Supercomplexes multifonctionnels chez les mitochondries, et chez E. coli

Les processus mitochondriaux tels que la réplication et la traduction sont effectués par des complexes multiprotéiques. Par contre, le métabolisme et la voie de maturation des ARN mitochondriaux (p. ex précurseurs des ARNt et des ARNr) sont habituellement traités comme une suite de réactions catalysées par des protéines séparées. L’exécution fidèle et optimale de ces processus mitochondriaux, exige un couplage étroit nécessaire pour la canalisation des intermédiaires métaboliques. Or, les évidences en faveur de l'interconnexion postulée de ces processus cellulaires sont peu nombreuses et proviennent en grande partie des interactions protéine-protéine. Contrairement à la perception classique, nos résultats révèlent l’organisation des fonctions cellulaires telles que la transcription, la traduction, le métabolisme et la régulation en supercomplexes multifonctionnels stables, dans les mitochondries des champignons (ex Saccharomyces cerevisiae, Aspergillus nidulans et Neurospora crassa), des animaux (ex Bos taurus), des plantes (B. oleracea et Arabidopsis thaliana) et chez les bactéries (ex E. coli) à partir desquelles les mitochondries descendent. La composition de ces supercomplexes chez les champignons et les animaux est comparable à celle de levure, toutefois, chez les plantes et E. coli ils comportent des différences notables (ex, présence des enzymes spécifiques à la voie de biosynthèse des sucres et les léctines chez B. oleracea). Chez la levure, en accord avec les changements dûs à la répression catabolique du glucose, nos résultats révèlent que les supercomplexes sont dynamiques et que leur composition en protéines dépend des stimulis et de la régulation cellulaire. De plus, nous montrons que l’inactivation de la voie de biosynthèse des lipides de type II (FASII) perturbe l’assemblage et/ou la biogenèse du supercomplexe de la RNase P (responsable de la maturation en 5’ des précurseurs des ARNt), ce qui suggère que de multiples effets pléiotropiques peuvent être de nature structurale entre les protéines. Chez la levure et chez E. coli, nos études de la maturation in vitro des précurseurs des ARNt et de la protéomique révèlent l’association de la RNase P avec les enzymes de la maturation d’ARNt en 3’. En effet, la voie de maturation des pré-ARNt et des ARNr, et la dégradation des ARN mitochondriaux semblent êtres associées avec la machinerie de la traduction au sein d’un même supercomplexe multifonctionnel dans la mitochondrie de la levure. Chez E. coli, nous avons caractérisé un supercomplexe similaire qui inclut en plus de la RNase P: la PNPase, le complexe du RNA degradosome, l’ARN polymérase, quatre facteurs de transcription, neuf aminoacyl-tRNA synthétases, onze protéines ribosomiques, des chaperons et certaines protéines métaboliques. Ces résultats supposent l’association physique de la transcription, la voie de maturation et d’aminoacylation des ARNt, la dégradation des ARN. Le nombre de cas où les activités cellulaires sont fonctionnellement et structurellement associées est certainement à la hausse (ex, l’éditosome et le complexe de la glycolyse). En effet, l’organisation en supercomplexe multifonctionnel représente probablement l’unité fonctionnelle dans les cellules et les analyses de ces super-structures peuvent devenir la prochaine cible de la biologie structurale. / It is known that processes such as transcription, translation and intron splicing require a multitude of proteins (plus a few non-protein components) organized in large ‘molecular machines’. But, according to traditional views, processing of RNA precursors (e.g., tRNA and rRNA) and metabolic pathways are pools of individual enzymes (single proteins or small complexes), with sequential enzymatic reaction steps connected via diffusible metabolites. This perception is incompatible with the ‘molecular crowding’ in most cellular compartments (e.g., 60% in the mitochondrial matrix). It is also not in line with the cumulating indirect evidence from comprehensive studies of protein-protein interactions and affinity purification, showing that numerous protein complexes involving different metabolic and regulatory processes are interconnected. However, direct evidence of extensive cross-talk among diverse cellular processes remains to be clearly demonstrated. Here we show that in mitochondria of yeast and other fungi (Neurospora crassa and Rhizopus oryzae), animal (Bos taurus), plant (Brassica oleracea), and in E. coli (standing for the “bacterial ancestor” of mitochondria), metabolism is physically interlinked (in supercomplexes) with translation, replication, transcription and RNA processing. Further, the supercomplexes also contain a variety of helper proteins, in support of earlier reports that describe such proteins as important structural units assisting complex assembly. Whereas the composition of supercomplexes in fungi (e.g., Neurospora crassa), animals and yeast is relatively similar, plants and E. coli present substantial compositional differences (e.g., plant-specific enzymes involved in the biosynthesis of sugars and secondary metabolites). In yeast, the supercomplex pattern of glucose-repressed cells is completely different from that of cells grown on galactose/glycerol, and the protein composition perfectly correlates with known regulatory changes under glucose repression. The destabilization of the complex organization is also illustrated by the deletion of genes in the mitochondrial fatty acid type II biosynthetic pathway (mutant strain oar1Δ). Mutants have both, a defect in fatty acid synthesis and in 5’ processing of mitochondrial tRNA, and no longer have a supercomplex containing Oar1p and components of RNase P (5’ tRNA processing). The pleiotropic mutant phenotype is best explained by a structural (assembly) defect. Also in yeast mitochondria, we demonstrate that RNase P and tRNA Z activities are part of a large complex, which further includes the RNA degradosome complex, five additional RNA processing proteins, and several other mitochondrial pathways. 5’ and 3’ tRNA processing enzymes are also associated in a large, multifunctional supercomplex in E. coli that includes six out of the seven proteins of the RNA degradosome, nine aminoacyl-tRNA synthases, RNA polymerase plus four transcription factors, eleven ribosomal proteins plus four translation factors, several components of protein folding and maturation, and a small set of metabolic enzymes. Apparently, not only is RNA processing coordinated, but it is also structurally connected to aminoacylation, transcription and other cellular functions. The number of documented cases where functionally related activities are structurally integrated is definitely increasing (e.g., editosome, glycolysis complex, etc). Indeed, structural integration of related functions and pathways may turn out to be a principle and the analyses of such super-structures may become a next structural biology frontier.

Evaluation of polycyclic amines as modulators of calcium homeostasis in models of neurodegeneration / Young L.

Compromised calcium homeostasis in the central nervous system (CNS) is implicated as a major contributor in the pathology of neurodegeneration. Dysregulation of Ca2+ homeostasis initiates downstream Ca2+–dependent events that lead to apoptotic and/or necrotic cell death. Increases in the intracellular free calcium concentration ([Ca2+]i) may be the result of Ca2+ influx from the extracellular environment or Ca2+ release from intracellular Ca2+ stores such as the endoplasmic reticulum (ER). Influx from the extracellular environment is controlled predominantly by voltage gated calcium channels (VGCC), such as L–type calcium channels (LTCC) and ionotropic glutamate receptors, such as the N–methyl–D–aspartate (NMDA) receptors. Ca2+ release from the ER occurs through the inositol–1,4,5–triphosphate receptors (IP3Rs) or ryanodine receptors (RyRs) via IP3–induced or Ca2+–induced mechanisms. Mitigation of Ca2+ overload through these Ca2+ channels offers an opportunity for pharmacological interventions that may protect against neuronal death. In the present study the ability of a novel series of polycyclic compounds, both the pentacycloundecylamines and triquinylamines, to regulate calcium influx through LTCC was evaluated in PC12 cells using calcium imaging with Fura–2/AM in a fluorescence microplate reader. We were also able for the first time to determine IC50 values for these compounds as LTCC blockers. In addition, selected compounds were evaluated for their ability to offer protection in apoptosis–identifying assays such as the lactate dehydrogenase release assay (LDH–assay), trypan blue staining assay and immunohistochemistry utilizing the Annexin V–FITC stain for apoptosis. We were also able to obtain single crystal structures for the tricyclo[6.3.0.02,6]undecane–4,9–dien–3,11–dione (9) and tricyclo[6.3.0.02,6]undecane–3,11–dione (10) scaffolds as well as a derivative, N–(3–methoxybenzyl)–3,11–azatricyclo[6.3.0.02,6]undecane (14f). We also evaluated the possibility that the polycyclic compounds might be able to modulate Ca2+ flux through intracellular Ca2+ channels. Computational methods were utilized to accurately predicted IC50 values and develop a QSAR model with marginal error. The linear regression model delivered r2 = 0.83, which indicated a favorable correlation between the predicted and experimental IC50 values. This model could thus serve as valuable predictor for future structural design and optimization efforts. Data obtained from the crystallographic analysis confirmed the NMR–data based structural assignments done for these compounds in previous studies. Obtaining structural information gave valuable insight into the differences in size and geometric constrains, which are key features for the LTCC activity of these compounds. vii In conclusion, we found that all of the compounds evaluated were able to attenuate Ca2+ influx through the LTCC, with some compounds having IC50 values comparable with known LTCC blockers such as nimodipine. Representative compounds were evaluated for their ability to afford protection against apoptosis induced by 200 ?M H2O2. With the exception of compound 14c (the most potent LTCC blocker in the series, IC50 = 0.398 ?M), most compounds were able to afford protection at two or more concentrations evaluated. Compound 14c displayed inherent toxicity at the highest concentrations evaluated (100 ?M). We concluded that compounds representing both types of structures (pentacycloudecylamines and triquinylamines) have the ability to attenuate excessive Ca2+ influx through the LTCC. In general the aza–pentacycloundecylamines (8a–c) were the most potent LTCC blocker which also had the ability to offer protection in the cell viability assays. However, NGP1–01 (7a) had the most favorable pharmacological profile overall with good activity as an LTCC blocker (IC50 = 86 ?M) and the ability to significantly attenuate cell death in the cell viability assays, exhibiting no toxicity. In addition to their ability to modulate Ca2+ influx from the extracellular environment, these compounds also displayed the ability to modulate Ca2+ flux through intracellular Ca2+ channels. The mechanisms by which they act on intracellular Ca2+ channels still remains unclear, but from this preliminary study it would appear that these compounds are able to partially inhibiting Ca2+–ATPase activity whilst possibly simultaneously inhibiting the IP3R. In the absence of extracellular Ca2+ these compounds showed the ability in inhibit voltage–induced Ca2+ release (VICaR), possibly by modulating the gating charge of the voltage sensor being the dihydropyridine receptors. In future studies it might be worthwhile to do an expanded QSAR study and evaluate the aza–pentacycloundecylamines. To clarify the mechanisms by which the polycyclic compounds interact with intracellular Ca2+ channels we should examine the direct interaction with the individual Ca2+ channels independently. The polycyclic compounds evaluated in this study demonstrate potential as multifunctional drugs due to their ability to broadly regulate calcium homeostasis through multiple pathways of Ca2+ entry. Analysis and Design of a Multifunctional Spiral Antenna

The Archimedean spiral antenna is well-known for its broadband characteristics with circular polarization and has been investigated for several decades. Since their development in the late 1950's, establishing an analytical expression for the characteristics of spiral antenna has remained somewhat elusive. This has been studied qualitatively and evaluated using numerical and experimental techniques with some success, but many of these methods are not convenient in the design process since they do not impart any physical insight into the effect each design parameter has on the overall operation of the spiral antenna. This work examines the operation of spiral antennas and obtains a closed-form analytical solution by conformal mapping and transmission line model with high precision in a wide frequency band. Based on the analysis of spiral antenna, we propose two novel design processes for the stripline-fed Archimedean spiral antenna. This includes a stripline feed network integrated into one of the spiral arms and a broadband tapered impedance transformer that is conformal to the spiral topology for impedance matching the nominally-high input impedance of the spiral. A Dyson-style balun located at the center facilitates the transition between guided stripline and radiating spiral modes. Measured and simulated results for a probe-fed design operating from 2 GHz to over 20 GHz are in excellent agreements to illustrate the synthesis and performance of a demonstration antenna. The research in this work also provides the possibility to achieve conformal integration and planar structural multi-functionality for an Unmanned Air Vehicle (UAV) with band coverage across HF, UHF, and VHF. The proposed conformal mapping analysis can also be applied on periodic coplanar waveguides for integrated circuit applications.

During this thesis, it has been proved that the electrical conductivity of multilayered and electrically anisotropic carbon fiber materials can be expressed by an equivalent second order tensor, which is equal to sum of each layer’s electrical conductivity tensor. The aforementioned equivalent electrical conductivity tensor is valid assuming that the material’s thickness is negligible compared to the other dimensions of the body. The mathematical expression for the prediction of the electrical conductivity of a multilayered material for any stacking sequence, is based on the electric current conservation, and was validated using different methods. Each layer’s electrical conductivity was experimentally studied at the two principal directions. Transverse to the fibers’ direction, an empirical model was developed for the prediction of the electrical conductivity as a function of the layer’s thickness, of the fibre volume fraction and of temperature. All cases involved the study of multidirectional and unidirectional carbon fiber materials without the presence of matrix (porous form – CF preform) as well as in the presence of polymeric matrix (CFRP). The validation of the equivalent tensor was achieved through three different ways: a) through the measurement of the electric resistance, for various stacking sequences, b) through the Joule heating effect, by recording and comparing the developing temperature field to the respective numerically calculated, c) through 3D numerical models which approximate the analytical solution of the 2D domain problem. Moreover using the finite difference method, certain electrothermal models were developed in order to study the temperature field for different stacking sequences. The electrical problem can be expressed by an elliptic PDE, for the case where the material is electrically anisotropic and homogeneous, or non-homogeneous. On the other hand, the transient heat transfer problem involves the case where the material is thermally anisotropic and homogeneous. Using the equivalent tensor, the 3D domain problem is simplified to a 2D domain problem resulting in less computational requirements for the solution of the problem. The present research study could be used in a plethora of application, such as the development of carbon fibre reinforced heating elements (direct heating CFRP molds) as well as damage detection in multidirectional composite materials with electrical conductive reinforcement. / Κατά τη διάρκεια της παρούσας διδακτορική διατριβής, αποδείχθηκε ότι η ηλεκτρική αγωγιμότητα των πολύστρωτων και ηλεκτρικά ανισότροπων υλικών με ίνες άνθρακα, μπορεί να εκφραστεί από έναν ισοδύναμο τανυστή δεύτερης τάξης, ο οποίος είναι το άθροισμα των τανυστών κάθε στρώσης. Ο ισοδύναμος τανυστής ισχύει υποθέτοντας ότι το πάχος συγκριτικά με τις υπόλοιπες διαστάσεις του υλικού είναι πολύ μικρό. Η μαθηματική έκφραση με την οποία μπορεί να προβλεφθεί η ηλεκτρική αγωγιμότητα ενός πολύστρωτου υλικού για οποιαδήποτε αλληλουχία στρώσεων αποδείχτηκε με συστηματικό τρόπο και βασίζεται στην αρχή διατήρησης του ηλεκτρικού φορτίου. Η ηλεκτρική αγωγιμότητα κάθε στρώσης μελετήθηκε πειραματικά στις δύο κύριες διευθύνσεις. Κάθετα στη διεύθυνση των ινών αναπτύχθηκε ένα εμπειρικό μοντέλο πρόβλεψης της ηλεκτρικής αγωγιμότητας συναρτήσει του πάχους της στρώσης, της περιεκτικότητας σε ίνες άνθρακα και της θερμοκρασίας. Σε όλες τις περιπτώσεις μελετήθηκαν πολύστρωτα υλικά ινών άνθρακα χωρίς μήτρα (πορώδης μορφή-CF preforms) και με πολυμερική μήτρα (CFRPs). Η επιβεβαίωση της εγκυρότητας του ισοδύναμου τανυστή έγινε με τρεις διαφορετικούς τρόπους: α) μέσω μετρήσεων της ηλεκτρικής αντίστασης, για διαφορετικές αλληλουχίες στρώσεων, β) μέσω του φαινομένου Joule, καταγράφοντας και συγκρίνοντας το αναπτυσσόμενο θερμοκρασιακό πεδίο με το θερμοκρασιακό πεδίο που υπολογίζεται αριθμητικά, γ) μέσω τρισδιάστατων αριθμητικών μοντέλων όπου τείνουν στην αναλυτική λύση του δισδιάστατου προβλήματος. Στη συνέχεια αναπτύχτηκαν ηλεκτροθερμικά μοντέλα με τη μέθοδο των πεπερασμένων διαφορών με σκοπό τη μελέτη του θερμοκρασιακού πεδίου για διαφορετικές αλληλουχίες στρώσεων. Το ηλεκτρικό πρόβλημα εκφράζεται από μία ελλειπτική διαφορική εξίσωση όπου το υλικό είναι ηλεκτρικά ανισότροπο και ομογενές ή μη ομογενές ενώ το θερμικό πρόβλημα είναι θερμικά ανισότροπο και ομογενές. Χρησιμοποιώντας τον ισοδύναμο τανυστή το τρισδιάστατο πρόβλημα μετατρέπεται σε ένα δισδιάστατο πρόβλημα με αποτέλεσμα να απαιτούνται λιγότεροι πόροι για την επίλυση του προβλήματος. Η συγκεκριμένη εργασία μπορεί να χρησιμοποιηθεί σε μία πληθώρα εφαρμογών όπως στην ανάπτυξη και στη λειτουργία θερμαινόμενων στοιχείων ενισχυμένων με ίνες άνθρακα (καλούπια όπου το θερμαντικό στοιχείο το αποτελούν οι ίνες άνθρακα) αλλά και στην ανίχνευση βλάβης συνθέτων υλικών με αγώγιμη ενίσχυση.

Nanoparticules hybrides thermosensibles pour la théranostique / Hybrid and thermosensitive nanoparticles for theranostic applications

Cette étude concerne le développement de nanoparticules hybrides offrant de nouvelles stratégies pour la thérapie et le diagnostic médical. Elles sont constituées d’un cœur magnétique jouant le rôle d’agent de contraste pour l’IRM et d’inducteur de chaleur par hyperthermie, d’une couronne de polymère thermosensible permettant d’encapsuler des principes actifs et de peptides de reconnaissance biologique. Une grande partie de l’étude a consisté à étudier les processus d'adsorption de copolymères poly(éther)-b-poly(L-lysine) de composition variable sur les particules magnétiques et à comprendre le rôle de la conformation des chaînes polymère à la surface des particules sur la stabilité des colloïdes en milieu physiologique. Un agent antitumoral a été encapsulé puis libéré de façon contrôlée sous l’effet d’un champ magnétique alternatif en exploitant le caractère thermosensible des blocs polyéthers. Des séquences peptidiques ciblant les zones d’inflammation de la barrière hémato-encéphalique ont été greffées sur les copolymères. L’efficacité du ciblage a été validée par IRM et fluorescence sur un modèle animal démontrant ainsi la multifonctionnalité des nanoparticules. / This work deals with the development of hybrid nanoparticles that could offer new strategies for therapy and diagnostic. These are based on a magnetic core which can play the role of contrast agent for MRI as well as heat inductor in AC magnetic field. This inorganic core is surrounded by a thermo-responsive polymeric brush that controls the loading and the release of drugs, and can be functionalized by specific ligands ensuring the targeting specificity. A large part of this work consists in studying the adsorption mechanism of poly(ether)-b-poly(L-lysine) based block copolymers onto magnetic particle and to better understand the influence of the polymer chain conformation at particles surface on the colloidal stability under physiological conditions. An anticancer drug has been loaded and released in a controlled manner under alternative magnetic field by taking advantage from the thermosensitivity of the polyether block. Targeting peptides specific of inflammation sites at the blood brain barrier have been grafted onto copolymers. The targeting specificity has been demonstrated by MRI and fluorescence imaging in rats attesting the multifunctionality of such nanoparticles.

Cette thèse décrit une stratégie de synthèse de nouvelles générations des nanoparticules (NPs) pour applications biomédicales, visant à une amélioration de leurs performances pour l’imagerie, le diagnostic thérapeutique. Ces NPs présentent plusieurs fonctionnalités leur permettant de réaliser des tâches multiples. Deux types de sondes bimodales ont été développés et étudiés afin d'évaluer leur potentiel comme agents (1) de contraste en IRM et (2) luminescents. Ces objetscombinent les propriétés des complexes de lanthanide (Ln3+) et celles des NPs de silice ou de type coeur-écorce Fe2O3@SiO2 pour une imagerie bimodale. Ces NPs testées sur des cellules vivantes ont permis d’illustrer la preuve du concept aussi bien en IRM avec une augmentation d'intensité des images et un impact significatif sur les relaxivities r1, r2 et r2* qu’en photoluminescence. L’étude du système coeur-écorce a montré que l’influence du contrôle fin de l’écorce autour du noyau d'oxyde de fer a pu être modélisée. / This thesis describes a strategy of synthesis of new generations of nanoparticles (NPs) for biomedical applications, aiming at an improvement of their performances for the imaging, and the therapeutic diagnosis. These NPs present several functionalities enabling them to carry out multiple tasks. Two types of bimodal probes were developed and studied so as to evaluate their potential as contrast agents (1) in MRI and (2) and luminescence. These objects combine the properties of the lanthanide complexes (Ln3+) and those of NPs of silica or core/shell Fe2O3@SiO2 for a bimodal imaging. These NPs tested on living cells were able to illustrate the proof of the concept not only in MRI with an increase of intensity of the images and a significant impact on the relaxivities r1, r 2 and r2* but also in photoluminescence. The study of the core/shell system showed that the influence of the fine control of the shell around the iron oxide core could be modeled.

The work is focused on the issue of parameterization of the enterprise capabilities of its multifunction, stabilisation and growth by means of the non-capital measures. The basic terms and categories of this issue are characterized in the search section such as the agrarian sector and its position in the national economy of the Czech Republic, the common agricultural policy, rural development programme, the marginality, the LFA, the sustainability of agriculture, business multifunctional potential and its diversification, the financial health of the company, creditworthy and bankruptcy models. Doctoral dissertation tasks were solved on the file of legal entities in the South Bohemia region (229 farms - i.e. 1,504 statistical units) in the years 2005-2010, after the correction then on 214 agricultural holdings with the initial year 2010 and a scientific experiment was performed on 5 model enterprises with the initial year 2012. The main outputs of the dissertation are as follows: definition of zoning formulas, new theoretical defining business multifunctional potential including the methodology for determining business potential and its parameterization. Investigation was carried out on the test set of agricultural enterprises through models of non-investment measures, including subsidies; consideration was given to progressive increases and size degression. Variants of optimization (profit maximization and advantages of livestock production) were tested at scientific experiment on five chosen enterprises. The result of multifunctional potential parameterization process is also expressed in differentiation of the test set on creditworthy businesses and non-performing businesses and businesses falling into, so-called, a gray area. Solving the dissertation was connected to the state task of the Institute of Agricultural Economics and Information (IAEI, Prague) in preparation for the new programming period, the Rural Development Programme for the years 2014-2020 and the government document Strategy of Agriculture and Food Industry of the Czech Republic.

Allied to the interests of organocatalysis of simplicity and low operating costs, resulting in products that do not have contaminants like metals, designed in order to be able to catalyze / promote the reaction of Morita-Baylis-Hillman (MBH). The synthesis was performed starting from cheap raw materials and good commercial availability, S-lactic acid and 2-N, Ndimetilaminoetan-1-ol in one step reaction (98%). Based on current mechanistic proposal of the MBH reaction, the mechanism of Cantillo and Kappe, we investigated the reaction conditions, where there is a hydrogen transfer in aprotic media and also in the presence of phenols as additives, checking the influence speeds, reaction yields and asymmetric induction. We investigated the reaction between pnitrobenzaldehyde and acrylonitrile as reaction MBH pattern, varying experimental conditions such as solvent, quantity of (S)-2-hydroxypropanoate 2-N, Ndimetilaminoetila time, temperature and addition of phenolic additives. In addition, a new method was developed for the separation of enantiomers for the racemic compound antiparasitic 2 - [Hydroxy (4-nitrophenyl) methyl] acrylonitrile by gas chromatography with high resolution capillary columns packed with chiral phases. Chemical yields were obtained 2 - [hydroxy (4-nitrophenyl) methyl] <1 to 98% and enantiomeric excesses up to 20.5%. Mechanistic aspects are discussed in this paper, based on the results and computations, using the theory DFT and QTAIM. / Este trabalho foi realizado com o intuito de sintetizar e avaliar uma nova amina terciária quiral, o (S)-2-hidroxipropanoato de 2-N,N-dimetilaminoetila de acordo ao conceito de catalisador multifuncional, o qual permite que uma mesma molécula apresente várias características como os de ácido de Bronsted e base de Lewis. Aliado aos interesses da organocatálise de simplicidade e baixo custo operacional, resultando em produtos que não apresentam metais como contaminantes, projetamos no intuito de ser capaz de catalisar/promover a reação de Morita-Baylis-Hilman (MBH). A síntese de foi efetuada a partir de matérias-primas baratas e de boa disponibilidade comercial; o ácido S-lático e o 2-N,Ndimetilaminoetan- 1-ol em uma etapa reacional (98%). Baseado na atual proposta mecanística da reação de MBH, o mecanismo de Cantillo e Kappe, foram investigados as condições reacionais, onde há a transferência de hidrogênio em meio aprótico e também na presença de fenóis como aditivos, verificando a influência nas velocidades, rendimentos de reação e indução assimétrica. Investigamos a reação entre o p-nitrobenzaldeído e a acrilonitrila como reação de MBH padrão, variando condições experimentais como: solvente, quantidade do(S)-2-hidroxipropanoato de 2-N,N-dimetilaminoetila, tempo, temperatura e adição de aditivos fenólicos. Em adição, foi desenvolvido um novo método de separação de enantiômeros para o composto racêmico antiparasitário 2-[Hidroxi(4-nitrofenil)metil] acrilonitrila via cromatografia gasosa com colunas capilares de alta resolução empacotadas por fases quirais. Foram obtidos rendimentos químicos do 2-[Hidroxi(4- nitrofenil)metil] <1 à 98% e excessos enantioméricos até 20,5%. Aspectos mecanísticos são discutidos neste trabalho, baseados nos resultados obtidos e em cálculos computacionais, usando a teoria DFT e QTAIM.
