Interventions thérapeutiques prometteuses dans un modèle in vivo de stéatohépatite non alcoolique

La stéatohépatite non alcoolique (NASH) est une pathologie du foie dont l’amplitude et les répercussions sont de plus en plus préoccupantes dans le monde médical ou biomédical. Elle est associée à l’obésité, au syndrome métabolique et au diabète sucré de type II. La recherche de la thérapie optimale pour le NASH est un domaine en plein essor puisqu’aucun traitement n’est suffisamment efficace à ce jour. La présente étude fait le point sur de nouvelles possibilités de traitements qui se sont avérés efficaces pour contrer les différentes lésions métaboliques et cellulaires rencontrées dans un modèle in vivo chez le rat où le NASH est induit par l’ingestion d’une diète riche en gras. Cette étude démontre, tout d’abord, que les traitements durant six semaines avec l’acide ursodéoxycholique (UDCA) et son dérivé le NCX 1000, possédant des propriétés donatrices de monoxyde d’azote, à doses équimolaires, protègent de manière équivalente le foie contre le stress oxydatif, l’hyperinsulinémie, l’inflammation et la fibrose causés par la stéatohépatite. De plus, la combinaison d’une plus faible dose de NCX 1000 avec un antioxydant lipophile tel que la vitamine E offre une protection similaire, particulièrement au niveau des paramètres du stress oxydatif. Par ailleurs, l’étude illustre aussi que la silibinine, composé polyphénolique actif du chardon marie (Silybum marianum) et utilisé en traitement pendant 5 semaines, possède un pouvoir hépatoprotecteur, des propriétés antioxydantes et un effet hypoinsulinémique dans ce modèle de stéatohépatite d’origine nutritionnelle. Le potentiel thérapeutique de ces composés en fait des candidats de choix pour le traitement du NASH qui méritent de faire l’objet d’études cliniques poussées. / Nonalcoholic steatohepatitis (NASH) is a serious liver condition related to the metabolic syndrome, obesity, and type II diabetes mellitus whose prevalence is drastically rising in developed countries and worldwide. Several remedies were investigated for the treatment of NASH but an efficient therapy has yet to be developed. In the present study, we explored novel therapeutic possibilities that were thought to be effective for the treatment of experimental high-fat diet-induced NASH the in rat. Our results show that a chronic six week treatment with a high dose of NCX 1000, a derivative of ursodeoxycholic acid (UDCA) with nitric oxide (NO) donating properties, is efficient at reversing steatosis, oxidative stress, inflammation, insulin resistance and fibrosis; major hallmarks of experimental NASH. We also demonstrated that the mother molecule, UDCA, is as efficacious in controlling the same parameters at equimolar doses. Moreover, our study demonstrates that NCX 1000 at lower doses can exert similar potent properties when combined with lipophilic antioxidants like vitamin E. On the other hand, we found that a 5-week treatment with silibinin, the major active component of milk thistle extract, improved liver steatosis and inflammation and decreased NASH-induced oxidative stress, insulin resistance, and fibrosis. These compounds have therefore the potential for being developed for the treatment of NASH. Clinical evidences are needed.

Etude des marchés d'assurance non-vie à l'aide d'équilibre de Nash et de modèle de risques avec dépendance

L'actuariat non-vie étudie les différents aspects quantitatifs de l'activité d'assurance. Cette thèse vise à expliquer sous différentes perspectives les interactions entre les différents agents économiques, l'assuré, l'assureur et le marché, sur un marché d'assurance. Le chapitre 1 souligne à quel point la prise en compte de la prime marché est importante dans la décision de l'assuré de renouveler ou non son contrat d'assurance avec son assureur actuel. La nécessitéd'un modèle de marché est établie. Le chapitre 2 répond à cette problématique en utilisant la théorie des jeux non-coopératifs pour modéliser la compétition. Dans la littérature actuelle, les modèles de compétition seréduisent toujours à une optimisation simpliste du volume de prime basée sur une vision d'un assureur contre le marché. Partant d'un modèle de marché à une période, un jeu d'assureurs est formulé, où l'existence et l'unicité de l'équilibre de Nash sont vérifiées. Les propriétés des primes d'équilibre sont étudiées pour mieux comprendre les facteurs clés d'une position dominante d'un assureur par rapport aux autres. Ensuite, l'intégration du jeu sur une période dans un cadre dynamique se fait par la répétition du jeu sur plusieurs périodes. Une approche par Monte-Carlo est utilisée pour évaluer la probabilité pour un assureur d'être ruiné, de rester leader, de disparaître du jeu par manque d'assurés en portefeuille. Ce chapitre vise à mieux comprendre la présence de cycles en assurance non-vie. Le chapitre 3 présente en profondeur le calcul effectif d'équilibre de Nash pour n joueurs sous contraintes, appelé équilibre de Nash généralisé. Il propose un panorama des méthodes d'optimisation pour la résolution des n sous-problèmes d'optimisation. Cette résolution sefait à l'aide d'une équation semi-lisse basée sur la reformulation de Karush-Kuhn-Tucker duproblème d'équilibre de Nash généralisé. Ces équations nécessitent l'utilisation du Jacobiengénéralisé pour les fonctions localement lipschitziennes intervenant dans le problème d'optimisation.Une étude de convergence et une comparaison des méthodes d'optimisation sont réalisées.Enfin, le chapitre 4 aborde le calcul de la probabilité de ruine, un autre thème fondamentalde l'assurance non-vie. Dans ce chapitre, un modèle de risque avec dépendance entre lesmontants ou les temps d'attente de sinistre est étudié. De nouvelles formules asymptotiquesde la probabilité de ruine en temps infini sont obtenues dans un cadre large de modèle de risquesavec dépendance entre sinistres. De plus, on obtient des formules explicites de la probabilité deruine en temps discret. Dans ce modèle discret, l'analyse structure de dépendance permet dequantifier l'écart maximal sur les fonctions de répartition jointe des montants entre la versioncontinue et la version discrète.

A game theoretic analysis of adaptive radar jamming

Advances in digital signal processing (DSP) and computing technology have resulted in the emergence of increasingly adaptive radar systems. It is clear that the Electronic Attack (EA), or jamming, of such radar systems is expected to become a more difficult task. The reason for this research was to address the issue of jamming adaptive radar systems. This required consideration of adaptive jamming systems and the development of a methodology for outlining the features of such a system is proposed as the key contribution of this thesis. For the first time, game-based optimization methods have been applied to a maritime counter-surveillance/counter-targeting scenario involving conventional, as well as so-called ‘smart’ noise jamming.Conventional noise jamming methods feature prominently in the origins of radar electronic warfare, and are still widely implemented. They have been well studied, and are important for comparisons with coherent jamming techniques.Moreover, noise jamming is more readily applied with limited information support and is therefore germane to the problem of jamming adaptive radars; during theearly stages when the jammer tries to learn about the radar’s parameters and its own optimal actions.A radar and a jammer were considered as informed opponents ‘playing’ in a non-cooperative two-player, zero-sum game. The effects of jamming on the target detection performance of a radar using Constant False Alarm Rate (CFAR)processing were analyzed using a game theoretic approach for three cases: (1) Ungated Range Noise (URN), (2) Range-Gated Noise (RGN) and (3) False-Target (FT) jamming.Assuming a Swerling type II target in the presence of Rayleigh-distributed clutter, utility functions were described for Cell-Averaging (CA) and Order Statistic (OS) CFAR processors and the three cases of jamming. The analyses included optimizations of these utility functions, subject to certain constraints, with respectto control variables (strategies) in the jammer, such as jammer power and spatial extent of jamming, and control variables in the radar, such as threshold parameter and reference window size. The utility functions were evaluated over the players’ strategy sets and the resulting matrix-form games were solved for the optimal or ‘best response’ strategies of both the jammer and the radar.

An investigation into Braess' paradox

Braess' paradox is a counter-intuitive phenomenon which can occur in congesting networks. It refers to those cases where the introduction of a new link in the network results in the total travel time on the network increasing. The dissertation starts by introducing the traffic assignment problem and the concept of equilibrium in traffic assignment. The concept of equilibrium is based on Wardrop's first principle that all travellers will attempt to minimize their own travel time regardless of the effect on others. A literature review includes details of a number of papers that have been published investigating theoretical aspects of the paradox. There is also a brief description of Game Theory and the Nash Equilibrium. It has been shown that the equilibrium assignment is an example of Nash Equilibrium. The majority of work that has been published deals with networks where the delay functions that are used to compute the travel times on the links of the network do not include explicit representation of the capacity of the links. In this dissertation a network that is similar in form to the one first presented by Braess was constructed with the difference being that the well-known BPR function was used in the delay functions. This network was used to show that a number of findings that had been presented previously using simpler functions also applied to this network. It was shown that when it occurs, Braess' paradox only occurs over a range of values at relatively low levels of congestion. Real-world networks were then investigated and it was found that similar results occurred to those found in the simpler test networks that are often used in discussions of the paradox. O presente trabalho propõe-se analisar a industrialização do Brasil no contexto de mudanças econômicas e políticas no primeiro governo de Getúlio Vargas, de 1930 até 1945. Na literatura de economia brasileira com relação ao surgimento da indústria no Brasil, duas visões se contrapõem: de um lado, a industrialização como subproduto da intervenção do governo no setor cafeeiro; de outro, a industrialização intencionalmente promovida pelo governo. Com base em uma análise de equilíbrio em um jogo dinâmico de informação imperfeita, é evidenciada a racionalidade da criação de instituições para o desenvolvimento industrial na década de 1930. Este resultado de equilíbrio mostra as mudanças pela qual o Brasil passou a partir da década de 1930, com mudanças da estrutura tributária, educacional, financeira e relações de trabalho, ou seja, essa mudança institucional gerou campo fértil para o surgimento do processo de industrialização que caracterizou o primeiro governo Vargas e também mostra a intencionalidade desse governo, quando se observa a transformação do sistema tributário, de tal modo a ficar imune das oscilações econômicas externas e também as mudanças educacionais, que passou a incentivar o ensino primário, secundário e técnico profissionalizante, medidas essas feitas para atender o novo panorama econômico brasileiro. Efeito do Yo Jyo Hen Shi Ko (YHK) no metabolismo lipídico na esteatohepatite experimental / Evaluation of Yo Jyo Hen Shi Ko (YHK) in hepatic lipid metabolism in experimental steatohepatitis

A Doença Hepática Gordurosa Não Alcoólica (DHGNA) é atualmente uma das formas mais comuns de doença hepática, e está diretamente relacionada com a obesidade. Estudos indicam uma prevalência por volta de 30% na população ocidental e 11% na população oriental. A DGHNA possui um largo espectro abrangendo desde casos de esteatose simples sem inflamação, até casos com esteatohepatite e fibrose, podendo evoluir para cirrose e carcinoma hepatocelular. A fisiopatogênese da DHGNA é baseada em múltiplos eventos: resistência insulínica, aumento da lipólise periférica, aumento da síntese de novo de lipídeos, estresse oxidativo, endotoxemia crônica no desencadeamento da inflamação e fibrose entre outros. A etapa inicial caracteriza-se pelo acúmulo de ácidos graxos no hepatócito, suplantando sua capacidade de metabolização e exportação conseqüente à ação lipogênica da insulina, desta forma o metabolismo de ácidos graxos está intimamente ligado ao desenvolvimento da DHGNA. O Yo Jyo Hen Shi Ko (YHK) é um composto natural usado na medicina japonesa para tratamento de doenças hepáticas e apresenta propriedades antioxidantes, antiinflamatórias e hipolipemiantes. O presente estudo teve como objetivo avaliar o efeito do YHK no metabolismo hepático de lipídeos. Para tanto, foram utilizados camundongos obesos (ob/ob) com esteatohepatite não alcoólica (ENA) induzida por dieta deficiente em colina e metionina (DCM) e foram analisados no tecido hepático, genes relacionados com a síntese de novo de lipídeos (SREBP1c, FASn), genes relacionados com a oxidação e exportação de lipídeos (CPT1a e SCD-1, MTP), assim como genes relacionados com o armazenamento de lipídeos (Perilipina e ADFP). O YHK apresentou um efeito citoprotetor hepático com melhora dos parâmetros histológicos neste modelo experimental de ENA. Associadamente, houve redução na expressão de genes relacionados à síntese de novo como SREBP e da FASn, quando se comparou o grupo tratado com DCM+YHK com o grupo não tratado DCM. Em contrapartida, houve aumento na expressão da MTP e da SCD-1 ocasionando uma maior exportação de triglicerídeos hepáticos nos animais que utilizaram o YHK. Ainda, o YHK modulou as proteínas Perilipina e ADFP. Por outro lado, não houve modificação na oxidação de lipídeos.. Conclui-se que o YHK pode ser uma droga promissora no tratamento da DHGNA, já que age modulando genes envolvidos na síntese e exportação de lipídeos hepáticos, reduzindo o acúmulo de gordura no hepatócito / Nonalcoholic fatty liver disease (NAFLD) is currently one of the most common forms of liver disease, related directly to the increase of obesity in the world. Previous studies indicate a prevalence of around 30% in the western population and 11% in the eastern. This disease covers cases from simple steatosis without inflammation to cases of steato hepatitis (NASH) and fibrosis and it may lead to liver cirrhosis and hepatocellular carcinoma. The pathophysiology of NAFLD is based on multiple events: insulin resistance, increased peripheral lipolysis, increased de novo lipogenesis, oxidative stress, endotoxemia in triggering chronic inflammation and fibrosis and others. The initial stage of the disease is characterized by the accumulation of fatty acids in hepatocytes caused by the lipogenic action of insulin. This condition results in the surpassing of the cells ability to metabolize and export. This way, the metabolism of fatty acids is closely linked to the development of NAFLD. The Yo Jyo Hen Shi Ko (YHK) is a natural compound used in Japanese medicine for the treatment of liver disease and it has antioxidant, anti-inflammatory and lipid lowering properties. This study aimed to evaluate the effect of YHK in the hepatic metabolism of lipids. An experimental model was designed using obese mice (ob/ob) affected by NASH induced by a deficient diet in choline and methionine (MCD). Then, genes from its hepatic tissue related to de novo lipogenesis (SREBP1c, FASN), to lipid oxidation and exportation (CPT1A and SCD-1, MTP) as well as genes related to lipid storage (Perilipin and ADFP) were analyzed. The YHK presented a cytoprotective effect, improving the hepatic histological parameters in this experimental model. Additionally, when comparing the group treated with MCD + YHK (diet + YHK) to the untreated MCD group (diet) there was a reduction in the expression of genes related to de novo synthesis like SREBP1c and FASn. However, an increased expression of MTP and SCD-1 were observed leading to a greater liver exportation of fatty acids in animals that were treated with YHK. Moreover, the YHK modulated the proteins Perilipin and ADFP. On the other hand, there was no changing in lipid oxidation. In summary, the YHK can be a promising drug for the treatment of NAFLD, as it acts by modulating genes involved in the synthesis and exportation of hepatic lipids, reducing the accumulation of fat in hepatocytes

Seat Allocation And Pricing in a Duopoly in The Airline Industry

Revenue Management (RM) is the practice of managing perishable assets by control-ling their availability and/or prices with an objective to maximize the total revenue. Seat inventory allocation falls in the purview of quantity-based RM. The liberalization of the aviation sector and the subsequent entrance of the low-cost carriers saw an ever-increasing customer base for the airline industry. Given the large number of buyers, firms were free to decide the price at which they would sell tickets. The low-cost carriers started to follow a third degree price discrimination and segmentation of the market, charging a higher price to the market with a relatively inelastic demand. Although a lot of work has been done in the area of seat inventory allocation under a monopolistic market scenario, we realized that not a lot of work had been done in a competitive market scenario. This thesis considers the problem of seat inventory allocation and pricing in a duopoly where each of the competing airlines have two fare-classes. We consider the possibility that the same fare-class may be priced differently by the two competing airlines and allow for the over flow of passengers between the airlines in the same fare-class. In the first part of our work, we develop a non-linear mathematical model for setting the booking limits for one of the two competing air-lines such that the revenue earned is maximized. We consider over flow of passengers from one airline to another in the same fare-class in response to a price differential and compare the results obtained from our model with the standard Expected Marginal Seat Revenue (EMSR) model under a monopolistic scenario. The results show that our model gives higher revenues than that obtained from the EMSR model. In the second part of our work, we consider a non-cooperative game between two competing airlines with price cutting as the strategy to increase their demand. Through numerical computations, we identify the pure strategy Nash equilibrium. From the results, we conclude that Nash equilibrium is achieved only when both the airlines follow the same pricing strategy indicating that individual price cutting will not be beneficial. This also indicates that unless the competitors enter into a cooperative coalition with each other, they would not benefit from deep discount offers. In the third and final part, we prove theoretically the existence of pure strategy Nash equilibrium in a two airline, two fare-class problem with price sensitive over flow of customers in the same fare-class that was computationally analysed earlier. The strategy / strategies at which Nash equilibrium is achieved are identified. We show that Nash equilibrium is only achieved when both the airlines price identically. Hence, our thesis concludes that differential pricing does not hold any significance for the competing airlines from an operational perspective.

Dynamic management of spectral resources in LTE networks / Gestion dynamique des ressources spectrales dans les réseaux LTE

La croissance exponentielle du nombre de dispositifs communicants et des services sans fils émergents fixe des objectifs toujours plus haut pour répondre à la demande de capacité sans cesse croissante des utilisateurs. Cela pose des défis constants pour atteindre les objectifs envisagés. La réutilisation spectrale élevée (High efficiency spectral reuse) a été adopté, cependant, elle conduit à des interférences accrues sur le réseau, ce qui dégrade les performances. L'OFDM (Orthogonal Frequency Division Multiplexing) est utilisé comme solution dans les réseaux de 4 G. Grâce à son orthogonalité, l'OFDM élimine l'interférence intra-cellulaire, mais l'interférence inter-cellule reste importante. Plusieurs méthodes connues sous le nom d'Inter-Cell interférences coordination (ICIC) ont été proposées pour les diminuer. L'ICIC permet la gestion des ressources radio coordonnée entre plusieurs cellules appelées ENodeB. Ces eNodeB peuvent partager les informations nécessaires grâce à l'interface X2 qui les relient, ces informations sont transmises par des messages LTE normalisés. Lorsque les ENodeBs sélectionnent égoïstement les ressources, la théorie de jeux non-coopératifs est largement appliquée pour trouver un juste équilibre. Dans cette thèse, nous mettons l'accent sur l'ICIC pour la liaison descendante d'un système OFDMA cellulaire dans le contexte du projet SOAPS (Spectrum opportuniste accès à la Sécurité publique). Ce projet a pour but l'amélioration de la planification des ressources de fréquences pour fournir des services à large bande dans les systèmes PMR (radiocommunications mobiles privées) en utilisant les technologies LTE. Nous adressons le problème d'ICIC en proposant quatre solutions différentes sous forme d'algorithmes entièrement décentralisés, ces algorithmes se basent sur la théorie des jeux non-coopératifs avec des équilibres de Nash purs des jeux considérés / The exponential growth in the number of communications devices has set out new ambitious targets to meet the ever-increasing demand for user capacity in emerging wireless systems. However, the inherent impairments of communication channels in cellular systems pose constant challenges to meet the envisioned targets. High spectral reuse efficiency was adopted as a solution to higher data rates. Despite its benefits, high spectral reuse leads to increased interference over the network, which degrades performances of mobile users with bad channel quality. To face this added interfence, OFDM (Orthogonal Frequency Division Multiplexing) is used for the new 4th generation network. Thanks to its orthogonality OFDM eliminates the intra-cellular interference, but when the same resources are used in two adjacents cells, the inter-cell interference becomes severe. To get rid of the latter, several methods for Inter-Cell Interference Coordination (ICIC) have been proposed. ICIC allows coordinated radio resources management between multiple cells. The eNodeBs can share resource usage information and interference levels over the X2 interface through LTE-normalized messages. Non-cooperative game theory was largely applied were eNodeBs selfishly selects resource blocks (RBs) in order to minimize interference. In this thesis, we stress on ICIC for the downlink of a cellular OFDMA system in the context of the SOAPS (Spectrum Opportunistic Access in Public Safety) project. This project focuses on the improvement of frequency resource scheduling for Broadband Services provision by PMR (Private Mobile Radio) systems using LTE technologies. We addressed this problem with four different solutions based on Non-cooperative game theory, three algorithms are devoted to RB selection in order to manage the interference, while the last one is a power control scheme with power economy and enhanced system performances
