Desinfektionsmittel sind ein elementarer Bestandteil der Tierseuchenbekämpfung und damit auch der Lebensmittelsicherheit. Die Prüfung chemischer Desinfektionsmittel ist Voraussetzung für deren zuverlässige Wirksamkeit und zielgerichteten Einsatz. In Deutschland geschieht dies nach den Richtlinien der Deutschen Veterinärmedizinischen Gesellschaft e.V. (DVG). Seit der ersten Fassung sind die Richtlinien einem ständigen Anpassungsprozess unterworfen. Im Zuge der europäischen Harmonisierung gilt es nun, sich gesamteuropäischen Richtlinien, verfasst durch das europäische Komitee für Normung (Comité Européen de Normalisation) (CEN) anzupassen. Das Thema dieser Arbeit entwickelte sich im Kontext der derzeitigen Diskussion über Verbesserungsvorschläge zu den bestehenden Richtlinien und deren Anpassung an die europäischen Normen. Es wurden je zwei Testviren für die Bereiche Tierhaltung und tierärztliche Praxis ausgewählt, um sie auf Eignung für die Viruzidieprüfung zu testen und gegebenenfalls zu etablieren. Des Weiteren wurde in einem zweiten Teil, in Anlehnung an die Forderungen der europäischen Normen die Prüfung zu vereinfachen, ein alternatives Zellkulturnachweissystem für das Newcastle-Disease-Virus (NDV) geprüft.
Die Prüfung der viruziden Wirksamkeit erfolgte mit fünf verschiedenen Grundsubstanzen, gewählt um ein möglichst breites Spektrum an Desinfektionsmittelwirkstoffen abzudecken. Es wurden Glutaraldehyd, Ethanol, Natronlauge, Natriumhypochlorit und Peressigsäure verwendet. Die Versuche wurden mit einer niedrigen Eiweißbelastung und bei einer Temperatur von 20°C durchgeführt. Um eine praxisnahe Situation zu simulieren wurde auf, bereits in den DVG-Richtlinien, verankerten Stahl- und Holzkeimträgertests zurückgegriffen.
Als mögliche Prüfviren für die Tierhaltung wurden das Equine Arteritis-Virus (EAV) und das Bovine Virus Diarrhoe Virus verwendet. Bei beiden Viren handelt es sich um weit verbreitete Tierseuchenerreger mit einer großen epidemiologischen Bedeutung. Die Untersuchung von fünf verschiedenen Desinfektionsmitteln erfolgte im Keimträgertest auf Holz. Sowohl EAV als auch BVD stellen ein weniger geeignetes Prüfvirus dar, da beide Viren enorme Titerverluste im Trocknungsvorgang der Holzkeimträger zeigten. Die Viren ließen sich zwar leicht vermehren, aber die erzielten Ausgangstiter reichten nicht aus um die Trocknungsverluste zu kompensieren und aussagekräftige Ergebnisse zu produzieren.
Für den Bereich tierärztliche Praxis wurden das Feline Coronavirus (FCoV) und das Murine Parvovirus (MPV) genutzt. FCoV ist ein weltweit in Hauskatzenpopulationen vorkommendes Virus mit einer hohen Seroprävalenz und wurde daher ausgewählt. MPV wurde als Stellvertreter für die, in der Praxis häufig vorkommenden Parvovirusinfektionen gewählt. Es schien ein ideales Modellvirus aufgrund seiner weiten Verbreitung in der Forschung zu sein. Bei beiden Viren erfolgte die Prüfung auf Stahlkeimträgern. Unter Laborbedingungen konnte FCoV ohne Probleme zu hohen Titern vermehrt werden. Es gab keine nennenswerten Trocknungsverluste. FCoV erwies sich als geeignetes Prüfvirus. MPV hingegen ist bedingt durch die langen Versuchszeiten und schwierig auszuwertenden Zellkulturen, sowie wegen der niedrigen Ausgangstiter weniger geeignet als Modellvirus für die Desinfektionsmittelprüfung.
Die Anzucht von NDV in Allantoisflüssigkeit von SPF Hühnereiern erschien sehr aufwendig und mit hohem Eiweißfehler belastet. In den Versuchen konnte ein deutlich höherer Eiweißgehalt als in den vergleichend geprüften, in Zellkultur angezogenen Viren nachgewiesen werden. Infolge der Probleme mit der Kultivierung der LMH-Zelllinie und den damit verbundenen langen Wartezeiten bis zur eigentlichen Versuchsdurchführung kann nur eine teilweise Empfehlung, von auf Zellkultur vermehrtem NDV (NDV (ZK)) gegeben werden. Nach Behebung dieser Probleme ist durchaus eine Ablösung, von in Allantoisflüssigkeit angezüchtetem NDV durch NDV (ZK) zu empfehlen. Die Verfälschung der Ergebnisse durch die höheren Eiweißgehalte bei Desinfektionsmitteln mit deutlichem Eiweißfehler könnten so vermieden werden.

Effect of naturally contaminated diet with deoxynivalenol (don) on vaccine response against Newcastle disease and infectious bronchitis virus in broiler chickenHamadi, Solaman 12 1900 (has links)
Le déoxynivalénol (DON) est l'une des mycotoxines trichothécènes les plus abondantes et les plus importantes produites par les espèces de Fusarium. Les aliments contaminés par les mycotoxines peuvent affecter négativement la santé des poulets de chair. La présente étude a été menée pour évaluer les effets du déoxynivalénol (DON) sur les performances des poulets de chair, le poids des organes, les paramètres biochimiques sériques, la réponse immunitaire et l'histologie intestinale. L’expérience a permis d’évaluer l'efficacité d'un additif alimentaire commercial, basé sur les synbiotiques (SFA), et ayant la capacité d'oxyder le DON. Au total, six cents poussins mâles d'un jour d'une souche commerciale (Ross 308) ont été vaccinés avec un vaccin vivant combiné contre le virus de la maladie de Newcastle (NDV) et le virus de la bronchite infectieuse (IBV) et ont été répartis de façon aléatoire entre 6 traitements (100 oiseaux dans chaque groupe) pour 5 semaines. Ces groupes ont été nourris avec la même nourritures naturellement contaminés par DON mais à des concentrations différentes : < 0,5 (régime témoin), 1,5 et 3 mg/kg avec et sans SFA à 250 g/tonne. Les paramètres, y compris le poids corporel, la consommation d'aliments, et le taux de mortalité, ont été enregistrés sur une base hebdomadaire. De plus, des échantillons de sang ont été prélevés sur quatre oiseaux de chaque groupe pour déterminer les caractéristiques biochimiques sériques, et les titres d'anticorps contre NDV et IBV. Au cours de la semaine 5, quatre des poulets sélectionnés ont été euthanasiés, et les organes, coeur, foie, rate, bourse de Fabricius, ont été pesés. Des tissus de l'intestin, segments de 2 cm du duodénum, du jéjunum, de l'iléon et du cæcum, et de la bourse de Fabricius et thymus ont été prélevés pour des examens histologiques. Les résultats obtenus démontrent que les oiseaux dont l’alimentation était contaminée au DON présentaient une diminution (P < 0,05) du poids moyen aux jours 28 et 35 par rapport aux groupe témoin et groupes traités avec le SFA. De plus, l'inclusion de DON dans l'alimentation a réduit le titre d'anticorps contre le NDV (P < 0,05) et l'IBV (P < 0,001). Les paramètres biochimiques sériques, et le poids des organes pendant toute la période expérimentale (P > 0,05) n’ont pas montré de différences significatives. De plus, chez les oiseaux nourris avec des aliments contaminés, le DON a provoqué un processus de nécrose au niveau des villosités du duodénum et du thymus. Cependant, cela n'a pas modifié la morphométrie du tissu intestinal et de la bourse de Fabricius. Il a été conclu que les performances des poulets de chair, ainsi que la réponse vaccinale et paramètres histologiques, étaient négativement affectées par une alimentation contaminée par le DON. La supplémentation alimentaire avec un additif alimentaire à base de synbiotiques, serait une alternative pour atténuer les effets causés par cette mycotoxine chez les oiseaux. / Deoxynivalenol (DON) is one of the most abundant and important trichothecene mycotoxins produced by Fusarium species. Mycotoxin-contaminated feeds may negatively affect broiler chickens’ health, a sustainable approach to achieve mycotoxin elimination is necessary. An experiment was conducted to study the effects of deoxynivalenol (DON) on the performance of broilers, organ weights, serum biochemical parameters, immune response, and intestinal histology and to evaluate the efficacy of a commercial feed additive, based on synbiotics (SFA), with the ability to deepoxidize DON. In total, six hundred 1-d-old male broiler of a commercial strain (Ross 308) were vaccinated with combined live Newcastle disease virus (NDV) and infectious bronchitis virus (IBV) vaccine and were randomly allotted to 6 dietary treatments (100 birds in each group) for 5 wk. These groups were fed 3 diets naturally contaminated with DON at concentrations of < 0.5 (control diet), 1.5, and 3 mg/kg with and without SFA at 250 g/ton. The parameters including body weight, feed intake, and mortality rate were recorded on a weekly basis. In addition, weekly blood samples were collected from four birds in each group to determine serum biochemical characteristics and antibody titers to NDV and IBV. In week 5, four selected chickens were euthanized, and organs (heart, liver, spleen, bursa of Fabricius) were weighed. Tissues from intestine (2-cm segments of duodenum, jejunum, ileum, and caecum) and immune system (bursa of Fabricius and thymus) were collected for further histological examinations. The results indicated that DON-challenged birds had decreased (P < 0.05) average body weight (ABW) at days 28 and 35 as compared to control and treated groups with SFA. Furthermore, the inclusion of DON in the diet reduced antibody titer against NDV (P < 0.05), and IBV (P < 0.001). However, there were no significant differences in serum biochemical parameters and organs weight during the whole experimental period (P > 0.05). Moreover, in birds fed contaminated diets, DON caused necrosis in duodenum villus and in the thymus but did not alter the intestinal or the immune system morphometrics. It was concluded that broiler performance, histological, and immunological parameters were adversely affected by feeding diets contaminated with DON. However, the dietary supplementation with Synbiotics as a detoxifying agent would be an alternative to alleviate adverse effects on birds.

Dissertation (PhD)--University of Stellenbosch, 2004 / ENGLISH ABSTRACT: Newcastle disease (NO) and mycoplasma infections in ostriches have considerable
economic implications for the South African ostrich industry in that NO is a limiting
factor in the export of ostrich products to the European Union and mycoplasma
infections cause stock losses, reduced production, reduced hatchability and
downgrading of carcasses. In the first section of this dissertation, the role of passively
acquired and mucosal immunity in protection of ostrich chicks against Newcastle
disease virus (NOV) was investigated. Ostrich hen serum IgG and yolk IgY were
isolated and characterized, and the transfer of maternal anti-NOV antibodies to the
egg yolk was determined using an enzyme-linked immunosorbent assay (ELISA).
Results indicated that anti-NOV antibodies were successfully transferred from the
ostrich hen to the egg yolk. In addition, ostrich IgA was isolated, characterized and
rabbit anti-ostrich IgA antibodies produced and used for measuring mucosal anti-
NOV IgA antibodies produced in response to mucosal vaccination. Results indicated
that the live La Sota vaccine stimulates IgA production and thus mucosal immunity in
ostrich chicks. In the second section of this dissertation, ostrich mycoplasmas were
isolated and identified using 16S rRNA gene sequencing. These sequences indicated
that ostriches carry three unique mycoplasmas, which are phylogenetically quite
divergent. The 16S rRNA gene sequences of the ostrich mycoplasmas were
subsequently used for the development of specific primers for the detection and
diagnosis of mycoplasma infections in ostriches by PCR.
The last section of this dissertation focuses on avian malaria in African penguins and
the management of this disease during rehabilitation. The Foundation for the
Conservation of Coastal Birds (SANCCOB) is a seabird rescue and rehabilitation
centre, which is largely dedicated to the rehabilitation of diseased, injured and oiled
penguins. Significant mortalities due to avian malaria occur at this facility. The aim of
this study was the development of an ELISA for the purpose of assessing the natural
levels of anti-Plasmodium antibodies in African penguins on entry into the SANCCOB
facility and during rehabilitation. Results indicated significant increases in anti-
Plasmodium antibody levels after entry, which was not influenced by oiling. Infection
with malaria and not parasite recrudescence was viewed to be the cause of this
increase, indicating a possible role of the SANCCOB facility in exposing penguins to
avian malaria. / AFRIKAANSE OPSOMMING: Newcastlesiekte (NS) en mikoplasmainfeksies in voltruise het geweldige ekonomiese
implikasies vir die Suid-Afrikaanse volstruisbedryf. Die rede hiervoor is dat NS 'n
beperkende faktor in die uitvoer van volstruisprodukte na die Europese Unie is, en
mikoplasmainfeksies tot kudde verliese, verlaagde produksie en uitbroei asook lae
gradering van karkasse lei. In die eerste gedeelte van hierdie proefskrif is die rol van
passiewe- en mukosale-immuniteit in die beskerming van volstruiskuikens teen NS
virus (NSV) ondersoek. Volstruishenserum IgG en eier IgY is geïsoleer en
gekarakteriseer en die oordrag van maternale anti-NSV antiliggame na die eier
ondersoek met behulp van 'n 'enzyme-linked immunosorbent assay' (ELISA).
Resultate het getoon dat anti-NSV antiliggame suksesvol van die hen na die eier
oorgedra is. Volstruis IgA is ook geïsoleer, gekarateriseer en konyn anti-volstruis IgA
antiliggame geproduseer wat gebruik is vir die bepaling van mukosale anti-NSV IgA
antiliggame in reaksie op mukosale immunisering. Resultate het getoon dat
lewendige La Sota entstof IgA produksie stimuleer en dus tot mukosale-immuniteit in
volstruiskuikens lei. In die tweede gedeelte van hierdie proefskrif is
volstruismikoplasmas geïsoleer en geïdentifiseer met behulp van 16S rRNA
geenopeenvolgingsbepalings. Hierdie volgordes het getoon dat drie unieke
mikoplasmas in volstruise voorkom wat filogeneties verskillend blyk te wees. Die 16S
rRNA geenopeenvolgings van die volstruismikoplasmas is gebruik vir die
ontwikkeling van spesifieke inleiers vir die PKR identifisering en diagnose van
mikoplasmainfeksies in volstruise.
Die laaste gedeelte van hierdie proefskrif fokus op voëlmalaria in die Afrika pikkewyn
en die bestuur van hierdie siekte gedurende rehabilitasie. Die 'South African
Foundation for the Conservation of Coastal Birds' (SANCCOB) is 'n seevoëlreddingsen

Enacting social accounting within a community enterprise : actualising hermeneutic conversationGibbon, Rebecca Jane January 2010 (has links)
The research was carried out using a participatory action research approach to develop social accounts with Jesmond Swimming Pool (JSP). The original motivation to carry out this project was to see what social accounts would look like and whether it was possible to develop them in this organisation. The experience of doing social accounts is further examined using Gray, et al., (1997) to explore whether these were either ‘ideal’ or ‘good’ social accounts for the organisation. A communitarian philosophical framework is used in order to examine the conceptualisation of ‘good’ social accounts. The first part of the thesis explores the social and environmental accounting (SEA) and accountability literature, with the second part exploring the experience of reaching initial agreement to do social accounts (SAs). The agreement to do the accounts was then followed by two years of social accounts developed with JSP. This empirical data provides a detailed account from the perspectives of insiders and other sources as to the experience of doing social accounts. This experience is then opened up to interrogation from a wider view point. The third part of the thesis examines the experience of JSP using third sector communitarian philosophy and a voluntary accountability framework in order to demonstrate that JSP could provide an example of a ‘good’ or ‘ideal’ social account. This raises the issue of whether or not ‘good’ or ‘ideal’ social accounts for third sector organisations are only possible within a communitarian paradigm. If it is possible to establish what ‘good’ social accounts entail then it may be appropriate to extend this approach to other contexts, for example, the public sector or the corporate world.

A Mutational Analysis of Structural Determinants Within the Newcastle Disease Virus Fusion Protein: a DissertationReitter, Julie N. 01 April 1994 (has links)
The fusion protein of the Newcastle Disease Virus (NDV) contains three hydrophobic domains. To explore the topogenic signals of these domains, mutants were constructed in which each of the hydrophobic domains was deleted. The membrane insertion and topology of these proteins was characterized in a wheat germ cell-free translation system supplemented with canine microsomal membranes. The results indicated that the first 13 amino acids of the fusion protein are necessary to confer translation inhibition by SRP. Translocation of the nascent chains containing all or part of the first hydrophobic sequence resulted in the appearance of a species of higher molecular weight consistent with glycosylation of at least four of the five potential N-linked glycosylation sites. When glycosylation was inhibited with a glycosylation competitor peptide, signal sequence cleavage was detected. Protease digestion of mutants missing the C-terminal hydrophobic domain indicated that the C-terminus has stop transfer activity. A comparison of membrane insertion of the wild-type fusion protein to that of a mutant missing the second hydrophobic domain, the fusion sequence, indicated that the fusion domain has stop-transfer activity when synthesized in vitro. Furthermore, the fusion domain shows little signal sequence activity when positioned near the amino terminus of the fusion protein.
The fusion protein has a highly conserved leucine zipper motif immediately upstream from the transmembrane domain of the F1 subunit. In order to determine the role that the conserved leucines have for the oligomeric structure and biological activity of the NDV fusion protein, the heptadic leucines at positions 481,488, and 495 were changed individually and in combination to an alanine residue. Whereas single amino acid changes had little effect on fusion, substitution of two or three leucine residues abolished the fusogenic activity of the protein although cell surface expression of the mutants and sedimentation in sucrose gradients was similar to that of the wild type. Furthermore, deletion of the C-terminal 91 amino acids, including the leucine zipper motif and transmembrane domain resulted in secretion of an oligomeric structure. These results indicate that the conserved leucines do not play a role in oligomer formation but are required for the fusogenic ability of the protein. When the polar face of the potential alpha helix was altered by nonconservative substitutions of a serine-to-alanine (position 473), glutamic acid-to-lysine (position 482) or an asparagine-to-lysine (position 485), the fusogenic ability of the protein was not significantly disrupted.
A phenylalanine residue is at the amino terminus of the F1 protein of all paramyxovirus fusion proteins with the exception of the avirulent strains which have a leucine residue in this position. To explore the role of this phenylalanine in the fusion activity of the protein, this residue was changed to leucine (F117L) or to glycine (F117G) by site-specific mutagenesis while maintaining the cleavage site sequence of virulent strains of NDV. Whereas both the wild-type and the F117G proteins were proteolytically cleaved and F1 was detected, the leucine subsitution abolished cleavage. When co-expressed with the HN protein, the fusion protein with either a phenylalanine and glycine residue at position 117, but not a leucine, was shown to stimulate membrane fusion. However, incubation in trypsin activated the fusion activity of the F117L protein. Thus the presence of a leucine at position 117 of the precursor sequence blocks cleavage, but not fusion acitivity, and indicated that the phenylalanine at the amino terminus of the F1 subunit is not conserved for the fusion activity of the protein.

Saudi Arabia has been accused of sportswashing, an attempt to improve its reputation through buying the English football club Newcastle United. The purpose of this paper is to explore through a discourse analysis how Saudi Arabia's action to acquire Newcastle United can be seen as an exercise of power, and whether and how the images presented construct and reinforce orientalist discourses about the Saudi state and the region. The discourse analysis is conducted using Laclau and Mouffe's methodology and the theoretical framework applied is Joseph Nye's soft power, Edward Said's Orientalism and hegemony. Based on the international newspaper The Guardian and the human rights organization Amnesty International, orientalist discourses have been identified and the purchase of Newcastle United has resulted in soft disempowerment effects for Saudi Arabia. Local newspaper The Chronicle also constructed orientalist discourses about the state of Saudi Arabia but legitimizes the purchase of Newcastle United making the soft power exercise successful.

The Membrane Integration of the Hemagglutinin-Neuraminidase Glycoprotein of Newcastle Disease Virus: A ThesisWilson, Cheryl Anne 01 May 1989 (has links)
The hemagglutinin-neuraminidase (HN) molecule of Newcastle disease virus (NDV) is an integral membrane glycoprotein that is oriented with its N-terminus in the cytoplasm and its C-terminus external to the infected cell. Single spanning membrane proteins with this type of topology (N-terminus in, C-terminus out) have been classified as Type II glycoproteins, in contrast to the more common Type I glycoproteins, which are oriented in the opposite direction. (C-terminus in, N-terminus out). The membrane integration of HN protein was investigated using a wheat germ translation system to synthesize and integrate HN protein into microsomal membranes in vitro. The insertion and translocation of HN protein into microsomal vesicles was found to occur cotranslationally without signal sequence cleavage. The membrane targeting required both signal recognition particle (SRP) and SRP receptor. Membrane binding assays utilizing HN nascent chain/ribosome/SRP complexes demonstrated that the membrane insertion of HN polypeptide required the presence of GTP, in a way similar to that described for secretory, multispanning and Type I proteins.
To investigate further the membrane translocation process of HN protein, the amino terminal region of HN was mutated to determine the role of this region in the membrane integration of HN. The cDNA sequence encoding the bulk of the cytoplasmic tail of the HN glycoprotein was deleted. When transcripts produced from the mutated cDNA were translated in wheat germ extract in the presence of membranes, several abnormalities were identified in the interaction of the mutant protein with membranes. Although translocation and glycosylation of the mutant protein was detected, the efficiency of membrane translocation and the stability of the mutant protein's membrane interaction were reduced. Even though a large proportion of the mutant products remained nontranslocated and unglycosylated, many of these products were inserted into membrane vesicles in a reverse orientation from the wild type HN protein. The aberrant insertion of the mutant protein required both SRP and SRP receptor. Ribosome-bound mutant nascent chains were able to insert into membranes without the addition of GTP or SRP, but this GTP-independent insertion was in reverse. Therefore, the cytoplasmic tail of the HN glycoprotein appears to playa critical role in the maintanence of faithful directionality of the protein's membrane insertion.

This study aims to conduct a framing analysis to explore how Saudi Arabia’s acquisition of Newcastle United can be perceived as an exercise of soft power and how the portrayed images in media construct and reinforce orientalist and eurocentric perspectives about Saudi Arabia and the region. This study analyzes the coverage from the media outlets The Guardian, Dagens Nyheter and the Saudi-based Al Arabiya. It focuses on how the media shapes and presents the image of Saudi Arabia in connection with the acquisition of Newcastle United and to what extent orientalist representations are reflected.
