Dispersions aqueuses de polyuréthanes bio-sourcés sans isocyanates / Aqueous dispersions of bio-based non-isocyanate polyurethanes

Les polyuréthanes forment une importante famille de polymère ; ils sont obtenus industriellement à partir d’isocyanates, des dérivés du phosgène. Dans l’optique de s’affranchir de ces composés toxiques et de promouvoir l’utilisation de la biomasse, cette thèse s’intéresse à la synthèse de dispersions aqueuses de polyuréthanes sans isocyanates et dérivés d’huiles végétales. Deux voies de synthèse de polyuréthane sans isocyanate sont étudiées : la transuréthanisation et l’aminolyse des carbonates cycliques. Des synthons biscarbamates et bis-carbonates cycliques dérivés d’acides gras ont ainsi été développés, et leur polymérisation en masse avec des diols ou diamines étudiée. Les deux voies de synthèse permettent d’obtenir des polyuréthanes pour des températures de 20 à 130°C en quelques heures ; le méthanolate de sodium est identifié comme un catalyseur efficace lors de la transuréthanisation tandis que l’aminolyse des carbonates cycliques ne nécessite pas de catalyse. Les masses molaires (Mn) obtenues sont de l’ordre de 5-17kg.mol-1, ces résultats sont cohérents avec la littérature. La polymérisation en mini-émulsion (aqueuse) a alors été utilisée pour la synthèse de polyuréthanes par aminolyse des carbonates cycliques. De nombreuses expériences de formulation des mini-émulsions ont permis d’aboutir à des mini-émulsions et latex de polyuréthanes stables. / Polyurethanes are a major polymer family; they are industrially obtained from phosgene derivatives: isocyanates. In order to avoid the use of such toxic compounds and to promote the use of biomass, this thesis investigates the synthesis of aqueous dispersions of nonisocyanate polyurethanes derived from vegetable oils. Two synthesis pathways have been studied; transurethanization and aminolysis of cyclic carbonates. Bis-carbamates and biscyclic carbonates were synthesized from fatty acids, and their polymerizations with diols or diamines were studied in bulk. The two routes allow the production of polyurethanes in a few hours at 20-130°C. Sodium methoxide is used as catalyst for transurethanization reactions while the other synthesis pathway does not require catalysts to proceed. Polyurethanes obtained have molar mass (Mn) around 5-17kg.mol-1, which is in accordance with the literature. The polymerization in aqueous miniemulsion was then investigated for the synthesis of polyurethane by aminolysis of cyclic carbonates. Many formulation experiments were necessary to achieve stable miniemulsion and latex; aqueous dispersions of bio-based non-isocyanate polyurethanes were then obtained.

Bisher getestete Knochenkleber zeigen häufig geringe Klebeeigenschaften auf Knochen bei Zutritt von Feuchtigkeit. Gegenstand dieser Arbeit war es, die Haftfähigkeit im feuchten Milieu zu verbessern. Hierfür wurde der Einfluss sternförmiger, mit Isocyanaten funktionalisierter Poly(ethylenglykol) Moleküle (NCO-sP(EO-stat-PO)) auf die Klebefestigkeit und Alterungsbeständigkeit einer photopolymerisierbaren Poly(ethylenglykol)dimethacrylat-Basis (PEGDMA) untersucht. Die Polymerisation mittels energiereicher Strahlung erlaubt hohe Reaktionsraten bei Körpertemperatur sowie zeitliche und örtliche Kontrolle über die Polymerisationsreaktion. Durch den Zusatz degradierbarer, keramischer Füllstoffe auf Calciumsulfat- und Magnesiumphosphat-Basis in die Matrix sollten durch Lösungsprozesse Poren geschaffen werden. Diese könnten das Einwachsen neuer Knochensubstanz in das ausgehärtete Material ermöglichen. Die Veränderungen der kristallinen Strukturen wurden mittels Röntgendiffraktometrie beobachtet. Zudem wurden die Proben infrarotspektroskopisch und mikroskopisch untersucht. Die Klebefestigkeit auf kortikalem Rinderknochen im Abscherversuch ebenso wie die Biegefestigkeit vor und nach Lagerung in feuchter Umgebung wurde unter Variation des NCO-sP(EO-stat-PO)-Gehaltes ermittelt. Anschließend sollten die mikroskopische Analyse und energiedispersive Röntgenspektrometrie (EDX) Aufschluss über das Bruchverhalten des Materials beim Klebeversuch geben. Es konnte gezeigt werden, dass durch die Zugabe von 20 bis 40 Gew.-% NCO sP(EO-stat-PO) zur Matrix die Klebefestigkeit auf Knochen von initial etwa 0,15 bis 0,2 MPa auf etwa 0,3 bis 0,5 MPa gesteigert werden kann. Während alle Referenzproben ihre Haftung an Knochen innerhalb von weniger als 24 Stunden verloren, zeigten Proben mit NCO sP(EO-stat-PO) auch nach 7-tägiger Lagerung noch Festigkeiten von 0,18 bis 0,25 MPa. Die höchste Festigkeit nach 7 Tagen war bei Proben mit dem Füllstoff Newberyit und einem NCO-sP(EO-stat-PO)-Anteil von 40 Gew.-% zu verzeichnen. Diese Proben wiesen auch in der mikroskopischen Analyse und im EDX eindeutig ein rein kohäsives Versagen auf. 20%-ige Proben zeigten zumindest in geringem Maße auch adhäsives Versagen. Die 3-Punkt Biegefestigkeit lag initial bei 3,5 bis 5,5 MPa. Durch die Lagerung in PBS sank die Festigkeit auf ~1 MPa. Die Zugabe von NCO-sP(EO-stat-PO) und die Art des eingesetzten Füllstoffes hatten kaum einen Einfluss auf diese. / Bone adhesives often show a significant loss in adhesive strength to bone when used in a wet environment. The aim of this work was to improve the adhesion to bone under wet conditions by addition of star-shaped, isocyanate-functionalized poly (ethylene glycol) molecules (NCO-sP (EO-stat-PO)) and to investigate the influence on bond strength and aging resistance of a photopolymerizable poly (ethylene glycol) dimethacrylate base (PEGDMA). Polymerization by high-energy radiation allows high reaction rates at body temperature as well as temporal and local control over the polymerization reaction. The addition of degradable, ceramic calcium sulfate and magnesium phosphate based fillers into the matrix was supposed to create pores through solution processes. These could allow the ingrowth of new bone into the cured material. The changes of the crystalline structures were observed by X-ray diffractometry. In addition, the samples were examined microscopically and by infrared spectroscopy. The adhesive strength on bovine cortical bone in shear tests as well as the flexural strength before and after storage in a wet environment was determined by varying the NCO-sP (EO-stat-PO) content. Subsequently, the microscopic analysis and energy dispersive X-ray spectrometry (EDX) should provide information about the failure mode of the material. It could be shown that by the addition of 20 to 40 wt .-% NCO sP (EO stat PO) to the matrix, the adhesive strength to bone can be increased from initially about 0.15 to 0.2 MPa to about 0.3 to 0,5 MPa. While all reference samples lost adhesion to bone within less than 24 hours, samples containing NCO sP (EO-stat-PO) still showed bonding strengths of 0.18 to 0.25 MPa after 7 days of storage. The highest bonding strength after 7 days was observed in samples with newberyit fillers and an NCO-sP (EO-stat-PO) content of 40 wt .-%. These samples also clearly showed cohesive failure in microscopic analysis and EDX, while samples containing 20% wt.-% showed at least slight adhesive failure. Initially tested, the 3-point bending strength was about 3.5 to 5.5 MPa. After storage in PBS, the strength decreased to ~ 1 MPa. The addition of NCO-sP (EO-stat-PO) and the type of filler used had only little effect on strength loss.

Synthesis of Beta-Aminocarbonyl Compounds and Hydrazine Derivatives Using Amino- and Imino-Isocyanates

Over the past recent years, β-aminocarbonyls have been of great interest to medicinal chemists. As a practical method to obtain these moieties, alkene aminocarbonylation, accounting for the formation of a C-N and a C-C bond, has been the subject of limited research efforts (very specific intramolecular metal-catalyzed variants have been reported). Direct aminocarbonylation of alkenes constitutes a challenging and an important potential innovation in the synthesis of β-aminocarbonyls such as β-amino acids. The research efforts described in the present thesis have been primarily directed towards the development of concerted pathways for the amination of alkenes using hydrazine derivatives as bifunctional reagents. Building on our previous report on the reactivity of hydrazides, progress on the aminocarbonylation of alkenes along with the synthetic scope of this reactivity are herein provided. Therefore, the first part of the present thesis (Chapter 2) focuses primarily on the development of thermolytic conditions for the intramolecular aminocarbonylation of alkenes using amino-isocyanates. Alongside, development of imino-isocyanates have provided complementary synthetic tools for aminocarbonylation. The second part (Chapter 3) describes the work accomplished towards intermolecular aminocarbonylation of alkenes and the synthesis of complex azomethine imine products (Chapter 3). Finally, the last part of the discussion (Chapter 4) will be on the development of new hydrazide reagents for the intramolecular Cope-type hydroamination of alkenes. In doing so, description of the synthetic utility of amino-isocyanates as amphoteric reagents for cascade reactions and heterocyclic synthesis will be provided.

Intermolecular [3+2] Cycloadditions of Imino-isocyanates to Access β-Amino Carbonyl Compounds

In modern synthetic organic chemistry, chemists are driven to develop efficient methods for important C-C and C-N bond formation reactions. The challenge lies with establishing new uses for readily available substrates. In this regard, the synthesis of β-aminocarbonyl compounds from alkenes remains a long-standing challenge. Innovation in reaction discovery often requires finding new reagents, or rare reagents with underappreciated value in synthesis. In Chapter 1, N-isocyanates and other heterocumulenes are introduced as versatile amphoteric reagents. Their amphoteric properties are valuable in the discovery of new synthetic approaches, especially in cycloaddition reactions. While C-isocyanates are bulk industrial chemicals, the formation and reactivity of N-isocyanates remains underexplored. Chapter 2 describes the development of reactivity with rare imino-isocyanates. This includes methods to access the reagent in situ with a blocking group approach, and the establishment of intermolecular cycloaddition reactivity with a variety of alkenes. This stereospecific reaction provides complex N,N’-cyclic azomethine imines, and enables access to β-aminocarbonyl compounds from alkenes. β-Amino amides and esters, pyrazolidinones, and pyrazolones were accessed by reductive derivatization of the aminocarbonylation products. Exploration into the limits of this reactivity gave insight into fundamental properties of imino-isocyanates. This includes the first detection of imino-isocyanates by IR spectroscopy. A kinetic resolution of the azomethine imines obtained from this alkene aminocarbonylation reaction was then developed, which gave access to enantioenriched β-amino carbonyl compounds (Chapter 3). This was accomplished by Brønsted acid catalysed reduction, with a selectivity factor of 13-43. This was the first example of the enantioselective reduction of azomethine imines, and represents a new activation mode for reactions of N,N’-cyclic azomethine imines. Using this reductive method, both enantiomers of the β-amino amide could be obtained from a racemic azomethine imine in ≥ 97% ee. The discovery of new reactivity of imino-isocyanates with imines in described in Chapter 4, which allowed the synthesis of eight new azomethine imines with the triazolone core. Our initial scope studies revealed different trends with imines than with alkenes, including increased reactivity, which led to investigation of the mechanism of this reaction. In addition, this was shown to be a valuable new approach for the synthesis of triazolones from imines.

Biomarkers of internal exposure/dose : Methods to quantify adducts to protein and DNA by LC/MS studied with benzo[a]pyrene and isocyanates

This thesis focuses on methods for quantification by liquid chromatography/mass spectrometry (LC/MS) of specific biomarkers for internal dose of chemicals which induce toxicity through their electrophilic reactivity. In vivo such compounds are short-lived, and could feasibly be measured as their reaction products (adducts) with biomacromolecules. Analysis by MS methods of stable adducts offers the specificity and accuracy required to generate data on internal dose useful in risk estimation. The primary aim was to develop a method for quantification by LC/MS of bulky adducts to serum albumin (SA) from polycyclic aromatic hydrocarbons, using the genotoxic diolepoxide (DE) of benzo[a]pyrene (BP) as a model. A method for analysis of the BPDE adducts to His146 in SA was developed which is robust, easy-to-use, has good reproducibility and which reached a high sensitivity. A method for quantification of BPDE adducts to N2-deoxyguanosine (dG) in DNA by LC/MS was also established. In mice exposed to BP, adducts to SA and DNA from stereoisomers of BPDE were identified and quantified. The adduct level was shown to be &gt;400 times higher in DNA than in SA, which from an in vitro study could be concluded to mainly depend on a large difference in the rates of adduct formation to His in SA and to dG in DNA. BPDE adduct levels to SA and DNA, and a biomarker of genotoxic effect (frequency of micronuclei), were compared in BP-exposed mice. The results were used to evaluate how these methods could be used in procedures for cancer risk estimation. An LC/MS method for analysis of valine hydantoins (VH) formed as adducts from isocyanates to N-termini in haemoglobin was established. VH, formed from urea/isocyanic acid, was investigated in mice as a potential biomarker of renal failure and for dose adjustment during treatment with a radioactive cytostatic drug. Fast Curing Phenol Formaldehyde and Isocyanate Based Hybrid Resin for Forest Products Application

The objective of this study is to develop a fast curing phenolic formaldehyde (PF) and polymeric diphenyl methane isocyanate (MDI) based hybrid resin system for wood products applications. Various formulas of PF resins were synthesized with different formaldehyde to phenol ratio, sodium hydroxide to phenol and isocyanate group (-NCO) to hydroxyl group (-OH) molar ratios. The shear bonding strength property was used to evaluate and optimize the formulations by appropriate sample preparation. The optimized resins were characterized by rheometer, Fourier transform infrared spectroscopy (FTIR) and other methods. In order to eliminate the influence of hydroxyl groups from water in the PF resin, a frozen dried method was applied to remove the water while keeping PF resin in liquid state. Acetone was used to disperse the hybrid co-polymer to improve the mobility of the mixture of frozen dried PF resins and MDI. An unexpected phenomenon was observed when the two resins were mixed in acetone and a sharp reaction occurred. This led to an assumption that acetone promoted the curing of hybrid resin. The effect of acetone on the curing behavior of hybrid resin was studied by differential scanning calorimetry (DSC), confocal laser scanning microscope (CLSM) and other techniques. It was confirmed that acetone promoted the curing of the co-polymer system. The gel time of hybrid resin with acetone decreases sharply compared to that of pure phenolic resin and original hybrid resin. Acetone also helped the hybrid resin to have better penetration behavior by improving the mobility and this also resulted in less variation of the strength distribution. Finally, lap shear samples were prepared at room temperature curing commercial polyurethane (PU), phenol-resorcinolormaldehyde resin (PRF) and laboratory made hybrid resins based on PF and MDI to compare the shear strength of different resins under different application conditions. The strength reduction of frozen dried PF with acetone/MDI is the lowest in humidity, temperature and humidity-temperature conditions. With just humidity condition, its shear strength reduction is significantly lower than that of any other resins including the solid wood control.

Species Dependence of pMDI/Wood Adhesion

Polymeric methylenebis(phenylisocyanate) (pMDI) has increasingly been used in the wood particulate composite industry. Wood composites, especially oriented strand board (OSB) are made with many variations of wood species. Little research has been done to investigate how pMDI adhesion has been affected by species. The present is divided into two parts. First, mode I fracture mechanics and surface free energy analysis was performed to investigate differences in adhesion between southern yellow pine and yellow-poplar bonded with pMDI. Secondly, an improvement in the synthesis of 13C, 15N labeled pMDI is discussed. Mode I fracture results show that pMDI adhesion was affected differently by southern yellow pine compared yellow poplar. The shear energy release rate was significantly higher in pine/pMDI composites than in yellow poplar/pMDI composites. The total surface energy of southern yellow pine was shown to be significantly greater than yellow poplar. The free energy of adhesion (DG) of the pine/pMDI and the poplar/pMDI was investigated. The DG indicated that the pine/pMDI system would take more energy to separate compared to the poplar/pMDI system. Lastly, a double-labeled 13C, 15N pMDI adhesive was successfully synthesized to produce Solid-State NMR composites. / Master of Science

The addition of isocyanates to C=N double bonds of imines gives triazindione heterocycle structures; their thermal properties are reported. Mono-isocyanates were used to form 2:1 adducts with the imine bases 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU), 1,5-diazabicyclo[4.3.0]non-5-ene (DBN) and 2-tert-butyl-1,1,3,3-tetramethylguanidine (tBuTMG). A 2:1 stoichiometry of the adducts was proven by NMR and IR spectroscopy, and single crystal X-Ray diffraction; certain cleavage temperatures (70 and 160 °C) were measured. Thermal analysis (TG-MS) of adducts indicates the release of free isocyanate during adduct cleavage. Furthermore, a new class of step-growth oligomers (MN = 750–7,000 g∙mol–1) composed of multi-functional isocyanates and these imine bases was introduced. Their systematic spectroscopic and thermal analysis is shown revealing the similarity in their chemical properties to the 2:1 adducts. Actuellement, les polyuréthanes (PUs) sont produits industriellement par polyaddition entre un diisocyanate et un polyol. Le caractère fortement sensibilisant des isocyanates, mettent ces composés sous forte pression réglementaire au niveau européen (REACH) et ont créé le besoin d'obtenir des PUs ne provenant pas d'isocyanates, lesquels sont plus communément appelés Non-Isocyanate PolyUréthanes (NIPUs). Les travaux de cette thèse visent ainsi à établir de nouvelles voies d'accès à des NIPUs. Des pré-polymères téléchéliques ont été synthétisés via la réaction de métathèse. Plusieurs groupements terminaux (jusqu'à 16), réagissant à température ambiante avec une amine primaire, ont été greffés avec succès à ces pré-polymères. La réaction avec plusieurs diamines a conduit à la synthèse de nouveaux matériaux NIPUs entièrement caractérisés. Les stratégies de synthèses développées au cours de ces travaux de thèse ouvrent de nouvelles perspectives quant à l'industrialisation de NIPUs. / Conventional polyurethanes (PUs) involve the use of isocyanates, which are considerably toxic and require phosgene for their manufacture. To tackle environmental issues, it is necessary to elaborate different routes to PUs. In this context, two isocyanate-free strategies towards the preparation of polythiourethanes (PTUs), i.e. non-isocyanate polyurethanes (NIPUs), have being developed. The first way involves the synthesis of α,ω-di(dithiocyclocarbonate) telechelic poly(propylene glycol) (bis(5DTCC)-PPG), poly(tetrahydrofurane diglycidyl ether) (bis(5DTCC)-PTG), upon chemical modification of the corresponding α,ω-diepoxide telechelic polymers (PPG, PTG, respectively) through cycloaddition of carbon disulfide. The second approach involves the ring-opening metathesis polymerization (ROMP), using Grubbs’ 2nd generation ruthenium catalyst, of cycloolefins using 16 differents chain-transfer agents. Bis(5DTCC) telechelic copolyolefins are thus synthesized. Reaction of the end-capping 5DTCC moieties with a diamine by ring-opening polyaddition ultimately affords at room temperature the corresponding NIPTU.
