Determination of in vitro effects of aqueous extract of camellia sinensis on human sperm functions

(Medical Sciences)) -- University of Limpopo, 2021 / Infertility, defined as the inability to conceive following one year of unprotected sexual intercourse, respectively affects 25% of couples globally. Oxidative stress (OS) has been greatly related to the idiopathic cause of infertility and Camellia sinensis contains antioxidants that may enhance reproductive functions. This study focussed on the effects of Camellia sinensis (green and black tea) on human sperm functions in both normal and abnormal samples. Semen samples (n= 59) collected from donors were liquefied, analysed, and classified as normal (n=40) and abnormal (n= 19) using the WHO criteria. Samples were washed and exposed to aqueous leaf extracts of green and black tea (0, 0.4, 4, 40, 405 μg/ml) for 1 hour. Human Tubular Fluid (HTF) served as the control. The respective sperm parameters were analysed (sperm motility, vitality, DNA fragmentation, mitochondrial membrane potential (MMP), capacitation and acrosome reaction (CTC) and reactive oxygen species (ROS). Green and black tea significantly increased vitality, and intact MMP, while it significantly reduced, CTC, and intracellular ROS as well as DNA fragmented spermatozoa in both normal and abnormal samples compared to the control (p<0.05). A significant increase in sperm CTC, ROS, with a decrease in sperm vitality, and intact MMP was observed in the abnormal compared to the normal samples (p<0.05). No significant change in motility was observed between normal and abnormal samples compared to their respective controls, in both green and black tea (p>0.05). Bakgrund: Att få besked om infertilitet kan beskrivas som en livskris. En fertilitetsutredning kan klargöra orsaken. Barnmorskans kompetensområde innefattar att främja förmågan att kunna fortplanta sig och möjligheten att kunna få friska barn. Syfte: Syftet var att beskriva kvinnors upplevelser av att genomgå IVF-behandling under tidsperioden då Covid-19 pandemin pågick. Metod: En kvalitativ metod, i form av netnografi har använts. En icke-deltagande observation valdes, genom en granskning av sju bloggar. Materialet analyserades med hjälp av Kozinets metod för analys av netnografiskt material. Resultat: Analysprocessen resulterade i sex teman och sex kategorier. Huvudkategorierna var följande; en lång väntan, att hålla hårt i hoppet, behandlingen i praktiken, betydelsefulla relationer, ständig oro och samla kraft.  Slutsats: Att gå igenom en IVF-behandling är en påfrestande upplevelse för kvinnorna och att lägga till en pandemi på en redan svår situation gav upphov till ökad stress, känsla av ensamhet och oro. Att få besked om att få beviljad behandling ledde till en känsla av ökat hopp. Upplevelserna av de praktiska momenten pekade på att injektionsbehandling och äggplock var särskilt svåra. Att inte få ha med sig en medföljare till följd av restriktioner, ledde till känslor av ensamhet och orättvisa. / Background: Being notified of infertility can be described as a life crisis. A fertility assessment can clarify the cause. The midwife's area of expertise includes promoting the ability to reproduce and the possibility of being able to have healthy children. Aim: The aim was to describe women's experiences of undergoing IVF treatment during the time period when the Covid-19 pandemic was ongoing. Method: A qualitative method, in the form of netnography has been used. A non-participatory observation was chosen, through a review of seven blogs. The material was analyzed using Kozinet's method of analysis of netnographic material. Results: The analysis process resulted in six themes and six categories. The main categories were as follows; a long wait, to hold tight to hope, treatment in practice, meaningful relationships, constant worry and gathering strength. Conclusion: Going through an IVF treatment is a stressful experience for the women and adding a pandemic to an already difficult situation gave rise to increased stress, feelings of loneliness and worry. Being told to receive treatment led to a sense of increased hope. The experiences of the practical elements indicated that injection treatment and egg picking were particularly difficult. Not being allowed to have a companion with you as a result of restrictions, led to feelings of loneliness and injustice.

Infertilitet är ett relativt vanligt hälsoproblem i dagens samhälle. Infertilitet kan uppstå som resultat av en sjukdom där en störning av reproduktiva kroppsfunktioner hindrar uppkomsten av en graviditet och födsel av ett barn. Flertalet kvinnor som senarelägger starten på barnafödandet löper en större risk att drabbas av primär och sekundär infertilitet. Barnlöshet går inte alltid att förebygga och alla kvinnor som önskar att få barn kan inte alltid få barn. Syftet med det här arbetet var att belysa kvinnors upplevelse av ofrivillig barnlöshet efter misslyckade försök att bli mamma. Litteraturöversikt är den metod som har valts för att besvara syftet med det här arbetet. Integrerad analys är den dataanalysmetod som har används. Resultatet visar att isolering från omgivningen är vanlig förekommande bland kvinnor som är ofrivilligt barnlösa. Ofrivilligt barnlösa kvinnor kunde uppleva sin kropp som ett hinder för konception, vilket gav dem känslan av att vara ofullständiga i jämförelse med andra kvinnor. Sorgen över barnlösheten var ständigt närvarande hos kvinnorna. När kvinnorna accepterar sin situation som barnlös sker en personlig mognad och de börjar lägga fokus på sig själva och mål som är uppnåbara. Slutsatsen som framkommer i det här arbetet är att kvinnorna i den här litteraturöversikten oavsett kulturell bakgrund har liknande upplevelser i sin längtan efter ett barn. Att vara ofrivillig barnlös är en stor påfrestning för den som är drabbad. Det är av stor vikt att ofrivilligt barnlösa kvinnor erbjuds emotionellt stöd under sorgeprocessen som följer barnlösheten samt att stötta dem att identifiera strategier för att hantera sorgen.

Orientador: Silvia Helena Venturoli Perri / Banca: José Fernando Garcia / Banca: José Antônio Visintin / Resumo: O crescente avanço da produção in vitro de embriões bovinos intensificou a utilização de outras biotecnologias da reprodução tais como a micro-manipulação embrionária e o diagnóstico genético pré-implantacional, sendo a identificação do sexo embrionário utilizada na rotina comercial de laboratórios de produção in vitro. O objetivo deste trabalho foi avaliar as interações de diferentes fatores sobre a taxa de mortalidade embrionária e a proporção do sexo de embriões bovinos submetidos ao processo de sexagem. Foi realizado levantamento no banco de dados da Transfix - Transplante de Embriões Ltda, Patrocínio Paulista / Brasil, referente a 4.650 embriões produzidos in vitro e sexados entre 2005 e 2007. Os embriões foram submetidos à micro-manipulação pela técnica de micro-aspiração, e as biópsias à reação em cadeia pela polimerase (PCR). Somente as fêmeas foram transferidas para receptoras previamente sincronizadas. O diagnóstico de gestação e a determinação do sexo fetal foram realizados por ultra-sonografia. As variáveis foram classificadas de acordo com o sexo dos embriões (macho, fêmea e indeterminado), cinco laboratórios (A, B, C, D e E), seis raças bovinas (Nelore, Brahman, Girolando, Simental, Holandês e Jersey), estágio embrionário (MO, BI, BL, BX e BE), qualidade embrionária (1, 2 e 3) e qualidade da biópsia ("dentro do padrão" e "fora do padrão"). As análises estatísticas foram realizadas pelos testes 2 de associação, 2 de aderência para proporção 1:1 e pela análise de regressão logística com o método de Hosmer-Lameshow utilizando o procedimento logístico (PROC LOGISTIC) programa computacional SAS. The present study aimed to evaluate the interactions among different factors on the mortality rate and sex ratio of in vitro produced bovine embryos. A survey was performed in the Transfix - Transplante de Embriões Ltda, Patrocínio Paulista / Brazil data base, referring to 4.650 in vitro produced bovine embryos sexed during years 2005/2007. Embryos were submited to the biopsy by the microaspiration technique, and biopsies to the polymerase chain reaction (PCR). Only female embryos were transferred to synchronized recipients. Pregnancy diagnosis and fetal sex determination were carried out by ultrasound. The variables were classified according embryo sex (male, female and indeterminate), five laboratories (A, B, C, D and E), six bovine breeds (Nellore, Brahman, Girolando, Simmental, Holstein and Jersey), embryo stage (MO, EB, BL, XB and HB), embryo quality (1, 2, and 3) and biopsy quality ("standard" and "non standard"). The statistical analysis was carried out by association 2 test, 2 for 1:1 ratio and logistic regression analysis with Hosmer- Lameshow method using logistic procedure (PROC LOGISTIC) of SAS package. The PCR showed 93.3% efficiency, 93.2% accuracy and male and female ratio of 52.9% and 47.1%, respectively. Mortality rate of biopsied embryos was 10.3% and pregnancy rate was 31.7%. Although no significant differences were observed between male and female ratio, indeterminate embryos possess greater possibility to die after micromanipulation.
