Přístupy tří učitelek ke stejnému učivu / Approaches of three teachers to the same subject matter

The thesis is focused on comparison of three teachers' approaches to the same subject matter in the area of curriculum, differentiation and high expectations. There are analysed topics in question it the theoretical part. The chapter about curriculum contains information about setting curriculum, where is it set, which role does the teacher play in setting curriculum and how he/she chooses the curriculum. We can find there also information about what types of curriculum and approaches to it do exist. The chapter about high expectations explains the term self-fulfilling prophecy and also the way how it is created and how it can be changed. The topic of the last chapter is differentiation, where there are types of differentiation and way of working with pupil according to their needs. There are concisely described two groups of pupils, pupils with special needs and gifted pupils. The approaches of three teachers are based on recordings and interviews. The results of this study give us information about, how the teachers work with textbook materials and what is their approach to chosen unusual topic. The results also tell us, if the teachers are differentiating and if we can observe different behaviour to various pupils. Changes in the intensity of road transportation in CZ - 1995-2010 Abstract This diploma thesis focuses on the development of intensity load of road network between 1995 and 2010. Theoretical part is devoted to general definition of transport and transport systems. The next part is the theoretical concept of research that is focused on the spatial interaction and road transport in terms of spatial structure. The conclusion of theoretical part is devoted to characteristics of the road network and transport resources of Czech Republic. The empirical part of the thesis is describing the methodology of data processing at first. The following is an initially analysis of the evolution of traffic intensity on the motorway and road network in Czechia. Then they are analyzed regional differences in the intensity of road traffic and their evolution over time. Finally, here are three studies. The first is focused on the road transport at inputs to districts with the regional centres. The second is focused on the development of transport in the border region and compared with the hinterland. This thesis is devoted to a relatively little explored issue - the social differentiation of literary characters and the transfer of its linguistic means. The language material for exploring this field of translatology was provided by Döblin's novel Berlin Alexanderplatz (1928) and by two translations into Czech (1935 and 1968). We analyzed utterances of two characters. The first one represents a lower social class and criminal environment. In his speech the analysis showed many elements of Berlin urbanolect which were used for his social characterization. The second character represents the Jewish ethnic group. His direct speech contains elements of literary Yiddish, alluding to his Jewish origins. The subsequent utterance analysis of the first character in Czech translations showed an appropriate use of common Czech as a substitute for Berlin urbanolect in both translations. The analysis was set in the context where the usage of nonstandard elements in Czech and translated literature used to be a convention. Both translators followed these conventions successfully. When translating the literary Yiddish in the direct speech of the second character, significant differences in both translations were discovered. This diploma thesis deals with the political-geographical analysis of the Czech extreme rights, its voters and electoral support in the period after 1990. The paper aims to evaluate the development of the Czech extreme rights in the period 1990-2010 in terms of its importance in the Czech political system, and especially from the perspective of regional differentiation of its electoral support, and to identify possible influencing factors that conduce the voters to support this political stream. The objective is to answer the question whether there are significant regional differences in the support of right-wing extremist parties and, if so, whether these differences are stable over time. Just monitoring of a longer period (1990-2010) will specify whether the regions with above-average support of far-right parties are stable, or whether the extraordinary support of these parties during the reported period varies regionally and some shifts occur for example, due to the social-economic changes in the transformation period. Regional differentiation of gender patterns Abstract It is common nowadays to view the problem of gender from all angles, but until now it has not been considered from a regional point of view. This dissertation focuses on the regional differentiation of gender patterns. The object of this dissertation is to show the biggest diferences between men and women from a regional standpoint by the use of selected indicators. This entails considering the selected gender patterns in distinct districts. As a suitable data basis for this survey results from the last two censuses were chosen, the criterion being permanent residence in an area. The dissertation focused on study of patterns of gender characteristics and their changes between the last two censuses, which took place in the Czech Republic, SLDB 2001 and SLDB 2011. In the first part the indicators are presented for the whole of the Czech Republic. Then follows an analysis of changes in the levels of the chosen patterns between individual censuses and then their structural transformation. Then the dissertation looks at the regional view in more detail. The last part focuses on the creation of district typology in the Czech Republic according to the most important patterns. Regulace alternativního sestřihu pomocí chromatinových modifikací / Regulation of alternative splicing via chromatin modifications

Alternative splicing (AS) is involved in expansion of transcriptome and proteome during cell growth, cell death, pluripotency, cell differentiation and development. There is increasing evidence to suggest that splicing decisions are made when the nascent RNA is still associated with chromatin. Here, I studied regulation of AS via chromatin modification with main focus on histone acetylation. First, we demonstrate that activity of histone deacetylases (HDACs) influences splice site selection in 700 genes. We provided evidence that HDAC inhibition induces histone H4 acetylation and increases RNA Polymerase II (RNA Pol II) processivity along an alternatively spliced element. In addition, HDAC inhibition reduces co-transcriptional association of the splicing regulator SRp40 with the target fibronectin exon. Further we showed that histone acetylation reader, Brd2 protein, affect transcription of 1450 genes. Besides, almost 290 genes change their AS pattern upon Brd2 depletion. We study distribution of Brd2 along the target and control genes and find that Brd2 is specifically localized at promoters of target genes only. Surprisingly, Brd2 interaction with chromatin cannot be explained solely by histone acetylation, which suggests that other protein-domains (in addition to bromodomains) are important for...

The aim of this thesis is the analysis of the position of Italy from the socio-economic point of view in the EU in context of its most important aspect -- differentiation of the regions of North-Centre and South. The thesis is divided into three chapters. The first chapter deals with the comparative profile of Italy in EU including the causes and the development of the Italian social and economic specifics -- the high public debt, the slow economic growth rate and labour market problems. The basic macroeconomic and labour market indicators are used to determine the position of Italy in EU. The second chapter is aimed at the analysis of the primary causes, development and the current state of the Italian dualism on a theoretical level. The third chapter is dedicated to the description of Italian dualism by means of the concrete socio-economic indicators and deals with the comparison on the regional level. The part of this chapter deals with the efectiveness of application of EU regional policy in Italy.

The thesis describes and analyses the development of foreign direct investments in Czech Republic with emphasis on state and regional level. The goal of my thesis, focusing on the topic "Regional differences of foreign direct investments", is to answer the question if foreign direct investments help to decrease the regional disparities. The first part of the thesis oves more detail sof FDI definition, types, factors of localizations and barriers. Second part of the thesis answers the question if FDI brings positive impact to economic development. The third part defines the regions supported by government. The main part of the thesis analyses the development of FDI in Czech Republic and two levels of districts during 2000-2008. In the last part of the thesis are mentioned two examples of foreign direct investment localizated in Czech Republic, in different districts.

The primary function of erythrocytes is transport of oxygen from lungs to various tissues of the body. Red blood cell mass, due to this important role, must be controlled at precise levels. The number of erythrocytes is primarilly increased by the glycoprotein hormon erythropoietin, which expression is controlled by HIF (hypoxia inducible factor). Transcriptional factor HIF consists of the two subunits, HIFα and HIFβ. Under normoxic conditions, alfa subunit of HIF is hydroxylated by PHD protein. This hydroxylation provides a recognition motif for the VHL protein, a part of an E3 ubiquitin ligase complex that targets hydroxylated HIF for proteasomal degradation. Under hypoxic conditions, the degradation is inhibited. The alfa subunit is translocated to the nucleus, where binds the beta subunit and regulates gene expression. HIF pathway regulates a broad spectrum of cellular functions - energy metabolism, angiogenesis, apoptosis and many others. This diploma thesis is focused on HIF2α and its role in erythropoiesis. In this present study, we used CRISPR/Cas9 technology and created HEL (human erythroleukemia) cell line with knock-out of the gene for HIF2α (EPAS1).
