Les activités humaines sont responsables de l’apport de nombreux micropolluants vers les systèmes aquatiques parmi lesquels les substances poly- et perfluoroalkylées (PFAS) ont été identifiées. Ces molécules sont utilisées depuis les années 1950 comme tensio-actifs dans de nombreuses applications industrielles et produits d’usage courant. Depuis deux décennies, certaines de ces substances, les acides perfluoroalkylés (PFAA) ont particulièrement attiré l’attention en raison de leur caractère ubiquiste et persistant dans l’environnement. Actuellement, il existe un réel manque de connaissance sur l’intégralité de la contamination environnementale par l’ensemble des PFAS. L’objectif global de ces travaux de thèse est de poursuivre les efforts menés depuis le début des années 2000 pour mieux comprendre la dynamique des PFAS depuis leurs sources en milieu urbain, leurs rejets dans les rivières et leur transfert trophique.La première partie consiste à optimiser une configuration de l’échantillonneur passif POCIS (Polar Organic Chemical Integrative Sampler) pour l’analyse ultra-trace de 25 PFAS dans les eaux de surface. Dans un second temps, il est question d’identifier les sources en PFAS sur la métropole de Bordeaux. L’analyse de 30 PFAS dans les eaux usées collectées en raiseau d’assainissement tend à montrer l’importance des apports industriels par rapport aux rejets domestiques pour la quasi-totalité des PFAS étudiés. A noter que les eaux de ruissellement sont également vectrices de contamination en PFAS. La caractérisation des effluents d’entrée et de sortie des quatre principales stations d’épuration des eaux usées (STEU) de la métropole met en évidence une faible efficacité des filières de traitement. L’utilisation d’une approche d’analyse non ciblée par oxydation, la méthode TOP (Total Oxidizable Precursors) révèle des quantités non négligeables de précurseurs de PFAA aussi bien en réseau d’assainissement que dans les effluents de STEU. L’impact de l’exutoire collectant les eaux de ruissellement en provenance de l’aéroport et d’une portion du périphérique de Bordeaux sur les niveaux en PFAS d’une petite rivière périurbaine est également montré. Enfin, la dernière partie renseigne la bioamplification des PFAS en milieu lotique. L’évaluation des concentrations le long de 5 réseaux trophiques du bassin hydrographique Rhône-Méditerranée permet d’une part de confirmer la bioamplification du perfluorooctane sulfonate (PFOS) et des perfluoroalkyles carboxylates (PFCA) à chaîne longue, et d’autre part d’évaluer la variabilité spatiale des facteurs d’amplification trophique (TMF). Le caractère bioamplifiable de PFAS d’intérêt « plus émergent » tels que les 8:2 et 10:2 fluorotélomères sulfonates est mis en évidence dans une rivière localisée à la périphérie de Paris. L’application de la méthode TOP à différents maillons de cette chaîne trophique permet de soutenir l’hypothèse de l’implication de la biotransformation des précurseurs dans la bioamplification apparente des PFAA. / Human activities are responsible for the release of multiple micropollutants into aquatic systems, such as poly- and perfluoroalkylated substances (PFASs). These molecules have been used since the 1950s as surfactants in many industrial applications and commonly used products. For two decades, some of these substances, perfluoroalkylated acids (PFAA), have generated a major concern due to their ubiquitous and persistent behavior in the environment. Currently, there is a real lack of knowledge about the full extent of environmental contamination by all PFASs. The overall objective of this thesis is to continue the efforts undertaken since the early 2000s to gain a better understanding of the dynamics of PFASs, from their sources in urban areas, their releases to aquatic systems, to their trophic transfer.The first part consisted in optimizing a configuration of the Polar Organic Chemical Integrative Sampler (POCIS) for the ultra-trace analysis of 25 PFASs in surface water. In a second time, the dynamics of the PFASs on the Bordeaux conurbation is studied. The analysis of 30 PFASs in wastewater collected in the sewerage network tends to show the importance of industrial inputs compared to domestic discharges for almost all the studied PFASs. Note that urban runoff is also a source of PFAS contamination. The characterization of the influents and effluents of the four main wastewater treatment plants (WWTP) in the metropolis shows a low efficiency of treatment channels. The use of a non-targeted analysis approach by oxidation, the TOP method (Total Oxidizable Precursors) reveals significant amounts of PFAA precursors in the sewerage network as well as in WWTP effluents. The impact of urban and airport storm water discharge on the contamination levels of a small peri-urban river has also been shown. The last part dealt with the biomagnification of PFASs in lotic systems. The evaluation of PFAS concentrations along 5 food webs from the Rhone-Mediterranean watershed enables to confirm the biomagnification of perfluorooctane sulfonate (PFOS) and long-chain perfluoroalkyl carboxylates (PFCA) and to evaluate the spatial variability of trophic magnification factors (TMF). The biomagnification character of PFASs of "more emerging" interest, such as the 8:2 and 10:2 fluorotelomer sulfonates, is observed in a river located on the outskirts of Paris. The application of the TOP method to different trophic levels supports the hypothesis of the involvement of the biotransformation of precursors in the apparent biomagnification of PFAAs.

Synthesis of Diazonium (Perfluoroalkyl) Arylsulfonimide Monomers from Perfluoro (3-Oxapent-4-ene) Sulfonyl Fluoride for Proton Exchange Membrane Fuel Cell

Two diazonium perfluoroalkyl arylsulfonimide (PFSI) zwitterionic monomers, 4-diazonium perfluoro(3-oxapent-4-ene)benzenesulfonimide (I) and 4-(trifluoromethyl)-2-diazonium perfluoro(3-oxapent-4-ene)benzenesulfonimide (II) have been synthesized from perfluoro(3-oxapent-4-ene) sulfonyl fluoride (POPF) for proton exchange membrane fuel cells. PFSI polymers are proposed as new electrolytes due to their better thermal stability, inertness to electrochemical conditions, and lower susceptibility to oxidative degradation and dehydration. For a better integration between the electrode and the electrolyte, the PFSI polymers are expected to be grafted onto the carbon electrode via the diazonium moiety. All the reaction intermediates and the final product were characterized with 1H NMR, 19F NMR and IR spectroscopies.

3-Diazonium- 4-(trifluorovinyloxy) - perfluorobutanesulfonyl benzenesulfonimide zwitterionic monomer (see figure 1) is proposed to be polymerized and further act as a new electrolyte for Polymer exchange membrane fuel cells (PEMFCs). One reason is that, the aromatic trifluorovinyl aryl ether (TFVE) group can easily be homopolymerized to aromatic perfluorocyclobutane (PFCB) polymer. Furthermore, the diazonium moiety in the monomer is expected to covalently attach the electrolyte to the carbon electrodes support. The perfluoroalkyl(aryl) sulfonimide (PFSI) pendant provides good chemical and mechanical stability as well as better proton conductivity. Several multi-step synthetic schemes are designed to obtain such monomer from perfluoroalkyl(aryl) sulfonimide (PFSI). Among them, the purified coupling product 4-OCF2CF2Br-3-NO2-PhSO2(M) SO2C4F9 from the first approach was successfully completed. The next stages of the work will involve dehalogenation, reduction, and diazotization to achieve the targeting monomer. All the intermediates were characterized by 1H and 19F NMR and FT-IR spectroscopy.

Prenatal Exposures to Perfluoroalkyl Acids and Serum Lipids at Ages 7 and 15 in Females

Background In some cross-sectional epidemiologic studies the shape of the association between serum concentrations of perfluoroalkyl acids (PFAAs) and lipids suggests departures from linearity. Objectives We used statistical approaches allowing for non-linearity to determine associations of prenatal exposures of perfluorooctane sulfonic acid (PFOS) and perfluorooctanoic acid (PFOA) with lipid concentrations. Methods PFAAs were measured in serum from pregnant women collected in 1991–1992 at enrollment in the Avon Longitudinal Study of Parents and Children and lipids in serum from their daughters at ages 7 (n = 111) and 15 (n = 88). The associations of PFAAs with lipids were first explored by cubic splines, followed by piecewise linear regressions by tertiles to obtain regression coefficients (β) and their 95% confidence limits (95% CL) (in mg/dL per 1 ng/mL). Results At age 7, total cholesterol was positively associated with prenatal PFOA concentrations in the lower tertile (β = 15.01; 95% CL = 2.34, 27.69) but not with PFOA concentrations in the middle (β = − 3.63; 95% CL = − 17.43, 10.16) and upper (β = − 1.58; 95% CL = − 4.58, 1.42) tertiles. At age 15, a similar pattern was noted as well. Positive associations between LDL-C and prenatal PFOA concentration in the lower tertile were observed in daughters at ages 7 (β = 14.91; 95% CL = 3.53, 28.12) and 15 (β = 13.93; 95% CL = 0.60, 27.26). LDL-C was not associated with PFOA concentrations in the middle or upper tertile at any age. Neither HDL-C nor triglycerides was associated with prenatal PFOA exposure. Non-linear patterns of association of total cholesterol and LDL-C with prenatal PFOS were less consistently noted. Conclusion Exposure to low levels of PFOA during prenatal development may alter lipid metabolism later in life. Given the small sample size further replication of the association in large independent cohorts is important.

Per- and polyfluoroalkyl substances (PFASs) is a group of man-made, highly persistent chemicals. Due to the specific surface-active attributes of these molecules, applications are numerous and feed an economically important industry. During the last decade, PFASs have been detected globally in the environment, living organisms and tap water. The combination of toxic properties and high bioaccumulative potential, together with the discovery that conventional water treatment methods do not remove PFAS, renders further research on purification methods highly needed.  Three techniques of purifying water from PFASs were examined. Nanofiltration technology (NF) is a membrane filtration technique, which produces a purified product (the permeate) by generating an effluent of high contaminant concentration (the reject water). To decontaminate the reject water, adsorption by granular activated carbon (GAC) or anion exchange (AE) have been proposed. The efficiency of these three technologies was studied at Bäcklösa drinking water treatment plant (DWTP) in Uppsala. A nanofiltration pilot with two 270NF membranes (Dow Filmtech™), connected in series, was used. A high removal efficiency (>90%) was found for all PFASs. Furthermore, it was confirmed that the concentration in the permeate water was a function of the concentration in the incoming raw water; increased PFAS raw water concentration resulted in increased PFAS permeate concentration. Size-exclusion and electrostatic repulsion were deemed important mechanisms. For the comparison of GAC (Filtrasorb 400®) and AE (Purolite® A-600), a column experiment was set up. The perfluoroalkane (-alkyl) sulfonic acids (PFSAs) and perfluorooctanesulfonamide (FOSA) had similar removal efficiencies using both GAC and AE, and the efficiency increased with increasing chain length. AE was found to have a higher average removal efficiency of perfluoroalkyl carboxylic acid (PFCAs) (62-95%) than GAC (49-81%). In conclusion, longer chain length PFASs were removed more effectively than shorter-chained, and the PFSAs and FOSA showed higher removal efficiency compared to the PFCAs. Furthermore, linear isomers were removed more effectively than branched for GAC and AE. In contrast, the opposite was found for the NF membrane, where branched isomers were better retained. / Per- och polyfluorerade alkylsubstanser (PFAS) är en grupp syntetiska, ytterst persistenta kemikalier. På grund av deras ytaktiva egenskaper är de lämpliga för användning i många produkter och tillverkningsprocesser, och är således viktiga för en ekonomiskt betydande industri. Under det senaste årtiondet har PFAS påträffats i miljön, levande organismer och kranvatten världen över.  Kombinationen av toxiska egenskaper, en hög bioackumuleringspotential och upptäckten att konventionella reningsmetoder inte avlägsnar substanserna från vatten, gör att vidare forskning av reningsmetoder för PFAS är mycket angelägen. Tre reningsteknikers förmåga att rena vatten från PFAS undersöktes. Nanofiltrering (NF) är en membranfiltreringsteknik som utöver den renade produkten, permeatet, även framställer en biprodukt av hög föroreningsgrad, rententatet. För att rena rententatet har adsorption till granulärt aktivt kol (GAC) eller jonbytarmassa (AE) föreslagits. Teknikerna utvärderades på Bäcklösa Vattenverk i Uppsala.  Nanofiltreringen undersöktes i en pilotanläggning där två 270NF (Dow Filmtech™) membran var seriekopplade. En hög reningsgrad (>90%) konstaterades för alla typer av PFAS. Vidare visades PFAS-koncentrationen i permeatet vara en funktion av PFAS-koncentrationen i råvattnet; en ökad råvattenkoncentration gav en ökad permeatkoncentration. Storleksseparation och elektrostatisk repulsion befanns vara viktiga mekanismer som påverkade reningsgraden. För att undersöka de mekanismer som påverkar PFAS-adsorption jämfördes GAC (Filtrasorb 400®) och AE (Purolite® A-600) i ett kolonnexperiment. Reningsgraden för GAC och AE av perfluorerade sulfonsyror (PFSA) och perfluorooktan sulfonamider (FOSA) var lika hög och reningseffektiviteten ökade med ökande kolkedjelängd. AE återfanns ha en högre genomsnittlig reningsgrad av perfluorkarboxylsyror (PFCA) (62-95%) än GAC (49-81%). Sammanfattningsvis avlägsnades PFAS av längre kolkedjelängd mer effektivt än kortare kolkedjor, och PFAS med sulfonsyror och sulfonamider som funktionella grupper uppvisade en högre reningsgrad än karboxylsyrorna. The proposed diazoniumPFSI zwitterionic polymer (I) is expected to enhance the thermal and chemical stability, increase the proton conductivity of electrolytes, and improve the catalyst efficiency for PEM fuel cells. Synthesis of the perfluorobenzoyl peroxide initiator, homopolymerization of perfluoro (3-oxapent-4-ene) sulfonyl fluoride,coupling reaction with4-sulfamonylacetanilide, couplingreaction with 4-nitrobenzene sulfonyl amide, n-deacetylation reaction, and diazotization reactionhave been carried outsuccessfully in the lab. The intermediate chemicals are characterized by GC-MS, IR, NMR, and GPC spectroscopies.

Stanovení vybraných perfluoroalkylových sloučenin v komplexních matricích / Determination of selected perfluoroalkyl compounds in complex matrices

Diploma work focuses on the determination of selected representatives of perfluoroalkyl substances in complex matrices, particularly, in sewage sludge and feed. In case of sewage sludge, samples were extracted into methanol and three extractions techniques were compared: accelerated Soxhlet, pressurized liquid extraction and Powley method. Powley method showed sufficient efficiency, the lowest matrix effect and minimal background. Perfluorooctane sulfonate was present in all tested samples of sewage sludge (0.74–38.02 ng.g-1). Other detected compounds were perfluorodecanoic acid and perfluorooctanoic acid. Three extraction techniques were tested on feed samples (QuEChERS, fast methanol extraction and Powley method into acetonitrile). Powley method provided the cleanest extracts and showed simultaneously the highest recovery of native perfluoroalkyl substances and the lowest matrix effects. Perfluorooctane sulfonate and perfluorooctanoic acid were rarely present in samples of complete and supplemental feeds, while other perfluoroalkyl substances were found at levels below limit of quantification or they were not detected at all. Perfluorooctane sulfonate (0.100–2.768 ng.g-1), perfluoroundecanoic acid and perfluorotridecanoic acid were determined in all fish meal samples.

Prenatal Exposure to Perfluoroalkyl Acids and Serum Testosterone Concentrations at 15 Years of Age in Female ALSPAC Study Participants

Background: Exposure to perfluorooctane sulfonic acid (PFOS) or to perfluorooctanoic acid (PFOA) increases mouse and human peroxisome proliferator–activated receptor alpha (PPARα) subtype activity, which influences lipid metabolism. Because cholesterol is the substrate from which testosterone is synthesized, exposure to these substances has the potential to alter testosterone concentrations. Objectives: We explored associations of total testosterone and sex hormone–binding globulin (SHBG) concentrations at age 15 years with prenatal exposures to PFOS, PFOA, perfluorohexane sulfonic acid (PFHxS), and perfluoronanoic acid (PFNA) in females. Methods: Prenatal concentrations of the perfluoroalkyl acids (PFAAs) were measured in serum collected from pregnant mothers at enrollment (1991–1992) in the Avon Longitudinal Study of Parents and Children (ALSPAC). The median gestational age when the maternal blood sample was obtained was 16 weeks (interquartile range, 11–28 weeks). Total testosterone and SHBG concentrations were measured in serum obtained from their daughters at 15 years of age. Associations between prenatal PFAAs concentrations and reproductive outcomes were estimated using linear regression models (n = 72). Results: Adjusted total testosterone concentrations were on average 0.18-nmol/L (95% CI: 0.01, 0.35) higher in daughters with prenatal PFOS in the upper concentration tertile compared with daughters with prenatal PFOS in the lower tertile. Adjusted total testosterone concentrations were also higher in daughters with prenatal concentrations of PFOA (β = 0.24; 95% CI: 0.05, 0.43) and PFHxS (β = 0.18; 95% CI: 0.00, 0.35) in the upper tertile compared with daughters with concentrations in the lower tertile. We did not find evidence of associations between PFNA and total testosterone or between any of the PFAAs and SHBG. Conclusions: Our findings were based on a small study sample and should be interpreted with caution. However, they suggest that prenatal exposure to some PFAAs may alter testosterone concentrations in females.

Calibration and Application of Passive Sampling in Drinking Water for Perfluoroalkyl Substances / Kalibrering och tillämpning av passiv provtagning i dricksvatten för perfluoroalkylsubstanser

Perfluoroalkyl substances (PFASs) are global environmental contaminants and a need for monitoring levels has arisen due to their persistency and their ability to bioaccumulate. One relatively novel method of monitoring for both long and short time intervals and generating time-weighted average (TWA) concentrations is passive sampling for which no power, maintenance and supervision is required. The polar organic compound integrative sampler (POCIS) with a weak anion exchange (WAX) sorbent and the POCIS with a hydrophilic-lipophilic balance (HLB) adsorbent were calibrated for PFASs in a laboratory uptake experiment, and applied at a drinking water treatment plant (DWTP) in Stockholm, Sweden.   In the calibration study, all of the 14 studied PFASs were taken up by both passive samplers. Two and three out of the 14 studied PFASs had reached equilibrium after 28 days using POCIS WAX (PFBA and PFTeDA) and POCIS HLB (PFBA, PFPeA and PFTeDA), respectively. The sampling rate (Rs), which is the extracted water in liters per day, ranged between 0.003 and 0.10 L day-1 for the POCIS WAX and between 0.00052 and 0.13 L day-1 for the POCIS HLB. In general, Rs increased with increasing perfluorocarbon chain-length (C4 to C8) and for a perfluorocarbon chain-length longer than C8, Rs decreased with increasing perfluorocarbon chain-length (C8 to C13) for both passive samplers. FOSA had the highest Rs-value (0.10 and 0.13 L day-1) for both POCIS WAX and POCIS HLB, respectively. The POCIS WAX had a higher uptake for the short-chained PFASs PFBA (134 ng after 28 days), PFPeA (410 ng) and PFHxA (834 ng), compared to the POCIS HLB (0.5 ng, 58 ng, and 373 ng, respectively). For all other compounds, the accumulated amounts in the POCIS HLB were in the same range as in the POCIS WAX.   The application of the passive samplers at the DWTP showed that both passive samplers could detect ultra-trace (pg to ng L-1) levels of PFASs. A comparison of the TWA concentration showed that the two passive samplers had a good linear correlation (R2 = 0.63), but the TWA concentrations derived by POCIS WAX was approximately 40% higher compared to POCIS HLB. A comparison between the passive samplers and the grab samples did not show a correlation (R2 = 0.24 for POCIS WAX and R2 = 0.10 for POCIS HLB). The application also included a comparison of the removal efficiency in the conventional DWTP and a pilot plant with additional treatments steps of granulated activated carbon (GAC) and nanofiltration (NF). For the full-scale DWTP the average removal efficiency was 32% and high removal efficiency was observed for PFBA (81%). For the pilot plant, the removal efficiency was 100% for all the detected PFASs in the raw water. / Perfluoroalkylsubstanser (PFAS) har blivit uppmärksammade som globala miljöföroreningar, och ett behov av att övervaka dessa ämnens förekomst i miljön har uppkommit på grund av hög persistens i kombination med hög förmåga att bioackumulera. En relativt ny metod för tidsintegrerad provtagning är så kallad passiv provtagning. En adsorbent placeras i fält och ackumulerar PFAS från vattnet. Tillgång till elektricitet behövs inte, och behov av övervakning och underhåll är minimalt. I denna studie kalibrerades en så kallad ’polar organic compound integrative sampler’ (POCIS) för mätning av PFAS genom upptagsexperiment med två olika adsorbenter: en svag anjons adsorbent (WAX) och en hydrofil-lipofil balanserad adsorbent (HLB). Metodiken tillämpades sedan på vatten från ett dricksvattenverk i Stockholm, Sverige.   Upptagsexperimenten utfördes med 14 PFAS och samtliga togs upp av båda adsorbenterna. Två respektive tre av de studerade PFAS uppnådde jämvikt efter 28 dagar för WAX (PFBS och PFTeDA) samt HLB (PFBA, PFPeA och PFTeDA). Upptagshastigheten (Rs), det vill säga den volym som extraheras per dag, varierade mellan 0,003 och 0,1 L dag-1 för WAX och mellan 0,00052 och 0,13 L dag-1 för HLB. Generellt ökade Rs med en ökande längd på kedjan av perfluorerade kol upp till C8, för att sedan avta med ökande kedjelängd. FOSA hade det högsta Rs-värdet (0,10 och 0,13 L dag-1) för både WAX och HLB. WAX hade ett högre upptag (upp till 134, 410 och 834 ng) för PFAS med kort perfluorerad kolkedja (PFBA, PFPeA respektive PFHxA) jämfört med HLB (upp till 0,5, 58, och 373 ng). Den ackumulerade mängden för alla andra PFAS överensstämde väl mellan de båda provtagarna.   Mätning av PFAS halter i dricksvattenverket med hjälp av POCIS WAX och POCIS HLB visade att även PFAS kunde detekteras även vid miljörelevanta halter. En jämförelse mellan de båda passiva provtagarna visade på ett linjärt samband (R2 = 0,63), men där POCIS WAX hade en tendens att överskatta koncentrationen med ca 40%. En jämförelse mellan de passiva provtagarna och traditionell uppsamlingsprovtagning visade på låg överrensstämmelse (R2 = 0.24 för POCIS WAX och 0.10 för POCIS HLB). Vid tillämpningen gjordes även en beräkning för reningseffektiviteten av PFAS i dricksvattenverket och i en pilotanläggning där ytterligare rening med granulerat aktivt kol (GAC) och nanofiltration (NF) används. I dricksvattenverket var den genomsnittliga reningen 32%, med den högsta reningseffektiviteten för PFBA (81%). I pilotanläggningen var reningen 100% för alla upptäckta PFAS i råvattnet.
