Untersuchungen zum Postantibiotischen Effekt bei Pseudomonas aeruginosa-Isolaten einer Intensivstation

In der vorliegenden Untersuchung wurde der Postantibiotische Effekt (PAE) von Amikacin und Ceftazidim alleinig und in Kombination beider Antibiotika nach einmaliger und mehrfacher Exposition bei Pseudomonas aeruginosa bestimmt. Es wurden verschiedene Stämme mit unterschiedlicher Resistenz gegen Amikacin und Ceftazidim untersucht. Die MHK-Werte bewegten sich für Ceftazidim zwischen < 0,25 µg/ml bis 4 µg/ml und für Amikacin zwischen < 2 µg/ml bis 32 µg/ml. Der PAE lag nach der einmaligen Inkubation von Amikacin bei 0 bis 2,65 Stunden, von Ceftazidim bei 0 bis 2,78 Stunden und in der Kombination beider bei 2,4 bis 5,37 Stunden. Außerdem ergab die mehrmalige Exposition bei Amikacin nach der 1. Inkubation einen PAE von 0,75 bis 2,25 Stunden und stieg auf 2,3 bis 3,5 Stunden nach der 3. Inkubation an; bei Ceftazidim von 1,1 bis 2,18 Stunden Anstieg auf 1,18 bis 2,5 Stunden und bei der Kombination von Amikacin und Ceftazidim von 2,3 bis 3,75 Stunden war ein geringer Abfall auf 1,25 bis 3,25 Stunden zu verzeichnen. Es wurde ein Zusammenhang zwischen dem PAE und der MHK beobachtet: je höher die Resistenz, desto kürzer der PAE. Die Dosierungsintervalle wurden so gewählt, daß sie der klinisch üblichen dreifach Applikation pro Tag entsprachen. Aus unseren Untersuchungen läßt sich theoretisch eine einmalige Applikation pro Tag für Aminoglykoside und eine Kombinationstherapie Aminoglykosid plus Beta-Lactamantibiotika ableiten; vor allem auch, daß bei Mehrfachapplikation der PAE nicht kürzer wird. Für Ceftazidim erscheint eine Dauerinfusion bei fehlendem PAE sinnvoll. / In the current study we examinated the postantibiotic effect (PAE) of the antimicrobial agents amikacin and ceftazidime in vitro. We analyzed the PAE using both agents alone and in combination and after once and several expositions of different isolates of Pseudomonas. Different resistant stains of Pseudomonas were explored versus amikacin and ceftazidime. The observed minimum inhibitory concentration values (MIC) for ceftazidime ranged from < 0.25µg/ml to 4.0µg/ml and for amikacin from < 2.0µg/ml to 32µg/ml. After unique incubation the PAE of amikacin ranged from 0 to 2.65 hours while ceftazidime ranged from 0 to 2.78 hours. In combination of both we observed PAEs between 2.4 to 5.37 hours. A several exposition of amikacin showed after the first incubation PAEs between 0.75 to 2.25 hours increasing after third incubation from 2.3 to 3.5 hours whereas ceftazidime ranged from 1.1 to 2.18 hours and 1.18 to 2.5 hours. Both antimicrobial agents in combination had PAEs between 2.3 and 3.75 hours after first incubation and decreased low after third incubation between 1.25 and 3.25 hours. There is significant correlation to be seen between PAE and MIC-values: the higher resistance is, the shorter PAE will become.

Immune modulation by parasites

Die Infektion mit Schistosoma mansoni resultiert in einer Th2-Immunantwort mit Eosinophilie und erhöhtem IgE-Titer, wobei der wasserlösliche Extrakt der S. mansoni Eier (SEA) ausreicht um diese Reaktion auszulösen. In der vorliegenden Arbeit konnte demonstriert werden, dass sich IL-4-produzierende CD4+ T-Lymphozyten in Zellkulturen mit SEA-konditionierten Dendritischen Zellen (DCs) trotz gleichzeitig vorkommenden IFN-gamma entwickeln und SEA die Expression von Faktoren in DCs, die üblicherweise mit einer Th1-Antwort einhergehen, auf Transkriptions- und Proteinebene selektiv hemmt. Um den Faktor aus S. mansoni Eiern zu isolieren, der zur Expression von IL-4 in CD4+ Zellen und zur Inhibition von IL-12 in DCs führt, wurde eine Gelfiltrationschromatographie der exkretorisch/sekretorischen Ei-antigene (ES) durchgeführt und die Fraktionen in vitro getestet. Darin wurde gezeigt, dass Fraktionen mit einer Proteinbande von 30 kD die Expression von IL-4 in CD4+ Zellen induzieren. Dieses ES-Protein wurde durch N-terminale Sequenzierung als hepatotoxische Ribonuclease Omega-1 identifiziert, welches ebenfalls die Expression von IL-12 in DCs inhibiert und die Produktion von IL-4 in CD4+ Zellen bei einer 10-fach geringeren Proteinkonzentration als mit dem Kontrollansatz SEA induziert. Zudem sollte untersucht werden, inwieweit Toll-like Rezeptoren in der Generierung einer Th2 Antwort gegen schistosomale Antigene involviert sind. Dazu wurden TLR2-, TLR3-, TLR4- und MyD88-defiziente Mäuse mit S. mansoni infiziert und immunologische und pathologische Daten in der akuten und chronischen Phase der Infektion analysiert. Demnach sind TLR2, TLR3, TLR4 und MyD88-abhängige Signaltransduktionswege nicht für eine-Th2 Antwort notwendig, jedoch ist letzteres Molekül in der Ausprägung der typischen Leberfibrose involviert. / Infection with Schistosoma mansoni results in the induction of a Th2 immune response, eosinophilia and increased levels of IgE. The water-soluble extract of S. mansoni eggs (SEA) is sufficient to promote TH2 polarization in a dendritic cell-dependent manner. In this thesis, it was demonstrated that IL-4+ CD4+ cells emerge in cultures with SEA-conditioned dendritic cells (DCs) in the presence of IFN-gamma and that SEA inhibits selectively the expression of IL-12 and co-stimulatory markers in DCs on the transcriptional and protein level. To identify the putative protein in S. mansoni eggs mediating a Th2 induction, a gel filtration chromatography of the excretory/secretory egg antigens (ES) was conducted and the fractions tested in vitro. Fractions containing a single band of 30 kD were sufficient to promote IL-4 induction in naïve CD4+ cells. Using N-terminal sequencing this ES-protein was identified as the hepatotoxic S. mansoni ribonuclease omega-1 which displayed both biological functions observed with SEA: inhibition of IL-12 in LPS-stimulated DCs and induction of IL-4+ CD4 cells at a 10 fold lower protein concentration than SEA. In order to understand, if the innate immune receptors TLR2, TLR3, TLR4 or the TLR adaptor molecule MyD88 are involved in the generation of the Th2 response against schistosomal antigens, the respective knock out mice were infected and immunological and pathological parameters were analyzed during acute and chronic phase of infection. This study showed that during S. mansoni infection TLR2, TLR3, TLR4 and TLR activation through the MyD88-dependent pathway are neither required for the induction (priming and polarization) nor for the down-regulation of Th2 responses, however, the fibrotic response against S. mansoni eggs was significantly reduced in MyD88-deficient mice suggesting a detrimental role of this pathway in liver pathology.

Untersuchung von intrazellulären Osmolytkonzentrationen im Hirn nach Dehydratation durch Ausdauerbelastung

Einleitung: Mit der vorliegenden Studie wurden zum ersten Mal beim Menschen die Auswirkungen von Dehydration (Dehy) und anschließender Rehydratation (Rehy) auf cerebrale volumenregulatorische Metabolite (myo-Inosit (mI), N-Azetylaspartat+N-Azetylaspartylglutamat (tNAA), Kreatin (Cr), Glyzerophosphocholin+Phosphocholin (Cho) und Glutamat+Glutamin (Glx)) sowie auf Flüssigkeitsverschiebungen untersucht. Methoden: 14 Radsportler (26.6 (22.7/29.8) Jahre, Median und 25./75. Perzentile) wurden mittels 1H-Spektroskopie (1H-MRS) in der okzipitoparietalen grauen Substanz (GM) und parietalen weißen Substanz rechts (WMR) und links (WML) untersucht (GE Signa Horizon 3T94; PRESS: TE 30ms, TR 6000ms, VOI 8ml). Die Messungen erfolgten vor, direkt nach Dehy und nach Rehy (180min, Zufuhr von 150\% der verlorenen Körpermasse (KM)). Zusätzlich wurde durch T2-Relaxationsmessungen der Atrophieindex alpha (Verhältnis cerebrales Gewebewasser (HW) zu Liquor (CSF)) bestimmt. Resultate: Die KM der Probanden reduzierte sich durch Dehy um 3.7 (3.4/4.1)% und stieg durch Rehy wieder um 4.5 (3.7/5.3)% an (Wilcoxon: p / Introduction: In the present study the influence of Dehy on cerebral volume regulatory metabolites (myo-Inositol (mI), N-Acetyl-aspartata+N-Acetyl-aspartyl-glutamate (tNAA), Creatine (Cr), Glycerophosphocholine+Phosphocholine (Cho) and Glutamate+Glutamine (Glx)) and fluid shifts has been investigated for the first time in humans. Methods: 14 cyclists (26.6 (22.7/29.8) y, median and 25./75. percentile) have been examined with proton NMR spectroscopy in the occipito-parietal gray matter (GM) and the right (WMR) and left (WML) parietal with matter (GE Signa Horizon 3T94; PRESS: TE 30ms, TR 6000ms, VOI 8ml). Spectra were acquired before, immediately after Dehy and after rehydration (Rehy). Rehy took place during 180min and 150% of lost body weight (BW) was substituted. Additionally the atrophy index alpha (ratio between cerebral water and liquor) was assessed (T2 signal decay as a function of echo time). O desenvolvimento da Medicina Nuclear, aliado à evolução dos equipamentos de diagnóstico e terapia, necessita, cada vez mais, da disponibilidade comercial de radioisótopos. Nesse contexto, o IPEN tem buscado atender e abastecer o mercado nacional. Um dos investimentos nesta área foi a aquisição de um ciclotron de 30 MeV, modelo Cyclone-30, que permitiu a produção dos radioisótopos tais como, o 18F, 67Ga, 201Tl e o 123I, sendo este último o foco do presente trabalho. Através de dados de produções rotineiras de 123I via irradiação com prótons em alvo gasoso de Xenônio com enriquecimento superior a 99,8% em 124Xe, foi realizado um estudo para identificar os fatores relevantes que influenciam diretamente o rendimento de obtenção de 123I com altíssimo grau de pureza. Embora a metodologia seja bem conhecida, quando se trata de produção comercial há uma escassez de dados sobre os parâmetros operacionais utilizados. Os parâmetros avaliados foram: pressão do gás 124Xe, intensidade de corrente de feixe de prótons, tempo de irradiação, temperatura de operação do sistema durante a irradiação, tempo de espera para formação de 123I, tempo de aquecimento do porta-alvo para recuperação do 123I formado, temperatura de aquecimento da solução de lavagem e influência do revestimento interno da câmara de irradiação com Ni. Com os resultados obtidos, foi possível alterar as condições operacionais nas produções rotineiras, conduzindo a um aumento de eficiência do processo em torno de 30%. / The development of diagnosis equipment and therapy procedures in nuclear medicine depends on the availability of commercial radioisotopes. IPEN is the most important institution that provides radioisotopes for national market. In order to achieve this function, IPEN had invested in the acquisition of a 30 MeV Cyclone-30 cyclotron to produce mainly 18F, 67Ga, 201Tl and 123I. The 123I production is the aim of the present work. With the 123I routine production data obtained by proton irradiation of Xe targets with an enrichment greater than 99.8%, it was possible to identify the important parameters that have direct influence on the production yield of high purity degree 123I. Even though the methodology for the commercial production of 123I, there are an scarcity of operational parameters data for this task. In this work the evaluated parameters were: 124Xe pressure, proton beam quality, irradiation time, operational temperature of the irradiation system under irradiation, waiting time to obtain 123I, temperature of washing solution and the impact of the internal Ni coating in the target. With the obtained results it was possible to modify the operational conditions for routine production and increasing the efficiency in about 30%.

Fragmentation in Proton-Nucleus Reactions from 100 to 1400 MeV

The heaviest fragments, recoils, have been studied in proton and deuteron induced 28Si reactions and proton-20Ne reactions at 100-300 MeV per nucleon. Inclusive charge and angular distributions and coincidences between He nuclei and recoils have been compared to two theoretical models, Dubna Cascade Model and JAERI Quantum Molecular Dynamics. The overall agreement was good for the reactions with 28Si, however the angular distributions of He fragments could not be reproduced. For the 20Ne reactions the recoil angular distributions were only reproduced for large angles. There was a significant underestimation at small angles and low recoil charge. α-clustering in the bombarding nucleus is a possible explanation for the deviations. In the 100 MeV per nucleon reactions all assumptions of the models may not be valid and the agreement was worst for these reactions. In proton-natXe reactions intermediate mass fragments have been studied from 200 to 1400 MeV. Slow ramping was used to scan the energy. Charge distributions and a caloric curve have been compared to Cascade Fragmentation Evaporation Model. Charge distributions showed good agreement for fragments with Z&lt;8 but the heavier fragments were underestimated.

Characterization of liquid crystals in porous materials by means of NMR of probe atoms and molecules

Abstract The present thesis describes a method for characterization of liquid crystals in confined spaces by means of NMR of probe atoms and molecules. 129Xe isotope enriched xenon gas and 13C isotope enriched methyl iodide and methane were used as probes. Behavior of solutes and liquid crystals confined to porous materials was investigated using 129Xe and 13C NMR spectroscopy. Uniaxial nematic liquid crystals Phase 4 and ZLI 1115 were used as a medium. Controlled pore glass with well defined and known properties was used as a porous material. The behavior of liquid crystals and solutes in various different size pores, temperatures and magnetic fields at different solute concentrations was explained. The average pore diameter of the material varied from mesopores to macropores. The studied temperature range covered solid, nematic and isotropic phases of bulk liquid crystals, and the highest magnetic field was 2.5 times stronger than the lowest one used (4.70 T). The chemical shifts, intensities, and line shapes of the resonance signals from the solutes were observed to contain lots of information about the effect of confinement on the state of the liquid crystals. Especially the line shape of the 13C resonances of methyl iodide was observed to be very sensitive to the liquid crystal orientation distribution in the pores. By varying experimental conditions the relative contribution of field and the surface forces of pore walls to the orientation of liquid crystal molecules inside the pores was seen to change quite drastically. In addition, it was also observed that when the sample is cooled very rapidly, xenon atoms do not squeeze out from the freezing medium but they are occluded inside the solid lattice, and their chemical shift is very sensitive to crystal structure. Furthermore, because solutes experienced on average isotropic environment inside the smallest pores, isotropic value of the shielding tensor could be determined at exactly the same condition and temperature as anisotropic counterpart between the pore particles. Thus, for the first time in the solution state, shielding anisotropies could be determined as a function of temperature.

NMR-SPEKTROSKOPIE AN FLEXIBLEN UND CHIRALEN METAL-ORGANIC FRAMEWORKS (MOFs)

Es wurden verschiedene NMR-spektrokopische Messungen an flexiblen und chiralen MOFs durchgeführt. Zur Untersuchung der Porensysteme kamen 129Xe-NMR und 13C-NMR an adsorbiertem CO2 zum Einsatz, während die MOF-Gitter und ihre Wechselwirkungen mit adsorbierten Gastmolekülen mittels 13C- und 1H-MAS-NMR-Spektroskopie studiert wurden. Während DUT-8(Ni) Flexibilität zeigt, weist DUT-8(Cu) ein starres Gitter auf. Die Flexibilität der sogenannten Solid-Solutions hängt in ausgeprägter Weise vom Verhältnis der funktionalisierten bdc-Linker 2,5-bme-bdc und db-bdc ab. Dieses Verhältnis hat zudem einen großen Einfluss auf die Orientierung der adsorbierten CO2-Moleküle. Es wurde erstmals eine Methode vorgestellt, die den Festkörper-NMR-spektroskopischen Nachweis chiraler Seitengruppen in chiralen MOFs erlaubt, wie anhand des chiral modifizierten UMCM-1 (ChirUMCM-1) demonstriert wurde. Die Chiralität kann einen NMR-spektroskopisch messbaren Einfluss auf die intrinsische Dynamik des MOF-Gitters ausüben, wie am chiral modifizierten DUT-32 deutlich wurde, dessen chirale Seitengruppe selektiv 15N- und 13C-isotopenmarkiert wurde.

Des années 50 à nos jours, la propulsion électrique n'a cessé d'évoluer afin de s'imposer dans le domaine de la propulsion spatiale. Les Propulseurs à effet Hall (PEH) sont principalement utilisés pour des missions de correction de trajectoire ou de maintien en orbite des satellites. Ils délivrent des faisceaux d'ions à forte densité de courant et à faible énergie, ce qui en font de bons candidats potentiels pour d'autres applications comme la microélectronique ou encore les traitements de surfaces. Le xénon est l'ergol le plus utilisé en raison de sa masse élevée et de son faible énergie d'ionisation. Cependant son coût élevé et la difficulté d’approvisionnement motivent la recherche d'alternatives pour le fonctionnement des MEH. C'est dans ce cadre que cette thèse s'est inscrite avec l'idée d'un développement d'une source de faible puissance fonctionnelle en argon. L'amorçage d'une telle décharge n'étant pas immédiat, une démarche progressive qui passe par des décharges de mélange de gaz a été adoptée. Les décharges Xe-Ar se sont révélées très intéressantes pour la compréhension des mécanismes physiques qui régissent les PEH. La caractérisation en vitesse des ions Xe II (par Fluorescence Induite par Laser) associée à l'analyse en énergie par RPA a permis de remonter à des informations utiles sur les zones d'ionisation et d'accélération. Une technique originale de résolution temporelle du RPA basée sur une interruption rapide de la décharge ou sur les oscillations naturelles du courant de décharge, a été développée et a permis l'identification et la quantification des différentes espèces présentes dans le jet d'ions. Grâce aux résultats de l'étude paramétrique des décharges de mélange Xe-Ar, une décharge d'argon pur a pu être amorcée et caractérisée pour la première fois dans un PEH de faible puissance. / Since the 50s, electric propulsion has improved in order to establish itself on space propulsion field. The Hall Effect Thruster (HET) are mainly used for trajectory correction or satellites orbit maintaining. The HET provide high current densities and low energy ion beam that making it a good candidate for other applications such as microelectronics or surface treatments. Xenon propellant is most commonly used due to its high atomic mass and its low ionization energy. However, the high cost and difficult supply of xenon, leads to looking for alternative propellant for HET operation. In this context, this PhD thesis had as goal the development of a functional Argon low power source. Argon discharge ignition is not immediate, that why a progressive approach which involves gas mixture discharges was adopted. The Xe-Ar discharge gives very interesting results for the understanding of physical mechanisms governing HET. The characterization of Xe II ions velocity (Laser Induced Fluorescence) associated to the energy analysis by RPA have provided access to useful information on ionization and acceleration areas. An original time resolved RPA technique, based on an ultra-fast discharge interruption or on the discharge current oscillations, has been developed. This technique allows the identification and the quantification of different species present in the ion beam. Thanks to the discharge Xe-Ar study, a pure argon discharge could be initiated and characterized for the first time in a low power HET.

Zur Untersuchung der Porensysteme kamen 129Xe-NMR und 13C-NMR an adsorbiertem CO2 zum Einsatz, während die MOF-Gitter und ihre Wechselwirkungen mit adsorbierten Gastmolekülen mittels 13C- und 1H-MAS-NMR-Spektroskopie studiert wurden. Während DUT-8(Ni) Flexibilität zeigt, weist DUT-8(Cu) ein starres Gitter auf. Die Flexibilität der sogenannten Solid-Solutions hängt in ausgeprägter Weise vom Verhältnis der funktionalisierten bdc-Linker 2,5-bme-bdc und db-bdc ab. Dieses Verhältnis hat zudem einen großen Einfluss auf die Orientierung der adsorbierten CO2-Moleküle. Es wurde erstmals eine Methode vorgestellt, die den Festkörper-NMR-spektroskopischen Nachweis chiraler Seitengruppen in chiralen MOFs erlaubt, wie anhand des chiral modifizierten UMCM-1 (ChirUMCM-1) demonstriert wurde. Die Chiralität kann einen NMR-spektroskopisch messbaren Einfluss auf die intrinsische Dynamik des MOF-Gitters ausüben, wie am chiral modifizierten DUT-32 deutlich wurde, dessen chirale Seitengruppe selektiv 15N- und 13C-isotopenmarkiert wurde.
