L'esthétique fonctionnelle de l'appareillage informatique comme ancrage phénoménologique de l'oeuvre à l'époque des immatériaux / The functional aesthetics of computer apparatus as a phenomenological anchoring of the artwork in the age of immateriality

Nous ne sommes plus à l’âge où les destructions poétiques d’appareils se voient car les régimes d’appareils sont devenus transparents : lorsque nous pressons un écran tactile, nous touchons ce que nous ne voyons plus, nos empreintes, et nous voyons ce que nous ne toucherons jamais, le plan numérique. Pour l’artiste contemporain, l’interface haptique – point d’orgue d’une technologie stéréotypée de la percussion – est le parergon d’une échelle inédite, toute tactile. De la machine à écrire aux claviers de plus en plus fins du design informatique, à mesure que l’on croit voir le bloc imaginaire céder sous la pression ergonomique de l’empire cybernétique, se palpe en fait un réel d’emblée en dehors des formes et à l’intérieur de notre doigté. C’est l’espace de la corne aux doigts. Ce transfert, cette remontée du point d’impact dans son élan, ce retour de la forme dans sa structure font du moment de touche – de l’appareillage comme manœuvre de départ – un isolat capital pour reposer la question de la sculpture à l’époque des immatériaux. Bien en face et à contre-courant de la surface informatique, une épaisseur sensible fonctionnelle aurait lieu et pourrait donc faire œuvre ; telle est la thèse proposée ici. Le comportement machinal de l’utilisateur d’interfaces sera considéré comme un modèle poïétique ou pensé comme une partie de l’imagination en attente de traduction plastique. Pendant que se joue l’expérience rythmique de l’interactivité, le corps ne doit-il pas fournir un effort perceptif pour ne pas s’absenter dans une partition photo-digitale jouée par avance ? Ne doit-il pas exprimer son ancrage particulier, calleux et bruissant – nous l’appellerons dactylo-phonique – au contact de la matière du monde ? Sans cette expression, jamais le design technologique contemporain, bi ou tridimensionnel, modélisé et assisté par ordinateur, n’entamera de rapport véritablement efficient avec le réel de nos habitudes et de nos démarches ; bref avec tout le poids incarné de notre conscience imageante : la perte d’indicialité instaurerait-elle un type introverti de phénoménalité ? / Poetic destructions of apparatus are no longer visible as regimes of apparatus have become transparent: when we press a tactile screen, we touch what we cannot see anymore – our fingerprints – and we see what we will never be able to touch – the digital space. For contemporary artists, haptic interfaces are the finger-tipping point of a technological dynamics of percussion but also the parergon of a new and entirely tactile scale. From the first typewriters to the latest ultra thin keyboards designed by computer manufacturers, we may think imagination is yielding under the ergonomic pressure of the cybernetic world. But in fact, existing outside of the shapes and right under our fingertips lies a palpable reality – the space of calluses. This transfer, this sensory feedback, this return of the form to its structure turns this casting off of the apparatus into an essential isolate to rethink sculpture in the age of immateriality. Right in front of the computer surface but working against it, there would be a functional sensitive thickness – an art-making place. This is our thesis. The machine-like behaviour of the interface user will be considered as a poietic model or regarded as a part of imagination waiting for plastic translation.We may wonder whether the rhythmic experience of interactivity does not entail a perceptual bodily effort on our side in order not to remain absent during the performance of a somewhat predictable photo-digital score.Doesn't this peculiar, callous and rustling – or dactylo-phonic – anchoring of the body to the substance of the world need to express itself? Without this expression, contemporary technological design — whether bi or tri-dimensional, modeled or computer-assisted — will never initiate a truly efficient relation with the real experience of our habits and actions, i.e. with all the embodied strength of our image-making consciousness: does the loss of indiciality create an introverted type of phenomenality ?

Avaliação da reconstituição da função tímica e a caracterização das subpopulações de linfócitos T e do perfil de citocinas em pacientes submetidos ao transplante alogênico de células-tronco hematopoiéticas que desenvolveram doença do enxerto-contra-hospedeiro / Evaluation of thymic function recovery and characterization of T lymphocyte subpopulations and cytokines profile in patients underwent to allogeneic hematopoietic stem cell transplantation that developed graft-versus-host disease

O transplante de células-tronco hematopoiéticas tem sido a melhor opção terapêutica para muitas doenças. Seu sucesso, no entanto, depende de alguns fatores que influenciam a taxa de mortalidade. A doença do enxerto-contra-hospedeiro é uma causa de mortalidade. Apresenta duas formas, a aguda e a crônica, e ambas têm linfócitos T na patofisiologia. Outra causa são as infecções, cujos patógenos variam conforme o tempo de recuperação do sistema imune. O presente estudo avaliou a recuperação linfoide T com base no timo e distinção das subpopulações e citocinas nos pacientes que desenvolveram doença do enxerto-contra-hospedeiro crônica no primeiro ano de transplante. Os pacientes foram alocados no Hospital de Transplante Amaral Carvalho (Jaú/SP) e tinham entre 18 e 60 anos de idade. No pré-transplante, foram comparados com indivíduos saudáveis, pareados em idade. Após o transplante, todos os pacientes foram acompanhados por um ano e seis meses, para observação de possíveis eventos de doença do enxerto-contra-hospedeiro e/ou infecções. A análise laboratorial foi feita com o sangue do paciente, sendo a primeira antes do transplante, seguidas por mais quatro, com intervalos de três meses. Tais avaliações laboratoriais tinham por objetivos caracterizar a função tímica por quantificação de sjTREC; as subpopulações de linfócitos T, por citometria de fluxo e a análise de citocinas, por Luminex. Foram estudados 172 indivíduos, sendo 75 do transplante alogênico, 43 do transplante autólogo, além de 54 pessoas saudáveis. Nossos resultados mostraram que os pacientes apresentaram função tímica diminuída antes mesmo da realização do transplante. A função tímica enfraquecida foi um fator de risco para doença do enxerto-contrahospedeiro crônica e a recuperação do sistema imune foi melhor nos pacientes que não apresentaram doença do enxerto-contra-hospedeiro crônica após um ano do transplante. No entanto, a função tímica não foi diferente entre os pacientes que faleceram e os que estão vivos. No geral, não houve distinção quanto à apresentação das subpopulações de linfócitos T e à produção de citocinas entre os pacientes submetidos ao transplante alogênico que desenvolveram doença do enxerto-contra-hospedeiro crônica, relativamente aos demais pacientes. Com o tempo, foi observada a recuperação gradual do compartimento linfoide T e diminuição na incidência de infecções. A taxa de infecções não influenciou a apresentação das subpopulações de linfócitos T e a produção de citocinas nos pacientes que desenvolveram doença do enxerto-contrahospedeiro crônica em relação aos demais pacientes. Como conclusão, a função tímica apresentou-se deteriorada antes da realização do transplante, porém não foi determinante para que houvesse piora na evolução clínica após o transplante. Os pacientes que desenvolveram doença do enxerto-contra-hospedeiro crônica, independentemente da incidência de infecções, apresentaram semelhança no perfil das subpopulações de linfócitos T e das citocinas, quando comparados aos demais / Hematopoietic stem cell transplantation has been the best therapeutic option for many diseases. Its success depends on some factors that influence the mortality rate. Graftversus- host disease is one cause of mortality. It has two forms, acute and chronic, and both have T lymphocytes in their pathophysiology. Other causes are infections, whose pathogens vary according to immune system recovery time. The present study evaluated the lymphoid T recuperation through the thymus and the differentiation of subpopulations and cytokines in patients who developed chronic graft-versus-host disease at the first year of transplantation. The patients were allocated at Hospital de Transplantes Amaral Carvalho (Jaú/SP). They were between 18 and 60 years old. At pre-transplant phase, patients were compared to healthy individuals, age-matched. After transplantation, they were all assisted for one year and six months, so that graft-versushost disease\'s and infections\' events could be observed. The laboratory analysis was done by blood sample; the first occurred before the transplant, followed by four more, every three months. Laboratory examinations were performed to characterize thymic function by quantification of sjTREC; T lymphocyte subpopulations, by flow cytometry, and cytokine analysis, by Luminex. A total of 172 individuals were studied: 75 allogeneic transplant patients, 43 autologous transplant patients and 54 healthy persons. Our results showed that patients presented thymic function impaired even before the transplant. The weakening of the thymic function was a risk factor for chronic graft-versus-host disease and immune system recovery was better in patients who did not develop chronic graft-versus-host disease after one year of transplantation. However, the thymic function was not different between patients who died and those who remained alive. In general, there was no distinction of the presentation of T lymphocyte subpopulations and cytokines production among patients who underwent allogeneic transplant and developed chronic graft-versus-host disease, in comparison to the other patients. Over time, a gradual recovery of the T lymphoid compartment and a decrease in the infections incidence were observed. The infection rate did not influence the presentation of the T lymphocyte subpopulations and the production of cytokines in patients who developed chronic graft-versus-host disease in comparison to the other patients. In conclusion, the thymic function was impaired before transplantation, but it was not relevant for clinical evolution worsening after transplantation. Patients who developed chronic graft-versus-host disease, regardless of the incidence of infections, showed similar profile of T lymphocytes subpopulations and cytokines, relatively to other patients

Pacientes submetidos ao transplante de medula óssea, apresentando hematúria/cistite hemorrágica, tiveram amostras de urina analisadas pela microscopia eletrônica. Esta foi a técnica escolhida de pesquisa viral pela sua confiabilidade. Noventa em 402 pacientes submetidos ao transplante de medula óssea neste serviço apresentaram hematúria/cistite hemorrágica (incidência de 22%). O estudo por microscopia eletrônica foi realizado em 72 destes pacientes com hematúria/cistite hemorrágica (grupo de estudo), identificando 55,6% (40/72) de positividade viral. Foram também estudadas amostras de urina de 12 pacientes submetidos ao transplante de medula óssea sem hematúria/cistite hemorrágica (grupo controle); houve associação significante entre a presença de vírus e hematúria/cistite hemorrágica (p<0,01 - Teste exato de Fisher). No grupo com hematúria/cistite hemorrágica, 65% (26/40) dos vírus detectados pertenciam à família Poliomaviridae, 30% (12/40) à Adenoviridae, e 5% (2/40) foram positivos para ambas as famílias. Houve associação entre a positividade para vírus e a presença da doença enxerto-contra-hospedeiro (p=0,05-x2) e o de início tardio (>dia+21) da H/CH (P=0,04-x2), bem como entre a doença enxerto-contra-hospedeiro e a severidade da H/CH (p=0,04-x2). O presente estudo mostra que a microscopia eletrônica é uma ferramenta útil para detecção de vírus na urina de pacientes submetidos ao transplante de medula óssea apresentando hematúria/cistite hemorrágica e que o vírus provavelmente tem um papel no desenvolvimento do sangramento urinário, o qual é agravado pelo doença enxerto-contra-hospedeiro. A microscopia eletrônica deveria ser realizada rotineiramente para guiar as outras técnicas de detecção viral, como a imunocitoquímica e biologia molecular. / Patients submitted to bone marrow transplantation presenting hematuria/hemorrhagic cystitis had the urine analyzed by electron microscopy. It was the elected technique for viral search because its reliability. Ninety from 402 patients submitted to bone marrow transplantation at this service showed hematuria/hemorrhagic cystitis (incidence of 22%). Electron microscopy study was performed in 72 of these patients with hematuria/hemorrhagic cystitis (study group), identifying 55.5% (40/72) of viral positivity. The present study shows that electron microscopy is a useful tool for detection of virus in urine of patients submitted to bone marrow transplantation presenting hematuria/hemorrhagic cystitis and that the virus probably play a role in the development of the urinary bleeding, which is aggravated by graft-versus-host disease. Electron microscopy should be performed routinely to guide the other techniques for viral detection, like immunocitochemistry and molecular biology.

O transplante de células tronco hematopoéticas alogênico e autogênico na leucemia mielóide aguda em primeira remissão completa: analise de 62 pacientes. Os pacientes foram submetidos a transplante de células tronco hematopoéticas alogênico e autogênico. Ao final do estudo estavam vivos no alogênico 43,3% e no autogênico 62,5%. Consolidação intensiva teve melhor sobrevida no alogênico. Os pacientes com DECH aguda grau II tiveram melhor sobrevida. Dois pacientes com DECH crônica extensa morreram. Óbito por infecção ocorreu com maior freqüência no alogênico seguido de recidiva. No autogênico a recidiva foi a principal causa de óbito. Morte por toxicidade ocorreu em 47% dos pacientes que foram a óbito no alogênico e em 8,3% no autogênico. Na analise múltipla de Cox a consolidação intensiva e DECH crônica, tiveram significância. / The allogeneic and autologous hematopoietic stem cell transplantation in acute myeloid leukemia in first complete remission: analyses of 62 patients. The patients were submitted to allogeneic and autologous hematopoietic stem cell transplantation. The end of the study were kept alive in allogeneic 43,3% and in autologous 65,2%. Patient in allogeneic who were consolidated had better survival. Patients with acute GVHD grade II had better survival. Two patients with chronic GVHD in intense, died. Infection was the most frequent dead cause in allogeneic following relapse. In autologous the relapse was the principal cause of death. Toxicity occurred in 47% of patients who died in allogeneic and 8,3% in autologous. In cox multiple analyses intensive consolidation and chronic GVHD had significance.

Untersuchungen zum differenzierten Wirkungsprofil von Glucocorticoiden in humanen mononukleären Zellen des peripheren Blutes

Qualitativ unterschiedliche genomische und nichtgenomische Mechanismen vermitteln die starken anti-inflammatorischen und immunmdulatorischen Eigenschaften der Glucocorticoide (GC). Der genomisch vermittelte Mechanismus ist bereits gut untersucht und dokumentiert, während der nichtgenomisch vermittelte Mechanismen noch einen Gegenstand vielseitiger Untersuchungen darstellt. Wir haben uns daher die Frage gestellt, ob Beclometason und Clobetasol besonders geeignet für die topische Applikation sind, weil sie sich in ihrem Wirkungsspektrum von systemisch zu applizierenden GC wie Dexamethason unterscheiden. Wir verglichen dazu die Effekte auf den Sauerstoffverbrauch mittels der Clark-Elektrode (nichtspezifisch nichtgenomischer Mechanismus), auf die IL-6-Synthese mittels ELISA (genomischer Mechanismus) und auf die Apoptose mittels Durchflusszytometrie (nichtgenomischer und genomischer Mechanismus) in ruhenden und stimulierten humanen PBMC. Dabei zeigten Beclometason und Clobetasol in sehr niedrigen Konzentrationen (10-10, 10-8 M) einen stärkeren Effekt auf den Sauerstoffverbrauch, waren aber in hohen Konzentrationen (10-5, 10-4 M) weniger potent im Vergleich zu Dexamethason. Auch hinsichtlich ihrer genomischen Potenz waren die topischen GC in einer Konzentration von 10-10 M und 10-8 M effektiver als Dexamethason, in höheren Konzentrationen unterschieden sie sich aber nicht. Alle drei GC induzierten Apoptose konzentrationsabhängig und unterschieden sich nicht in Konzentrationen zwischen 10-8 M und 10-5 M. In einer Konzentration von 10-4 M war die Induktion von Apoptose durch die topischen GC in PBMC und Jurkat-T-Zellen aber signifikant stärker im Vergleich zu Dexamethason. Diese Ergebnisse zeigen, dass sich topische und systemische GC in ihrer genomischen und nichtgenomischen Potenz signifikant unterscheiden. Es ist daher davon auszugehen, dass nichtgenomische Effekte eine deutlichere klinische Relevanz besitzen als bisher angenommen. / Several different genomic and non-genomic mechanisms mediate the important anti-inflammatory and immunomodulatory effects of glucocorticoids (GCs). The genomic effects are the most important while the clinical relevance of non-genomic actions is still a matter of debate. We therefore investigated whether beclomethasone and clobetasol are particularly suitable for topical application because they differ in their spectrum of activity from systemically administered GCs such as dexamethasone. We compared effects on oxygen consumption as measured with a Clark electrode (nonspecific non-genomic glucocorticoid effects), on interleukin-6 synthesis by means of ELISA (genomic effects) and on apoptosis using flow cytometry (non-genomic and genomic effects) in quiescent and mitogen-stimulated PBMCs. Beclomethasone and clobetasol had stronger effects on the oxygen consumption of quiescent and stimulated cells at lower concentrations (10-10, 10-8 M) but were less potent at higher concentrations (10-5, 10-4 M) in comparison with dexamethasone. Also in terms of genomic potency, topical GCs were more effective than dexamethasone at 10-10 M and 10-8 M but gave similar results at higher concentrations. The ability of all three GCs to induce apoptosis was found to be concentration-dependent and similar at concentrations between 10-8 and 10-5 M but, compared with 10-4 M dexamethasone, 10-4 M beclomethasone or clobetasol was significantly more effective at inducing apoptosis in both PBMCs and Jurkat T cells. These results show that systemic and topical GCs differ significantly in their ability to induce genomic and non-genomic effects. This suggests that non-genomic effects are more therapeutically relevant in certain clinical conditions than currently assumed.

Auditory immersion and the believability of a first-person perspective in computer games : Do players have a preference between mono and stereo foley, and is one perceived as more believable?

Based on previous research on spatial attributes in foley and the concept that auditory immersion in first-person perspective computer games is enhanced by believable sound effects, this study explores if there is a connection between stereo foley and the believability of the first-person perspective, and regardless, if there is a preference to either mono or stereo foley. An interactive listening test was created in unreal engine 4, where 20 subjects, all considered gamers, played three levels that differed visually and in auditory content. In these levels, subjects auditioned two versions of avatar-related foley sounds. One version was mono, the other stereo. The test prompted the subjects to complete two tasks for each level, whereupon the foley version changed upon completion of the first task. The subjects then answered questions in between each level, regarding the foley version. They were asked to rate believability and choose a preference, as well as provide motivations for their choices. The quantitative data showed next no evidence that either mono or stereo was generally perceived as more believable or preferred. However, the qualitative data indicates that the majority of players tend to prefer and rate stereo foley as more believable in certain game environments. Furthermore, the data indicates that some subjects prefer a sensory replication of reality in foley. It is also shown that preference for stereo width vary between subjects and therefore argued that there cannot be a perfect standardized setting for stereo foley.

Óbito por infecção ocorreu com maior freqüência no alogênico seguido de recidiva. No autogênico a recidiva foi a principal causa de óbito. Morte por toxicidade ocorreu em 47% dos pacientes que foram a óbito no alogênico e em 8,3% no autogênico. Na analise múltipla de Cox a consolidação intensiva e DECH crônica, tiveram significância. / The allogeneic and autologous hematopoietic stem cell transplantation in acute myeloid leukemia in first complete remission: analyses of 62 patients. The patients were submitted to allogeneic and autologous hematopoietic stem cell transplantation. The end of the study were kept alive in allogeneic 43,3% and in autologous 65,2%. Patient in allogeneic who were consolidated had better survival. Patients with acute GVHD grade II had better survival. Two patients with chronic GVHD in intense, died. Infection was the most frequent dead cause in allogeneic following relapse. In autologous the relapse was the principal cause of death. Toxicity occurred in 47% of patients who died in allogeneic and 8,3% in autologous. In cox multiple analyses intensive consolidation and chronic GVHD had significance.

Acting and its refusal in theatre and film.

This thesis examines works of theatre and film that explore a refusal of acting. Acting has traditionally been considered as something false or as pretending, in opposition to everyday life, which has been considered as something real and truthful. This has resulted in a desire to refuse acting, evident in the tradition of the anti-theatrical prejudice where acting is considered to be seductive and dangerous. All the works that I examine in this thesis are relatively recent and all of them explore the paradox that in our (postmodern) times a gradual reversal has occurred where everyday life is seen as more and more false or as pretending or simulating (ie. containing acting and theatricality) and conversely, acting in theatre and film has become the place where people have begun searching for reality and truth and where ‘acting’ and pretending in life can be revealed and refused. The result of this paradox - and what I also discuss as a confusion of acting and living - is that the place in which acting can be refused has shifted; the ethical desire to refuse acting (in theatre and in life) is turning up in the aesthetic domain of acting itself. In my first chapter I study works by filmmaker István Szabó and playwright Werner Fritsch, who represent the desire to refuse acting in the context of fascism where theatrical and filmic spectacle was used by the Nazis to seduce the population and where actors during this period also experienced an inability to separate their political and artistic lives. In my second chapter I look at the way Genet’s The Balcony and Ang Lee’s Lust, Caution explore the desire to refuse acting as a result of a confusion of acting and living in the context of sexual (sadomasochistic) role-play. And in my third chapter I examine the way Warhol’s The Chelsea Girls, von Trier’s The Idiots and Affleck’s I’m Still Here represent a refusal of acting and theatricality altogether, responding to the way that ‘acting’ in life may have become an all-pervasive substitute (a simulation) for living. Foundational to the development of this thesis and a major source of material is my analysis of three theatrical productions with Free Theatre Christchurch, directed by Peter Falkenberg, in which I was involved as an actor and in which a refusal of acting was explored.

Chapter 1: In Search of Innate Leadership : Discovering, Evaluating and Understanding Innateness

Every individual is born with different natural competencies that can be honed by both voluntary and involuntary environmental stimuli. The response our genotype decides to make, if any, towards those stimuli, determines how well our competencies develop. Each person’s coding and variations of genes will result in unique qualities in their phenotype, or physical structure. As a result, a person has various traits that are displayed through their behavior. DNA is genetically shown to express itself through traits by up to 75%. This leaves a sort of buffer of around 25%. This region is available for us to adapt to our environmental stimuli. Your innate qualities will not reach their full potential without stimulation from the environment, in a leadership case, with education and training and therefore it can be argued that environmental exposure is necessary to fully expose the potentials and capabilities of an individual, rather than instill a new skill or develop a talent that was not existent before. Innate leadership is not a permanent state, on the contrary, it is a continuously adaptive situation demanding contextual evolutionary changes or resignation from the subject occupying the role. When the needs and demands of a society or era outweigh the relevance of the innate leaders' traits and competencies, an evolution of leadership is needed to maintain a positive relationship between all parties involved. As a result, the innate leader will begin to lose their innateness in their role and unless they evolve and adapt (because the two actions are not the same) to new contextual needs, their tenure as leader will begin to be detrimental and counter-functional. What we want to put forward is a real, universal and constructive understanding of what makes a human happy, motivated and productive and how an innate person in context is a much better solution in the short and long run, for those around them when put to a task.
