This study aims to evaluate the effect of base saturation and mineral fertilization on crop crambe. Experiments were set in two growing seasons. In the first year were montadsos experiments in pots in the greenhouse and in the year 2010 which evaluated independently, the effect of doses of (N, P, Zn and B). The doses evaluated in mg kg-1 of soil were: N (control, 20, 40 and 80), P (control, 20, 40 and 80), Zn (control, 1, 2, and 4), B (control, 0.5, 1, and 2.) We evaluated the following parameters: root dry weight per plant, shoot dry mass per plant, grain yield per plant and oil content of the grain. In the year 2011, which was the second year of cultivation, were assembled in three experiments to evaluate field conditions, in isolation, in a factorial design the effect of raising the base saturation and nitrogen levels, base saturation and doses Phosphorus and base saturation and potassium levels in the culture of crambe. Each of these experiments was formed by a factorial design (4x3) with 4 replicates totaling 48 installments, the common witness to all nutrients. In each experiment, four levels of saturation and three levels of each mineral fertilizers. The base saturation levels were: (34 - natural soil, 40, 50 and 60%). The mineral fertilizers were evaluated: a) Nitrogen - 1 = control, 2 = 40 kg ha-1 nitrogen, 3 = 80 kg ha-1 nitrogen, b) Phosphorus - 1 = control, 2 = 40 kg ha-1 P2O5, 3 = 80 kg ha-1 P2O5, c) potassium - 1 = control, 2 = 40 kg ha-1 of K2O, 3 = 80 kg ha-1 of K2O. The experimental plots were formed by a rectangle of 9 m2 with 5 lines of planting spacing of 0.45 m between them. The planting was held using the cultivar 'Bright FMS ". The results obtained in greenhouse experiments showed that the addition of nitrogen increased linearly all measured parameters. The addition of phosphorus levels increased linearly the development of root, shoot and grain production. Addition of zinc doses increased linearly shoot growth and grain yield. The addition of boron doses quadratically influenced the development of the root system. The experiments in the field showed that the addition of nitrogen increased the dry weight of roots and shoots, productivity and no influence on oil content. The phosphorus levels significantly increased all parameters evaluated. Potassium significantly increased all parameters except the oil content. The base saturation influenced all parameters in a linear or quadratic. / Ainda não há recomendações específicas para a cultura do crambe em relação a adubação com macro e micronutrientes. Este trabalho tem como objetivo avaliar o efeito da saturação por bases e adubação mineral na cultura do crambe. Foram instalados experimentos em dois anos de cultivo. No primeiro ano foram instalados experimentos em vasos e em casa de vegetação no ano de 2010 avaliando, de forma independente, o efeito de doses de (N, P, Zn e B). As doses avaliadas em mg kg-1 de solo, foram: N (testemunha, 20, 40 e 80), P (testemunha, 20, 40 e 80), Zn (testemunha, 1, 2, e 4), B (testemunha, 0,5, 1, e 2). Foram avaliados os seguintes parâmetros: massa seca de raiz por planta, massa seca da parte aérea por planta, produção de grãos por planta e teor de óleo dos grãos. No ano de 2011, que foi o segundo ano de cultivo, foram instalados três experimentos em condição de campo para avaliar, de forma isolada, em um esquema fatorial o efeito da elevação da saturação por bases e doses de nitrogênio, saturação por bases e doses de fósforo e saturação por bases e doses de potássio na cultura do crambe. Cada um desses experimentos foi formado por um fatorial (4x3) com 4 repetições totalizando 48 parcelas, sendo a testemunha comum para todos os nutrientes. Em cada experimento foram avaliados quatro níveis de saturação por bases e três níveis de cada uma das adubações minerais. Os níveis de saturação por bases foram: (34 - natural do solo, 40, 50 e 60 %). As adubações minerais avaliadas foram: a) Nitrogênio - 1=testemunha; 2= 40 kg ha-1 de nitrogênio; 3 = 80 kg ha-1 de nitrogênio, b) Fósforo - 1= testemunha; 2= 40 kg ha-1 de P2O5; 3= 80 kg ha-1 de P2O5, c) potássio - 1=testemunha; 2= 40 kg ha-1 de K2O; 3= 80 kg ha-1 de K2O. As parcelas experimentais foram formadas por um retângulo de 9 m2 com 5 linhas de plantio com espaçamento entre elas de 0,45m. O plantio foi realizado no dia 08/03/2011 utilizando a cultivar “FMS Brilhante”. Os resultados obtidos nos experimentos de casa de vegetação mostraram que a adição das doses de nitrogênio elevou de forma linear todos os parâmetros avaliados. A adição das doses de fósforo elevou de forma linear o desenvolvimento do sistema radicular, da parte aérea e a produção de grãos. A adição das doses de zinco elevou de forma linear desenvolvimento da parte aérea e a produção de grãos. A adição das doses de boro influenciou de forma quadrática o desenvolvimento do sistema radicular. Os experimentos em condições de campo mostraram que a adição das doses de nitrogênio aumentou a massa seca do sistema radicular e da parte aérea, a produtividade e não influenciou no teor de óleo. As doses de fósforo elevaram significativamente todos os parâmetros avaliados. O potássio elevou significativamente todos os parâmetros avaliados com exceção do teor de óleo. A saturação por bases influenciou todos os parâmetros avaliados de forma linear ou quadrática.

Characterization of heat shock protein 70-z (PfHsp70-z) from plasmodium falciparium

PhD (Biochemistry) / Department of Biochemistry / Malaria is a parasitic disease that accounts for more than 660 thousand deaths annually, mainly in children. Malaria is caused by five Plasmodium species P. ovale, P. vivax, P. malariae, P. falciparum and P. knowlesi. The most lethal cause of cerebral malaria is P. falciparum. The parasites have been shown to up-regulate some of their heat shock proteins (Hsp) in response to stress. Heat shock protein 70 (called DnaK in prokaryotes) is one of the most prominent groups of chaperones whose role is central to protein homeostasis and determines the fate of proteins. Six Hsp70 genes are represented on the genome of P. falciparum. The Hsp70 genes encode for proteins that are localised in different sub-cellular compartments. Of these two occur in the cytosol, PfHsp70-z and PfHsp70-1; two occur in the endoplasmic reticulum, PfHsp70-2 and PfHsp70-y; one in the mitochondria, PfHsp70-3 and one exported to the red blood cell cytosol, PfHsp70-x. PfHsp70-1 is a well characterized canonical Hsp70 involved in prevention of protein aggregation and facilitates protein folding. Little is known about PfHsp70-z. PfHsp70-z was previously shown to be an essential protein implicated in the folding of proteins possessing asparagine rich repeats. However, based on structural evidence PfHsp70-z belongs to the Hsp110 family of proteins and is thought to serve as a nucleotide exchange factor (NEF) of PfHsp70-1. The main aim of this study is to elucidate the functional roles of PfHsp70-z as a chaperone and its interaction with PfHsp70-1. In the current study, PfHsp70-z was cloned and expressed in E. coli JM109 cells. This was followed by its purification using nickel chromatography. The expression of PfHsp70-z in parasites cultured in vitro was investigated and its association with PfHsp70-1 was explored using a co-immuno precipitation assay. PfHsp70-z expression in malaria parasites is up regulated by heat stress and the protein is heat stable based on investigations conducted using Circular Dichroism. Furthermore, the direct interaction between recombinant forms of PfHsp70-z and PfHsp70-1 were investigated using slot blot and surface plasmon resonance assays. PfHsp70-z was observed to exhibit ATPase activity. In addition, the direct interaction between PfHsp70-z and PfHsp70-1 is promoted by ATP. Based on limited proteolysis and tryptophan fluorescence analyses, PfHsp70-z binds ATP to assume a unique structural conformation compared to the conformation of the protein bound to ADP or in nucleotide-free state. PfHsp70-z was able to suppress the heat-induced aggregation of malate dehydrogenase and luciferase in vitro. Interestingly, while ATP appears to modulate the conformation of PfHsp70-z, the chaperone function of PfHsp70-z was not influenced by ATP. Altogether, these findings suggest that Characterization of Heat Shock Protein 70-z (PfHsp70-z) from Plasmodium falciparum iii PfHsp70-z serves as an effective peptide substrate holding chaperone. In addition, PfHsp70-z may also serve as the sole nucleotide exchange factor of PfHsp70-1. The broad spectrum of functions of this protein, could explain this PfHsp70-z is an essential protein in malaria parasite survival. This is the first study to show that PfHsp70-z possess independent chaperone activity and that it interacts with its cytosolic counterpart, PfHsp70-1 in a nucleotide dependent fashion. Furthermore, the study shows that PfHsp70-z is a heat stable molecule and that it is capable of forming high order oligomers.

The thesis' aim is to explore the changes of the anarchofeminist zine Bloody Mary and how it was influenced by the Internet, as well as by the specific blogging platform and computer graphics software used in the zine's production. The chief sources of data used in my research are the specific issues of the zine, articles and archived documents published on the blog. Moreover, I have undertaken several interviews with the zine's authors and readers, giving me insight into the zine's production. The theoretical section of the thesis focuses on the function of zines as alternative media, as well as describing zines within the larger context of the key feminist views on the media industry and the history of independent feminist publications. The following section describes the formation of the Riot Grrrl movement, which has led the proliferation of zines within feminist discourse, as well as serving as a chief inspiration to Bloody Mary's authors. The next chapter describes the context of Czech anarchist activism and names some of the key groups such as the "Feminist Group of the 8th March" or the "Anarcho-feminist Group", which Bloody Mary's authors belonged to. Essays on redistributive policies and household finance with heterogeneous agents

The overall objective of the thesis is to investigate needs and incentives of all income/wealth groups in order to explore ways and means to remedy the excessive economic inequality. A closer examination of individual decisions across richer and poorer households allows us to recognize conflicts of wants, needs and values and subsequently to draw recommendations for future policies. The first chapter examines households' preferences over the redistribution of wealth resources. The preferences of voting households are restricted by agents' present and future resource constraints. The wealth resources vary over the business cycle, which affects the grounds for speculations of voting households. We augment the standard Real-Business-Cycle (RBC) model by the majority voting on lump-sum redistribution employing a balanced government budget. Our findings indicate that for the usual elasticity of labor supply both transfers' level and share of output are procyclical, with the procyclicality increasing in the discrepancy between richer and poorer households. In the second chapter we analytically demonstrate that all economic agents face subsistence costs that hinder economic and financial decisions of the poor. We find that the standard two-asset portfolio-selection model with a time-invariant subsistence component in the common-across agents Stone-Geary utility function is capable of explaining qualitatively and quantitatively three empirical regularities: (i) increasing saving rates in wealth, (ii) rising risky portfolio shares with wealth, (iii) more volatile consumption growth of the richer. On the contrary, "keeping-up-with-the-Joneses" utility with a time-varying weighted mean consumption produces identical saving rates and portfolio asset shares across richer and poorer agents, failing to match the micro data. Finally, in the third chapter we use Epstein-Zin-Weil recursive preferences altered to include subsistence costs, as this form of utility function enables trade-off between stability and safety. We pursue an analytical investigation of a more complex multi-asset portfolio-choice model with perfectly insurable labor risk and no liquidity constraints and find further support of the data evidence. If households' total resources are anticipated to increase over time, poorer agents can afford to gradually escape subsistence concerns by choosing lower saving rates and accepting only minor portfolio risks as their consumption hovers close to the subsistence needs. The calibration part of the model economy shows that analytical results can quantitatively reconcile the data, too.

Nanocristaux luminescents de phosphures d'indium et de zinc : synthèse, enrobage et caractérisation / Indium phosphide and zinc phosphide luminescent nanocrystals : synthesis, coating and characterization

Ce travail de thèse porte sur la synthèse organo-métallique de nanoparticules (NPs) semi-conductrices colloïdales de phosphures d’indium (InP), de zinc (Zn3P2) et de structures cœur/coquille obtenues par la croissance d’une couche de sulfure de zinc (ZnS) à la surface des NPs. Les objectifs consistent à comprendre et maîtriser la synthèse dans le but de décaler les longueurs d’onde d’absorption et d’émission vers le proche infra-rouge, domaine spectral intéressant pour l’imagerie biomédicale.Le premier chapitre présente l’état de l’art sur les nanocristaux (NCx) d’InP et d’InP/ZnS. Un bref rappel sur les propriétés physico-chimiques des NCx semi-conducteurs est présenté et différentes synthèses sont décrites. Une attention toute particulière a été portée sur la taille des NCx, le décalage de l’émission de fluorescence vers les plus grandes longueurs d’onde et l’optimisation des rendements quantiques. Les potentialités offertes par ces objets soit pour les diodes électroluminescentes (LED) blanches soit pour l’imagerie biomédicale montrent l’intérêt d’utiliser les NCx de type InP/ZnS plutôt que d’autres matériaux à base d’éléments toxiques (Cd, Pb, …).Le deuxième chapitre porte sur une synthèse à partir des carboxylates d’indium connue de la littérature. Le but est alors de caractériser la structure des NPs pour comprendre le déroulement de la synthèse et de l’enrobage. Des mesures par résonance magnétique nucléaire (RMN) en phase solide et spectroscopie photo-électronique par rayons X (XPS) révèlent l’oxydation des NPs d’InP. La couche d’oxyde qui se forme durant la synthèse des NPs d’InP s’épaissit lors de l’enrobage. Cette oxydation provient d’un couplage décarboxylant des acides carboxyliques à haute température en présence des NPs. Elle serait à l’origine de l’inhibition de croissance des objets, ce qui limiterait les gammes de longueurs d’onde atteignables.Le troisième chapitre concerne une nouvelle synthèse à partir d’amidinate d’indium au lieu des carboxylates d’indium. L’intérêt de cette approche est la possibilité d’abaisser considérablement la température de réaction (150°C au lieu de 280°C) et ainsi d’éviter la réaction secondaire de décarboxylation. Un enrobage à basse température (150°C) est aussi mis en place. La synthèse induit également une oxydation de la surface des NPs d’InP. Un nouveau couplage a lieu entre les ligands, l’acide palmitique et l’hexadécylamine, et donne de nouvelles conditions oxydantes. Le jeu sur les ratios des ligands montre qu’en bouleversant le milieu réactionnel, les NPs d’InP ne présentent pas de réponse en luminescence concluante. La synthèse et l’enrobage sont alors réalisés sous atmosphère de dihydrogène (H2) en réacteur Fisher-Porter dans le but de contrer ces conditions oxydantes. La synthèse et l’enrobage donnent des tailles de NPs de l’ordre de 3,4 nm (condition nécessaire pour s’approcher d’une émission dans l'infra-rouge) et un rendement quantique de 18-20 %, résultats encore jamais atteints lors de cette thèse.Le dernier chapitre est consacré à une étude exploratoire sur les NPs de Zn3P2. Le phosphure de zinc est un matériau prometteur du fait de l’abondance de ses constituants non toxiques et des longueurs d’onde potentiellement accessibles. Différents paramètres de synthèse sont étudiés et les propriétés structurales et optiques sont caractérisées. Des résultats préliminaires sur l’enrobage montrent des difficultés liées à la stabilité des NPs de Zn3P2. L’utilisation de l’oxyde de trioctylphosphine (TOPO) semble permettre la passivation de ces NPs à l’air et en travaillant sous H2 une meilleure stabilité est envisageable / Résumé de la thèse en anglais : This PhD investigation focuses on organometallic synthesis of indium phosphide (InP), zinc phosphide (Zn3P2) colloidal semiconductor nanoparticles (NPs) and core/shell structures which were obtained by the growth of a layer of zinc sulfide (ZnS) on the surface. The objectives are to understand and control the synthesis in order to shift the absorption and emission wavelengths to the near infra-red range, interesting for biomedical imaging.The first chapter presents the state of the art on the InP and InP/ZnS nanocrystals (NCx). A brief recall on the physical and chemical properties of semiconductor NCx is presented and various syntheses are described. Particular attention was paid to the size of NCx, the shift of the fluorescence emission to higher wavelengths and the optimization of quantum yields. The potential of these objects for white light emitting diodes (LED) or biomedical imaging shows the value added of using InP/ZnS NCx rather than other materials based on toxic elements such as cadmium, lead elements…The second chapter focuses on a synthesis from indium carboxylates known in the literature. The goal is to characterize the structure of NPs to understand the procedure of the synthesis and the coating. Measurements by Nuclear Magnetic Resonance (NMR) in solid state and Photoelectronic X-ray spectroscopy (XPS) revealed the oxidation of InP of the NPs. This oxide layer increases during the coating. This originates from a decarboxylating coupling of carboxylic acids at high temperature in the presence of NPs. This oxidation is believed to inhibit the growth of the object, which restricts the attainable range of wavelengths.The third chapter provides a novel synthesis from indium amidinate instead of indium carboxylate. The advantage of this approach is the potential to lower significantly the reaction temperature (150°C instead of 280°C) and to avoid secondary decarboxylation reaction. A coating with ZnS at low temperature (150°C) is also developed. The synthesis of InP NPs also causes an oxidation of the surface. A coupling takes place again between the ligands, palmitic acid and hexadecylamine providing new oxidizing conditions. The study of different ratios of ligands shows that when the reaction medium is modified, the InP NPs do not exhibit a conclusive luminescence response. Synthesis and coating are carried out under an atmosphere of hydrogen (H2) in Fisher-Porter reactor in order to counter these oxidizing conditions. NPs with diameters of the order of 3,4 nm (a necessary condition to approach the infra-red emission) and a quantum yield of 18-20% are thus obtained. These had never been observed before during this thesis.The last chapter is devoted to an exploratory study on Zn3P2 NPs. Zinc phosphide is a promising material because of non-toxic and abundant constituents, and potential access to near infra-red wavelengths. Different synthesis parameters are studied and the structural and optical properties are characterized. Preliminary results on the coating show instabilities of the Zn3P2 NPs. The use of trioctylphoshine oxide (TOPO) appears to allow the passivation of the NPs in the air and a better stability is possible under an atmosphere of H2
