The aim of this thesis is to evaluate the territorial differentiation of the European Union and selected member countries. In this context I observe the differences in major socio-economic indicators (in GDP per capita and employment rates) across the European Union and in more detail in the Czech Republic and Slovakia. Furthermore, I focus on explaining the development and the current setting of EU regional policy at EU level and the CR. The thesis is structured into three main chapters. The first chapter focuses on assessing the economic, social and regional differentiation of the Member States and EU regions. The second chapter is devoted to the analysis of regional structure and differentiation of the Czech Republic and Slovakia. The final chapter covers the development and current form of regional and structural policy of the European Union.

The diploma thesis deals with the generation Y and its approach to employment. The theoretical part is devoted to generational theories and characteristics of existing generations and detailed focus on generation Y. Practical part focus on surveys realized on the topic generation Y in the labour market. There is also own survey in the practical part. The goal of the survey is the comparison of generation Y in Czech Republic and French and its approach to labour market. Based on the results of the researches, proposals are recommended for employers to introduce new instruments for adaptation generation Y in the workplace. Consequently there is assessment of costs for these proposals.

Theoretical part is paying attention to difference in salary, legislative measure of equal pay, gender pay gap index, theories and studies in economics. Practical part of my diploma thesis deals with employment of men and women, analyzes particular factors which affects these salaries differences. Additionally, I focus on position of women in leading and political functions. The last chapter considers differences in salaries between men and women in coun-tries of Visegrad group.

Eurasian Reed Warbler (Acrocephalus scirpaceus) is a long-distance migrant wintering in sub-Saharan Africa and breeding in reedbeds of the Western Palaearctic. European populations migrate in two main directions (SW and SE). Though there is little morphological variation across the breeding range, Asian populations are traditionally separated from the nominate subspecies as the subspecies fuscus. My aim was to explore genetic differentiation among Eurasian Reed Warbler populations in respect to the factors which may have influenced the genetic diversity and divergence of the populations, such as a migratory divide, high migratory connectivity or isolation by distance. I used samples of 495 individuals from 36 populations across the breeding range using microsatellites. Pairwaise fixation indexes FST showed significant differences among populations from the Iberian peninsula, Finland and the Middle East and the rest of populations. Exact tests of differentiation did not confirm non-random distribution between pairs of populations. Overall FST was statistical significant but very small (FST = 0,018). Analysis of molecular variance (AMOVA) revealed that 98% of variation was ascribable to variability of allele frequencies within populations. On the other hand, differences between populations contributed to overall variation with only 2%. Isolation by distance showed only weak relationship between geographical and genetic distances. Main analysis using Bayesian clustering approach implemented in software Structure 2.2. detected no genetic structure of population. Sampled Eurasian Reed Warbler populations seem to form one genetic population. Relatively low genetic diversification indicates large dispersal potential of the studied Reed Warbler populations, when gene flow successfully counters differentiation of population irrespective of large breeding area and high migrate connectivity between breeding and wintering area.

The main function of mesenchymal stem cells in the body is to facilitate the restoration and regeneration of damaged tissues. They are known for the ability to differentiate into tissue originating from the mesoderm, which among others includes connective tissues. Due to this feature are MSCs being intensively examined. Different directions of differentiation can be induced by treatment of specific polypeptides, so called growth factors. In the field of tissue engineering are growth factors used to induce and accelerate the healing processes. They may be incorporated into the nanofiber carrier which is inserted into the site of injury. Cells in this area would thus be stimulated by surrounding 3D microenvironment. At the same time the scaffold provides a supply of growth factors which are able to affect metabolism, motility and differentiation of present cells. In order to induce osteogenic differentiation of human MSCs the following bioactive substances were used: TGF-β, bFGF, HGF, IGF-1, VEGF and the BMP-2 and the organic acid taurine. During 21 days lasting experiments, were these molecules added to the medium in various combinations and in the case of taurine also at two different concentrations. Cells were cultured on plastic. Dispatched 3 (DISP3), sterol - sensing domain (SSD) - containing protein, is a key focus of our laboratory. It was described as a gene regulated by thyroid hormone and its expression is mainly localized within neural tissue. Our preliminary data suggested increased DISP3 expression in medulloblastoma, a highly common pediatric cerebellar tumour, therefore we wanted to examine DISP3 role in human cancer cells. The aim of this thesis is to perform DISP3 overexpression and downregulation in human medulloblastoma cell lines and in mouse neural progenitors and analyse its effect on cell proliferation and differentiation. For this purpose, we chose DAOY and D341, human medulloblastoma cell lines with low and high expression of DISP3 and mouse multipotent neural progenitor cell line, C 17.2, with low DISP3 expression. We showed, that DISP3 ectopic expression leads to increase in cell proliferation in both DAOY and C 17.2 cells. Next, we examined the ability of C 17.2 cells to differentiate into neurons and astrocytes and observed, that cells overexpressing DISP3 reveal delay in differentiation, what we proved by analysis of cell specific markers. Regionálna diferenciácia príčin úmrtnosti vo vybraných krajinách strednej Európy / Regional differentiation of cause-specific mortality in selected countries of Central Europe

The aim of this thesis is to analyse the differentiation in mortality rates at the international and regional level (NUTS 2) in selected countries of Central Europe (Czech Republic, Slovakia, Hungary, Germany, Austria, Switzerland). At the country level, it examines the development of total mortality and cause- specific mortality since 1990. At the regional level, it evaluates the regional differences in total mortality and cause-specific mortality in the period 2008-2010. The thesis makes use of demographic indices (life expectancy, dispersion measure of mortality, lifespan disparity, standardised death rate) and tools of statistical analysis (Poisson regression, cluster analysis). The analysis reveals that mortality rates have decreased in Central European countries, although significant differences in the intensity of mortality can still be observed at the international as well as regional level.
