Host-parasite interaction between the potato tuber rot nematode (Ditylenchus destructor), stem nematode (Ditylenchus dipsaci) and potato

ZUSAMMENFASSUNG Nematoden rufen weltweit erhebliche Verluste in der Kartoffelproduktion hervor. Der Knollenfäule-Nematode Ditylenchus destructor Thorne 1945 und das Stock-und Stangelälchen Ditylenchus dipsaci Kühn 1857 Filipjev, 1936, verursachen Läsionen an der Kartoffelknolle, was deren Qualität und Marktwert verringert. Diese Nematoden sind aufgrund ihres umfangreichen Wirtspflanzenspektrums schwierig zu kontrollieren und werden daher in vielen Ländern als Quarantäne-Nematoden geführt. In der hier vorliegenden Doktorarbeit wurden Experimente durchgeführt, um die Wechselwirkung von Kartoffel mit jeder dieser beiden Nematodenarten zu untersuchen. Verschiedene Populationen von D. destructor und D. dipsaci wurden molekular und morphometrisch charakterisiert. Die Gensequenz kodierend für die Untereinheit I der Cytochrom Oxidase (COI) auf der mitochondrialen DNA (mtDNA) wurde analysiert, um eine phylogenetische Beziehung zwischen den untersuchten Populationen darzustellen. Die Ergebnisse zeigten zwei deutlich getrennte Cluster für die D. destructor und die D. dipsaci Populationen. Eine Diskriminanzanalyse der morphometrischen Daten von Männchen und Weibchen von D. destructor und D. dipsaci Populationen verdeutlichte, dass diese beiden Arten mittels a-ratio und ihrem Durchmesser unterschieden werden können. Die sich ergänzende Kombination von molekularen und morphometrischen Untersuchungen ermöglichte die Identifikation der untersuchten Arten. Es wurden zwei Gewächshausexperimente mit 25 angebauten Kartoffelsorten durchgeführt, um diese auf Resistenz und Toleranz gegenüber Nematoden zu testen. Dazu wurde ein Standard-Screening Protokoll entwickelt. Resistenz und Toleranz wurden gemäß der derzeitigen Definition (Nematologie) bewertet. Die relative Anfälligkeit und äußere Verletzung der Kartoffelknolle stellten sich als beste Parameter für die Beurteilung der Resistenz respektive der Toleranz heraus. Alle 25 untersuchten Kartoffelsorten waren weder resistent noch tolerant gegenüber D. destructor oder D. dipsaci. Jedoch wurden Unterschiede in der Toleranz der Sorten festgestellt. Im Vorfeld zum Pflanzenexperiment wurden die Populationsdichten von D. destructor und D. dipsaci und deren Einfluss auf Ertragsverlust untersucht. Die initiale Populationsdichte hatte einen signifikanten Effekt auf den Schaden an der Kartoffelknolle und den Fortpflanzungsfaktor der Nematoden unter Gewächshausbedingungen. D. destructor verursachte Schäden bereits bei einer geringeren initialen Populationsdichte als D. dipsaci. Unter Klimakammerbedingungen wurde gezeigt, dass die Temperatur Einfluss auf die Schadensrate von D. destructor und D. dipsaci an Kartoffelknollen nimmt. Im Gegensatz zu einer Temperatur von 16°C waren 26°C sowohl optimal für die Vermehrung von D. destructor als auch für D. dipsaci. Obwohl für D. destructor und D. dipsaci unterschiedliche Temperaturanforderungen beschrieben wurden, verursachten beide Arten in ähnlichen Temperaturbereichen äußere Verletzungen an der Kartoffelknolle. Beauveria bassiana ist ein weltweit verbreiteter Pilz, der vor allem zur Bekämpfung von Insektenschädlingen in der Kartoffelproduktion Anwendung findet. Doppelbefall von Kartoffeln mit B. bassiana Sporensuspensionen im Boden mit D. destructor oder D. dipsaci begünstigte die Nematoden, was zu einer erhöhten Nematodenfortpflanzung und Knollenbeschädigung führte. B. bassiana allein war nicht schädlich für die Kartoffel. Es wurde vermutet, dass B. bassiana eine indirekte Rolle in der Wechselwirkung Nematode-Kartoffel spielt. Vorschläge für zukünftige Experimente werden angebracht, die einen weiteren Beitrag zu den hier vorgestellten Studien leisten können.

Agricultural businesses face many internal and external factors that significantly affect their business. Aspects such as management, marketing, internal and external environments are increasingly important for agricultural businesses in the Czech Republic. This thesis analyses the external environment of the company, including PEST analysis and Porter's five forces model. Both models are explored in detail including explanations of their main role and functionality within the given environment. Also explained is the Boston matrix to define the internal environment of the company. This thesis provides a SWOT analysis- techniques focused on the evaluation of internal and external factors influencing the organization's success. The practical part of this thesis will implement the theoretical aspects previously explored within a selected agricultural business of Central Bohemia. This thesis will explain and analyse the external and internal environment of the selected agriculture business. A general overview of the models will be defined and then implemented upon the agricultural business of Klučenice a.s.

Arbetsgivarvarumärket inom en kommun : Vad är en attraktiv arbetsgivare? / The employer brand within a municipality : What is an attractive employer?

Studiens syfte var att i samarbete med en kommun i Mellansverige genomföra en studie om kommunens arbete vad gällde deras arbetsgivarvarumärke samt upplevelsen av en attraktiv arbetsgivare. Detta har undersökts ur arbetsgivarens respektive medarbetarnas perspektiv. Studien genomfördes med kvalitativ metod och byggde på semistrukturerade intervjuer. Data analyserades med hjälp av en induktiv tematisk analys. I resultatet framkom olika teman till respektive frågeställning. De teman som framkom angående hur kommunen arbetar med sitt arbetsgivarvarumärke var Kommunens varumärke och Värdegrundsarbetet. De teman som framkom kring frågeställningen om vad som kännetecknar en attraktiv arbetsgivare enligt medarbetarna var Kollegor och stöttande chefer, Arbetsmiljö och trygghet, Personlig utveckling, Möjlighet till att påverka, förändra och bidra, Personalvård, Kommunikation och struktur samt Introduktion. De teman som framkom enligt kommunens representanter var Stöttande chefer och kommunikation, Personlig utveckling och möjlighet att påverka samt Arbetets innehåll. Arbetsgivarens och medarbetarnas uppfattning av en attraktiv arbetsgivare visade sig ha många gemensamma nämnare, däremot lyfte medarbetarna ytterligare teman. De teman som utkristalliserades som faktorer för att vara en attraktiv arbetsgivare stämde väl överens med synen på ett starkt arbetsgivarvarumärke, något som stärkte syftet med studien. / The aim of this study was to, in collaboration with a municipality in Central Sweden, carry out a study on the municipality's work regarding their employer brand and the experience of an attractive employer. This has been examined from the perspective of the employer and the employees. The study was conducted with a qualitative method and was based on semi-structured interviews. Data was analyzed using an inductive thematic analysis. In the result, various themes emerged from each question. The themes that emerged regarding how the municipality works with its employer brand were the Municipality's brand and the overall Values. The themes that emerged about the issue of what characterizes an attractive employer according to the employees were Colleagues and supportive managers, Work environment and security, Personal development, Opportunity to influence, change and contribute, Staff welfare, Communication and structure, and Orientation. The themes that emerged according to the municipality's representatives were Supportive managers and communication, Personal development and the opportunity to influence, and Work content. The employer's and employees' perception of an attractive employer turned out to have many common themes. However, the employees highlighted additional themes. The themes present in the study for being an attractive employer matched well with the view of a strong employer brand, which strengthened the purpose of the study.

The objective of this research was to evaluate the efficiency of the hydrocooling in the fruit post-harvest quality of Cantaloupe melon Torreon . An experiment was carried out in a completely randomized design in a split plot scheme with four replications. In the main plots were assigned the treatments with and without fruit immersion in cold water at 5ºC and to subplots were assigned the storage times of 0, 14, 21, 28 and 35 days at 3ºC. From all analyzed characteristics, it was only observed a significant interaction between treatments and storage times on external (EA) and internal appearances (IA), and reducing sugars (RA). The fruits kept marketing pattern until the storage time of 35 days, with score above three for both external and internal appearance, without symptom of sunken patche (brown spots in the fruit epidermis) in both treatments. The hydrocooling produced a high fruit firmness, but it did not alter the soluble solids and titrable acidity contents. Moreover, the hydrocooling was not efficient to prolong the storage life of melon fruits / O hidroresfriamento é um método de pré-resfriamento adotado por alguns produtores como alternativa para remover mais rapidamente o calor dos justos de melão visando restringir seu metabolismo, reduzindo as perdas pós-colheita. O objetivo deste trabalho foi avaliar a eficiência do hidroresfriamento na qualidade póscolheita do melão Cantaloupe Toneon . Um experimento foi realizado no delineamento inteiramente casualizado em esquema de parcelas subdivididas com quatro repetições. Nas parcelas foram distribuídos quatro repetições. Nas parcelas foram distribuídos os tratamentos com e sem imersão dos justos em água fria a 5ºC e nas subparcelas foram alocados os tempos de armazenamento de 0, 14, 21, 28 e 35 dias a 3ºC. Para todas as características avaliadas apenas foi observado interação significativa entre os tratamentos e o tempo de armazenamento nas aparências externa (AE) e interna (AI), e conteúdo de açúcares redutores (RA). Os frutos se mantiveram comercializáveis até o tempo de armazenamento de 35 dias, com nota acima de três para ambas as aparências externa e interna, sem sintomas de sunken patche (manchas amarronzadas na epiderme dos frutos) em ambos os tratamentos. O hidroesfriamento promoveu maior firmeza de frutos, mas não alterou os conteúdos de sólidos solúveis e acidez titulável. After more than twenty years since the birth of those initiatives, discussions about the schools own internal assessment by teachers have been intensified. Bringing near the outer large-scale evaluation to the school’s internal evaluation arises as a problem today in the Brazilian educational scenario. In this context, we have delimited the object of this research: the evaluator-teacher. We have defined as general objective of this research the discussion on the ethical identity of evaluator-teacher. The specific objectives that guided this study were planned as follows: (1) to discuss the ethical identity of evaluator-teacher in the context of the paradigm docimológico school evaluation, (2) to discuss the ethical identity of evaluator-teacher in the context of the pedagogical paradigm of school evaluation, (3) to identify the reference of powers attributed to the evaluator-teacher on Aprender pra valer (Learning for sure) program, by SEDUC-CE. This research falls under the qualitative approach by stating – among other features – the descriptive and interpretative matter, the interest on the use of the results, use various data sources. The methodological procedures substantiated in documentary analysis and literature review. The data were interpreted by content analysis. For purposes on this study we defined as theoretical references Bloom, Hastings and Madaus (1983), Perrenoud (1999), Afonso (2000), Hadji (1994.2001), Tyler (1949), Bonniol and Vial (2001). The survey results indicated that external evaluation on a large scale will influence on ethical identity of evaluator-teacher by proposing a framework for the construction of assessment instruments: the Reference Matrix. The Learning For Sure Program (Aprender Pra Valer) indicates the external evaluation on a large scale as the main agent of the management system, school management and the management of the classroom. However, this program does not have a specific project for the evaluator-teacher’s education evaluator and to confirm the importance of internal school evaluation. The docimologic paradigm allows us to identify the duties of the evaluator-teacher from the perspective of evaluation instruments. While the formation of the evaluator-teacher, based on educational paradigm, lies in their education in a complex frame of the systemic approach initially announced by Tyler (1977) and confirmed by Bloom, Hastings and Madaus (1983), Perrenoud (1999, 2000, 2002) and Hadji (1994,2001). / Diversas iniciativas de avaliação externa em larga escala surgiram no Brasil durante a década de 1990, a destacar: Sistema Nacional da Avaliação Básica (SAEB) e Exame Nacional do Ensino Médio (ENEM). Passados mais de vinte anos, desde o nascimento dessas iniciativas, intensificaram-se as discussões em torno da própria avaliação escolar interna realizada pelos professores. Aproximar a avaliação externa em larga escala da avaliação interna escolar coloca-se como um problema da atualidade no cenário educacional brasileiro. Nesse contexto, delimitamos o objeto desta pesquisa: o professor-avaliador. Definimos como objetivo geral deste trabalho discutir a identidade deontológica do professor-avaliador. Os objetivos específicos que orientaram este trabalho foram assim planificados: (1) discutir a identidade deontológica do professor-avaliador no contexto do paradigma docimológico da avaliação escolar, (2) discutir a identidade deontológica do professor-avaliador no contexto do paradigma pedagógico da avaliação escolar, (3) identificar o referencial de competências imputado ao professor-avaliador do programa Aprender pra Valer SEDUC-CE. Esta pesquisa se inscreve na abordagem qualitativa por manifestar, entre outras características, o caráter descritivo e interpretativo, o interesse pela utilização dos resultados, a busca por diversas fontes de dados. Os procedimentos metodológicos consubstanciaram-se em análise documental e pesquisa bibliográfica. Os dados foram interpretados pela análise de conteúdo. Para efeito deste estudo, como referenciais teóricos, demarcamos Bloom, Hastings e Madaus (1983), Perrenoud (1999), Afonso (2000), Hadji (1994, 2001), Tyler (1949), Bonniol e Vial (2001). Os resultados da pesquisa indicaram que a avaliação externa em larga escala exerce influência sobre identidade deontológica do professor-avaliador ao propor um referencial para a construção de instrumentos de avaliação: a Matriz de Referência. O Programa Aprender Pra Valer indica a avaliação externa em larga escala como principal agente da gestão do sistema, da gestão da escola e da gestão da sala de aula. No entanto, esse programa não apresenta um projeto específico para a formação do professor-avaliador nem confirma a importância da avaliação escolar interna. O paradigma docimológico nos permite identificar os deveres do professor-avaliador na perspectiva dos instrumentos de avaliação, enquanto a formação do professor-avaliador, baseada no paradigma pedagógico, situa a formação dele em um quadro complexo da abordagem sistêmica anunciada inicialmente por Tyler (1977) e confirmada por Bloom, Hastings e Madaus (1983), Perrenoud (1999, 2000, 2002) e Hadji (1994, 2001).

Passados mais de vinte anos, desde o nascimento dessas iniciativas, intensificaram-se as discussÃes em torno da prÃpria avaliaÃÃo escolar interna realizada pelos professores. In this context, we have delimited the object of this research: the evaluator-teacher. We have defined as general objective of this research the discussion on the ethical identity of evaluator-teacher. The specific objectives that guided this study were planned as follows: (1) to discuss the ethical identity of evaluator-teacher in the context of the paradigm docimolÃgico school evaluation, (2) to discuss the ethical identity of evaluator-teacher in the context of the pedagogical paradigm of school evaluation, (3) to identify the reference of powers attributed to the evaluator-teacher on Aprender pra Valer (Learning for Sure) program, by SEDUC-CE. This research falls under the qualitative approach by stating â among other features â the descriptive and interpretative matter, the interest on the use of the results, use various data sources. The methodological procedures substantiated in documentary analysis and literature review. The data were interpreted by content analysis. For purposes on this study we defined as theoretical references Bloom, Hastings and Madaus (1983), Perrenoud (1999), Afonso (2000), Hadji (1994.2001), Tyler (1949), Bonniol and Vial (2001). The survey results indicated that external evaluation on a large scale will influence on ethical identity of evaluator-teacher by proposing a framework for the construction of assessment instruments: the Reference Matrix. The Learning For Sure Program (Aprender Pra Valer) indicates the external evaluation on a large scale as the main agent of the management system, school management and the management of the classroom. However, this program does not have a specific project for the evaluator-teacherâs education evaluator and to confirm the importance of internal school evaluation. The docimologic paradigm allows us to identify the duties of the evaluator-teacher from the perspective of evaluation instruments. While the formation of the evaluator-teacher, based on educational paradigm, lies in their education in a complex frame of the systemic approach initially announced by Tyler (1977) and confirmed by Bloom, Hastings and Madaus (1983), Perrenoud (1999, 2000, 2002) and Hadji (1994.2001).

The diploma thesis deals with a strategic analysis of the agricultural company AGRO Dolní Kralovice, s.r.o. The aim of the thesis is to process the strategic analysis of the company, evaluate its results and suggest some recommendations or possible changes. Strengths, weaknesses, opportunities and threats are identified in order to fulfil the aim. The thesis uses analytical methods, namely the analysis of external, competitive and internal environment. The methodology of the thesis builds on a qualitative research. The first part includes theoretical knowledge of the strategic management and selected strategic analysis. The second part is based on the aim of the thesis to process the strategic analysis, carry out the external and internal analysis and evaluate their results. The thesis also includes information about the agricultural sector itself, captures its progressive development and positive or negative changes. It is possible to fully use the resulting recommendations for management's needs.
