Development of a stock cube with functional food characteristics

Thesis submitted in fulfilment of the requirements for the degree Master of Technology: Consumer Science: Food and Nutrition in the Faculty of Applied Sciences at the Cape Peninsula University of Technology 2012 / The chronic diseases of lifestyle, tuberculosis and Human immunodeficiency virus/Acquired immunodeficiency syndrome have a high prevalence in South Africa. These diseases are characterised by oxidative stress and a chronic inflammatory state that contribute to both the development and the acceleration of these diseases. Research into the phytochemical plant food components suggest that these substances could possibly play a vital role in the prevention of such disease. Corn steep liquor (CSL) is a waste product with an exceptionally high polyphenol content and total antioxidant capacity (TAC). This led to the suggestion that it could be utilised in the development of food products with functional food characteristics. Stock cubes, due to the widespread use among consumers of different socio-economic backgrounds, were identified as vehicle for the delivery of the CSL with its phytochemical content. This lead to the development of a stock cube utilising CSL as a source of phytochemical polyphenolic antioxidants with the micronutrients zinc, selenium and copper as added support to immunonutrition, along with iron due to the wide spread prevalence of iron deficiency in the South African population. The acceptability of the developed stock cube was tested by preparing savoury rice and pea soup and having blue collar (n = 50) and white collar (n = 49) participants rate the acceptability on a 9-point hedonic scale, ranging from “dislike extremely” to “like extremely”. The savoury rice received a 41% “like very much” rating followed by a 24% “like moderately” rating. The pea soup was rated even more positively as it received a 42% “like very much” rating and a 29% “like extremely” rating. In addition, among the blue collar participants, significant (p < 0.05 for each) findings occurred with a greater liking of the sample dishes prepared with the developed stock cube by participants of the ethnic Black grouping, with Xhosa as home language and being married, as well as those participants who habitually prepare the meals in the household. A significant (p < 0.05) finding with the white collar participants was a greater liking of the sample dishes prepared with the developed stock cube among those participants who would be willing to make use of a stock cube with health benefits. The developed stock cube was also subjected to six months accelerated shelf-life stability testing, reflecting twelve months real time storage. The parameters measured were microbial growth, oxidative rancidity, antioxidant status and organoleptic changes. All microbial growths tested remained within the acceptable specification ranges. Oxidative rancidity, measured as the peroxide value, was detected analytically at the fifth and sixth samplings, but was not detectable organoleptically. The antioxidant status, measured as the TAC and the total polyphenol content, remained relatively stable during the testing period. Though no noticeable organoleptic changes were observed during the stability testing, there was a darkening of the colour by the second sampling. The results of the acceptance testing, as well as the shelf-life stability testing, support the conclusions that the study objectives of developing a stock cube with functional food characteristics and it being received positively by consumers were achieved. Effect of phytochemicals on estrogen biosynthesis in human breast cancer and placental cells. / CUHK electronic theses & dissertations collection

A breast cancer cell line stably transfected with the CYP19 gene had been employed for aromatase inhibition. Among the phytochemicals tested, the major dietary flavonoids, such as genistein and daidzein, produced very weak inhibition. On the other hand, the red clover isoflavone biochanin A, the hydroxychalcone butein and the red grape phytoalexin resveratrol were found to be effective aromatase inhibitors. Cell proliferation assay had shown that they could inhibit ER-positive cell proliferation induced by testosterone, and the inhibitory effect was specifically attributed to the reduction of estrogen synthesis. In another breast cancer cell line SK-BR-3, resveratrol, biochanin A and genistein inhibited CYP19 both in enzyme and promoter I.3/II transcriptional levels. The element responsible for the inhibition of aromatase by these phytoestrogens should fall within the region between -556 to -446 by upstream of exon II. / Breast cancer is one of the most common cancers affecting women. Estrogen plays an important role in breast cancer initiation and development. The majority of breast tumors are initially dependent upon estrogen to support their growth. Most breast cancers occur in the postmenopausal period. However, the intra-tumoral estradiol (E2) is maintained at a high level equivalent to the pre-menopausal status. High intra-tumoral E2 level in postmenopausal women is sustained by the biosynthesis of estrogens in the tumorous tissue. / Genistein and Biochanin A, ranged from 0.1 to 10 muM, might act as estrogen agonist and induced aromatase activity and promoter I.1 transactivation in ERalpha-transfected SK-BR-3 cells. Pharmacological, phytochemical and safety evaluaton of commercial herbal preparations common in South Africa.

Herbal formulations claimed to cure several medical conditions including skin eruptions, chest pains, wounds, gout, menstrual pains, stress, nervous disorders, microbial and viral infections as well as stomach ailments have recently appeared as part of South African traditional medicine. The formulations consist of mixtures of extracts of different plant parts from several different plant species packaged in labelled bottles or boxes. The mixtures are available for sale in herbal shops and public places. While there has been widespread use of these herbal mixtures, there has been no scientific evidence to support their use. This project was aimed at documenting, validating claimed efficacy and testing the safety of fourteen unregistered commercial herbal preparations commonly sold in Pietermaritzburg. A phytochemical and biological investigation of Sutherlandia Frutescens

Since ancient times, indigenous plants have been used by traditional healers for treating various ailments. Sutherlandia frutescens is one of the most commonly used medicinal plants of southern Africa. This widely distributed plant has been traditionally used to treat cancer and HIV patients; however scientific validation is still in high demand. This research aimed to phytochemically characterise the various extracts prepared and to determine if any chemotypes were present. Subsequent biological characterisation was carried out to preliminary ascertain whether this medicinal plant could have anti-cancer and/or immunemodulating properties and which compounds might be responsible for these actions. Various traditional and organic extracts were prepared. Extracts, fractions and compounds generated were analysed and chemical profiles obtained. Column chromatographic techniques were used to isolate and purify compounds and structure elucidation was carried out using various analytical techniques. Heavy metal content absorption and medicinal potential of Egeria densa (Planch.) Casp

The contamination of heavy metals in the environment is a looming concern worldwide. Egeria densa (Planch) (Submerged aquatic plant) from two ponds: Site A with co-ordinates (32º 48’22.04”S; 26°48’58.79” E) and Site B with co-ordinates (32°48’33.25”S; 26°48’33.25”S) in Alice (Eastern Cape) was evaluated for its ability to absorb heavy metals, phytochemical constituents, antimicrobial activity and ultra-structure using standard analytic procedures. Cadmium (Cd), copper (Cu), iron (Fe), lead (Pb), manganese (Mn), and zinc (Zn) were measured in water, sediments and plant. The concentrations of these metal elements were determined with use of Inductively Coupled Plasma- Optical Emission Spectrometry (ICP-OES). In sediments, the heavy metals (mg/kg) decreased in the order of their average concentration as follows: Fe (40.320) > Zn (1.259) > Pb (0.564) > Mn (0.186) > Cu (0.037) in Pond 1 whereas in Pond 2 Fe (61.527) > Cd (0.999) > Mn (0.648) > Pb (0.586) > Zn (0.156) > Cu (0.045). The highest concentration of Fe was detected in both sites and Cu being the least. The concentrations of the metals in the plants sample (from Pond 1) were found in order of Mn > Pb > Cu > Fe whereas cadmium and zinc were not detected, while the concentration in Pond 2 decreases in order of Zn > Mn > Pb > Cd > Fe > Cu. In the water samples, concentrations of heavy metals (mg/L) decreased in the order of their average concentrations as follows: Pb (35.36) > Fe (3.07) > Mn (0.238) > Cu (0.104), both cadmium and zinc were below the limit of detection in Pond 1, whereas in Pond 2 the concentrations decreased as follows: Pb (13.033) >Fe (1.69) > Cu (0.270) > Mn (0.248) > Cd (0.004) and Zinc was not detected. Phytochemical analyses of the plant extracts revealed the presence of phenols, flavonoids, proanthocyanidin, flavonols, saponins, alkaloid and tannins in all the extracts (water, acetone and n-hexane). Both acetone and water extracts, showed high concentration of proanthocyanidin, while tannin was the lowest in acetone extract. Antimicrobial evaluation using, Gram positive (Staphylococcus aureus, Bacillus pumilus, Bacillus cereus, Streptococcus pyogenes, Enterococcus faecalis) and Gram negative (Klebsiella pneumonia, Escherichia coli, Pseudomonas aeruginosa, Proteus vulgaris and Serratia marcescens) bacteria showed negative results for all the strain, except Streptococcus pyogenes which was inhibited at MIC of 0.1 mg/ml. Scanning electron microscopy (SEM) of ultra-structure of Egeria densa, showed that certain bacteria attached to the leaf, However more work has to be done on E. densa to verify the mechanism by which it accumulates heavy metals. The study shows that E. densa has a potential of accumulating heavy metals especial Manganese in plant.

Antimicrobial, cytotoxic and prelimenary phytochemical analysis of four medicinal plants and their formulation

MSc (Microbiology) / Department of Microbiology / BACKGROUND: Medicinal plants form an important part of the Southern African cultural heritage. Indigenous populations, for example the Vha-Venda people, tend to use medicinal plants in formulations rather than western medicines for health and survival. In order to certify and give scientific credibility to the use of medicinal plants formulations used by Vha-Venda people for the treatment of diseases, several assays were carried out. The present study was aimed at assessing phytochemical content, antimicrobial, antioxidant and cytotoxic activities of four indigenous Venda medicinal plants in a formulation and compare their activity with each plant used individually. METHODS: Peltophorum africanum (roots), Pterocarpus angolensis (bark), Terminalia sericea (roots) and Ximenia caffra (roots) were collected from the Thohoyandou area. The collected plant parts were extracted with methanol and water respectively. Individual plant extracts and Five designed formulations were tested for their antimicrobial activity against Staphylococcus aureus ATCC 25923 (Methicillin Resistant), Staphylococcus aureus ATCC 33591(Methicillin Susceptible), beta lactamase producing Klebsiella pneumonia (ATCC 700603) and extended spectrum beta lactamase producing E. coli (ATCC 35218), four clinical isolates of Candida spp and Cryptococcus neoformans using the Broth dilution method. Minimum bactericidal concentration (MBC) of the extracts was determined by culturing the contents of minimum inhibitory concentration (MIC) on nutrient agar. Similarly, minimum fungicidal concentration (MFC) was also determined by culturing contents of MIC in sabouraud dextrose agar (SDA). Extracts were further assessed for their total phenolic content, total flavonoid content and Qualitative phytochemical analysis. The antioxidant ability of the plants extracts and formulations to scavenge free radical DPPH was also determined. The plant formulations were assessed for their anti-HIV activity using the reverse transcriptase colorimetric assay kit. Cytotoxicity against human lymphatic endothelial cells (HLEC) was determined using MTT assay. RESULTS: Methanolic and aqueous extracts of T. sericea exhibited the best antifungal and antibacterial activities whilst P. angolensis and X. caffra showed poor activities. Methanolic plant formulations showed good activities compared to aqueous formulations. However, Fractional Inhibition Concentration Index showed that there was 1 synergistic interaction, 25 additive interactions and 14 antagonistic interactions between the plant extracts. The methanolic formulation 3 showed the best overall phenolic content at 11.85±0.109 mgGAE/g whilst aqueous X. caffra extract showed the least content at 4.546±0.104 mgGAE/g. Higher total flavonoid contents were seen in methanolic formulation 4 at 2.75±0.02 mgQE/g. Qualitative phytochemical analysis revealed the presence of flavonoids, phenolics, terpenoids, tannins, saponins and steroids in 80% of the tested plant extracts and formulations. All plant extracts and formulations exhibited good antioxidant activity against DPPH. The methanolic formulation showed the best antioxidant activity with IC50 of 0.094 ± 0.33μg/ml. For anti- HIV inhibition, all formulations at 200μg/ml exhibited higher percentage of HIV-1 reverse transcriptase inhibition with methanolic mixture 3 being the best overall at 97.5% activity whilst aqueous mixture5 was the least active with 63.03% inhibition activity. Moreover, the best anti-HIV activity at 100μg/ml was exhibited by methanolic mixture 3 at 71% inhibition. Furthermore, aqueous X. caffra, mixture 2 inhibited 26% and 51% at 12.5mg/ml and 3.125mg/ml respectively. Peltophorum africanum and mixture 5 inhibited 34%, 54% and 43% at 3.125mg/ml, 6.25mg/ml and 12.5 mg/ml respectively of Human Lymphatic Endothelial cells growth. CONCLUSIONS: The results from the study indicated that most of the commonly used traditional medicinal Plants in the Venda region when mixed together have merit for use in traditional medical practice as they have shown good antimicrobial activities, good antioxidant xviii activities, good phytochemical activities and good cell proliferation activity. However some formulations showed antagonistic interaction against bacteria. Some Individual medicinal plants showed toxicity at higher concentrations against immune cells. Whereas formulations promoted cell proliferation, therefore, the use of such individual plants in the treatment of infections should be highly monitored as they may pose a health threat to normal immune cells. Evaluation of phytochemical constituents and mutagenic properties of Coccinia rehmanni And Jatropha zeyheri Plant Extracts

MSc (Microbiology) / Department of Microbiology / Background: The medicinal value of plants lies in some chemical substances that produce a definite physiological action in the human body. The secondary metabolites help the plants to survive hash conditions and could be used by humans as supplements of their health, as foods additives or for medicinal purposes. This bioactive compounds are not always beneficial to human beings, and some of this plants bioactive compounds can be toxic or genotoxic to human cells. This study used several methods to evaluate of phytochemical constituents and mutagenic properties of Coccinia rehmanni and Jatropha zeyheri plant extracts. Methodology: Methanol was used for extraction of the bioactive compounds from the two selected plants, filtered with Whatman filter paper and evaporated with rotary evaporator. The extracts were fractionated using open column chromatography. Chemical and TLC methods were used to determine phytochemicals of the study plants extracts and fractions. The plants extracts and fractions were tested against Vero cell lines in order to evaluate cytotoxicity and genotoxicity of the plants. NucRed and LTR Hoechst 33342 dyes were used for cytotoxicity and genotoxicity respectively. For the evaluation of cytotoxicity and genotoxicity Quantification of live and dead cells for the screening assay was performed using the ImageXpress Micro XLS Widefield Microscope and acquired images analyses using the MetaXpress software and Multi-Wavelength Cell Scoring Application Module. Antimutagenicity of plants extracts was observed using PARP universal colorimetric assay kit. Acquired data was transferred to an EXCEL spreadsheet and data was analyzed. Results and discussion: C. rehmanni (12.03%) yielded more extract than J. Zeyheri (8.20%). the two plants had different compound composition and were in different stages of maturity. The study revealed the domination of Terpenoids, Cardiac glycosides, Phenolic and tannis. With an exception of two fraction fractions all the fractions was found to be toxic to an extent were genotoxicity of such fraction could not be concluded. The reason for such extreme toxicity could be due to the influence of the retained alcohol during rotary evaporation. xvi | P a g e Conclusion: this study provides and add to existing knowledge on the phytochemicals mutagenicity and anti-mutagenicity of C. rehmanni and J. Hydroponic cultivation allows optimizing the greenhouse vertical area, leading to a higher yield, reduces the diseases occurrence and also makes easier crop management and, as a consequence, enhances farmer life quality. However, there are few studies on the adaptation of the crop to this cultivation system, especially focusing concentrations of the nutrient solution and, overall, plant densities. Aiming to study growth, yield and fruit quality of strawberry crop in hydroponics system, two trials were conducted in a plastic greenhouse at Universidade Federal de Pelotas , RS. NFT (nutrient film technique) hydroponic system was employed. It consists of cement tiles cultivation benches. The first trial aimed to study different ionic concentrations of the nutrient solution for cultivar Camarosa and was replicated in two consecutive years (2009/2010). The analyzed experimental factor was the nutrient solution electrical conductivity (EC) at four levels: 0.9, 1.5, 2.1, 2.7dS m-1 (2009) and five levels: 0.7, 1.2, 1.7, 2.2, 2.7 dS m-1 (2010). The second trial was conducted in order to verify the effect of different plant densities on cultivar Camino Real. The experimental factor was plant density at four levels: 15.0, 12.5, 10.7, and 9.3 plants m-2. Fresh and dry matter of leaves, crown, roots and fruit, leaf area, fruit number and average fruit weight were evaluated. Samples of fruits were analyzed regarding to total soluble solids content (TSS), titratable total acidity (TTA)  for both trials of 2010  concentrations of anthocyanins, L-ascorbic acid, total phenolics compounds and 12 antioxidant capacity only for EC trial in 2010. The obtained results of the first trial indicated that EC from 1.2 to 1.5dS m-1 promotes plant growth and fruit yield. Growth and yield decreased when EC is below or above these values. The fruits total soluble solids content, antocyanin and L-ascorbic acid concentrations were increased at 1.7dS m-1 EC. The concentration of total phenolic compounds was increased reducing the EC to 1.2dS m-1. While soluble solids, anthocyanin and L-ascorbic acid content, respectively, decreased by 17, 17 and 15%, fruit yield was increased by 52% and phenolic compounds concentration by 22 %, when the EC decreased from 1.7 to 1.2dS m-1.The average fruit weight was reduced with increasing EC in 2009 and it was not affected in 2010, as well as the acidity and the antioxidant capacity. We can conclude that nutrient solution from 1.2 to 1.5 dS m-1 EC can be used to achieve higher yieldy, without great losses in fruit quality of cultivar Camarosa grown in NFT hydroponic system. The results obtained from the second trial indicated that growth and yield of individual plants of strawberry cultivar Camino Real was not affect by increasing plant density in the rage from 9.3 to 15.0 plants m-2. Growth and yield per square meter, as well as chemical characteristics related to the organoleptic quality of fruits (TSS and TSS/TTA), present linear response to increasing plant density. We suggest the adoption of the plant density of 12.5 plants m-2, which provides high yield, with appropriate organoleptic characteristics of fruits, and reduction in the number of plants required in relation to 15.0 plants m-2 plant density. However, it is necessary to carry out additional studies on cultivar Camino Real in order to promote changes in the system and its management and check the economic feasibility of its cultivation in hydroponics. / A cultura do morangueiro possui grande importância para o setor hortigranjeiro da Região Sul do Rio Grande do Sul, pois permite atingir altas produtividades e alto rendimento econômico, uma vez que as frutas tem mercado garantido e um alto valor. O cultivo hidropônico permite a otimização do espaço vertical do ambiente protegido, resultando em maior produtividade, diminuindo a ocorrência de doenças e, ainda, facilitando os tratos culturais e beneficiando a qualidade de vida do agricultor. Porém, há poucos estudos sobre a adaptação da cultura a este tipo de sistema de produção, principalmente em se tratando de concentrações da solução nutritiva e, sobretudo, de densidades de plantio. Com o objetivo de se estudar o crescimento, a produtividade e a qualidade das frutas de morangueiro em sistema hidropônico, dois experimentos foram realizados em condições de estufa plástica, no Campus da Universidade Federal de Pelotas, RS. O sistema hidropônico empregado era do tipo NFT (técnica da lâmina de nutrientes), constituído de bancadas de cultivo construídas com telhas de fibrocimento. O primeiro experimento teve o objetivo de estudar diferentes concentrações iônicas da solução nutritiva para a cultivar Camarosa e foi repetido em dois anos consecutivos (2009/2010). O fator experimental estudado foi a condutividade elétrica da solução nutritiva (CE) em quatro níveis: 0,9; 1,5; 2,1; 2,7dS m-1 (2009); e cinco níveis: 0,7; 1,2; 1,7; 2,2 e 2,7 dS m-1 (2010). O segundo experimento foi conduzido a fim de verificar o efeito de 9 diferentes densidades de plantio sobre a cultivar Camino Real. O fator experimental estudado foi a densidade de plantio em quatro níveis: 15,0; 12,5; 10,7 e 9,3 plantas m-2. Avaliaram-se a matéria fresca e seca de folhas, coroa, raízes e frutas, a área foliar, o número de frutas e o peso médio das frutas. Amostras das frutas foram analisadas em relação ao teor de sólidos solúveis totais (SST), à acidez total titulável (ATT)  em ambos os experimentos realizados em 2010, às concentrações de antocianinas, ácido L-ascórbico, compostos fenólicos totais e à capacidade antioxidante  somente no experimento sobre CE, em 2010. Os resultados obtidos no primeiro experimento indicam que a CE entre 1,2 e 1,5dS m-1 favoreceu o crescimento da planta e a produtividade das frutas. O crescimento e a produtividade diminuíram quando a CE foi mantida abaixo ou acima destes valores. Os teores de SST, de antocianinas e de ácido L-ascórbico das frutas foram aumentados com a elevação da CE para 1,7dS m-1. A concentração de compostos fenólicos totais elevou-se com a diminuição da CE para 1,2dS m-1. Enquanto os teores de SST, de antocianinas e de ácido L-ascórbico, respectivamente, diminuíram em 17, 17 e 15%, a produtividade foi aumentada em 52% e a concentração de compostos fenólicos totais em 22%, quando a CE diminuiu de 1,7 para 1,2dS m-1. O peso médio das frutas foi reduzido com a elevação da CE em 2009 e não foi afetado em 2010, assim como a ATT e a capacidade antioxidante. Conclui-se que a solução nutritiva com CE entre 1,2 e 1,5dS m-1 pode ser indicada para obter maior produtividade, sem grandes perdas na qualidade das frutas da cultivar Camarosa em sistema hidropônico NFT. Os resultados obtidos no segundo experimento indicam que o crescimento e a produtividade individual das plantas da cultivar de morangueiro Camino Real não foram afetados pela elevação da densidade de plantio, no intervalo entre 9,3 e 15,0 plantas m-2. O crescimento e a produtividade por unidade de área, bem como as características químicas relacionadas à qualidade organoléptica das frutas (concentração de SST e relação SST/ATT), apresentaram resposta linear ao incremento da densidade de plantio. Sugere-se a adoção da densidade de plantio de 12,5 plantas m-2, que proporciona rendimento elevado e frutas com adequadas características organolépticas e redução no número de mudas necessárias, em relação à densidade de 15,0 plantas m-2. Porém, existe a necessidade de desenvolver novos estudos com a cultivar Camino Real, a fim de promover adaptações no sistema e seu manejo e verificar a viabilidade econômica de seu cultivo em hidroponia.

Synthesis, Characterization and Evaluation of Novel Treatment against Resistant Pathogenic Bacteria

MSc (Microbiology) / Department of Microbiology / BACKGROUND: Antibiotic resistance amongst microbial pathogens has become a challenge over past decades, bringing about genuine and frequently deadly contaminations that can't be dealt with by ordinary means. This has led to a search on developing solutions to this problem by searching for new source of antimicrobial agents or chemically altering the existing ones. Traditional medicinal plants and nanoparticles are highly targeted as promising agents to address the challenge. Pyrenacantha grandiflora Baill from Icacenaceae family possess pharmaceutical activities and is used by Vhavenda people to cure gastrointestinal related infections, diarrhea and tooth pain. OBJECTIVES: The present study aimed to synthesize, characterize and evaluate the efficacy of Pyranacantha grandiflora extracts alone and when conjugated with selected nanoparticles against pathogenic microorganisms. Furthermore, this study investigated the efficacy of selected antibiotics when conjugated with nanoparticles against selected pathogenic microbes. METHODS: Pyrenacantha grandiflora Baill (tubers) were collected from Masisi area. Bioactive compounds were extracted using different solvents such as methanol, acetone, hot water, dichloromethane and chloroform. Preliminary phytochemical screening was done to identify different phytochemicals in the extracts and their functional groups were identified by Fourier Transform Infrared spectroscopy (FTIR). Extracts were further assessed for their total phenolic and flavonoids content. Thin layer chromatography was used to separate the compounds from the plant extracts and active compounds/group of compounds were identified by bioautography. The antioxidant ability of the extracts to scavenge free radical DPPH was also determined. Silver and gold nanoparticles were synthesized using chemical and biological methods, characterized by Ultraviolet-Visible Spectrophotometry (UV-VIS), Transmission Electron Microscopy (TEM) and Energy dispersive X-ray analysis (EDX). Plant extracts, nanoparticles and antibiotics were xix conjugated differently, and conjugates were analyzed by FTIR and their antimicrobial activities were evaluated against different bacteria and fungi. The conjugates were tested for antimicrobial activity against extended beta-lactamase producing Escherichia coli (ATCC 35218), Escherichia coli (ATCC 25922), methicillin-resistant Staphylococcus aureus (ATCC 25923), methicillin-susceptible Staphylococcus aureus (ATCC 33591) and beta-lactamase producing Klebsiella pneumonia (ATCC 700603) using agar diffusion assay and the minimum inhibitory concentrations (MIC) were determined using the microdilution method. The minimum bactericidal concentration (MBC) and minimum fungicidal concentration (MFC) were determined by sub-culturing from the MIC plates on Mueller-Hinton Agar. RESULTS: Pyrenacantha grandiflora was found to contain phenolics, saponins, alkaloids, tannin, steroids, terpenoids and flavonoids. FTIR spectroscopic studies revealed different characteristic peak values with various functional compounds similar in most extracts but differed with transmittance values. The total phenolic contents in the examined extracts ranged from 14.167 to 19.02 mg GA/g. The total flavonoid content in the examined extracts ranged from 26.603 to 34.621 mg QE/g. Thin-layer chromatography revealed various Rf values and when analyzed with bioautography, well-defined inhibition zones within the Rf value of 0.236 was identified against E. coli and K. pneumonia. The MICs of the extracts were determined, and all the extracts showed some antimicrobial activity against all tested strains ranging from 0.06-7.5 mg ml/g. Some extracts appeared to be fungicidal and hot water extracts were more active against Cryptococcus neoformans with the MFC value of 0.06 mg/ml. The methanol extract was also active against most tested strains including Candida tropicalis with the minimum fungicidal concentration value of 3.75 mg/ml. Pyrenacantha grandiflora tuber extracts conjugated with silver or gold nanoparticles exhibited a good antibacterial activity against all bacterial strain used and very few were able to exhibit bactericidal activity. Penicillin showed improvement of antibacterial activity xx when conjugated with compounds from the acetone extracts and vancomycin was found to be more effective when conjugated with silver nanoparticles and water extracts. CONCLUSION: The present study validated the efficacy of conjugated P. grandiflora tuber extracts which is used in traditional medicine. The results revealed that water extracts which are generally used by the traditional healers are active against most microorganisms tested as well as methanol and acetone extracts and the synergistic effect was observed when they were conjugated to gold and silver nanoparticles. The results of the present investigation clearly indicate that antimicrobial activity of Pyrenacantha grandiflora Baill tuber when conjugated with selected nanoparticles and antibiotics vary with test strain and the type of solvent used during extraction, thus giving hope for future development of drug leads. / NRF

The nutritional knowledge and consumption of blackjack by hypertensive patients in Vhembe District, Limpopo Province, South Africa

Indigenous vegetables, Blackjack in particular, have been shown to possess anti-hypertensive properties that are attributed to bioactive chemical substances such as phytochemicals, fiber, antioxidants as well as micronutrients that include potassium, magnesium, calcium, vitamin C, zinc, iron and copper. The use of indigenous vegetables in combination with the conventional hypertension treatment may reduce hypertensive conditions; hence, the purpose of this study was to determine Blackjack nutritional knowledge and consumption by hypertensive patients. A cross-sectional study employing both quantitative and qualitative methods was conducted on 275 hypertensive participants at Mphambo and Mutale Health Centers, Vhembe District of Limpopo Province, South Africa. A questionnaire was used to collect data on participants’ Blackjack nutritional knowledge, Blackjack availability around their households as well as their Blackjack consumption patterns and nutritional status. The Blackjack nutritional knowledge data was triangulated with focus group discussions and participants’ nutritional status was assessed by anthropometric and dietary intake methods. The body mass index (BMI) was calculated and dietary intake frequency from selected foods was determined. Blood pressure was measured using Microlife automatic blood pressure monitor. The findings indicated that most hypertensive participants had poor knowledge of the nutritional importance of Blackjack and its associated medicinal contribution on hypertension management. Participants were unable to mention different diseases that can be prevented or managed by Blackjack, which was mostly consumed once a week with porridge during the summer season. Hypertensive patients were consuming high amounts of sodium and less amounts of minerals that are known to regulate blood pressure, such as potassium and magnesium. Hypertensive patients had high prevalence of overweight and obesity and most were either in hypertension stage 1 or stage 2. Lack of Blackjack nutritional knowledge may have contributed to a decreased consumption of Blackjack by hypertensive patients. The health professionals should promote the nutritional benefits of Blackjack in rural areas, particularly to hypertensive patients to raise awareness of Blackjack’s benefits in disease prevention and management. / NRF
