Le rôle des microARNs dans la mise ne place de la maladie chez les plantesEl Mnouchi, Salma January 2015 (has links)
La réussite de la mise en place d’une résistance efficace chez la plante hôte requiert la présence d’un dialogue entre les différentes voies signalétiques menant à l’induction de la défense. L’implication de l’extinction génique post-transcriptionnelle dans la résistance contre des agents pathogènes, autres que les virus, est une nouvelle avenue qui va permettre de disséquer le lien qui peut exister entre les deux voies immunitaires de la plante : celle qui est déjà importante pour lutter contre les bactéries et les champignons (voie induite à la suite de la reconnaissance des éliciteurs) et celle de l’extinction génique post-transcriptionnelle. Les résultats que nous avons obtenus ont une signification très importante concernant l’implication de l’extinction génique post-transcriptionnelle dans la résistance des plantes contre les agents pathogènes d’une manière générale et Botrytis cinerea, Alternaria brassicicola et Pseudomonas syringae en particulier. Mon travail de recherche a permis d’identifier le miR472 comme étant un acteur qui favorise la mise en place de la maladie causée par des agents pathogènes du type nécrotrophe et hémibiotrophe. De plus j’ai pu, en utilisant le modèle P. syringae pv tomato DC3000-Arabidopsis, disséquer dans le détail, comment la bactérie manipule la machinerie de la plante pour activer la voie des microARNs menant à l’accumulation du miR472, responsable de la suppression de la résistance basale et du PTI. Cette suppression passe par l’inhibition de sa cible, la protéine NB-LRR; RDS1 un nouveau régulateur impliqué dans le PTI induit par la Flg22.
Cette étude permettra une avancée remarquable dans les connaissances reliées à l’immunité des plantes, ce qui constituerait une étape clé dans la découverte de nouvelles stratégies de lutte contre les agents phytopathogènes. Sur le plan fondamental, cette étude a permis de consolider le dialogue qui existe entre les voies immunes de la plante et de mettre en évidence un nouvel acteur impliqué dans la résistance basale. En matière d’application, comprendre les mécanismes de défense et de résistance des plantes aux agents pathogènes est primordial pour développer des plantes génétiquement résistantes et des stimulateurs de défense naturelle (Vaccin de plantes). Ceci constitue un enjeu majeur dans l'objectif de concevoir de nouveaux moyens de lutte phytosanitaire, à la fois plus respectueux de l’environnement et ayant moins d’impact sur la santé humaine.

The Alternaria genomes database: a comprehensive resource for a fungal genus comprised of saprophytes, plant pathogens, and allergenic speciesDang, Ha X., Pryor, Barry, Peever, Tobin, Lawrence, Christopher B. January 2015 (has links)
BACKGROUND: Alternaria is considered one of the most common saprophytic fungal genera on the planet. It is comprised of many species that exhibit a necrotrophic phytopathogenic lifestyle. Several species are clinically associated with allergic respiratory disorders although rarely found to cause invasive infections in humans. Finally, Alternaria spp. are among the most well known producers of diverse fungal secondary metabolites, especially toxins. DESCRIPTION: We have recently sequenced and annotated the genomes of 25 Alternaria spp. including but not limited to many necrotrophic plant pathogens such as A. brassicicola (a pathogen of Brassicaceous crops like cabbage and canola) and A. solani (a major pathogen of Solanaceous plants like potato and tomato), and several saprophytes that cause allergy in human such as A. alternata isolates. These genomes were annotated and compared. Multiple genetic differences were found in the context of plant and human pathogenicity, notably the pro-inflammatory potential of A. alternata. The Alternaria genomes database was built to provide a public platform to access the whole genome sequences, genome annotations, and comparative genomics data of these species. Genome annotation and comparison were performed using a pipeline that integrated multiple computational and comparative genomics tools. Alternaria genome sequences together with their annotation and comparison data were ported to Ensembl database schemas using a self-developed tool (EnsImport). Collectively, data are currently hosted using a customized installation of the Ensembl genome browser platform. CONCLUSION: Recent efforts in fungal genome sequencing have facilitated the studies of the molecular basis of fungal pathogenicity as a whole system. The Alternaria genomes database provides a comprehensive resource of genomics and comparative data of an important saprophytic and plant/human pathogenic fungal genus. The database will be updated regularly with new genomes when they become available. The Alternaria genomes database is freely available for non-profit use at http://alternaria.vbi.vt.edu.

Incidence and epidemiology of apple core rot in the Western Cape of South AfricaBasson, Elaine 12 1900 (has links)
Thesis (MScAgric)--Stellenbosch University, 2012. / ENGLISH ABSTRACT: This study looked at the incidence, etiology and epidemiology of core rot of apples in orchards situated in the Western Cape, South Africa. Core rot is a post-harvest disease, with three symptoms, namely mouldy core (MC), dry core rot (DCR) and wet core rot (WCR). These symptoms are caused by various pathogenic fungi, including Alternaria and Penicillium. Although MC is not economically important, DCR and WCR are, as they affect the flesh of the fruit. Core rot occurs worldwide in susceptible apple cultivars such as ‘Starking’ and ‘Red Delicious’. These cultivars have a wider, open calyx tube which results in an open core area. In South Africa, core rot of apples are important post-harvest diseases and losses of between 5 and 12% occur in apple cultivars.
An in depth literature search was done on core rot including literature on each core rot symptom, the genuses Alternaria and Penicillium, molecular identification and techniques, disease incidence and its economic importance, various inoculum sources, pathogenicity of core rot organisms and integrated management of core rot. This study included two research chapters, with seven objectives, namely, to 1, determine the incidence of core rot in apples from commercial orchards both pre- and post-harvest; 2, to identify the causal organisms associated with core rot symptoms; 3, to identify potential sources of inoculum of core rot pathogens and determine whether there is synergism between Alternaria and Tarsonemus mites associated with core rot; and 4, to determine whether the fungicide Bellis®, used a full bloom application, can be used to manage core rot in South Africa; 5, to identify the species of Alternaria and Penicillium sampled from core rot symptomatic fruit and inoculum sources (air, apple mummies and mites), using morphological and molecular methods; 6, to compare Penicillium species isolated from pre- and post-harvest WCR symptomatic fruit, using molecular species identification methods and 7, to compare and to select the most reliable pathogenicity test for use in future research.
The total decay incidence for Ceres is considerably higher than the previous losses indicated in literature. Pre-harvest core rot, which was confirmed by previous studies, had a higher incidence of each core rot symptom than previously indicated. The two most frequently isolated causal organisms were Alternaria and Penicillium. Other organisms isolated and then identified from the symptoms were Fusarium, Cladosporium, Epicoccum, Ulocladium, Stemphylium, Phoma, Botryosphaeria, Botrytis, Trichoderma, Verticillium, Paecilomyces and Gliocladium.
Three inoculum sources, air, mummies and mites, were regarded as potential sources of infection for core rot. During this study the sources of infection were verified and core rot causing organisms were isolated from these sources. Alternaria was isolated from air inoculum samples, but was not found on the other two sources. This dismissed the hypothesis that there was a possible synergism between Alternaria and Tarsonemus mites. Penicillium species were isolated from all three sources, more frequently from the mummies and mites. Bellis® was applied three times during the bloom period. The subsequent results showed a significant difference between the control and Bellis® treated treatments with the treated fruit having a significant higher incidence than the controlled fruit. No control was observed with this result and managing core rot with only Bellis® is not advisable.
Alternaria species were identified using the following genetic loci, ITS, OPA1-3, 2-1 and 10-2 as well as endoPG. Isolates from pre- and post-harvest symptoms and air inoculum were identified using each of the genetic loci. Alternaria arborescens was one of the species that was identified. The other isolates obtained were A. alternata, A. tenuissima, A. gaisen, A. dumosa, A. turkisafria and A. perangusta. Separating combined species was not possible. Another molecular technique, ISSR, was used to identify Alternaria species. This technique, after multiple re-runs, did not give consistent results and species could not be identified. Penicillium species were identified using the genetic loci ITS for isolates collected from pre- and post-harvest symptoms and inoculum sources. Thirteen clades were identified, including the species P. ramulosum, P. sp. (aff. cecidicola), P. sp (aff. dendriticum), P. expansum, P. paneum, P. solitum, P. crustosum , P. brevicompactum, P. novae-zeelandiae, P. glabrum and P. rugulosum. Penicillium expansum and P. ramulosum had the highest distribution between the isolates. Pre- and post-harvest WCR isolates were identified using the partial beta-tubulin PCR-RFLP method, and comparing different banding-patterns. The species identified using this method were P. expansum, P. ramulosum, P. sp. (aff. cecidicola), P. sp (aff. dendriticum), P. rugulosum, P. chermesinum and P. glabrum. Penicillium ramulosum and P. expansum had the highest incidence with P. ramulosum occurring more frequently pre-harvest than post-harvest and P. expansum occurring more frequently post-harvest.
Five methods, previously published, were compared to select the most reliable pathogenicity test. The methods included surface wounding of an apple with colonised toothpicks, surface wound inoculated with a pipette, inoculation of an open mesoderm core cavity, deep and non-wounding of apple fruit with colonised toothpicks. The surface wounding with a colonised toothpick gave the most reliable results and can be used in industry as a pathogenicity test for Alternaria in apples.
This study contributed to our understanding on the incidence and etiology of core rot in the Western Cape as well as in identifying inoculum sources from where infection can take place in the orchard. The results for the fungicide trial were not as anticipated and more research is required on selecting fungicides for the control of core rot in South African orchards. Although molecular techniques reduce the time in identifying fungal species, it is costly and mistakes can occur due to contamination. Identification of species can be incorrect when using a Genbank as the sequence information may be incorrect. Molecular techniques, though a good tool in identifying species, should be combined with morphological characteristics to ensure more accurate results. / AFRIKAANSE OPSOMMING: Hierdie studie het gekyk na die insidensie, etiologie en epidemiologie van kernvrot in appels vanuit boorde in die Wes-Kaap, Suid-Afrika. Kernvrot is ‘n na-oes iekte, met drie simptome, naamlik beskimmelde kern, droë kernvrot en nat kernvrot. Hierdie simptome word veroorsaak deur verskillende patogeniese swamme, insluitend Alternaria en Penicillium. Alhoewel beskimmelde kern nie ekonomies belangrik is nie, is droë en nat kernvrot wel belangrik, omdat hulle die vrug se vlees affekteer. Kernvrot kom wêreldwyd voor in vatbare kultivars soos ‘Starking’ en ‘Red Delicious’. Hierdie kultivars het ‘n wye, oop kelkbuis wat ‘n oop kern area veroorsaak. Vyf metodes, wat voorheen gepubliseer is, is vergelyk om die betroubaarste patogenisiteits toets te selekteer. Die resultate vir die fungisied proef was nie wat ons verwag het nie en meer navorsing word benodig om fungisiede te selekteer vir die beheer van kernvrot in Suid-Afrikaanse boorde. Alhoewel molekulêre tegnieke die tyd verminder om ‘n swam spesie te identifiseer, is dit wel duur en foute kan voorkom as gevolg van kontaminasie. Identifikasie van spesies kan verkeerd wees indien Genbank gebruik is, omdat die informasie daar nie altyd korrek is nie. Systematic Revision within the Pleosporaceae and Identification of Processes that Occlude Phylogenetic ReconstructionLawrence, Daniel January 2012 (has links)
The hypothesis of horizontal gene transfer of a hybrid gene from bacteria to the filamentous Ascomycota was tested using constrained phylogenetic analyses and tests of topological congruence. Results suggest that the hybrid gene was acquired from bacteria by a single transfer before the radiation of the Leotiomyceta. The phylogenetic relationship among Nimbya and Embellisia was investigated using both morphological and molecular data. Examination of conidia morphology revealed that Nimbya and Embellisia comprise two and four distinct morphological groups, respectively. Molecular analyses support all morphological groups of Nimbya and Embellisia and reveal that both genera are polyphyletic. Results suggest the circumscription of these genera is based upon convergent morphological characters. To further understand the evolutionary relationship among Alternaria and closely related genera, ten protein-coding genes were sequenced across 176 species. Three genes possessed significant substitution saturation and two other genes did not possess sufficient phylogenetic signal to assess relationships among the asexual Alternaria. The remaining five loci revealed strong support for asexual Alternaria and the order of divergence among eight asexual Alternaria species-groups. The current polyphyly of Alternaria was resolved using morphological and molecular data. Morphological examination revealed that most members of the infectoria clade (sexual Alternaria) produce diagnostic colony characters on dichloran rose bengal yeast extract sucrose agar (DRYES) and weak potato dextrose agar (WPDA) that are fundamentally different as compared to other small-spored Alternaria species. These data also revealed that all members of the infectoria clade produce arachnoid vegetative hyphae with multiple primary conidiophores, whereas other small-spored Alternaria species do not. Phylogenetic analyses revealed that the sexual infectoria clade clusters with other sexual genera phylogenetically distant to the asexual Alternaria. Lastly, the validity of taxonomy and the phylogenetic relationship among three small-spored Alternaria species was examined. Total ortholog comparisons and whole-genome comparisons revealed that the DNA sequence of A. alternata ATCC 11680 has a higher percent similarity to A. tenuissima EGS 34-015 than to A. alternata EGS 34-016. This suggests that these two isolates share a more recent common ancestor and A. alternata EGS 34-016 is more distantly related.

Assessing the genetic diversity of Alternaria Bataticola in South Africa using molecular markersChalwe, Joseph Musonda January 2015 (has links)

(Biotechnology, Department of Biotechnology, Faculty of applied and computer sciences) Vaal University of Technolog / Sweetpotato (Ipomoea batatas) is an important food crop that is grown in many countries. A number of viral and fungal sweetpotato diseases have been reported worldwide. One of the major and most economic diseases of the sweetpotato is Alternaria blight which is caused by the fungal pathogen Alternaria bataticola. This disease can be managed in a short term using fungicides and cultural practices. However, a long term and inexpensive approach is through the development of resistant cultivars. A prerequisite to this approach is the knowledge of the genetic diversity of this fungal pathogen. This study assessed the genetic diversity of 25 South African isolates of A. bataticola from naturally infected leaves and stems collected from different sweetpotato growing regions in South Africa by (i) characterising the isolates based on their morphology (ii) pathogenicity tests (iii) random amplified polymorphic DNA (RAPD) (iv) variation of the ITS2 sequences and (v) prediction of the ITS2 secondary structures. The isolates revealed some variation in colony colour pigments after culturing but Koch’s postulates were confirmed by their pathogenicity tests. The analysis of RAPD and variation of the ITS2 sequences showed high levels of variation (100%) among the isolates. Dendrograms generated from these analyses had many subclusters and did not cluster the isolates according to their geographic origins. The ITS2 secondary structures were predicted and can be used to identify and distinguish the isolates. This information in addition to the genetic diversity of the A. bataticola isolates will aid plant breeders in the development of resistant sweetpotato cultivars and early management of blight disease in South Africa.

Prospecção de compostos bioativos no produto de fermentação de fungos endofíticos isolados de Handroanthus impetiginosusAGOSTINI, Sandra Barbosa Neder 26 February 2016 (has links)
Os fungos endofíticos têm sido cada vez mais pesquisados como fonte de moléculas bioativas. Neste trabalho, foram isolados fungos endofíticos de uma árvore produtora de compostos medicinais, Handroanthus impetiginosus, com o objetivo de verificar se produziam metabólitos com atividade biológica e de identificar as espécies fornecedoras de princípios bioativos. Todas as 148 cepas isoladas foram triadas segundo a atividade antimicrobiana contra Staphylococcus aureus ATCC 6538, Candida albicans ATCC 10231 e Escherichia coli ATCC 25922 pelo método do ágar em bloco. Dentre essas, cinco fungos foram submetidos a fermentação em pequena escala, em caldo Czapek, durante 15 dias, a 28°C, tanto sob agitação quanto em modo estático. Ao final desse período, as culturas foram filtradas e submetidas a extração líquido-líquido. Os extratos obtidos foram avaliados quanto à capacidade de inibir o crescimento dos micro-organismos patogênicos citados anteriormente, por teste de difusão em ágar e determinação da concentração inibitória mínima (CIM). As menores faixas de CIM observadas foram 125-250 μg.mL-¹, de Alternaria alternata, e 250-500 μg.mL-¹, de Lasiodiplodia sp. Também avaliou-se o efeito sobre a viabilidade de linhagens celulares derivadas de câncer humano de mama (MCF-7), fígado (HepG2) e pulmão (A549). Os extratos de A. alternata e de Lasiodiplodia sp. reduziram a menos que 10% a viabilidade das células tumorais MCF-7 e HepG2. O primeiro também reduziu a menos que 5% a viabilidade de células A549. Por isso, tais fungos endofíticos foram submetidos a fermentação submersa em maior escala. O extrato de Alternaria alternata e algumas de suas frações reduziram expressivamente a viabilidade das três linhagens celulares testadas. Dentre tais frações, a F3 se destacou por apresentar menores valores de IC50 para MCF-7 e HepG2 (10,28±5,22 e 9,75±1,31 μg.mL-¹, respectivamente) do que o fármaco cisplatina (21,12±1,56 e 27,15±1,73 μg.ml-¹, respectivamente). O fracionamento dos extratos por cromatografia em coluna e cromatografia líquida de alta eficiência permitiram o isolamento de dois compostos, provenientes de A. alternata, e de um produzido por Lasiodiplodia sp. Tais amostras foram analisadas por ressonância magnética nuclear, e foi identificado o composto 4-hidróxi-benzaldeído no extrato de A. alternata. Ficou comprovado que os extratos de fermentação de fungos endofíticos isolados de H. impetiginosus exerceram importante atividade biológica, reduzindo a viabilidade de células derivadas de câncer de mama, fígado e pulmão. / Endophytic fungi have been prospected as source of active metabolites. In this work, endophytic fungi had been isolated from a tree that produces medicinal compounds, Handroanthus impetiginosus, in order to check if they produce biological active metabolites and to identify the biologically active species. All the 148 strains isolated were screened by agar piece method to investigate their antibiotic activity against Staphylococcus aureus ATCC 6538, Candida albicans ATCC 10231 e Escherichia coli ATCC 25922 . Among them, five endophytic fungi were fermented on small scale. The fermentation was performed in a Czapek broth, for 15 days, 28oC, under either stirring or static conditions. After this period, the cultures were filtered and subjected to liquid-liquid extraction. The obtained extracts were tested against the microorganisms mentioned above and the cancer cells MCF-7, HepG2 and A549. The lowest minimum inhibitory concentrations (MIC) exhibited were 125-250 μg.mL-¹ (from Alternaria alternata) and 250-500 μg.mL-¹ (from Lasiodiplodia sp.) both against S. aureus . Two extracts appreciably reduced tumor cells viability and, therefore, these endophytic fungi were fermented on large scale. The extract from A. alternata significantly reduced the viability of all three cell lines tested to less than 10% and its chemical study was biomonitored. One of its fractions, F3, exhibited lower IC50 against MCF-7 and HepG2 (10.28±5.22 and 9.75±1.31 μg.mL-¹, respectively) than cisplatin, a pharmaceutical compound commercially available (21.12±1.56 and 27.15±1.73 μg.mL-¹, respectively). The extract from Lasiodiplodia sp. diminished the viability of MCF-7 and HepG2 cells to less than 10%. The partitioning of the extracts by column chromatography and high performance liquid chromatography provided two isolated compounds from A. alternata and one from Lasiodiplodia sp. The samples were analyzed by nuclear magnetic resonance technique and the compound 4-hydroxybenzaldehyde was identified for A. alternata extract. L'induction par un hôte d'un stress oxydatif chez un parasite représente l'une des stratégies de défense chimique les plus répandues. Chez les Brassicacées, famille de plantes comprenant des espèces à fort intérêt agronomique comme le chou ou le colza, ou d'intérêt scientifique comme Arabidopsis thaliana, le stress oxydatif peut être généré soit par des formes actives d'oxygène libérées au niveau de la zone d'agression lors du burst oxydatif soit par des molécules issues du métabolisme secondaire de la plante telles que les isothiocyanates (ITC). Bien que fortement exposé à un tel stress lors de l'infection, Alternaria brassicicola, champignon pathogène nécrotrophe inféodé aux Brassicacées, est capable d'accomplir son cycle parasitaire et donc de s'y adapter. Cette thèse met en évidence que la tolérance du champignon aux stress oxydatif est partiellement dépendante du métabolisme du mannitol qui participe à la résistance des conidies lors du burst oxydatif in planta ou à la protection contre les dommages intracellulaires des FAO générées par les ITC lors des phases précoces de l'interaction. Les travaux réalisés montrent également que la charge fongitoxique des dérivés des glucosinolates peut être inhibée par leur conjugaison au glutathion via des Glutathion-Stransférases (GST) spécifiques. L'altération du métabolisme du mannitol ou l'inactivation de ces GST diminue significativement le pouvoir pathogène d'A. brassicicola et la régulation de ces voies métaboliques est donc à considérer dans l'optique de développer de nouvelles stratégies de protection des cultures.

EFFICACY OF ORGANICALLY CERTIFIABLE MATERIALS AND NATURAL COMPOUNDS AGAINST FOLIAR HEMIBIOTROPHIC AND NECROTROPHIC FUNGI IN CANTALOUPE AND TOMATOFeliciano-Rivera, Merari 01 January 2011 (has links)
Kentucky reported a solid 13.1% growth in certified organic land from 1997 to 2002. The relative lack of research on disease management practices in Kentucky consistent with organic regulations is an issue that needs to be addressed to provide more reliable information to local farmers. Thus, the first objective of this research was to investigate the potential disease control obtained with natural, organically certifiable spray materials against Colletotrichum orbiculare in vitro and in vivo. The second objective was to test certifiable spray materials in combinations to identify synergistic interactions. The third objective was to evaluate Organic Material Review Institute (OMRI)-certified materials for managing Septoria leaf spot and early blight in tomato under field conditions. The fourth objective was to evaluate chitosan-based products against C. orbiculare in vitro and in vivo.
Essential oils, Trilogy®, and Actinovate®, failed to suppress C. orbiculare in vitro as well as cucurbit anthracnose. Bicarbonate salts, Regalia®SC, Sonata®, copper based-products, lime sulfur and water-soluble chitosan showed high antifungal activity in vitro. Bicarbonate salts, Sonata®, Serenade Max®, Soil Gard 12G®, copper based-products and lime sulfur reduced anthracnose disease severity in vivo.
In the synergism experiments only a limited number of mixtures showed synergistic interactions, but even in those cases, the effect was not consistent between experiments. The main response obtained was antagonism.
In field experiments the most effective fungicides for managing Septoria leaf spot and early blight of tomato were copper-based fungicides. None of the biological-based products (Sonata® and Serenade Max®)), plant-based extracts (Trilogy® and Regalia® SC), chitosan, ammonium bicarbonate nor horticultural lime sulfur provided a significant reduction in disease severity.
For the fourth objective, water-soluble chitosan with a molecular weight between 3 to 10 kDa (80 and 85% deacetylated) showed the highest antifungal activity among all chitosan-based products evaluated in vitro. Also, combining the in vitro and in vivo results suggest that the antifungal activity of chitosan-based products is molecular weight- and concentration-dependent. These results provide a significant advance in the evaluation of the efficacy of OMRI-certified materials and natural materials to help organic farmers in Kentucky and the USA to manage diseases.

Previous issue date: 2002-12-03 / Fundação de Amparo à Pesquisa do Estado de Minas Gerais / A busca de alimentos produzidos sob um sistema de manejo menos agressivo ao meio ambiente vem sendo adotado por um número cada vez maior de produtores. Entretanto, apesar de existirem diversos benefícios na redução ou até eliminação do uso de defensivos, a grande diversidade de doenças em tomateiro (Lycopersicon esculentum Mill.), capazes de limitar a produção, torna necessária a busca por alternativas viáveis, eficientes e tecnicamente comprovadas. Dentre os organismos mais estudados, bactérias têm sido relatadas como agentes de biocontrole capazes de atuar por meio de mecanismos como antibiose, parasitismo, competição e indução de resistência. O presente trabalho teve como objetivos selecionar bactérias do filoplano do tomateiro, baseado em uma estratégia de seleção in vivo, verificando se há um método de isolamento que permita obter antagonistas eficientes no controle da pinta-preta, causada por Alternaria solani, requeima por Phytophthora infestans, mancha-bacteriana pequena por Pseudomonas syringae pv. tomato e mancha-bacteriana por Xanthomonas vesicatoria. Objetivou ainda estudar a possibilidade dos mecanismos de antibiose e indução de resistência serem responsáveis pelo controle destas doenças e se testes de antibiose in vitro são adequados como critério de seleção. Caracterizar aspectos biológicos dos antagonistas que podem otimizar sua aplicação como agente de biocontrole. Determinar a quais produtos antimicrobianos os isolados são insensíveis, visando fornecer subsídios para o desenvolvimento de meios semi-seletivos e estudos de dinâmica populacional. Verificar sua compatibilidade com antibióticos e fungicidas registrados para o controle de enfermidades do tomateiro, a fim de inseri-lo no sistema de manejo integrado e estudar a eficiência de antagonistas selecionados em condições de campo. Os resultados demonstram que, em folíolos mais jovens, os métodos de isolamento que visam obter bactérias da população total e da superfície do filoplano, foram os que permitiram obter a maioria dos antagonistas. O único obtido de folíolos mais velhos foi proveniente da população capaz de habitar sítios protegidos do filoplano e/ou resistir a fatores de estresse. Não se observou relação entre características biológicas dos antagonistas e dos patógenos testados. Nos testes de antibiose com os antagonistas selecionados, o isolado UFV-STB 6 foi capaz de produzir compostos voláteis e inibir a germinação de cistos de Phytophthora infestans, o que possivelmente deve estar envolvido no controle da requeima. O isolado UFV-IEA 6 produziu quitinase, havendo uma tendência em reduzir a taxa de crescimento de Alternaria solani por compostos voláteis. Ficou demonstrado que os testes de antibiose in vitro são inadequados como critério para seleção de agentes de biocontrole do filoplano de tomateiro. A caracterização dos melhores antagonistas demonstrou que três são bactérias Gram-positivas, em forma de bastonete, e uma Gram-negativa, pleiomórfica. Dentre as Gram-positivas todas são anaeróbias facultativas e uma forma endósporos. Nenhum antagonista foi capaz de causar reação de hipersensibilidade (HR) em fumo e produzir pigmento fluorescente in vitro. Os períodos de geração calculados a partir das curvas de crescimento revelaram que três isolados são capazes de se multiplicar rapidamente em meio de cultura, o que é uma característica desejável. Os resultados obtidos a partir dos antibiogramas, mostraram que existem antibióticos que podem ser utilizados para elaboração de meios semi-seletivos, adequados a cada antagonista e os testes de compatibilidade com antibióticos e fungicidas utilizados na cultura do tomateiro revelaram que os antagonistas podem ser expostos aos fungicidas benomyl, enxofre, dimetomorph e tiofanato-metílico. Verificou-se também a inadequação de se utilizarem compostos antimicrobianos em meio de cultura para isolamento de agentes bacterianos de controle biológico, uma vez que os antagonistas selecionados foram sensíveis à maioria dos produtos testados. Os testes com as enzimas indicadoras do estado de indução de resistência, β-1,3-glucanases, Fenilalanina amônia-liase (PAL), Peroxidases (PO), Polifenoloxidases (PPO) e Lipoxigenases (LOX), indicaram que o isolado UFV-IEA 6 foi capaz de promover aumento significativo na atividade das PO, evidenciando a possibilidade do antagonista agir como indutor de resistência. Esse parece ser o primeiro caso que se tem conhecimento de uma bactéria não fitopatogênica do filoplano induzindo resistência na mesma cultura de onde foi obtida. Os testes com os dois antagonistas em condições de campo demonstraram que UFV-STB 6 foi o mais eficiente em reduzir a severidade da requeima no terços médio e superior das plantas, enquanto UFV-IEA 6, somente no terço superior. Houve tendência na redução do progresso da septoriose por UFV-STB 6 e capacidade em diminuir o número de frutos com sintomas de requeima. Os resultados demonstram o potencial de uso dos agentes de biocontrole selecionados para as doenças da parte aérea de tomateiro estudadas. / Farmers are increasingly adapting environmentally less aggressive management systems for food production and there are many benefits in reducing or even eliminating pesticide use. Due to a large number of production limiting diseases on tomato (Lycopersicon esculentum), it is desirable to find technically viable and proven alternatives for their control. Among the microorganisms, bacteria have been reported as biocontrol agents capable of acting through antibiosis, parasitism, competition and induced resistance. The present study aimed at selecting the tomato phylloplane bacteria, based on in vivo isolation strategy, and to determine if this method permits obtaining efficient antagonists to control Alternaria solani leaf spot, Phytophthora infestans blight, small bacterial leaf spot caused by Pseudomonas syringe pv tomato, and bacterial leaf spot caused by Xanthomonas vesicatoria. The study also aimed at determining mechanisms of action, such as antibiosis and induced resistance, involved in disease control, and to determine if the antibiosis tests are sufficient selection criteria. The study also included biological characterization of the antagonists that may optimize their use. To help develop selective or semi-selective media for population dynamic studies, insensitivity of selected isolates to some antimicrobial compounds was also determined. The compatibility of select antagonists with antibiotics and fungicides registered for control of tomato diseases was elucidated so that the antagonist can be inserted in the integrated management. Field studies were done to determine the efficiency of select antagonists. The isolation method that obtain total bacterial population from the phylloplane of the young leaflets permitted obtaining maximum number of antagonists. The only one isolate obtained from the older leaflets originated from the population capable of inhabiting protected sites of phylloplane and/or that resist stress factors. There was no relation between biological characteristics of the antagonists and of the pathogen tested. In the antibiosis testes, the isolate UFV-STB 6 produced volatile compounds that inhibited germination of P. infestance cyst and may be involved in the control of blight. The isolate UFV-IEA 6 produced chitinase and showed a tendency to reduce A. solani growth by the volatile compounds. In vitro antibiosis testes were inadequate criteria to select biocontrol agents from tomato phylloplane. The characterization of promising antagonists showed that three were Gram positive bacilli and one was gram negative pleiomorphic bacteria. Among the Gram positives all were facultative anaerobes and the one formed endospores. None of the antagonists caused hypersensitive reaction (HR) in tobacco, and did not produce fluorescent pigment in vitro. The generation period calculated from the growth curve revealed that three isolates are capable of multiplying rapidly in the culture media, which is a desirable characteristic. The antibiograms showed that there are antibiotics that can be used for elaboration of semi-selective media for each of antagonists and the compatibility testes with antibiotics and fungicides used on tomato crop revealed that the antagonists can be exposed to fungicides such as benomyl, sulfur, dimethomorph and thiophante-methyl. Many antimicrobial compounds were inhibitory to the selecte antagonists in culture media used for isolation of bacterial biocontrol agents, therefore were inadequate for use in selective media. The analysis of enzymes involved in induced resistance, like -1,3-gluconase, phenylalanine ammonia lyase (PAL), peroxidase (PO), polyphenol oxidase (PPO) and lipoxigenase (LOX), showed that the isolate UFV-IEA 6 was capable of increasing PO activity, showing the possibility of being a resistance inducer . This appears to be the first case of a non-pathogenic phylloplane bacterium inducing resistance in a plant of origin. The field testes with two antagonists, UFV-STB 6 was more efficient in reducing the blight severity in the middle and upper third of the plant, while UFV-IEA 6 only in the upper third. The latter isolate also showed a tendency for reducing the Septoria leaf spot progress and the number of fruits with the blight symptoms. Previous issue date: 2003-03-14 / Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / Visando entender os mecanismos envolvidos na resistência de quatro cultivares de batata à pinta preta, esse trabalho teve por objetivos: 1) quantificar os componentes epidemiológicos da resistência dos cultivares Aracy, Delta, Desirée e Bintje que apresentam diferentes níveis de resistência à pinta preta, e estudar o efeito da idade dos tecidos nesses componentes; 2) realizar a análise histológica do processo infeccioso nesses cultivares, visando identificar possíveis mecanismos estruturais e relacionados com os níveis de resistência dos cultivares, e com a resistência de tecidos jovens; 3) verificar se a expressão diferencial de genes homólogos aos genes que codificam as proteínas PR gluB (glucanase básica de tabaco) e chiB (quitinase básica de feijão) respectivamente, se correlaciona com os níveis de resistência dos cultivares analisadas, e com a maior resistência dos tecidos jovens. Os componentes epidemiológicos da resistência, período de incubação (PI), número de lesões (NL), a severidade, taxa de expansão das lesões (TEL), período latente (PL) e o número de esporos por área foliar afetada (NEAA) foram quantificados nos terços inferior, médio e superior de plantas dos cultivares Aracy (resistente), Delta (resistência intermediária), Desirée(suscetível) e Bintje (suscetível) inoculadas com Alternaria solani numa concentração de 10 3 conídios.mL -1 . No cultivar Aracy constatou-se maior PI, redução do NL, da TEL e conseqüentemente da severidade em todos os terços avaliados. Já o menor NL, TEL, e severidade no terço médio explicam a resistência moderada do cultivar Delta. O cultivar Desirée, classificado como suscetível, apresentou, no terço superior, NL e severidade similar ao cultivar Aracy. O PL e o NEAA não foram adequados para a avaliação da resistência em condições de casa de vegetação e, provavelmente, foram influenciados pela falta de molhamento foliar. Todos os componentes foram influenciados pela idade dos tecidos. Maior PI e menores valores de NL, TEL, severidade e AACPD foram observados no terço superior, comprovando a menor suscetibilidade desses tecidos. Foram efetuados estudos histológicos do processo infeccioso de A. solani nas diferentes cultivares e terços usando a técnica de diafanização de tecidos. Foi quantificado o número de conídios germinados, número de apressórios, número de penetrações diretas e número de sítios de penetração exibindo células com reação de hipersensibilidade (HR). Não se constatou mecanismos de resistência que pudessem ser associados com os níveis de resistência dos cultivares estudados, nem com a maior resistência dos tecidos jovens. Constatou-se uma relação positiva entre o número de sítios de penetração mostrando HR e a resistência do cultivar Aracy. Houve associação desse componente com a resistência dos tecidos jovens. A expressão de genes homólogos à glucanase básica de tabaco (gluB) e à chitinase básica de feijão (chiB) como resposta a infecção de A. solani, foi analisada nos terços inferior e superior dos cultivares Bintje e Aracy, mediante a técnica de "Northern Blotting". Um aumento na expressão de transcritos a gluB e chiB foi constatado no terço inferior do cultivar Aracy, mas não em Bintje. Esse padrão de expressão sugere um possível envolvimento de isoformas destes genes com a resistência de Aracy a A. solani. Essa expressão diferenciada, não foi observada em folhas do terço superior desse cultivar sugerindo um padrão de regulação gênica dependente da idade dos tecidos, e o envolvimento de outros mecanismos bioquímicos na resistência dos tecidos jovens. / The resistance components of four potato cultivars to Alternaria solani were studied in three different levels: 1) quantification of epidemiological components related to resistance in cultivars Aracy, Delta, Desirée, and Bintje, and the effect of age-related resistance in these components; 2) histological analysis of the infectious process in these cultivars, aiming at identifying possible structural mechanisms related to resistance; 3) differential expression of genes homologous to gluB (basic glucanase of tobacco) and chiB (basic chitinase of bean) that encode pathogenesis-related proteins (PR) and the correlation between gene expression levels and cultivar/tissue age resistance levels. The epidemiological components related to resistance, incubation period (PI), number of lesions (NL), early blight severity, lesion expansion rate (TEL), latent period (PL), and the number of spores produced by lesion area (NEAA) were quantified in the lower, middle, and upper parts of cultivars Aracy (resistant), Delta (intermediate resistance), Desirée (susceptible), and Bintje (susceptible) inoculated with a suspension of 103 conidia of A. solani.mL-1. Longer PI, reduced NL, low TEL, and low severity were recorded in Aracy in all parts of the plant. For 'Delta', the reduced NL, low TEL and severity on the middle part of the plant corroborates its intermediate resistance level. Desirée is a susceptible cultivar, but NL and severity on the upper part did not differ from Aracy. The PL and NEAA were not informative when assessing resistance under greenhouse conditions. Probably, PL and NEAA were influenced by the shortage or lack of leaf wetness. All components were influenced by tissue age. Longer PI and smaller values of NL, TEL, and severity were observed in the upper leaves, indicating higher resistance of these tissues. Histological analyses of the infectious process of A. solani in different cultivars were carried out by tissue discoloration. The number of germinated conidia, number of apressoria, number of direct penetrations, and number of penetration sites with cells exhibiting hypersensitive reaction (HR) were quantified. No mechanisms of resistance were associated with cultivar resistance levels or with tissue age. For cultivar Aracy there was a direct relationship between number of penetration sites showing HR and cultivar resistance. The number of sites with HR was also associated with resistance of young tissues. The expression of gluB and chiB in response to A. solani infection was analyzed in the lower and upper parts of 'Bintje' and 'Aracy' plants by Northern blotting analysis. An increase in the level of transcripts related to gluB and chiB was recorded in the lower part of Aracy, but not in Bintje. Differential expression suggests the involvement of isoforms of these genes in Aracy resistance. Differential expression was not observed in leaves of the upper part of Aracy suggesting an age-dependent gene regulation process and/or the involvement of other biochemical mechanisms associated with young tissue high resistance level. / Tese importada do Alexandria
