Representation of asynchronous communication protocols in Scala and Akka

This thesis work investigates how to represent protocols for asynchronous communication in the Scala programming language and the Akka actor framework, to be run on Java Virtual Machine (JVM). Further restrictions from the problem domain - the coexistence of multiple protocol instances sharing the same Java thread - imply that neither an asynchronous call waiting for response nor anything else can block the underlying Java threads. A common way to represent asynchronous communication protocols is to use state machines. This thesis seeks a way to shrink the size of and to reduce the complexity of the protocol implementations by representing sequences of asynchronous communication calls (i.e. sequences of sent and received messages) as a type of procedure. The idea is find a way to make the procedures that contain asynchronous calls look like synchronous communication procedures by hiding the asynchronous details. In other words, the resulting procedure code should show what to do and not so much focus on how to overcome the impediment of the asynchronous calls. With the help of an asynchronous communication protocol toy example, this report shows how such an protocol can be implemented with a combination of a state machine and a procedure representation in Scala and Akka. The procedure representation hides away the asynchronous details by using the Scala capability to use CPS-transformed delimited continuations. As a sub-problem, this thesis also shows how to safely schedule asynchronous communication timeouts with help of Scala and Akka within the restrictions of the thesis problem domain.

Syftet med denna studie är att undersöka huruvida ett avgränsat tillvägagångssätt med få, tydliga regler att förhålla sig till kan främja eller hämma min improvisationsförmåga inom jazzgenren bebop och hur det påverkar min lärandeprocess. För att kunna svara på dessa frågor så har videodokumentation tillsammans med loggbok använts som datainsamlingsmetod. Studien utgår från ett sociokulturellt perspektiv med fokus på mästarlära. I kapitlen som följer så kommer relevant litteratur och forskningsartiklar att presenteras tillsammans med metod och resultat. I resultatkapitlet presenterar jag min process och drar slutsatsen att ett avgränsat tillvägagångssätt är till min fördel och främjar min lärandeprocess inom improvisation och bebop. Detta resultat kommer avslutningsvis att diskuteras i det sista kapitlet i relation till den litteratur och de forskningsartiklar som jag har använt mig utav. / The purpose of this study is to investigate whether a delimited approach with few, distinct rules to adhere to can support or obstruct my ability to improvise within the jazz genre of bebop and how it affects my learning process. In order to be able to answer these questions, video documentation together with a logbook has been used as a data collection method. The study is based on a socio-cultural perspective with a focus on master – apprentice teaching. In the chapters that follow, relevant literature and research articles will be presented together with method and results. In the results chapter, I present my process and conclude that a limited approach is to my advantage and promotes my learning process in improvisation and bebop. This result will finally be discussed in the last chapter in relation to the literature and research articles that I have used.

Insurance contract corresponds to the social-economical development, however it does not translate one legal transaction only as it can mean an infinity of contractual relations, that involve many different people, in innumerable branches of the insurance market. It occurs that the insurance contract can be translated by contract of civil liability insurance, contract of safety from contractual guarantees, contract of residential insurance, contract of health insurance, contract of enterprise insurance, contract of bail insurance, contract of educational insurance, and the contract of vehicule insurance, the target of this study. In this context, the risk should be deeply analyzed, it will have to be declared by the insured, being certain that it is consolidated through the questionnaire of risk evaluation, which should be part of the the insurance proposal. The questionnaire of risk evaluation is also known as a clause of insured profile and its major goal is to personalise and delimit the risk so that the insurer can accept or refuse the insurance proposal and establish the prize of the insurance. This study also analyzes the positive and negative aspects of the clause of profile ahead of the rules established for the Consumer Defense Code / O avanço tecnológico, o progresso científico, o parque industrial, e os aglomerados populacionais acarretaram o desenvolvimento sócio-econômico, contudo aumentaram a incidência dos riscos sobre o patrimônio. O contrato de seguro corresponde ao desenvolvimento sócio-econômico, contudo não traduz apenas um negócio jurídico, uma vez que pode significar uma infinidade de relações contratuais, que envolvem uma diversidade de pessoas, em inúmeros ramos do mercado securitário. Ocorre que o contrato de seguro pode ser traduzido pelo contrato de seguro de responsabilidade civil, contrato de seguro de garantias contratuais, contrato de seguro residencial, contrato de seguro saúde, contrato de seguro empresarial, contrato de seguro fiança, contrato de seguro educacional e o contrato de seguro automóvel, objeto do presente estudo. Nesse contexto, impende analisar com profundidade o risco que deverá ser declarado pelo segurado, sendo certo que atualmente ele se consolida por meio do questionário de avaliação do risco, constante da proposta de seguro. O questionário de avaliação do risco é também conhecido como cláusula de perfil do segurado e tem o objetivo de individualizar e delimitar o risco, para que o segurador possa aceitar ou recusar a proposta de seguro e estabelecer o prêmio do seguro. O presente estudo analisa ainda os aspectos positivos e negativos da cláusula de perfil diante dos preceitos estabelecidos pelo Código de Defesa do Consumidor

Direct Activation of TRPC3 Channels by the Antimalarial Agent Artemisinin

(1) Background: Members of the TRPC3/TRPC6/TRPC7 subfamily of canonical transient receptor potential (TRP) channels share an amino acid similarity of more than 80% and can form heteromeric channel complexes. They are directly gated by diacylglycerols in a protein kinase C-independent manner. To assess TRPC3 channel functions without concomitant protein kinase C activation, direct activators are highly desirable. (2) Methods: By screening 2000 bioactive compounds in a Ca2+ influx assay, we identified artemisinin as a TRPC3 activator. Validation and characterization of the hit was performed by applying fluorometric Ca2+ influx assays and electrophysiological patch-clamp experiments in heterologously or endogenously TRPC3-expressing cells. (3) Results: Artemisinin elicited Ca2+ entry through TRPC3 or heteromeric TRPC3:TRPC6 channels, but did not or only weakly activated TRPC6 and TRPC7. Electrophysiological recordings confirmed the reversible and repeatable TRPC3 activation by artemisinin that was inhibited by established TRPC3 channel blockers. Rectification properties and reversal potentials were similar to those observed after stimulation with a diacylglycerol mimic, indicating that artemisinin induces a similar active state as the physiological activator. In rat pheochromocytoma PC12 cells that endogenously express TRPC3, artemisinin induced a Ca2+ influx and TRPC3-like currents. (4) Conclusions: Our findings identify artemisinin as a new biologically active entity to activate recombinant or native TRPC3-bearing channel complexes in a membrane-confined fashion.

Vliv projektové výuky na postoje žáků ve výuce / The influence of project based learning on students' attitudes to instruction

The dissertation is focused on changes in an attitude and initiative students at secondary special school with gastronomy orientation to study special subjects after realization a creative project: "Welcome Day - Day of Open Door - Halloween." The target group are students of Secondary School and College for Tourist Industry in České Budějovice. The dissertation is divided into fifth chapters. I the first two chapters different opinions of experts in teaching methods, classical and alternative methods of education and teaching, teachers competence and authority participation, experimental education are compared. The thirds chapter contains proposals for topics of project learning in schools with gastronomy orientation. There is an enumeration of folk traditions and their connection with the culture and gastronomy. The fourth chapter describes the project realization in Secondary School and College for Tourist Industry in České Budějovice. The fifth chapter contains the research targets, the research methods and interpretation of research results. The target of the dissertation is questionnaire comparison of students' attitudes after realization the project learning. The research assumptions are that students appreciate extension special subjects of folk traditions and do not reject to participation in the pro?ject realization.
