Previous empirical research of co-workership focuses on experiences of implementation or development programs. This study particularly intends to examine co-workership itself as there are no studies with the same focus. The purpose of this study was to shed light on employees’ perceptions of co-workership after an implementation of the topic and describe employees’ perceptions of sustainable and health-promoting working life factors in relation to co-workership. This is a qualitative study with semi-structured interviews based on ten employees from the same organization. The data collection is analyzed according to a phenomenographic approach to highlight significantly different perceptions. The outcome space describes employees’ perceptions of co-workership. Categories which highlight interactions & relationships and organizational insight & awareness as central perceptions of co-workership in this study. The category harmony emerged from perceptions of sustainable and health-promoting working life factors. Finally, the category symbiotic relationship, refers to perceptions of the relationship between sustainable and health-promoting working life factors and co-workership. This research contributes to increasing knowledge about how employees and work groups perceive co-workership where the implementation has taken place. Which also highlight the topics importance in promoting a sustainable and healthy working life.

Evaluating comprehension of temporary traffic control

There are over 5 million reported motor vehicle collisions annually in the United States, and while crash rates and fatality rates have declined in the past decades, rates in work zones are disproportionately high. There are strict standards for evaluating the crashworthiness of temporary traffic control devices, but not for evaluating drivers’ comprehension of existing or novel device deployments. This dissertation presents a series of three experiments evaluating driver comprehension for existing and novel traffic control devices conducted in a work zone setting. This evaluation is further expanded by decomposing the task of comprehending traffic control into the three subtasks of detection, localization, and identification. Methods are proposed for conducting a computer-based experiment with still image stimuli to measure participant performance at each of these subtasks. Next, procedures for categorizing localization responses and accounting for variation in participants physical responses are explored. Lastly, an application of Item Response Theory toward the evaluation and comparison of participant comprehension is demonstrated. It is hoped that these methods and procedures can be used by future researchers and experimenters to compare novel temporary traffic control devices and systems to inform future design.

The study investigated the cognitive opinions expressed by consumers in an online shopping environment in the sport supplementation industry as well as the influence certain factors identified through literature, had on the decision-making of these consumers. This was done in an attempt to better understand the online consumer in order to improve marketing campaigns within an e-Commerce environment. Primary data was collected by means of a snowball sampling strategy. Respondents were asked to complete an online questionnaire that consisted of 68 questions. 118 respondents initialized the online questionnaire and 102 fully completed it. Initial analysis proved the data to be sufficient for factor analysis where after confirmatory factor analysis was performed. A new framework to measure influential factors in an online environment was established. 6 out of 10 factors were found to contain sub-groups of influence which suggests that certain questions pertaining to those factors need improvement or rephrasing in order to measure the stand-alone factor more accurately. The reliability of all the factors was measured adequately, noting that Service Quality, Product Risk and Vendor Familiarity could be improved by further developing their underlying variables. Overall it was concluded that Price and Convenience were seen as the greatest motivational factors when a consumer decided to shop online, and Vendor Familiarity and Website Usability were regarded as two crucial components in the comfort level portrayed by consumers while shopping online. Trust and Security were also pointed out to be a major concern for online shoppers in a South African context. The findings of this study are of great value to entrepreneurs and businesses alike seeking to expand or increase their product offering into an online environment. Given the research results from this study and future studies on this topic, marketing strategies can be constructed more precisely in order to reach a target audience in an optimum and more productive manner. / MBA, North-West University, Potchefstroom Campus, 2014

The effects of growth factors on bovine satellite cells

This study examined the effects of basic fibroblast growth factor (bFGF), insulin-like growth factor I (IGF-I) and transforming growth factor beta (TGF-β) on the proliferation and differentiation of bovine satellite cells (BSC) in vitro. Cells were treated with serum-free defined media containing varying concentrations of bFGF, IGF-I and TGF-β. On day 3 of treatment total cell nuclei and myotube nuclei were determined. bFGF stimulated BSC proliferation in a dose-dependent fashion with half-maximal stimulation observed at a concentration of 5 ng/ml (p < .05). Similar results were found for IGF-I and TGF-β in the presence of FGF, with half-maximal stimulation observed at 5 ng/ml and 1 ng/ml, respectively. With regard to differentiation, TGF-beta inhibited myotube formation at concentrations above 0.05 ng/ml. IGF-I stimulated myotube formation at concentrations as low as 10 ng/ml (p < .05). These results demonstrate that proliferation and differentiation of BSC in vitro are affected by growth factors, and consequently, similar effects may be found in vivo. This information may prove to be useful in future methods of manipulating muscle growth in vivo.

Régulation des gènes C/EBPS par des voies de signalisation intracellulaire dans les cellules épithéliales intestinales

Nous avons utilisé la lignée épithéliale intestinale de crypte de rat IEC-6 afin de mieux comprendre le rôle des C/EBPs dans la différenciation et la prolifération de l'intestin. Nous avons vérifié l'expression des ARNm de C/EBP$\alpha$, C/EBP$\beta$ et C/EBP$\delta$ suite à la stimulation des cellules IEC-6 par la forskoline et l'IBMX qui stimulent la PKA, le TPA qui stimule la PKC et la thapsigargine qui augmente la concentration de Ca$\sp{2+}$ cytoplasmique. La forskoline et l'IBMX induisent les niveaux d'ARNm des trois C/EBPs avec différentes cinétiques tandis que le TPA induit seulement les niveaux d'ARNm de C/EBP$\alpha$ et de C/EBP$\beta.$ La thapsigargine induit les niveaux d'ARNm des trois isoformes par un mécanisme de régulation post-transcriptionnel sans affecter les niveaux de protéines. Les variations des niveaux d'ARNm, induits par les trois voies de signalisation, étaient indépendantes d'une synthèse nouvelle de protéines. Les analyses des niveaux de protéine par Western nous ont permis de constater une induction des trois isoformes en accord avec celle des ARNm suite à une stimulation par la forskoline et l'IBMX. Nous avons observé que le patron d'expression des protéines C/EBP$\alpha$ et C/EBP$\beta$ concorde avec celui des ARNm après une stimulation par le TPA. La capacité de liaison à l'ADN des C/EBPs n'est pas affectée significativement par les voies de la PKA, de la PKC et du Ca$\sp{2+}.$ Les voies de la PKA et de la PKC peuvent moduler l'expression de certains gènes de la réponse inflammatoire comme l'haptoglobine et le thiostatin. La stimulation de ces différentes voies inhibe la prolifération cellulaire des cellules IEC-6 en absence et en présence de sérum. Finalement, nous avons observé que ces différentes voies pouvaient induire à des niveaux variables l'ARNm de p21, un gène très important dans l'arrêt de la prolifération cellulaire. Factors influencing adolescents' attitude towards biology in Gazankulu

The purpose of this study was to investigate if teacher, pupil, curriculum and classroom environment variables were significantly related to adolescents' attitudes towards Biology. The study also investigated whether certain factors (as moderator variables) significantly influenced the relationships between pupils' attitudes towards Biology and each of the independent variables. For the investigation a sample of 426 standard eight, nine and ten pupils from three school-types completed an attitude questionnaire. Parametric statistics were used to analyse the results. Findings from the correlation and stepwise regression analysis revealed that the curriculum was the strongest correlate of attitude and accounted for most of the variance in pupils' attitudes towards Biology. An analysis of variance revealed no significant difference in pupils' attitudes towards Biology when gender, standard, parental levels of formal education and type of school were used as moderator variables. Thesis (PhD)--Stellenbosch University, 2013. / ENGLISH ABSTRACT: Although the effect of inhaled corticosteroids (ICS) on the hypothalamic- pituitary-adrenal axis (HPA) has been regarded as a “benign physiological response”, a survey published in 2002 suggested that adrenal crisis is more common in asthmatic children on ICS than previously thought. Relying on clinical features to detect chronic adrenal insufficiency secondary to corticosteroids may not be wise, as these are non-specific and can therefore easily be missed. Accurate biochemical assessment of the whole axis to detect subclinical HPA suppression (HPAS) is thus desirable. A review of the literature indicates that basal adrenal function tests, including plasma cortisol profiles, do not identify which children can appropriately respond to stress. There is no evidence to suggest that the degree of the physiological adjustment of the HPA to ICS and/or nasal steroids (by reducing basal cortisol production), predicts HPAS. Cortisol profiles should therefore only be used to demonstrate differences in systemic activity of various ICS and delivery devices. Only two tests, considered as gold standard adrenal function tests [the insulin tolerance test (ITT) and the metyrapone test] can assess the integrity of the whole axis. / AFRIKAANSE OPSOMMING: Die outeurs van ´n opname wat in 2002 gepubliseer is stel voor dat ´n bynierkrisis meer algemeen by asmatiese kinders, wat inhalasie kortikosteroïede ontvang, voorkom as wat voorheen gedink is.
