Influence of Primary Somatosensory Cortex on Hand Motor Circuitry and the Role of Stimulation Parameters

<p>The primary somatosensory cortex (SI) is important for hand function and influences motor circuitry in the primary motor cortex (M1). Areas 3a, 1 and 2 of SI have direct connectivity with M1. Much of our present knowledge of this connectivity and its relevance to hand function is based on animal research. However, less is known about the neural mechanisms that underpin hand function in humans. The present study investigated the influence of SI on corticospinal excitability as well as inhibitory and excitatory neural circuitry within M1 before and after continuous theta-burst stimulation (cTBS). Additionally, stimulation parameters influence the direction and magnitude of cTBS after-effects. Thus, current direction and frequency of cTBS were manipulated. Two experiments were performed. In Experiment 1, motor-evoked potentials (MEPs) were recorded from the first-dorsal interosseous (FDI) muscle bilaterally before and after 50 Hz cTBS over left SI. In a second condition, the orientation of cTBS was reversed. Experiment 2 measured MEPs, short-latency intracortical inhibition (SICI) and intracortical facilitation (ICF) from the right FDI following a modified 30 Hz cTBS over left SI or M1. The results of Experiment 1 and 2 demonstrate that SI influences M1 circuitry such that MEPs are facilitated following cTBS over SI. However, MEPs are suppressed when the current direction is reversed. CTBS at 30 Hz delivered over M1 suppressed excitatory circuitry that generates MEPs and ICF. The findings from the thesis suggest that SI influences hand motor circuitry and is likely a mechanism by which somatosensory information modulates hand motor function.</p> / Bachelor of Science (BSc)

Determining ideal staple size for small intestinal surgery in cats

Background: The use of stapling equipment for intestinal surgery in cats is rarely reported, and appropriate staple sizes for cat intestine are unknown. Objective: To determine staple cartridge sizes for thoracoabdominal (TA) and endoscopic gastrointestinal anastomosis (EndoGIA) that will simultaneously prevent leakage of small intestinal contents while also allowing for sufficient vascular permeability past the staple lines for intestinal healing. Methods: Two sizes of EndoGIA cartridges (2.0/2.5/3.0mm and 3.0/3.5/4.0mm) and two sizes of TA cartridges (2.5mm and 3.5mm), applied in a transverse manner across fresh cadaveric cat jejunum, were evaluated via intestinal burst pressure testing for maximum intraluminal pressure prior to leaking, and via infusion of an intravascular dye at normal arterial pressures to determine percentage of vascular patency past the staple lines. Vascular patency was compared not only from pre-and post-staple segments of the same intestinal sample, but also EndoGIA vascular patency was evaluated against TA vascular patency. Results: Two cats met study criteria. All samples had intraluminal burst pressures over twice the chosen minimum (of 30mmHg). Vascular patency post- staple line ranged from 0-90.8%, with the most consistently high numbers noted with the TA 3.5mm cartridges. No EndoGIA cartridge had a post- staple line vascular patency higher than 31.1%, and no intravascular dye was noted in any post- staple line sample in the EndoGIA 2.0/2.5/3.0mm group. Conclusions: While statistical analysis of the dataset was unable to be performed due to low numbers of samples for comparison, both intestinal intraluminal burst pressure trends and intravascular dye patterns suggested both the TA 3.5mm cartridge and (to a lesser extent) the 3.0/3.5/4.0mm EndoGIA cartridge could provide the ideal combination of intraluminal seal without restriction of vascular access for healing. The intravascular dye infusion technique, developed during this research, shows promise as a future instrument to determine vascular patterns around intestinal implants in cadaveric cat specimens. / Master of Science / Despite the regularity of feline small intestinal surgery, few reports exist of stapled anastomoses in cats, in part due to stapler size limitations. However, the recently developed endoscopic gastrointestinal anastomosis (EndoGIA) stapler shows promise as a future surgical tool for cats because it fits into cat intestine. In dogs, 3.5mm staples are often chosen for intestinal surgery; however, dog intestine is considerably thicker than cat intestine. The study goal was to evaluate not only intestinal burst pressures (the pressure at which repaired intestine leaks), but also the ability of fluids to flow through blood vessels that cross the staple lines of four stapler cartridge types from two staple lineages (EndoGIA 2.0/2.5/3.0mm, EndoGIA 3.0/3.5/4.0mm, TA 2.5mm, and TA 3.5mm). The central hypothesis was twofold. First, smaller stapler cartridge sizes (the EndoGIA 2.0/2.5/3.0mm and TA 2.5mm) would have higher intraluminal burst pressures when compared to the larger sizes (the EndoGIA 3.0/3.5/4.0mm). Second, larger stapler cartridges (the EndoGIA 3.0/3.5/4.0mm and the TA 3.5mm) would allow for increased flow of fluids in blood vessels past the stapler lines compared to the smaller cartridges (the EndoGIA 2.0/2.5/3.0mm and the TA 2.5mm). Two cats were included in the study. Trends in the data suggested that all components of the hypothesis might be proven with further data. However, due to the low number of cats acquired during the study period, the hypotheses could not be verified with statistics. The dye infusion technique to evaluate flow of fluids in blood vessels, developed during this research, shows promise as a future instrument to determine vascular patterns around intestinal implants. Future research should focus on acquiring more cats to have the ability able to perform statistical analyses (and prove the hypothesis), before proceeding with additional related studies.

Introdução. A Anemia das Doenças Crônicas (ADC) é uma desordem comum em idosos, freqüentemente multifatorial, e exacerbada por citocinas pró-inflamatórias. Nesta população, o diagnóstico de anemia por deficiência de ferro (ADF) é difícil utilizando-se os testes laboratoriais convencionais, devido à prevalência destes estados crônicos. Espécies reativas de oxigênio (ERO) são produzidas por fagócitos durante o burst oxidativo em defesa do hospedeiro, mas são também implicadas como agentes deletérios em um grande número de desordens inflamatórias. Objetivos. Verificar a eficiência do receptor de transferrina sérico (sTfR) e índice sTfR-log ferritina (sTfR-F) no diagnóstico da ADF e ADC; determinar os efeitos das anemias e de estados inflamatórios crônicos sem anemia no burst oxidativo, fagocitose, produção de óxido nítrico (NO) por monócitos, e produção de ácido hipocloroso (HOCl) por neutrófilos. Métodos. Participaram do estudo cinqüenta e três indivíduos (42 mulheres e 11 homens) idosos recrutados do departamento de Cardio-Geriatria da Rede Pública de Saúde de Ribeirão Preto-SP. A proteína C reativa (PCR), utilizada como marcador inflamatório foi analisada pela metodologia ultra-sensível. O status férrico foi estabelecido pelos níveis do sTfR (enzimaimunoensaio, kit Quantikine soluble transferrin receptor, R&D Systems, USA), ferritina sérica (ensaio quimioluminescente, kit Ferritin Immulite®- DPC, England) e índice sTfR-F. Foram compostos os seguintes grupos, com base nos valores de normalidade dos parâmetros PCR, concentração de hemoglobina, ferritina e sTfR: controle (n=15), ADC (n=12), inflamação (n=08), anemia inexplicável (n=06) e grupo ADF (n=12). Nenhum paciente apresentou função renal prejudicada, hemoglobinopatias, diabetes mellitus descompensada, doenças infecciosas ou malignas. Estudou-se o efeito da ADC, ADF, anemia inexplicável e inflamação no burst oxidativo, fagocitose de neutrófilos e monócitos por citometria de fluxo, produção de HOCl por neutrófilos purificados e de NO produzido em cultura de monócitos em meio RPMI 1640, estimulados por lipopolissacáride (LPS). Resultados. Os níveis de sTfR foram capazes de diagnosticar ADF em sete (58,34%) dos doze pacientes do grupo ADF. A intensidade de fluorescência (IF) do burst oxidativo de neutrófilos, que reflete a atividade celular, no grupo ADF foi significativamente menor que a do grupo controle (p<0,05), mas a IF de monócitos não foi estatisticamente diferente. As percentagens de monócitos e neutrófilos realizando burst oxidativo e fagocitose dos grupos ADF, anemia inexplicável e inflamação não foram estatisticamente diferentes do grupo controle. As percentagens de monócitos e neutrófilos realizando fagocitose do grupo ADC foram estatisticamente maiores que as do grupo controle (p<0,05). A produção de HOCl nos grupos ADC e inflamação foram estatisticamente menores comparadas ao grupo controle (p<0,05), enquanto houve uma superprodução de NO por monócitos no grupo ADC (p<0,05). Conclusão. No presente estudo, o sTfR foi uma ferramenta diagnóstica adicional à ferritina no diagnóstico da ADF, mas o valor do índice sTfR-F não aumentou a utilidade diagnóstica do sTfR em indivíduos idosos. NO parece modular a produção de HOCl, provavelmente por regular a atividade enzimática da mieloperoxidase (MPO). Estes resultados mostraram o papel importante do ferro na ADC e ADF em manter a resposta imunológica de fagócitos durante o processo de envelhecimento. / Background: Anemia of Chronic Disease (ACD) is a very common disorder in elderly and it is often multifatorial, exacerbated by pro-inflammatory cytokines. In these people, the diagnostic of iron deficiency anemia (IDA) or iron deficiency is difficult in these patients using the conventional iron status tests, because of the prevalence of these chronic states. Reactive oxygen species (ROS) are produced by phagocytic cells during the oxidative burst in defense of the host, but it has also been implicated as a very harmful agent in an increasing number of inflammatory-mediated disorders. Objectives: to elucidate the use of serum transferrin receptor (sTfR) and sTfR-log ferritin index (TfR-F index) to diagnose IDA and ACD; to determinate the effects of anemia and chronic inflammatory states without anemia in some function of oxidative stress, such as oxidative burst, phagocytosis, nitric oxide (NO) production by monocytes and the hypochlorous acid (HOCl) production by purified neutrophils. Methods: Fifty three (42 women and 11 men) elderly subjects recruited from geriatric department of healthy public system of Ribeirão Preto city were selected. The high sensitivity C-Reactive Protein (CRP) was analyzed as an inflammatory marker. The iron status was confirmed by sTfR (enzyme-linked immunosorbent assay, ELISA, kit Quantikine soluble transferrin receptor R&D Systems, USA), serum ferritin levels (chemiluminescence assay, kit Ferritin Immulite®- DPC, England) and TfR-F index. The groups were established according to the criteria CRP assay, hemoglobin analysis, ferritin and sTfR: control (n =15), ACD (n =12), inflammation (n =08), unexplained anemia (n=06), and IDA (n=12). Patients with impaired kidney function, hemoglobinopathy, decompensate diabetes mellitus or infectious and malignancies diseases were excluded. We also studied the effect of ACD, IDA, unexplained anemia and inflammation on oxidative burst, phogocytosis of neutrophils and monocytes by flow cytometry and HOCl generation of neutrophils and NO produced by the monocytic cell culture, cultured in RPMI 1640 medium, stimulated by lipopolysaccharide (LPS). Results: Our results showed that sTfR could diagnose IDA in seven (58.34%) of twelve patients in IDA group. Oxidative burst fluorescence intensity of neutrophil in IDA group was lower statistically compared to control group, p<0.05. Oxidative burst fluorescence intensity of monocytes did not differ significantly. The percentages of monocytes and neutrophils expressing oxidative burst and the phagocytosis did not differ significantly amongst control, IDA, unexplained anemia and inflammatory groups. However, the percentages of neutrophils and monocytes expressing phagocytosis in ACD group were statistically higher when compared to control group (p<0.05). The HOCl generation in ACD, and inflammation groups were lower significant statistically than control group, p<0.05, while there was an overproduction of NO by monocyte in ACD group (p<0.05). Conclusion: In the present study, the sTfR was an additional diagnostic tool to ferritin for the diagnosis of IDA, but the value of the sTfR-F index did not increase the utility diagnostic of sTfR in elderly patients. NO seems modulate the HOCl production, perhaps by regulates the myeloperoxidase (MPO) enzyme activity. These results showed the important role of iron in ACD and IDA to maintaining phagocyte immune defense of organism during the aging process.

Πρωτόκολλα και αρχιτεκτονικές σε δίκτυα μεταγωγής οπτικής ριπής

Η μεγάλη διαθεσιμότητα ευρυζωνικής πρόσβασης και η πληθώρα νέων διαδικτυακών υπηρεσιών οδήγησε στην ενσωμάτωση του διαδικτύου στην καθημερινότητά μας. Οι χρήστες του διαδικτύου μέσα στο 2011 έφτασαν το 1.5 δισεκατομμύριο με έντονα αυξητικές τάσεις, ιδιαιτέρως σε χώρες της Ασίας. Αυτή η αύξηση έχει γίνει εφικτή χάρη στο τεράστιο εύρος ζώνης και την υψηλή αξιοπιστία των οπτικών τηλεπικοινωνιακών δικτύων κορμού σε συνδυασμό με το όλο και μειούμενο κόστος ανά bit της μεταδιδόμενης πληροφορίας. Πρόσφατες μελέτες έχουν καταδείξει ότι η πλειοψηφία του μεταδιδόμενου όγκου δεδομένων στο διαδίκτυο (>50%) μεταδίδεται μέσω του πρωτοκόλλου HTTP, με ισχυρές ενδείξεις ότι το ενδιαφέρον των χρηστών απομακρύνεται από τις P2P εφαρμογές σε υπηρεσίες video/audio streaming και κοινωνικής δικτύωσης. Όμως, ενώ σταδιακά εμφανίζεται η ανάγκη για επόμενης γενιάς υπηρεσιών δικτύου, (e- science, τηλεργασία, HD-IPTV, VOD) με ιδιαίτερες απαιτήσεις σε εύρος ζώνης, ποιότητα υπηρεσίας και καθυστέρηση τα τηλεπικοινωνιακά δίκτυα στις μέρες μας είναι προσανατολισμένα στο να παρέχουν χαμηλού κόστους «best effort» υπηρεσίες χωρίς κανενός είδους εγγυήσεις. Η έλευση καινοτομικών διαδικτυακών υπηρεσιών έχει σαν αποτέλεσμα σταδιακά η χωρητικότητα των (ηλεκτρονικών) συσκευών μεταγωγής να γίνεται το σημείο συμφόρησης, αφού η μετατροπή των πακέτων στο ηλεκτρονικό πεδίο και η ένα-προς-ένα επεξεργασία των επικεφαλίδων τους δεν είναι κλιμακώσιμη σε ρυθμούς διαμεταγωγής της τάξης των Tbps. Η αρχιτεκτονική μεταγωγής οπτικής ριπής (OBS) μπορεί να δώσει λύση σε αυτό το πρόβλημα, χάρη στα σημαντικά της πλεονεκτήματα. Στην αρχιτεκτονική OBS, τα πακέτα δεδομένων συναθροίζονται σε ριπές (bursts) οι οποίες μεταδίδονται χωρίς προ-εγκατάσταση κυκλώματος, και μετάγονται με διαφανή-οπτικό τρόπο από την πηγή στον προορισμό. Επίσης η αρχιτεκτονική OBS είναι η μόνη (πρακτικά εφαρμόσιμη) αμιγώς οπτική τεχνολογία μεταγωγής με sub-lambda ικανότητες δέσμευσης πόρων. Η επίλυση ενός αριθμού ανοιχτών προβλημάτων που αντιμετωπίζει η αρχιτεκτονική OBS θα επιτρέψει την εμπορική αξιοποίησή της στα δίκτυα κορμού των τηλεπικοινωνιακών παρόχων. Στόχος μας στα πλαίσια αυτής της διδακτορικής έρευνας είναι η πρόταση καινοτομικών πρωτοκόλλων και αρχιτεκτονικών που δίνουν λύσεις σε ανοιχτά προβλήματα της τεχνολογίας OBS. Στην παρούσα διδακτορική διατριβή αρχικά στρέφουμε την προσοχή μας στην αποδοτική μετάδοση TCP κίνησης πάνω από OBS δίκτυα. Πρόκειται για ένα δύσκολο πρόβλημα που έχει μελετηθεί εκτενώς στη βιβλιογραφία, λόγω της ευαισθησίας του TCP πρωτόκολλου στις απώλειες πακέτων, οι οποίες συμβαίνουν στα OBS δίκτυα λόγω ανταγωνισμού ριπών ακόμα και σε συνθήκες χαμηλού φορτίου. Στη βιβλιογραφία έχουν προταθεί πληθώρα λύσεων για την αντιμετώπιση του προβλήματος, τόσο στο OBS domain (π.χ. με τη χρήση κατάλληλων πρωτόκολλων χρονοπρογραμματισμού) όσο και στο TCP domain (βελτίωση του TCP πρωτοκόλλου). Στα πλαίσια της παρούσας διδακτορικής διατριβής πραγματοποιήθηκε μια σειρά εκτενών προσομοιώσεων σε ρεαλιστικές συνθήκες με χιλιάδες ενεργές TCP πηγές, που εστιάζουν στην κατανομή TCP πακέτων και συνόδων στις μεταδιδόμενες ριπές καθώς και στη συμπεριφορά του παράθυρου συμφόρησης. Το συμπέρασμά μας ήταν ότι οι αλγόριθμοι συναρμολόγησης με μοναδικό κατώφλι εμφανίζουν βέλτιστη απόδοση μόνο για συγκεκριμένες TCP πηγές με κοινά χαρακτηριστικά, προτείνοντας τους αλγόριθμους συναρμολόγησης πολλαπλών κλάσεων σαν πιθανή λύση. Οι τελευταίοι δείξαμε ότι μπορούν να οδηγήσουν σε βελτίωση του TCP throughput, όταν κάθε κατηγορία TCP πηγής ανατίθεται σε διαφορετική κλάση συναρμολόγησης με διαφορετικό assembly timer. Ένα ακόμα πρόβλημα που παρατηρήθηκε κατά τη μεταφορά TCP κίνησης σε OBS δίκτυα και μελετήθηκε στις προσομοιώσεις μας ήταν ο συγχρονισμός των TCP πηγών. Η απώλεια πολλαπλών TCP πακέτων με μία χαμένη ριπή συχνά οδηγούσε σε συγχρονισμό των TCP πηγών που μεταφέρονταν στη ριπή, ένα πρόβλημα που χειροτέρευε όσο αύξανε η πιθανότητα απώλειας ριπής. Για την αντιμετώπιση του συγχρονισμού των TCP πηγών, προτάθηκαν μια σειρά από αλγόριθμοι συναρμολόγησης πολλαπλών κλάσεων, οι οποίοι εισάγουν ένα βαθμό τυχαιότητας στην ανάθεση TCP πηγών σε κλάσεις συναρμολόγησης για την αποφυγή του συγχρονισμού. Στη συνέχεια στρέψαμε το ενδιαφέρον μας στο πρόβλημα παροχής ποιότητας υπηρεσίας σε ένα OBS δίκτυο με υποστήριξη πολλαπλών κλάσεων, στοχεύοντας στην αποδοτική μετάδοση βίντεο κίνησης. Συγκεκριμένα, προτείναμε ένα νέο σχήμα παροχής ποιότητας υπηρεσίας το οποίο βασίζεται στην τεχνική των burst preemptions. Για να έχουμε μια ρεαλιστική εκτίμηση της βελτίωσης ποιότητας της βίντεο κίνησης, χρησιμοποιήσαμε μετρικές σχετικά με την αντιληπτή ποιότητα της video κίνησης, όπως το MOS score και το PSNR. Στη συνέχεια προτείνουμε μια νέα αρχιτεκτονική για δημιουργία προφίλ και πρόβλεψη κίνησης σε δίκτυα μεταγωγής οπτικής ριπής, με στόχο τη βελτιστοποίηση της μετάδοσης TCP κίνησης. Σε αυτή την αρχιτεκτονική, ένας TCP profiler υπολογίζει ένα σύνολο στατιστικών στοιχείων για τις ενεργές TCP πηγές παράλληλα με τη διαδικασία συναρμολόγησης ριπής. Αυτά αξιοποιούνται για την κατασκευή του μοντέλου κίνησης στο οποίο βασίζεται η πρόβλεψη του μεγέθους ριπών σε ένα χρονικό παράθυρο μήκους τουλάχιστον ίσου με RTT. Η πρόβλεψη του μεγέθους των ριπών επιτρέπει την εκ των προτέρων ενημέρωση των χρονοπρογραμματιστών του δικτύου κομού για επερχόμενες μεταβολές στην κίνηση του δικτύου, ώστε να επανα-χρονοπρογραμματίσει τις δεσμεύσεις πόρων με βέλτιστο τρόπο. Για την εκ των προτέρων ενημέρωση των κόμβων κορμού προτείνεται ένα νέο πρωτόκολλο προληπτικής δέσμευσης πόρων, του οποίου η συνεισφορά την απόδοση του δικτύου και τη χρησιμοποίηση των τηλεπικοινωνιακών γραμμών αξιολογείται με ένα σύνολο λεπτομερών προσομοιώσεων. Στο τελευταίο μέρος της διδακτορικής διατριβής στρέφουμε το ενδιαφέρον μας στις υβριδικές οπτικές αρχιτεκτονικές. Επειδή καμία οπτική δικτυακή τεχνολογία δε δίνει λύση σε όλα τα προβλήματα και τις απαιτήσεις των μελλοντικών οπτικών τηλεπικοινωνιακών δικτύων, πολλά υποσχόμενες είναι οι λεγόμενες υβριδικές τεχνολογίες οι οποίες συνδυάζουν χαρακτηριστικά από περισσότερες της μίας βασικές αρχιτεκτονικές (OBS, OCS και OPS). Σε αυτά τα πλαίσια προτείνεται η νέα υβριδική αρχιτεκτονική HOBS (Hybrid Optical Burst Switching) η οποία εκμεταλλεύεται τις κενές περιόδους κατά την εγκατάσταση κυκλωμάτων ενός Dynamic Circuit Switching δικτύου για τη μετάδοση ριπών δεδομένων. / The wide availability of broadband access has allowed users to tightly integrate network use into their daily lives. There will be 1.5 billion people with Internet access in 2011 and a big growth rate in the online population, primarily occurring in Brazil, Russia, India and China. This growth has been made possible due to the vast capacity of optical core networks, and the decreasing cost per bit. New research has shown that P2P no longer dominates internet traffic; HTTP is now dominant in terms of bytes transmitted by a big margin for a significant fraction of the Internet. User’s interest has shifted to new video/audio streaming services and social networking sites. However while new generation services are emerging, such as e-science, HD-IPTV and VOD, with high bandwidth requirements and special QoS needs modern commercial networks are engineered for low cost, best effort access. The advent of innovative web services along with the phenomenal increase of internet traffic has resulted in the capacity of electronic routers becoming the bottleneck point of modern telecommunication networks. With terabit-per-second bandwidths, IP routing that converts optical signals into electric signals and identifies IP headers one-by-one cannot meet the demands. Optical Burst Switching (OBS) architecture has beed proposed as a near- term solution to this problem. Data packets in OBS are assembled to bursts which are immediately transmitted without acknowledgements and switched all-optically in the network core. OBS is the only practical optical switch architecture that can offer sub-wavelength resource reservation. However, there are a set of limitations and open problems that hinder the wider adoption of OBS architecture. The contribution of this thesis is a set of innovative algorithms, protocols and architectures for OBS networks to enhance performance in real world networks. These include –but are not limited to– burst assembly and scheduling algorithms, reservation protocols and QoS differentiation. The efficient transmission of TCP traffic over OBS networks is a challenging problem in OBS networks, due to the high sensitivity of TCP congestion control mechanism to burst losses. Various burst assembly and burst scheduling algorithms have been proposed in the literature, to enhance the efficient transmission of TCP over OBS networks. However it still remains an open problem, since the relatively high burst loss ratio experienced in OBS networks is incompatible with TCP congestion control mechanism. It has been observed that burst losses have a significant impact on the TCP end-to-end performance. One burst loss typically results in many sources timing out and subsequently entering the slow start phase. In this thesis, we study the problem of TCP over OBS transmission through extensive simulations in a real-world scenario, with thousands of active TCP connections. We conclude that fixed timer-based burstifiers are not appropriate, since they do not provide maximum performance but only optimal performance for individual flows with similar characteristics. To address this problem, in this thesis we propose multi-class burst assembly algorithms. We have shown that these can lead to an enhanced TCP throughput, by assigning a different assembly timer to each class of service. In our simulations we observed that burst losses would often result in synchronizing TCP transmissions with an imminent effect on link utilization. This phenomenon was caused by multiple packet losses from different TCP flows in a single round and was exaggerated with high burst loss ratios. Thus, to avoid flow synchronization we proposed a set of multi-class burst assembly algorithms, where the assignment of flows to classes of service was non-deterministic. We then turned our attention to the problem of QoS provisioning in a multi-class OBS network. Our goal was to provide QoS differentiation to the transmission of video traffic over OBS networks. We propose a QoS-aware scheduling algorithm that supports QoS differentiation based on preemptions, which are controlled by a novel preemption policy. Performance evaluation of the proposed scheme is based on throughput measurements for the best effort class, and perceived quality metrics like PSNR and MOS scores for the video streaming class. We then turn our attention to a new TCP-specific traffic profiling and prediction scheme, which is proposed with the aim to optimize TCP transmission over one-way OBS networks. Traffic prediction is an interesting approach for the enhancement of TCP performance over OBS. If it would be possible to accurately predict TCP flows’ throughput, it would also be possible to predict burst sizes. That would allow making reservations of the appropriate resources in advance, enhancing network performance and improving bandwidth utilization. In the proposed scheme the burst assembly unit inspects TCP packet headers in parallel to the assembly process, keeping flow-level traffic statistics. These are then exploited to derive accurate traffic predictions, in at least one RTT-long prediction window. This allows in-advance notifying traffic schedulers of upcoming traffic changes, in order to optimally re-schedule their resource reservations. In this paper, we detail the traffic profiling and prediction mechanism and also provide analytical and simulation results to assess its performance. The performance gains, when using the prediction scheme are shown with a modified one-way OBS reservation protocol, which efficiently and in advance reserves resources at the burst level. In the final part of this thesis we turn our attention to hybrid optical networks. Since no single optical switch architecture covers all requirements of future telecommunication networks, hybrid architectures that combine the merits of two or more optical switch architectures are considered very promising. In this thesis, we present a radically different hybrid optical burst switch (HOBS) architecture that combines one-way with two-way reservation under a single, unified control plane (hybrid signaling) for QoS differentiation. It takes advantage of the idle, round-trip time delay during lightpath establishment phase to transmit one-way data bursts of a lower class of service, while high priority data explicitly requests and establishes end-to-end connections (lightpaths), as in wavelength-routed OBS.

Introdução. A Anemia das Doenças Crônicas (ADC) é uma desordem comum em idosos, freqüentemente multifatorial, e exacerbada por citocinas pró-inflamatórias. Nesta população, o diagnóstico de anemia por deficiência de ferro (ADF) é difícil utilizando-se os testes laboratoriais convencionais, devido à prevalência destes estados crônicos. Espécies reativas de oxigênio (ERO) são produzidas por fagócitos durante o burst oxidativo em defesa do hospedeiro, mas são também implicadas como agentes deletérios em um grande número de desordens inflamatórias. Objetivos. Verificar a eficiência do receptor de transferrina sérico (sTfR) e índice sTfR-log ferritina (sTfR-F) no diagnóstico da ADF e ADC; determinar os efeitos das anemias e de estados inflamatórios crônicos sem anemia no burst oxidativo, fagocitose, produção de óxido nítrico (NO) por monócitos, e produção de ácido hipocloroso (HOCl) por neutrófilos. Métodos. Participaram do estudo cinqüenta e três indivíduos (42 mulheres e 11 homens) idosos recrutados do departamento de Cardio-Geriatria da Rede Pública de Saúde de Ribeirão Preto-SP. A proteína C reativa (PCR), utilizada como marcador inflamatório foi analisada pela metodologia ultra-sensível. O status férrico foi estabelecido pelos níveis do sTfR (enzimaimunoensaio, kit Quantikine soluble transferrin receptor, R&D Systems, USA), ferritina sérica (ensaio quimioluminescente, kit Ferritin Immulite®- DPC, England) e índice sTfR-F. Foram compostos os seguintes grupos, com base nos valores de normalidade dos parâmetros PCR, concentração de hemoglobina, ferritina e sTfR: controle (n=15), ADC (n=12), inflamação (n=08), anemia inexplicável (n=06) e grupo ADF (n=12). Nenhum paciente apresentou função renal prejudicada, hemoglobinopatias, diabetes mellitus descompensada, doenças infecciosas ou malignas. Estudou-se o efeito da ADC, ADF, anemia inexplicável e inflamação no burst oxidativo, fagocitose de neutrófilos e monócitos por citometria de fluxo, produção de HOCl por neutrófilos purificados e de NO produzido em cultura de monócitos em meio RPMI 1640, estimulados por lipopolissacáride (LPS). Resultados. Os níveis de sTfR foram capazes de diagnosticar ADF em sete (58,34%) dos doze pacientes do grupo ADF. A intensidade de fluorescência (IF) do burst oxidativo de neutrófilos, que reflete a atividade celular, no grupo ADF foi significativamente menor que a do grupo controle (p<0,05), mas a IF de monócitos não foi estatisticamente diferente. As percentagens de monócitos e neutrófilos realizando burst oxidativo e fagocitose dos grupos ADF, anemia inexplicável e inflamação não foram estatisticamente diferentes do grupo controle. As percentagens de monócitos e neutrófilos realizando fagocitose do grupo ADC foram estatisticamente maiores que as do grupo controle (p<0,05). A produção de HOCl nos grupos ADC e inflamação foram estatisticamente menores comparadas ao grupo controle (p<0,05), enquanto houve uma superprodução de NO por monócitos no grupo ADC (p<0,05). Conclusão. No presente estudo, o sTfR foi uma ferramenta diagnóstica adicional à ferritina no diagnóstico da ADF, mas o valor do índice sTfR-F não aumentou a utilidade diagnóstica do sTfR em indivíduos idosos. NO parece modular a produção de HOCl, provavelmente por regular a atividade enzimática da mieloperoxidase (MPO). Estes resultados mostraram o papel importante do ferro na ADC e ADF em manter a resposta imunológica de fagócitos durante o processo de envelhecimento. / Background: Anemia of Chronic Disease (ACD) is a very common disorder in elderly and it is often multifatorial, exacerbated by pro-inflammatory cytokines. In these people, the diagnostic of iron deficiency anemia (IDA) or iron deficiency is difficult in these patients using the conventional iron status tests, because of the prevalence of these chronic states. Reactive oxygen species (ROS) are produced by phagocytic cells during the oxidative burst in defense of the host, but it has also been implicated as a very harmful agent in an increasing number of inflammatory-mediated disorders. Objectives: to elucidate the use of serum transferrin receptor (sTfR) and sTfR-log ferritin index (TfR-F index) to diagnose IDA and ACD; to determinate the effects of anemia and chronic inflammatory states without anemia in some function of oxidative stress, such as oxidative burst, phagocytosis, nitric oxide (NO) production by monocytes and the hypochlorous acid (HOCl) production by purified neutrophils. Methods: Fifty three (42 women and 11 men) elderly subjects recruited from geriatric department of healthy public system of Ribeirão Preto city were selected. The high sensitivity C-Reactive Protein (CRP) was analyzed as an inflammatory marker. The iron status was confirmed by sTfR (enzyme-linked immunosorbent assay, ELISA, kit Quantikine soluble transferrin receptor R&D Systems, USA), serum ferritin levels (chemiluminescence assay, kit Ferritin Immulite®- DPC, England) and TfR-F index. The groups were established according to the criteria CRP assay, hemoglobin analysis, ferritin and sTfR: control (n =15), ACD (n =12), inflammation (n =08), unexplained anemia (n=06), and IDA (n=12). Patients with impaired kidney function, hemoglobinopathy, decompensate diabetes mellitus or infectious and malignancies diseases were excluded. We also studied the effect of ACD, IDA, unexplained anemia and inflammation on oxidative burst, phogocytosis of neutrophils and monocytes by flow cytometry and HOCl generation of neutrophils and NO produced by the monocytic cell culture, cultured in RPMI 1640 medium, stimulated by lipopolysaccharide (LPS). Results: Our results showed that sTfR could diagnose IDA in seven (58.34%) of twelve patients in IDA group. Oxidative burst fluorescence intensity of neutrophil in IDA group was lower statistically compared to control group, p<0.05. Oxidative burst fluorescence intensity of monocytes did not differ significantly. The percentages of monocytes and neutrophils expressing oxidative burst and the phagocytosis did not differ significantly amongst control, IDA, unexplained anemia and inflammatory groups. However, the percentages of neutrophils and monocytes expressing phagocytosis in ACD group were statistically higher when compared to control group (p<0.05). The HOCl generation in ACD, and inflammation groups were lower significant statistically than control group, p<0.05, while there was an overproduction of NO by monocyte in ACD group (p<0.05). Conclusion: In the present study, the sTfR was an additional diagnostic tool to ferritin for the diagnosis of IDA, but the value of the sTfR-F index did not increase the utility diagnostic of sTfR in elderly patients. NO seems modulate the HOCl production, perhaps by regulates the myeloperoxidase (MPO) enzyme activity. These results showed the important role of iron in ACD and IDA to maintaining phagocyte immune defense of organism during the aging process.

Chez l’humain, différents protocoles de stimulation magnétique transcrânienne répétée (SMTr) peuvent être utilisés afin de manipuler expérimentalement la plasticité cérébrale au niveau du cortex moteur primaire (M1). Ces techniques ont permis de mieux comprendre le rôle du sommeil dans la régulation de la plasticité cérébrale. Récemment, une étude a montré que lorsqu’une première session de stimulation SMTr au niveau de M1 est suivie d’une nuit de sommeil, l’induction subséquente de la plasticité par une deuxième session SMTr est augmentée. La présente étude a investigué si ce type de métaplasticité pouvait également bénéficier d’une sieste diurne. Quatorze sujets en santé ont reçu deux sessions de intermittent theta burst stimulation (iTBS) connue pour son effet facilitateur sur l’excitabilité corticale. Les sessions de stimulation étaient séparées par une sieste de 90 minutes ou par une période équivalente d’éveil. L’excitabilité corticale était quantifiée en terme d’amplitude des potentiels évoqués moteurs (PEM) mesurés avant et après chaque session de iTBS. Les résultats montrent que la iTBS n’est pas parvenue à augmenter de manière robuste l’amplitude des PEMs lors de la première session de stimulation. Lors de la deuxième session de stimulation, la iTBS a produit des changements plastiques variables et ce peu importe si les sujets ont dormi ou pas. Les effets de la iTBS sur l’excitabilité corticale étaient marqués par une importante variabilité inter et intra-individuelle dont les possibles causes sont discutées. / In humans, various repetitive transcranial magnetic stimulation (rTMS) protocols can be used to modulate motor cortical plasticity. These techniques have shed light on the role of sleep in neural plasticity regulation. Recent work has demonstrated that when a night of sleep follows one session of rTMS over the hand motor cortex (M1), the capacity to induce subsequent plasticity by another rTMS session in M1 is enhanced. The present study investigated whether such metaplasticity could also benefit from a day nap. Fourteen healthy participants received two sessions of intermittent theta burst stimulation (iTBS) known for its excitatory effects on cortical excitability over M1 spaced by either a 90-minute nap or an equivalent amount of wake. Motor cortical excitability was measured in terms of amplitude of motor evoked potentials (MEP), which were assessed before iTBS and after the stimulation. Results show that the first iTBS session did not induce significant change in MEP amplitude. The second iTBS session induced variable plastic changes regardless of whether participants slept or stayed awake. The effects of iTBS on motor cortical excitability were highly variable within and between individuals. The possible causes of such variability are discussed.

Effet de la source du sélénium sur le statut du sélénium, de la GSH-Px et sur le système immunitaire des bovins de boucherie

Résumé L’objectif de cette étude était de déterminer les effets de la source de sélénium sur les concentrations de Se et de GSH-Px des vaches de boucherie (n =33) et leurs veaux et sur des paramètres immunitaires des veaux. Deux groupes de vaches ont reçu 3 mg/j/animal de Se organique ou inorganique dans le minéral. Le troisième groupe n'a pas été supplémenté en Se et leurs veaux ont été divisés en deux sous-groupes, l’un des deux a reçu une injection de sélénite de sodium (0,087 mg/Kg) à la naissance. Le Se et la GSH-Px ont été respectivement mesurés par HPLC-UV et par cinétique enzymatique. La phagocytose, la flambée respiratoire et le ratio CD4:CD8ont été évalués par des kits commerciaux et les IgG totales ont été mesurés par immunodiffusion radiale. La supplémentation de Se a augmenté significativement le Se sérique et colostral (P<0,02) et la GSH-Px(P≤0,04) pour les vaches et leurs veaux avec un effet significativement plus élevé pour le Se organique. Le Se du lait a augmenté de façon significative uniquement avec la source organique du Se (P≤0,0007). L’injection du Se chez les veaux a permis une augmentation significative mais temporaire (P<0,0001) du Se sérique. La supplémentation en Se n’a pas influencé les paramètres immunitaires mesurés (P>0,01, non significatif après correction de Bonferroni). Nous concluons que la supplémentation en Se améliore le niveau du Se colostral, lacté et sérique ainsi que la GSH-Px pour les vaches et leurs veaux sans effet sur les paramètres immunitaires mesurés des veaux. Mots clés: Sélénium, veaux de boucherie, phagocytose, flambée respiratoire, anticorps, ratio CD4:CD8, GSH-Px. / Abstract The aims of this study were to determine the effects of selenium (Se) supplementation sources (organic and inorganic) on Se and GSH-Px concentrations of beef cows (n=33) and their calves and on immune parameters of the calves. Two groups of cows were given daily 3 mg of either organic or inorganic Se in mineral supplement starting from 12 weeks before calving until weaning. The third group had no Se added into the diet and their calves were divided into two subgroups either injected or not with 0.087 mg/kg of sodium selenite after birth. Serum Se and whole blood GSH-Px were respectively measured by HPLC-UV and by kinetic-enzymatic technique. Calves immune parameters were evaluated using commercial kits for phagocytosis, respiratory burst and CD4:CD8 ratio and radial immunodiffusion for total IgG concentrations. In cows and calves, Se supplementation increased significantly serum and colostrum Se concentrations (P<.02) with significant higher effect for organic source. However, milk Se concentrations increased significantly only with the organic source (P≤.0007). Se supplementation increased GSH-Px concentrations in cows (P≤.04) and their calves (P≤.0004); organic source induced a higher effect than inorganic one in calves (P≤.0004). Se injection in calves allowed a temporary increase (P<.0001) of serum Se concentrations. No significant differences were noticed throughout the experiment for all of the immune parameters measured (P>.01, not significant after Bonferroni adjustment). Our results showed that Se supplementation improved colostrum, milk and serum Se and GSH-Px concentrations in cows and their calves without effect on the measured immune parameters in calves. Manufacture, modelling and characterisation of novel composite tubes

This thesis primarily focused on the development of novel composite tubes by braiding. The objective was to use hierarchical scale technique, i.e., micro, meso and macro scales, with the transfer of information from one scale to another to develop novel braided composite tubes. This research was conducted and reported in three journal papers. The aim of the first paper was to predict plane elastic properties for E-glass/epoxy braided composite structures at different braid orientations, by analytical and finite element techniques. The lenticular shape has been used to describe the geometry of the tow. Modified lenticular geometric model was developed to improve an existing geometric model, in terms of tow parameters, thereafter, plane elastic properties from Chamis micromechanical model for E-glass fibre and epoxy matrix without any knockdown effects were used as benchmark to develop predictive models, namely; Lekhnitskii's methodology and braided unit cell meso-scale finite element model to account for the effects of tow geometry, undulations/crimp, cross-over and braid orientations on the plane elastic properties of E-glass/epoxy composite. The results showed agreement in trend between the predictive models, Chamis micromechanical model, and a similar existing model. However, the plane elastic properties were knocked down in predictive models by 30% in the E11 direction and 32% in the E22 direction, when compared with Chamis micro-mechanical model for largest ±65° braid angle, among the braid angles, considered. The aim of the second paper was to manufacture E-glass/epoxy braided tubes at different braid orientations by vacuum bag infusion technique, conduct internal pressure tests, and determine the hoop and axial moduli of the infused tubes. Lekhnitskii's methodology was also used to develop plane elastic moduli by experiment using microscopy results, and by calculation. The experimental elastic moduli of the infused tubes and the experimental elastic moduli from Lekhnitskii's methodology were used to compare the predictive elastic moduli for E-glass/epoxy braided structures by Chamis micro-mechanical model, and the braided unit cell meso-scale finite element model. The two were from another paper. Results showed a perfect agreement in trend between the experimental results and the predictive results. However, the values of the experimental results were close but lower than the predicted results. Optical microscopy was performed on braided tube cross-section to evaluate the level of crimp or undulation. This was done by the determination of tow centreline crimp angle and aspect ratio. Results show that when compared with the predicted crimp, there was an agreement in trend, although the experimental results were lower than the predicted. Also, the knockdown factor was evaluated and used to quantify the reduction in experimental elastic moduli when compared with the predicted. Results showed that the absences of crimp in the Chamis model caused a tremendous difference between it, other predicted models and the experiment results. The elastic moduli of Chamis were by far higher than all others, including other predictive models. The purpose of the third paper was to manufacture E-glass/epoxy braided tube at ±31°, ±45°, ±55°, ±65° braid orientations using vacuum bagging and resin infusion technique, to design and manufacture a rig for tube internal pressures experiment, to determine the hoop and axial stress performances of the tubes by internal pressure experiment, to compare experimental results with laminate analysis predictions to evaluate the effect of crimp on the internal pressure performance of the braided tubes. To use E-glass braided tow meso-scale unit cell finite element model to predict the tow critical stresses, and the optimum braided tube architecture, using tube hoop and axial failure stresses or strains. The tubes were manufactured and subjected to internal pressure test (2:1), to failure. Failure mode was by weeping and bursting. Hoop stress was twice the axial stress. The highest value of hoop stress was at the ±65° braid angle, higher than the hoop stresses at the ±31°, ±45°, and ±55 ° braid angles by 50%, 39%, and 28% respectively. Hoop stress increased with increase in braid angle. The experimental results were validated by laminate analysis predictions by Chamis micro-mechanical model and Lekhnitskii's methodology, and the trend of the laminate analysis prediction matched that of the experimental results. However, the predicted values were higher than the experimental results by 21%, 14%, 11%, 10% for the ±31°, ±45°, ±55°, ±65° braid angles for the Chamis micro-mechanical model and 5%, 7%, 7%, 5% for the ±31°, ±45°, ±55°, ±65 braid angles respectively for the Lekhnitskii's model, showing the severe effect of crimp in the experimental tube, mostly when compared with Chamis micro-mechanical model. Braided tow unit cell finite element model prediction, showed that tow axial stresses increased with increase in braid angle, while the tow transverse stresses decreased with increase in braid angle. The predictions showed that the tow critical stresses and the tube optimum braided architecture lie between the ±65° and 90° braid angles. The tow critical stresses are the stresses at which the tow decreasing transverse stress and the tow increasing axial stress causes the tube to fail.
