Specific Cation Effects in Biological Systems: Thermodynamic and Spectroscopic Insights

Very specific protein-salt interactions are involved in a multitude of biological phenomena such as protein folding/stability, enzymatic activity, and signal transduction events. In this work, we used two very simple, protein-mimic model biopolymers to obtain a better understanding of specific cation effects operating in aqueous protein environments. The two biopolymers used were Elastin-like Polypeptides (ELPs) and poly(N-isopropylacrylamide) (PNIPAM). ELPs are an especially an ideal model system as these polypeptides can be easily genetically engineered to observe the effect of specific amino acid residues and peptide chain length on these salt interactions. Both of these biopolymers are also highly thermoresponsive as their aqueous solutions undergo a hydrophobic collapse/aggregation induced phase transition process above a lower critical solution temperature (LCST). Thermodynamic measurements of these biopolymers were carried out under various salt solution conditions. Additionally, both of these biopolymers are suitable for making surface specific spectroscopic measurements. Vibrational sum frequeny spectroscopy (VSFS), a non-linear interface sensitive spectroscopic technique, was employed here to investigate biologically relevant cation interactions which occur at peptide/protein surfaces. First, the LCST response of a non-polar ELP and a neutral biopolymer, PNIPAM, was investigated in the presence of 12 different alkali, alkaline-earth metal and transition metal chloride salts. Even though the salt interactions for uncharged proteins are dominated by anions, subtle specific cation effects were also observed. The results followed a direct Hofmeister series for cations. Most alkali cations are excluded from the polar amide regions of proteins. More polarizable cations, however, can solvate the hydrophobic moieties and somewhat counter the salting-out effect of the chloride anion. More charged and hydrated ions like lithium and divalent cations showed a weak interaction to the amide moiety through their hydration shell. The role of acidic amino acid residues in inducing cation specificities was investigated using an aspartate-rich ELP system. Both thermodynamic and spectroscopic data conclusively proved that the negative charge on protein surfaces is the main driving force for cation partitioning and specificity under physiological relevant concentration regimes. Apparent binding constants of carboxylate moieties with cations were determined. This is the first quantitative and thoroughly systematic study of such biologically relevant cation-carboxylate interactions prevalent in enzyme active sites and protein surfaces.

Microgéis termo-responsivos preparados a partir dos polímeros do amido

Esta tese apresenta os estudos realizados quanto à síntese e caracterização de microgéis termo-responsivos de N-isopropilacrilamida (NIPAM) e macromoléculas de origem amilácea em sua composição, sistemas ainda pouco explorados na literatura. Inicialmente, nanopartículas de amido (SNP) foram preparadas por nanoprecipitação para incorporação nos microgéis de interesse, através da dissolução dos grânulos do amido em DMSO/H2O, aplicação de ultrassom e precipitação em não-solvente. As SNPs preparadas foram utilizadas como copolímero para formação de microgéis híbridos do tipo SNP-co-p(NIPAM) através de polimerização em emulsão na presença e na ausência de surfactante. A influência das SNPs nas propriedades dos microgéis foi estudada por MEV, DLS, PZ e SAXS, onde foram observadas mudanças no comportamento físico-químico e estrutura supramolecular dos microgéis. No geral, observaram-se deslocamentos da temperatura de transição de fase para valores maiores (até 35 °C, cerca de 3 °C acima do valor determinado para sistemas contendo p(NIPAM) puro), aumento da estabilidade coloidal e organização dos polímeros presentes no microgel em uma arquitetura caroço-casca. As propriedades de inchamento dos microgéis também foram estudadas através da teoria de Flory-Rehner, onde a temperatura theta e o grau de polimerização entre dois pontos de reticulação foram determinados através do ajuste apropriado do parâmetro de interação (χ). Paralelamente, visando um sistema diferente do descrito acima, foi estudado um novo agente de reticulação baseado nos polímeros do amido, a amilose e a amilopectina. Para isso, os polímeros do amido que compõem as nanopartículas foram modificados para a utilização como agente de reticulação na síntese de microgéis de p(NIPAM). Nesta etapa, uma reação de substituição dos grupos hidroxila dos polímeros do amido foi realizada com o cloreto de acriloíla de forma a inserir insaturações na estrutura dos polímeros, para que estes fossem aplicados como reticulante. O sucesso da reação de modificação foi verificado através de RMN 1H e FTIR, no qual um maior grau de substituição foi obtido quando um tempo maior de reação foi empregado. Microgéis de p(NIPAM) foram então preparados através de polimerização em emulsão utilizando os reticulantes sintetizados. Observou-se a influência do grau de substituição e da concentração do reticulante nos microgéis, analisados por MEV, DLS e UV-Vis com controle de temperatura. As principais mudanças foram relativas à temperatura de transição de fase e tamanho dos microgéis. Utilizando o agente de reticulação com maior grau de substituição, foi possível deslocar a temperatura de transição de fase dos microgéis preparados para valores maiores (35 °C, cerca de 3 °C acima do valor determinado para sistemas contendo p(NIPAM) puro). Observou-se também uma resposta mecânica da rede do microgel, que refletiu no tamanho das partículas, influenciada pela concentração dos agentes de reticulação e do grau de substituição. Reaction performances were evaluated through 1H NMR and FTIR. Then, p(NIPAM) microgels were prepared by emulsion polymerization using the crosslinker synthesized. The degree of substitution and the crosslinker concentration influence were verified and analyzed by SEM, DLS and UV-Vis with temperature control. The main observed changes were related to the phase transition temperature e microgels size. Using the crosslinker with higher substitution degree, it was possible the shift of the phase transition temperature to higher values (~35 °C). A mechanical response, observed trough microgels size, was found, driven by the crosslinker concentration and substitution degree. As higher the concentration and substitution degree of the crosslinker, higher was the swelling resistance observed in microgel structure.

Non-Collinear Second Harmonic Generation in Strontium Barium Niobate

Refractive index measurements of the Strontium-Barium-Niobate (SBN) crystals show that none of the known second-harmonic-generation scheme (SHG) can be hold responsible for the SHG in SBN. Based on observations of the SHG experiments carried out with several compositions of the crystals in different setup-geometries a new model of second harmonic generaion was developed. The new SHG model, domain-induced second-harmonic generation (DISHG), which considers that the needle-like domain structure of this material plays an active role in the quasi phase matching of the produced second harmonic light has been experimentally proved using two different experiments. The new SHG process in the SBN crystals is a potential light source of cylindrically polarized light. The easy way of obtaining cylindrically polarized light with the SBN crystal broadens the potential applications for this material. The (DISHG) allows to investigate several properties of the ferroelectric domains. Using SHG measurements it was possible to analyze the development of the domain densities for domains of different sizes during the poling of the crystal. SHG measurements allow us to determine the minimum length of the ferroelectric domains. It was shown that this does not depend on the [Sr]/[Ba] ratio and domains are not getting longer after the sample was poled, except for the case of doped SBN. The ferroelectric-paraelectric phase transition has also been investigated. From the inflection point of the nonlinear susceptibility as function of the temperature the phase transition temperature was determined. The non fully-linear dependence of the phase transition temperature as function of the [Sr]/[Ba] ratio can be explained by a system of three different sublattices at the crystallographic positions of Strontium and Barium atoms.

Optical and Dielectric Properties of Sr(x)Ba(1-x)Nb(2)O(6)

Several SBN-x crystals of different composition have been investigated using the following methods: Optical absorption in the band gap spectral region, optical absorption of the OH-stretch-mode in the near infrared, Raman scattering, pyroelectric and dielectric measurements.The band edge position depends on the crystal composition in a non-linear manner, thus showing band bowing, typical for mixed systems. A new method has been developed to increase the hydrogen content in the bulk. This doping depends on the composition in an almost linear manner. The observed OH stretch mode spectra have been deconvoluted into three sub bands which can be attributed to different sites in the lattice. The composition dependent spectra have been modelled with a few parameters, using different line shapes and both linear and quadratic dependences of the band position.Raman spectra of several crystals of different composition were recorded for four different scattering configurations. Changes for wave numbers below 500 have been found, but could not attributed to particular modes. A prominent feature at about 600 wave numbers was not disturbed by other modes allowing a decomposition and an assigned of this mode to a certain vibration. It was found that the behaviour of this mode is governed by the [Sr]/[Ba] ratio in the pentagonal channel of SBN-x.The ferroelectric relaxor phase-transition of SBN-x has been studied with pyroelectric measurements. From the nonlinear susceptibility as a function of temperature the phase-transition temperature was deduced using the inflection point. The non fully-linear dependence of the phase-transition temperature as a function of the [Sr]/[Ba] ratio can be explained by a system of three different sublattices for the Strontium and Barium atoms.First results obtained with a setup for measuring the dielectric constant confirmed already reported data of other groups.

Studium membránových vlastností liposomálních systémů pomocí fluorescenční spektroskopie / Study of membrane properties of liposomal systems using fluorescence spectroscopy

The presented diploma thesis is focused on the preparation, characterization and study of membrane properties of liposomal systems which were composed of the neutral phosphatidylcholine (DPPC), cholesterol, negatively charged phosphatidylglycerol (DPPG), polyethylenglycol bounded to phosphatidylethanolamine (PEG5000–PE) and polycation N,N,N-trimethylchitosan (TMC). The influence of individual components and their concentrations on the average particle size, zeta potential and changes in the outer and inner part of the bilayer was investigated. In this matter, methods of dynamic and electrophoretic light scattering and fluorescence spectroscopy with the application of laurdan and DPH probes were used. Based on the above-mentioned parameters, concentrations of components that most suitably influence properties of liposomes in terms of the intended application were selected for the definite complex. It was managed to prepare a liposomal complex stealth liposome–N,N,N-trimethylchitosan, which, due to the optimized composition, could have suitable attributes as a drug delivery system for inhalation administration of biologically active substances.

The Diploma thesis is focus on polymer modified bitumen for warm mix asphalt. The theoretical part of the thesis describes additives which influence the properties of bitumen for their use and reduction of production and service temperatures. Further, the theoretical part describes the conclusions from the researches. This part also describes empirical and functional tests of bituminous binders. The practical part presents the results of individual tests. Tests were performed on samples of non-aged, aged by RTFOT and RTFOT + PAV methods. The results of the work and the assessment of the suitability of bituminous binders are summarized at the end of the thesis.
