Deep Water Mixing Prevents Harmful Algal Bloom Formation: Implications for Managed Fisheries Refugia

Inflows affect water quality, food web dynamics, and even the incidence of harmful algal blooms. It may be that inflows can be manipulated to create refuge habitat for biota trying to escape poor environmental conditions, such as fish populations in lakes during times of toxic Prymnesium parvum blooms. Water availability sometimes can be an issue, especially in arid climates, which limits this approach to management. Utilizing source water from deeper depths to displace surface waters, however, might effectively mimic inflow events. I test this notion by conducting in-lake mesocosm experiments with natural plankton communities where I manipulate hydraulic flushing. Results show that P. parvum cell density is reduced by 69%, and ambient toxicity completely ameliorates during pre-bloom conditions in the lake. During conditions of bloom development, population density is reduced by 53%, toxicity by 57%, and bloom proportions are never reached. There is minimal effect of these inflows on total phytoplankton and zooplankton biomass, and little effect on water quality. Shifts toward more rapidly growing phytoplankton taxa are observed, as are enhanced copepod nauplii. In other words, while inflows using deep waters suppress P. parvum bloom initiation and development, they are benign to other aspects of the lower food web and environment. The results from using deep lake water to suppress harmful algal blooms indicate this may be a promising management approach and further studies are recommended to test whether this mitigating effect can translate to a large-scale in-lake treatment.

Culture, Abstinence, and Human Rights: Zulu Use of Virginity Testing in South Africa’s Battle against AIDS

Virginity Testing, a traditional Zulu pre-nuptial custom that determines the worth of a bride, has been resurrected in communities in KwaZulu-Natal in South Africa as a response to the HIV/AIDS epidemic. The practice takes place during large community festivals when young girls have their genitals physically examined to determine whether they are virgins and results are made public. Supporters of the tradition claim that in fostering a value of chastity among its youth, it encourages abstinence from sexual intercourse which leads to a lower HIV infection rate and prevents the disease from spreading. Human rights activists disagree; Rather than slowing the spread of a disease, they argue, the practice instead endangers girls. Those who fail are often shunned and turn to prostitution, while those who pass may be exposed as potential targets for rape (due to a myth that says intercourse with a virgin cures HIV/AIDS). Despite a ban on the practice in 2005, the testing festivals continue, and are described by supporters as an important part of the preservation of Zulu culture. This thesis examines the ways in which human rights may be re-negotiated for young girls in Zulu communities while maintaining a respect for local culture. It moves beyond the traditional debate between relativism and universalism in order to propose solutions to rights violations in culturally diverse contexts by exploring ideas of inclusive human rights and capabilities theories.

Holistic approach to the evaluation of the anthropocentric influence on domoic acid production and the corresponding impact on the California Sea Lion (Zalophus californianus) population

Domoic acid (DA) is a neurotoxin produced by the harmful algae Pseudo-nitzschia that has been directly linked to mass stranding events of the California Sea Lion (CSL). The purpose of this paper is to review the anthropogenic influence on the production of this neurotoxin and examine how human activities are impacting this marine mammal species. A comprehensive and interdisciplinary literature review was conducted to evaluate the future sustainability of the CSL population. It was found that while Pseudo-nitzschia bloom developments are vulnerable to anthropogenic influences, the incontestable existence of natural contributing factors adds a certain complexity to the determination of causalities and the development of solutions. Strong evidence exists to show that DA can cause major and irreversible neurological damage in CSLs. Rehabilitation of DA-impacted CSLs is a polarizing issue in the U.S. and presents interesting implications for sustainable development. While the CSL population is currently healthy and plenteous, the strong abundance of future uncertainties warrants concern. A balance must be found between the involving social, economic, and environmental factors to ensure a promising future for the CSL species.

Risk and resources in the plankton: effects on copepod population growth and zooplankton community dynamics

The focus of my thesis research is on the interplay between individual behavior, population dynamics and community-level processes within zooplankton communities in coastal Maine. The target organisms of my thesis work are marine copepods. Copepods are small (1-10 mm) crustaceans that perform the essential ecosystem function of consuming and assimilating primary production (phytoplankton) making it available to higher trophic levels such as commercially important fishes. Therefore, copepod population growth is of critical importance to marine food webs. Fertilization limitation has been suggested as a constraint on copepod population growth but field surveys describing the prevalence of fertilization limitation are lacking. During my doctoral research, I explored the in situ fertilization success of two marine copepod species, Temora longicornis and Eurytemora herdmani in coastal Maine. I collected monthly zooplankton samples and analyzed clutches from field-caught females using an egg-staining technique. My results indicate that both species exhibit fertilization limitation in nature and the factors correlated with their fertilization span population, community and ecosystem level factors. To determine a causal relationship between predator density and copepod mating success, I conducted laboratory experiments to assess the effects of a common mysid shrimp predator, Neomysis americana on Eurytemora herdmani mating success. I subjected males and females to predators or predator cues. I found that the presence of a mysid predator, or only a predator cue, reduced copulation frequency and spermatophore transfer leading to a 38-61% decrease in E. herdmani nauplii production. These results suggest that mysid predators can constrain copepod population growth through non-consumptive processes. To determine the effects that resources can impose on copepod behavior, I explored the behavioral and fitness consequences of Temora longicornis ingesting Alexandrium fundyense, a phytoplankton species that forms harmful algal blooms in coastal Maine. My results suggest that ingesting A. fundyense causes copepods to swim faster and with more directional persistence compared to control algae. Temora longicornis increased their average swimming velocity by 24%, which leads to a 24-54% increase in their theoretical encounter rate with predators. Therefore, these findings suggest behaviorally mediated copepod-algal interactions may have significant impacts on harmful algal bloom dynamics and the fate of toxins in marine food webs.

Molecular Detection of the Toxic Marine Diatom Pseudo-nitzschia multiseries

The marine diatom genus Pseudo-nitzschia includes species that produce domoic acid, a neurotoxin responsible for illness and mortality in both humans and marine wildlife. Because of the expertise and time required for the microscopic discrimination of species, molecular methods that monitor environmental concentrations of Pseudo-nitzschia provide a rapid alternative for the early detection of blooms and prediction of toxin accumulation. We have developed a nucleic acid sequence-based amplification with internal control RNA (IC-NASBA) assay and a quantitative reverse transcription PCR (qRT-PCR) assay for the detection of the toxic species P. multiseries targeting the ribulose- 1,5-biphosphate carboxylase/oxygenase small subunit (rbcS) gene. Both methods use RNA amplification and fluorescence-based real-time detection. Due to a limited rbcS sequence database, primers were designed and used to sequence this gene from 14 strains of Pseudo-nitzschia (including four P. multiseries) and 19 other marine diatoms. The IC-NASBA and qRT-PCR assays had a limit of detection of one cultured cell of P. multiseries and were linear over four and five orders of magnitude, respectively (r2 ! 0.98). Neither of the assays detected closely related organisms outside the Pseudo-nitzschia genus, and the qRT-PCR assay was specific to P. multiseries. While cross-reactivity of primers with unknown species prevented reliable detection of P. multiseries in spiked environmental samples using IC-NASBA, the qRT-PCR assay had positive detection from 107 cells/L to 103 cells/L. Nearly a 1:1 relationship was observed between predicted and calculated cell concentrations using qRT-PCR. Based on a diel expression study, the rbcS transcript copy number per cell ranged from 2.16 x 104 to 5.35 x 104, with the highest expression during early to mid photoperiod. The rbcS qRT-PCR assay is useful for the detection and enumeration of low concentrations of P. multiseries in the environment.

Comparative effects of the toxic dinoflagellate, Karenia brevis, on bivalve molluscs from Florida

The effects of the toxic dinoflagellate, karenia brevis (Wilson clone), on larval survival and development of the northern quahog (=hard clam, Mercenaria mercenaria), eastern oyster (Crassostrea virginica) and bay scallop (Argopecten irradians) were studied in the laboratory. The effects of K. brevis on feeding activities of juveniles from these species plus the green mussel (Perna viridis) were also examined. Finally, adult bay scallops were exposed to K. brevis for two weeks to investigate possible cytotoxic effects. Survival of 3-day-old larvae was generally > 85% for all shellfish species at Karenia brevis densities of 100 cells . ml-1 or less, and not significantly different between whole and lysed culture. At 1,000 cells . ml-1, survival was significantly less in lysed culture than whole culture for both M. mercenaria and C. virginica. Survival of 7-day-old larvae in all species was not significantly affected at densities up to 1,000 cells . ml-1. At 5,000 cells . ml-1, however, survival was reduced to 37, 26 and 19% for A. irradians, M. mercenaria and C. virginica, respectively. Development of C. virginica and M. mercenaria larvae was protracted at K. brevis densities of 1,000 cells . ml-1. Clearance rates of juveniles were determined under static and flow-through conditions using whole and lysed cultures of K. brevis. The bay scallop was most sensitive, exhibiting a 79% reduction in clearance rate at 1,000 cells . ml-1 of whole culture. The eastern oyster was least responsive, showing a 38% reduction in clearance rate between the same treatments. The green mussel and the northern quahog displayed intermediate responses. Similar results were observed during longer (2 day) exposures to a continuous supply of K. brevis. Bay scallops showed a significant decline in clearance rate at 100 cells . ml-1 after 24 hr exposure; clearance rate of oysters was not affected by K. brevis at this concentration. No mortality was observed for any species during these brief exposures. Adult bay scallops exposed to K. brevis for two weeks showed degenerative and inflammatory changes in the digestive gland, including reduced thickness of the epithelium, increased size of digestive tubule lumens and hemocytic infiltration. The prospect for recovery of bay scallop populations in Florida may be hampered by recurring blooms of K. brevis.

Monitoring and modeling water quality at the C.W. Young Regional Reservoir

This work explores the relationship between nutrient loading and changes in water quality in a sub-tropical, above-ground, off-stream municipal water supply reservoir, the C.W. Bill Young Regional Reservoir. The three source waters for the reservoir have varied but high levels of nutrients such as phosphorus and nitrogen. In other reservoirs, these nutrients have been linked to deterioration of water quality and increased expense in water treatment. The need to minimize excess nutrients results led to the primary research question: what allocation of withdrawals from the three sources will minimize the deterioration of water quality? To answer this question, the relationship between nutrient and other water quality data, such as temperature,phosphorus, chlorophyll a, Secchi depth, and trophic state indices were explored. Results indicate that temperature had a correlation with observed water quality. 27.9% of the variability in trophic state index as a function of Chlorophyll a was correlated with average temperature at one foot below water level. Correlation and regression models were developed using available time-series of linear and log-transformed water quality data to predict Chlorophyll a response. The parameters used in the model were selected from correlation matrices and from the P value in the multiple regression. The models developed were significant at P < 0.05. In the developed models, temperature was found to have greater predictive strength than nutrients indicating that this reservoir may be more strongly influenced by season and light than by nutrient limitation. Lastly, the US Army Corps of Engineers' eutrophication model, BATHTUB, was used to simulate different loading conditions and trophic response. The model results indicate that use of water the middle pool or lower pool of the Tampa Bypass Canal yield similar trophic states with the middle pool slightly lower. Use of water from the Alafia River yielded the highest trophic state and would be expected to have negative impacts on water quality.

Molecular and phytochemical investigations of the harmful, bloom-forming alga, Prymnesium parvum Carter (Haptophyta)

This dissertation includes molecular and phytochemical investigations of the harmful, bloom-forming alga, Prymnesium parvum, including analysis of known polyketide metabolites as a function of salinity and growth. Initially, the development of molecular and phytochemical tools was necessary for the detection and quantification of P. parvum and its associated toxins. Suites of oligonucleotides and molecular beacons were designed for conventional and quantitative multiplex PCR to amplify four species- and gene-specific products simultaneously that were used for the detection and quantitation of P. parvum. This built-in redundancy provided increased confidence in reactions with the positive confirmation of four discrete products. Techniques were also developed for the chemical enrichment of toxins produced by P. parvum. Until now, isolation of “prymnesins” has never been reproduced. Polyketide prymnesins possess unique spectral properties that were used to generate an LC-MS fingerprint that comprised 13 ion species. Preliminary investigations using chemifluorimetric methods were also capable of detecting prymnesins in the pico- and nano-molar range. Environmental samples were tested as an independent assessment of these methods. Lastly, the roles of polyketide prymnesins were analyzed with respect to total hemolytic activity (HA) as a function of culture age and salinity. Variation in HA of supernatants was statistically significant relative to both variables (p << 0.05). Salinity was inversely related to HA wherein cultures growing in 5-25 psu were 150-200% more hemolytic. Total HA was inversely related to culture age during the first three weeks, but positively related to it during the next three weeks. Interestingly, no hemolysis was detected in fractions containing prymnesins from culture supernatants and the majority of hemolysins remained in the aqueous phase. Prymnesins extracted from cells varied significantly over the 6-week observation period (p << 0.05); HA was positively correlated during the first half and inversely related during the last half of the study. Salinity was directly related to HA from cell extracts, but these effects were not significantly different until the last three weeks. These investigations suggest that polyketide prymnesins are present at much lower quantities than previously believed, and they may not be the key compounds associated with hemolysis due to P. parvum. / text

Viena aktualiausių šiandienos problemų - moksleivių fizinių ir sveikatos rodiklių blogėjimas. Fizinis aktyvumas vienas iš svarbiausių moksleivių vystymosi ir sveikatos stiprinimo veiksnių. Tačiau fizinis aktyvumas, vis dar nepakankamai vertinamas. Atlikti tyrimai Lietuvoje rodo, kad vaikai nėra pakankamai aktyvūs. Taip pat nustatyta, kūno masės didėjimas, sutrikusios medžiagų apykaitos požymiai,bei, kad anksti įprantama rūkyti, vartoti alkoholį, netinkamai maitintis bei nepakankamai judėti (Zaborskis, 1997). Tyrimo tikslas: Išanalizuoti 12 -17 metų moksleivių psichologinę savijautą, požiūrį į savo sveikatą ir fizinį aktyvumą lyties aspektu. Tyrimo uždaviniai: 1. Palyginti 12 - 17 metų berniukų ir mergaičių psichologinę savijautą; 2. Palyginti 12 - 17 metų berniukų ir mergaičių požiūrį į savo sveikatą; 3. Palyginti 12 – 17 metų berniukų ir mergaičių fizinio aktyvumo lygį. Tyrimo dalyviai: Iš viso buvo apklausti 214 moksleivių (100 berniukų, 114 mergaičių), besimokančių Anykščių Antano Vienuolio gimnazijoje. Tiriamųjų amžiaus vidurkis 15 ± 3metų. Tyrimo metodai: 1. Anketinė apklausa. Išvados : 1. Dauguma 12 – 17 metų moksleivių būna blogos nuotaikos, jaučia nerimą. Berniukų (11,0 proc.) ir mergaičių (18,4 proc.) pasiskirstymas panašus. 2. Dauguma 12 – 17 metų moksleivių savo sveikatą vertina kaip labai gerą. Mergaitės savo sveikatą vertino blogiau nei berniukai. Dažniausi nusiskundimai regėjimo bei laikysenos sutrikimai. 3... Physical activeness is one of the most important factors in pupil’s development and strengthening of health, but it is still not appreciated enough. Researches in Lithuania show that children are not physically active. Also, it is determined the increase of body mass, the signs of metabolic disorder, that teenagers start drinking alcohol or smoking in early age, they keep to improper diet and move not enough (Zaborskis, 1997). The aim of the research: To analyze low 12 -17 years old school children’s rates their psychological being, their health and physical activity – by gender. The goals of the research: 1. To compere 12 – 17 years boys’ and girl’s psychological being. 2. To compere how 12 -17 years old boys and girls rate their approach to their health. 3. To compere 12 – 17 boys and girls of physical activeness of level. The participants of the research: 214 pupils (100 boys and 114 girls) from Anyksciai Antanas Vienuolis Gymnasium were questioned.
