Yield losses due to mildew are caused by reduction in photosynthesis, increase in respiration and transpiration, impairment of heading and grain filling, and loss of plant vigor and growth.

Interaction Chêne-oïdium : caractérisation moléculaire et adaptation locale du parasite, résistance génétique de l’hôte

L’oïdium est une des maladies les plus communes sur chêne en Europe. Cette maladie a été signalée à partir du début du 20ème siècle. Toutefois, peu de données sont disponibles sur l’identité de l’agent causal et de son interaction avec l’hôte. L’objectif de cette étude est d’améliorer les connaissances sur ce pathosystème : (1) caractériser son agent causal avec un marqueur moléculaire (ADN ribosomal) et étudier la distribution spatiale des différentes lignées ; (2) étudier l’adaptation locale du parasite à son hôte ; (3) explorer le déterminisme génétique de la résistance de l’hôte. L’étude de l’ADN ribosomal (ITS=Internal transcribed region et IGS= Intergenic spacer) a mis en évidence l’existence de quatre haplotypes, dont trois seulement avaient été associés à des agents d’oïdium sur chêne en Europe. Erysiphe alphitoïdes, classiquement considéré comme responsable de l’épidémie du XXème siècle est bien l’espèce prépondérante (environ 80% des détections), E. hypophylla et Phyllactinia sp étant détectés à fréquence beaucoup plus faible. Un résultat inattendu est la détection d’une quatrième séquence, présentant 100% d’homologie avec celle de plusieurs agents d’oïdium d’arbres tropicaux et avec Erysiphe quercicola, très récemment décrit sur chêne en Asie et Australie mais jusqu’alors inconnu en Europe. L’ITS de E. quercicola est détecté dans toutes les régions de France, à une fréquence non négligeable (de l’ordre de 15% en moyenne). On le trouve dans les mêmes parcelles et parfois sur les mêmes arbres, voire dans les mêmes lésions que E. alphitoides. La mise au point d’une méthode d’inoculation artificielle a permis d’étudier de façon quantitative l’interaction chêne-oïdium. Une expérimentation d’inoculations réciproques montre que les souches d’oïdium sont plus performantes sur les descendants des arbres dont elles ont été isolées que sur ceux d’autres arbres, suggèrant une adaptation trans- générationnelle. Des inoculations artificielles, complétées par des observations en conditions naturelles, ont également permis de démontrer un déterminisme génétique de la résistance des chênes, avec l’identification de plusieurs QTL. Certains de ces QTL co-localisent avec des QTL de phénologie, en accord avec l’importance de la résistance ontogénique des chênes à l’oïdium. L’ensemble de l’étude amène à reconsidérer l’oïdium du chêne comme un probable complexe d’espèces cryptiques, dont l’histoire de l’invasion et la co-existence en Europe restent à préciser. Les résultats acquis sur l’héritabilité de la résistance et l’adaptation locale du parasite constituent une première étape dans la compréhension des interactions démo-génétiques entre hôte et parasite dans ce pathosystème. / Powdery mildew is the most common disease on oaks in Europe where it was first recorded at the beginning of the 20th century. Yet, little is known about the identity of the causal agent and his interaction with its host. The objective of this study was: (1) to characterize the species responsible of oak powdery mildew with a molecular marker (ribosomal DNA); to study the spatial distribution of these different lineages and (2) to study local adaptation of the parasite to its host (3) to explore the genetic determinism of host resistance. The study of the ITS (internal transcribed region) and IGS (intergenic spacer) diversity revealed the existence of four haplotypes, only three had been already associated with oak powdery mildew in Europe. Erysiphe alphitoïdes, for a long time considered as sole responsible for the epidemic of the XXth century, was the predominant species (~ 80% of detections), E. hypophylla and Phyllactinia sp. were detected at lower frequencies. An unexpected result is the detection of a fourth sequence which show 100% homology with ITS sequences of several powdery mildew agents of tropical trees and Erysiphe quercicola, recently described on oak in Asia and Australia but previously unknown in Europe. E. quercicola ITS was detected in all French regions, at a significant frequency (~ 15%). The study showed that E. alphitoides was often found in association with different ITS types in the same region, the same tree, and even in the same lesion. The development of an artificial inoculation method allowed the quantitative evaluation of the oak-powdery mildew interaction. A reciprocal inoculation experiment showed that powdery mildew strains were more efficient on their mother tress and their descendants than on the other trees, suggesting a trans-generational adaptation. Artificial inoculations, supplemented by observations in natural conditions, have also demonstrated a genetic determination of resistance of oak trees, with the identification of several QTL. Some of these QTL co-localize with QTL controlling phenology, in agreement with the importance of oaks ontogenic resistance to powdery mildew. The entire study leads to reconsider oak powdery mildew as a probable complex of cryptic species; the invasion history and the co-existence in Europe are still to be determined. The results achieved on the heritability of resistance and localization of the parasite are a first step in understanding the demo-genetic interactions between host and parasite in this pathosystem.

Nuovi approcci per la valutazione dell'effetto dei fungicidi nei confronti dell'oidio della vite / NEW APPROACHES FOR THE EVALUATION OF FUNGIDES' EFFECT ON GRAPEVINE POWDWRY MILDEW / New approaches for the evaluation of fungicides’ effect on grapevine powdery mildew

In questo lavoro si intende indagare l'effetto degli anti-oidici più diffusi al fine di individuare nuovi approcci per la ricerca sui fungicidi. Le analisi statistiche mostrano che meptildinocap, zolfo, metrafenone e penconazolo, somministrati a dosi di etichetta, prevengono le infezioni di Erysiphe necator qualora applicati fino a 12 giorni prima dell'inoculazione. Meptildinocap e zolfo conservano una buona efficacia anche se applicati fino a 9 e 6 giorni dopo l'inoculazione. Meptildinocap neutralizza il massimo numero di conidi entro 6 giorni dal trattamento. La meta-analisi multivariata eseguita su prove di campo dimostra che tutti i fungicidi riducono la gravità delle infezioni su grappolo oltre l'80%, qualora applicati da 3 a 13 volte a dosi di etichette secondo il criterio fenologico. Nonostante cyflufenamid, quinoxyfen, fenarimol, fenarimol+zolfo, propiconazolo e kresoxim-metil+boscalid siano significativamente più efficaci dello zolfo, la loro applicazione ripetuta rappresenta una condizione favorevole all’insorgenza di resistenze. L’ approccio chemio-metrico all’ analisi dei dati ottenuti da microanalisi in microscopia SEM a raggi X mostra che meptildinocap induce modificazioni nei rapporti tra Ca e Al e P e S sulla parete dei Chasmoteci trattati. Tali modificazioni sono probabilmente dovute al dissolvimento della membrana plasmatica. Questi nuovi approcci possono rappresentare un'utile integrazione ai metodi già noti. / In this work the effect of the most widespread fungicides used for Erysiphe necator management is investigated in order to test new approaches to fungicide research. ANCOVA and Tukey’s post hoc tests showed that meptyldinocap, sulfur, metrafenone and penconazole administrated at label rates prevent E. necator infections when applied up to 12 days before inoculation. Meptyldinocap and sulfur preserve a good efficacy even when applied until 9 and 6 days after inoculation. Meptyldinocap significantly increases E. necator conidia mortality within 6 days exposure. The multi-treatment meta-analysis performed on field trials shows that all fungicides reduce disease severity over 80% on vine bunches when applied from 3 to 13 times at label rates with the phenological criterion. Although cyflufenamid, quinoxyfen, fenarimol, fenarimol+sulfur, propiconazole and kresoxim-methyl+boscalid are significantly more effective than sulfur their repeated application in vineyard represents an important E. necator resistance risk. The application of chemo-metric approach to data earned by SEM X-ray microanalysis shows that meptyldinocap leads to modifications in the relationships between Ca and Al and P and S in treated Chasmothecia wall. Such modifications are probably due to the plasma membrane disruption. The new approaches tested here may represent a useful integration of already known methods.

Baseado no tamanho da lesão, o porta-enxerto „Dinero‟ foi susceptível, „Strong Tosa‟ moderadamente resistente, „Ojakkyo‟ e „Calabash TI-191‟ resistentes e o isolado Dbr 37 se mostrou mais agressivo dentre os isolados. No segundo experimento, o híbrido de meloeiro „Bônus II‟ foi enxertado nos porta-enxertos „Dinero‟ e „Strong Tosa‟, inoculadas com o isolado Dbr 37 do fungo e submetidas a diferentes concentrações de potássio (0; 62,5; 125; 187,5; 250 mg L-1), tanto plantas enxertadas como pé-franco. As plantas do híbrido „Bônus II‟ enxertadas foram resistentes ao fungo e as pé-franco susceptíveis. As concentrações de potássio não influenciaram no tamanho da lesão e na sobrevivência das plantas, porém influenciaram na condutância estomática e transpiração das plantas. / Chemical control of the fungus Didymella bryoniae, causal agent of gummy stem blight of melon, is frequently inefficient. Alternative methods as grafting on resistant rootstocks and nutrition handling must be studied. Different rootstocks and potassium concentrations were tested to control the disease on net melon grown under plastic house conditions. In a first experiment four rootstocks (melon „Dinero‟, watermelon „Ojakkyo‟, squash „Strong Tosa‟ and calabash „TI-191‟) were tested for resistance inoculated by disks mycelium (7 mm diameter) with four isolates ( Dbr 34, Dbr 36, Dbr 37 and Dbr 39) of D. bryoniae in stem previously injured. Based on lesion size „Dinero‟ proved to be susceptible to the fungus, „Strong Tosa‟ moderately resistant and „Ojakkyo‟ and „TI-191‟ resistant and the isolated Dbr 37 proved more aggressive among isolates. In a second experiment hybrid net melon „Bonus II‟ was grafted on rootstocks „Dinero‟ and „Strong Tosa‟, inoculated with isolate Dbr 37 of the fungus and submitted to different potassium concentration (0; 62,5; 125; 187,5; 250 mg L-1), inoculated both grafted plants as non grafted. Grafted plants of „Bonus II‟ were resistant to the fungus and the non grafted ones susceptible. Potassium concentrations did not influenced size of the lesion on the stem and also in the plants survival, however influence on stomatal conductance and transpiration plants.

O objetivo deste trabalho foi estudar a herança de caracteres qualitativos e quantitativos e mapear genes de resistência dessas doenças, utilizando populações de plantas F2 e F2:3 derivados dos cruzamentos entre os genótipos resistentes à ferrugem asiática PI 200487 e PI 200526 com o genótipo suscetível BRI98-641. Ambos os genótipos apresentaram um gene dominante para a resistência, os quais foram mapeados no mesmo grupo de ligação da soja (GL-N). Adicionalmente, as análises quantitativas demonstraram que o modelo aditivo-dominante foi suficiente para explicar a herança dos dois caracteres, número de urédias e severidade, mostrando que alguns genes adicionais de efeito menores podem contribuir para a resposta da severidade da ferrugem asiática nas duas populações. No entanto, o número de urédias demonstrou ser menos influenciado pelas condições ambientais, indicando que os genes podem estar concentrados apenas nos parentais resistentes, evidenciando o caráter de resistência. A severidade continua sendo importante na seleção, pois demonstra melhor o caráter seletivo e pela facilidade de avaliação. Para oídio da soja o gene de resistência foi mapeado em uma população F2:3, derivada do cruzamento entre os genótipos BR01-22106 (resistente) e PI 200487 (suscetível). A resistência ao oídio nesta população foi determinada por um único gene dominante e permitiu o mapeamento de um loco red no grupo de ligação C2 da soja. Os marcadores associados ao Rpp e Red identificados neste trabalho mostraram-se potencialmente úteis na seleção assistida de plantas resistência para o desenvolvimento de cultivares elites carregando estes genes e para manter... / The pathogenic fungi Phakopsora pachyrhizi and Erysiphe diffusa cause the Asian rust and powdery mildew diseases, respectively, in soybean and are responsible for severe yield reduction around the world. The aim of this work was to study the inheritance of qualitative and quantitative resistance characters and to map disease resistance genes in F2 e F2:3 soybean populations, derived from the crosses between Asian rust resistance genotypes, PI 200487 and PI 200526, with the susceptible genotype BRI98-641. Both genotypes revealed a dominant resistance gene that was mapped in the same soybean linkage group (LG-N). In addition, quantitative analysis demonstrated that the additivedominant model was sufficient to explain the inheritance of both characters, number of uredinia and severity, showing that some additional genes can contributed to soybean response to Asian rust severity in the two populations. However, the number of uredinia showed to be less influenced by environmental conditions, demonstrating that gene could be concentrated the in PI 200487 and PI 200526 and be evidence for resistance character. The severity remains important in the selection process because showing the best selective character and the available to evaluation. For powdery mildew, the resistance gene was mapped on linkage group C2 in a F2:3 populations derived from the cross between BR01-22106 (resistant) and PI 200487 (susceptible) genotypes. The powdery mildew resistance was determined by one dominant gene, mapped on linkage group C2 of soybean. The associate markers to Rpp and Red genes will be very useful to assist the selection of resistant plants on the development of elite cultivars carrying these genes and keep the competitiveness and sustainability of soybean Brazilian agribusiness.

La vigne est soumise à la pression de nombreux bioagresseurs dont des parasites obligatoires tels que l’oïdium et le mildiou. La lutte contre les maladies causées par les pathogènes biotrophes nécessite une utilisation souvent intensive de fongicides. Le vignoble consomme à lui seul 16% des fongicides commercialisés chaque année en France. Pour réduire leur impact environnemental qui conduit à l’acquisition de la résistance aux pesticides des pathogène et la présence de résidus dans les vins et dans l’atmosphère, des efforts doivent être entrepris pour développer des stratégies de protection innovante de remplacement ou complémentaire permettant de réduire les intrants pesticides.Les stimulateurs des défenses des plantes permettent de limiter le développement des bioagresseurs en conditions contrôlées. Toutefois, leurs efficacités in natura sont variables et souvent décevantes. Suite au grand nombre de produits potentiellement stimulateurs des défenses des plantes, et à l’intérêt que leur portent les viticulteurs, il est nécessaire de disposer de connaissances et d’outils qui permettent d’évaluer leus efficacités et mieux connaitre leurs potentiels de protection du vignoble. Pour ce faire, une méthode d’évaluation de l’efficacité de produits potentialisateurs ou éliciteurs a été développée au niveau biologique, moléculaire (expression de gènes impliqués dans les défenses) et biochimique (analyses qualitatives et quantitatives des polyphénols), nommée "BioMolChem". Cette méthode a permis d’évaluer l’efficacité de deux phosphonates et d’un analogue de l’acide salicylique, sur différents génotypes et phénotypes de mildiou de la vigne et d’oïdium. Cette approche méthodologique "BioMolChem" a permis d’établir des corrélations entre l’expression de gènes de défense, la présence de certains stilbènes et une efficacité des défenses de Vitis vinifera cv. Cabernet-Sauvignon vis-à-vis de l’oïdium et du mildiou. Les modifications des patrons d’expression des 19 gènes suivis dans les feuilles de vigne et les profils HPLC de polyphénols révèlent des mécanismes de défense multigéniques et complexes. Ainsi, les réactions de défense de la plante sont-elles modulées, en fonction de l’éliciteur considéré, mais aussi en fonction de la diversité phénotypique et génétique des agents pathogènes contre lesquels elle se défend. Ces défenses se caractérisent par une sur-expression d’un ensemble de gènes de défense et une accumulation de composés phénoliques spécifiques.Les marqueurs (gènes et molécules) ainsi identifiés, la méthode "BioMolChem" a été appliquée in natura et a conforté, pour partie, les résultats obtenus au laboratoire. Dans des conditions de fortes pressions parasitaires, il est donc possible de protéger les feuilles et les grappes, à l’aide de SDP et des essais d’association ou d’alternance avec des fongicides conventionnels montrent l’intérêt potentiel de l’emploi des SDP au vignoble. Chemin faisant, dans le cadre d’une viticulture innovante et durable, les SDP et la méthode "BioMolChem" ont été appliqués sur des génotypes hybrides (Vitis vinifera x Muscadinia rotundifolia). Nous révélons que selon le niveau de résistance intrinsèque des génotypes (plus ou moins résistants à l’oïdium et au mildiou), il est possible d’augmenter le niveau de la résistance exprimée par élicitation. Ainsi, les SDP pourraient-ils s’avérer des alliés d’intérêt pour l’utilisation de variétés partiellement résistantes et limiter potentiellement le contournement des QTL de résistance. L’ensemble de ce travail, à but appliqué, a conduit à l’obtention de résultats qui nous permettent de mieux comprendre comment la vigne réagit aux SDP dans son environnement agronomique. Leur exploitation et leur finalisation devraient nous permettre d’exploiter et de mettre en place une utilisation des éliciteurs mieux adaptée, à des stratégies alternatives ou complémentaires de la gestion des bioagresseurs de la vigne. / Powdery (Erysiphe necator) and downy mildew (Plasmopara viticola) are very important grapevine diseases (Vitis vinifera). These two biotrophic pathogens, which are native to the United States, infect green vine tissues and cause significant economic loss as well as environmental damage through the repetitive applications of fungicides. To reduce their environmental impact efforts should be made to develop strategies to protect innovative alternative or complementary to reduce pesticide inputs.In this study, the efficacy and the role of Benzothiadiazole (BTH), a salicylic acid analogue, and two phosphonate derivatives strengthen plant defence mechanisms against various isolates of downy and powdery mildews (Plasmopara viticola and Erysiphe necator). These compounds showed differences in their efficacy depending on the variability of mildews and highly dependent on plant genetics, environmental conditions and selection pressure. The plant defense stimulation could be an alternative or additional method to traditional pest management in the grapevine.Tools “BioMolChem” were developed to better assess the defence status of the plant defences in vitro and in natura. Transcript kinetics of selected defence-related genes and polyphenol contents profiles, during Vitis vinifera-biotrophic pathogen interaction, were characterized, and the impact of pathogen diversity was investigated in the absence or presence of elicitation. In vineyard, under strong pathogen pressures, it is thus possible to protect leaves and clusters, with SDP and assays of association or alternation with conventional fungicides show the potential interest of the use of these SDP in the vineyard.The grapevine defense mechanisms are complex, depending on the elicitor, leading to the coordinated accumulation of pathogenesis-related proteins (PR), the production of phytoalexins, and the reinforcement of plant cell walls.On the way, within the framework of an innovative and sustainable viticulture, the SDP was applied to hybrid genotypes (V. vinifera x M. rotundifolia). We reveal that according to the level of intrinsic resistance of the genotypes (more or less resistant to powdery and to downy mildew), it is possible to increase the level of the expressed resistance. The SDP could become allies of interest in the use of partially resistant grapevine varieties.The present findings provide insights into the potential use of transcripts and stilbenes as markers of the defense status of grapevine leaves with or without elicitation or infection, which should allow us to exploit and develop a better use of elicitors in alternative or complementary strategies in grapevine pest management.

Uma das principais doenças que ocorrem na cultura é o oídio, causada por Erysiphe pisi DC. O controle da doença tem sido realizado por meio de pulverizações com fungicidas, no entanto, o uso de cultivares resistentes deve ser pensado como uma alternativa viável e de efetivo controle no manejo da doença. O trabalho objetivou elucidar a herança da resistência ao oídio na cultivar de ervilha MK-10 e caracterizar alguns aspectos histológicos da infecção de E. pisi nessa cultivar. Para o estudo da herança, as gerações F1, F2, retrocuzamentos e geração F3 de MK-10 com duas populações suscetíveis foram avaliadas em ambiente protegido e a epidemia foi aberta. Nas avaliações histológicas, os seguintes parâmetros foram avaliados: % de germinação dos conídios, % de conídios germinados que formaram apressório, % de conídios germinados com apressório que estabeleceram colônia (% colonização) e número de haustórios por colônia. Para comparar as razões de segregação obtidas no estudo da herança da resistência, adotou-se o teste do Qui-quadrado (X²) e para os dados das análises histológicas, utilizou-se o teste Tukey a 5% de probabilidade. Com base nas segregações obtidas no estudo da herança e nas análises histológicas realizadas, concluiu-se que a resistência de MK-10 ao oídio é devida a um par de alelos recessivos, provavelmente o er1er1 e que a resistência é expressa na fase de pré-penetração, completada por uma morte celular localizada pós-penetração. / The pea (Pisum sativum L.) is the most produced foods in the world. One of the main diseases that occur in the culture is the powdery mildew, caused for Erysiphe pisi DC. The control of the disease has been carried through by means of sprayings with fungicides, however, the use to cultivars resistants must be thought as a viable alternative and of effective control about the handling of the disease. The work objectified to elucidate the inheritance of the resistance to powdery mildew in cultivar of pea MK-10 and to characterize some histological aspects of the infection of E. pisi in this cultivar. For the study of the inheritance, the F1, F2, generations, backcrosses and F3 generations of MK-10 with two susceptible populations had been evaluated in protected environment, the epidemic were open. In the histological evaluations, the following parameters had been evaluated: percentage of germination, percentage of germinated sporelings forming aprressorium, percentage of germinated sporelings with appressorium that established a colony (% colonization) and number of haustoria for colony. To compare the gotten reasons of segregation in the study of the inheritance of the resistance, the test of Qui-square (X²) was adopted and for the data of the histological analyses, the Tukey test was used 5% of probability. On the basis of the segregations gotten in the study of the inheritance and the carried through histological analyses, were concluded that the resistance of MK-10 to the powdery mildew must to a pair of recessive alleles probably er1er1 and that the resistance is express in the pre-penetration phase, completed for a cellular death located post-penetration.

Dois experimentos foram instalados no município de Pariquera-Açú, em solo orgânico-álico, utilizando-se 10 plantas por introdução, em linhas contínuas com 1,0 x 0,5 m de espaçamento e 3 repetições. No experimento 1 foram utilizadas 31 introduções e no experimento 2, 23 introduções. As avaliações constaram de medidas de número de folhas caídas e presentes na planta, produção, número de ramos laterais e incidência de oídio na folha. A colheita dos frutos foi realizada três vezes por semana, sendo estes colhidos a partir de um comprimento mínimo de 7 cm. A análise dos dados permitiu concluir que houve variação das cultivares quanto a vários fatores estudados: não foi possível encontrar introduções totalmente resistentes; ao oídio, as introduções que mostraram melhores condições de serem aproveitadas em programas de melhoramento, foram N45 (Exp.1) e N25 (Exp.2) quanto à produção, N20 e N50 (Exp1) e N9, N358 (Exp.2) quanto à resistência a oídio, N20, N50 (Exp.1) e N9, N49, N62, N63, N101, N339, N358, N4374, CBVE, CBVC e Sta. Cruz, quanto ao número de folhas caídas... / In the present work, 54 okra (Abelmoschus esculentus (L. ) Moench) germoplasms ware evaluated related to the resistance against Erysiphe cichoracearum, in autumn-winter cultivation in the Vale do Ribeira-SP, Brazil. The appraised material consisted of 03 commercial cultivars, 43 introduction from germoplasm bank of the National Center of Vegetables Researches (CNPH), 13 from the Federal University of Viçosa and 02 from V. O. P. R. D. C. (Japan Min. Agric.). The experiment was installed at Pariquera-Açú, in organic-alic soil; it consisted of 10 plants/introduction, in line with 1,0 x 0,5 m of spacing with 03 repetitions. The evaluations consisted of measures of height of the plants, number of fallen leaves, according to the powdery mildew incidence in the leaf, and yield. Thirty one introductions were evaluated in the experiment 1 and 23 introductions in experiment 2. Fruits harvest was done in a frequency of three times a week. Fruits were picked with a minimum length of 7 cm. Analysis of data allowed the following conclusions: a) there was variation among cultivars with relationship to several studied characteristics; b) it was not possible to find a cultivar completely resistant to powdery mildew, appearing presence of polygenic resistance; c) cultivars that showed able conditions to be used at breeding programs were N45 (Exp.1) and N25 (Exp.2), high powdery mildew resistance: N20, N50 (Exp.1), N9,N358 (Exp.2), CBVE and N29; e) fallen leaves N20, N50 (Exp.1) and N45, N49, N62, N63. Molecular marker analysis of adult plant resistance to powdery mildew in common wheat

Powdery mildew, caused by Blumeria graminis (DC.) E.O. Speer f. sp. tritici E'm. Marchal (syn. Erysiphe graminis f. sp. tritici), is one of the major diseases of wheat (Triticum aestivum L.) worldwide. The use of cultivars with resistance to powdery mildew is an efficient, economical and environmentally safe way to control powdery mildew. Race-specific resistance has been extensively used in breeding programs; however, it is ephemeral. Adult plant resistance (APR) to powdery mildew is more durable as demonstrated by the cultivar Massey, which has maintained its APR to powdery mildew since its release in 1981. To develop an efficient breeding strategy, it is essential to understand the genetic basis of APR. The objectives of this study were to identify molecular markers associated with APR to powdery mildew in common wheat Massey and to verify their association using recombinant inbred (RI) lines. A cross was made between the powdery mildew susceptible cultivar Becker and Massey. One hundred and eighty F2:3 lines were rated for disease severity under natural pressure of powdery mildew in field. Using both restriction fragment length polymorphism (RFLP) and microsatellite markers, three quantitative trait loci (QTL), designated as QPm.vt-1B, QPm.vt-2A and QPm.vt-2B, were identified in the Becker x Massey F2:3 generation. These loci are located on chromosomes 1B, 2A and 2B, respectively, and explained 17%, 29% and 11% of the total variation among F2:3 lines for powdery mildew resistance, respectively. Cumulatively, the three QTLs explained 50% of the phenotypic variation among F2:3 lines in a multi-QTL model. The three QTLs associated with APR to powdery mildew were derived from Massey and displayed additive gene action. QPm.vt-2B also fits a recessive model for APR to powdery mildew. In the second part of this study, 97 RI lines were developed from the Becker x Massey cross. The RI lines were evaluated for APR to powdery mildew under natural disease pressure for three years. Both single marker analysis and interval mapping confirmed the presence of the three QTLs identified in the F2:3 generation. The three QTLs, QPm.vt-1B, QPm.vt-2A and QPm.vt-2B, accounted for 15%, 26% and 15% of the variation of mean powdery mildew severity of the RI lines over three years. In a multi-QTL model, the three QTLs explained 44% of the phenotypic variation of the RI lines. The RI lines were grouped according to the genotype of the three QTLs, represented by markers GWM304a, KSUD22 and PSP3100, respectively. The RI lines with Massey alleles at all three loci had a mean disease severity of 3.4%, whereas the RI lines with Becker alleles at all three loci had a mean disease severity of 22.3%. These severity values are similar to those of the corresponding parents. The molecular markers identified and verified as to their association with APR to powdery mildew in Massey have the potential for use in marker-assisted selection for resistance to powdery mildew and in pyramiding powdery mildew resistance genes, as well as facilitating a better understanding of the molecular basis of APR to powdery mildew. / Ph.
