DesidrataÃÃo de polpa de manga e suco de caju integral por atomizaÃÃo e caracterizaÃÃo dos produtos obtidos / Dehydration of mango pulp and cashew juice integral in atomization and characterization of products obtained

Inicialmente, avaliou-se a influÃncia das condiÃÃes de processo sobre o rendimento e a umidade desses pÃs atravÃs de planejamentos experimentais do tipo composto central rotacional completo. Foram produzidos pÃs da polpa de manga integral e do suco de caju integral que foram submetidos a uma caracterizaÃÃo fÃsico-quÃmica e higroscÃpica, sendo analisados os seguintes parÃmetros: atividade de Ãgua, umidade, pH, sÃlidos solÃveis, acidez titulÃvel, vitamina C, carotenoides, aÃÃcares redutores e totais, parÃmetros colorÃmetricos (L*, a*, b*, C* e h0), higroscopicidade, grau de caking, reidrataÃÃo e isotermas de sorÃÃo. As amostras produzidas por este processo de secagem apresentaram-se com boa qualidade, tanto fÃsico-quÃmicos quanto higroscÃpica e, para as isotermas de sorÃÃo, ambos os pÃs (manga e caju), apresentaram-se como do tipo III (Formato J), que caracteriza os alimentos com alto teor de aÃÃcar, sendo o modelo Henderson, em todas as temperaturas analisadas, o que melhor se ajustou. Em seguida, avaliou-se a estabilidade destes pÃs, em perÃodo de armazenamento de 180 dias sob duas embalagens diferentes (laminada e transparente). Assim, concluiu-se que, apÃs este perÃodo de armazenamento, tanto a polpa de manga integral quanto o suco de caju integral em pÃ, armazenados nas diferentes embalagens, ainda apresentavam-se na forma de pà e com boas caracterÃsticas fÃsico-quÃmicas e higroscÃpicas, sendo a embalagem laminada a que melhor apresentou barreira de proteÃÃo Ãs amostras. / The main aim of the study was to obtain powder of whole mango pulp and whole cashew juice by drying in a Spray-dryer. Innitially, it was made as assessment of all conditions in the process related to the output and humidity of those powders through experimental designs such as complete rotational central compound. From the results were produced powder of mango and cashew which underwent a physical-chemical and hygroscopic characterization, being analyzed the following parameters: water activity, humidity, pH, soluble solids, acidity, vitamin C, carotenoids, total and reducing sugars, colorimetric parameters (L *, a *, b *, C * and h0), hygroscopicity, degree of caking, rehydration and sorption isotherms. The samples produced by this drying process presented good quality both physicochemical and hygroscopic and, for the parameter sorption isotherms, both the powders (mango and cashew), presented as the type III (Format J) which characterizes the foods with high sugar content, and the model of Henderson, at all temperatures which were analyzed, was the best that fit. Next, were evaluated the stability of these powders in storage period of 180 days under two different packages (laminated and transparent). Thus, it was concluded that, after this storage period, both mango and cashew powder enclosed in those different packages, still were in the form of powder with good physico-chemical and hygroscopic characteristics, being the laminated packaging the one which best showed the barrier protection to the samples.

The aim of this research was to evaluate the biology and infestation levels of A. woglumi in three host plants (orange Pêra, C. sinensis, acid lime Tahiti, Citrus latifolia and mango, M. indica) and biometry evaluations. For this, experiments were assembled in the period of January to June of 2006 at laboratory conditions. It was verified that acid lime Tahiti had been shown as the most suitable host for A. woglumi. Orange Pêra and mango are similar, being the citrus genus the most favorable to the complete development of A. woglumi. Mango is an appropriate host for A. woglumi. The duration of embrionary development was 15 days on average for the three hosts. The fourth nymph stadium was the longest when compared to the other stages of evolutive cycle. The higher viabilities were on third nymph stages, eggs and second nymph stages. The duration of egg-adult cycle was 70 days on average (2 months and 10 days) for the three hosts evaluated. / A mosca-negra-dos-citros (Aleurocanthus woglumi Ashby) uma importante praga dos citros de origem asiática, foi detectada no Brasil em 2001. O objetivo deste estudo foi avaliar a biologia e os níveis de infestação de A. woglumi em três plantas hospedeiras (laranja Pêra, C. sinensis; lima ácida Tahiti, Citrus latifolia e manga, M. indica) e avaliações de biometria. Para isto, foram montados experimentos no período de janeiro a junho de 2006 em condições de laboratório. Foi verificado que lima ácida Tahiti se mostrou o hospedeiro mais favorável para A. woglumi. Laranja Pêra e manga assemelham-se; sendo que o gênero Citrus se mostrou mais favorável ao desenvolvimento completo de A. woglumi. Manga é um hospedeiro adequado para A. woglumi. A duração do desenvolvimento embrionário foi de 15 dias em média para os três hospedeiros. O estádio de ninfa 4 (pupário) foi o mais longo quando comparado com as outras fases do período desenvolvimento. As maiores viabilidades foram nas fases de ninfa 3, ovos e ninfa 2. A duração do ciclo ovo-adulto foi de 70 dias em média (2 meses e 10 dias) para os três hospedeiros avaliados.

She was the first women to receive a D.Sc. from Unisa'a predecessor, the University of the Cape of Good Hope in 1914 / This thesis researched a mango disease which caused considerable loss to mango growers in South Africa at the beginning of the twentieth century, particularly around Barberton, Warm baths and the coastal region of Natal. The disease caused dark angular spots on the leaves. This did not noticeably affect the general health of the tree, but served as a source of infection for the fruit. The infected fruit is detached from the tree by the slightest air movement and falls rotting to the ground. The infection is carried by wind and rain. Several spraying experiments were carried out at Barberton in an orchard placed at the author's disposal by Messrs Winter Brothers during an exceptionally dry season, which showed that spraying with Bordeaux mixture, iron sulphide or Hyco/ was useless in checking the disease. The disease had not been described in the literature before. According to the author, the cause of the disease was a flagellate bacillus Bacillus mangiferae. It invades the parenchyma, wedging apart and killing the cells and causing gummosis, but it does not touch the lignified tissues. El objetivo de esta tesis fue determinar las propiedades antihiperglicemiantes del liofilizado del extracto acuoso de alta temperatura de la corteza del tronco de Mangifera indica (EMI), en ratas hiperglicémicas agudas. Determinamos el efecto antihiperglicemiante utilizando tres concentraciones (50, 250 y 750 mg/Kg/ml.) de EMI en un modelo de hiperglicemia aguda in vivo en ratas normoglicémicas. Se midió la evolución temporal de la glicemia tras la administración de EMI (30 minutos antes de la glucosa) por vo, utilizando un sistema de bioamperometría. / The objective of this thesis was to determine the antihyperglycemic properties of the lyophilization of the aqueous extract of high temperature of the crust of the trunk of Mangifera indica (EMI) in acute hyperglycemic rats. We determined the antihyperglycemic effect using three concentrations (50, 250 and 750 mg/Kg/ml.) of EMI in an acute model of hyperglycemia in live normoglycemic rats. The temporary evolution of glycemia mediated after the administration of EMI (30 minutes before the glucose) orally, using a bioamperometry system.

The Evolution and Domestication Genetics of the Mango Genus, Mangifera (Anacardiaceae)

Domesticated species are vital to global food security and have also been foundational to the formulation and advancement of evolutionary theory. My dissertation employs emerging molecular genomic tools to provide an evolutionary context for crop improvement. I begin by providing a contemporary perspective on two components of domestication biology that have long been used to improve crop production: wild relatives of crop species and grafted rootstocks. First, I propose a method to systematically introgress crop wild relative diversity into crop breeding programs. Then, I explore rootstocks, the lesser-known half of the perennial crop equation, documenting prevalence and diversity, cataloging rootstock traits under selection, and discussing recent advances in rootstock biology. Both crop wild relatives and rootstocks remain largely underutilized resources and hold great promise for agricultural innovation. While humans have domesticated thousands of plant species, research has largely focused on annual crops, to the exclusion of perennials. To improve our understanding of how tree species respond to domestication, I examine the evolution and domestication of one of the world’s most important perennial tropical fruit crops, the mango, Mangifera indica, and its wild and semi-domesticated relatives. I generated a dataset suitable for studying Mangifera across evolutionary time using double digest restriction site associated DNA sequencing (ddRADseq). I present a multilocus phylogeny that informs the classification of Mangifera and reveals, for the first time, the evolutionary relationships of wild, semi-domesticated, and domesticated species in the genus. Narrowing my focus to the intraspecific level, I examine how the introduction of M. indica into regions of the world impacted its genetic diversity. My results show M. indica maintained high levels of genetic diversity during its introduction into the Americas. However, the novel diversity I detect in Southeast Asian mango cultivars suggests that M. indica has a more complex domestication history than previously assumed. I also find evidence that M. indica hybridized with multiple congeners following its introduction into Southeast Asia, forming two hybrid lineages that may be maintained by clonal polyembryonic reproduction. Collectively, my research provides a comprehensive framework for understanding the evolution and domestication of a tropical tree crop of global economic importance.

Embora esforços têm sido empregados na seleção e identificação de materiais resistentes à doença, a base genética da resistência à murcha-de-Ceratocystis em mangueira, informação essencial para direcionar programas de melhoramento que visem desenvolver cultivares resistentes, não é conhecida. Nos estudos sobre o controle genético da resistência, devem-se usar preferencialmente famílias segregantes oriundas de cruzamentos controlados. No entanto, em mangueira, o pequeno tamanho das flores associado à alta abscisão dos frutos torna os cruzamentos manuais muito trabalhosos e com baixo rendimento. Assim, tem-se adotado o uso de polinização aberta a qual pode apresentar indivíduos contaminantes ou oriundos de autofecundação. Portanto, o presente estudo teve como objetivos: 1) confirmar por meio de marcadores microssatélites a origem híbrida de seis progênies obtidas via cruzamentos naturais; 2) estudar o controle genético da resistência à murcha-de-Ceratocystis em mangueira. Na primeira parte, plantas isoladas das cultivares Coquinho, Espada, Haden, Keitt, Palmer e Van Dyke foram identificadas dentro de pomares comerciais estabelecidos com a cultivar Tommy Atkins. Frutos de cada uma destas seis cultivares foram colhidos para a produção das mudas que posteriormente foram levadas para o campo. Folhas das progênies e dos genitores foram coletadas, o DNA foi extraído e a caracterização dos híbridos foi feita utilizando seis marcadores microssatélites. A percentagem de híbridos confirmados nos cruzamentos (33 a 89 %) foi maior que a relatada na literatura usando polinização manual e a estratégia adotada mostrou-se eficiente na obtenção e caracterização dos híbridos. Na segunda parte do estudo, avaliou-se a resistência à murcha-de- Ceratocystis nas cultivares Coquinho, Espada, Haden, Keitt, Palmer, Van Dyke e Tommy Atkins e estudou-se o controle genético da resistência à doença em 197 plantas derivadas de cruzamentos de cada uma dessas cultivares com a “Tommy Atkins”. As cultivares Keitt, Palmer, Tommy Atkins e Van Dyke foram mais resistentes e as cultivares Coquinho, Espada e Haden mais suscetíveis à doença. Os resultados deste estudo mostram que a herança da resistência à murcha-de-Ceratocystis em mangueira é poligênica com uma prevalência de genes com efeitos de dominância e epistasia. O ganho genético obtido com a seleção das 10 melhores plantas, ou seja, as mais resistentes, foi de 46%, o que significa uma redução de 46% na severidade da doença. Em geral, observou-se baixa frequência de alelos favoráveis à resistência a doença na população estudada o que indica a necessidade de introdução de novos materiais genéticos. / The mango (Mangifera indica L.) is one of the most consumed tropical fruit in the world and Ceratocystis wilt caused by Ceratocystis fimbriata Ellis & Halsted is a limiting factor for its production. Although efforts have been employed in the selection and identification of materials resistant to Ceratocystis wilt, the genetic basis of resistance to the disease in mango is unknown. This information is essential to guide breeding programs aiming the development of resistant cultivars. In studies on the genetic control of resistance, segregating families, derived from controlled crossings, should be used, preferably. However, the small size of mango flowers associated with a high fruit abscission make artificial crossings laborious and with low yield. Therefore, this study aimed to: 1) to confirm by microsatellite markers the hybrid origin of six progenies obtained via natural crossings; 2) study the genetic control of resistance to Ceratocystis wilt in mango. In the first part of this work, isolated plants of cultivars Coquinho, Espada, Haden, Keitt, Palmer and Van Dyke were identified in commercial orchards established with the cultivar Tommy Atkins. Open pollinated progenies from each cultivar were planted in the field. Leaves from these progenies and parents were collected, DNA was extracted and the characterization of the hybrids was done using six microsatellite markers. The percentage of confirmed hybrids for the crossings (33-89%) was higher than expected by manual pollination, which indicates that the adopted strategy is efficient in obtaining and characterization of the hybrids. In the second part of the work, we evaluated the resistance to Ceratocystis wilt in the cultivars Coquinho, Espada, Haden, Keitt, Palmer, Van Dyke and Tommy Atkins. Subsequently, we studied the genetic control of resistance to the pathogen on 197 plants derived from crossings between the remaining cultivars and “Tommy Atkins”. The cultivars Keitt, Palmer, Tommy Atkins and Van Dyke were more resistant and the cultivars Coquinho, Espada and Haden were more susceptible. The results of this study show that the inheritance of resistance in mango is polygenic with a prevalence of genes with effects of dominance and epistasis. The genetic gain by selecting the ten more resistant plants was 46%, which means a 46% reduction in disease severity. In general, there was a low frequency of favorable alleles for resistance to the disease in the population studied indicating the need to introduce new sources of genetic materials.

It has high nutritional and commercial value, being used for the production of juices, sweets, jams, ice creams etc, and for fresh consumption. It is a rich source of bioactive compounds, among which stand out the polyphenols, carotenoids and vitamin C. This study aimed to evaluate the anti-inflammatory and antioxidant of hydroalcoholic extracts of peel and pulp variety of mango 'Tommy Atkins' against oxidative damage in the gastric injury model induced by naproxen. Mangoes were acquired in the ripe stage, separate the peel and the pulp, and freeze-dried these parts. The following procedures were performed: hydroalcoholic extract of lyophilized peel and pulp; Quantification of bioactive compounds; the antioxidant property of the compounds of hydroalcoholic extracts of mango peel and pulp; evaluating the cytotoxicity of the extracts in vitro analysis (MTT test); Identification of phenolic compounds present in the extracts by analysis by ultra performance liquid chromatography (UPLC®-QTOF); and antioxidant in vivo evaluation of hydroalcoholic extracts of mango peel and pulp by determining of peroxidation levels of membrane lipids, with the quantification of malondialdehyde (MDA), determination of myeloperoxidase activity (MPO) and quantification of levels reduced glutathione (GSH). The extracts have a high content of bioactive compounds, contributing to its antioxidant activity. The cytotoxicity test showed that the extracts have no toxic effect to cells (IEC-6 culture). It was detected by ultra-performance liquid chromatographic analysis (UPLC®-QTOF), the presence of compounds that provide important antioxidant activity, such as quercetin, mangiferin, and gallic acid. This can be shown by in vivo analysis in an experimental model of a naproxen-induced gastric injury, where the hydroalcoholic extracts of mango peel and pulp were able to significantly reduce the levels of MPO and MDA, and to conserve GSH levels. Therefore, by presented antioxidant activity in the result of pretreatment with hydroalcoholic extracts of mango peel and pulp, it was concluded that both protected the gastric mucosa against injury caused by oxidative agents triggered using naproxen, significantly reducing the lesions, which contributes to an anti-inflammatory action. / A manga (Mangifera indica L.) é uma fruta tropical originária da Índia, adaptada à região Nordeste do Brasil desde o século XVI. Possui elevado valor nutricional e comercial, sendo utilizada para produção de sucos, doces, compotas, sorvetes etc, e para consumo in natura. É uma rica fonte de compostos bioativos, dentre os quais se destacam os polifenóis, os carotenóides e a vitamina C. Neste estudo, objetivou-se avaliar o efeito anti-inflamatório e o antioxidante dos extratos hidroalcoólicos da casca e da polpa da manga variedade ‘Tommy Atkins’ contra danos oxidativos no modelo de lesão gástrica induzida por naproxeno. Foram adquiridas mangas no estádio de maturação maduro, separadas as cascas da polpa e liofilizadas essas partes. Os seguintes procedimentos foram realizados: extração hidroalcoólica de casca e de polpa liofilizadas; quantificação dos compostos bioativos; determinação da propriedade antioxidante dos compostos dos extratos hidroalcoólicos de casca e polpa de manga; avaliação da citotoxicidade dos extratos em análise in vitro (teste do MTT); identificação dos compostos fenólicos presentes nos extratos através de análise por cromatografia líquida de ultra performance (UPLC®-QTOF); e avaliação antioxidante in vivo dos extratos hidroalcoólicos da casca e da polpa de manga através da determinação do grau de peroxidação dos lipídios da membrana, com a quantificação do malondialdeído (MDA), determinação da atividade da enzima mieloperoxidase (MPO) e quantificação dos níveis de glutationa reduzida (GSH). Os extratos obtidos apresentam elevado conteúdo de compostos bioativos, contribuindo para sua atividade antioxidante. O teste de citotoxidade demonstrou que os extratos não possuem efeito tóxico às células (cultura IEC-6). Foi detectada, através da análise cromatográfica (UPLC®-QTOF), a presença de compostos importantes que propiciam atividade antioxidante, como quercetina e mangiferina e ácido gálico. Esta pode ser comprovada através da análise in vivo, no modelo experimental de lesão gástrica induzida por naproxeno, onde os extratos hidroalcoólicos da casca e da polpa de manga foram capazes de reduzir de forma significativa os níveis de MPO e de MDA, e de preservar os níveis de GSH. Portanto, através da atividade antioxidante apresentada no resultado do pré-tratamento com os extratos hidroalcoólicos de casca e de polpa de manga, conclui-se que ambos protegeram a mucosa gástrica contra injúrias causadas por agentes oxidativos desencadeados pelo uso do naproxeno, reduzindo significativamente as lesões, o que contribui para uma ação anti-inflamatória.

Aplicação de calcário e seus efeitos no sistema radicular, estado nutricional, produção e qualidade tecnológica de frutos de mangueiras

O estudo foi realizado de março/2005 a fevereiro de 2008, em um Latossolo Vermelho distrófico, em um pomar implantado em 1992, espaçamento de 10 x 10 m, na Fazenda de Ensino, Pesquisa e Extensão da Faculdade de Engenharia/UNESP, Campus de Ilha Solteira, localizada no Município de Selvíria-MS. O delineamento experimental utlilizado foi em blocos casualizados, com 5 tratamentos, 4 repetições na ‘Haden’ e 3 na ‘keitt’, com 5 e 4 plantas por parcela, respectivamente, nas duas variedades. Os tratamentos foram compostos de doses crescentes de calcário, tomando-se como referência a dose calculada (3,1 t ha-1), como segue: T1 = testemunha, sem aplicação de calcário; T2 = metade da dose; T3 = a dose total; T4 = 1,5 vezes a dose e T5 = 2 vezes a dose calculada, correspondendo respectivamente a zero; 1,55; 3,10; 4,65 e 6,20 t ha-1. Foram avaliadas duas colheitas consecutivas de cada variedade (2006 e 2007 na mangueira ‘Haden’, 2007 e 2008 na ‘Keit’) e realizadas amostragens de folhas em quatro épocas diferentes. A calagem promoveu melhoria dos atributos químicos do solo (pH, Ca, Mg, SB, CTC, V (%) e H+Al), com efeitos significativos na produtividade, número de frutos planta, acidez titulável, sólidos solúveis e pH de frutos, havendo aumento nos teores foliares de Ca e Mg e diminuição nos teores de K, Fe, Mn e Zn. No sistema radicular promoveu aumento de 15,73% de raízes de absorção nas plantas que receberam calcário em relação à testemunha. As raízes de absorção de maior ocorrência foram aquelas com diâmetros menores do que 2 mm. / The present work aimed to evaluate the surface lime application, in the clay soil of the cerrado region, and to observe the effects in the production, fruits quality and nutrition state of two mangoes variety, Haden and Keitt, grafted on the variety Coquinho, as well to evaluate the distribution of the root system of the mangoes Haden variety in plants that received no calcareous and plants that received calcareous. The study was conducted between March 2005 and February 2008, in a dystrophic Red Latosol (Haplustox), in an orchard implanted in 1992, spacing of 10 x 10 m, at Experimental Station/UNESP/Ilha Solteira, located in Selvíria-MS. The experimental design was randomized block, with 5 treatments, 4 replicates in the ‘Haden’ and 3 in the ‘Keitt’, with 5 and 4 plants by parcel, respectively. The treatments were constituted by increasing doses of liming, taking as reference of the recommended dose (3.1 t of lime ha-1) as follows: T1 = zero; T2 = half of the recommended dose; T3 = a time the recommended dose; T4 = 1.5 times the recommended dose e T5 = 2 two times the recommended dose, corresponding to zero; 1.55; 3.10; 4.65 e 6.20 t ha-1, respectively. Two harvests of each variety (2006 e 2007 in the ‘Haden’, 2007 and 2008 in the ‘Keitt’) were evaluated and the leaves were sampled in four separate periods of time. Lime application promoted an improvement on the chemical properties of the soil (pH, Ca, Mg, SB, CTC, V (%) e H+Al), with significant effects on productivity, number of fruits by plants, soluble solid, acidity and pH of fruits, having increased in the tenors of Ca and Mg and reduced in the tenors of K, Fe, Mn and Zn on the leaves. In the root system, the plants that received lime presented 15.73% of the roots of absorption more than plants no lime. The roots of absorption of bigger occurrence had a smaller diameter by 2mm.
