RNA polymeráza: průsečík regulačních sítí / RNA polymerase: The "meeting point" of regulatory networks

Bacterial RNA polymerase (RNAP) is a multisubunit complex essential for transcription of DNA into RNA. As a key enzyme responsible for regulation of gene expression it interprets regulatory signals from the cell and based on these cues RNAP adjusts transcription level of particular genes. This process is affected both by the regular subunits of RNAP as well as other transcription factors (TFs) directly or indirectly interacting with RNAP. The general focus of this Thesis was to extend the knowledge about the complex transcriptional regulatory networks and about the connections between individual pathways. The main specific topic and the main publication of the thesis are focused on the HelD protein, a novel binding partner of RNAP in Bacillus subtilis. We showed that HelD binds between the secondary channel of RNAP and alpha subunits of the core form of the enzyme. We proved that HelD stimulates transcription in an ATP dependent manner by enhancing transcriptional cycling and elongation. We revealed a new connection in the transcription regulatory machinery when we demonstrated that the stimulatory effect of HelD can be amplified by delta, a small subunit of RNAP specific for gram positive (G+) bacteria. Não obstante os empresários tenham autonomia para decidir pela celebração ou não de um contrato e pela determinação de seu conteúdo, tem se verificado, hodiernamente, uma crescente limitação dessa autonomia. As companhias abertas, objeto de análise do presente trabalho, resultantes da celebração de contrato de sociedade, devem observar, além das disposições aplicáveis ao direito contratual e ao direito societário, as regulamentares emanadas pela CVM. Conforme aplicável, devem observar, ainda, as normas decorrentes da autorregulação e imposições decorrentes de contratos celebrados com as entidades administradoras de mercados. Tais regras têm sua razão de ser: surgiram em contextos econômicos, sociais e políticos específicos visando a tutelar determinados valores elencados como primordiais. Assim, o presente trabalho tem por objetivo analisar as limitações à autonomia privada impostas aos empresários na constituição das sociedades anônimas, especificamente no que se refere ao estabelecimento de cláusulas estatutárias de companhias abertas. Para tanto, serão feitas análises sobre autonomia privada e suas limitações e sobre o desenvolvimento das companhias e do mercado de capitais brasileiro, permitindo-se a avaliação de quais interesses e valores foram considerados merecedores de tutela pelo ordenamento jurídico. Com esses fundamentos, será possível compreender a estrutura dos estatutos sociais das companhias abertas e quais as limitações impostas no estabelecimento das cláusulas estatutárias. / Although entrepreneurs have autonomy to celebrate or not a contract and to determine its content, it has been noted a gradually restriction of the mentioned autonomy. The publicly-held companies, object of the present study, originated from the conclusion of association contract, must observe, in addition to the contract law and corporate law provisions, the regulation issued by the Securities Commission. Besides, when applicable, publicly-held companies must be considered in accordance with self-regulation rules and other requirements as stated in listing contracts. The arise of these rules can be explained by the occurrence of specific economic, social and political contexts, which demanded the protection of certain values considered as primaries. Thus, the present dissertation aims to analyse the restrictions of the private autonomy entailed to entrepreneurs in setting up publicly-held companies, specifically when it concerns the establishment of by-laws clauses. For that reason, we will proceed to the analysis of private autonomy and its limitations, as well as the development of Brazilian publicly-held companies and capital markets, in order to examine which interests and values must be protected by judicial and institutional system. With these considerations, we expect to reach a better understading about by-laws framework and which are the restrictions imposed to the establishment of the referred clauses.

Metal and oxide nanoparticles : green biosynthesis using Portobello Mushroom Spores (PMS) for nanocomposites and replicas, properties and applications

This thesis describes novel research using Portobello mushroom spores (PMS) as a biotemplate and reducing agent that may be used to produce nanocomposites and replicas with societal benefits. First, the use of PMS as a green eco-friendly bioreagent for the biosynthesis of Ag, Au and TiOx nanoparticles (NPs) and their hybrids is described. These have been characterised using SEM, TEM, XRD, FTIR, UV, SIMS, DLS and TGA (where a full list of acronyms is to be found on page iv). The spherical shape, location and mean diameter of the PMS-held (5-30nm) Ag and Au NPs were confirmed. PMS functional groups are comparable with those in plant sources and allow them to act as reducing/capping/stabilising agents. Second, the sequential biosynthesis of PMS-held Ca2+, CaCO3 and nanohydroxyapatite (nHAp) is described where surface nucleation and growth are facilitated by biotemplate surface hydrophilic polar groups (-OH and -COO-). XRD and HRTEM suggest that the nHAp so formed has lower crystallinity and greater directed growth in the (002) direction than commercial nHAp (e.g. Bio-Oss), making PMS-routes useful for the delivery of artificial bone (after subsequent PMS removal). Furthermore, PMS-held nHAp showed more rapid in-vitro mineralization in simulated body fluid (SBF). Third, it is shown that ascorbic acid (AA), haemoglobin (Hb) and insulin (In) can be loaded into/released by PMS through its cell walls faster than with synthetic hydrogels (PVA); the transdermal use of PMS nanocomposites is considered in the context of drug delivery nanotechnology, avoiding size-dependent toxicity. Fourth, it is shown by traditional diffusion and novel methods that the antimicrobial activity especially of Ag/PMS, but also TiOx/PMS and Ag-TiOx/PMS (but not Au/PMS) is good in the inhibition of the growth of E.coli and S.aureus bacteria. The highest activity of Ag/PMS is attributed to Agx+ release. Fifth, the synthesis of NPs/PMS and biomimetic TiOx/PMS has shown to lead to good photocatalysis for the removal of coloured organic pollutants (e.g. methyl orange) from water, with faster rates of removal in molecules/mg/s and turnover numbers (TON) (s-1) than commercial anatase-rutile TiO2 (P25) possibly as the texture of the titania replicas raises the number of multiple reflections of light.

This diploma thesis deals with the presentation and disclosure requirements of non-current assets held for sale and discontinued operations in accordance with rules and requirements of IFRS. The theoretical part is divided into several areas. The most important chapter is Non-current assets held for sale, in which are detailed rules for their classification, measuring and reporting and the chapter Discontinued operations. Conclusion of the theoretical part is devoted to Czech accounting legislation, what criteria are required in connection with this issue. The practical part is focused on illustration application of IFRS 5 in selected companies. The aim of this analysis is to determine whether these companies comply with the rules and requirements of IFRS 5 in their financial statements.

I take the position that only a <em>democratic</em> world government is a desirable political goal, as it is only possible to promote individual autonomy consequently solving global problems. While accepting that there could be different answers to this condition, I analyze three approaches to the subject by Tannajo, Nielsen and Held and argue that shared sovereignty within a world federation defined by a democratic global constitution is a necessary condtion to call a world order a democratic one.</p>

NMDAR-dependent Synaptic Plasticity at the Calyx of Held Synapse

NMDARs are indispensable for developmental plasticity in the mammalian brain, but their roles in vivo are difficult to ascertain as NMDAR-knockouts are lethal. To circumvent this problem, we utilized NR1-knockdown mice to examine plasticity at the calyx of Held-MNTB synapse in the auditory brainstem. Previous work shows NMDARs at this synapse are rapidly down-regulated following the onset of hearing (P12), leading to the hypothesis that transient NMDAR expression is indispensable for modulating functional and morphological remodelling during development. We tested this by performing electrophysiological recordings, fluorescence tracing in vitro, and auditory brainstem responses in vivo, and surprisingly found that reducing postsynaptic NMDARs appears to promote functional maturation via presynaptic mechanisms in the absence of morphological and acoustic transmission differences in the mature calyx. This suggests a novel role for NMDARs to function as an activity-dependent control for setting the pace of consolidation and maturation in the calyx of Held synapse.
