A gênese da bossa nova : João Gilberto e Tom Jobim

Este trabalho estuda a gênese do estilo musical bossa nova a partir da análise das quatro canções de Tom Jobim com letra dele mesmo que integram o repertório dos três primeiros discos de João Gilberto. A análise propriamente dita é precedida de contextualizações e definições. Situa-se a bossa nova como gênero no contexto histórico da canção, estabelecedo-se o recorte específico do trabalho. Procura-se aplicar o conceito de sistema literário, ao estudo da canção, em especial para clarificar o papel relevante das novas tecnologias disponíveis de gravação e radiodifusão na definição da canção popular do século XX. É feita uma análise resumida do que é a canção popular norte-americana, em especial aquilo que convencionou-se de chamar standards. Tendo alguns compositores – Noel Rosa, Pixinguinha, Dorival Caymmi e Ary Barroso – como eixo diacrônico, mostram-se os traços da modernização da música brasileira e, ao mesmo tempo, discutesse a dicotomia tom elevado/tom rebaixado aplicada à essa música. Finalmente, pesquisa-se na análise de Outra vez, Só em teus braços, Este teu olhar e Corcovado, os traços característicos do estilo. Considerando a relevância em cada caso, são analisados: a voz, o violão, a forma, os baixos, a harmonia, as letras e as melodias. / This work studies the birth of the bossa nova style from the analysis of the four Tom Jobim‟s songs where he is also the lyricist in the first three albuns of João Gilberto. The analysis itself is preceeded of contextualizations ano definitions. The bossa nova is placed as a genre in the historic context of the “song”. The concept of literary sistem is applied to the study of the song, particularly concernig the role of the new recording and broadcasting tecnologies that appeared in the 20th century. A brief analysis of the american popular song, specially the jazz standards, a historical contextualization of the bossa nova in brazilian popular music and the discussion of the dicotomy between high and low levels of art are included. Whith the analysis of the songs Outra vez, Só em teus braços, Este teu olhar and Corcovado, the main characteristics of the style are pointed. The voice, the guitar, the form, the basses, the harmony the lyrics and the melodies are analysed.

To get the expected results of the study addressed characteristics that locality, in order to try to demonstrate some peculiarities about the population as a whole. Through various analyzes, we realized that beyond the means slaves and economic activities, family relationships between free and slave also informed us that the slave society constitutes a remote rural parish of large shopping plazas and focused on the domestic supply site. Elucidating the functional interplay between Parkinson’s disease-related proteins and the mitochondrion / Etude de l’interaction fonctionnelle entre les protéines impliquées dans la maladie de Parkinson et la mitochondrie

La maladie de Parkinson (MP) est une affection neurodégénérative fréquente d’étiologie inconnue, touchant environ 5% de la population mondiale après 80 ans. Environ 10% des cas correspondent à des formes familiales à transmission mendélienne. Pendant longtemps, un dysfonctionnement mitochondrial a été soupçonné jouer un rôle dans la physiopathologie de la MP. Cette possibilité a été récemment corroborée par des découvertes majeures réalisées dans le cadre des formes autosomiques récessives. Parkine et PINK1, les produits de deux gènes associés à ces formes familiales, participent au sein d’une même voie moléculaire au contrôle de la qualité mitochondriale, par la régulation du transport, de la dynamique, de la biogenèse et de la clairance de ces organites.L’objectif de ce travail a été d’élucider certains des mécanismes moléculaires sous-jacents à la régulation de l’homéostasie mitochondriale par Parkine et PINK1. Nous avons utilisé un ensemble d’approches de biologie moléculaire et cellulaire, de biochimie et de microscopie confocale, afin d’identifier et de caractériser des interacteurs moléculaires de Parkine et PINK1 à la membrane mitochondriale externe (MME).Dans la première partie de ce travail, nous avons découvert que la Parkine et PINK1 s’associent sur la MME de mitochondries dysfonctionnelles à proximité de la translocase de la MME (TOM), un complexe dédié à l’import de la grande majorité des protéines mitochondriales. Nous avons montré que ces interactions protéiques jouent un rôle clé dans l’activation du programme de dégradation mitochondriale régulé par la voie PINK1/Parkine. Nous avons également observé que la GTPase de type dynamine Drp1, impliquée dans la fission mitochondriale, est recrutée au niveau de mitochondries endommagées à proximité de Parkine et PINK1 ; ainsi, les processus de fission et de dégradation mitochondriales pourraient être spatialement coordonnés. Dans la deuxième partie de ce projet, nous avons caractérisé l’interaction fonctionnelle entre la Parkine et l’enzyme neuroprotectrice multifonctionnelle de la matrice mitochondriale, 17B-hydroxystéroïde déshydrogénase de type 10 (HSD17B10), dont les taux s’étaient révélés être diminués chez la souris déficiente en Parkine. Nous avons mis en évidence un effet protecteur d’HSD17B10 vis-à-vis de la mitochondrie qui était indépendant de son activité catalytique. Nous avons de plus montré que la Parkine interagit directement avec HSD17B10 à proximité de la machinerie TOM et qu’elle régule positivement l’abondance mitochondriale de cette protéine ; cela suggère qu’elle pourrait promouvoir son import.Dans l’ensemble, ces résultats approfondissent notre connaissance des mécanismes moléculaires mis en jeu par la Parkine et PINK1 dans le contrôle de la qualité mitochondriale, élargissant ainsi notre compréhension de leur rôle dans la physiopathologie des formes autosomiques récessive de MP. / Parkinson’s disease (PD) is a common neurodegenerative disorder of unknown etiology, affecting nearly 5% of the world population over the age of 80. Nearly 10% of PD cases are familial forms with Mendelian inheritance pattern. Mitochondrial dysfunction has long been suspected to play a role in the physiopathology of sporadic PD. This possibility has been recently corroborated by major discoveries in the field of autosomal recessive PD. Parkin and PINK1, the products of two genes associated with these forms, participate in a common molecular pathway focused on maintenance of mitochondrial quality, with roles in mitochondrial transport, dynamics, biogenesis and clearance.The aim of this work was to elucidate some of the molecular mechanisms underlying the regulation of mitochondrial homeostasis by Parkin and PINK1. We used a combination of approaches in molecular and cell biology, biochemistry and confocal microscopy to identify and characterize molecular interactors of Parkin and PINK1 on the outer mitochondrial membrane (OMM).In the first part of my project, we discovered that Parkin and PINK1 associate on dysfunctional mitochondria in proximity of the translocase of the OMM (TOM), a complex devoted to the mitochondrial import of the vast majority of the mitochondrial proteins. We provided evidence that these associations play a key role in activation of the mitochondrial degradation program mediated by the PINK1/Parkin pathway. We also observed that the dynamin-related GTPase Drp1, involved in mitochondrial fission is recruited to defective mitochondria in proximity of Parkin and PINK1, suggesting that mitochondrial fission occurs at sites where mitochondrial clearance is initiated.In the second part of my project, we characterized the functional interaction between Parkin and the multifunctional neuroprotective mitochondrial matrix enzyme 17B-hydroxysteroid dehydrogenase type 10 (HSD17B10), previously found by the team to be altered in abundance in Parkin-deficient mice. We demonstrated that HSD17B10 exerts a mitochondrion-protective function independent of its enzymatic activity. In addition, we provided evidence that Parkin directly interacts with HSD17B10 at the TOM machinery and that it positively regulates its mitochondrial levels, possibly through the regulation of its mitochondrial import.Altogether, these results provide novel insights into the molecular mechanisms by which Parkin and PINK1 control mitochondrial quality, and deepen our understanding of the role of these proteins in the physiopathology of autosomal recessive PD.

Os frutos do tomateiro (Solanum lycopersicum) apresentam elevada diversidade fenotípica, grande parte da qual deve-se a mutações em cinco genes, SUN (SUN), OVATE (O), LOCULE NUMBER (LC), FRUIT SHAPE 8.1 (FS8.1) e FASCIATED (FAS). Enquanto mutações em LC, O e FAS são responsáveis pela alteração no número de lóculos no fruto, mutações em FS8.1, SUN e O resultam em frutos mais alongados, sendo a última mutação o foco de estudo do presente trabalho. Apesar de ovate ser uma mutação importante tanto do ponto de vista agronômico quanto biológico, não é conhecido ainda o efeito exato da nova classe de gene regulatório codificado por esse locus. Por promover a drástica alteração no formato do fruto, característica esta que pode influenciar a preferência do consumidor, essa mutação é de grande interesse agronômico. Sob o ponto de vista biológico, o maior conhecimento da função desse gene resultaria em um maior entendimento do processo de frutificação. A análise de duplos mutantes ovate DR5::GUS e ovate ARR5::GUS mostrou que essa mutação altera o padrão de resposta a auxina e citocinina, respectivamente, durante o desenvolvimento do fruto. Além disso, foram realizados estudos que mostraram seu impacto no teor de sólidos solúveis, produtividade, tempo de prateleira, teor de matéria seca, peso das sementes e partenocarpia. Tais resultados não somente contribuíram para um melhor entendimento do efeito gênico da nova classe de genes regulatórios codificado por OVATE, como também fornecem importantes implicações agronômicas, já que a mutação ovate vem sendo utilizada na obtenção de variedades e híbridos do tipo \"grape\". / The fruits of tomato (Solanum lycopersicum) present a high phenotypic diversity, most of which can be attributed to mutations in five genes: SUN (SUN), OVATE (O), LOCULE NUMBER (LC), FRUIT SHAPE 8.1 (FS8.1) and FASCIATED (FAS). Whereas mutations in LC, O and FAS alter fruit locule number, mutations in FS8.1, SUN and O result in elongated fruits. The latter is the focus of the present work. ovate is a biologically and agronomically relevant mutation, however, the exact effect of the new class of regulatory gene it represents is unknown. The drastic alteration in fruit shape produced by the mutation can influence consumer preference, thus rendering it agronomically important. Biologically, a thorough characterization of its gene function would result in deeper understanding of fruit formation. Crosses to the marker lines DR5::GUS and ARR5::GUS showed that ovate alters the response pattern of auxin and cytokinin, respectively, during fruit development. Furthermore, studies are described showing its effect on total soluble solids content, yield, shelf life, dry mass rate, seed weight and parthenocarpy. The results contribute not only to a better understanding of the effect of the new class of regulatory genes encoded by OVATE, but also have important agronomic implications, given that the ovate mutation has been used in the obtention of \'grape/plum\' tomato varieties and hybrids.

Notre recherche consiste à comprendre quels sont les effets des jeux vidéo de simulation d’animaux virtuels sur les conceptions du vivant, en particulier sur la distinction entre ce qui est vivant et ce qui ne l’est pas. Pour ce faire, il s’agit d’identifier les différences conceptuelles entre les populations de joueurs et de non-joueurs aux jeux vidéo de simulation d’animaux virtuels. Tout d'abord, nous avons fait une enquête exploratoire identifiant les pratiques vidéo ludiques des enfants. Cette enquête a permis de sélectionner les deux jeux les plus utilisés par les enfants de cet âge : Talking TOM et POU. Notre étude porte plus particulièrement sur ces deux jeux. Un questionnaire a été diffusé au Liban et en France pour étudier la façon dont les joueurs et les non-joueurs à ces deux jeux conçoivent le vivant. Au total 916 enfants âgés de 9 à 12 ans ont rempli ce questionnaire. Au Liban, le questionnaire a été suivi de 21 entretiens avec des élèves. Les questionnaires ont été traités statistiquement afin de faire émerger les différences et les similitudes entre les populations de joueurs et de non-joueurs en France et au Liban. Les entretiens ont été analysés par une analyse thématique de contenu suivie par une analyse lexicale et ont permis une exploitation plus qualitative des résultats. Les résultats montrent que les entretiens et les questionnaires convergent fortement vers l’hypothèse d’une influence des jeux sur la construction de la notion de vie chez les joueurs des deux pays. En effet, au Liban et en France les utilisateurs de TOM et POU ont des conceptions significativement plus anthropomorphiques, animistes et anthropocentriques que ceux qui n’y jouent pas. / Our research is to understand the effects of video games simulating virtual animals on the concepts of life, especially the distinction between what is alive and what is not. To do this, it is a question of identifying the conceptual differences between the populations of players and non-players in virtual animal simulation video games. First, we did an exploratory survey identifying children's playful video practices. This survey allowed selecting the two most used games by children of this age: Talking TOM and POU. Our study focuses on these two games. A questionnaire has been distributed in Lebanon and France to study how players and non-players in TOM and / or POU design the living. A total of 916 children aged 9 to 12 completed this questionnaire. In Lebanon, 21 interviews with students followed the questionnaire. The questionnaires were statistically analyzed to reveal the differences and similarities between the populations of players and non-players in France and Lebanon. The interviews had been analyzed by a thematic analysis of content followed by a lexical analysis and allowed a qualitative exploitation of the results. The results show that interviews and questionnaires strongly converge on the hypothesis of an influence of games on the construction of the notion of life among players of both countries. Indeed, in Lebanon and France users of TOM and POU have significantly more anthropomorphic, animistic and anthropocentric conceptions than those who do not play it.

La sempiternelle recherche de compréhension par le visuel, des émotions générées par la musique, ouvre un horizon de paradoxes au cinéma qui mérite que l'on s'y attarde. L'esthétique du réalisateur-compositeur Tom Tykwer qui réalise ses films comme Wagner compose ses opéras en écrivant partition et livret, et dont l'inflexibilité sur sa double fonction est probablement unique au cinéma, nous sert de modèle pour consolider notre révision de quelques pensées communément admises sur la relation entre musique et images. Nous y revisitons la définition de la musique en tant que matière sonore du film, la question de sa concordance avec l'image, de son indépendance alors qu'elle intègre un tout unifié, ainsi que de son rapport aux sens et à leur transcendance. Les images comme substrat matériel et la musique comme élément de conscience chez ce réalisateur-compositeur, conservent leur autonomie de sens alors même que la partition est intimement liée à la structure du film. Elles s'inscrivent dans un rapport de complémentarité qui remet en cause tout un paradigme basé sur l'idée d'analogie ou de convergence, en somme, de dualité entre images et musique au cinéma. / The perennial search of comprehension by the visual, of emotions generated by music, opens an horizon of paradoxes in cinema that merits further consideration. Filmmaker-composer Tom Tykwer's aesthetics that allows him to direct movies as Wagner composes his operas by writing scores and libretto, and whose inflexibility of his double function is probably unique in cinema, serve us as model to strengthen our revision of some commonly admitted thoughts about the relationship between music and images. We reassess the definition of music as sound material of films, the question of its concordance with the image, of its indépendence even though it integrates an unified whole, as well as its connection with the senses and their transcendence. Images as material substrate and music as element of consciousness in this director-composer's work retain their autonomy of sense while the score is closely connected to the film'structure. They are consistent with a complementary relation that questions an entire paradigm based on the idea of analogy or convergence, in short, of a duality between images and music in cinema.

Por promover a drástica alteração no formato do fruto, característica esta que pode influenciar a preferência do consumidor, essa mutação é de grande interesse agronômico. Sob o ponto de vista biológico, o maior conhecimento da função desse gene resultaria em um maior entendimento do processo de frutificação. A análise de duplos mutantes ovate DR5::GUS e ovate ARR5::GUS mostrou que essa mutação altera o padrão de resposta a auxina e citocinina, respectivamente, durante o desenvolvimento do fruto. Além disso, foram realizados estudos que mostraram seu impacto no teor de sólidos solúveis, produtividade, tempo de prateleira, teor de matéria seca, peso das sementes e partenocarpia. Tais resultados não somente contribuíram para um melhor entendimento do efeito gênico da nova classe de genes regulatórios codificado por OVATE, como também fornecem importantes implicações agronômicas, já que a mutação ovate vem sendo utilizada na obtenção de variedades e híbridos do tipo \"grape\". / The fruits of tomato (Solanum lycopersicum) present a high phenotypic diversity, most of which can be attributed to mutations in five genes: SUN (SUN), OVATE (O), LOCULE NUMBER (LC), FRUIT SHAPE 8.1 (FS8.1) and FASCIATED (FAS). Whereas mutations in LC, O and FAS alter fruit locule number, mutations in FS8.1, SUN and O result in elongated fruits. The latter is the focus of the present work. ovate is a biologically and agronomically relevant mutation, however, the exact effect of the new class of regulatory gene it represents is unknown. The drastic alteration in fruit shape produced by the mutation can influence consumer preference, thus rendering it agronomically important. Biologically, a thorough characterization of its gene function would result in deeper understanding of fruit formation. Crosses to the marker lines DR5::GUS and ARR5::GUS showed that ovate alters the response pattern of auxin and cytokinin, respectively, during fruit development. Furthermore, studies are described showing its effect on total soluble solids content, yield, shelf life, dry mass rate, seed weight and parthenocarpy. The results contribute not only to a better understanding of the effect of the new class of regulatory genes encoded by OVATE, but also have important agronomic implications, given that the ovate mutation has been used in the obtention of \'grape/plum\' tomato varieties and hybrids.
