Sociální a lidský kapitál jako faktor rozvoje periferních oblastí Česka / Social abd human capital as a factor of development of peripheral areas of Czechia

Within the framework of the research focused on the polarization of space, or peripheral areas of Czechia respectively, approaches of cultural geography have been applied to a significant extent recently. As far as the issue of such areas development is concerned, the importance of the so- called "soft" socio-cultural development factors (including social and human capital) has been accentuated. The way-out is the belief that with the limited development potential of the peripheral (and rural) areas, activity and activism of local communities (quality of social and human capital) as the important internal (endogenous) development factor become more important. Despite the number of attempts, which have been made, we are not able to find exhaustive answers to questions relating to the actual importance of social and human capital (and different forms thereof) in the (socio-economic) development of these areas or the mutual relation (conditionality) thereof. For this reason, problems can be seen both in the area of theory and methodology (conceptualization) and, in particular, in insufficient verification of such theoretical concepts by means of empirical research, be specific in particular at the micro-regional up to local level. Inactivation of tumor supressor gene BRCA1 causes a life-long risk of breast carcinoma development. Genetic screenings of indicated individuals from high-risk families help to identify large number of sequence variants in known predisposing genes. Majority of discovered variants doesn't have clinical significance yet which causes a big problem for diagnostics. Some of these variants are found within regulatory non-coding regions of gene. A part of the clinical classification of variants is their functional characterization. The goal of this thesis was to create a model system for functional characterization of variants in non-coding regions and to verify its function. Model system was based on targeted gene manipulation by co-transfecting CRISPR-Cas9 construct and donor construct that contained a portion of BRCA1 gene sequence with analyzed modifications, into U2 OS cells. The cells have stably integrated DR-GFP system which allows the activity of homologous recombination (HR) to be determined. Monoallelic modifications were induced into U2 OS cells. These modifications were in a Kozak sequence region of BRCA1 gene. Analýza miRNA u nádorů asociovaných s lidskými papilomaviry / Analysis of miRNAs in HPV-associated carcinomas

Papillomaviruses are small DNA viruses that are associated with the induction of epithelial tumors. HPV is an important infectious agent causing almost 100 % of cervical tumors but it can also cause tumors in other anogenital and head and neck locations in both men and women. Active HPV infection induces changes in miRNA expression that contribute to the tumor formation and progression. It is already known that papillomaviruses do not encode their own viral miRNAs but they affect the expression of cellular miRNAs. In my thesis I have in selected epithelial tumors (vulva, cervix, anus and tonsils) determined their etiology and analyzed the presence of miRNAs in tissues by next generation sequencing. From these data I determined the expression profiles of deregulated miRNAs in tumors relation to healthy tissues of corresponding location. Even though, sufficient number of samples was analyzed, it was not possible to detect HPV-core miRNA common to all analyzed HPV-induced tumors due to the absence of statistically relevant differentially expressed miRNAs in HPV positive vulvar tumors. The aim of this thesis is to investigate gas interdependence for political relations between the European Union and the Russian Federation. Despite of the fact that relations in the gas sphere still have particular importance for both parties, after the Ukrainian crisis, sanctions and counter-sanctions applied, they have reached the lowest point of cooperation since the end of the Cold War. This thesis is based on the interdependence model of international relations theory, asymmetries in sensitivity and vulnerability of the EU and Russia were connected to their strategies as part of the political power play. Therefore, conflict situation, high level of sensitivity and vulnerability, and interest in finding alternatives of both parties lead to the situation of 'negative interdependence'. In this context, this kind of interdependence where partners would rather avoid relations logically pushes the EU and Russia apart and stimulates reconsideration of their relationship in gas sphere.

This thesis deals with the conflict of nature and the landscape protection with recreational use of nature. This conflict is examined in relation to "conservationist subject" and "bivouacking subject" in the context of ecogovernmentality, i.e. the shift of nature to the government sphere. The aim of this thesis is to introduce more of cosmology of nature protection, besides the dominant one - conservational, and to understand their relation. The thesis is divided into two parts - a theoretical part and an empirical part. The theoretical part is devoted to the theory of subjectivity and the concept of ecogovernmentality in the context of chosen ethnographic studies. The empirical part is dedicated to ethnographic research of bivouacking subjects. The results of the research are based on the comparison of data acquired from the ethnographic research with abstracted figures of "conservationist subject" - as defined by visitor rules of the protected areas and the law of nature and landscape protection and "bivouacking subject" - made by stays in nature or scout education. (in English): The diploma thesis examines the system of accompanying of foster families as a profession, which is done by the so called Key workers. The first part of the thesis focuses on foster care, its typology and the overview of foster care benefits. The accompanying system is presented in detail, including a legislative overview and the content of the foster care agreement. The theoretical part specifies the roles, prerequisites and working conditions of the Key workers and discusses the limits of this profession. The aim of the thesis is to find out how these workers evaluate the current system of accompaniment in selected organizations which are authorized to conclude foster care agreements in Prague and Central Bohemia. The empirical part includes research, analysis, and evaluation. Methodology of the research is qualitative and the data is collected by semi-structured interviews.

This diploma thesis is focused on corporate social responsibility in selected multinational organizations in the information technology sector, namely on Apple, Google, IBM and Microsoft. Recommendations for those organizations are based on qualitative analysis. Based on quantitative analysis and selected organizations recommendations were developed recommendation for small and middle sized IT companies in Czech Republic. Research has shown that multinational IT organizations are socially responsible. They are paying attention to all areas of CSR and are leaders in enviromental field.

Understanding the mechanisms of transcriptional regulation and the constraints that operate in gene promoter sequences is the key step in understanding the evolutionary conservation of transcriptional regulation. It is well known that regulatory regions with the same expression outputs do not have to share the sequence similarity. The most important elements in regulatory sequences are transcription factor binding sites and their position relocation does not usually influence the expression output. The least complex transcriptional regulation is characteristic for housekeeping genes. For their expression they require only basal core promoter elements (sometimes only CpG islands are sufficient) and general transcription factors, so they can be transcribed easily and immediately whenever they are needed. In this study we focused on transcriptional regulation of invertebrate amphioxus (Branchiostoma floridae) housekeeping genes in vertebrate systems. We prepared a set of constructs with amphioxus regulatory regions for testing their activity in different mammalian cell lines and a set of constructs with the same amphioxus regulatory regions for observing their spatial recognition in developing medaka fish embryo. It focuses on the development between the last two censuses, of the fastest growing metropolitan areas in the United States. The economic crisis of the late 2000s has been a major turning point of the decade. The goal of the research was to create a developmental typology of the fastest growing metros. The text analyzes "classifiers" data in order to decide what developmental types have been among the metros. A major factor contributing to high population gains in the fastest growing metros was the migration induced by the housing bubble.

The diploma thesis deals with the image of a social worker in the Czech media. More specifically, it deals with how social workers are described in the press, television and the radio. The work has two goals. The first goal searches for the widest possible image of a social worker in the media. The second goal seeks to study how social workers participate in the media image and how they think about the media in relation to their work. The work consists of a theoretical and a practical part which are connected. The theoretical part forms the main basis for the second, research part of the work. It begins by defining the media, their functions, types, effects. It also touches on public opinion and the term 'social reality' which is an important part of describing the media image and its understanding. A necessary chapter of the theoretical part is also a description of the media product in the application of content analysis. Finally, the social worker and his media skills are described. The practical part is accompanied by two research methods. With the help of content analysis, the work tried to find out the widest possible picture of social workers, which consists of the press, television and the radio.
