Lucie Cahlíková, Ph.D. Title of diploma thesis: Derivatives of Amaryllidaceae Alkaloids as Drugs Plants of the family Amaryllidaceae belong to the widespread species. They contain a large amount of Amaryllidaceae alkaloids (AA) which are known for their biological activity. AA possess a broad spectrum of biological activities including an antiviral, antimalarial, antitumor, cholinesterase's inhibitory activity and others. An interesting AA is ambelline which occurs mainly in plants of the genus Crinum and Nerine. So far the biological activity of this compound has been studied only marginally. In the studies conducted, this substance appears to be less interesting. A series of aliphatic and aromatic derivatives of ambelline has been prepared in the framework of this thesis. Subsequently, their cholinesterase inhibitory activity and GSK-3β inhibitory activity were studied. Cytotoxic activity on a panel of selected tumor and resting cell lines was also screened. The thesis describes practical work as one of the teaching methods with a focus on its goals, effectiveness and different types of assignment. This thesis analyses lifestyles of female students from the whole University of South Bohemia, focusing on physical activities, their regularity, opinion of negative addictive drugs, relationship towards healthy diet and drinking water habits, and interest in relaxation. The theoretical part of the thesis deals with the definition of the term 'physical activity' and its classification. In the practical part, the following areas are observed: results regarding the quality of physical activities, regularity of physical activities, evaluation of the physical activities of female students followed by the evaluation according to the stated hypotheses. Based on the data obtained, it is possible to conclude how big the interest in regular physical activities is nowadays, whether the students have enough exercise during the week, why they do physical activities and what physical activities are the most popular.

The thesis is focusing on the influence of organized physical activities on the smoking and alcohol drinking by the kids between 12 and 16 years of age. The basic characteristics of organized physical activities, smoking and alcohol drinking are described in the theoretical part of the thesis where emphasis is put on children and youth.The sociological, psychological and biological views are included in the basic characteristics. The research in the practical part of the thesis was done by a questionnaire. The total number of interviewee was 216. The amount of physical activities, smoking and alcohol drinking by kids between 12 and 16 years of age was charted, once the data collected was statistically evaluated and put into connection with membership in a sports club.The research clearly shows that children registered in a sports club are less likely to smoke or consume alcohol.

The dissertation deals with the iss. of physical activity in connection with primary prevention. It concerns particularly primary prevention of non-infectious diseases with massive incidence. It is important to know the principles that can contribute to the efficiency of nursing intervention in the area of physical activity. The model of stages of readiness for change plays a significant role here. The goal of the dissertation consisted in mapping the quality and quantity of exercise in ealthy population and its differences related to gender, age, education, and its role as a tool influencing the quality of health condition. Based on that goal, the following hypotheses were stated: Men and women differ in physical activity quality. Men and women differ in physical activity quantity. The quantity of physical aktivity decreases with increasing age in healthy population. The quality level of physical activity changes with the degree of education achieved. The quantity level of physical activity changes with the degree of education achieved. Further, the following research question was asked: How does physical activity contribute to bio-psycho-social aspects of life? The research within the dissertation was designed as quantitative-qualitative research. The selection set of the quantitative part consisted of 1122 respondents who constituted a representative set of adult population for the Czech Republic, according to the preset socio-demographic indicators. The data collection was performed in form of questionnaire research through the standardized short version of IPAQ questionnaire. The degree of dependence of selected categorial features was determined by chi-square test. Ordinal and cardinal data, after verification of normality by Shapiro-Wilk test, were processed with the help of non-arametric Mann-Whitney test, Wilcoxon paired test and Spearman non-parametric correlation. The qualitative research took place in form of a six-month experiment in which the research group (20 probands) was subject to intervention of physical activity, while the control group (34 probands) lived common life. Before and after intervention, subjectively perceived quality of life was measured in both groups with the help of selected questions of the standardized WHOQOL 100 questionnaire, and body composition was measured with the help of biolectrical impedance with Bodystat 1500 device. The research found that both men and women spend most of the day sitting. That fact is very probably related also to the Body Mass Index, showing an average value of 25.6 kg/m2 in the 1122 respondents, which is a value ranking in the category of overweight. It was proved that men are more active than women, preferring more intensive physical activity, which complies fully with statements of other authors. But an interesting fact is that the intensity and frequency of physical activity are influenced by education. Particularly respondents with university education are more active. On the contrary, age is related to decreasing intensity and frequency of physical activity, which is stated by other authors as well. The dissertation had four goals set and all of them were met. The five hypotheses were verified too. It can be stated that men and women differ in the quality and quantity of physical activity. It can also be stated that the level of quality and quantity of physical activity decreases with increasing age. And finally, it was shown that the quality and quantity of physical activity change also with the level of the education achieved. Regular physical activity contributes to improve the subjectively perceived psycho-social aspects of quality of life. They influence the body composition of the probands too. Nursing, considered an irreplaceable and self-standing scientific discipline, can contribute essentially to the development of active lifestyle not only of ill but also of healthy individuals.

Studie možností rozvoje vybraných obcí mikroregionu Slezská Harta

The aim of the diploma thesis is to find out landscape vision of the population -- citizens and mayors of selected municipalities of microregion Slezská Harta. Four villages, which are in the process urban planning, were selected. The municipalities were analysed by socioeconomics analysis and SWOT analysis. Also questionnaires were made -- both for citizens and mayors of the villages. Finally the measures and program activities are proposed -- they will help with local development of the municipalities. Survey shows, that 2/3 respondents wish space for their peaceful life, 1/2 wish space for tourism and 1/3 wish space for environment protection. Urban plans disagree with landscape vision of citizens. Therefore are proposed changes of the problematic points and other specific measures which lead to the development of communities.

The diploma thesis is composed on the topic Management of corporate social responsibility in an international environment and has as main objective to propose an effective way of managing social responsibility at IBM on five continents. The partial objective is to determine whether the concept of CSR is driven by retrograde, progressive or bi-directionally. The basis for evaluating and designing effective way to control the social responsibility is qualitative research within five continents with managers in selected branches. The diploma thesis concludes that IBM has clearly defined CSR activities, all activities are controlled by retrograde planning (Top-Down) and this control method works effectively across cities, countries and continents. In the recommendation were as suitable way of communicating corporate values and employee involvement in their own creation selected team building activities in the form of corporate volunteering.

This diploma thesis deals with the use of the TPR method (Total Physical Response) where the principle is a student who does not speak, only reacts nonverbally to verbal instructions given by a teacher e.g. by moving, drawing, colouring. This method is based on a similarity with learning the mother tongue. The work is divided into a theoretical part consisting of the theoretical background of the given method and the comparison with the process of learning the mother tongue, and a practical part which contains a research survey about the knowledge of using the TPR method at a particular primary school, it also contains general typology of TPR activities and specific examples and an analysis to each type of an activity.

In the theoretical part are defined basic terms of marketing, marketing services, quality of services and marketing research. The practical part is started by a short description of the company, its history and current activities. Analysis and exploration are performed in Prague`s sales network of ŠKODA AUTO by using methods of Mystery Activity. At the conclusion of the evaluation of research are identified weaknesses of ŠKODA AUTO and proposed measures to eliminate these deficiencies and thereby increase the quality of provided services.

The thesis describes business project based on franchising. For this type of business had been choosen the company The Little Gym which is deals with providing services mainly exercise classes in specialized gyms for children aged from 3 months to 12 years. The aim of this thesis is create a business and economic evalution of the project in the field of fitness with the use of franchising. The thesis is devided into theoretical and practical parts. In practical part the authoress used data from the Czech statistics authority and created a comparison of development birth of childrens, income of families, proportion of minor against the population and others namely between regional cities as Hradec Králové where a new branch will come and Ostrava where the branch was recently opened. Marketing research of potential customers was focused on their opinions and attituteds to physical activitites. Further was done a market segmentation and analysis of competition, especially Porter model. The financial plan was developed based on calculation from the available data. In conclusion the bussines plan was evaluated including evaluation of the economic aspects of the project.
