This diploma thesis deals with the optimalization of synthesis of gold nanoparticles and their surface modification allowing their use as contrast agents for in vivo imaging by CT. Gold nanoparticles were prepared by the Turkevich method and characterized by TEM, DLS, MADLS and UV -Vis. Their surface was functionalized with polyethylene glycol containing a thiol group forming a bond with the Au atoms in the surface of gold nanoparticles. The terminal end of the polymer was methylated or containing an aminooxy group forming an orthogonal bond with hyaluronic acid using click-chemistry. The eligibility for in vivo application of the prepared nanoparticles was verified with stability and cytotoxicity tests. The nanoparticles modified by methylated polyethyleneglycol were injected intravenously into a mouse and their application potential as contrast agents were verified by CT.

This work deals with the metal surface wetting problems of molten lead-free solder and monitoring of ongoing processes at the inter-phase interface using the method of evaluation of the height of the molten solder deducted from the video sequences. The work is aimed at evaluating the metal surface wettability, wetting angle determined. Wettability of the metal surface is compared for different types of surface treatments and for different ages of the measured samples. Measurement is performed at the improved workplace, is used to evaluate the newly derived formula for calculating the wetting angle and created program for automatic evaluation of Picture is used to.

The project aim is an optimalization of production of pressure die casting, which serves as a one part of handle in office furniture area. The main emphasis was for mass finishing operation, which serves for rounding of very sharp part edges after die casting operation. A few tests of single mass finishing parameter was carried out and as result was a few partial recommendations for an increasing of production efficiency and improving of manufacturing process, namely an adjustment of separation cycle, proposal of optimum amount of pieces in each single batch and an innovation of chips type and dimensions. The project includes a proposal of a new and more efficiency layout of pieces on the transport pallet.

Selektivní růst gallium-nitridových tenkých vrstev na substráty pokryté maskou z pyrolyzovaného rezistu / Selective gallium nitride thin-film growth on substrates covered by pyrolyzed resist mask

This thesis deals with deposition of GaN thin films and GaN selective growth utilizing pyrolyzed resist masks. Carbon masks were prepared on silicon substrates by electron-beam litography and resist pyrolysis. As a further step, Ga and GaN were deposited on the masked substrates by Moleculer Beam Epitaxy (MBE) method. A selective growth of Ga droplets was achieved. These results were used for preparation of GaN crystallites by pulse deposition. It is also shown that direct MBE deposition of GaN on the masked substrates leads to a selective growth of GaN thin films with GaN film growing only on the areas which are not covered by the carbon mask. The results are explained by enhanced surface diffusion of gallium atoms on the surface of the carbon mask.

Pharmaceuticals are biological active compounds with different functional groups, physico-chemical and biological properties. These chemical compounds are called as “new contaminants” which cumulate in various environmental components. These contaminants input to environment from industrial processes, hospitals and health care institutions or household sources and these pollutants can to negatively interact with environmental components. Pharmaceuticals are separated according to structure and their effects to live organisms. On the basis of drug consumption in Czech Republic the non-steroidal anti-inflammatory drugs are the most using pharmaceuticals, because these drugs can be obtain without prescriptions. The choosing analgesics for study were monitored in waste water from waste waters treatment plant (WWTP Brno – Modřice) and in surface water from two sampling sites of river Kretinka. Pharmaceuticals were monitored in surface waters in Czech Republic, in Scotland (river Thurso) and in Taiwan (river Erren, Agongdian, Yanshuei and canal of Tainan) Solid phase extraction (SPE) is used for extraction of study pharmaceutical from water system. The high performance liquid chromatography (HPLC) with diode array detection or mass spectrometry was used for determination of drugs in surface waters from Czech Republic. The water samples from Scotland and Taiwan were analysed and the high performance liquid chromatography with mass spectrometry (HPLC/MS) was used for determination of pharmaceuticals. The method of HPLC/MS enables the determination of study compounds in the very low range of concentrations (ng ? l-1). All monitored pharmaceuticals were identified and quantified in water samples from river Kretinka in Czech Republic, river Thurso (Scotland) and rivers Erren, Agongdian, Yanshuei and canal of Tainan in Taiwan and the obtained data were compared.

The doctoral thesis is focused on reliability of lead-free solder SAC 305. Knowledge in the field of fatigue models used in determining the lifetime of solder joints are observed in this thesis. Also such methods of predicting reliability as numerically-analytical methods or reliability experimental tests are mentioned. Practical results of reliability measurement are presented. Experimental data served as the foundation for determining empirical coefficients for the fatigue model based on deformation induced by creep of the solder, which was implemented in the ANSYS environment. Results from different methods were compared and conclusions discussing the suitability of the presented prediction methods are formulated.

In metallurgy it is important to know a cooling efficiency of a product as well as cooling efficiency of working rolls to maximize the quality of the product and to achieve the long life of working rolls. It is possible to examine this cooling efficiency by heat transfer coefficients and surface temperatures. The surface temperature is hardly measured during the cooling. It is better to compute it together with heat transfer coefficient by inverse heat conduction problem. The computation is not easy and it uses estimated values which are verified by direct heat conduction problem. The time-consuming of this task can be several days or weeks, depends on the complexity of the model. Thus there are tendencies to shorten the computational time. This doctoral thesis considers the possible way of the computing time shortening of inverse heat conduction problem, which is the parallelization of this task and its transfer to a graphic card. It has greater computing power than the central processing unit (CPU). One computer can have more compute devices. That is why the computing time on different types of devices is compared in this thesis. Next this thesis deals with obtaining of surface temperatures for the computation by infrared line scanner and using of inverse heat conduction problem for the computing of the surface temperature and heat transfer coefficient during passing of a test sample under cooling section and cooling by high pressure nozzles.

Využití separačních metod pro studium vybraných léčiv a pesticidů ve vodách / The Use of Separation Methods for the Study of Selected Drugs and Pesticides in Water

This work is focused on the study of penetration of pesticides and pharmaceuticals in the aquatic environment. The aim was to develop and verify the optimal method for long-term monitoring of the occurrence of selected pesticides and pharmaceuticals in surface waters. Two frequently used analgesics ibuprofen and diclofenac, and carbamazepine that is used to treat epilepsy, were chosen from the group of drugs. A group of 41 pesticides were selected for monitoring too. The attention was focused on a group of urea pesticides and phenoxyacetic acids, as well as on selected pesticides falling to the group of priority substances or identified as environmental quality standards. Some of pesticides were selected because their consumption in the Czech Republic exceeds 30 tons a year. Liquid chromatography with tandem mass spectrometry (LC / MS / MS) was used for the determination of selected analytes in surface waters. Two optimized analytical method were developed. Ten analytes were determined in negative ion mode and 34 analytes in positive ion mode. Solid phase extraction (SPE) was used for purification and concentration of the samples. Developed methodology was validated and used for monitoring of selected compounds in samples of surface waters from the Morava River basin. Samples were collected on the rivers Morava and Svratka monthly for two years. Samples from the river Blata and stream Nivnička were collected for one year. Surface water samples were also collected from the river Litava. Four samples were taken throughout the length of the flow during one day. Smaller streams were chosen for monitoring because of the probability of occurrence of pesticides. They flow abundantly agriculturally cultivated areas. Drugs ibuprofen and diclofenac were detected in all taken samples, carbamazepine mostly. Among the most commonly occurring pesticides were bentazon, diuron, isoproturon, carbendazim, MCPA, MCPP, propiconazole, tebuconazole and 2,4-D.

Stanovení polárních pesticidů v odpadních a povrchových vodách / Determination of polar pesticides in waste and surface waters

This work is focused on the determination of polar pesticides in surface and waste waters. In this study Mecoprop-P, MCPA, Dichlorprop-P, 2,4-D and MCPBA in differently loaded surface waters and communal waters from sewage treatment plants were analyzed. These herbicides are included in the group identified as environmental quality standards. Some of herbicides (2,4-D and MCPA) were selected because their consumption in the Czech Republic exceeds 30 tons a year. Samples were collected prom three rivers near town of Jaroměř (Elbe, Úpa and Metuje Rivers), from one river near the town of Brno (Svratka River) and from inflow and outflow of two sewage treatment plants (WWTP Jaroměř, WWTP Brno – Modřice). Particular rivers were chosen for monitoring due to the probability of occurrence of herbicides, because they flow through agriculturally cultivated areas. Solid phase extraction (SPE) was used for the isolation of target compounds and their concentration. Gas chromatography with tandem mass spectrometry (GC/MS/MS) was used for the determination of target analytes in surface and waste waters. Selected herbicides were detected in all collected samples.
