Δ-9-Tetrahydrocannabinol: Effect on Macromolecular Synthesis in Human and Other Mammalian Cells

The principal psychoactive component of marihuana is Δ-9-tetrahy-drocannabinol. This compound at 10-5 molar concentration in the medium of human cell cultures appeared to inhibit DNA, RNA, and protein synthesis by 50, 40, and 30% respectively, as measured by incorporation of radioactive precursors into acid-insoluble cell fractions in human diploid fibroblasts, human neuroblastoma cells, and mouse neuroblastoma cells. While Δ-9-tetrahydrocannabinol inhibited semiconservative DNA synthesis, it had no effect on DNA repair synthesis in human cells as assayed by the photolysis of 5-bromodeoxyuridine incorporation into DNA during repair after ultraviolet radiation damage. Δ-9-tetrahydrocannabinol also had no effect on rejoining of DNA single-strand breaks induced by γ-rays. The nonspecificity of the inhibition of macromolecular synthesis by Δ-9-THC suggested a possible interference with uptake of radioactive precursors. However, experimentation has shown that this depression of macromolecular synthesis cannot be accounted for by reduced transport of radioactive precursors into the cell because the rate of transport of these precursors into the cell is essentially the same in the presence or absence of Δ-9-THC. Pool sizes of macromolecular precursors as measured radioisotopically (3Hthymidine, 3H-uridine, 14C-leucine) appear to be reduced about 50%, and this reduced pool size could fully account for the reduced macromolecular synthesis seen in the presence of Δ-9-THC. We do not know what causes this apparent reduction of pool sizes in the presence of Δ-9-THC.

The diploma thesis deals with microclimate of a pool hall. This is divided into three parts. The first part describes the theoretical aspects of the environment in swimming pools in general. The second part of the diploma thesis deals with the calculation of the course of specific moisture concentration during the day in the pool hall. Different humidity concentration curves are assessed based on different electable input parameters. Subsequently, suitable setting options are evaluated in order to achieve acceptable environment in the hall in terms of humidity and optimal operating costs. The last part relates to the experimental part. This examines the measurement of evaporation under different boundary conditions and the subsequent comparison with some calculation methods for the calculation of evaporation.

The diploma thesis deals with the design of a new sports and recreational complex in Velké Němčice. The goal is to create a leisure area with a suitable layout, attractive appearance and a wide range of possible activities to be done here. All of these matters would help to improve the public space of the township for its inhabitants and increase the attractiveness of the place for visitors. The thesis looks for suitable functional content for the area, presents proposals for newly built elements and gives an overall proposal for a new sports and recreational complex, which has to deal with the preservation of several existing objects. The individual diploma thesis was preceded by an collectively prepared project dealing with general analysis of Velké Němčice. The strategic plan for development of the township was created as an output of this project. Several goals were set out in the strategic plan. If these goals were fulfilled, it would help to restore and revitalize unsuitable public spaces, to find better use for buildings and to improve the overall image of Velké Němčice. The project did not bring a specific solution to individual problems, but inspired solutions and new perspectives on the issue were presented here. One of these ideas of the strategic plan considered the possibility of creating a new sports and recreational complex. At this point the individual diploma thesis follows up on the project. The thesis deals with the preparation of an urban-architectural study of the defined area and the creation of a new sports and recreational complex. The solved area is logical and functional divided into individual zones with mutual interconnection of these zones and the creation of a new comprehensive public space serving for various activities. In addition to these urban tasks, the thesis also touches on the architectural design of newly designed buildings.

Concept In the measure of town Yverdon Les Bains the baths park is understood like one of the important spot which influences its surroundings. The surroundings give the impression of neutrality because of the differentness of its composition. So the baths park and three historical buildings in it create the most valuable element. It is independent world where people could relax and feel their senses. Free shape in the “free landscape”. It is seemingly haphazard like its surroundings. Although the building has its own inner composition which I call “Small world under large rooftop”. I would like to create atmosphere where would be unique inner space, light and intimate darkness, warm and cold, atmosphere full of nice smell. Each material has its own sound, temperature and surface. The basic element of the baths is water. It is going through the building, connecting all the parts of the baths and creates special atmosphere. People have to know they entered in to the baths world.
