Spectroscopic Investigation into Minor Groove Binders Designed to Selectively Target DNA Sequences

Recently, there has been increasing focus toward the development of small molecules designed to target a specific sequences of double stranded DNA for therapeutic purposes1. Minor groove binding compounds have been shown to be capable of selectivity target GC sites in AT tract DNA2. In this research, binding selectivity was investigated using absorption, fluorescence and circular dichroic properties of selected DB minor groove binders in the presence of two unique DNA sequences. Further insight was gained by comparing the electrostatic potential maps and optimized structures of the compounds of interest. Using the results presented, potential selective minor groove binders can be selected for further investigation and kinetic studies.

This work was created in cooperation with company Mubea. Introduction part describes purpose of automotive headrest and function of special grooves, which are fitted on headrest frame. Overview of manufacturing technologies of these grooves with short summary of their advantages and disadvantages follows. The main part of this work consists of development of new alternative technology for manufacture special grooves in thin-walled tubes with following experimental verification of technological conditions and shape of grooves. This work also contains evaluation of tool life and wear. Interpretation of the results is performed by calculations and graphically.

This master's thesis considers by single-phase asynchronous motor. The first part discuss about construction, principle of operation and basic parameters of this motor type. It also discussed the emergence of torque and torque characteristics of the different types of engines. The second part explains the basic ways of obtaining grip moment including outline the principles of functions particulars constructions types, which is produce in practice. In the third part is minutely analyse single-phase asynchronous motor with auxiliary phase and permanently conect capacitor, including method of assign size of a capacitor. Fourth part section provides the principles of the procedure for calculating basic parameters of the single asynchronous motor with auxiliary phase. At the last part is accomplished a calculation of single-phase asynchronous motor with permanently conect capacitor at auxiliary phase including comparation with already manufacturing motor.

The current large concentration of wireless networks brings new horizons, but also new concerns. Failure to follow basic rules may produce far-reaching problems that could make more wrinkles to all affected managers and administrators. The aim of this thesis was to measure and compare the radio parameters of selected products and how they behave in neighboring interference and the speed they have while there are hidden nodes. The resulting values were measured in the laboratory network Wificolab and compared with the various support protocols. Possible effects on the specific situation are also analyzed in this thesis.

This diploma thesis deals with project of shaft manufacturing using drop forging technology. This technology is manufacturing using cubic forming on vertical forging press. Work includes detail data for working out technical drawings of suggested component. Thanks to modification of parameters and forging a whole we reduce material waste which is compared to turning technology. Production process is adapted to established production process in company Kovarna ZETOR Brno. Further the work includes cost price of one forging from the set of 30 000 products a year. The work also contains background research of drop forging technology, technical drawings, production process list and simulation outcome for drop forging of the suggested komponent by software FormFEM.

This diploma thesis deals with experimental study of the effect of surface grooves on distribution of lubricant film in elastohydrodynamic lubricated point contact. Lubricant film thickness was measured by ball-on-disc apparatus using an optical interferometry technique - thin film colorimetric interferometry. The effects of groove geometry, lubricant and operating conditions were studied in this diploma thesis. The effects of these variables are evaluated based on newly suggested parameter Rgi (groove influence ratio). Groove influence ratio is defined as an influenced film thickness influenced by groove divided by film thickness for smooth surfaces.

Design, Synthesis and Study of DNA-Targeted Benzimidazole-Amino Acid Conjugates

Indiana University-Purdue University Indianapolis (IUPUI) / The DNA minor groove continues to be an important biological target in the development of anticancer, antiviral, and antimicrobial compounds. Among agents that target the minor groove, studies of well-established benzimidazole-based DNA binders such as Hoechst 33258 have made it clear that the benzimidazole-amidine portion of these molecules promotes an efficient, site-selective DNA association. Building on the beneficial attributes of existing benzimidazole-based DNA binding agents, a series of benzimidazole-amino acid conjugates was synthesized to investigate their DNA recognition and binding properties. In this series of compounds, the benzimidazole-amidine moiety was utilized as a core DNA “anchoring” element accompanied by different amino acids to provide structural diversity that may influence DNA binding affinity and site-selectivity. Single amino acid conjugates of benzimidazole-amidines were synthesized, as well as a series of conjugates containing 20 dipeptides with the general structure Xaa-Gly. These conjugates were synthesized through a solid-phase synthetic route building from a resin-bound amino acid (or dipeptide). The synthetic steps involved: (1) the coupling of 4-formylbenzoic acid to the resin-bound amino acid (via diisopropylcarbodiimide and hydroxybenzotriazole); followed by (2) introduction of a 3,4-diaminobenzamidoxime in the presence of 1,4-benzoquinone to construct the benzimidazole ring; and, finally, (3) reduction of the resin-bound amidoxime functionality to an amidine via treatment with 1M SnCl2•2H2O in DMF before cleavage of final product from the resin. The synthetic route developed and employed was simple and straightforward except for the final reduction that proved to be very arduous. All target compounds were obtained in good yield (based upon weight), averaging 73% mono-amino acid and 78% di-amino acid final compound upon cleavage from resin. Ultimately, the DNA binding activities of the amino acid-benzimidazole-amidine conjugates were analyzed using a fluorescent intercalator displacement (FID) assay and calf thymus DNA as a substrate. The relative DNA binding affinities of both the mono- and di-amino acid-benzimidazole-amidine conjugates were generally weaker than that of netropsin and distamycin with the dipeptide conjugates showing stronger binding affinities than the mono-amino acid conjugates. The dipeptide conjugates containing amino acids with positively charged side chains, Lys-Gly-BI-(+) and Arg-Gly-BI-(+), showed the strongest DNA binding affinities amongst all our synthesized conjugates.

Four chord fenomenet : En studie i att återanvända en ackordföljd

I denna studie har jag undersökt den välkända ackordfunktionen I–V–vi–IV inom populärmusiken, även kallat ”four chord” fenomenet. Undersökningen har gått ut på att skriva tre låtar med samma ackordföljd (med ackordfunktionen I–V–vi–IV) i syfte att försöka göra låtarna så olika varandra som möjligt, för att komma fram till vilka musikaliska parametrar som gör störst skillnad för att återvinna en ackordföljd och göra låten så unik som möjligt, samt undersöka på vilket sätt musiken påverkas av valen som görs utifrån de musikaliska parametrarna. Jag valde detta ämne då jag har skrivit musik i många år och själv använt mig av denna ackordfunktion och därmed känt att det är lätt att hamna i samma bana och tankesätt när jag använder den, vilket ofta resulterar i låtar som har fler likheter med varandra än vad jag skulle önska. Jag kände att denna undersökning skulle kunna konkretisera och bryta ned processen i teoretiska byggstenar, för att kunna analysera vad jag kan göra och vilka redskap samt hjälpmedel jag kan använda mig av för att undvika att hamna i samma fälla när jag skriver låtar med denna ackordfunktion. Men jag valde också detta ämne då jag anser att det inte finns så mycket konkret forskning eller undersökningar om det, trots fenoments räckvidd, så jag kände att detta skulle kunna vara intressant information för andra än bara mig att ta del av och gynnas av.Det jag har kommit fram till i denna studie är att de musikaliska parametrarna som påverkar och gör störst skillnad för att låtar med samma ackord blir så unika som möjligt är rytm (rytmisering av ackord) och taktart. En annorlunda rytmisering av ackorden med synkoperingar och olika fokusgrader på ackorden gör att ”four chord” känslan lättare försvinner, och taktart placerar mig som låtskrivare i andra tankegångar kopplat till genre under skrivandeprocessen vilket resulterar i ett annorlunda slutresultat.Studien har försett mig med ny kunskap bland annat om mig själv som låtskrivare och vad som fungerar bra och mindre bra för mig, men även om ämnet som helhet. Jag tar med mig redskap och tips som jag fått från denna undersökning som jag kommer kunna använda mig av i mitt fortsatta låtskrivande. Jag hoppas även att andra låtskrivare som kanske också befinner sig i samma sits som jag befann mig i innan denna undersökning också kan dra nytta av den och komma till egna personliga insikter som för deras låtskrivande framåt samt utvecklar deras förståelse för fenomenet.
