits pMEL1AS and pAP4AS, indicating that it is negatively regulated by ethylene.
For the other variables evaluated (total soluble solids, color, carotenoids, chlorophylls, pulp firmness) during the ripening on the plant, there were no marking differences between pMEL1AS and pAP4AS. After harvesting and treatment of
pMEL1AS fruits with ethylene, there was a degreening of the peel and increase of the production of esters, correlated with higher levels of transcripts of the genes of HPL, LOX, AAT1, AAT2, AAT3 and AAT4. In pAP4AS fruits, the levels of these
transcripts was significantly lower, not having been observed degreeningor yellowing, nor restoration of the synthesis of esters. In these fruits (pAP4AS) it was detected a higher concentration of zeatin and zeatin ribose than in pMEL1AS and WT. For tomato plants, in which was done application of cytokinins, there was prolongation of the vegetative cycle and delay in the maturation. The fruits from the treated plants
also prolonged the maturation cycle, but did not decrease sensitivity to ethylene action. It is believed that cytokinins may be the responsible ones for lower sensitivity to ethylene in melons, but this hypothesis was not confirmed and must be tested with greater profundity. The attempt to prove it using tomato plants as model was not efficient.
principais pigmentos desse fruto (β-caroteno e licopeno).

[en] The paper proposes to examine the compositional process of the musician Antonio Carlos Jobim, in partnership with Vinícius de Moraes, Newton Mendonça and Dolores Duran in the early years of the movement known as Bossa Nova (1958 and 1959). During this period you can see some movement in the songs produced, showing both approaches and departures with the previous musical repertoire (samba-canção). Based on the notions of tradition / modernity, classical / popular, we reflect on the role of the musician as a kind of social chronicler.

Evoluce importu proteinů do mitochondrií / Evolution of mitochondrial protein importDohnálek, Vít January 2020 (has links)
1 Abstract Even though mitochondria possess their own genome and ribosomes, majority of mitochondrial proteins is encoded in the nucleus and translated by cytosolic ribosomes. Hence it was necessary to establish transport complexes allowing the import of proteins from the cytosol. These complexes are best described in yeast. However, we are encountering organisms lacking many of the subunits of these complexes with increasing frequency. Therefore, we are presenting the overview of the distribution of the subunits within eukaryotic organisms. We specifically take a closer look at parasitic protist Giardia intestinalis that is well known for its extreme reductions of the import complexes. There have been only few subunits identified so far. Porin Tom40, that is responsible for translocating all the incoming proteins across the outer mitochondrial membrane, has been identified despite the high divergence, while homolog of Sam50 hasn't been successfully identified yet. Sam50 is however believed to be necessary for insertion of Tom40 into the membrane. Vast part of this thesis is dedicated to this phenomenon that is highly uncommon and maybe unique among the eukaryotic organisms.

The Hydrogeology and Hydrochemistry of the Mt. Tom Price Mine, Pilbara, Western Australia – A Groundwater Flow Model.Manewell, Neil January 2008 (has links)
The Mt. Tom Price Mine, located in the Pilbara region of Western Australia, has been the site of major iron ore mining since the 1960s by Rio Tinto Iron Ore/Pilbara Iron. The thesis project area covers approximately 121 km², covering the Mt. Tom Price Mining area and the surrounding catchment boundary. The climate in the Pilbara region is arid, with rainfall driven by seasonal cyclonic events, producing 300 mm/year net rainfall on average. The geology of the Mt. Tom Price area consists of a series of banded iron formations (BIF) and shales that are generally low in hydraulic conductivity values. Iron ore in the region is produced through the process of supergene enrichment whereby gangue minerals are dissolved and replaced with haematite and goethite. Mining is focused in a series of open cast pits including, North Deposit, West Pits, Centre Pits, Southern Ridge, South East Prongs, Section Six, Section Seven, and the proposed Marra Mamba Pits. Due to the impermeable nature and complex geology of the BIF sequence, groundwater flow is dominated by bedrock aquifer flow, with compartmentalization occurring in several areas of the mine. Highly faulted and folded units can also have increased hydraulic conductivity values. Pit floor lowering began to encounter the regional water table in early 1994. A series of dewatering bores and depressurization measurements have been utilized to ensure dry mining practice. This data was used to help understand regional groundwater flow and create the Mt. Tom Price Groundwater Model (MTPGM). A 3D geological model of the project area was created to aid visualisation of semi-regional hydrogeology. From this model, accurate template files were created so that geological detail loss is kept to a minimal when entering hydrogeological parameters into the MTPGM. The MTPGM was setup using PMWIN Pro, a graphical user interface for use with MODFLOW. Stresses such as recharge and pumping were entered via software packages within MODFLOW. The model was run to simulate measured 1994-2007 responses to dewatering and high rainfall events. A Parameter Estimation (PEST) software package and trial and error calibration was used to lower stress response variances that were observed in the model output files. This was achieved by the adjustment of hydrogeological parameters such as hydraulic conductivity and specific yield values. A prediction simulation of final pit lake recovery was created Using the calibrated MTPGM. Recovery curves predicted that full recovery of the water table of the pit voids varied from 96 to 120 years, recovering to levels close to the initial heads measured in 1994 before large-scale pumping commenced. The hydrochemistry of the groundwater in the mining area is highly influenced by geological hosts, with clearly defined hydrochemical signatures approximated for each screened geological unit. Due to the sulphur rich, acid- forming Mt. McRae Shale, regular monitoring of pit and groundwater is essential. Final pit lake water quality was estimated using final pit levels and recovery rates approximated from the MTPGM, combined with historical data and previous groundwater quality reports. Pit lake water quality is dominantly driven by evaporation concentration, caused by high evaporation rates and low throughflow. Pit waters are expected to be brine waters (greater than 100,000 mg/L TDS), with high levels of acidity values occurring in the South East Prongs and Section Six pits due to the exposure of the acid forming Mt. McRae Shale above the pit lakes at these localities. Future studies should focus on more detailed modelling of the compartmentalised aquifer systems. This would produce much more accurate final pit lake levels. Further study of the Mt. McRae Shale formation and its implications on acidity should also be undertaken. Seasonal fluctuations in lake levels will affect acidity due to the continual re-exposure and oxidation of the Mt. McRae Shale. This could be studied to help understand short term pit lake quality conditions and help to predict long term acidity conditions in the pit lakes.

Performing masculinity : the star persona of Tom CruiseO'Donnell, Ruth January 2012 (has links)
Tom Cruise was one of the most financially successful stars of the 1980s and 1990s and remains an important Hollywood player. In his time at Paramount, he negotiated new deals which redefined how stars earned revenue from their films. Yet little substantial study has been made of him or his persona. Cruise came to prominence in the 1980s, an epoch which redefined the terms of masculinity, moving away from a particular martial ideal following American defeat in Vietnam. His persona addresses the consequent anxieties surrounding these significant social changes. In addition to this, his image can be understood within a psychoanalytic framework to be offering a 'compromise formation' to the difficulties of psychosexual development. Cruise appears to offers a challenge to the father, but in reality recedes from Oedipal threat. Chapter One examines the nascent Cruise star persona and its development across his films. The chapter provides a historical overview and contextualisation of the persona. Chapter Two explores how the persona offers a 'performative' model of masculinity based on a heroic martial ideal, as well as his positioning as an object of spectacle and the onscreen homoerotic relationships he enjoys. Chapter Three looks at the star's association with the racial 'other' and preoccupation with male friendships at the expense of heterosexual romance. Objectification through spectacle, which black characters and Cruise share in his films, indicate a symbolic shared powerlessness within the hegemony. Chapter Four argues that this lack of status is characterised by Cruise's struggle against male authority figures, which represent both corporate America and the punishing father. This relays the trauma of Oedipal struggle, which Cruise attempts to evade. Chapter Five looks in further depth at the Cruise persona's preoccupation with male bonds and lack of interest in women, both indicative of a regression from the phallic to the earlier anal/cannibalistic psycho-sexual stage, as he is unable to overcome the symbolic father . The thesis concludes by suggesting Cruise's persona is a compromise: it presents a triumphant young man who in reality is oppressed by patriarchal forces. This compromise is premised on youth and is threatened as Cruise ages, exaggerated by social changes within America. Biologically and socially, Cruise's persona is no longer tenable.
