Détection électrochimique de gradients de concentration ou de gouttes générés à l'intérieur d'un canal microfluidique : approche théorique et expérimentale / Electrochemical detection of concentration gradients and droplets generated within microfluidic channel : theoretical and experimental approaches

L'électrochimie permet la mise en œuvre de techniques de détections pertinentes et adaptées aux contraintes de miniaturisation imposées par la conception de laboratoires sur puce. L'objectif de la thèse a été d'étudier la détection d'espèces électroactives circulant à l'intérieur d'un canal microfluidique, sous la forme de gradients de concentration localisés, ou d'espèces contenues à l'intérieur de gouttes. Pour cela, deux approches ont été menées au moyen d'électrodes microbandes intégrées dans des microcanaux. La première a été d'étudier la possibilité de générer puis de détecter électrochimiquement de façon contrôlée des gradients de concentration en écoulement monophasique. Les réponses ampérométriques ont été analysées en fonction des caractéristiques des gradients de concentration après les phases de génération et de propagation. Deux comportements limites ont été mis en évidence par simulations numériques puis vérifiés expérimentalement. La seconde approche a été de mettre en œuvre une détection électrochimique du contenu de gouttes en écoulement diphasique. L'enjeu a été à la fois de démontrer la faisabilité des mesures mais aussi d'établir des relations entre les courants mesurés et les concentrations ou quantités d'espèces à l'intérieur des gouttes. Dans ce cadre, un microdispositif innovant a été proposé puis testé expérimentalement, démontrant la possibilité d'effectuer des électrolyses totales de gouttes. / Electrochemistry enables the implementation of relevant and appropriate detection techniques to the miniaturization constraints imposed by the design of labs-on-a-chip. The aim of this thesis was to study the detection of electroactive species flowing within microfluidic channels under the form of concentration gradients or microdroplets. Therefore, two approaches were undertaken by means of microband electrodes integrated within microchannels. The first one was to study the opportunity to control the electrochemical generation and detection of concentration gradients in continuous flow. The amperometric responses were analyzed as a function of the characteristics of concentration gradients after the generation and propagation processes. Two boundary behaviours were evidenced by numerical simulations and validated experimentally. The second approach was to implement the electrochemical detection of droplet content in segmented flow. The challenge was both to demonstrate the feasibility of the experiments and to introduce relationships between currents and concentration or amount of species inside droplets. In this context, an innovative microdevice was designed and tested experimentally allowing the total electrolysis of the droplets.

EFFETS DES NANO- ET MICRO-STRUCTURATIONS DE SURFACE SUR LE TRANSFERT THERMIQUE EN ÉBULLITION

Ce travail concerne l'étude du transfert thermique par ébullition dans des microcanaux avec l'objectif de développer des systèmes thermiques compacts adaptés aux composants de puissance miniaturisés. Les techniques de nano- et micro-structurations de surface ont été utilisées comme des méthodes novatrices pour améliorer les performances de transfert thermique ainsi que pour retarder l'assèchement intermittent qui apparaît dans les microcanaux. Dans un premier temps, des expériences d'ébullition en vase ont été réalisées afin de mettre en évidence l'impact d'un revêtement à l'échelle nanométrique sur les mécanismes d'ébullition nucléée. Il a été observé que la mouillabilité de surface, qui est modifiée par le dépôt de nanoparticules, a des effets significatifs sur les processus d'ébullition. Dans un deuxième temps, une campagne expérimentale a été réalisée pour étudier l'ébullition convective dans un microcanal avec des échantillons nano- et microstructurées. Ces études ont mis en évidence les effets de la mouillabilité et de la microstructuration de surface sur les régimes d'écoulement, les pertes de charge et le coefficient de transfert thermique. En particulier, de fortes améliorations du coefficient de transfert thermique et le retard d'assèchement intermittent ont été obtenus avec des surfaces microstructurées.

LbL (layer-by-layer) films were fabricated by the self-assembly technique throughout the dipping, spray and dynamic methodologies, available at Laboratory FINEP1, UFSCar, campus Sorocaba. When compared with other bottom-up strategies employed in the build-up of nanostructured ultrathin films, the LbL technique is simple, cheap and easy to handle, beside the incorporation of distinct materials in the film structure, not limited to the form and size of substrates. The times used for the polyelectrolytes in the dipping and dynamic methods were based on the kinetic growth of the (PDDA-GPSS)8 films. In the spray methodology the time used was based initially in the literature and further confirmed by experimental data considering pressure and spray time. All nanostructures formed were characterized by UV-vis spectroscopy, atomic force microscopy, scanning electron microscopy, profilometry and impedance spectroscopy. In all cases it was observed a linear growth of the LbL structures, pointing that the same amount of material was adsorbed at each deposition step. Films obtained from the dynamic methodology indicated higher amount of material adsorbed in the nanostructures, and by an adequate control of the parameters used in the self-assembly methods by physical adsorption one can make a fine tuning regarding to the spontaneous aggregation of the nanoplatelets at solid interfaces. / Neste trabalho comparamos a forma com a qual as nanofolhas de grafeno são nanoestruturadas em bicamadas de poli(cloreto de dialildimetilamônio) (PDDA) e grafeno estabilizado em poli(estireno sulfonato de sódio) (GPSS), designados por (PDDA-GPSS)n sendo n o número de bicamadas depositadas. Os filmes foram fabricados com a técnica de automontagem por adsorção física (LbL, do inglês Layer-by-Layer), utilizando os métodos de imersão, spray e dinâmico, disponíveis no Laboratório Finep1 da UFSCar, campus Sorocaba. Comparada a outras estratégias bottom-up empregadas na construção de filmes ultrafinos nanoestruturados, a técnica LbL é simples, barata e de fácil aplicação, além de permitir a incorporação de diferentes materiais, e não estar limitada quanto à forma e o tamanho dos substratos. Os tempos para a aplicação dos polieletrólitos por imersão e pelo método dinâmico foram estabelecidos com base nas cinéticas de crescimento de filmes (PDDA-GPSS)8. Para o método de spray os tempos foram estabelecidos inicialmente com base na literatura, e depois confirmados em ensaios de crescimento, tendo como variáveis a pressão de ar e o tempo de spray. As nanoestruturas foram caracterizadas por espectroscopia na região do ultravioleta e do visível, microscopia de força atômica, microscopia eletrônica de varredura, perfilometria e espectroscopia de impedância. Em todos os casos foi observado crescimento linear das estruturas LbL, indicando mesma quantidade de material adsorvido a cada etapa de deposição. Os filmes obtidos pelo método dinâmico indicaram maior quantidade de material agregado nas nanoestruturas, e pelo controle adequado nos parâmetros utilizados nos métodos de automontagem por adsorção física podemos realizar um ajuste fino em relação à agregação espontânea de nanofolhas de grafeno em interfaces sólidas

Mise au point d'une méthode de fonctionnalisation de microcomposants par vois photochimique.

Le travail ici exposé a pour finalité le développement d'une méthode efficace de fonctionnalisation et d'adressage de biomolécules sur support solide fermé tel que des microcanaux ou capillaires. La fonctionnalisation de la surface de verre ou de silicium est réalisée en utilisant la formation d'un lien covalent oxime entre la molécule et le support. L'adressage est réalisé en utilisant une déprotection photochimique de la fonction oxyamine protégée par un groupe photolabile (NPPOC). Nous avons démontré en format plan et en format capillaire que la surface oxyamine protégée par un groupe photolabile est efficace pour l'immobilisation de plusieurs oligonucléotides de façon localisée. La stratégie a été étendue pour l'immobilisation dans les capillaires d'autres molécules tels que les sucres, peptides ou molécules hydrophobes et hydrophiles. Les résultats montrent l'efficacité de notre stratégie pour l'immobilisation de ce type de molécules. Nous avons également montré que l'utilisation d'une surface hydrophile (PEG) améliore sensiblement le bruit de fond.

Improved techniques for CE-MALDI-MS off-line coupling and MALDI-MS analysis of primarily hydrophobic proteins and peptides

<p>Due to the hydrophobic nature of integral membrane proteins (IMP) they give rise to several difficulties concerning handling and analysis, which is not the case for the most water soluble proteins. New analysis methods are needed, where the insolubility problems of the hydrophobic proteins due to aggregation and adhesion are tackled. Those problems also affect digestion performance and equipment compatibility for the analysis.</p><p>Protocols for analysis and separation specified for IMP are presented in <b>Paper I</b> and<b> III</b>.</p><p>The instrumentation used in this work was capillary electrophoresis (CE) and matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS). Both instruments are suitable for peptide/proteins analysis.</p><p>In <b>Paper I</b>, protocols for a CE separation of bacteriorhodopsin (BR) peptides as model IMP peptides are established. Also, a partially automated manufacturing procedure of a concentration MALDI-target is presented, suitable for fractions from CE. The MS analysis detected 9 out of 10 cyanogen bromide (CNBr) digested BR peptides. A novel technique for the off-line integration of CE to MALDI-MS using a closed-open-closed system is presented in <b>Paper II</b>, where the open part is a microcanal functioning as a MALDI target window. Investigation of the microcanal electro-osmotic flow (EOF) properties and band broadening characteristics was performed. A protein separation was obtained and detected with MALDI-MS analysis in the microcanal. Different protein digestion methods were evaluated using BR in <b>Paper III</b> through MALDI-MS. Several digestion methods as well as MS media were investigated alongside different MALDI matrices. For example, matrices as the hydrophobic 2,6-dihydroxyacetophenone (DHAP) and 2-Hydroxy-3-methoxybenzoic acid (2H3MBA) or 2-Hydroxy-5-methoxybenzoic acid (2H5MBA) mixed with DHB, appeared to be promising matrices for analysis of BR.</p> / <p>Med anledning av integrala membranproteiners (IMP) hydrofoba egenskaper uppstår flera svårigheter vid hantering och analys av IMP, vilket inte är fallet för vattenlösliga proteiner.
