Um sujeito tem outro nome além dos que lhe foram destinados pelos seus pais ou pela linhagem a que pertence. É portador também de um nome desconhecido que lhe é mais próprio e singular, que o marca no real do corpo e fala do modo como goza das marcas dos restos vistos ou ouvidos, registradas no seu inconsciente. Utilizando a metodologia da marca do caso, realizamos uma leitura clínica, a partir dos pressupostos da psicanálise, dos depoimentos de um sujeito, que apresenta a peculiaridade de ancorar seu ser em suas alterações corporais, a fim de mostrar que elas são sua tentativa de resposta ao excesso traumático que foi para ele o encontro com um gozo sem ordenamento fálico. Reconhecer o nome a que responde o gozo, para saber o que fazer com ele, é essencial ao reordenamento da economia de gozo, na maioria das vezes mortificante, como no caso objeto deste estudo. O resultado obtido aponta para questões inesperadas com as quais a clínica contemporânea tem se deparado, especialmente a das psicoses, no sentido das invenções singulares com que os sujeitos buscam se estabilizar ou construir um modo de laço social. Faz avançar o debate sobre a clínica do gozo, em jogo, nas construções de cada sujeito em particular. / A subject has another name besides those who were given by their parents or by the lineage to which it belongs. Therefore, it was selected 30 individuals through the procedure of Snow Ball (15 men and 15 women), aged between 22 and 40, who had at least 30% of their body surface covered by tattoos. On data collection, it was utilized a semi-directed interview, a picture of a chosen tattoo by the participant and the free association from this image. The data were analyzed according to the jungian psychosomatics. The results allowed to observe 11 categories: perception of aching, rising self-esteem, rising of attractiveness feeling, prejudices, regret on the choice of the image, religious factor, financial independency, moment of life they made the chosen tattoo, motivation for acquisition of the chosen tattoo, repercussion on the individual’s life after making the tattoo, symbols and images chosen. The results pointed out the incidence of prejudice, influence of the religious and financial factor at the time of tattooing, besides the perception of pain sometimes related to sacrifice, sometimes associated to self-mutilation behaviours. It was observed that the tattoo increases the self-esteem and eases extroversion on social relationships. It can be said that unconscious contents are transducted to skin through the tattoo. It is concluded that body marks help the individual on the appropriation of his own identity, contribute to the organization of psychological conflicts, and act as an important component on rites of passage. Finally, the tattoo can promote the self-knowledge / A presente pesquisa teve por objetivo investigar as representações de tatuagens e realizar uma análise simbólica das mesmas. Para tanto, foram selecionados 30 sujeitos através do procedimento Bola de Neve (15 homens e 15 mulheres), entre 22 e 40 anos de idade, que possuíam, no mínimo, 30% de sua superfície corpórea tomada por tatuagens. Na coleta de dados, foram utilizados uma entrevista semidirigida, a foto de uma tatuagem escolhida pelo participante e a associação-livre a partir desta imagem. Os dados foram analisados à luz da psicossomática junguiana. Os resultados permitiram observar 11 categorias: percepção de dor, aumento da autoestima, aumento do sentimento de atratividade, preconceito, arrependimento da escolha da imagem, fator religioso, independência financeira, momento de vida em que realizou a tatuagem escolhida, motivação para aquisição da tatuagem escolhida, repercussão na vida do indivíduo após a realização da tatuagem, símbolos e imagens escolhidas. Os resultados apontaram para a ocorrência de preconceito, influência de fatores religiosos e financeiros no ato de se tatuar, além da percepção de dor, ora relacionada a sacrifício, ora associada a comportamentos automutilantes. Observou-se que a tatuagem melhorou a autoestima e facilitou a extroversão nos relacionamentos sociais. Pode-se dizer que conteúdos inconscientes são transduzidos para a pele através da tatuagem. Concluiu-se que as marcas corporais auxiliaram o indivíduo na apropriação de sua identidade, contribuíram para a organização de conflitos psíquicos, e atuou como componente importante nos ritos de passagem. Design and synthesis of quinoxaline derivatives for medicinal application against breast cancer cells

Thesis (M.Sc.(Chemistry)) -- University of Limpopo, 2020 / Breast cancer is a malignant tumour that starts in the cells of the breast. Many studies revealed aromatase (CYP19A1) and cyclin-dependent kinase 2 (CDK2) as possible therapeutic targets regarding breast cancer treatment, because they play crucial roles in anti-apoptotic processes during cell proliferation. Quinoxaline derivatives have attracted a great deal of attention due to their biological activities against fungi, virus, bacteria and cancer. Computer modelling was employed in order to reduce time and cost by searching the library of molecules and identifying those which are likely to bind to the drug target. A library of new one hundred (100) nitro and amino quinoxaline alkyne derivatives were successfully designed and screened against target proteins (CYP19A1 and CDK2) using virtual screening technique and thirteen (13) molecules were identified to be hit compounds against both targets with the docking score ranging from -6.143 to -8.372 kcal/mol as a measure of binding affinity. The hit compounds were subjected to IFD in order to identify tight binding through intermolecular interactions with active site residues of the binding pocket of the target proteins. All identified nitro and amino quinoxaline alkyne derivatives were successfully synthesised in a multi-step reaction sequence and their spectroscopic analysis (NMR, FTIR and MS) were in good agreement with the proposed structures in a good to moderate yield. The newly synthesised novel amino and nitro-quinoxaline derivatives were evaluated for anti-proliferative activity against breast cancer (MCF-7). Compound 59 showed to possess good inhibition against MCF-7 with an IC50 of 9.102 μM, whereas compounds 34, 54, 56 and 61 showed promising activity against MCF-7 with an IC50 value of < 50 μM. However, the MTT assay results showed that 59 was found to be toxic with an IC50 value of 0.205 μM against Raw 264.7 cell line. The dose response investigations showed that 31 and 34 have the promising anti-cancer activity against CYP19A and the correlation between molecular modelling (in-silico) and CYP19A inhibition activities (in- vitro), was established as compounds 31 and 34 were identified to bind to the drug target (CYP19A) with the docking score of -8.372 and 7.630 kcal/mol respectively. All the synthesized compounds were evaluated for the antitubercular activity against Mtb H37Rv strain as a secondary study. Compounds 57-62 with nitro-quinoxaline derivatives exhibited stronger inhibitory effects on Mtb H37Rv strain. In addition, compounds 60 and 62 were found to be most active against Mtb H37Rv with the high activity at MIC90 of <0.65 and <0.64 μM respectively. Suspensão corporal e algumas implicações intersubjetivas / Body suspension and some intersubjective implications

Em seu uso original, a suspensão corporal era parte de rituais realizados em culturas milenares na índia e onde hoje são os Estados Unidos. Essa prática envolve a suspensão de uma pessoa por ganchos cravados em sua pele. A partir dos anos 1960, ela foi reapropriada e tem sido realizada especialmente em centros urbanos, geralmente, por pessoas que possuem modificações corporais - piercings, tatuagens, escarificações, implantes, brandings etc. Neste trabalho, pretendemos compreender os sentidos em torno dessa retomada, suas diferentes funções na vida dos praticantes contemporâneos e o que ela diz de nossa cultura. Com base em uma leitura inspirada em Winnicott, contrapomos a análise de relatos com o desenvolvimento infantil e sugerimos que essas práticas permitem avivar o sentimento de si como parte de um processo que possibilita novas formas de estar no mundo. Ao final, discutimos como a suspensão praticada atualmente só pode ser compreendida em sua inserção na cultura pós-moderna, seja por utilizar elementos extremamente contemporâneos - como a busca por uma performance esteticamente fascinante -, seja por tentar solucionar alguns impasses atuais - como o lugar destinado ao corpo em uma sociedade bastante mediada por imagens. Enfim, compreendemos que as mudanças culturais, de alguma forma, alteram o modo de experimentação do corpo. Contudo, há algo na experiência de suspensão corporal que não pode ser dito, um mistério compartilhado apenas por quem a viveu na própria carne. Como forma de circundar esse mistério, nós procuramos não apenas descrever e conceitualizar a suspensão, mas explorar a experiência de pesquisa do tema como via de acesso à prática propriamente dita. / Body suspension, in its origins, was part of rituals carried out in ancient societies like India and Native American cultures. This practice involves the suspension of the person by means of pierced hooks in the skin. It has been going under a rebirth since the sixties, especially in urban centers, and mostly by people whom already possess some kind of body modifications - piercing, tattooing, scarifications, implants, brandings, etc. The present study dedicates itself to understand the meanings around this rebirth and the different functions it may have in the life of contemporary practitioners, as well as to inquire if it has something to say about our own culture. Based on Winnicott\'s approach, we match the analysis of stories with the infantile development and suggest that these practices allow awakening the feeling of the self as part of a process that makes possible new forms to be in the world. Finally, we argue that the contemporary practice of suspension can be understood only in its insertion in the postmodern culture, either for using extremely contemporary elements - such as the search for a fascinating aesthetic performance - either for trying to solve some current setbacks - such as the place destined to the body in a society strongly mediated by images. At the end, we understand that cultural changes somehow modify the way we experiment our bodies. However, there\'s something left unsaid in the experience of body suspension, something of a mystery shared only by these whom experienced it in their own flesh. We tried to circle around this mystery by means of describing the suspension in its very concept, but also by means of exploring the research experience as a gateway to the practice itself.
