<p>Between 1990 and 1992/1993 there was a dramatic change in the Swedish labor market, resulting in an increased number of unemployed and social assistance recipients. As a response to this situation, many municipalities developed local activation programs. One of these programs, the “Uppsala model” – practiced in the City of Uppsala and characterized by a “paternalistic discourse” with sharp means tests, control and sanctions – was quite controversial. The general purpose of this dissertation is to study<b> </b>the effects of this activation method on the claimants and to study these effects with regard to the intentions underlying the method. The central questions are whether this method contributed to a greater number of claimants leaving the system for work or education, after a shorter period of time and on a more permanent basis compared to an alternative and more “traditional” method characterized by a “redistributive discourse”. Another central question is whether the method was effective under different economic conditions. The data used in the analyses were taken from two social welfare offices and included 509 able-bodied individuals who applied for allowances in either 1990 or 1992, reporting unemployment as the main reason. The main results indicated no systematic differences between the two methods with respect to outcome variables such as length of time on social assistance, probability of obtaining a regular job or an education. There were some indications (non-significant) of an increased probability of returning with claims for social assistance and after a shorter period of time, associated with the paternalist Uppsala model. The great importance of the labor market to recipients` possibilities to leave the social assistance system for work, irrespective of working method, was also shown in the study.</p>

Within these years, in order to effectively arrange the limited resource that the government has owned and been able to allocate, a detailed financial plan and a well-structured evaluation system for any project is essential. Normally the most research topics issued are related to the economic effect of natural resources. In Taiwan, the article about analyzing the economic effect about the tour schemes proposed for the development of National Scenic Area is few and only at the beginning stage and the cause mainly is because of the common property of such schemes, i.e. the involvement of different departments needed. At this situation, effected by the different control froces from a varity of government departments and varied industry structures and developing progress in different areas, it is difficult to clarify and analyze the achievement for each department. Owing to this character, the economic effect related to such development schemes is difficult to conclude and it is even harder to evaluate whether or not the economic benefit is definitely resulted from the improving projects.
The research topic is going to comprehend the economic effect caused by the medium-range scheme launched by National Scenic Areas, then is able to propose some feasible items to evaluate the economic effect and collect the figures the Cost-Efficiency Evaluation relaed to the effect. As to the intangible benefit, it is assessed by the Contingent Valuation Method (CVM). The study takes the case of Penghu National Scenic Area to predict the economic effect between 2008 to 2011. From the research, it shows that the figure of Willing To Pay (WPA) among the tourists already visited the Penghu National Scenic Area is NT$6,943 and within 2008 and 2011, after the development implemented, the amount is going to raise to NT$9,638. Based on this raise, the economic revenue would be NT$415,691,000 caculated from the Prediction equation in 2008, NT$428,984,000 in 2009, NT$442,397,000 in 2010 and NT$ 455,933,000 in 2011. Within 10 year of available analytic fixed number of years, the 4-year Net Present Value¡]NPV¡^ ¡]valuta of 2008 year ¡^ will be NT$1,766,586,000 based on the discount rate at 2% and the 4 year Benefit-Cost Ratio, B/C ratio is 3.05>1. From this result, the 2008-2011 development plan itself has demostrated its economic feasibility.

Bakgrund: En viktig fråga hos företag rör belöningssystem och hur dessa ska utformas, såväl hos chefer som hos anställda. Belöningssystem inkluderar allt från lön och bonus till kompetensutveckling. Belöningar finns för att motivera de anställda, men är belöningar alltid bra? Det är inte bara förändringen av belöningssystem som kan leda till negativa reaktioner. En faktor kan vara att implementeringen i sig kan få negativa konsekvenser. Syfte: Huvudsyftet med denna studie är att utvärdera det nya belöningssystemet och hur det har påverkat lastbilschaufförerna inom Södra Skogs åkeri. Ett delsyfte är också att utvärdera vilka effekterna är för Södra Skogs åkeri. Vi ska även beskriva vilka faktorer av förändringen som har påverkat lastbilschaufförerna. Metod: En kvalitativ forskningsmetod med en abduktiv forskningsansats har präglat studien. Två chefer, en ekonomiassistent, en controller och elva lastbilschaufförer har intervjuats genom semi-strukturerade intervjuer. Slutsats: Förändringen av belöningssystemet har skapat ett stort missnöje hos lastbilschaufförerna. De anser att lönen minskat med det nya systemet och att deras motivation har blivit sämre. Förändringen var dock nödvändig för Södra Skog på grund av att det tidigare systemet triggade till olagligheter. Vår egen beräkning visar på att grunderna i systemet faktiskt är bättre för lastbilschaufförerna med det nya belöningssystemet, samtidigt som det nu är mer rättvist. Det största problemet med förändringen är att implementeringen varit bristfällig och att det nu förbjudna övertidsarbetet inte stoppats tidigare. / Background: A major concern of companies is about reward systems and their design, both of directors and of the employees. Reward systems include everything from salary and bonuses to skills development. Rewards are motivating employees, but are the rewards always good? It is not just the change in the reward system that may lead to negative reactions. One factor may be that the implementation itself, which can have negative consequences. Purpose: The main purpose of this study is to evaluate the new reward system and how it has affected the truck drivers in Södra Skog trucking company. A subsidiary aim is to evaluate the effects of Södra Skog trucking company. We will also describe the factors of change that has affected the truck drivers. Method: A qualitative research method with an abductive research approach has characterized the study. Two managers, one assistant, one controller and eleven truck drivers were interviewed through semi-structured interviews. Conclusion: The change in the reward system has created great dissatisfaction among truck drivers. They consider that the salary has been reduced by the new system and their motivation has become worse. However, the change was necessary for Södra Skog because the previous system triggered for illegal acts. Our own calculations show that the basics of the system is actually better for truck drivers with the new reward system since it is now more fair. The biggest problem with the change is that the implementation was inadequate and the now banned overtime work was not stopped earlier.

Custom Fluorophores for Investigating the Cellular Uptake Mechanisms and Side-Effects of PharmaceuticalsChu, Yu-Hsuan 18 May 2015 (has links)
There is a significant current need to elucidate the molecular mechanisms of the side-effects caused by widely-used pharmaceuticals. Examples include the acute nephrotoxicity and irreversible ototoxicity promoted by the cationic drugs gentamicin and cisplatin. Gentamicin is an aminoglycoside antibiotic used for the prevention and treatment of life-threatening gram-negative bacterial infections, such as tuberculosis and meningitis. Cisplatin is used to treat a broad spectrum of cancers including head and neck, ovarian, cervical, stomach, bladder, sarcoma, lymphoma, testicular cancer and others.
The objective of this study is to design and synthesize rhodamine derivatives that can be used for the construction of geometrically well-defined cationic drug conjugates. The long-term goal is to use the conjugates as tools to aid in elucidating the properties and identities of ion channels involved in the uptake of cationic pharmaceuticals into kidney and cochlear hair cells. This will shed light on the origin and potential prevention of unwanted side effects such as nephrotoxicity and ototoxicity associated with specific cationic drugs.
A series of extended rhodamine analogs with reactive groups for biomolecule conjugation has been synthesized. These fluorophores show similar spectral properties to their prototype, Texas Red succinimidyl ester (TR-SE). However, they contain rigid linkers between the fluorophore and amine-reactive moiety. The resultant gentamicin conjugates of these materials are rigidified enabling one to assess channel pore dimensions without the confounding issue of conjugate folding. Preliminary cell studies are promising, as one observes reduced gentamicin uptake in both kidney and sensory hair cell upon systematically increasing the dimension of the fluorophore. This work has enabled us to tentatively assign the maximum dilated MET channel pore size as between 1.44 nm to 1.56 nm. However, this preliminary finding, though encouraging, needs further validation via ongoing studies with larger diameter fluorophore conjugates,
A cisplatin-Texas Red conjugate has also been synthesized to enable studies of cellular uptake mechanisms. This conjugate preserves not only the spectral properties of Texas Red after conjugation, but also the cytotoxicity of cisplatin. This has been validated in zebrafish. The series of rhodamine probes that have been conjugated to gentamicin should be similarly useful for cisplatin studies. These studies are planned. Additional future work includes the synthesis of semi-flexible (glycol) and flexible (alkyl) linkers to evaluate structure-activity relationships.

Intra-nucleus accumbens shell injections of R(+)- and S(-)- baclofen bidirectionally alter binge-like ethanol, but not saccharin, intake in C57Bl/6J miceKasten, Chelsea Rae January 2014 (has links)
Indiana University-Purdue University Indianapolis (IUPUI) / It has been proposed that the GABAB receptor subtype plays a role in alcoholism and alcohol use disorders (AUDs) (Cousins et al., 2002; Agabio et al., 2012). Specifically, the GABAB agonist baclofen has been looked at extensively in clinical and pre-clinical studies. In various animal models of chronic and intermittent consumption, baclofen has been shown to both increase (Petry, 1997; Smith et al., 1999; Czachowski et al., 2006; Moore et al., 2007) and decrease (Colombo et al., 2000; 2002; 2005; Stromberg, 2004; Moore et al., 2009) drinking. A critical issue in determining pharmacological effects of a drug is using the appropriate animal model. The drinking-in-the-dark (DID) model, developed by Rhodes et al. (2005, 2007), produces high levels of drinking in a binge-like paradigm and has been used to assess many pharmacological targets (e.g. Kamdar et al., 2007; Gupta et al., 2008; Moore et al., 2007; 2009).
While DID produces high-levels of binge drinking, it is unclear what areas of the brain are involved in this behavior. A direct way to target areas that are believed to be involved in the circuitry of particular behaviors is through microinjection of drugs (Kiianmaa et al., 2003). Of particular recent interest involving motivated behaviors and addiction is the nucleus accumbens (Acb) (Everitt & Robbins, 2005); specifically the
accumbens shell (AcbSh) (e.g. Rewal et al., 2009, 2012; Nie et al., 2011; Leriche et al., 2008).
The current study aimed to investigate the role of GABAB receptors in the AcbSh by examining the ability of two different enantiomers of baclofen to alter ethanol and saccharin intake in male C57BL/6J (B6) mice. B6 mice underwent bilateral cannulation surgery targeting the AcbSh. After 48 hours of recovery time, animals began a five day Drinking-in-the-Dark (DID) procedure where they received 20% ethanol or 0.2% saccharin for two hours, three hours into the dark cycle, each day. Throughout the five drinking sessions, animals were kept in home-cage locomotor activity chambers to monitor activity throughout the drinking cycle. Day 4 drinking was immediately preceded by a mock microinjection, whereas Day 5 drinking was immediately preceded by a drug microinjection. Microinjection of one of five doses of baclofen was given in ng/side dissolved in 200 µl of aCSF (aCSF alone, 0.02 R(+)-, 0.04 R(+)-, 0.08 S(-)-, or 0,16 S(-)-). Intake was recorded every twenty minutes on Days 4 and 5. Retro-orbital sinus blood samples were taken from ethanol animals immediately following the Day 5 drinking period to determine blood ethanol concentrations (BECs).
A one-way ANOVA on total Day 4 ethanol consumption revealed no baseline differences between dose groups. A one-way ANOVA on total Day 5 ethanol consumption revealed that the 0.04 R(+)- baclofen dose reduced total drinking, but the 0.16 S(-)- baclofen dose increased total drinking (p’s<0.05). This pattern was reflected in the BECs; 0.04 R(+)- baclofen reduced BECs, whereas 0.16 S(-)- baclofen increased BECs (p’s<0.05). These results were also time-dependent, with R(+)-baclofen reducing drinking in the first 20 minutes of the session and S(-)- increasing drinking in the last 40 minutes of the session. There were no effects on saccharin intake. An issue with the locomotor activity boxes led to unreliable locomotor activity counts. However, because there were no drug effects on saccharin consumption, it was concluded that locomotor effects did not contribute to the decreases or increases in ethanol consumption. These results further implicate the role of GABAB receptors in modulating ethanol intake. The bidirectional effects shown highlight the importance of considering enantioselective drug effects when interpreting data. Finally, these results also support previous conclusions that the AcbSh plays an important role in modulating use of drugs of abuse, but not other reinforcers.
