Development of Structural Equations Models of Statewide Freight Flows

The modeling of freight travel demand has gained increasing attention in the recent past due to the importance of efficient and safe freight transportation to regional economic growth. Despite the attention paid to the modeling of freight travel demand, advances in modeling methods and the development of practical tools for forecasting freight flows have been limited. The development of freight demand models that incorporate the behavioral aspects of freight demand face significant hurdles, partially due to the data requirements, which are a consequence of the inherent complexity of the mechanisms driving freight demand. This research attempts to make a contribution in this context by proposing a relatively data simple, but behaviorally robust statewide modeling framework for the state of Florida, in the spirit of an aggregate level four-step planning process. The modeling framework that is developed in this research can be applied to the modeling of freight travel demand using data contained in readily available commercial databases such as the Reebie TRANSEARCH database and the InfoUSA employer database. The modeling methodology consists of a structural equations modeling framework that can accommodate multiple dependent variables simultaneously. This framework predicts freight flows on various modes between two zipcodes based on the socio-economic characteristics and the modal level of service characteristics. Separate models have been developed for various commodity groups. The estimated models for various commodity groups are found to offer statistically valid indications and plausible interpretations suggesting that these models may be suitable for application in freight transportation demand forecasting applications. The sensitivity analysis conducted on these models clearly added evidence to the fact that employment is the key factor influencing freight flows between two regions.

A Study on Information Transmission and Volume-price Relationship in Taiwan Stock Index and Industrial Stock Index

The purpose of this study is to research the volume-price relationship and information transmission among Taiwan Stock Index, Electronic Industry Index, Financial Industry Index and Plastic Industry Index. This study uses the time series methods of ADF unit root test, variance decomposition, Granger causality and impulse response analysis to proceed empirical research. It covers the period June 2, 2003, through December 29, 2006 and uses the daily data for sample. The empirical results can be summarized as follows¡G (1) All the trading volume and stock return series are trend stationary at level, therefore, they are integrated of order 0 ~ I (0). (2) The variance decomposition shows that the major change of every variable comes from by itself. The explanatory power of trading volume is higher than stock returns. Among the stock returns of Taiwan Stock Index, Electronic Industry Index, Financial Industry Index and Plastic Industry Index, Taiwan Stock Index has the highest explanatory power. (3) According to the Granger causality test, it expresses that trading volume leads stock returns. Taiwan Stock Index is the leading indicator of the Electronic Industry Index and Financial Industry Index. (4) As to the impulse response functions, neither persistent nor overall. The effect of shocks on all variables is transitory.

Representações de escola por alunos evadidos e reinscritos em turmas de Educação de Jovens e Adultos (EJA)

A presente pesquisa visa a analisar as representações de `escola materializadas nos depoimentos de alunas pertencentes a classes sociais menos favorecidas e que se evadiram ainda durante o ensino fundamental para, posteriormente, retomarem seus estudos via turmas de Educação de Jovens e Adultos (EJA). O aluno que inicia sua vida escolar, ao se relacionar com as diversidades (e adversidades) da língua e da educação, pode oferecer, como resposta, movimentos de auto exclusão, indicando, em situações agudas, renúncia perante o seu papel social como estudante, gerando, dessa forma, alienação do processo educativo e consequentemente do universo formal escolar. Em última instância, este estudo procurou compreender os mecanismos de auto exclusão e os fatores que contribuíram para a evasão escolar. O aporte teórico da pesquisa se fundamenta na Análise do Discurso de Linha Francesa, sobretudo nos estudos postulados por Pêcheux e Foucault na interface com a psicanálise. Foram realizadas entrevistas orais, informais, semiestruturadas, para que se pudessem investigar, a partir das regularidades enunciativas, marcas ideológicas que revelassem os modos de subjetivação e efeitos de sentido, indicando as posições do sujeito desejante em relação ao Outro. Levantamos como Hipótese que o aluno egresso, ao retornar à escola, o faz conforme motivação proveniente do desejo do Outro (seja do Estado, ou do contexto econômico-social), afetado pelas metanarrativas que concebem a educação formal como único meio de se atribuir poder ao saber do aluno em questão. Entretanto, esse discurso sócio historicamente compartilhado não produz os mesmos efeitos de verdade para o pesquisado, uma vez que este não consegue significar/singularizar a educação formal, resultando em novas situações de auto exclusão de sua vida escolar. Em última instância, desconstruímos esses mecanismos de auto exclusão, de modo que os depoentes conseguissem singularizar e significar seu processo de aprendizagem na escola. / This research aims to analyze the representations of `school materialized in the testimonies of students belonging to lower social classes and that evaded even during elementary school to subsequently resume their studies via classes from EJA (School for Youngs and Adults). The student who starts his school life, when dealing to the differences (and adversities) of language and education, can offer as an answer, movements of self exclusion, indicating, in acute situations, resignation before its social role as a student generating thus alienation regarding the educational process and consequently the academic formal universe. Ultimately, this study sought to understand the mechanisms of self exclusion and the factors contributing to school dropout. The theoretical research is based on French Analyses of Speech, especially in studies postulated by Pecheux and Foucault, interfacing with psychoanalysis. Oral, informal, semi-structured interviews were conducted, so that it could be investigated, from the enunciative regularities, ideologic marks that reveal modes of subjectivity and meaning effects, indicating the positions of the desiring subject in relation to the Other. One raised the hypothesis that the evaded student, returning to school, does it according to the motivation from the desire of the Other (whether of the State, or the economic and social context, or someone elses desire), affected by metanarratives that conceive formal education as the only means to empower upon learning of the student in question. However, this social discourse historically shared does not produce the same signification effects according to the interviewee, since he can not singularize formal education, resulting in new situations of self exclusion of his academic life. Ultimately, one wants to deconstruct these mechanisms of self-exclusion, so that the interviewed ones can singularise their learning process in school.

Part of the work also includes the evaluation of the simulator FlitePro for the purposes of its certification as a training device.

Identificação e caracterização de proteínas que se ligam a actina (ABPs) no apicomplexa Neospora caninum / Identification and characterization of actin binding proteíns (ABPs) from the apicomplexan Neospora caninum

Neospora caninum é um parasita intracelular obrigatório pertencente ao filo Apicomplexa, conhecido por ser uma das principais causas de aborto parasitário em bovinos e por apresentar transmissão transplacentária. Para locomoverem-se e acessarem o conteúdo intracelular de células hospedeiras, organismos apicomplexas fazem uso de um mecanismo não convencional que se utiliza de uma maquinaria celular cujo papel central é exercido pelo motor actina-miosina, auxiliado por proteínas intermediárias e de acoragem, que realiza a propulsão do parasita na direção do movimento. Para o funcionamento dessa maquinaria, é essencial que actina esteja em sua forma filamentosa (actina-F). Porém, actinas de apicomplexas são conhecidas por serem funcional e estruturalmente não convencionais, formando filamentos pequenos e instáveis in vitro, assim como pelo predomínio de grande maioria de actina monomérica (actin-G) nas células in vivo. Desse modo, para formar e manter actina-F a dinâmica de actina desses organismos requer uma regulação precisa, que, em apicomplexas, é conduzida por um arsenal conhecidamente pequeno de proteínas que se ligam a actina (ABPs). Nosso objetivo neste estudo foi identificar e caracterizar ABPs de N. caninum. Para isso, duas ABPs de N. caninum foram estudadas: fator de despolimerização de actina (NcADF) e proteína associada a ciclase (NcCAP); também, foi gerado e caracterizado soro contra região de actina de N. caninum entre aminoácidos 201 e 310 (anti-NcAct201-310). NcADF (correspondente ao acesso NCLIV_012510 em ToxoDB) foi submetida a caracterização molecular e bioquímica. A sua estrutura terciária foi gerada por modelagem molecular baseada em homologia, apresentando folding conservado, porém com F-loop de menor tamanho, quando comparada a ADF/cofilinas canônicas. A forma recombinante de NcADF foi expressa E. coli BL21 por plasmídeos pET32a(+) e pET28a(+) e solubilizada em tampão desnaturante e nativo, respectivamente. NcADF_pET32 foi purificada e utilizada para geração de soro anti-NcADF, que detectou ambas NcADF recombinantes, assim como proteínas endógenas em western blot 1-D e 2-D com peso molecular e pI próximos aos preditos. O soro anti-NcADF também localizou NcADF difusa no citoplasma, com menos intensidade nos polos de taquizoítas de N. caninum extracelulares. NcADF_pET28 foi purificado na forma nativa e utilizado para caracterização funcional para avaliação de seu papel na dinâmica de actina liofilizada de coelho. Ensaios de cossedimentação, cinética de polimerização e despolimerização, viscosimentria de baixo cisalhamento (queda de bola), estado estacionário e ligação entre actina-G e NcADF, em conjunto, mostraram que NcADF causa despolimerização de actina-F, realiza sequestro de monômeros de actina e quebra de filamentos. NcCAP foi submetida a caracterização molecular e foi identificada como produto de expressão do gene de acesso NCLIV_054140. NcCAP recombinante foi expressa em pET32a(+) e pET28a(+) predominantemente em corpos de inclusão e foi solubilizada em tampão desnaturante. A forma purificada de pET32_NcCAP, identificada por espectrometria de massas, foi utilizada para imunização e o soro resultante detectou NcCAP recombinante e endógena por western blot 1-D e 2-D, apresentando bandas e spots de peso molecular e pI próximos ao esperado. O soro anti-NcCAP também localizou NcCAP em taquizoítas ii extracelulares de N. caninum difusa no citoplasma e/ou com predomínio na região periplasmática da célula. Por fim, o soro anti-NcAct201-310 foi gerado, sendo capaz de detectar proteínas em sua forma nativa e realizar marcação na região periférica e, possivelmente, nuclear de taquizoítas de N. caninum extracelulares. A caracterização de ABPs de N. caninum feita neste trabalho amplia o conhecimento sobre a conservação dessas proteínas ao longo do filo Apicomplexa. Ademais, representa uma contribuição para o entendimento da dinâmica de actina e, por consequência, futuramente, pode colaborar para a elucidação de mecanismos-chave para a sobrevivência e disseminação dos parasitas pelo seu hospedeiro / Neospora caninum is an obligate intracellular parasite that belongs to the phylum Apicomplexa. It is known as one of the main causes of infectious abortion in cows and for its efficient transplacentary transmission. Apicomplexan organisms use a phylum-specific mechanism of invasion and gliding motility, which use an unusual cellular machinery based on an actin myosin motor assisted by intermediary and anchoring proteins that creates the traction force to impulse the parasite forward. Filamentous actin (F-actin) is essential to the appropriate functioning of this machinery, although apicomplexan unconventional actin forms small and unstable filaments in vitro and is found preponderantly as monomer (G-actin) in cells. Thus, the parasites need actin-binding proteins (ABPs) to strictly regulate actin dynamics and to form and maintain F-actin when it is necessary to the cell. Here, we aimed at identifying and characterising ABPs from N. caninum. Two ABPs were characterised: actin-depolymerising factor (NcADF) and cyclase-associated protein (NcCAP) from N. caninum. In addition, a serum against the actin region between amino acids 201 and 310 (anti-NcAct201-310) was raised. NcADF, which corresponds to identification NCLIV_012510 on ToxoDB, was molecular and biochemically characterised. Firstly, the tertiary structure of NcADF was generated by molecular modelling based on homology. Comparing to canonical ADF/cofilins, NcADF presented a conserved folding, albeit its smaller F-loop. The recombinant form of NcADF was expressed in E. coli BL21 using pET32a(+) and pET28a(+) plasmids and solubilized in denaturing and native buffers, respectively. Polyclonal antibodies were raised in mice against purified NcADF_pET32, which was able to detect both forms of recombinant NcADF as well as proteins in 1-D and 2-D western blot with expected molecular weight and isoelectric point (pI). Additionally, NcADF was localised in extracellular N. caninum tachyzoites as a diffuse pattern on cytoplasm with less intensity in both poles. NcADF_pET28 was successfully purified in native form and used for functional characterisation to evaluate the role of recombinant NcADF on lyophilised rabbit actin dynamics. Together, co-sedimentation, polymerisation and depolymerisation kinetic, low shearing viscometry (falling ball), steady state, and G-actin and NcADF binding assays showed that NcADF was able to depolymerise actin-F, sequester actin monomers, and sever filaments. Moreover, NcCAP (identification NCLIV_054140) was also characterised. Recombinant NcCAP was expressed in pET32a(+) and pET28a(+) plasmids predominantly in inclusion bodies and was solubilised in denaturing buffer. NcCAP_pET32 was purified and identified by mass spectrometry. Then, the polyclonal antibodies against this recombinant protein was generated in mice. It was able to detect recombinant and endogenous NcCAP, presenting bands and spots in 1-D and 2-D western blot with molecular weight and pI quite near to the predicted ones. NcCAP was localised as a diffuse pattern on cytoplasm and/or predominantly on periplasmic regions of extracellular taclyzoites of N. caninum. Finally, the serum containing anti-NcAct201-310 polyclonal antibodies was raised in mice. It detected endogenous proteins mainly in native form and localised them on periplasmic and possibly nuclear region in extracellular N. caninum tachyzoites. The characterisation of N. caninum ABPs iv extends our understanding of these proteins conservation and their function throughout the Apicomplexa phylum. Furthrmore, it represents a contribution to the field towards the comprehention of actin dynamics and in the future might provide information for important mechanisms of dissemination and survival of the parasite at its host

Bakgrund och Problem: Aktiemarknaden påverkas både av inhemska och utländska faktorer. Därför är det av intresse att se vilka makroekonomiska variabler som påverkar den svenska aktiemarknaden. Anledningen till att Sverige har valts som den geografiska punkten är att det är av intresse att se hur ett litet land som Sverige, som har en öppen ekonomi påverkas av de utvalda makroekonomiska variablerna. Syfte: Syftet med uppsatsen är att med hjälp av information samt analys, studera hur de olika makroekonomiska variablerna påverkar den inhemska aktiemarknaden. Olika faktorer som påverkar aktiemarknaden kommer att lyftas fram för att i sin tur även se till de olika branscherna. Metod: Då data enbart består av hämtning av tidigare information fokuseras det enbart på sekundärdata i form av historiska siffror samt historiska undersökningar. De statistiska tester som tillämpas är Granger Causality test, Johansens Cointegration test, Impulse Response Function test, ADF test, KPSS test, Mulitpel regression. Slutsats: Med de resultat som presenterades i denna studie, skulle vi nog inte kunna säga att vi har ett svar över vilka aktier en investerare ska införskaffa. Dock skulle vi kunna poängtera att den potentiella investeraren bör ha dessa variabler i beaktning vid beslut. Genom att studera dessa variabler kan man få en känsla om vilket håll variablerna kommer att röra sig och på så sätt säga att de kan påverka aktieindexen. Att bara kolla på de makroekonomiska variabler som denna studie belyser räcker inte för att förstå hur aktieindex kommer att se ut i framtiden, men det är en bit på vägen till att förstå aktiemarknadens rörelse.

Bitcoin - Monero analysis: Pearson and Spearman correlation coefficients of cryptocurrencies

In this thesis, an analysis of Bitcoin, Monero price and volatility is conducted with respect to S&P500 and the VIX index. Moreover using Python, we computed correlation coefficients of nine cryptocurrencies with two different approaches: Pearson and Spearman from July 2016 -July 2018. Moreover the Pearson correlation coefficient was computed for each year from July2016 - July 2017 - July 2018. It has been concluded that in 2016 the correlation between the selected cryptocurrencies was very weak - almost none, but in 2017 the correlation increased and became moderate positive. In 2018, almost all of the cryptocurrencies were highly correlated. For example, from January until July of 2018, the Bitcoin - Monero correlation was 0.86 and Bitcoin - Ethereum was 0.82.

Evaluation of development platforms : building financial online applications

CRM Treasury Systems AB is one of the Nordic Countries’ leading companies within treasury management. Their main product CRM Finance is a software program for financial administrators with roots back to the early 90s. Computer usage and software has changed considerably since then and the company is currently considering choosing a new technology to base future products upon. In this thesis Oracle ADF, Microsoft Lightswitch beta 1 and Silverlight with WCF RIA Services, are compared in order to select a suitable technology to base future applications on. The frameworks are used to implement a simple application with a small number of screens displaying database data in order to get to know the frameworks and capture the time required to get started. After the basic applications have been implemented in all the three frameworks and evaluated it is decided to continue by implementing a number of more advanced authorization and internationalization scenarios. It is decided to continue with only two of the selected frameworks due to lack of time once the simple application has been implemented successfully. Lightswitch beta 1 is chosen for discontinuation due to an uncertain future for the application as it is the first public available beta of the product. Based on the experience from the implementations performed, the time required for them as well as how the platforms fit the company’s requirements the .NET platform with Silverlight + WCF RIA Services is recommended as the base for future applications.
