Perfil oxidativo e bioquímico em pacientes que apresentam anemia falciforme ou traço falciforme / Oxidative and biochemistry profile in sickle cell trait subjects and sickle cell anaemia patientsManfredini, Vanusa January 2008 (has links)
Sickle cell anaemia (SCA) is an autossomal recessive disease and, among the hemoglobinopaties, is the most common of the known hereditary hematologic alterations in man. Its distribution is ample, enclosing all the continents. Sickle cell anaemia (SCA) was the first disease to be characterized on the molecular level. It is basically a red blood cell (RBC) disorder in which the gene encoding the human β-globin subunit presents a mutation with the resulting replacement of β6 glutamic acid (Glu) by valine (Val). This exchange of the amino acids that results in the HbS modifies the molecule structurally, and under determined conditions, the polymeration occurs, bringing serious consequences to the symptomatic individual. The reactive oxygen species (ROS) can cause deep injuries in erythrocytes, diminishing its period of useful life, specially in patients with sickle cell anaemia. Sickled erythrocytes are under constant oxidative stress and thus liberate products of degradation of the HbS, contributing for the progression of the disease. Because of the oxidative damage aggravates the pathophysiology of sickle cell patients. Using spectrophotometrically technique, catalase (CAT), glutathione peroxidase (GPx), superoxide dismutase (SOD), were determined the activities of antioxidant enzymes and total glutathione (GSH) quantified in the erythrocytes of the patients. We also determined oxidative damage of plasma proteins and in hemolysate using carbonyl assay at 360 nm. The levels of lipid peroxidation (MDA) and vitamin C were determined by high-performance liquid chromatography (HPLC). Finally, we quantified the levels of high sensitivity Creactive protein (hsCRP) using turbidimetry technique. The participants of the research (30 HbAA, 28 HbAS and 20 HbSS) were selected from Centro de Apoio ao Portador de Anemia Falciforme (CAPAF/RS) and/or registered in cadastre in Laboratório de Hematologia da Faculdade de Farmácia (UFRGS). Sickle cell patients had been identified by HPLC and confirmed by molecular study, using the polymerase chain reaction amplification (PCR). All individuals signed the term of free and clarified assent and were submitted to a nutricional questionnaire. Data were expressed as average ±SD and analyzed using ANOVA test of one way via with the post ad hoc test. Results show that sickle cell trait subjects (HbAS) had significantly high activity of CAT when compared to healthy controls. On the other hand, sickle cell patients had greater activity of GPx and SOD. The GSH levels was proportionally higher in the followed HbSS of the HbAS and HbAA. We also observe oxidative damage in plasma proteins, but not in the cellular hemolysate. HbSS have significantly higher oxidative damage in plasma proteins than other groups. A significant increasing of the production of MDA in the serum of the HbSS was observed as an indicative of the increasing of the auto-oxidation of the lipids under oxidative estress. Serum vitamin C levels in HbSS were significantly higher than healthy control. The hsCRP was presented significantly higher in HbSS than HbAA. These results reinforce the idea that patients with SCA are subjected to chronic oxidative stress, that contributes for the progression of this hemolytic anaemia. On the other hand, sickle cell trait subjects have higher antioxidant defenses that are able to reduce oxidative damage in biological macromolecules such as proteins and lipids.

Avaliação do estresse oxidativo em pacientes idosos com anemia ferropênica / Oxidative stress evaluation in old patients with iron deficiency anaemiaBaccin, Aline Coghetto January 2008 (has links)
Iron deficiency is the outcome of a long period of negative iron balance, culminating in exhaustion of the body’s iron stores. This is revealed when hemoglobin concentration declines to values below normal levels and anaemia is detected, due to a deficient synthesis of hemoglobin, with decrease in eritrocitary proliferation. Iron deficiency anaemia is considered the most important health problem in elderly and it is associated with various complications, including cardiovascular disease, cognitive dysfunction, increased risk of death and, mostly, it has a meaningful effect in the quality of life. Studies reveal that iron deficiency anaemia cause higher susceptibility to oxidant agents. Using spectrophotometric techniques, the activities of antioxidant enzymes, catalase (CAT), glutathione peroxidase (GPx), superoxide dismutase (SOD) and total glutathione (GSH) were quantified in the erythrocytes of the patients. We also determined oxidative damage of plasma proteins and in hemolysate using the carbonyl assay at 360 nm. The levels of lipid peroxidation (MDA) and vitamin C were determined by highperformance liquid chromatography (HPLC). Participants in this study were selected in Ambulatories of Basic Health Attention in the North of the State of Rio Grande do Sul. 17 individuals presented iron deficiency anaemia and primary hypertension and 18 only primary hypertension, because it was difficult to select patients without any associated deficiency. All individuals signed the term of free and clarified consent to participate in this study. Data were analyzed using t- Student Test. Results show that individuals with iron deficiency anaemia show significantly higher activity of SOD and CAT when compared with those from the control group, but they do not show difference in the activity of GPx or GSH. Oxidative damage was also observed in plasma proteins, but not in the cellular hemolysate. Patients with iron deficiency anaemia showed oxidative damage in plasma proteins significantly higher when compared with the control group. And they also showed a significant increase in the production of MDA in the serum as an indicative of the increased auto-oxidation of lipids under oxidative stress. Serum vitamin C levels did not show significant difference between the groups. These data reveal the involvement of the reactive oxygen species in carential anaemia. Therefore, we reinforce the need to diagnose and treat these patients early to reduce the magnitude of the effects of anaemia on the heath of the elderly.

This dissertation presents a study of a class of materials called Magneto-Rheological Elastomers (MRE) with main focus on its mechanical and magnetic response, where the properties of the iron carbonyl micro particles functionalize rubber silicone. Samples were prepared from the dispersion of iron carbonyl particles in silicone rubber, and that vulcanization of the material took place by means of a catalyst, and the type of shear tests were performed. We carried out experimental shear tests in which the samples were arranged in the form of "sandwich" formed by Brass / MRE / Brass / MRE / brass and attached to the universal test machine between permanent magnets, which examined the relationship tension strain samples with different concentrations of 18%, 33% and 48% by volume iron carbonyl with and without magnetic field. Considering the characterization of the MRE, they are also displayed results using the Infrared Spectroscopy technique of Fourier Transform aimed at building an understanding of the relationship between the properties of the samples by concentration and scanning electron microscopy technique in order to observe the characterization morphological particles dispersed in the silicone. The results point to the possibility of use of the MRE system as Dynamic Vibration Absorbers tunable since the results obtained are within 50% variation of the stiffness of the system with and without magnetic field.

Previous issue date: 2009-10-14 / Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPES / The intramolecular hydrogen bond occurs when the same molecules has both proton donor and proton acceptor groups in satisfactory configuration space for the formation of this interaction. It is important to note the changes in the structural, electronic and vibrational properties that occur due to the formation of this interaction. In the hydrogen bonding formation is an important phenomenon called charge transfer , where part of the electronic density of the proton acceptor species, Y, is transferred o the proton donor specie, HX. With respect to the vibrational spectrum are observed changes in the way of straightening of donor and acceptor proton species. Di-carbonyl compounds (C3H2O2R2) with their substituent groups (R=CH3, CN, H, NH2, OH and SH) were studied focusing on the energetic, structural, vibrational and electron density analysis. Initially the energy and structural analysis were carried out starting from the molecules optimized geometry. We also evaluated of the strength s hydrogen bonding and the length s intramolecular bond. The QTAIM study was performed to obtain the electron density s values and the electron density s Laplacian values and verify the existence of the bond critical point in the intramolecular hydrogen bond. From the harmonic vibrational spectra was possible to identify changes in the vibrational modes, related the intramolecular interaction s formation.

Age Related Macular Degeneration (AMD) and Genetic Stargardt’s disease share a common cytotoxic mechanism involving carbonyl and oxidative stress. Hence both stresses represent promising therapeutic targets. A lipophenol lead molecule has been previously highlighted. This molecule is composed by an alkylphloroglucinol moiety and a lipid part which is a polyunsaturated fatty acid, the docosahexaenoic acid (DHA, C22:6, ω3). In order to improve the antioxidant activity of this lipophenol, the total synthesis of two deuterated DHA on bis-allylic positions was performed. The cross-coupling between the deuterated DHA and the alkylphloroglucinol moiety led to the corresponding deuterated lipophenols. The impact of the deuterium to limit the cell toxicity and the lipid peroxidation was highlighted thanks to in vitro biological studies performed on retinal pigment epithelium cell line.

Avaliação ambiental de compostos orgânicos voláteis (VOC) provenientes da queima da cana-de-açúcar / Environmental assessment of volatile organic compounds (VOC) from sugar cane burninMarcelo Luiz Araujo Lopes 09 June 2010 (has links)
Biomass burning is an activity widely practiced in many tropical developing countries. This activity has implications at regional and global scales due to the emissions of significant quantities of trace gases to the atmosphere such as CO, non-methane hydrocarbons (NMHC) and particulate matter (PM). The sugar cane residues represent 11% of worldwide production of agricultural residues whose burning releases significant amounts of gases and particles that influence the atmospheric chemistry. In the last years, in Brazil, there has been an increase of the use of the ethanol as fuel and an increase of the sugar cane plantations in the State of São Paulo. Although the sugar cane burning prohibition, burning episodes are still very frequent, as 50% of the sugar cane plantations of São Paulo are burned in the the pre-harvest stage. The aim of this study was to evaluate VOC emissions of sugar cane burning. In order to estimate the environmental impact caused by the combustion process in urban regions near fires laboratory and field studies were conducted. The contribution of biogenic carbonyl compounds in the area of sugar cane was evaluated and results showed that the cane sugar leaves, in the most of the experiments, do not act as emission source. Laboratory experiments were carried out to determine incomplete combustion products in plume. Measurements of CO, CO2, and VOC were characterized by the emission ratio (ER) and emission factor (EF). Mixture ratio, in ppbv, of formaldehyde (175 - 309), acetaldehyde (2 - 71), acetone (1 - 36), acrolein (0,3 - 3), propionaldehyde (0,2 - 10), crotonaldehyde (3 -10), butyraldehyde (1 - 3), benzaldehyde (0,5 - 6), valeraldehyde-total (2 - 60) and hexaldehyde (4 - 136) were identified and quantified; and other VOCs, such as aromatic hydrocarbons, alkyl halides, nitrogen containing compounds and furans were only identified. The efficiency of the sugar cane combustion was evaluated (ΔCO/CO2 = 4.0±0.4%) in the laboratory and the predominant burning phase was the flaming phase. By comparing statistically filed measurements done before and during fire in the atmosphere of urban areas near fires, it was possible to distinguish two different groups of the measurements suggesting that the origin of the compounds is different. Direct emission sources and mainly photochemical reactions were responsible by the presence of formaldehyde and acetaldehyde in samples collected before fire (daytime measurements), whereas only direct emission sources were responsible by the presence of these compounds in samples collected during the fire (night measurements). Trajectories of air masses that reached the urban area and spots of burning recorded in the sampling days showed that the region studied was affected by fires.

Palladium catalysed asymmetric hydroxy- and alkoxycarbonylation of alkenesDurrani, Jamie T. January 2015 (has links)
Palladium catalysed asymmetric hydroxy- and alkoxycarbonylation reactions of alkenes have the potential to deliver valuable chiral carboxylic acid and ester building blocks from cheap feedstocks: alkenes, carbon monoxide and water (alcohols in the case of alkoxycarbonylation). Despite the attractive nature of these reactions, extensive research has so far been unable to produce effective catalysts which are capable of controlling both regio- and enantioselectivity. Building on exciting recent results involving the use of highly enantioselective palladium catalysts derived from Phanephos-type ligands, this research focuses on paracyclophane-diphosphines and their use in asymmetric hydroxy- and alkoxycarbonylation reactions. An investigation into reaction conditions analysed several factors, including solvents, CO-pressure, acidic additives and halide sources, to provide optimal activity and selectivities. Two novel electron-poor paracyclophane-diphosphines and their mono- and di-palladium complexes were synthesised and shown to provide exceptional levels of regioselectivity while maintaining high levels of asymmetric induction. These are the first such examples of hydroxy- or alkoxycarbonylation catalysts to facilitate simultaneous control over both regio- and enantioselectivity. The most effective catalyst was used to promote the reactions of a selection of aryl alkenes and was shown to be tolerant of several different functional groups. A selection of non-symmetric paracyclophane-diphosphine ligands and their palladium complexes were also synthesised and assessed for their performance in hydroxy- and alkoxycarbonylation. We also report the use of Phanephos-type ligands to promote the highly enantioselective hydroxycarbonylation of N-(p-toluenesulfonyl)-3-pyrroline to deliver a chiral proline derivative in high ee.

Fenton Pre-treatment of a Birch Kraft Pulp for MFC preparationHellström, Pia January 2015 (has links)
The potential to use acidic hydrogen peroxide in the presence of ferrous ions (Fenton’s reagent) as a pre-treatment when producing microfibrillar cellulose (MFC) from a fully bleached birch (Betula verucosa) kraft pulp was investigated and the properties of the produced MFC was compared to the properties of a MFC produced with enzymatic pre-treatment with a monocomponent endoglucanase (FiberCare® R). The mechanical treatment to MFC was performed in a laboratory colloid mill or in a pilot high-pressure homogeniser and the pre-treated pulps as well as the produced MFCs were chemically and morphologically characterised. Additionally, the MFCs produced in the colloid mill were evaluated as strength enhancers in test sheets representing the middle ply of paperboard. From the chemical characterisation, it was concluded that the Fenton pre-treatment caused a decrease in the degree of polymerisation (DP) and an increase in both carboxyl- and carbonyl groups. The increase in carbonyl groups could not be explained by the formation of new reducing end groups due to depolymerisation which indicates that carbonyl groups are introduced along the cellulose chain. The enzymatic pre-treatment as performed in this study caused less impact on the cellulosic material, i.e. resulted in a pulp with a higher DP and a much lower amount of carbonyl- and carboxylic groups compared with the Fenton pre-treated pulps. In the subsequent mechanical treatment in a colloid mill, the Fenton pre-treated pulps were easier to process mechanically i.e. reached a higher specific surface area and a higher surface charge at a given mechanical treatment time compared to enzymatic pre-treated pulps and pulps not subjected to any pre-treatment. These findings were confirmed when MFCs were produced by homogenisation at high pressure in multiple passes; the birch kraft pulp was either pre-treated with Fenton’s reagent or the combined mechanic and enzymatic pre-treatment methodology used at the Centre Technique du Papier (CTP, France). By size fractionation, rheological measurements and scanning electron microscopy, it was revealed that Fenton pre-treatment resulted in MFC suspension containing a significantly higher proportion of small sized material (< 0.2 mm). When the MFCs were evaluated as strength enhancers in test sheets produced from a furnish consisting of a spruce (Picea abies) chemithermomechanical pulp, MFC and a retention system containing cationic starch and an anionic silica sol, Fenton pre-treated MFCs increased the strength properties more than the enzymatic pre-treated MFCs. Addition of 5 wt% Fenton pre-treated MFC resulted in an increase in z-directional strength of about 50%, an increase in tensile stiffness index of about 25% and an increase in tensile index of 35% compared to test sheets prepared without MFC addition. / The potential to use acidic hydrogen peroxide in the presence of ferrous ions (Fenton’s reagent) as a pre-treatment when producing microfibrillar cellulose (MFC) from a bleached birch kraft pulp was investigated and the properties of the produced MFC was compared to the properties of a MFC produced with enzymatic pre-treatment. Kinetics Studies of Substituted Tungsten Carbonyl ComplexesWang, I-Hsiung, 1950- 08 1900 (has links)
Thermal reactions and flash photolysis are used to study the olefin bond-migration promoted by tungsten carbonyls. Substitution of piperidine (pip) by 2- allylphenyldiphenylphosphine (adpp) in the cis-(pip)(η^1- adpp)W(CO)-4 complex was investigated, and no olefin bond-migration was observed. This suggests that a vacant coordinated site adjacent to the coordinated olefin is an essential requirement for olefin bond rearrangement.
The rates of olefin attack on the photogenerated coordinatively unsaturated species, cis-[(CB)(η^1-ol- P)W(CO)-4] (CB = chlorobenzene, p-ol = Ph-2P(CH-2)-3CH=CH-2; n = 1-4) were measured. Kinetics data obtained both in pure CB
and in CB/cyclohexane mixtures support a dissociative mechanism in which the W-CB bond is broken in the transition state. In contrast to results observed in studies of other related systems, no olefin bond-migration is noted. This observation is attributed to P-W coordination at all stages of the reaction, which precludes formation of a reactive intermediate containing a vacant coordination site adjacent to a P-ol bond.
