Determinação de ácidos graxos polinsaturados em fórmulas infantis: comparação de metodologias na análise por cromatografia em fase gasosa / Determination of polyunsaturated fatty acid in infant formulas: comparison of quantization methods by gas chromatography

Os ácidos graxos polinsaturados atuam no organismo humano em diversos processos fisiológicos e metabólicos, além de serem importantes na nutrição infantil. A quantificação dos ácidos graxos polinsaturados, devido à presença de vários sítios reativos na molécula, deve envolver processos de extração da gordura em condições amenas. Este trabalho teve como objetivos a comparação dos métodos analíticos para determinação de lipídios totais e ácidos graxos polinsaturados (ácido linoléico, ácido α-linolênico, ácido araquidônico e ácido docosahexaenóico) em fórmula infantil, a quantificação dos ácidos graxos polinsaturados nas fórmulas infantis comerciais e o acompanhamento da estabilidade destes ácidos graxos neste alimento. Foram analisadas 15 amostras, sendo uma amostra da Nacional Institute of Standards and Techonology (NIST 1849) e 14 fórmulas infantis comercializadas no Estado de São Paulo. Os métodos analíticos comparados para extração de lipídios foram: gravimétricos (Bligh Dyer, Roese Gottlieb e hidrólise ácida AOAC 963.15) e por cálculo (AOAC 996.06 e método direto adaptado de Golay et al. (2006)). Para a preparação dos ésteres metílicos de ácidos graxos utilizaram-se metodologias descritas pela IUPAC, Hartman e Lago e método direto adaptado de Golay et al. (2006). Compararam-se diferentes padrões interno, sendo estes dos ésteres metílicos de ácidos graxos 13:0, 21:0 e 23:0 e fatores de resposta para quantificação dos ácidos graxos polinsaturados em relação aos ácidos graxos 16:0, 18:0 e 23:0. O estudo de estabilidade durou 8 meses, e as análises foram realizadas nos meses de março, abril, julho e outubro de 2008. Os melhores resultados, para gordura total e para ácidos graxos polinsaturados, foram obtidos pelo método oficial (Roese Gottilieb). Quanto ao cálculo dos ácidos graxos polinsaturados, o uso do padrão interno 23:0 e o fator de resposta de correção teórico em relação ao 23:0 revelam resultados mais satisfatórios. Das fórmulas infantis comerciais analisadas, 85,7% apresentaram pelo menos um analito em desacordo com a informação nutricional e 100% com relação à legislação brasileira e o Codex Alimentarius. Em relação à estabilidade dos ácidos graxos polinsaturados, apenas três fórmulas infantis não apresentaram diferença estatisticamente significativa (p>0,05) nos teores de ácido graxos no período de 8 meses. / Long chain polyunsaturated fatty acids are involved in several physiological and metabolic process of human organism. The quantification of polyunsaturated fatty acid, must involve the fat extraction in mild conditions, due the reactive sites in the molecule. This work had as objective the comparison of the analytical methods for determination of polyunsaturated fatty acid (linoleic acid, α-linolenic acid, arachidonic acid and docosahexaenoic acid) and lipids in infant formula, quantification of polyunsaturated fatty acids in commercial infant formulas and polyunsaturated fatty acids stability evaluation in these foods. A 15 samples of infant formulas were analyzed, these one from NIST and 14 commercial infant formulas. The analytical methods to lipids extraction were: Bligh and Dyer, Roese Gottlieb, acid hydrolyze AOAC 963.15, acid hydrolyze AOAC 996.06 and direct method adapted from Golay et al. (2006). To prepared esters metilics fatty acids utilized the methods: IUPAC, Hartman and Lago and direct method. The quantization of polyunsaturated fatty acid was realized with different standards internal, like fatty acid methyl ester 13:0, 21:0 and 23:0, and flame ionization detector response factors of correction in relation fatty acids 16:0, 18:0 and 23:0. The stability study during 8 months and analysis were analyzed in the months March, April, July and October. The best results, for lipids and polyunsaturated fatty acid, were obtained by the official method, Roese Gottlieb and metilation followed Hartman and Lago. In accordance with the present study the internal standard 23:0, and the theoretical correction factors with relation 23:0 showed satisfactory trueness and precision for calculation of polyunsaturated fatty acid. Among of commercial infant formula analyzed, 85.7% had at least one analyte in disagreement with the nutritional label facts and 100% with respect to Brazilian Legislation and the Codex Alimentarius. Regarding polyunsaturated fatty acids stability, three infant formulas showed no statistically significant difference (p > 0.05) in levels of these fatty acids during the period of analysis (8 months).

The essential fatty acids omega-3 and omega-6, consumed during pregnancy can benefit maternal and offspring health. The nuclear receptor peroxisome proliferator-activated receptor α (PPARα), when activated by PUFAs enhances lipolytic genes. Liver X receptor α (LXRα) and sterol regulatory element binding protein 1c (SREBP-1c) are nuclear receptors that enhance lipogenic genes, and are repressed by PUFAs. The present study aimed to investigate the effects of diets, rich in omega-3 or omega-6, consumed throughout three generations, on biochemical parameters and the expression level of some genes related to lipid metabolism. For this study we used adult Wistar/UFPel rats, composing the founding generation, females that received diet with flaxseed oil (OM group, n=18) or soybean oil (CTL group, n=18) during pregnancy. At F1 weaning, 48 females offspring were selected and allocated in three groups, females from OM group, that continued receiving diet with flaxseed oil (OM/OM group, n=16), females from OM group, that started to receive diet with soybean oil (OM/CTL group, n=16) and females from CTL group, that continued receiving diet with soybean oil (CTL/CTL group, n=16). At F2 weaning, 16 females offspring of each group were selected and continued receiving the same diets (OM/OM/OM group, OM/CTL/CTL group and CTL/CTL/CTL group). The females were evaluated in the pre-partum (19±1 days of pregnancy) and at postpartum period (21 days after parturition). The dietary fatty acid profile did not affect the diet daily intake and the pregnancy rate (P>0.05), however, dams that received the diet rich in omega-3 had heaviest offspring (P=0.01). Diets with higher omega-3 level decreased triglycerides serum levels, and maintained constant the non-esterified fatty acids (NEFA) levels (P=0.04), while the animals fed with high proportion of omega-6 had fluctuant NEFA levels, and an outstanding increase in the postpartum period of F2 generation (P=0.02). PPARα, RXRα, LXRα and SREBP-1c had been regulated by the diets throughout generations. In the pre-partum moment of F2 generation, the animals fed with high omega-3 had a lower expression of LXRα (P=0.01) indicating its effectiveness on inhibiting lipogenic action. Our results indicate that the PUFAs effect on the control of lipolysis and lipogenesis is cumulative throughout generations and the omega-3 exerts a better control. / Dietas contendo ácidos graxos poli-insaturados (AGPIs) exercem controle negativo sobre a lipogênese hepática e seus efeitos são observados ao nível transcricional. Os ácidos graxos essenciais ômega-3 e ômega-6, quando consumidos durante a gestação podem beneficiar a saúde materna e da prole. O receptor nuclear peroxisome proliferator-activated receptor α (PPARα), quando ativado pelos AGPIs favorecem a atividade de genes lipolíticos. O liver X receptor α (LXRα) e sterol regulatory element binding protein 1c (SREBP-1c) são receptors nucleares que ativam genes lipogênicos, e são reprimidos pelos AGPIs. O presente estudo teve como objetivo investigar os efeitos de dietas, ricas em ômega-3 ou ômega-6, consumidas ao longo de três gerações, sobre parâmetros bioquímicos e o nível de expressão de alguns genes relacionados ao metabolismo lipídico. Para este estudo foram utilizados ratas adultas, da linhagem Wistar/UFPel, compondo a geração fundadora, fêmeas que receberam ração com óleo de linhaça (Grupo OM, n=18) ou óleo de soja (Grupo CTL, n=18) durante toda a gestação. No desmame da F1 foram selecionadas 48 progênies fêmeas, então divididas em três grupos, fêmeas oriundas do Grupo OM, que continuaram a receber dieta com óleo de linhaça (Grupo OM/OM, n=16), fêmeas oriundas do Grupo OM, que passaram a receber dieta com óleo de soja (Grupo OM/CTL, n=16) e fêmeas oriundas do Grupo CTL, que continuaram a receber dieta com óleo de soja (Grupo CTL/CTL, n=16). No desmame da F2 foram selecionadas 16 progênies fêmeas de cada grupo que seguiram recebendo o mesmo tratamento (Grupo OM/OM/OM; Grupo OM/CTL/CTL e Grupo CTL/CTL/CTL). As fêmeas foram avaliadas no período pré parto (19±1 dias de gestação) e no pós-parto (21 dias após o parto). O perfil de ácidos graxo da dieta não afetou o consumo de ração e nem a taxa de prenhez (P>0,05), porém as fêmeas alimentadas com dieta rica em ômega-3 tiveram filhotes mais pesados (P=0,01). A dieta com alto nível de ômega-3 diminuiu os níveis séricos de triglicerídeos (P=0,04), e mantiveram constante os níveis de ácidos graxos não-esterificados (AGNEs), enquanto animais alimentados com uma proporção maior de ômega-6 tiveram níveis flutuantes de AGNEs, e um notável aumento no perído pós-parto da geração F2 (P=0,02). PPARα, RXRα, LXRα and SREBP-1c foram regulados pelas dietas ao longo das gerações. No período pré-parto da geração F2, os animais alimentação com alto ômega-3 apresentaram uma menor expressão de LXRα (P<0,001), indicando sua efetividade em inibir a ação lipogênica. Nossos resultados indicam que os efeitos dos AGPIs no controle da lipólise e lipogênese é cumulativo ao longo das gerações e o ômega-3 exerce um controle mais efetivo.

Etude de la cinétique de l’adaptation respiratoire à la naissance par la résonance magnétique : les effets des acides gras polyinsaturés oméga 3 sur la transition périnatale / Study of the kinetics of respiratory adaptation at birth by magnetic resonance : the effects of omega 3 polyunsaturated fatty acids on the perinatal transition

L’adaptation à la vie extra-utérine requiert la résorption rapide du liquide pulmonaire, la création de la capacité fonctionnelle résiduelle (FRC), ainsi que l’augmentation du débit pulmonaire. Bien que ces mécanismes aient été largement étudiés, leurs cinétiques restent mal connues faute de méthode d’étude non invasive in vivo. L’échec de l’adaptation cardiorespiratoire à la naissance est la principale cause de mortalité et de morbidité de la période néonatale, malgré l’utilisation de stratégie préventive comme la corticothérapie anténatale, et l’amélioration des techniques de réanimation néonatale. Les acides gras polyinsaturés oméga 3 (AGPI n3) que l’on retrouve dans les huiles de poisson ont des effets cardiovasculaires et respiratoires qui pourraient améliorer la transition périnatale. Par ailleurs, leurs propriétés anti-inflammatoires peuvent réduire les conséquences des effets iatrogènes des techniques de réanimation dont la mise en oeuvre est nécessaire en cas de maladaptation cardiorespiratoire à la naissance. Les objectifs de ce travail ont été de : Mettre au point et valider une méthode d’exploration de la cinétique de réabsorption du liquide pulmonaire à la naissance par résonnance magnétique nucléaire Etudier les effets des AGPI n3 sur : l’adaptation ventilatoire à la naissance la circulation pulmonaire périnatale la prévention des lésions pulmonaires induites par une hyperoxie prolongée Les études ont été réalisées sur les modèles expérimentaux de foetus de brebis et de ratons nouveau-nés exposés à une hyperoxie prolongée. Nous montrons que la technique par IRM permet d’évaluer la cinétique de clairance du liquide pulmonaire in vivo en situations physiologique et pathologique. Alors que les AGPI n3 améliorent la circulation pulmonaire, ils n’ont pas d’effet sur la clairance du liquide pulmonaire. Par contre, ils réduisent les lésions pulmonaires induites par l’oxygène. Ces résultats expérimentaux suggèrent que les AGPI n3 pourraient prévenir les échecs d’adaptation cardiorespiratoire à la naissance et ses conséquences à long terme. / Perinatal transition requires clearance of pulmonary fluid, creation of functional respiratory capacity (FRC), and the multiplication of pulmonary blood flow by ten, to ensure gas exchange and blood oxygenation. Physiology of perinatal transition has been broadly studied but the kinetics of its evolution is less known. The aim of our study was to assess the kinetics of the respiratory transition. Besides, we supposed that polyunsaturated fatty-acids omega 3 (n3 PUFA) would improve this transition. To study the kinetic of respiratory transition, we used the MRI in 3 groups of lambs: premature group (Preterm) with a term of 122-124 days, two groups of late preterm lambs (134-136 days), including one group who received antenatal steroids (Late preterm + steroids), and a group without steroids (Late preterm). Surprisingly, we observed a rapid increase of lung fluid in preterm lambs reaching 30% at the end of the experimentation. Creation of FRC was also impaired in this group. In the Late preterm group, clearance of pulmonary fluid was slowed down with a moderate fall of 10%, whereas, in the Late preterm + steroids group, lung fluid dropped by 50% (p<0.05). Similarly, FRC in the Late preterm + steroids group was higher than the other 2 groups (p <0.05). Respiratory functions were closely related to lung fluid content and to FRC. We hypothesized that n3 PUFA would improve perinatal transition using two models: - An acute model: in a chronically instrumented model, catheters were introduced in the jugular and the carotid vessels of lamb fetuses at 122 days of gestation. Lamb fetuses received either Omegaven (n3 PUFA), or Intralipide (n6 PUFA), or isotonique physiological serum (SSI) for 4 days. Respiratory transition was explored by MRI. There was no significant difference between these three groups. Total lung fluid increased by about 20% in Omegaven group and 18% in SSI group (p> 0.05). - A model of Bronchopulmonary dysplasia: pups were exposed to hyperoxia for 10 days, and feeding rats received supplementation by either n3 PUFA (Omacor), n6 PUFA (sunflower oil), or SSI, with control groups that received the same feeding supply but which were exposed to air. Histological studies showed bronchopulmonary dysplasia lesions in the Hyperoxia groups, characterized by decrease of alveolar number, decrease in the number of secondary alveolar septa, and widening of the interstitial space. These lesions were similar in SSI and n6 PUFA groups, whereas n3 PUFA improved these lesions (p <0.05). To assess the effects of n3 PUFA in pulmonary vascular resistances (PVR), we studied their effects on fetal circulation characterized by elevated RVP using a chronically instrumented experimental model of lamb fetus. Lamb fetuses received either n3 PUFA (Omegaven), or n6 PUFA (Intralipid). We then explored the action mechanisms of n3 PUFA: 1) NO pathway using L-Nitro Argenine (LNA), 2) potassium channels pathway using Tetraethylamonium (TEA) and 3) epoxides produced by cytochrome P450 using MS-PPOH. Finally, we investigated the effect of Eicosapentaenoic Acid (EPA) perfusion, or Docosahexaenoic Acid (DHA) perfusion in pulmonary circulation. We showed that, unlike Intralipid, Omegaven induced pulmonary vasodilation, and an increase in pulmonary flow. An identical effect was reproduced by EPA infusion, while DHA did not modify PVR. This response was not modified by LNA. But it was reduced by MS-PPOH, and abolished by TEA. These results indicate that n3 PUFA induced pulmonary vasodilation, mediated by epoxides which act on potassium channels. To conclude, we demonstrated that respiratory transition in preterm lambs even in late preterm ones is altered with the presence of pulmonary edema. n3 PUFA did not improve lung fluid clearance but should be considered in the tretment of pulmonary hypertension, and in the prevention of brochopulmonary dysplasia of the newborns.

Obesity is tightly connected with metabolic diseases including insulin resistance, type 2 diabetes or dyslipidemia. Peroxisome proliferator-activated receptor (PPAR)-α is a key transcription factor involved in the regulation of lipid metabolism, while its activity is stimulated by a variety of hypolipidemic drugs. n-3 polyunsaturated fatty acids (PUFA), including eicosapentaenoic (EPA) and docosahexaenoic (DHA) acid, are endogenous ligands of PPARα, and they are used in the form of fish oil as dietary supplements in order to lower blood lipid levels and to prevent cardiovascular disease. Wax esters represent a novel lipid form of EPA and DHA, and according to recent studies they could exert more potent effects than the classical fish oil (i.e. triacylglycerols). Mice of the 129S1/SvImJ inbred strain were used in the present experiment, and included wild-type (WT) mice, as well as transgenic mice either with the exclusive expression of the human form of PPARα (hPPARα) or mice completely lacking PPARα (PPARα-KO). Differential effects of arachidonic acid and docosahexaenoic acid on cell biology and osteoprotegerin synthesis in osteoblast-like cells

The purpose of the study was to elucidate the mechanisms by which polyunsaturated fatty acids (PUFAs) prevent bone loss. MG-63 human osteoblasts and MC3T3-E1 murine osteoblasts were exposed to the n-6 PUFA arachidonic acid (AA) and the n-3 PUFA docosahexaenoic acid (DHA) as well as oestrogen (E2) and parathyroid hormone (PTH) and the effects thereof tested on a variety of biological parameters characteristic of osteoblasts. These parameters included prostaglandin E2 (PGE2) synthesis, proliferation, differentiation to mature mineralising osteoblasts as well as osteoprotegerin (OPG) and receptor activator of nuclear factor êB ligand (RANKL) secretion. Results showed that AA stimulates PGE2 production significantly in both cell lines. Stimulated PGE2 production by MC3T3-E1 cells however, was significantly higher, which might be attributed to auto-amplification by PGE2 itself in this cell line. Pre-incubation of the MG-63 cells with cyclo-oxygenase (COX)-blockers inhibited PGE2 production significantly, suggesting that both COX enzymes were involved in PGE2 synthesis. The number of functional osteoblasts is important for bone formation therefore in vitro osteoblastic cell proliferation was investigated. In contrast to the hormones E2 and PTH, both AA and DHA inhibited proliferation significantly. The AA-mediated anti-proliferative effect is possibly independent of PGE2 production, as PGE2 per se had little effect on proliferation. DHA inhibited proliferation of MG-63 cells more severely, which might be attributed to the osteosarcoma nature of the MG-63 cells. The anti-proliferative effect of these PUFAs might be attributed to modulation of cell cycle progression or anti-mitotic effects of PUFA peroxidation products. Morphological studies showed apoptotic cells after DHA exposure in MG-63 cells. There is a reciprocal relationship between reduced proliferation and the subsequent induction of cell differentiation in vitro. High basal levels of alkaline phosphatase (ALP) activity, a marker of the mature mineralising osteoblastic phenotype, were detected in MC3T3-E1 cells. Long-term exposure to AA inhibited ALP activity in these cells. This process might be PGE2-mediated. Exposure to PUFAs, however, did not compromise the ability of the MC3T3-E1 cells to differentiate to mature mineralising osteoblasts. In contrast with MC3T3-E1 cells, MG-63 cells demonstrated low basal ALP activity and were unable to differentiate to mature mineralising osteoblasts. In the absence of osteogenic-inducing supplements, PUFAs induced adipocyte-like features that might be due to the expression of high levels of PPARã in this cell line. Lipid-filled vacuoles were absent in the MC3T3-E1 cells suggesting that the MC3T3-E1 cell line may not express PPARã mRNA. The study furthermore demonstrated that PUFAs are able to modulate OPG and RANKL secretion in osteoblasts. AA inhibited OPG secretion dose-dependently in both cell lines, this could be PGE2-mediated. AA dose-dependently stimulated soluble RANKL (sRANKL) secretion in MC3T3-E1 cells thereby affecting the OPG/RANKL ratio in a negative way, supporting various reports that AA and PGE2 do cause bone resorption. No sRANKL could be detected after exposing the MC3T3-E1 cells to DHA suggesting that DHA could be protective to bone. Les accidents cardio-vasculaires représentent la première cause de décès en France. Parmi les facteurs de risque de ces pathologies figure la structure de l’apport lipidique, excédentaire en acides gras saturés, et déficitaire en acides gras polyinsaturés n-3. L’objectif de cette thèse est la mise au point d’un corps gras alimentaire, dont le profil lipidique permet de participer au rééquilibrage des apports en acides gras, avec comme cible la prévention des maladies cardio-vasculaires.Quatre études ont été réalisées pour répondre à cette problématique. Après la mise au point du profil en acides gras optimal, l’impact de la formulation de la phase concrète sur la texture du produit a été étudié afin de concilier les exigences nutritionnelles et les contraintes techniques de sa fabrication et de sa conservation.La mise au point de la formulation de la phase fluide a engendré une étude sur le développement de composés volatils délétères suite à l’addition d’une proportion importante d’huile de poisson dans la formule. Ce travail a mis en évidence deux phénomènes liés au dégagement de ces molécules, l’un attribué à l’oxydation des lipides, l’autre au contact avec l’eau.Le besoin de protéger les nutriments d’intérêt dans la matrice alimentaire a conduit à la mise au point d’une technologie de microencapsulation, originale de par son procédé de fabrication et sa finalité.Enfin, une étude de la biodisponibilité chez l’animal a montré que ce produit alimentaire est un vecteur efficace de ces nutriments d’intérêt que sont les acides gras polyinsaturés à longue chaîne. Apportés dans une telle matrice, ces composés ont modifié le profil lipidique des cellules circulantes et cérébrales, ce qui permet potentiellement d’améliorer le statut inflammatoire et coagulant des animaux / Cardiovascular events are the major cause of death in France. Among the known risk factors, lipid intake is of growing interest, saturated fatty acids being too much consumed, and n-3 polyunsaturated fatty acids not enough. The aim of this PhD study is to design a spread with a lipid profile that can improve the whole lipid intake, and with cardiovascular events prevention as main goal.Four studies were conducted regarding this issue. Following the design of the optimal fatty acid profile for the product, it appeared necessary to study the solid phase recipe impact on the product texture, to achieve both nutritional and technological restrains.The design of the liquid phase recipe generated a study on the development of deleterious volatile compounds following the addition of a high proportion of fish oil in the formula. This work highlighted two phenomena linked to the emission of these molecules, one related to lipid oxidation, and the other to a contact with water.A technology of microencapsulation, which is original because of its production process and aim, was designed to ensure the protection of the active nutrients in the product matrix.Finally, a bioavailability study in rats showed that this food product is a powerful way of providing these nutrients. Incorporated in such a matrix, these molecules modified the lipid profile of the circulating and cerebral cells membrane, potentially improving the inflammatory and clotting status of the animals

Les zones littorales à macrophytes ont un rôle fonctionnel important dans les hydrosystèmes fluviaux. Outre leur forte productivité, ces milieux se caractérisent également par une forte biodiversité. Cependant, les liens pouvant exister entre hétérogénéité spatiale, biodiversité et flux de matière restent encore peu connus. En milieu aquatique, les microcrustacés représentent un maillon clef entre les microorganismes eucaryotes, qui sont les principaux producteurs d’acide gras polyinsaturés (AGPI), et le compartiment piscicole. Dans ce travail de thèse, nous avons donc cherché à savoir comment la diversité des cladocères rencontrée dans une zone littorale à macrophytes pouvait affecter le transfert des AGPI dans les réseaux trophiques. L’approche vers laquelle s’est orienté ce travail combine à la fois des études menées en milieu contrôlé, permettant des expériences de nutrition, ainsi que des études menées en milieu naturel. Cette double approche permet de faciliter l’interprétation des résultats issus des analyses lipidiques, isotopiques et isotopiques de composés spécifiques. Nos résultats montrent ainsi qu’il n’existe pas de variabilité dans les capacités d’accumulation et de bioconversion des AGPI chez les cladocères coexistant dans une zone littorale à macrophytes. D’après ces résultats, il semble donc que le principal facteur influençant la variabilité des transferts d’AGPI vers le compartiment piscicole soit la stratégie alimentaire et le compartiment trophique exploité par les différentes espèces de microcrustacés. En effet, outre le seston, notre étude montre que certaines espèces de cladocères rencontrées en zone littorale sont capables d’exploiter l’épiphyton et le neuston. Les analyses lipidiques mettent de plus en évidence, que la diversité des compartiments trophiques exploités s’accompagne d’une variabilité d’apports en AGPI. Dans notre étude, l’épiphyton est ainsi significativement plus concentré en AGPI que le seston. A l’interface air-eau, le neuston se caractérise de plus, par une forte accumulation de matière organique d’origine allochtone qui, dans le cas d’une pluie de pollen, représente une source de carbone et d’AGPI non négligeable pour certaines espèces de microcrustacés. Dans les zones littorales à macrophytes, la complémentarité des espèces de cladocères entraine donc une utilisation plus complète des ressources. La diversité des compartiments trophiques, associée à la diversité des microcrustacés, permet donc probablement une optimisation du transfert des AGPI vers les niveaux trophiques supérieurs. / Areas with littoral macrophytes play an important functional role in freshwater systems. In addition to the high productivity recorded in such areas, they also shelter a high diversity of organisms. However, possible links between spatial heterogeneity, biodiversity and energy pathways are still poorly known. As they constitute the major link between microorganisms and species higher in the food web, microcrustaceans play a key role in the transfer of polyunsaturated fatty acids (PUFA) to organisms at higher trophic levels. In this study, we wanted to assess if microcrustaceans diversity encountered in macrophytes littoral zones would lead to a variability of PUFA transfer in the food web. This work combined controlled conditions experiment and studies run in natural environments in order to help the interpretation of results from lipid analysis, isotopic analysis and fatty acid isotope analysis. Our results indicate that there are no differences of PUFA concentrations between cladocerans from a macrophyte littoral zone when they were exposed to the same pool of dietary PUFA. Hence, in heterogeneous feeding habitats such as macrophytes zones where these cladocerans often co-exist, foraging behavior of cladoceran species more than differences of metabolism may be crucial for determining PUFA transfer to upper trophic levels. In addition to seston, our study shows indeed that some cladoceran species are able to forage on the epiphytic and neustonic compartments. Lipid analyses highlight moreover that the diversity of trophic compartments lead to a variability of PUFA inputs to primary consumers. In our study, the epiphytic compartment is indeed significantly more concentrated in PUFA than seston. At the air-water interface, neuston is moreover characterized by important allochthonous organic matter accumulation. During a pollen rain, this organic matter represents an important source of carbon and PUFA for some microcrustacean species. In macrophytes littoral zones, cladocerans complementarity leads to a more complete use of PUFA sources. The association of trophic compartment diversity and microcrustacean diversity probably allows an optimization of PUFA transfer to higher trophic levels.

Modulation de l’apport en acides gras polyinsaturés n-3 : intérêt chez le sujet sain et au cours de l’insuffisance rénale chronique / Metabolic effect of omega 3 fatty acids in health and chronic kidney disease

Les omégas trois ont un bénéfice prouvé dans la prévention de maladie cardiovasculaire et l’inflammation. Un apport optimal peut être réalisé avec des modifications diététiques simples permettant d’avoir un enrichissement des membranes cellulaires et un effet métabolique. Le tissu adipeux de part son rôle important dans la genèse du syndrome métabolique semble être une cible importante du traitement par oméga trois. Les patients avec une maladie rénale chronique (MRC) ont un risque cardiovasculaire accru et cumulent les perturbations métaboliques comme le syndrome métabolique et un état micro inflammatoire. Des doses supra physiologiques d’oméga trois ont été utilisés dans le passé dans des études de prévention rénale ou traitement de dyslipidémie. Or l’effet métabolique en fonction de la dose d’oméga 3 n’est pas connu. En accord, avec les études chez le sujet sain, en fonction de la dose administrée, les omégas 3 ont un impact différent métabolique et sur l’expression génique. Des études complémentaires sont nécessaires pour vérifier la faisabilité et l’impact métabolique d’une modification de régime afin de diminuer le rapport n-6/n-3, ainsi que l’effet à long terme des omégas trois chez ces patients. Par ailleurs, les mécanismes impliqués dans les différences de dose réponse devront être caractérisés sur un modèle animal / Omega 3 fatty acids play an important modulatory role in metabolic and inflammatory responses, the progression of atherosclerosis and gene expression. Recent studies suggest their beneficial impact on adipocyte morphology and function. Chronic kidney disease (CKD) patients have an increased cardiovascular morbi-mortality and suffer from a cluster of metabolic disorders. On the basis of previous studies there are reasons to suggest that omega 3 supplementation may offer a host of benefits to CKD patients. Unfortunatly, published studies on the effect of such supplementation are characterized by supra physiological omega 3 doses, that may be difficult to implement for extended periods in one hand and in the other hand the metabolic effect of different doses of omega 3 hasn’t been studied in detail. Simple dietary modifications can help achieve the recommended n-6/n-3 ratio in healthy subjects. In CKD patients supplementation with n-3 shows a differential dose response effect. Further studies are required to test the faisability and metabolic impact of dietary modifications in order to decrease n-6/n-3 ratio and to assess the long term effect of omega supplementation in CKD patients. Regulation of the flow of fatty acids (FA) by futile cycle (TAG/FA cycle) in white adipose tissue (WAT) is an important mechanism of controlling metabolism of FA and therefore its regulation is in the interest of research as a possible therapeutic target in the treatment of obesity and insulin resistance. The study of the murine model suggests that the treatment of the n-3 PUFA with mild caloric restriction (CR) probably induces the TAG/FA cycle mainly in epididymal depot. It also reduces inflammation in WAT and the risk of cardiovascular diseases. We focused on monitoring the gene expression encoding key enzymes of the TAG/FA cycle (PEPCK, ATGL, HSL, DGAT1 and DGAT2) in dorzolumbar (DL), gonadal (GON) and mesenteric (MEZ) depot in a murine model C57BL/6 using qPCR. We were interested in the influence of the composition of fats in the diet, the influence of administration of n-3 PUFA with combination of 10% CR and the difference of gene expression among depots of WAT. The results indicate that the supplementation of high-fat diet with n-3 PUFA contributes mainly to reduction of gene expression for DGAT2 and the combination of n-3 PUFA and CR increases expression of genes influencing the TAG/FA cycle. In the human organism PUFA participate in many physiological and pathological processes this way, where they cause the large spectrum of actions. Onwards the thesis describes the gas chromatography method that is used in chemical analysis of lipids for the fatty acid determination as the gold standard. Experimentally, the fatty acid representation was determined in the blood, muscle, heart, liver and kidney among 26 cadavers divided into two groups according to the presence/absence of inflammatory process at the time of death.
