Vliv hyaluronanu na transdermální penetraci vybraných léčivých látek / Hyaluronan effect on transdermal penetration of selected pharmaceutical substances

This work deals with the transmittance of a family of non-steroidal anti-inflammatory drugs across biological membranes in combination with the use of hyaluronan. Hyaluronic acid (hyaluronan, HYA) is a linear polysaccharide formed from disacharide units containing N-acetyl-D-glucosamine and glucuronic acid. HYA is present in almost all biological fluids and tissues, so there is an assumption that could affect the penetration of substances through the skin. Standards were prepared by anti-inflammatory drugs in admixture with various concentrations hyaluronan and subsequently measured transmittance of such substances through the skin. Standards were applied to the skin obtained from pig auricle. The drug content was determined by HPLC.

Este trabalho tem como principal objetivo contribuir para o desenvolvimento de novos potenciais metalofármacos de rutênio. Nele são descritas a síntese, a caracterização e a avaliação da ação antiproliferativa de alguns complexos de dirutênio (II,III) com os fármacos antiinflamatórios não-esteróides (AINEs): ibuprofeno (ibp), ácido acetilsalicílico (aas), naproxeno (npx) e indometacina (ind) e também com o ácido &#947;-linolênico (lin), sobre células cancerígenas. Os compostos obtidos foram caracterizados por análise elementar, espectroscopia de absorção eletrônica, medidas de susceptibilidade magnética, espectroscopia vibracional FTIR e Raman, difratometria de raios X de pó, medidas de condutividade molar e análise térmica (TG, OTAe OSC). Todos os complexos sintetizados apresentam estrutura em gaiola, com os carboxilatos derivados dos fármacos AINEs coordenados à unidade dimetálica Ru2( (II,III), em ponte equatorial, estabilizando assim a ligação direta rutênio-rutênio. As posições axiais são ocupadas por íons cloreto, no caso dos complexos [Ru2(O2(CR)4(Cl] (O2(CR = ibp, aas, npx ou ind), ou por moléculas de água, nas espécies do tipo [Ru2(O2(CR)4(H2O)2]PF6(O2CR =npx e ind). Ensaios biológicos demonstraram que os compostos [Ru2(ibp) 4Cl]&#8226;&#189;H2O e [Ru2(npx)4(H2O)2]PF6 apresentam ação antiproliferativa sobre células de glioma de rato C6 in vitro, dependendo do tempo de exposição do meio celular ao complexo. O complexo [Ru2 (lin)4Cl] também apresenta efeito sobre a proliferação de células C6; entretanto, nesse caso, efeitos significativos são observaçlos já nas primeiras 24 h de exposição. Estudos mostraram que as bases adenina e adenosina reagem com o complexo [Ru2(OAc)4(H2O)2]PF6 sem que ocorra quebra da estrutura em gaiola. As bases nitrogenadas substituem axialmente as moléculas de água, formando pontes axiais entre duas unidades de dirutênio (II,III) no estado sólido. / The main objective of this work is to make a contribution to the development of new ruthenium metallodrugs. Synthesis, characterization and evaluation of antiproliferative action in tumor cells of some diruthenium(II,III) complexes with Nonsteroidal Anti-inflammatory Drugs (NSAIDs) such as ibuprofen, acetylsalicylic acid, naproxen, indomethacin and also &#947;-linolenic acid are described. The obtained compounds were characterized by elemental analysis, electronic absorption spectroscopy, magnetic susceptibility, FTIR and Raman vibrational spectroscopy, x-ray powder diffraction, molar conductivity and thermal analysis (TG, DTA and DSC). All complexes exhibit a paddlewheel structure with the NSAID carboxylates coordinated to the metal ions bridging two Ru2(II,III) dimetal units and stabilizing the ruthenium-ruthenium bound. The axial positions are occupied by chloride ions for [Ru2(O2CR)4Cl] (O2CR = ibp, aas, npx, ind), or by water molecules for [Ru4(O2CR) 4(H2O) 2]PF6(O2CR = npx, ind) complexes. Biologic assays in vitro show that antiproliferative action in C6 rat cells depends on the time of exposing the cell culture to [Ru2(ibp)4Cl]&#8226;&#189;H2O and [Ru2 (npx)4(H2O)2]PF6 complexes. The [Ru2(1in)4Cl] compound also exhibits proliferative changes in C6 rat cells, however, significant results are observed for the first 24 hours of treatment. Some results show that adenine and adenosine bases can react with [Ru2(OAc)4(H2O)2]PF6 without breakage of the cage structure. The bases substitute the axial water molecules forming axial bridges between diruthenium units in the solid state.

L’apparition de plusieurs maladies, telles que le cancer, le diabète, l'hypertension artérielle et la propagation d'infections de type virus mutagènes peuvent être liées à la qualité et au mode de vie que nous menons aujourd’hui. En effet, plusieurs études sur les facteurs déclenchant ces maladies dites "morbides" à long ou à court terme, sont liées au stress et à la qualité des aliments consommés, qu'ils soient d'origine végétale ou animale. Ces maladies deviennent un phénomène courant, elles touchent différentes races et toutes les catégories de la société. D'après les recherches ethnobotaniques, les substances d’origine naturelle, ont permis à des civilisations de survivre à des maladies mortelles. A titre d'exemple, on retrouve ainsi des références à des périodes de fièvre paludique en Chine et à des symptômes de cette maladie dans le «Huangdi Neijiang» Le Canon de Médecine datant des environs du premier siècle avant notre ère, plus de 2000 ans, qui relate de l'emploi de plantes médicinales, pour soulager les fièvres (Desgrouas et al., 2014).Vers 186 avant J.-C. apparaît, dans certaines régions de Chine, l'utilisation en tisane, du Qing hao su, appelé plus tard artémisinine en Occident et extrait d'une plante médicinale utilisée comme antipyrétique appelée "Qing hao", Artemisia annua ou Armoise annuelle. L'artemisinine bloque une enzyme qui permet au parasite de pomper le calcium et l'empêche ainsi de se développer. Au jour d'aujourd'hui l’Artemisinin-based combination therapy, en français Thérapie combinée à base d'artémisinine et en sigle ACT, est une thérapie et une prévention tertiaire dans les cas de paludisme simple.Dans cette optique notre étude vient s'ajouter à une longue série d'études menées sur les plantes médicinales et les substances naturelles extraites. Elle a pour objectif de révéler de nouvelles biomolécules, de mettre en évidence leurs activités biologiques grâce à des techniques de biotechnologies d'une part. D'autre part ces investigations permettront de valoriser les ressources naturelles qui se distinguent par leur endémicité.Pour se faire, notre choix s'est porté sur deux plantes médicinales endémiques à l'Algérie Cyclamen africanum Boiss. & Reuter et Zygophyllum cornutum Coss. , après une recherche ethnobotanique sur la pharmacopée traditionnelle du Nord de l'Afrique, et qui a révélé l’efficacité de ces plantes dans les problèmes inflammatoires minimes chez les autochtones, nous avons entrepris des investigations pharmaco- biochimiques.Ces dernières nous ont permis d'isoler : cinq composés à partir de l'extrait méthanolique des racines de l'espèce Cyclamen africanum Boiss. & Reuter, deux nouvelles saponines triterpéniques de type Oleanane, Afrocyclamin A et B (1, 2), ainsi que trois saponines triterpénoïdes connus sous le nom de lysikokianoside (3), deglucocyclamin I (4) et de son dérivé d'acide dicrotalique (5); et Sept saponosides connus à partir de l'extrait méthanolique de la plante entière de Zygophyllum cornutum Coss., ces saponosides sont de type ursane, ce type de triterpène est rapporté dans cette espèce pour la première fois et peuvent être considérés comme un marqueur chimio-taxonomique (chemotype) du genre Zygophyllum. Les structures ont été élucidées, sur la base de l'analyse des spectres de l'expérience RMN-1D et RMN- 2D (COSY, TOCSY, NOESY, HMBC et HSQC) et spectrométrie de masse en source FAB mode ion négatif. Des activités biologiques, des fractions saponosidiques Fr.1 et Fr.2, ont été testées sur des lignées de Rats mâles et femelles, de la race Winstar pour évaluer l'activité anti inflammatoire.La fraction saponosidique Fr.1 de Cyclamen africanum à la dose 5 mg, a montré un effet significatif sur l'inflammation causé par la carragénine, en réduisant l'oedème et la réponse immunitaire, qui s'est traduite par la concentration des protéines de la réponse inflammatoire (PRI) à travers leurs action sur les pro-médiateurs de l'inflammation (COX-2, PGE2, TNF -α, iNOS). / The appearance of several diseases, such as cancer, diabetes, high blood pressure and spread of infections mutagenic virus type can be linked to the quality and lifestyle that we lead today. Indeed, several studies on the factors triggering these so-called "morbid" long-or short-term illnesses are related to stress and quality of food consumed, whether of plant and animal origin. These diseases are becoming a common occurrence, they affect different races and all classes of society. According ethnobotanical research, naturally occurring substances, allowed civilizations to survive deadly diseases. For example, we thus find references to periods of malarial fever in China and one of the symptoms of this disease in the "Huangdi Neijiang" The Canon of Medicine dating from around the first century BC, more than 2000 years, which relates to the use of herbal medicines to relieve fevers (Desgrouas et al., 2014).Around 186 BC appears, in some parts of China, the use in herbal tea, Qing hao su, later known as artemisinin in the West and extracted from a medicinal plant used as antipyretic called "Qing hao" Artemisia annua or annual wormwood. Artemisinin blocks an enzyme which enables the parasite to pump calcium and prevents it from developing. As of today the Artemisinin-based combination therapy in French Combination therapy of artemisinin and ACT acronym, is a therapy and tertiary prevention in cases of uncomplicated malaria.From this perspective our study adds to a long series of studies on medicinal plants and natural substances extracted. It aims to reveal new biomolecules, highlighting their biological activities through techniques of biotechnology on the one hand. Moreover, these investigations will develop natural resources that are characterized by endemic.To do this, our choice is focused on two endemic medicinal plants in Algeria Cyclamen africanum Boiss. & Reuter and Zygophyllum cornutum Coss. After an ethnobotanical research on traditional medicine in Northern Africa, which showed the effectiveness of these plants in minimal inflammatory problems among Aboriginal, we undertook biochemical pharmacological investigations.The latter allowed us to isolate, five compounds from the methanol extract of the roots of the species Cyclamen africanum Boiss. Reuter & two new oleanane triterpene saponins type, Afrocyclamin A and B (1, 2) and three triterpenoid saponins known lysikokianoside of (3), deglucocyclamin I (4) and its derivative dicrotalique acid (5) September and known from the methanol extract saponins from the whole plant of Zygophyllum cornutum Coss. these saponins are ursane type, type triterpenes are reported in this species for the first time and can be considered a chemotherapy marker Taxonomic (chemotype) of Zygophyllum kind. The structures were elucidated on the basis of the analysis of NMR spectra of the experience-1D and 2D-NMR (COSY, TOCSY, NOESY, HSQC and HMBC) and mass spectrometry method negative ion FAB source. The biological activities of saponosidiques FR.1 and Fr.2 fractions were tested on lines of male and female rats of the Winstar rats to evaluate the anti-inflammatory activity. The saponosidique fraction FR.1 Cyclamen africanum the 5 mg dose, showed a significant effect on inflammation caused by carrageenan, reducing edema and immune response, which resulted in the concentration of protein the inflammatory response (PRI) through their action on the pro-inflammatory mediators (COX-2, PGE2, TNF -α, iNOS). The fraction of Fr.2 saponosidique Zygophyllum dose 20 mg did not show a significant effect on inflammation in general.

Estudo físico-químico da sorção de diclofenaco em resina colestiramina / Physical-chemical study of the sorption of diclofenac onto cholestyramine resin

Os seguintes parâmetros foram investigados para a síntese do resinato de diclofenaco colestiramina: sal precursor, concentração inicial, tempo de contato, velocidade de agitação e temperatura. Dos estudos de equilíbrio, foi verificado que os íons diclofenaco foram sorvidos em dois tipos de sítios diferentes da resina e puderam ser adequadamente representados pela soma de dois termos, derivado do modelo de Boyd, para quimissorção, e Langmuir, para fisissorção. Os dados cinéticos foram ajustados pelos modelos de: pseudo-primeira ordem, pseudo-segunda ordem e difusão intrapartícula. O modelo de pseudo-segunda ordem ajustou-se aos dados experimentais para todo o período de sorção com alto coeficiente de correlação linear. Equações foram desenvolvidas a partir do modelo de pseudo-segunda ordem e puderam predizer a quantidade de íons diclofenaco sorvida em qualquer tempo para a concentração inicial de 7,5 g.L-1. Os resultados dos parâmetros termodinâmicos sugeriram que o processo de sorção é resultante de difusão e reação química em relação à temperatura. / The following parameters were investigated for the synthesis of diclofenac-cholestyramine resinate: diclofenac salt precursor, initial diclofenac concentration, contact time, stirring speed and temperature. From equilibrium studies was verified that the diclofenac ions were adsorbed on two different sites of the resin and may be adequately represented by the sum of two terms, derived from the Boyd model, for chemisorption, and Langmuir, for physisorption. The kinetic data were fitted to the pseudo-first, pseudo-second and intraparticle diffusion models. The pseudo-second order model was found to fit the experimental data for the entire sorption period with high coefficient of determination. Equations were developed using pseudo-second order model and could predict the amounts of diclofenac ions adsorbed at any contact time for the initial concentration of 7,5 g.L-1. The results of thermodynamic parameters suggest that the sorption process results for both diffusion and chemical bonding processes in relation to the temperature.

Conventional non-steroidal anti-inflammatory drugs (NSAIDs) may cause nephrotoxicity, interfering with renal hemodynamics and fluid and eletrolyte homeostasis. Recently, new selective COX-2 inhibitors have been developed producing less side effects (gastric, cardiac and renal) related to COX-1 inhibition. The increasing use of FK 506 and the intensive use of NSAIDs with analgesic or ani-inflammatory purposes, increases the possibility of a drug combination, potentiating the nephrotoxic risk of the two drugs. Objective : Compare the renal function of rats receiving combination therapy with FK and a non-selective COX inhibitor, sodium diclofenac (SD) with those receiving FK and a selective COX-2 inhibitor, rofecoxib (RO). Material and Methods : Male Munich-Wistar rats receiving a low sodium diet (0.06%) for 7 days and gavage treatment for 7 days with FK (2 mg/kg/day), SD (10 mg/kg/day), RO (3 mg/kg/day), FK+SD, FK+RO and vehicle (control) were used. Glomerular filtration rate (GFR, inulin clearance, ml/min/100g); renal blood flow (RBF, Doppler ultrasound, ml/min); mean blood pressure (MBP, intracarotid probe, mmHg); renal vascular resistence ( RVR, mmHg/ml/min); hematocrit (Htc); urinary volume ( UV, &#956;l/min); solute clearance; renal histology; animal weight (g) and FK serum concentration (SCFK, ng/ml) were assessed. Results are presented as a mean±standart deviation and compared by ANOVA followed by Student-Neuman-Keuls test. Results : The GRF of the SD group was 0.98±0.03, RO 1.06±0.04 and FK 0.99±0.06 similar to control values (1.10±0.05). GRF values decreased in the FK+RO (0.86±0.06;p<0.05 vs RO and Control) and FK+SD (0.63±0.06;p<0.001 vs control, FK and SD groups and p<0.01 vs FK+RO) groups. RBF, MBP, RVR and Htc values were similar in all groups. Diuresis was lower in the groups with drug combination, but there was a statistically significant difference only between FK+SD and RO groups (8.38±0.46 vs 12.99±1.22;p<0.05). There were no significant histological chan ges in the treatment groups. The FK+SD group showed statistically significant weigth changes (-18±5) when compared to the Control and RO groups (6±2 and 5±2, respectively; p<0.001) and to the SD an FK+RO groups (0.2±4 and 1±2, respectively; p<0.01). SCFK was significantly decreased (p<0.05) for FK+SD and FK+RO (1.7±0.3 and 1.8±0.4) groups when compared to the FK group (3.2±0.4. Conclusions: The combination of FK and a non-selective COX inhibitor significantly decreased GFR regardless of a RBF decrease or RVR increase, and is probably a result of Kf decrease. The trend to antidiuresis was similar for the combination of FK with both classes of NSAIDs. FK combined to a non-selective COX inhibitor caused a mild systemic toxicity when compared with the COX-2 selective inhibitor. Serum FK concentrations were significantly lower in NSAIDs treated animals. / Introdução: O tacrolimus (FK 506) é uma potente droga imunossupressora, pode causar nefrotoxicidade aguda com diminuição do fluxo sanguíneo renal (FSR) e ritmo de filtração glomerular (RFG). Os antiinflamatórios não-hormonais (AINHs) convencionais podem causar nefrotoxicidade, interferindo na hemodinâmica renal e na homeostase de fluidos e eletrólitos. Recentemente surgiram novas drogas do grupo coxib que são inibidores seletivos da COX-2, e portanto teriam menos efeitos colaterais relacionados à inibição da COX-1 (gástricos, cardíacos e renais). O crescente uso do FK 506 e o intenso uso de AINHs com finalidade analgésica e ou antiinflamatória aumenta a possibilidade de utilização em conjunto, potencializando o risco de nefrotoxicidade das duas drogas. Objetivo: Comparar a função renal de ratos sob os efeitos do uso simultâneo do FK e de um inibidor não-seletivo da COX, o diclofenaco sódico (DS) e do FK e de um inibidor seletivo da COX-2, o rofecoxib (RO). Materiais e Método: Utilizaram-se ratos Munich-Wistar machos em dieta hipossódica (0,06%) por 7 dias e tratamento por gavagem por 7 dias com FK (2 mg/kg/dia), DS (l0mg/kg/dia), RO (3mg/kg/dia), FK+DS, FK+RO e veículo (Contr). Aferidos ritmo de filtração glomerular (RFG, depuração de inulina, ml/min/l00g); o fluxo sanguíneo renal (FSR, ultrasom Doppler, ml/min); a pressão arterial média (PAM, probe intracarotídeo, mmHg); a resistência vascular renal (RVR, mmHg/ml/min); hematócríto (Ht); o volume urinário (VU, pl/min); a depuração de solutos; a histologia renal; o peso dos animais (g) e a concentração sanguínea de FK CSFK, ng/ml). Os resultados são apresentados com médiaserro padrão da média e comparados por ANOVA seguido do teste Student-Neuman-Keuls. Resultados: O RFG do grupo DS foi 0,980,03, do RO foi 1,060,04 e do FK 0,990,06 similares ao controle (1,100,05). Houve queda do RFG nos grupos FK+RO (0,860,06;p<0,Os vs RO e Contr) e FK+DS (0,630,06;p<0,001 vs Contr,DS, RO e FK; p<0,01 vs FK+RO) Nota de Resumo O FSR, a PAM, a RVR e o Ht foram semelhantes em todos os grupos. A diurese foi menor nos grupos com associação de drogas, mas houve diferença estatisticamente significante apenas entre os grupos FK+DS e RO (8,380,46 vs l299l,22;p<0,05). Não ocorreram modificações histológicas significativas nos grupos estudados. O grupo FK+DS apresentou variação de peso (-185) estatisticamente significante em relação aos grupos Contr 62 e RO 52 (p<0,001) e DS 0,24 e FK+RO -12 (p<0,01). A CSFK diminuiu significativamente (p<0,05) para os grupos FK+DS e FK+RO (1,70,3 e 1,80,4) em relação ao grupo FK (3,20,4). Conclusões: A associação do FK com um inibidor não-seletivo da COX causou diminuição mais acentuada do RFG independentemente da diminuição do FSR ou aumento da RVR, sendo provavelmente decorrente da diminuição do Kf. A tendência à antidiurese foi similar para a associação do FK com as duas classes de AINHs. O FK associado com um inibidor não-seletivo da COX causou discreta toxicidade sistêmica quando comparado com inibidor seletivo da COX-2. Nos animais tratados com AINHs, as concentrações sanguíneas do FK foram significativamente menores.

Ensaios biológicos demonstraram que os compostos [Ru2(ibp) 4Cl]&#8226;&#189;H2O e [Ru2(npx)4(H2O)2]PF6 apresentam ação antiproliferativa sobre células de glioma de rato C6 in vitro, dependendo do tempo de exposição do meio celular ao complexo. O complexo [Ru2 (lin)4Cl] também apresenta efeito sobre a proliferação de células C6; entretanto, nesse caso, efeitos significativos são observaçlos já nas primeiras 24 h de exposição. Estudos mostraram que as bases adenina e adenosina reagem com o complexo [Ru2(OAc)4(H2O)2]PF6 sem que ocorra quebra da estrutura em gaiola. As bases nitrogenadas substituem axialmente as moléculas de água, formando pontes axiais entre duas unidades de dirutênio (II,III) no estado sólido. / The main objective of this work is to make a contribution to the development of new ruthenium metallodrugs. Synthesis, characterization and evaluation of antiproliferative action in tumor cells of some diruthenium(II,III) complexes with Nonsteroidal Anti-inflammatory Drugs (NSAIDs) such as ibuprofen, acetylsalicylic acid, naproxen, indomethacin and also &#947;-linolenic acid are described. The obtained compounds were characterized by elemental analysis, electronic absorption spectroscopy, magnetic susceptibility, FTIR and Raman vibrational spectroscopy, x-ray powder diffraction, molar conductivity and thermal analysis (TG, DTA and DSC). All complexes exhibit a paddlewheel structure with the NSAID carboxylates coordinated to the metal ions bridging two Ru2(II,III) dimetal units and stabilizing the ruthenium-ruthenium bound. The axial positions are occupied by chloride ions for [Ru2(O2CR)4Cl] (O2CR = ibp, aas, npx, ind), or by water molecules for [Ru4(O2CR) 4(H2O) 2]PF6(O2CR = npx, ind) complexes. Biologic assays in vitro show that antiproliferative action in C6 rat cells depends on the time of exposing the cell culture to [Ru2(ibp)4Cl]&#8226;&#189;H2O and [Ru2 (npx)4(H2O)2]PF6 complexes. The [Ru2(1in)4Cl] compound also exhibits proliferative changes in C6 rat cells, however, significant results are observed for the first 24 hours of treatment. Some results show that adenine and adenosine bases can react with [Ru2(OAc)4(H2O)2]PF6 without breakage of the cage structure. The bases substitute the axial water molecules forming axial bridges between diruthenium units in the solid state.

Estudo físico-químico da sorção de diclofenaco em resina colestiramina / Physical-chemical study of the sorption of diclofenac onto cholestyramine resin

Os seguintes parâmetros foram investigados para a síntese do resinato de diclofenaco colestiramina: sal precursor, concentração inicial, tempo de contato, velocidade de agitação e temperatura. Dos estudos de equilíbrio, foi verificado que os íons diclofenaco foram sorvidos em dois tipos de sítios diferentes da resina e puderam ser adequadamente representados pela soma de dois termos, derivado do modelo de Boyd, para quimissorção, e Langmuir, para fisissorção. Os dados cinéticos foram ajustados pelos modelos de: pseudo-primeira ordem, pseudo-segunda ordem e difusão intrapartícula. O modelo de pseudo-segunda ordem ajustou-se aos dados experimentais para todo o período de sorção com alto coeficiente de correlação linear. Equações foram desenvolvidas a partir do modelo de pseudo-segunda ordem e puderam predizer a quantidade de íons diclofenaco sorvida em qualquer tempo para a concentração inicial de 7,5 g.L-1. Os resultados dos parâmetros termodinâmicos sugeriram que o processo de sorção é resultante de difusão e reação química em relação à temperatura. / The following parameters were investigated for the synthesis of diclofenac-cholestyramine resinate: diclofenac salt precursor, initial diclofenac concentration, contact time, stirring speed and temperature. From equilibrium studies was verified that the diclofenac ions were adsorbed on two different sites of the resin and may be adequately represented by the sum of two terms, derived from the Boyd model, for chemisorption, and Langmuir, for physisorption. The kinetic data were fitted to the pseudo-first, pseudo-second and intraparticle diffusion models. The pseudo-second order model was found to fit the experimental data for the entire sorption period with high coefficient of determination. Equations were developed using pseudo-second order model and could predict the amounts of diclofenac ions adsorbed at any contact time for the initial concentration of 7,5 g.L-1. The results of thermodynamic parameters suggest that the sorption process results for both diffusion and chemical bonding processes in relation to the temperature.

Implication des métabolites de l'APP dans les troubles mnésiques précoces chez la souris TgCRND8, un modèle de la maladie d'Alzheimer / Differential contribution of APP metabolites to early memory deficits in a TgCRND8 mouse model of Alzheimer’s disease

La maladie d’Alzheimer (MA) est une pathologie neurodégénérative communément caractérisée par une perte progressive de la mémoire. L’étiologie de la MA demeure incertaine à ce jour ce qui complique l’élaboration de stratégies thérapeutiques permettant de l’éradiquer. L’accumulation des échecs thérapeutiques pourrait en partie s’expliquer par le fait que l’hypothèse amyloïde, qui met en avant l’implication prépondérante du peptide bêta-amyloïde (Aβ) dans la physiopathologie de la MA, serait incomplète. En utilisant un modèle murin transgénique de la MA, la souris TgCRND8, j’ai pu compléter l’hypothèse amyloïde en proposant l’implication, en plus de l’Aβ, du fragment carboxy-terminal bêta (β-CTF). Ces deux métabolites amyloïdogéniques de l’APP seraient responsables de l’altération de formes différentes de mémoire. Le dosage de ces métabolites dans l’hippocampe, suite au traitement chronique des souris avec un inhibiteur de β ou de γ-secrétase, a mis en évidence que le β-CTF serait responsable de l’atteinte de la mémoire impliquée dans la détection du remplacement d’un objet, alors que l’Aβ perturberait la mémoire permettant la détection du déplacement d’un objet. Ces travaux suggèrent qu’il serait judicieux de développer de nouvelles stratégies thérapeutiques qui diminuent à la fois les niveaux cérébraux des deux fragments amyloïdogéniques, le β-CTF et l’Aβ. / Alzheimer’s disease (AD) is a neurodegenerative pathology commonly characterized by a progressive memory loss. To these days, AD’s etiology has remained unclear which complicates the development of therapeutic strategies enabling to eradicate the pathology. The accumulation of therapeutic failures could partly be explained by the fact that the amyloid hypothesis, which highlights the leading involvement of the amyloid beta peptide (Aβ) in the physiopathology of AD, could be incomplete. Using a transgenic mouse model of AD, the TgCRND8 mice strain, I expanded the amyloid hypothesis, suggesting the involvement of the beta carboxy-terminal fragment (β-CTF), in addition to Aβ. These two amyloidogenic metabolites could be responsible for the alteration of different forms of memory. The dosage of these metabolites, after mice chronic treatment with either a β- or a γ-secretase inhibitor, highlighted the fact that β-CTF could be responsible for the deterioration of the memory involved in the detection of the replacement of an object. As for Aβ, it could disrupt the memory allowing the detection of the displacement of an object. This work suggests that it would be judicious to develop therapeutic strategies reducing brain levels of both amyloid fragments, β-CTF and Aβ.

Stanovení reziduí léčiv pomocí separačních metod / Determination of drug residuals using separation methods

Nonsteroidal anti-inflammatory substances became a very frequently used and constantly evolved group of the drugs, particularly in a human medicine. The risk of their penetration into the environment, especially to the water environment, is rising during the recent years. The diploma thesis is focused on a selection of appropriate separation method and optimization of analytical procedure for the identification and determination of the non-steroidal anti-inflammatory drugs in sewage and surface waters. Salicylic acid, ketoprofen, diclofenac and ibuprofen were chosen from the wide range of NSAIDs as the most widespread agents of this drugs group in the medical practice. The method was optimized and elaborated using the model water samples. This method was used for determination of the analytes contained in the real water samples. Waste water samples were taken from the large-scale wastewater treatment plant in Brno-Modřice and surface water samples were taken from the Highlands river Křetínka. Solid phase extraction (SPE) was used for the real samples treatment and preconcentration, the determination was performed using the high performance liquid chromatography with mass spectroscopy method (LC-MS).
