The aim of this study was to evaluate antiketogenic prophylaxis of the veterinary medicine Kexxtone, in breeding of Holstein cows, in the company Agrodam Hořepník, s.r.o. There were then evaluated the parameters of a metabolic state in the herd monitored. Furthermore I evaluated the milk yield, amount of fats, proteins, lactose and somatic cells in the milk during 100 days of lactation and subsequently, the level of pregnancy after the 1st and the 2nd insemination was evaluated. The observation took place from July 2014 to December 2014 in a way that every two months was selected 8 cows and 8 heifers with a similar calving date. After that they were split into two groups i.e. into the experimental group (the kexxtone medicine was used) and into the control one. These cows were bled and milked. Subsequently, those biological samples were sent to a laboratory to be diagnosed. The higher milk yield was found within the experimental group and so as the content of milk constituents, i.e. fats and proteins. A beneficial effect of this antiketogenic medicine on the amount of ketones in blood and milk of animals was also confirmed within the experimental group. There were not found any significant statistical differences in other monitored parameters.

Para avaliar o metabolismo oxidativo, perfil bioquímico e o efeito da suplementação com vitamina E intramuscular no período periparto, foram utilizadas 24 ovelhas, hígidas, da raça Santa Inês, no último mês de gestação. As ovelhas foram distribuídas em dois grupos que receberam, com intervalo de 14 dias, duas aplicações pela via intramuscular profunda de 2 mL de solução fisiológica (grupo controle-GC), ou 200 UI de vitamina E (grupo tratado-GT). Estes grupos foram subdivididos em P1 e P2. No P1, as ovelhas receberam a segunda dose de solução fisiológica ou vitamina entre 1 e 7 dias da data do parto. No P2, a segunda dose foi administrada entre 15 e 25 dias da data do parto. As amostras de sangue foram coletadas nos seguintes momentos: previamente à primeira aplicação (M0), 15 dias após a primeira aplicação (M1), no momento do parto (M2), 7 dias após o parto (M3), duas semanas após o parto (M4) e 4 semanas após o parto (M5). Foram analisadas as variáveis do perfil bioquímico: proteína total, albumina, globulina, uréia, creatinina, creatinofosfoquinase (cK), ácido úrico, aspartatoaminotransferase (AST), gamaglutamiltransferase (GGT), colesterol, triglicérides, glicose, beta hidroxibutirato (BHB) , ácidos graxos não esterificados (NEFA). Do metabolismo oxidativo foram determinas as atividades da superóxido dismutase (SOD), glutationa peroxidase (GSH-Px), glutationa reduzida (GSH) e habilidade de redução férrica plasmática (HRFP). Em P1, não foram observadas diferenças entre os grupos tratado e controle nas concentrações de proteína total, globulina, cK, ácido úrico, glicose, triglicérides, BHB, NEFA, SOD, GSH-Px e GSH. Porém em P1, foram observadas maiores concentrações em de albumina em M0 (P=0,039); uréia em M1 (P=0,018), M2 (P=0,005) e M3 (P=0,040); a creatinina em M2 (P=0,030) e M3 (P=0,047); GGT em M1 (P= 0,01) e M2 (P=0,024); colesterol em M2 (P=0,041) e HRFP em M3 (P= 0,022) para as ovelhas tratadas em relação às controle. Em P1, a AST foi maior para o controle em relação ao tratado em M2 (P=0,030). Em P2, foram observadas maiores atividades para o grupo controle nas variáveis SOD em M3 (P=0,013) e GSH-Px em M4 (P=0,027) e maior HRFP em M4 (P=0,023) para o grupo tratado. A aplicação de duas doses de vitamina E (200 UI, via IM) aumentou as concentrações de HRFP no pós-parto tanto em P1 como em P2. / Oxidative metabolism, biochemical profile and the effect of intramuscular vitamin E supplementation on sheeps per partum period were evaluated using 24 healthy Santa Ines sheep, in the last month of pregnancy. The lambs were randomly divided into two groups, control group-CG and treated group-TG. CG received 2 mL of saline and TG received 200 IU of vitamin E, both treatments were done with two doses within interval of 14 days, by deep intramuscular injection of. These groups were further divided into P1 and P2. In P1 the sheep received the second or vitamin saline dose between 1 and 7 days before delivery date. In P2, the second dose was administered between 15 and 25 days before delivery. Blood samples were collected at the following times: before the first application (M0), 15 days after (M1), at birth (M2), 7 days postpartum (M3), two weeks after delivery (M4) and four weeks after delivery (M5). The variables of biochemical profile analyzed were: total protein, albumin, globulin, urea, creatinine, creatine kinase (CK), uric acid, aspartate aminotransferase (AST), gamma-glutamyltransferase (GGT), cholesterol, triglycerides, glucose, beta hydroxybutyrate (BHB), acids non-esterified fatty acids (NEFA). Oxidative metabolism variable were: activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), reduced glutathione (GSH) and ferric reducing ability of plasma (HRFP). No differences were observed between the P1 treated and control groups in the concentrations of total protein, globulin, CK, uric acid, glucose, triglycerides, BHB, NEFA, SOD, GSH-Px and GSH. However higher concentrations of albumin in M0 (P = 0.039), urea in M1 (P = 0.018), M2 (P = 0.005) and M3 (P = 0.040), creatinine in M2 (P = 0.030 ) and M3 (P = 0.047), GGT in M1 (P = 0.01) and M2 (P = 0.024), cholesterol in M2 (P = 0.041) and HRFP at M3 (P = 0.022) for ewes treated we observed. AST concentration was greater for control in M2 (P = 0.030). In P2, higher activities were observed for the control group in the variables SOD in M3 (P = 0.013), GSH-Px in M4 (P = 0.027) and higher FRAP in M4 (P = 0.023) for the treated group. The application of two doses of vitamin E (200 IU, im) increased the concentrations of FRAP postpartum in both P1 and P2.

This thesis presents Pathway Informed Analysis (PIA), a classification method for predicting disease states (diagnosis) from metabolic profile measurements that incorporates biological knowledge in the form of metabolic pathways. A metabolic pathway describes a set of chemical reactions that perform a specific biological function. A significant amount of biological knowledge produced by efforts to identify and understand these pathways is formalized in readily accessible databases such as the Kyoto Encyclopedia of Genes and Genomes. PIA uses metabolic pathways to identify relationships among the metabolite concentrations that are measured by a metabolic profile. Specifically, PIA assumes that the class-conditional metabolite concentrations (diseased vs. healthy, respectively) follow multivariate normal distributions. It further assumes that conditional independence statements about these distributions derived from the pathways relate the concentrations of the metabolites to each other. The two assumptions allow for a natural representation of the class-conditional distributions using a type of probabilistic graphical model called a Gaussian Markov Random Field. PIA efficiently estimates the parameters defining these distributions from example patients to produce a classifier. It classifies an undiagnosed patient by evaluating both models to determine the most probable class given their metabolic profile.
We apply PIA to a data set of cancer patients to diagnose those with a muscle wasting disease called cachexia. Standard machine learning algorithms such as Naive Bayes, Tree-augmented Naive Bayes, Support Vector Machines and C4.5 are used to evaluate the performance of PIA. The overall classification accuracy of PIA is better than these algorithms on this data set but the difference is not statistically significant. We also apply PIA to several other classification tasks. Some involve predicting various manipulations of the metabolic processes performed in experiments with worms. Other tasks are to classify pigs according to properties of their dietary intake. The accuracy of PIA at these tasks is not significantly better than the standard algorithms.

The aim of the thesis was the evaluation of commercial properties, physiological state and selected blood parameters in sheep suffolk breed with focus on monitoring increases, blood parameters and coprological examination which prooved parasitological settlement. Monitoring was conducted in 2013 and early 2014 when 17 ewes and 16 lambs were observed. For thesis purpose two samples were made on 25.3.2013 and 14.10.2013. Several elevated values have been detected among which were glucose, urea, total protein, albumin, 1 and 2 globulin and partially cholesterol. ß globulin was below the physiological range, globulin was initially below the physiological range, but in the second sampling was above the physiological range. Decreased values were observed in case of calcium concentration in blood plasma. The parasitological findings prooved presence of coccidia Eimeria and gastrointestinal nematodes.

Sensibilidade à insulina e metabolismo oxidativo em cordeiros provenientes de ovelhas suplementadas com vitamina E no período pré-parto / Insulin sensitivity and oxidative metabolism in lambs born from ewes supplemented with vitamin E in the peripartum periodCarvalho, Débora Dias de 06 March 2018 (has links)
O objetivo deste trabalho foi estudar os efeitos da administração parenteral de vitamina E no desempenho produtivo e na sensibilidade à insulina de cordeiros nascidos de ovelhas suplementadas ou não com vitamina E no 108º dia de gestação. Foram utilizados 28 cordeiros em delineamento de blocos casualizados, considerando o sexo dos cordeiros e tipo de gestação, se única ou gemelar; com planejamento fatorial 2X2, em que foram considerados cordeiros de ovelhas que receberam ou não vitamina E e cordeiros que receberam ou não vitamina E na dosagem de 60 UI/kg PV até 24 horas após o nascimento, distribuindo em quatro grupos: EE: Cordeiros e ovelhas que receberam vitamina E; EC: Cordeiros que receberam veículo oleoso (controle) nascidos de ovelhas que receberam vitamina E; CC: Cordeiros e ovelhas que receberam veículo oleoso; CE: Cordeiros que receberam vitamina E nascidos de ovelhas que receberam veículo oleoso. Para detecção da sensibilidade à insulina, aos 28 dias de vida (T1) os cordeiros foram submetidos ao teste de tolerância à glicose (TTG). Foram determinados aos dois (T0) e aos 28 (T1) dias de vida as atividades de CK, AST, GGT, SOD, GSH-Px; o status antioxidante total (TAS); a habilidade de redução do ferro plasmático (HRFP), as substâncias reativas ao ácido tiobarbitúrico (TBARs); as concentrações de ácido úrico, glicose, ácidos graxos não esterificados (AGNE), betahidroxibutirato (BHB), triglicerídeos, colesterol, cálcio total, cálcio ionizável, proteína total, albumina, ureia, creatinina e a concentração plasmática de vitamina E (alfa-tocoferol). Foram calculados os valores de RQUICKI e de RQUICKI BHB em T0 e em T1. Semanalmente foram acompanhados o peso e o escore de condição corporal dos cordeiros (ECC). Os dados foram analisados utilizando-se o pacote estatístico STATISTICAL ANALYSIS SYSTEM (SAS) e para todos os testes foi adotado o nível de significância de 5%. A administração parenteral de 60 UI/kg de vitamina E em ovelhas no 108o dia de gestação e/ou nas primeiras 24 horas de vida de cordeiros não influenciou as concentrações de cálcio total, proteína total, ureia, albumina, TAS, cortisol, insulina; as concentrações plasmáticas de glicose, colesterol, triglicérides, AGNE, HRFP; as atividades de AST, GGT, GSH-Px e SOD; e os valores de RQUICKI, peso e ECC. Foi observada diminuição da sensibilidade à insulina e concentração de alfa tocoferol no T0 para cordeiros nascidos de ovelhas suplementadas com vitamina E. A suplementação dos cordeiros melhorou o status antioxidante no T0 e a produção de BHB no T1. A suplementação com alfa tocoferol não influenciou no peso vivo e no ECC até a quarta semana de vida do cordeiro. / The objective of this work was to study the effects of parenteral administration of vitamin E on the productive performance and insulin sensitivity of lambs born from ewes supplemented or not with vitamin E on the 108th day of gestation. Twenty-eight lambs received intramuscularly 60 UI/kg BW of vitamin E or oily vehicle until 24 hours after birth and were distributed according to their sex and type of pregnancy (single or twin) into four groups using a 2x2 factorial design: EE: lambs and ewes treated with vitamin E; EC: Lambs that received oily vehicle (control) born from ewe treated with vitamin E; CC: Lambs and ewe that received oily vehicle; EC: Lambs that received vitamin E born from ewe treated with the oily vehicle. At 28 days old, all lambs were submitted to the glucose tolerance test (GTT). At 28 days old, all lambs were submitted to the glucose tolerance test (GTT) to detect insulin sensitivity. The activities of CK, AST, GGT, SOD, GSH-Px; total antioxidant status (TAS); the ability to reduce plasma iron (HRFP), thiobarbituric acid reactive substances (TBARs); the concentrations of uric acid, glucose, non-esterified fatty acids (NEFA), betahydroxybutyrate (BHB), triglycerides, cholesterol, total calcium, ionizable calcium, total protein, albumin, urea, creatinine and plasma concentration of vitamin E (alpha-tocopherol) were determined at 2 (T0) and at 28 (T1) days of life. The RQUICKI and RQUICKI BHB values were calculated in T0 and T1. The body condition score (BCS) and the live weight were measured every week. The data were analyzed using the statistical package STATISTICAL ANALYSIS SYSTEM (SAS) and for all the tests was adopted the level of significance of 5%. Parenteral administration of 60 IU / kg vitamin E in sheep on the 108th day of gestation and / or during the first 24 hours of lamb life did not influence the concentrations of total calcium, total protein, urea, albumin, TAS, cortisol, insulin; plasma concentrations of glucose, cholesterol, triglycerides, NEFA, FRAP; the activities of AST, GGT, GSH-Px and SOD; and the RQUICKI, weight and BSC values. There was only effect related to the age of the lambs for these variables. Decreased insulin sensitivity and increased alpha tocopherol concentration was observed in T0 for lambs born from ewe supplemented with vitamin E. The supplementation of the lambs improved the antioxidant status in T0 and the BHB production in T1. Alfa Tocopherol supplementation did not influence the live weight and BSC until the fourth week of life of the lambs.

Desempenho de cabritas Saanen submetidas a diferentes níveis de energia na dieta: perfil metabólico e hormonal / Performance of Saanen female kids submitted to different levels of dietary energy: hormonal and metabolic profileBomfim, Gabriela Facholi 14 December 2012 (has links)
Considerando que a quantidade de energia ingerida pelo animal pode antecipar ou retardar o início da puberdade, este estudo tem por objetivo avaliar como diferentes níveis de energia influenciam o perfil metabólico, hormonal, consumo de alimento e o peso vivo de cabritas da raça Saanen em início de puberdade (entre 227 dias de idade e peso vivo de 28,38 ± 0,91kg até 304 dias de vida e peso vivo de 39,12 ± 1,47). Desse modo, as 21 cabritas experimentais foram submetidas a três dietas, sendo: T1) dieta controle, na qual atendeu 100% do requerimento dos animais (contendo níveis nutricionais preconizados segundo o NRC (2006)); T2) decréscimo de 20% de óleo de soja em relação à dieta controle e T3) acréscimo de 20% de óleo de soja em relação à dieta controle. Durante o período experimental foram realizadas pesagens e colheita de sangue semanalmente avaliação do desempenho dos animais bem como, de suas condições fisiológicas e reprodutivas, sendo essas obtidas através da dosagem de metabólitos (glicose, colesterol, triglicerídeo, proteína, ureia) e hormônios (progesterona (P4) e estradiol (E2). Ao avaliar o desempenho dos animais verificou-se que o tratamento controle apresentou peso vivo final e consumo da matéria seca superior (p≤0,05) aos demais tratamentos, sendo o tratamento com acréscimo de 20% de óleo de soja em relação à dieta controle apresentou pior desempenho. Porém, ao analisar as médias dos hormônios E2 e P4 ao final do período experimental foi constatado que os animais que receberam dieta com acréscimo de energia tiveram médias superiores (p≤0,05) aos demais quanto à concentração plasmática de progesterona, percentual de ovulação (considerando níveis de P4 acima de 1 ng/ml) e percentual de manifestação de estro (concentração de E2 acima de 30 pg/ml). Quanto aos metabólitos, os animais que foram submetidos ao decréscimo de energia em sua dieta apresentaram níveis de ureia e colesterol superiores (p≤0,05) aos da dieta com acréscimo de energia. Contudo, com base nos resultados obtidos conclui-se que o ideal é manter a dieta controle, pois os índices para percentual de estro, ovulação e perfil metabólico são satisfatórios, bem como o consumo e peso vivo final dos animais. / Whereas the amount of energy intake by the animal can advance or delay the start of puberty, this study has for objective evaluate how different energy levels influence the metabolic profile, hormonal, food intake and body weight of female goat kids Saanen in early puberty (between 227 days of age and body weight of 28.38 ± 0.91 kg to 304 days of age and body weight of 39.12 ± 1.47). In evaluating the performance of the animals was found that the control treatment showed higher final body weight and intake of dry matter (p ≤ 0.05) than other treatments, the treatment with an increase of 20% soybean oil in relation to diet control showed poorer performance. However, when analyzing the means of hormones E2 and P4 at the end of the experimental period it was found that animals fed the diet with an increase of energy had higher mean (p ≤ 0.05) to the other as the plasma progesterone concentration, percentage of ovulation (P4 levels more than 1 ng / ml) and percentage of expression of oestrus (E2 concentrations more than 30 pg / ml). As for metabolites, animals that were subjected to the decrease of energy in their diet had higher levels of urea and cholesterol (p ≤ 0.05) to the diet with an increase of energy. However, based on the results obtained gives to conclude that the ideal is to maintain the control diet, because the indices for percentage of oestrus, ovulation and metabolic profiles are satisfactory, and the consumption and final body weight of the animals.
