Two species of cyanobacteria (Fischeralla sp. 29161 and Phormidium autumnale) and one species of green algae (Fritschiella tuberosa) were found to promote survival of Legionella pneumophila in mineral salts medium cocultures. During the early stage of incubation Fischerella sp. supported growth of Legionella pneumophila even though the bacteria would not grow in the algae-free basal medium.

The search for alternative water disinfectants to those commonly used, such as chlorine, probably began when the disadvantages of those disinfectants became known. Soluble disinfectants have short half-lives and need to be replenished periodically which requires monitoring the determination of appropriate concentrations for waters being treated. This disadvantage may be balanced by the ability to alter dose concentrations of soluble disinfectants to meet changes in demands. Maintenance of a residual disinfectant concentration which can act throughout a water distribution system is another advantage of soluble disinfectants. Disadvantages due to reaction of organic materials include the loss of disinfecting capability, health hazards related to the disinfectant and the potential loss of aesthetic water qualities such as taste, smell and color. The purpose of this study was to investigate different surface contact disinfectants which, while having an inflexible dose concentration capability and releasing no residual disinfectant concentration, do not require monitoring and do not significantly leach into the waters they contact. Powdered aluminum when mixed with a loamy sand reduces virus concentration 3-4 orders of magnitude better than controls while not significantly altering the pH or aluminum concentration of waters that pass through the soil columns. Labeled poliovirus 1 was found to adsorb onto the aluminum surface in batch experiments and undergo degradation or dissociation of the capsid proteins with release of viral fragments and clumps within 76 hours. Freon dispersion of the viral clumps showed the clumps to be non-infective. Polyacrylamide gel electrophoresis of poliovirus 1 incubated with aluminum for 76 hours suggested degradation or dissociation of viral capsid proteins 1, 2 and 3. Magnesium peroxide and magnesium oxide, while effectively inactivating viruses, significantly increase the pH of water which they contact. Polyhalex resin and I5 resin reduce virus concentration, but release functional groups into the surrrounding water. Contact disinfectants such as aluminum, may find point-of-use application for drinking water, use in septic tank leachfields or sewage treatment infiltration basins to reduce the threat of spreading potential disease-causing organisms.

O presente estudo teve como objetivos observar a influência de programas de limpeza e desinfecção sobre o desempenho de frangos de corte e na microbiologia das instalações. Foram realizados dois experimentos, ambos com 960 pintos de corte criadosaté 42 dias. No primeiro experimento o piso foi forrado com maravalha nova. No segundo a cama foi reutilizada, proveniente do primeiro experimento. Os tratamentos foram: comum: retirada da matéria orgânica e lavagem da instalação com água; europeu: remoção seca da matéria orgânica, lavagem com água sob pressão, aplicação do detergente alcalino, enxague, secagem, e aplicação de dois desinfetantes(glutaraldeído+formaldeído e paraclorometacresol). Os dados de desempenho avaliados foram consumo de ração, ganho de peso, peso final, conversão alimentar, viabilidade e índice de eficiência produtiva. As análises microbiológicas realizadas foram contagem total de microrganismos, quantificação de Clostridium Spp., E. Coli, Salmonella Spp. e Aspergillus Spp. através de suabes de piso, parede, comedouros, bebedouros e cortinas. Foram ainda coletadas amostras de cama, ração e água para avaliação microbiológica. As analises estatísticas foram realizadas pelo teste F para desempenho e teste de Kruskal-Wallis para microbiologia ambos com índice de significância (p>0,05)através do procedimento GLM do programa SAS . No primeiro experimento não houve diferença estatística significativa para desempenho e avaliações microbiológicas. No segundo experimento notou-se melhor desempenho dos animais do tratamento Europeu e redução da carga microbiológica. Orientador: Elisabeth Loshchagin Pizzolitto / Banca: Jonas Contiero / Banca: Cristina Paiva de Sousa / Resumo: Avaliou-se o perfil de suscetibilidade de Staphylococcus aureus ATCC 6538, Escherichia coli ATCC 25922 e Listeria monocytogenes ATCC 7644, em suspensão e em biofilme formado in vitro, frente à ação dos desinfetantes: ácido iodado, à base de biguanida, à base de quaternário de amônio, à base de ácido peracético e clorado, empregados comumente em indústrias alimentícias brasileiras. As técnicas de microscopia eletrônica de varredura e microbiológica de viabilidade celular foram utilizadas para esta pesquisa. Os resultados demonstraram que as células microbianas em suspensão foram suscetíveis a ação de todos os desinfetantes estudados, não apresentando células viáveis após 10 minutos de tratamento com os desinfetantes. O desinfetante clorado foi o mais eficaz na eliminação das células microbianas em biofilme, proporcionando baixos valores numéricos na contagem das células viáveis e poucas células aderidas à superfície do aço inoxidável após submetido ao tratamento de 10 minutos, enquanto o desinfetante à base de biguanida mostrou ser o menos eficaz na eliminação das células microbianas em biofilme após um tratamento de 10 minutos. A idade do biofilme em horas (24, 75, 120) não mostrou ser um fator influente na resistência das células microbianas aos desinfetantes testados. Comparando-se os resultados da suscetibilidade das células microbianas em suspensão e em biofilme, foi verificado que as células em biofilme são mais resistentes à ação dos desinfetantes. O conjunto dos resultados sugerem uma deficiência de todos os desinfetantes estudados na eliminação de S. aureus, E. coli e L. monocytogenes quando em biofilme. / Abstract: The susceptibility of Staphylococcus aureus ATCC 6538, Escherichia coli ATCC 25922 and Listeria monocytogenes ATCC 7644, in suspension and in biofilm formed in vitro, was studied against the action of disinfectants: iodine, biguanide, quaternary ammonium, peracetic acid and chlorine, commonly used in Brazilian food industries. The scanning electron microscopy and cellular viability techniques were used in this research. The results showed that the microbiological suspension cells were susceptible after 10 minutes of treatment with the disinfectants. The chlorine disinfectant was the most effective against microbiological cells in biofilm, showed low numbers of viable cells and few adhered cells on stainless steel surface after 10 minutes of treatment, while the biguanide disinfectant showed to be the least effective against microbiological cells in biofilm after 10 minutes of treatment. The biofilm age in hours (24, 72 and 120) didn't show to be an influential factor on resistance of microbiological cells to studied disinfectant. The comparison between the results of susceptibility of microbiological suspension cells and microbiological biofilm cells showed that the biofilm cells are more resistant to disinfectants action. The results suggest that the studied disinfectants could not eliminate S. aureus, E. coli and L. monocytogenes biofilms. / Mestre
