Le cancer constitue l’une des principales causes de mortalité, plus particulièrement dans l’ensemble des pays développés, et représente, de ce fait aujourd’hui un problème de santé publique majeur.Depuis quelques années, les alcaloïdes marins représentent une source d’inspiration importante pour les chimistes en vue d’obtenir de nouveaux médicaments anticancéreux.Dans cette optique, des recherches effectuées au sein de notre laboratoire ont fait état de la synthèse d’analogues de ces alcaloïdes possédant une structure tris aromatique.Nous avons développé une approche synthétique originale de ces composés disposant d’un hétérocycle central sur lequel sont greffés deux noyaux phényles ou indoles substitués avec des groupes hydroxy ou hydrophiles. Ils sont reliés entre eux par une liaison carbone-carbone ou carbone-amine ce qui a généré une bibliothèque de près de 150 molécules originales.Différentes évaluations pharmacologiques ont été réalisées. L’inhibition des kinases (DYRK1A, CDK5, GSK3) par nos produits et l’évaluation de leur cytotoxicité sur diverses lignées cellulaires cancéreuses humaines ont été réalisées.L’obtention de ces composés a été également l’occasion de nous intéresser à la synthèse et la réactivité du 2-indolyltrifluoroborate de potassium dans les réactions de Suzuki-Miyaura. Un travail méthodologique a été réalisé dans ce sens. / Cancer, one of the leading causes of death, particularly in the developed countries, represents today a major public health problem.Over the last few years, marine alkaloids represent a source of inspiration for chemists in order to obtain new anticancer drogs.For this purpose, as a part of our laboratory researches, analogues of marine alkaloids were synthesized possessing a tris-aromatic structure.We developed an original approach to synthesize these compounds formed by a central heterocycle core on wich is graft two arylic or indoles patterns with hydroxylic or hydrophilic side chains. They are related by a carbon-carbon or carbon-amine bond that generated a library of 150 original molecules.Various pharmacological analyses were carried out. The inhibition of kinases (DYRK1A, CDK5, GSK3) by our products and the evaluation of their cytotoxicity on various human cancer cell lines have been performed.Obtaining these compounds was also an opportunity to be interested in the synthesis and reactivity of potassium 2-indolyl trifluoroborate towards Suzuki-Miyaura reaction. A new methodology was developed.

Chapter 1: Metal carbenoids in organic synthesis The chapter describes the phenomena of metal carbenoid insertion reactions in two parts: Part A, and Part B. The study of N-tosylhydrazones as diazo precursor was commenced by Jose Barluenga in 2007,1 which demonstrated an in-situ generation of diazo species and trapping of that with low valent palladium catalyst (Scheme 1). Later, this palladium-carbenoid assumption was supported by few reports. Some of these discoveries were by D. F. Taber in 1986 followed by van Vranken in 1999 & 2001.2 These studies of palladium carbenes were supplemented by several groups in subsequent years. The consequent developments with N-tosylhydrazones as diazo source were very fruitful and produced exceptional chemical transformations in recent years. Modes of reactivity of a metal-carbene Scheme 1 Cascade carbene migratory insertion process Part A: Ligand-free coupling of tosylhydrazones with aryl & heteroaryl halides In this part, Palladium catalysed cross-coupling reaction of hydrazones with aryl halides in absence of an external ligand is reported. The versatility of this coupling reaction has been demonstrated by showcasing the selectivity of coupling reaction in presence of hydroxyl and amine functional groups. This method allows synthesizing a variety of heterocyclic compounds, which are otherwise difficult to access from traditional methods. Application of the present methodology is validated in tandem reaction of ketones to the corresponding substituted olefins in a single pot experiment. Few examples are illustrated below in Scheme 2.3 Scheme 2: Scope of aryl halide coupling with tosylhydrazones Part B: Pd-catalysed Synthesis of Highly Branched Dienes The regioselective formation of highly branched dienes is a challenging task. Design and exploration of alternative working models to achieve such a regioselectivity to accomplish highly branched dienes is considered to be a historical advancement of Heck reaction to construct branched dienes. On the basis of the utility of carbene transfer reactions, in the reaction of hydrazones with Pd(II) under oxidative conditions, we envisioned obtaining a Pd-bis-carbene complex with α-hydrogens, which can lead to branched dienes. Herein, we report a novel Pd catalyzed selective coupling reaction of hydrazones in presence of tert-BuOLi and benzoquinone oxidant to form corresponding branched dienes (Scheme 3).4 The utility of the Pd catalyst for cross-coupling reactions for synthesizing branched conjugated dienes are rare. The reaction is very versatile and compatible with a variety of functional groups and is useful in synthesizing heterocyclic molecules. This commenced with the Myers’s report in 2004,5 which simplifies the Barton vinyl halide preparation with a remarkable revision on synthesis of alkyl-silyl-hydrazones and its applications. Improved methods of using tosylhydrazones were demonstrated by Aggarwal in successive years. Cycloadditions were implemented by Douglass F. Taber. 6 This study was enriched in a quite fascinating way by several groups such as Jose Barluenga, with many reductive coupling reactions and 1, 3-dipolar reactions. Thomson, in a very interesting report shows the traceless petasis reaction with hydrazones and also worked in many other prospects such as three component reactions and the acid catalysed [3+3] sigmatropic reactions of hydrazones. 7 Wang has also impressed with very attractive transformations in the past decade. 8 7) Thomson, R. Scheme 4: Trapping diazo species in intermolecular fashion Part A: Synthesis of vinyl halides Trapping diazo species in an intermolecular fashion by attack of two independent ions (a cation followed by an anion) in tandem at the carbene center is unprecedented. As part of our efforts on the utility of tosylhydrazones, herein we report a novel approach of using ambiphilic diazo species to perform a tandem attack of a nucleophile followed by an electrophile in an intermolecular fashion for synthesizing various types of vinyl halides. A few representative examples are shown in Scheme 5.9 Scheme5: Synthesis if vinyl halides Part B: Synthesis of vinyl sulfones Vinyl sulfones are potential synthetic targets due to their presence in biologically and pharmaceutically important molecules ranging from small natural metabolites to proteins, and have found widespread applications in biological research as covalent protease inhibitors. Vinyl sulfones represent one of the important sulfur containing functional groups in organic chemistry, which are generally synthesized through elimination reactions, oxidation of vinyl sulfides or witting reactions using multistep sequence. Following this technique, we were able to synthesize a variety of vinyl sulfones with rich mechanistic features in a single step. A few such examples are documented in Scheme 6.9 Scheme 6: synthesis of vinyl sulfones Part C: Synthesis of alkynes The functional group conversion to achieve alkyne frameworks are generally a difficult transformation. There are very few limited and tedious processes are available in literature, mainly containing multi-step procedures. Additionally these reactions are require harsh conditions. Considering all these factors, there is a need for developing methods to synthesize alkynes from common functional groups under mild reactions conditions. In a similar way, to introduce different halogens at the same carbon, we expected the eliminations of the leaving groups in tandem formed alkynes. After extensive screening studies, it was pleasing to find that the reaction of tosylhydrazones with NCS−BTEAC, NBS−TBAB, or NIS−TBAI combination in presence of K2CO3 in dioxane as solvent at 110 °C can furnish corresponding acetylene derivatives in good yields. Few examples are shown in Scheme 7.9 Scheme 7: Trapping diazo species in intermolecular fashion Chapter 3: Pd catalysed hydroboration This chapter shows a hydroboration study of terminal alkynes in a highly regioselective manner (Scheme 8). Organoboron derivatives have become essential intermediates in organic and medicinal chemistry. Scheme 8: possibility of site selectivity in hydroboration Part A: Pd-catalysed regioselective borylation of alkynes: A ligand controlled synthesis of α and β – vinyl boronates The metal catalyzed borylations of alkynes proceeds in a two-step process. Initially M-Bpin species undergo an addition onto the alkynes to generate organometallic species followed by quenching of the organometallic species with electrophiles. The addition M-Bpin species is regioselective governed by the steric and electronics factors of both metal complex as well as alkyne substituents. In this direction, a palladium catalysed α-selective borylation was achieved for terminal alkynes. A broad range of substrates were successfully borylated under optimized reaction conditions with very high selectivity. Interestingly, the selectivity was reversed to terminal site by using a NHC ligand. A few examples are shown in Scheme 9.11 Scheme 9: α & β-vinyl boronates Chapter 4: Pd/borane unit: Behavior towards isomerization vs reduction of alkenes This study presents a unique behaviour of palladium-boronate unit responsible for olefin chain walking and olefin reduction reactions (Scheme 10). The catalytic system stands efficient against both functionalized and unfunctionalized olefin isomerization as well as reductions. This study has been presented in two parts. Scheme 10: isomerization vs reduction Part A: Pd/ boronates or borane unit as efficient catalytic systems for olefin chain walk This study presents the behaviour of palladium-boronate unit responsible for olefin chain walking. The catalytic system is efficient for both functionalized and unfunctionalized olefin isomerizations (Scheme 11). Cycloisomerization of transient conjugated alkenes to synthesize heterocycles are prominent applications of this technique. The system describes a concept of olefin activation by coordination with Pd-borane complex, this complex assists in a facile [1,3]-hydride shift. This technique allows us to facilitate an isomerization in functionalized as well as unfunctionalized olefinic systems. Considering the substrates scope, the catalytic cycle tolerates various sensitive functional groups and shows good selectivity. In the following Scheme 11 few examples are depicted.12 12) Palladium/boron catalytic unit for olefin chain-walk (manuscript under preparation). Scheme 11: chain-walking of olefins. Part B: Palladium catalysed boronate promoted alkene reduction in water In this work, water has been employed as a source of hydrogen. The reduction of alkenes was achieved using Pd catalyst in presence of bis(pinacolato)diboron and H2O. In this aspect, the utility of water as hydrogen equivalent is the pertinent as well as beneficial with many advantages. Copper-Catalyzed Novel Oxidative Transformations : Construction of Carbon-Hetero Bonds

The thesis entitled “Copper-Catalyzed Novel Oxidative Transformations: Construction of Carbon-Hetero Bonds” is divided into two main sections. Section A deals with the utility of azide as a nitrogen source for C-N bond formation, which is further divided into 4 chapters, and section B presents decarboxylative radical coupling reaction for C-heteroatom bond formation which is further divided in to two chapters. Section A Chapter 1 describes an approach for the direct synthesis of nitrile from the corresponding alcohols using azide as a nitrogen source. Nitrile functionality is a versatile and ubiquitous which occurs in a variety of natural products. Nitrile functionality can be easily transformed into a variety of functional groups and products such as aldehydes, ketones, acids, amines, amides and nitrogen-containing heterocycles, such as tetrazoles and oxazoles. In this chapter a successful attempt for developing a novel methodology to oxidize benzylic and cinnamyl alcohols to their corresponding nitriles in excellent yields has been described. This strategy uses DDQ as an oxidant and TMSN3 as a source of nitrogen in the presence of a catalytic amount of Cu(ClO4)2·6H2O. A few representative examples are highlighted in Scheme 1.1 Scheme 1. Oxidative conversion of alcohols to nitriles Second chapter represents a protocol for the synthesis of 1,5-disubstituted tetrazoles from the corresponding secondary alcohols. Among heterocyles, tetrazole and its derivatives are important class of nitrogen containing molecules. Due to their well-known biological activities as well as vast applications in pharmaceuticals and material science, they are potential targets for synthetic organic chemists. Therefore, a simple and user-friendly method for the synthesis of tetrazole is desirable. In this chapter, a mild and convenient method to synthesize 1,5-disubstituted tetrazoles using easily accessible secondary alcohols by employing TMSN3 as a nitrogen source is developed. This reaction is performed in the presence of a catalytic amount of Cu(ClO4)2·6H2O using DDQ as an oxidant under ambient conditions (Scheme 2).2 Scheme 2. Oxidative conversion of secondary alcohols to tetrazoles Third chapter presents a method for synthesizing amides from their corresponding secondary alcohols. Amide functionality is a crucial backbone in peptide chemistry, it also serve as an important precursor or intermediate for variety of organic transformations. In this contention, a mild and convenient method to synthesize amides using easily accessible secondary alcohols by employing TMSN3 as a nitrogen source is developed. This reaction is performed in the presence of a catalytic amount of Cu(ClO4)2·6H2O using DDQ as an oxidant under ambient conditions (Scheme 3).3 Scheme 3. Oxidative conversion of secondary alcohols to amides Additionally, the application of this methodology has also been revealed for the synthesis azides directly from their alcohols. Some of the representative examples are shown in the Scheme 4.3 Scheme 4. Direct conversion of alcohols to their azides. Fourth chapter describes highly chemoselective Schmidt reaction. The classical Schmidt reaction involves the formation of new carbon-nitrogen bonds in a reaction of a carbon-centred electrophile with hydrazoic acid followed by loss of nitrogen, which usually occurs via a rearrangement. It is well known that under the Schmidt reaction conditions, ketones and carboxylic acids are converted into their corresponding amides and amines respectively, whereas aldehydes furnish a mixture of formanilides and nitriles. In this chapter, Schmidt reaction of aldehydes to obtain their nitriles without formation of the corresponding formanilide is presented (Scheme 5).4 It was also observed that aromatic ketones and acids functionalities were intact under the reaction condition, unlike the conventional Schmidt reaction. Highly chemoselective Schmidt reaction Section B It is divided into two chapters, describes a copper catalyzed decarboxylative radical coupling for the synthesis of vinyl sulfones and nitroolefins (Scheme 6). Scheme 6. General strategy for the second part First chapter narrates a strategy for synthesizing nitroolefins from the α,β-unsaturated carboxylic acids. Nitroolefins represent a unique class of nitro compounds, which have multifaceted utility in organic synthesis. They possess antibacterial, rodent-repelling, and antitumor activities. They serve as important intermediates in organic synthesis. Nitroolefins also react with a variety of nucleophiles, and their electron-deficient character renders them as a powerful dienophiles in Diels-Alder reactions. In our attempt to use the decarboxylative strategy, this chapter describes a method for the nitrodecarboxylation of substituted cinnamic acid derivatives to their corresponding nitroolefins. This nitrodecarboxylation reaction is performed using catalytic amount of CuCl in the presence of air using TBN as a nitrating source (Scheme 7).5 Besides, the reaction provides a useful method for the synthesis of β,β-disubstituted nitroolefin derivatives which are generally difficult to access from other conventional methods. Scheme 7. Decarboxylative nitration Second chapter presents a new protocol for the synthesis of vinyl sulfones from the α,β-unsaturated carboxylic acid. Vinyl sulfones are versatile building blocks, which find their utility as Michael acceptors and used in cycloaddition reactions. This functional group has also been shown to potently inhibit a variety of enzymatic processes, and thus provides unique properties for drug design and medicinal chemistry. Vinyl sulfones are prominent in medicinal chemistry owing to their wide presence in pharmaceutically active molecules, such as enzyme inhibitors and biological activity. In this chapter, we report a method for the construction of C-S bonds via ligand promoted decarboxylative radical sulfonylation of ,-unsaturated carboxylic acids to synthesize vinyl sulfones using Cu catalysis (Scheme 8).6 This is the first report for this particular conversion. Le paludisme et la leishmaniose sont les deux plus importantes infections parasitaires au monde, en termes de mortalité. La recherche de nouvelles molécules actives contre les protozoaires responsables de ces « maladies tropicales négligées », Plasmodium sp et Leishmania sp, est un enjeu majeur de santé publique. Après une première partie dressant un état des lieux des connaissances disponibles en matière de chimiothérapie antiplasmodiale, une seconde partie s’est intéressée à l’étude des propriétés anti-infectieuses du noyau 2-trichlorométhyl-quinazoline, en introduisant en position 4 des motifs alcynyles par couplage de Sonogashira, optimisés par LC/MS. Une troisième partie a porté sur la pharmacomodulation en positions 2 et 4 du même noyau, notamment par réactions de SNAr. Une quatrième partie a consisté à rechercher le mécanisme d'action des meilleures quinazolines antiplasmodiales, via une approche de chromatographie d'affinité sur inhibiteur immobilisé. La fonctionnalisation multi-étapes des molécules les plus puissantes, par un bras espaceur, a été suivie de leur ancrage sur divers supports solides, pour constituer des matrices biocompatibles spécifiques. L’une d’entre-elles a permis la mise en évidence de 2 cibles plasmodiales protéiques originales : la GTPase Pfrab6 et la pyruvate-kinase PfpyrK1. Enfin, une cinquième partie relate la pharmacomodulation antileishmanienne du noyau 8-nitroquinoléin-2(1H)-one. Les travaux se sont intéressés à l'étude de la substitution de la position 4 de ce noyau par des réactions de SNAr, ainsi que des réactions de couplages pallado-catalysés optimisées à l’aide de la technologie micro-ondes. / Malaria and leishmaniasis are the two most important parasitic infections worldwide, in terms of mortality. Thus, the research for new molecules targeting the protozoa parasites responsible for these “neglected tropical diseases”, Plasmodium sp and Leishmania sp, constitute a major challenge in public health. Our work focused first on a current state of knowledge about antiplasmodial chemotherapy. In a view to develop the study of the anti-infective properties of the 2-trichloromethylquinazoline scaffold, a second part presented antiplasmodial pharmacomodulation at position 4 using Sonogashira cross-coupling reaction, optimized with the LC/MS technology. A third part concerned other pharmacomodulation reactions, especially at positions 2 and 4, using especially SNAR reactions. A fourth part consisted in the research of the mechanism of action of the best antiplasmodial quinazolines by using the affinity chromatography on immobilized inhibitor approach. The multi-step functionalization of the most potent derivatives by a spacer side chain was followed by their anchoring onto various solid supports, so as to generate different biocompatible specific matrices. One of them, put in contact with a parasitic lysate, allowed the identification of two original plasmodial targets: the GTPase Pfrab6 and the pyruvate-kinase PfpyrK1. Finally, a fifth part presented the antileishmanial pharmacomodulation of the 8-nitroquinolin-2(1H)-one scaffold, especially at position 4 of the quinoline ring, involving SNAr reactions (with amines, phenols or thiophenols) or pallado-catalyzed coupling reactions (in particular Suzuki-Miyaura), some of them being optimized under microwave irradiation.

Growth optimization and characterization of regular arrays of GaAs/AIGaAs core/shell nanowires for tandem solar cells on silicon / Optimisation de la croissance et caractérisation de réseaux ordonnés de nanofils cœur/coquille GaAs/AlGaAs pour cellules solaires tandem sur silicium

L'objectif de cette thèse est de réaliser l'intégration monolithique de nanofils (NFs) à base de l’alliage Al0.2Ga0.8As sur des substrats de Si par épitaxie par jets moléculaires via la méthode vapeur-liquide-solide (VLS) auto-assistée et de développer une cellule solaire tandem (TSC) à base de ces NFs.Pour atteindre cet objectif, nous avons tout d'abord étudié la croissance de NFs GaAs, étape clé pour le développement des NFs p-GaAs/p.i.n-Al0.2Ga 0.8As coeur/coquille, qui devraient constituer la cellule supérieure de la TSC. Nous avons montré, en particulier, l'influence de l'angle d'incidence du flux de Ga sur la cinétique de croissance des NFs GaAs. Un modèle théorique et des simulations numériques ont été réalisées pour expliquer ces résultats expérimentaux.Nous avons ensuite utilisé le savoir-faire acquis pour faire croître des NFs p-GaAs/p.i.n-Al0,2Ga0,8As coeur/coquille sur des substrats de Si prêts pour l'emploi. Les caractérisations EBIC réalisées sur ces NFs ont montré qu'ils sont des candidats potentiels pour la réalisation d’une cellule photovoltaïque. Nous avons ensuite fait croître ces NFs sur des substrats de Si patternés afin d'obtenir des réseaux réguliers de ces NFs. Nous avons développé un protocole, basé sur un pré-traitement thermique, qui permet d'obtenir des rendements élevés de NFs verticaux (80-90 %) sur une surface patternée de 0,9 x 0,9 mm2.Enfin, nous avons consacré une partie de notre travail à définir le procédé de fabrication optimal pour la TSC, en concentrant notre attention sur le développement de la jonction tunnel de la TSC, l'encapsulation des NFs et le contact électrique supérieur du réseau de NFs. / The objective of this thesis is to achieve monolithical integration of Al0.2Ga0.8As-based nanowires (NWs) on Si substrates by molecular beam epitaxy via the self-assisted vapour-liquid-solid (VLS) method and develop a NWs-based tandem solar cell (TSC).In order to fulfil this purpose, we firstly focused our attention on the growth of GaAs NWs this being a key-step for the development of p-GaAs/p.i.n-Al0.2Ga0.8As core/shell NWs, which are expected to constitute the top cell of the TSC. We have shown, in particular, the influence of the incidence angle of the Ga flux on the GaAs NW growth kinetic. A theoretical model and numerical simulations were performed to explain these experimental results.Subsequently, we employed the skills acquired to grow p-GaAs/p.i.n-Al0.2Ga0.8As core/shell NWs on epi-ready Si substrates. EBIC characterizations performed on these NWs have shown that they are potential building blocks for a photovoltaic cell. We then committed to growing them on patterned Si substrates so as to obtain regular arrays of NWs. We have developed a protocol, based on a thermal pre-treatment, which allows obtaining high vertical yields of such NWs (80-90 %) on patterned Si substrates (on a surface of 0.9 x 0.9 mm2).Finally, we dedicated part of our work to define the optimal fabrication process for the TSC, focusing our attention to the development of the TSC tunnel junction, the NW encapsulation and the top contacting of the NWs.

Synthèse et fonctionnalisation des furo[3,2-b]- et [2,3-c]pyridines par voie organométallique / Synthesis and functionalisation of furo [3,2-b]- et [2,3-c]pyridines by using an organometallic way

Le travail décrit dans ce mémoire concerne l'étude de la métallation régiosélective de deux bicycles fusionnés complexes : les furo[3,2-b]- et [2,3-c]pyridines. L'influence de la nature du système basique sur le cours de la réaction et sur la régiosélectivité de la lithiation a été étudiée avec différentes bases : n-BuLi, LTMP, LDA et [n-BuLi/LiDMAE]. D'un point de vue fondamental, cette étude nous a permis d'établir quelques règles pour la fonctionnalisation des hétérocycles complexes, ce qui nous a conduit à l'obtention efficace et rapide d'une vaste chimiothèque de furo[3,2-b]- et [2,3-c]pyridines polyfonctionnalisés et refonctionnalisables. Quelques composés préparés ont ainsi été mis en jeu dans des réactions de couplage catalysées par les métaux de transition (Pd, Ni). Les difficultés rencontrées au cours de ce travail nous ont permises de mettre en évidence une séquence de double fonctionnalisation des hétérocycles ?-déficients par une séquence "one-pot" combinant le piégeage d'un intermédiaire lithié par un électrophile et l'addition nucléophile d'une espèce réactive générée in-situ. Le développement de cette séquence nous a conduits à l'obtention directe de pyrazines, pyridines et furo[3,2-b]pyridines disubstituées / The work described in this PhD thesis concerns a regioselective metalation study of two fused heterocycles : the furo[3,2-b]- et [2,3-c]pyridines. The influence of the lithiated agent on the reaction course and on the selectivity of the lithiation has been studied with several bases : n-BuLi, LTMP, LDA and [n-BuLi/LiDMAE]. From a fundamental point of view, this study allowed us to establish some rules concerning the functionalisation of fused heterocycles, and then, a chemical library of polyfunctionalised furo[3,2-b]- and [2,3-c]pyridines has been designed. Some of the compounds obtained in this way were engaged in metallo-catalysed coupling reactions. The difficulties we met during this work allowed us to discover a double functionalisation sequence of ?-deficient heterocycles via a cascade process combining the trapping of lithiated intermediates and the nucleophilic addition of some in-situ released species. The development of this sequence afforded direct access to difunctionalised pyrazines, pyridines and furo[3,2-b]pyridines

In der heutigen Wirkstoffforschung stellen vor allem multiresistente Krankheitserreger eine große Herausforderung an die Wissenschaft dar. Noch sterben z.B. an der Tuberkulose jährlich etwa 1.7 Millionen Menschen, davon 69000 allein in Europa (WHO-Angaben, 2004). Die Tuberkulose verursacht neben AIDS die meisten Todesfälle unter den Infektionskrankheiten. Ein großes Problem sind die zahlreichen Neuerkrankungen, die mit herkömmlichen Mitteln nicht mehr behandelt werden können, sowie zunehmende Medikamentenallergien. Somit ist ein wichtiges Gebiet der Wirkstoff-forschung, neben der Weiterentwicklung bekannter Medikamente, deren Neuentwicklung. Dabei leistet die Natur eine unentbehrliche Orientierungshilfe. So konnte aus asiatischen Medizinalpflanzen in letzter Zeit eine Reihe von Carbazolalkaloiden isoliert werden, die zweifelsfrei gegen das HI-Virus oder das Mycobakterium tuberculose, dem Erreger der Tuberkulose, aktiv sind.[1] Ziel der vorliegenden Arbeit war die regioselektive Synthese oxygenierter Carbazole. Dabei wurde das Konzept der eisenvermittelten Carbazolsynthese angewendet und darüber hinaus als Alternative die palladiumvermittelte Carbazolsynthese weiterentwickelt. Die für den Palladiumweg benötigten N,N-Diarylamine konnten über die BUCHWALD-HARTWIG-Aminierung, die Eisensalzkomplexe über eine katalytische Komplexierung ausgehend von Cyclohexadienen mit Pentacarbonyleisen hergestellt werden. Beide Methoden wurden gegenübergestellt und besonders hinsichtlich ihrer Ausbeuten und Syntheseeffizienz verglichen.

Les imidazo[1,2-b]pyrazoles tout comme les imidazo[1,2-a]imidazoles sont des entités présentant diverses applications intéressantes notamment dans le domaine pharmacologique. Cependant, malgré ce potentiel, ces structures hétérobicycliques ont été, jusqu’à ce jour, relativement peu étudiées tant au niveau de leur préparation que de leur fonctionnalisation. De ce fait, ces travaux de thèse ont pour objet la mise au point de nouvelles voies d’accès à ces systèmes bicycliques et ce, au départ de substrats facilement accessibles. Des stratégies de fonctionnalisation de ces charpentes moléculaires ont ensuite été développées dans le but de concevoir des librairies diversifiées de ce type de composés, librairies destinées à être évaluées biologiquement. Les premiers résultats d’évaluation sur des lignées cancéreuses de dérivés imidazo[1,2-b]pyrazoliques sont également présentés. / Imidazo[1,2-b]pyrazoles and imidazo[1,2-a]imidazoles are entities with some interesting applications in pharmacology. However, despite this potential, few methods of preparation and direct functionalisation of the heterocyclic moiety have been described. In this context, the overall goal of our research is to develop new routes to these bicyclic systems from readily available starting materials. Strategies of functionalisation of the heterocyclic moiety were then explored in order to design diversified libraries for the evaluation of potential biological activities. Herein, the results of the tests of imidazo[1,2-b]pyrazole series against various cancer lines are reported.
